204. Phylogenetics and phylogeography of human mitochondrial DNA in Island Southeast Asia

Abstract

For more than 20 years, a model for the colonisation ofIsland Southeast Asia (ISEA), involving as two waves of dispersal, has assumed the ~status of near-consensus. The first colonisation of the region by modem humans occurred around 50,000 years ago, but according to the 'Out of Taiwan' model, these populations were assimilated or replaced in the mid-Holocene by rice agriculturists from China, who moved to Taiwan and from there into ISEA and later the Pacific. This theory originated in the analysis of the tree of the Austronesian language family, spoken by most of these populations. In this thesis, mitochondrial DNA variation was analysed using a phylogeographic approach in order to test this hypothesis against other proposed models for the colonization of ISEA. Two improved analytical tools were employed to do this. One was an upgrade of the previously used founder analysis approach, and the second a reassessment of human mitochondrial DNA clock. I developed a time-dependent mutation rate that allows for the effect of purifying selection on the more recent nodes of the human complete mitochondrial tree (control region as well as coding region), increasing both the accuracy and precision of the age estimates. The founder analysis revealed that the Late Pleistocene and Early Holocene were the key periods that shaped the mitochondrial diversity of ISEA, possibly due to events related to climate change and sea-level rise, and that the mid-Holocene Neolithic signal traces mainly to Mainland Southeast Asia, rather than Taiwan. The analysis of complete mitochondrial geno~es suggested a first arrival of modem humans 50,000-60,000 years ago, confmned the importance of sea-level rises in the region, and highlighted the significance of interaction networks in establishing current genetic patterns in ISEA, the Pacific and Taiwan. The analysis offers little support for 'the 'Out of Taiwan' model, pointing to a more complex demographic history for the region in which climatic change appears to have played a crucial role.

Abstract

The aim of this study was to examine the association of social activities with cognitive performance in older adults in the southern area of Brazil, considering the important moderating role of physical vulnerability. A prospective population-based study was conducted in the rural area of Rio Grande, Rio Grande do Sul, Brazil. Self-reported social activities were collected at baseline. Cognitive performance and physical vulnerability were measured in the second wave of data collection. The association of social activities with cognitive performance was determined using robust generalized linear models. In adjusted analysis, the social activities were positively associated with cognitive performance in physically vulnerable older adults. However, this association was not found in those who were nonvulnerable. Our findings may contribute to future investigations of possible explanatory avenues for the association between social activities and cognitive performance as well as the development of interventions aimed at improving cognitive skills. Abstract

Cavernous sinus and superior ophthalmic vein thrombosis is a rare clinical condition, and little described in the literature. The clinical presentation is nonspecific and highly variable, and symptoms may include red eye, ophthalmoplegia, coma, and death. The main etiology results from infection of the paranasal sinuses. The final diagnosis must be made through imaging tests such as magnetic resonance imaging. We describe a case of cavernous sinus and superior ophthalmic vein thrombosis after COVID-19 infection in a 64-year-old patient with persistent ocular hyperemia and pain on eye movement. Ophthalmological examination showed preserved visual acuity, conjunctival hyperemia, dilation of episcleral vessels and retinal vascular tortuosity in the right eye. Magnetic resonance imaging confirmed the diagnosis. The association with the COVID-19 was raised, excluding other infectious causes. Enoxaparin and Warfarin were started with significant improvement in the ocular clinical presentation and maintenance of initial visual acuity after 12 months of follow-up. RESUMO A trombose de seio cavernoso e veia oftálmica superior é uma condição clínica rara e pouco descrita na literatura. A apresentação clínica é inespecífica e altamente variável. Os sintomas podem incluir olho vermelho, oftalmoplegia, coma e morte. A etiologia principal resulta da infecção dos seios paranasais. O diagnóstico final deve ser efetuado por meio de exames de imagem, como ressonância magnética. Descrevemos um caso de trombose de seio cavernoso e veia oftálmica superior após COVID-19 em paciente de 64 anos e com quadro de hiperemia ocular persistente e dor à movimentação ocular. Ao exame oftalmológico, observou-se acuidade visual preservada, hiperemia conjuntival, dilatação de vasos episclerais e tortuosidade vascular retiniana em olho direito. A ressonância confirmou o diagnóstico. A associação com a COVID-19 foi levantada, excluindo-se demais causas infecciosas. Prescrevemos enoxaparina e varfarina, com melhora do quadro clínico ocular e manutenção da acuidade visual inicial após 12 meses de acompanhamento.
