Moso bamboo is excellent candidate for cadmium (Cd)/lead (Pb) phytoremediation, while rhizosphere microbiome has significant impact on phytoremediation efficiency of host plant. However, little is known about the rhizosphere bacterial communities of moso bamboo in Cd/Pb contaminated soils. Therefore, this study investigated the assembly patterns and key taxa of rhizosphere bacterial communities of moso bamboo in Cd/Pb polluted and unpolluted soils, by field sampling, chemical analysis, and 16S rRNA gene sequencing. The results indicated α-diversity between Cd/Pb polluted and unpolluted soils showed a similar pattern ( p  > 0.05), while β-diversity was significantly different ( p  < 0.05). The relative abundance analysis indicated α-proteobacteria (37%) and actinobacteria (31%) were dominant in Cd/Pb polluted soils, while γ-proteobacteria (40%) and α-proteobacteria (22%) were dominant in unpolluted soils. Co-occurrence network analysis indicated microbial networks were less complex and more negative in polluted soils than in unpolluted soils. Mantel analysis indicated soil available phosphorus, organic matter, and available Pb were the most important environmental factors affecting microbial community structure. Correlation analysis showed 11 bacterial genera were significantly positively related to Cd/Pb. Overall, this study identified the bacterial community composition of bamboo rhizosphere in responding to Cd/Pb contamination and provides a theoretical basis for microbe-assistant phytoremediation in the future.

The intensive use of chemical fertilizers in China to maintain high crop yields has led to significant environmental degradation and destabilized crop production. Returning straw to soil presents a potential alternative to reduce chemical fertilizer requirements and enhance soil fertility. This study investigates the effects of different nitrogen (N) input levels and straw additions on crop phosphorus (P) uptake and soil P availability based on a long-term N-fertilizer trial. The treatments included no fertilizer input (CK), conventional (NPK), reduced NPK (0.75NPK), and straw-amended (SNPK) treatments. Results indicate that SNPK significantly enhances shoot P uptake and crop yields by 43.7-61.9% and 29.3-39.6%, respectively. The SNPK treatment improved rhizosphere P availability and increased the phosphorus activation coefficient (PAC) by 1.72-fold compared to NPK alone. The enhanced soil P availability under SNPK was primarily attributed to an abundance of functional microbes, leading to higher P storage in the microbial biomass P pool and its turnover. Additionally, SNPK promoted root exudate and phosphate-mobilizing microbes, enhancing P mobilization and uptake. Nitrogen fertilization primarily influenced root functional traits related to P acquisition. These findings provide valuable insights for developing effective fertilizer management strategies in maize-oilseed rape rotation systems, emphasizing the benefits of integrating straw with chemical fertilizers.

It is unknown why roses are terpene-rich, what the terpene biosynthetic pathways in roses are, and why only a few rose species produce the major components of rose essential oil. Here, we assembled two high-quality chromosome-level genomes for Rosa rugosa and Rosa multiflora. We also re-sequenced 132 individuals from the F1 progeny of Rosa chinensis and Rosa wichuraiana and 36 of their related species. Comparative genomics revealed that expansions of the 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR) and terpene synthases (TPSs) gene families led to the enrichment of terpenes in rose scent components. We constructed a terpene biosynthesis network and discovered a TPS-independent citronellol biosynthetic pathway in roses through gene functional identification, genome-wide association studies (GWASs), and multi-omic analysis. Heterologous co-expression of rose citronellol biosynthetic genes in Nicotiana benthamiana led to citronellol production. Our genomic and metabolomic analyses suggested that the copy number of NUDX1-1a determines the citronellol content in different rose species. Phenylpropanoid biosynthesis plays crucial roles in the adaptation to cadmium (Cd) stress. Nevertheless, few reports have dabbled in physiological mechanisms of such super pathway regulating Cd accumulation in plants. Herein, by integrating transcriptomic, histological and molecular biology approaches, the present study dedicated to clarify molecular mechanism on how rice adapt to Cd stress via phenylpropanoid biosynthesis. Our analysis identified that the enhancement of phenylpropanoid biosynthesis was as a key response to Cd stress. Intriguingly, POD occupied a significant part in this process, with the number of POD related genes accounted for 26/29 of all upregulated genes in phenylpropanoid biosynthesis. We further used SHAM (salicylhydroxamic acid, the POD inhibitor) to validate that POD exhibited a negative correlation with the Cd accumulation in rice tissues, and proposed two intrinsic molecular mechanisms on POD in contributing to Cd detoxification. One strategy was that POD promoted the formation of lignin and CSs both in endodermis and exodermis for intercepting Cd influx. In detail, inhibited POD induced by external addition of SHAM decreased the content of lignin by 50.98-66.65 % and delayed percentage of the D TIP-CS to root length by 39.17-104.51 %. The other strategy was expression of transporter genes involved in Cd uptake, including OsIRT1, OsIRT2, OsZIP1 and OsZIP, negatively regulated by POD. Flowering is a vital agronomic trait that determines the economic value of most ornamental plants. The flowering time of rose (Rosa spp.) is photoperiod insensitive and is thought to be tightly controlled by light intensity, although the detailed molecular mechanism remains unclear. Here, we showed that rose plants flower later under low-light (LL) intensity than under high-light (HL) intensity, which is mainly related to the stability of PHYTOCHROME-INTERACTING FACTORs (RcPIFs) mediated by OPEN STOMATA 1-Like (RcOST1L) under different light intensity regimes. We determined that HL conditions trigger the rapid phosphorylation of RcPIFs before their degradation. A yeast two-hybrid screen identified the kinase RcOST1L as interacting with RcPIF4. Moreover, RcOST1L positively regulated rose flowering and directly phosphorylated RcPIF4 on serine 198 to promote its degradation under HL conditions. Additionally, phytochrome B (RcphyB) enhanced RcOST1L-mediated phosphorylation of RcPIF4 via interacting with the active phyB-binding motif. RcphyB was activated upon HL and recruited RcOST1L to facilitate its nuclear accumulation, in turn leading to decreased stability of RcPIF4 and flowering acceleration. The petals of rose (Rosa sp.) flowers determine the ornamental and industrial worth of this species. The number of petals in roses was previously shown to be subject to fluctuations in ambient temperature. However, the mechanisms by which rose detects and responds to temperature changes are not entirely understood. In this study, we identified short interstitial telomere motifs (telo boxes) in the second intron of AGAMOUS (RcAG) from China rose (Rosa chinensis) that play an essential role in precise temperature perception. The second intron of RcAG harbors two telo boxes that recruit telomere repeat binding factors (RcTRBs), which interact with CURLY LEAF (RcCLF) to compose a repressor complex. We show that this complex suppresses RcAG expression when plants are subjected to low temperatures via depositing H3K27me3 marks (trimethylation of lysine 27 on histone H3) over the RcAG gene body. This regulatory mechanism explains the low-temperature-dependent decrease in RcAG transcript levels, leading to the production of more petals under these conditions. Autonomous driving has the potential to revolutionize transportation, but developing safe and reliable systems remains a significant challenge. Reinforcement learning (RL) has emerged as a promising approach for learning optimal control policies in complex driving environments. However, existing RL-based methods often suffer from low sample efficiency and lack explicit safety constraints, leading to unsafe behaviors. In this paper, we propose a novel framework for safe reinforcement learning in autonomous driving that addresses these limitations. Our approach incorporates a latent dynamic model that learns the underlying dynamics of the environment from bird's-eye view images, enabling efficient learning and reducing the risk of safety violations by generating synthetic data. Furthermore, we introduce state-wise safety constraints through a barrier function, ensuring safety at each state by encoding constraints directly into the learning process. Experimental results in the CARLA simulator demonstrate that our framework significantly outperforms baseline methods in terms of both driving performance and safety. Our work advances the development of safe and efficient autonomous driving systems by leveraging the power of reinforcement learning with explicit safety considerations.

Carbon-fixing functional strain-loaded biochar may have significant potential in carbon sequestration given the global warming situation. The carbon-fixing functional strain Bacillus cereus SR was loaded onto rice straw biochar pyrolyzed at different temperatures with the anticipation of clarifying the carbon sequestration performance of this strain on biochar and the interaction effects with biochar. During the culture period, the content of dissolved organic carbon (DOC), easily oxidizable organic carbon, and microbial biomass carbon in biochar changed. This finding indicated that B. cereus SR utilized organic carbon for survival and enhanced carbon sequestration on biochar to increase organic carbon, manifested by changes in CO 2 emissions and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) enzyme activity. Linear regression analysis showed that the strain was likely to consume DOC on 300 °C biochar, although the Rubisco enzyme activity was higher. In contrast, the strain had a higher carbon sequestration potential on 500 °C biochar. Correlation analysis showed that Rubisco enzyme activity was controlled by the physical structure of the biochar. Rice (Oryza sativa) is one of the major cereal crops and takes up cadmium (Cd) more readily than other crops. Understanding the mechanism of Cd uptake and defense in rice can help us avoid Cd in the food chain. However, studies comparing Cd uptake, toxicity, and detoxification mechanisms of leaf and root Cd exposure at the morphological, physiological, and transcriptional levels are still lacking. Therefore, experiments were conducted in this study and found that root Cd exposure resulted in more severe oxidative and photosynthetic damage, lower plant biomass, higher Cd accumulation, and transcriptional changes in rice than leaf Cd exposure. The activation of phenylpropanoids biosynthesis in both root and leaf tissues under different Cd exposure routes suggests that increased lignin is the response mechanism of rice under Cd stress. Moreover, the roots of rice are more sensitive to Cd stress and their adaptation responses are more pronounced than those of leaves. Quantitative PCR revealed that OsPOX, OsCAD, OsPAL and OsCCR play important roles in the response to Cd stress, which further emphasize the importance of lignin. The "Big Five" European football leagues, comprising England's Premier League, Germany's Bundesliga, Spain's La Liga, Italy's Serie A, and France's Ligue 1, command significant attention. While the occurrence of goals, substitutions, and fouls in football games is often considered random, of the presence of an inherent inevitability is unclear. To investigate, we analyzed a public dataset detailing timing of goals, substitutions, and yellow cards in regular time from WhoScored across three seasons (2018-2019, 2019-2020, 2020-2021) in the top five European football leagues. We employed various mathematical descriptive models (including linear, sigmoid, and gaussian functions) to measure the temporal tendency of goals, substitutions, and yellow cards. Our results indicate that, whether in the first or second half of the match, the temporal distribution of these elements exhibits evenness a (indicative of randomness). However, specific characteristics were discerned through distinct model parameters, capturing novel phenomena that were intuitively illustrated. Furthermore, we explored the interaction of the timing of goals, substitutions, and yellow cards. In this analysis we found that scoring in the second half leads to more substitutions and yellow cards. Changing players in the second half corresponded with more goals, while the impact of yellow card fouls showed no differences in goals in the first and second halves. Crop straw return is a widely used agricultural management practice. The addition of crop straw significantly alters the pool of dissolved organic matter (DOM) in agricultural soils and plays a pivotal role in the global carbon (C) cycle, which is sensitive to climate change. The DOM concentration and composition at different soil depths could regulate the turnover and further storage of organic C in terrestrial systems. However, it is still unclear how crop straw return influences the change in DOM composition in rice paddy soils. Therefore, a field experiment was conducted in which paddy soil was amended with crop straw for 10 years. Two crop straw-addition treatments [NPK with 50% crop straw (NPK+1/2S) and NPK with 100% crop straw (NPK + S)], a conventional mineral fertilization control (NPK) and a non-fertilized control were included. Topsoil (0-20 cm) and subsoil (20-40 cm) samples were collected to investigate the soil DOM concentration and compositional structure of the profile. Soil nutrients, iron (Fe) fraction, microbial biomass carbon (MBC), and concentration and optical properties (UV-Vis and fluorescence spectra) of soil DOM were determined. Here, we found that the DOM in the topsoil was more humified than that in the subsoil. The addition of crop straw further decreased the humidification degree of DOM in the subsoil. In crop straw-amended topsoil, microbial decomposition controlled the composition of DOM and induced the formation of aromatic DOM. In the straw-treated subsoil, selective adsorption by poorly crystalline Fe(oxyhydr)oxides and microbial decomposition controlled the composition of DOM. In particular, the formation of protein-like compounds could have played a significant role in the microbial degradation of DOM in the subsoil. Cadmium (Cd) is readily absorbed by rice and enters the food chain, posing a health risk to humans. A better understanding of the mechanisms of Cd-induced responses in rice will help in developing solutions to reduce Cd uptake in rice. Therefore, this research attempted to reveal the detoxification mechanisms of rice in response to Cd through physiological, transcriptomic and molecular approaches. The results showed that Cd stress restricted rice growth, led to Cd accumulation and H 2 O 2 production, and resulted cell death. Transcriptomic sequencing revealed glutathione and phenylpropanoid were the major metabolic pathways under Cd stress. Physiological studies showed that antioxidant enzyme activities, glutathione and lignin contents were significantly increased under Cd stress. In response to Cd stress, q-PCR results showed that genes related to lignin and glutathione biosynthesis were upregulated, whereas metal transporter genes were downregulated. Further pot experiment with rice cultivars with increased and decreased lignin content confirmed the causal relationship between increased lignin and reduced Cd in rice. Gene regulation via intragenic sequences is becoming more recognized in many eukaryotes. However, the intragenic sequences mediated gene expressions in response to environmental stimuli have been largely uncharacterized. Here, we showed that the first intron of RrKSN from the Rosa rugosa cultivar 'Purple branch' had a positive effect on RrKSN expression, and the effect depends on its position and orientation. Further analyses revealed that the four adjacent cis-elements (T)CGATT/AATCG(A) within the first intron were critical for the positive regulation, and the RrKSN promotion was significantly suppressed with mutations of these elements. These cis-elements were further evidenced as binding sites for RrARR1, the homologous of Arabidopsis type-B ARABIDOPSIS RESPONSE REGULATOR 1 (ARR1) transcription factor. The first intron-mediated RrKSN expression was enhanced with over-expressing of RrARR1, but abolished with RrARR1 silencing in rose seedlings. Moreover, the expression difference of RrKSN between 16°C and 28°C was eliminated along with RrARR1-silencing. Taken together, these results suggested both RrARR1 and its binding elements are required for the first intron-mediated RrKSN expression in response to varying temperatures. Ferromanganese oxide biochar composite (FMBC) is an efficient remediation material for cadmium -contaminated soils. However, the effect of FMBC under varied water managements on the remediation of Cd-polluted soil is unclear. In this study, we conducted both incubation and field experiments to investigate the combined effects of corn-stover-derived biochar modified with ferromanganese on the immobilization and uptake of Cd by rice under continuous aerobic (A), aerobic-flooded (AF), and flooded-aerobic (FA) water management regimes. The results showed that loading iron-manganese significantly increased the maximum sorption capacity (Qm) of Cd on FMBC (50.46 mg g -1 ) due to increased surface area, as compared to the pristine biochar (BC, 31.36 mg g -1 ). The results revealed that soil Eh and pH were significantly affected by FMBC and it's synergistic application with different water regimes, thus causing significant differences in the concentrations of DTPA-extractable Cd under different treatments. The lowest DTPA-extractable Cd content (0.28-0.46 mg -1 ) was observed in the treatment with FMBC (2.5 %) combined FA water amendment, which reduced the content of available Cd in soil by 2.63-28.4 %. Moreover, the treatments with FMBC-FA resulted the proportion of residual Cd increased by 22.2 % compared to the control. Variations in the content and fraction of Cd had a significant influence on its accumulation in the rice grains. The FMBC-FA treatments reduced the Cd concentration in roots, shoots and grains by 37.97 %, 33.98 %, and 53.66 %, respectively, when compared with the control. Predominantly because of the reduction in Cd biological toxicity and the improved soil nutrient content, the combined application increased the biomass and yield of rice to some extent. Ecosystem succession and pedogenesis reshuffle the composition and turnover of dissolved organic matter (DOM) and its interactions with soil microbiome. The changes of these connections are especially intensive during initial pedogenesis, e.g. in young post-glacial areas. The temporal succession and vertical development of DOM effects on microbial community structure remains elusive. Using Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR MS), high-throughput sequencing, and molecular ecological networks, we characterized the molecular diversity of water-extractable DOM and identified its links to microbial communities in soil profiles along deglaciation chronosequence (12, 30, 40, 52, 80, and 120 years) in the southeastern Tibetan Plateau. Low-molecular-weight compound content decreased, whereas the mid- and high-molecular-weight compounds increased with succession age and soil depth. This was confirmed by the increase in double bond equivalents and averaged oxygen-to‑carbon ratios (O/C), and decrease in hydrogen-to‑carbon ratios (H/C), which reflect DOM accumulation and stabilization. Microbial community succession shifted towards the dominance of oligotrophic Acidobacteria and saprophytic Mortierellomycota, reflecting the increase of stable DOM components (H/C < 1.5 and wider O/C). Less DOM-bacterial positive networks during the succession reduced specialization of labile DOM production (such as lipid- and protein-like compounds), whereas more DOM-fungal negative networks increased specialization of stable DOM decomposition (such as tannin- and condensed aromatic-like compounds). Introduction: This purpose of this work is to give a detailed description of a surgical technique for frameless robot-assisted asleep deep brain stimulation (DBS) of the centromedian thalamic nucleus (CMT) in drug-resistant epilepsy (DRE)., Methods: Ten consecutively enrolled patients who underwent CMT-DBS were included in the study. The FreeSurfer "Thalamic Kernel Segmentation" module and experience target coordinates were used for locating the CMT, and quantitative susceptibility mapping (QSM) images were used to check the target. The patient's head was secured with a head clip, and electrode implantation was performed with the assistance of the neurosurgical robot Sinovation ® . After opening the dura, the burr hole was continuously flushed with physiological saline to stop air from entering the skull. All procedures were performed under general anesthesia without intraoperative microelectrode recording (MER)., Results: The mean age of the patients at surgery and onset of seizures was 22 years (range 11-41 years) and 11 years (range 1-21 years), respectively. The median duration of seizures before CMT-DBS surgery was 10 years (2-26 years). CMT was successfully segmented, and its position was verified by experience target coordinates and QSM images in all ten patients. The mean surgical time for bilateral CMT-DBS in this cohort was 165 ± 18 min. The mean pneumocephalus volume was 2 cm 3 . The median absolute errors in the x-, y-, and z-axes were 0.7 mm, 0.5 mm, and 0.9 mm, respectively. The median Euclidean distance (ED) and radial error (RE) was 1.3 ± 0.5 mm and 1.0 ± 0.3 mm, respectively. No significant difference was found between right- and left-sided electrodes regarding the RE nor the ED. After a mean 12-month follow-up, the average reduction in seizures was 61%, and six patients experienced a ≥ 50% reduction in seizures, including one patient who had no seizures after the operation. All patients tolerated the anesthesia operation, and no permanent or serious complications were reported., Conclusions: Frameless robot-assisted asleep surgery is a precise and safe approach for placing CMT electrodes in patients with DRE, shortening the surgery time. The segmentation of the thalamic nuclei enables the precise location of the CMT, and the flow of physiological saline to seal the burr holes is a good way to reduce the influx of air. Flowering is critical for sexual reproduction and fruit production. Several pear (Pyrus sp.) varieties produce few flower buds, but the underlying mechanisms are unknown. The circadian clock regulator EARLY FLOWERING3 (ELF3) serves as a scaffold protein in the evening complex that controls flowering. Here, we report that the absence of a 58-bp sequence in the 2nd intron of PbELF3 is genetically associated with the production of fewer flower buds in pear. From rapid amplification of cDNA ends sequencing results, we identified a short, previously unknown transcript from the PbELF3 locus, which we termed PbELF3β, whose transcript level was significantly lower in pear cultivars that lacked the 58-bp region. The heterologous expression of PbELF3β in Arabidopsis (Arabidopsis thaliana) accelerated flowering, whereas the heterologous expression of the full-length transcript PbELF3α caused late flowering. Notably, ELF3β was functionally conserved in other plants. Deletion of the 2nd intron reduced AtELF3β expression and caused delayed flowering time in Arabidopsis. AtELF3β physically interacted with AtELF3α, disrupting the formation of the evening complex and consequently releasing its repression of flower induction genes such as GIGANTEA (GI). AtELF3β had no effect in the absence of AtELF3α, supporting the idea that AtELF3β promotes flower induction by blocking AtELF3α function. Background and Aims: Metabolic dysfunction-associated fatty liver disease (MAFLD) was proposed to substitute NAFLD in 2020. This new term highlights the systematic metabolic disturbances that accompany fatty liver. We evaluated the correlations between MAFLD and subclinical carotid atherosclerosis (SCA) based on a nationwide health examination population in China., Methods: We performed a nationwide cross-sectional population and a Beijing retrospective cohort from 2009 to 2017. SCA was defined as elevated carotid intima-media thickness. The multivariable logistic and Cox models were used to analyze the association between MAFLD and SCA., Results: 153,482 participants were included in the cross-sectional study. MAFLD was significantly associated with SCA in fully adjusted models, with an odds ratio of 1.66; 95% confidence interval (CI): 1.62-1.70. This association was consistent in the cohort, with a hazard ratio (HR) of 1.31. The association between baseline MAFLD and incident SCA increased with hepatic steatosis severity. Subgroup analysis showed an interaction between age and MAFLD, with a higher risk in younger groups (HR:1.67, 95% CI: 1.17-2.40)., Conclusion: In this large cross-section and cohort study, MAFLD was significantly associated with the presence and development of SCA. How crop domestication mediates root functional traits and trait plasticity in response to neighboring plants is unclear, but it is important for selecting potential species to be grown together to facilitate P uptake. We grew two barley accessions representing a two-stage domestication process as a sole crop or mixed with faba bean under low and high P inputs. We analyzed six root functional traits associated with P acquisition and plant P uptake in five cropping treatments in two pot experiments. The spatial and temporal patterns of root acid phosphatase activity were characterized in situ with zymography at 7, 14, 21, and 28 days after sowing in a rhizobox. Under low P supply, wild barley had higher total root length (TRL), specific root length (SRL), and root branching intensity (RootBr) as well as higher activity of acid phosphatase (APase) in the rhizosphere, but lower root exudation of carboxylates and mycorrhizal colonization (MC), relative to domesticated barley. In response to neighboring faba bean, wild barley exhibited larger plasticity in all root morphological traits (TRL, SRL, and RootBr), while domesticated barley showed greater plasticity in root exudates of carboxylates and colonization by mycorrhiza. Wild barley with greater root morphology-related trait plasticity was a better match with faba bean than domesticated barley, indicated by higher P uptake benefits in wild barley/faba bean than domesticated barley/faba bean mixtures under low P supply. Our findings indicated that the domestication of barley disrupts the intercropping benefits with faba bean through the shifts of root morphological traits and their plasticity in barley. The impact of atmospheric deposition of cadmium (Cd) in cereal crops has become a global concern. Enhanced lignin content was expected to benefit the plant performance against Cd exposure. To date, however, the underlying mechanisms of lignin regulating foliar Cd absorption in rice (Oryza sativa L.) and its effect on grain yield remains unclear. In present study, the effect and mechanism of rice in response to leaf Cd exposure were investigated using 113 Cd stable isotope and a lignin-increased rice mutant. The highest Cd uptake efficiency and uptake amount was observed in wild type (WT) plant grown in the maturity period, which were 3-fold higher than in mutant plant. Compared to WT, the mutant exhibited 14.75% and 25.43% higher contents in G- and S-unit of lignin monomers. Lignin biosynthesis and polymerization related genes (OsPAL/OsCOMT/Os4CL3/OsLAC5/OsLAC15) were significantly up-regulated in mutants. In addition, the enzyme activities involved in the above process were also significantly increased by 1.24-1.49-fold. The increased Cd retention in cell wall and decreased gene expression levels of OsNRAMP5, OsHMA3 and OsIRT1 in mutant indicated that lignin effectively inhibited Cd transportion in plant tissues. Moreover, the antioxidant capacity and photosynthesis efficiency in mutant plant were obviously improved, leading to higher Cd tolerance and increased grain yield. Fertilization plays an important role in changing soil microbial diversity, which is essential for determining crop yields. Yet, the influence of organic amendments on microbial diversity remains uncertain, and few studies have addressed the relative importance of microbial diversity versus other drivers of crop yields. Here, we synthesize 219 studies worldwide and found that organic amendments significantly increased microbial diversity components (i.e., Shannon, richness, and phylogenetic diversity) and shifted microbial community structure compared to mineral-only fertilization. The performance of microbial alpha diversity varied substantially with organic amendment types, microbial groups and changes in soil pH. Both microbial diversity and community structure exhibited significantly positive relationships with microbial functionality and crop yields. In addition, soil abiotic properties and microbial functionality had a much stronger impact on crop yields than microbial diversity and climate factors. Partial least squares path modeling showed that soil microbial diversity was an important underlying factor driving crop yields via boosting soil microbial functionality. Agricultural intensification driven by land-use changes has caused continuous and cumulative soil acidification (SA) throughout the global agroecosystem. Microorganisms mediate acid-generating reactions; however, the microbial mechanisms responsible for exacerbating SA feedback remain largely unknown. To determine the microbial community composition and putative function associated with SA, we conducted a metagenomic analysis of soils across a chronosequence that has elapsed since the conversion of rice-wheat (RW) to rice-vegetable (RV) rotations. Compared to RW rotations, soil pH decreased by 0.50 and 1.56 units (p < 0.05) in response to 10-year and 20-year RV rotations, respectively. Additionally, acid saturation ratios were increased by 7.3% and 36.2% (p < 0.05), respectively. The loss of microbial beta-diversity was a key element that contributed to the exacerbation of SA in the RV. Notably, the 20-year RV-enriched microbial taxa were more hydrogen (H + )-, aluminium (Al 3+ )-, and nitrate nitrogen (NO 3 - -N) -dependent and contained more genera exhibiting dehydrogenation functions than did RW-enriched taxa. "M00115, M00151, M00417, and M00004" and "M00531 and M00135" that are the "proton-pumping" and "proton-consuming" gene modules, respectively, were linked to the massive recruitment of acid-dependent biomarkers in 20-year RV soils, particularly Rhodanobacter, Gemmatirosa, Sphingomonas, and Streptomyces. To explore the effects of biochar application on CO 2 and CH 4 emissions as well as the temperature response of CO 2 emissions, a 1-year experiment was conducted with three treatments (control; CF, chemical fertilizer only; BCF, biochar combined with chemical fertilizer) in a vegetable field. The results showed that (1) compared with CF, short-term application of biochar significantly enhanced the cumulative CO 2 emissions by 27.5% from a soil-plant system by increasing the soil microbial biomass (e.g., MBC) and C substrates (e.g., SOC); (2) lowest emissions of CH 4 were observed in the BCF treatment, and an increase in CH 4 consumption and reduced competition with NH 4 + may be responsible for the significant reduction in CH 4 source strength in biochar-amended soil; and (3) activation energy (E a ) was identified as an important factor influencing the temperature sensitivity (Q 10 ) of CO 2 emissions. Fertilization (CF and BCF) reduced the average Q 10 and E a values of CO 2 emissions by 9.0-26.7% and 23.5-10.1%, respectively, relative to the control. In addition, the average E a value in the BCF treatment (51.9 kJ mol -1 ) was significantly higher than those in the control and CF treatments. The increase in Q 10 and E a values following biochar application possibly contributed to the supplementation of limited labile C and nutrients but highly resistant C following biochar application. Soil pH and crop cultivation may play key roles in influencing the change in E a . Increasing soil petroleum hydrocarbons (PHs) pollution have caused world-wide concerns. The removal of PHs from soils mainly involves physical, chemical, biological processes and their combinations. To date, most reviews in this field based on research articles, but limited papers focused on the integration of remediation technologies from the perspective of patents. In this study, 20-years Chinese patents related to the remediation of soil PHs were comprehensively analyzed. It showed an increasing number of patent applications and the patents' quantity were positively correlated with Chinese GDP over the years, suggesting the more the economy developed the more environmental problems and corresponding solutions emerged. In addition, chemical technologies were mostly used in a combination to achieve faster and better effects, while the physical technologies were often used alone due to high costs. In all PHs remediation techniques, bacteria-based bioremediation was the most used from 2000 to 2019. Bacillus spp. and Pseudomonas spp. were the most used bacteria for PHs treatment because these taxa were widely harboring functions such as biosurfactant production and hydrocarbon degradation. Objective: The purpose of this study was to explore the function and mechanism of LncRNA CERS6-AS1 on papillary thyroid cancer., Materials: CERS6-AS1, miR-497-5p, and LASP1 expression in papillary thyroid cancer tissues and cells were detected by RT-PCR. The relationship between CERS6-AS1 expression and clinical characteristics was analyzed, and overall survival was evaluated via Kaplan-Meier analysis. Cell activity was tested by cell counting kit-8, cell reactive oxygen species was detected by DCFH-DA method, and cell iron ion was detected by iron analysis kit. The relationship among CERS6-AS1, miR-497-5p, and LASP1 was confirmed by luciferase reporter gene detection, RNA pull-down detection, and RIP detection. The expression of related proteins was assessed by western blot or immunohistochemistry., Results: High level of CERS6-AS1 and LASP1 was detected in papillary thyroid cancer tissues and cells and predicted poor prognosis. In contrast, miR-497-5p was decreased in papillary thyroid cancer tissues and cells, which was positively correlated with prognosis. Silencing CERS6-AS1 suppressed cell viability and increased ferroptosis in papillary thyroid cancer. LASP1 was modulated by CERS6-AS1 through sponging miR-497-5p. Up-regulation of LASP1 or silencing miR-497-5p could weaken the effect of CERS6-AS1 on papillary thyroid cancer cells. Silencing CERS6-AS1 restrained the growth of xenografted tumors., Conclusion: Our findings demonstrated that down-regulation of CERS6-AS1 reduced cell viability and amplified cell ferroptosis by modulating the miR-497-5p/LASP1 axis in papillary thyroid cancer., (© 2022 by the Association of Clinical Scientists, Inc.)

Although Cd concentration of grains is generally lower in japonica than in indica subspecies, the effects of root endodermal barriers on the subspecific differences in Cd accumulation in rice (Oryza sativa L.) are poorly understood. Here, we characterized the differences in endodermal differentiation between japonica and indica subspecies and their effects on Cd radial transport. Casparian strips (CSs) and suberin lamellae (SL) in japonica subspecies were initiated at the 6%- 7% and 21%- 27% position from the root tip, respectively, which were 65% and 26% earlier than in indica subspecies, respectively. The lignin/suberin content in japonica subspecies was 47%/42% greater than that in indica subspecies because of the higher expression of lignin/suberin biosynthesis-related genes (OsCASP1, OsPAL, OsCYP86A1 and OsKCS20). Cd exposure induced endodermal plasticity in both subspecies, but the changes in japonica were greater than in indica subspecies. The earlier formation of CSs/SL in japonica subspecies significantly restricted the flow of radial transport tracer to reach the xylem and decreased Cd influx into roots, that is, endodermal barriers inhibited Cd radial transport via both apoplastic and cell-to-cell pathways, thus decreasing the root-to-shoot transport of Cd in japonica subspecies. Soil Cd pollution is a serious environmental issue associated with human activities. However, the factors determining exogenous Cd dynamics in the soil profile in a complex environment are not well understood. Based on regional observations from 169 soil profiles across the Chengdu Plain, this study explored the key factors controlling Cd accumulation in the soil profile under actual field conditions. Results showed that total soil Cd contents decreased from 0.377 to 0.196 mg kg -1 with increasing soil depth. The effects of phosphate fertilizer rates, road density and precipitation on the difference in total soil Cd content were only observed in topsoil, while agricultural land-use type and topography had no impact. In contrast, significant differences in the total soil Cd content among different parent material types were found in the 0-20, 40-60 and 60-100 cm soil depths. One sample t-tests showed that significant Cd accumulation occurred in the whole soil profile in soils formed from Q4 (Quaternary Holocene) grey alluvium, while soils formed from Q3 (Quaternary Pleistocene) old alluvium and Q4 grey-brown alluvium showed significant Cd accumulation only in the 0-40 cm soil layers. In the topsoil, acid soluble Cd accounted for the largest proportion of the total Cd in soils formed from Q4 grey alluvium, reducible Cd was the main fraction in soils formed from Q4 grey-brown alluvium, while reducible Cd and residual Cd contributed the largest proportion of the total soil Cd in soils formed from Q3 old alluvium. The above results indicated that parent material was the decisive factor determining the magnitudes and depths of exogenous Cd accumulation in the soil profile due to its impacts on the Cd fraction distributions. Members of the REM (Reproductive Meristem) gene family are expressed primarily in reproductive meristems and floral organs. However, their evolution and their functional profiles in flower development remain poorly understood. Here, we performed genome-wide identification and evolutionary analysis of the REM gene family in Rosaceae. This family has been greatly expanded in rose (Rosa chinensis) compared to other species, primarily through tandem duplication. Expression analysis revealed that most RcREM genes are specifically expressed in reproductive organs and that their specific expression patterns are dramatically altered in rose plants with mutations affecting floral organs. Protein-protein interaction analysis indicated that RcREM14 interact with RcAP1 (one of the homology of A class genes in ABCDE model), highlighting the roles of RcREM genes in floral organ identity. Finally, co-expression network analysis indicated that RcREM genes are co-expressed with a high proportion of key genes that regulate flowering time, floral organ development, and cell proliferation and expansion in R. chinensis., (Copyright © 2021 Elsevier Inc. The recycling of agricultural waste is a global challenge to the sustainable development of agriculture. By using corn stalks, we studied the feasibility of combining anaerobic fermentation and pyrolysis processes to produce both fermentated liquid feed and biologically modified biocharas cadmium adsorbents. Anaerobic ensiling enhanced the biodegradation of corn stalks by increasing crude protein and reducing fiber contents. After 24-h anaerobic fermentation, corn stalks silage was decomposed into the liquid filtrate and non-fermented residue. Fermented liquid feed (FLF) was prepared by storing feed and liquid filtrate (1:4.0, wt/wt) in a closed tank at 20 °C for 4 days, which showed desired properties (pH < 4.5, lactic acid bacteria greater than 9.0 lg cfu g -1 , lactic acid greater than 100 mmol L -1 ). The non-fermented residue was pyrolyzed at 500 °C to prepare biologically modified biochar (BCB24). In comparison with pristine biochar produced from corn stalks (CB), anaerobic ensiling and anaerobic fermentation significantly increased the surface area, oxygen-containing functional groups, as well as mineral components in BCB24. The maximum sorption capacity of Cd(II) for BCB24 was 2.1 times of CB, suggesting that BCB24 is an effective adsorbent for Cd(II) removal from water. Controlled release fertilizer (CRF) has been shown to increase crop yield and N use efficiency (NUE) compared with traditional chemical fertilizer (TF). However, few studies examined the effects of CRF on CH 4 , N 2 O, and NO emissions simultaneously in alkaline paddy fields under rice-wheat rotation. In the present study, we conducted a 2-year field experiment to compare the effects of different CRF application strategies on these gas emissions with those of TF and explored the effects of CRF on global warming potential (GWP), crop yields, and greenhouse gas emission intensity (GHGI). Results showed that CRF can reduce 0.98-14.3%, 13.3-21.1%, and 8.22-16.3% of CH 4 , N 2 O, and NO emissions, respectively, in the studied alkaline paddy field. CRF reduce CH 4 emission probably by regulating soil NH 4 + concentration. CRF reduce N 2 O and NO emissions probably by regulating inorganic N content in the studied alkaline paddy soil. CRF had the same effect on annual crop yield as TF, especially when CRF was applied twice in each season and had the same N application rate as TF. Annual crop yields and the agronomic efficiency of N (AE N ) increased by 8.24% and 21.6%, respectively. On the average of the two rice-wheat rotation cycles, GHGI significantly decreased by up to 14.1% after the application of CRF as relative to that after the application of TF (P < 0.05). These results suggest that CRF is an environment-friendly N fertilization strategy for mitigating GWP and ensuring high crop yield in an alkaline paddy field under rice-wheat rotation.

Rose (Rosa spp.) plants flower via two contrasting methods: once flowering (OF) and continuous flowering (CF). Purple branch is a rare continuously flowering variety of Rosa rugosa that is extensively cultivated in China. However, the genetic basis of its CF behavior is unknown. We demonstrated that Purple branch is heterozygous for the TFL1 homolog KSN. One KSN allele with a 9 kb Copia insertion was found to be identical to that from continuously flowering Rosa chinensis Old blush. The other allele was found to be a functional wild-type allele. The overall expression of KSN was closely linked to the floral transition, and it was significantly repressed in continuously flowering Purple branch compared with OF Plena. The promoter region of the normal KSN allele was hypermethylated, and histone methylation at H3H4, H3K9, and H3K27 of the KSN gene locus was modified in continuously flowering Purple branch. Silencing of the DNA methyltransferase genes MET1 and CMT3 and the histone methyltransferase gene SUVR5 in Purple branch led to enhanced KSN expression, but silencing of the histone demethylase gene JMJ12 suppressed KSN expression. Therefore, the CF habit of Purple branch may be due to reduced expression of KSN caused by the halved dose and may be associated with epigenetic modifications together with retrotransposon insertions along the chromosome. Our study revealed a novel mechanism underlying the CF behavior of rose plants.

Agricultural production of Ligusticum chuanxiong Hort. is often affected by heavy metal pollution in soil, especially mixtures of cadmium (Cd) and lead (Pb). We assessed metal-induced phytotoxicity in L. chuanxiong by exposing the plants to soil treated with Cd, Pb, or Cd/Pb mixtures. A combined Cd/Pb treatment alleviated the inhibition in plant growth, photosynthesis, and secondary metabolite generation seen in single-metal exposures in three of the four combinations. Most combined Cd/Pb treatments resulted in preferential uptake of magnesium, copper, and nitrogen in underground plant parts and accumulation of phosphorus and calcium in aboveground plant parts, thereby leading to improvements in photosynthetic potential. Compared with single-metal exposures, combined Cd/Pb treatment significantly decreased the contents of Cd by 16.67%-40.12% and Pb by 10.68%-21.70% in the plant, respectively. At the subcellular level, the Pb presence increased the Cd percentage associated with cell wall from 64.79% to 67.93% in rhizomes and from 32.76% to 45.32% in leaves, while Cd reduced Pb contents by 9.36%-46.39% in the subcellular fractions. A combined Cd/Pb treatment decreased the contents of water- and ethanol-extractable metal forms and increased the contents of acetic acid- and hydrochloric acid-extractable forms. The lower toxic effects of the Cd/Pb mixture in L. chuanxiong were associated with photosynthetic potential, subcellular distribution, the chemical forms of Cd and Pb, and synthesis of secondary metabolites. Key Message: We genome-wide identified 28 JmjC domain-containing genes, further spatio-temporal expression profiling and genetic analysis defined them as epigenetic regulators in flowering initiation of Rosa chinensis. The JmjC domain-containing histone demethylases play critical roles in maintaining homeostasis of histone methylations, thus are vital for plant growth and development. Genome-wide identification of the JmjC domain-containing genes have been reported in several species, however, no systematic study has been performed in rose plants. In this paper, we identified 28 JmjC domain-containing genes from the newly published genome database of Rosa chinensis. The JmjC domain-containing proteins in R. chinensis were divided into seven groups, KDM3 was the largest group with 13 members, and JmjC domain-only A and KDM5B were the smallest clades both with only one member. Although all the JmjC domain proteins having a conserved JmjC domain, the gene and protein structure experienced differentiation and specification during the evolution, especially in KDM3 clade, one gene (RcJMJ40) was found carrying site deletions for cofactors Fe (II) and α-KG binding which were crucial for demethylase activities, three genes (RcJMJ41, RcJMJ43 and RcJMJ44) had no intron while two of them had tandem JmjC domains. Spatial expression pattern analysis of these JmjC domain-containing genes in different tissues showed most of them were highly expressed in reproductive tissues such as floral meristem and closed flowers than vegetative tissues, demonstrating their important functions in developmental switch from vegetative to reproductive growth of roses. Temporal expression profiling indicated majority of JmjC domain-containing genes from R. chinensis fluctuated along with floral bud differentiation and development, further proving their essential roles in flower organogenesis. VIGS induced silencing of RcJMJ12 led to delayed flowering time, and decreased the expression levels of flowering integrator such as RcFT, RcSOC1, RcFUL, RcLFY and RcAP1, therefore providing the genetic evidence of RcJMJ12 in flowering initiation. Collectively, spatio-temporal expression profiling and genetic analysis defined the JmjC domain-containing genes as important epigenetic regulators in flower development of R. chinensis.

Bletilla striata (Thunb.) Reichb. f. (Orchidaceae), also known as Bai-ji, is a traditional Chinese herb that is widely used in Asia to treat hematemesis, hemoptysis, traumatic bleeding and other similar disorders. Most studies have focused on the pharmacological activities of polysaccharide extracts from B. striata. Our previous studies found that the nonpolysaccharide fraction from B. striata extract also has a hemostatic effect; however, the active constituents responsible for this pharmacological action are unclear. Thus, the metabolic profiles of the nonpolysaccharide fraction were investigated in Sprague-Dawley rats and intestinal bacteria models using ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry. Mass data were acquired by the MS E method. Eight components including five prototypes and three metabolites were identified in rat biofluids after oral administration of the nonpolysaccharide fraction. The parent compounds underwent various metabolic processes, including hydrolysis, deglucosylation, glycosylation and sulfate conjugation. The results not only reveal the possible metabolic pathway, but also indicate the potential pharmacological components. Seed germination is a fundamental process in the plant life cycle and is regulated by functionally opposing internal and external inputs. Here we explored the role of a negative regulator of photomorphogenesis, a B-box-containing protein (BBX19), as a molecular link between the inhibitory action of the phytohormone abscisic acid (ABA) and the promoting role of light in germination. We show that seeds of BBX19-overexpressing lines, in contrast to those of BBX19 RNA interference lines, display ABA hypersensitivity, albeit independently of elongated hypocotyl 5 (HY5). Moreover, we establish that BBX19 functions neither via perturbation of GA signaling, the ABA antagonistic phytohormone, nor through interference with the DELLA protein germination repressors. Rather, BBX19 functions as an inducer of ABA INSENSITIVE5 (ABI5) by binding to the light-responsive GT1 motifs in the gene promoter. Biologically modified biochars derived from digestion residue of corn straw silage at different pyrolysis temperature (300-700 °C) were prepared for removing Cd from water. Compared with the pristine biochar derived from corn straw (CB), transabdominal transformation of corn straw silage (TCB) significantly increased surface area (4.24-56.58 m 2  g -1 ), oxygen-containing functional group (COC, MgO, SiO) and mineral components (CaCO 3 , KCl). The sorption isotherms could be well described by Langmuir model, the kinetic data was best fitted by the Pseudo second order model. The maximum sorption capacity (Qm) obtained from Langmuir model for TCB700 (175.44 mg g -1 ) was 3 times of CB700 (56.82 mg g -1 ). Precipitation with minerals, ion exchange and complexation with oxygen-containing functional groups were the main mechanisms of Cd(II) sorption on TCB. These results imply that biologically modified biochar derived from digestion residue of corn straw silage at ≥600 °C is an effective sorbent for Cd removal from water., (Copyright © 2019 Elsevier B.V. In situ remediation and assessment of sediments contaminated with both antibiotics and heavy metals remains a technological challenge. In this study, MgCl 2 -modified biochar (BCM) was obtained at 500 °C through slow pyrolysis of Thalia dealbata and used for remediation of sediments contaminated by sulfamethoxazole (SMX) and Cd. The BCM showed greater surface area (110.6 m 2  g -1 ) than pristine biochar (BC, 7.1 m 2  g -1 ). The SMX sorption data were well described by Freundlich model while Langmuir model was better for the Cd 2+ sorption data. The addition of 5.0% BCM significantly increased the sorption of SMX (by 50.8-58.6%) and Cd (by 24.2-25.6%) on sediments in both single and binary systems as compared with 5.0% BC. SMX sorption in sediments was significantly improved by addition of Cd 2+ , whereas SMX has no influence on Cd sorption on sediments. The addition of BCM distinctly decreased both SMX (by 51.4-87.2%) and Cd concentrations (by 56.2-91.3%) in overlying water, as well as in TCLP extracts (by 55.6-86.1% and 58.2-91.9% for SMX and Cd, respectively), as compared with sediments without biochar. Both germination rate and root length of pakchoi increased with increasing doses of BCM in contaminated sediments, 5.0% BCM showed greater promotion on pakchoi growth than 5.0% BC. Overall, BCM in the sediments does not only decrease the bioavailability of SMX and Cd, but it also diminishes the phytotoxicity, and thereby shows great application potential for in situ remediation of sediments polluted with antibiotics and heavy metals., (Copyright © 2018 Elsevier B.V. Vestibulo-ocular reflex (VOR) responding to acceleration stimuli is originated from the vestibular apparatuses and thus widely used as an in vivo indicator of the vestibular function. We have developed a vestibular function testing (VFT) system that allows to evaluate VOR response with improved efficiency. The previously required surgical procedure has been avoided by using a newly designed animal-immobility setup. The efficacy of our VFT system was demonstrated on the mice with vestibular abnormalities caused by either genetic mutations (Lhfpl5 -/- or Cdh23 -/- ) or applied vestibulotoxicant (3,3'-iminodipropionitrile, IDPN). Daily longitudinal inspection of the VOR response in the IDPN-administered mice gives the first VOR-based daily-progression profile of the vestibular impairment. The capability of VOR in quantifying the severity of toxicant-induced vestibular deficits has been also demonstrated. The acquired VOR-measurement results were validated against the corresponding behavioral-test results. Further validation against immunofluorescence microscopy was applied to the VOR data obtained from the IDPN-administered mice. We conclude that the improved efficiency of our surgery-free VFT system, firstly, enables the characterization of VOR temporal dynamics and quantification of vestibular-impairment severity that may reveal useful information in toxicological and/or pharmaceutical studies; and, secondly, confers our system promising potential to serve as a high-throughput screener for identifying genes and drugs that affect vestibular function., (Copyright © 2018. Semi-agricultural ecosystems in peri-urban areas are susceptible to contamination. The spatial distribution and influencing factors of such pollution are unclear and poorly constrained in many areas worldwide. Therefore, studying the problems of soil pollution in peri-urban areas is critical for environmental management and agricultural production. In this paper, with cadmium (Cd) as the target pollutant, the spatiotemporal variations of soil cadmium pollution and the relative importance of the affecting factors were analyzed at a peri-urban area from the Jingyang District, Sichuan, China. Statistical results showed that the farmland in the study area could be considered moderately soil Cd-polluted, under the dual influence of natural factors and human activity. In particular, the soil Cd concentration in Tianyuan and Bajiaojing exceeded 0.5 mg kg -1 , for intensive industrial enterprises are distributed in these areas. Correspondingly, the geoaccumulation index also showed that the contamination of Cd in this area was moderately polluted. Moreover, the ecological risk index was 80% in the study area, indicating that the soil Cd pollution potential risk was moderate to high. High geological background values (soil Cd = 0.29 mg kg -1 ), river migration, industrial enterprises, and traffic significantly influenced soil Cd pollution, with natural geological factors playing greater roles. The significant horizontal-spatial effective distances away from Shiting River, Deyang-Aba Highway, and chemical plants were 200, 400, and 100 m, respectively. These results will be useful in guiding farmland cultivation and pollution remediation effectively in the peri-urban areas.

Determination of soil nitrogen distributions and the factors affecting them is critical for nitrogen fertilizer management and prevention of nitrogen pollution. In this paper, the spatiotemporal variations of soil nitrogen and the relative importance of their affecting factors were analysed at a county scale in the purple hilly area of the mid-Sichuan Basin in Southwest China based on soil data collected in 1981 and 2012. Statistical results showed that soil total nitrogen (TN) increased from 0.88 g kg(-1) in 1981 to 1.12 g kg(-1) in 2012, whereas available nitrogen (AN) decreased from 84.22 mg kg(-1) to 74.35 mg kg(-1). In particular, AN showed a significant decrease in agricultural ecosystems but remained stable in woodland and grassland. Correspondingly, most of the study area exhibited increased TN content and decreased AN content in space. The nugget/sill ratios of TN and AN increased from 0.419 to 0.608 and from 0.733 to 0.790, whereas spatial correlation distances decreased from 12.00 km to 9.50 km and from 9.50 km to 9.00 km, respectively, suggesting that the spatial dependence of soil nitrogen became weaker and that the extrinsic factors played increasingly important roles in affecting the soil nitrogen distribution. Soil group and land use type were the two dominant factors in 1981, followed by topographic factors, vegetation coverage and parent material, whereas land use type became the most important factor in 2012, and the relative contribution of topographic factors declined markedly. The results suggested that land use related to cultivation management and fertilizer application was the decisive factor for soil nitrogen change. The increase in TN content and the decrease in AN content over the study period also suggested improper use of nitrogen fertilizer, which can result in nitrogen loss through increasing nitrification rates.
