218. Periostin identified as a potential biomarker of prostate cancer by iTRAQ-proteomics analysis of prostate biopsy

Abstract

Abstract Background Proteomics may help us better understand the changes of multiple proteins involved in oncogenesis and progression of prostate cancer(PCa) and identify more diagnostic and prognostic biomarkers. The aim of this study was to screen biomarkers of PCa by the proteomics analysis using isobaric tags for relative and absolute quantification(iTRAQ). Methods The patients undergoing prostate biopsies were classified into 3 groups according to pathological results: benign prostate hyperplasia (BPH, n = 20), PCa(n = 20) and BPH with local prostatic intraepithelial neoplasm(PIN, n = 10). Then, all the specimens from these patients were analyzed by iTRAQ and two-dimensional liquid chromatography-tandem mass spectrometry (2DLC-MS/MS). The Gene Ontology(GO) function and the transcription regulation networks of the differentially expressed were analyzed by MetaCore software. Western blotting and Immunohistochemical staining were used to analyze the interesting proteins. Result A total of 760 proteins were identified from 13787 distinct peptides, including two common proteins that enjoy clinical application: prostate specific antigen (PSA) and prostatic acid phosphatase(PAP). Proteins that expressed differentially between PCa and BPH group were further analyzed. Compared with BPH, 20 proteins were significantly differentially up-regulated (>1.5-fold) while 26 were significantly down-regulated in PCa( Conclusion Our study indicates that the iTRAQ technology is a new strategy for global proteomics analysis of the tissues of PCa. A significant up-regulation of periostin in PCa compared to BPH may provide clues for not only a promising biomarker for the prognosis of PCa but also a potential target for therapeutical intervention.

Abstract

We engineered a machine learning approach, MSHub, to enable auto-deconvolution of gas chromatography-mass spectrometry (GC-MS) data. We then designed workflows to enable the community to store, process, share, annotate, compare and perform molecular networking of GC-MS data within the Global Natural Product Social (GNPS) Molecular Networking analysis platform. MSHub/GNPS performs auto-deconvolution of compound fragmentation patterns via unsupervised non-negative matrix factorization and quantifies the reproducibility of fragmentation patterns across samples.

Abstract

Minority Stress in lesbian, gay, and bisexual people (LGBs) might have detrimental effects on relationship satisfaction. However, less is known regarding the potential mechanisms among the association above in LGBs' romantic relationships. In the present study, we investigated the spillover and buffering impact of minority stress on relationship satisfaction among LGB individuals. A targeted sampling strategy was used to recruit LGBs (N= 1481) for a cross-sectional, online survey. Participants (M age = 35.05 years; 53% men) completed a survey that captured minority stress (i.e., internalized homonegativity, stigma consciousness, and concealment of sexual orientation); intra-/extra-dyadic stress and dyadic coping; and relationship satisfaction. Rigorous latent moderated structural equations was used to test associations between variables. Results indicated that the final model demonstrated acceptable fit. Minority stress was negatively associated with relationship satisfaction only via heightened intra-dyadic stress. Dyadic coping moderated the spillover effect between minority stress and intra-dyadic stress. This study highlights the importance of investigating dyadic coping in romantic relationships in LGBs to gain an insight into these relationships and the associated processes. Abstract

The naturally persistent flow of hundreds of dust particles is experimentally achieved in a dusty plasma system with the asymmetric sawteeth of gears on the electrode. It is also demonstrated that the direction of the dust particle flowcan be controlled by changing the plasma conditions of the gas pressure or the plasma power. Numerical simulations of dust particles with the ion drag inside the asymmetric sawteeth verify the experimental observations of the flow rectification of dust particles. Both experiments and simulations suggest that the asymmetric potential and the collective effect are the twokeys in this dusty plasma ratchet.With the nonequilibrium ion drag, the dust flow along the asymmetric orientation of this electric potential of the ratchet can be reversed by changing the balance height of dust particles using different plasma conditions., Comment: 5 pages,5 figures

Abstract

Superradiance and subradiance concerning enhanced and inhibited collective radiation of an ensemble of atoms have been a central topic in quantum optics. However, precise generation and control of these states remain challenging. Here we deterministically generate up to 10-qubit superradiant and 8-qubit subradiant states, each containing a single excitation, in a superconducting quantum circuit with multiple qubits interconnected by a cavity resonator. The $\sqrt{N}$-scaling enhancement of the coupling strength between the superradiant states and the cavity is validated. By applying appropriate phase gate on each qubit, we are able to switch the single collective excitation between superradiant and subradiant states. While the subradiant states containing a single excitation are forbidden from emitting photons, we demonstrate that they can still absorb photons from the resonator. However, for even number of qubits, a singlet state with half of the qubits being excited can neither emit nor absorb photons, which is verified with 4 qubits. Abstract

We report on deterministic generation of 18-qubit genuinely entangled Greenberger-Horne-Zeilinger (GHZ) state and multi-component atomic Schr\"{o}dinger cat states of up to 20 qubits on a quantum processor, which features 20 superconducting qubits interconnected by a bus resonator. By engineering a one-axis twisting Hamiltonian enabled by the resonator-mediated interactions, the system of qubits initialized coherently evolves to an over-squeezed, non-Gaussian regime, where atomic Schr\"{o}dinger cat states, i.e., superpositions of atomic coherent states including GHZ state, appear at specific time intervals in excellent agreement with theory. With high controllability, we are able to take snapshots of the dynamics by plotting quasidistribution $Q$-functions of the 20-qubit atomic cat states, and globally characterize the 18-qubit GHZ state which yields a fidelity of $0.525\pm0.005$ confirming genuine eighteen-partite entanglement. Our results demonstrate the largest entanglement controllably created so far in solid state architectures, and the process of generating and detecting multipartite entanglement may promise applications in practical quantum metrology, quantum information processing and quantum computation., Comment: 10 pages, 6 figures, 1 table including supplementary material
