203. Simultaneous Determination of Ractopamine, Chloramphenicol, and Zeranols in Animal-Originated Foods by LC-MS/MS Analysis with Immunoaffinity Clean-up Column.

Abstract

A novel analytical method employing immunoaffinity column (IAC) clean-up coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for simultaneous determination of ractopamine, chloramphenicol, and zeranols (α-zearalanol, β-zearalanol, zearalanone, α-zearalenol, β-zearalenol, and zearalenone) in animal-originated foods. The sample was first digested by β-glucuronidase/sulfatase and then extracted with ethyl acetate-diethyl ether (9:1, v/v). The extracted solution was evaporated to dryness and then the residue was dissolved by 2 mL of 50% acetonitrile solution. After filtration, 1 mL filtrate was diluted to 10 mL with PBS. The reconstituted solution was cleaned up with immunoaffinity column and then analyzed by high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS). The established method was shown to be sensitive efficient and reliable as indicated by the linearity ( r ≥ 0.9994), precision (RSD ≤ 1.7%), average recovery (72.3-103.2%), and the limit of detection (0.05-0.10 μg/kg). Abstract

Au nanoparticles (NPs) can endow titania nanotubes (Au@TiO 2 -NT) with light-independent antibacterial properties which bode well for in vivo application because of the dark environment inside tissues. In this work, the long-term antibacterial bactericidal properties and cytocompatibility of Au@TiO 2 -NT without photocatalytic effects are studied in details. The materials exhibit antibacterial effects against Staphylococcus aureus according to antibacterial tests carried out for a total time of 21 days, which are normally long enough for early stage tissue healing after surgery. In addition, adhesion and proliferation of MC3T3-E1 osteoblasts on Au@TiO 2 -NT reveal cytocompatibility comparable to that of TiO 2 -NT. No reactive oxygen species (ROS) are detected from either the bacteria or MC3T3-E1 cells cultured on the Au@TiO 2 -NT surface. The absence of ROS, long-term antibacterial properties, and cytocompatibility make Au@TiO 2 -NT promising biomaterials in orthopedic devices and implants. [ABSTRACT FROM AUTHOR]

205. Apparent limitations in the ability of CMIP5 climate models to simulate recent multi-decadal change in surface temperature: implications for global temperature projections.

Abstract

Observed surface temperature trends over the period 1998-2012/2014 have attracted a great deal of interest because of an apparent slowdown in the rate of global warming, and contrasts between climate model simulations and observations of such trends. Many studies have addressed the statistical significance of these relatively short-trends, whether they indicate a possible bias in the model values and the implications for global warming generally. Here we re-examine these issues, but as they relate to changes over much longer-term changes. We find that on multi- decadal time scales there is little evidence for any change in the observed global warming rate, but some evidence for a recent temporary slowdown in the warming rate in the Pacific. This multi-decadal slowdown can be partly explained by a cool phase of the Interdecadal Pacific Oscillation and a short-term excess of La Niña events. We also analyse historical and projected changes in 38 CMIP climate models. All of the model simulations examined simulate multi-decadal warming in the Pacific over the past half-century that exceeds observed values. This difference cannot be fully explained by observed internal multi-decadal climate variability, even if allowance is made for an apparent tendency for models to underestimate internal multi-decadal variability in the Pacific. Models which simulate the greatest global warming over the past half-century also project warming that is among the highest of all models by the end of the twenty-first century, under both low and high greenhouse gas emission scenarios. Abstract

Naturally occurring multiyear to decadal variability is evident in rainfall, temperature, severe weather, and flood frequency around the globe. It is therefore important to understand the cause of this variability and the extent to which it can be predicted. Here internally generated decadal climate variability and its predictability potential in an ensemble of CMIP5 models are assessed. Global hot spots of subsurface ocean decadal variability are identified, revealing variability in the southern Tasman Sea that is coherent with variability in much of the Pacific Ocean and Southern Hemisphere. It is found that subsurface temperature variability in the southern Tasman Sea primarily arises in response to preceding changes in Southern Hemisphere winds. This variability is multiyear to decadal in character and is coherent with surface temperature in parts of the Southern Hemisphere up to several years later. This provides some degree of potential predictability to surface temperature in the southern Tasman Sea and surrounding regions. A few models exhibit significant correlation between subsurface variability in the southern Tasman Sea and zonally averaged precipitation south of 50°S; however, the multimodel mean does not exhibit any significant correlation between subsurface variability and precipitation. Models that exhibit stronger subsurface variability in the southern Tasman Sea also have a stronger interdecadal Pacific oscillation signal in the Pacific. [ABSTRACT FROM AUTHOR]

Abstract

Although titanium embedded with silver nanoparticles (Ag-NPs@Ti) are suitable for biomedical implants because of the good cytocompatibility and antibacterial characteristics, the exact antibacterial mechanism is not well understood. In the present work, the antibacterial mechanisms of Ag-NPs@Ti prepared by plasma immersion ion implantation (PIII) are explored in details. The antibacterial effects of the Ag-NPs depend on the conductivity of the substrate revealing the importance of electron transfer in the antibacterial process. In addition, electron transfer between the Ag-NPs and titanium substrate produces bursts of reactive oxygen species (ROS) in both the bacteria cells and culture medium. ROS leads to bacteria death by inducing intracellular oxidation, membrane potential variation, and cellular contents release and the antibacterial ability of Ag-NPs@Ti is inhibited appreciably after adding ROS scavengers. Even though ROS signals are detected from osteoblasts cultured on Ag-NPs@Ti, the cell compatibility is not impaired. This electron-transfer-based antibacterial process which produces ROS provides insights into the design of biomaterials with both antibacterial properties and cytocompatibility. Abstract

The apoptosis in primary oral squamous cell carcinomas (OSCCs) without lymph node (LN) metastases and its relation with clinical stages and pathological grades was investigated. The terminal deoxynucleotidyl trasferase (TdT)-mediated dUTP nick end labeling (TUNEL) was used to detect the apoptotic cells in 15 cases of OSCCs. The percentage of apoptotic cells among tumor cells were calculated as apoptotic index (AI). The results showed that in all 15 cases of OSCCs, apoptotic cells could be visualized by TUNEL with AI ranging from 0.03 to 0.92 (average 0.32). Abstract

El Niño events are characterized by surface warming of the tropical Pacific Ocean and weakening of equatorial trade winds that occur every few years. Such conditions are accompanied by changes in atmospheric and oceanic circulation, affecting global climate, marine and terrestrial ecosystems, fisheries and human activities. The alternation of warm El Niño and cold La Niña conditions, referred to as the El Niño–Southern Oscillation (ENSO), represents the strongest year-to-year fluctuation of the global climate system. Here we provide a synopsis of our current understanding of the spatio-temporal complexity of this important climate mode and its influence on the Earth system. Our current understanding of the spatio-temporal complexity of the El Niño–Southern Oscillation phenomenon is reviewed and a unifying framework that identifies the key factors for this complexity is proposed.

Abstract

Genome-wide association studies (GWAS) and fine-mapping efforts to date have identified more than 100 prostate cancer (PrCa)-susceptibility loci. We meta-analyzed genotype data from a custom high-density array of 46,939 PrCa cases and 27,910 controls of European ancestry with previously genotyped data of 32,255 PrCa cases and 33,202 controls of European ancestry. Our analysis identified 62 novel loci associated (P< 5.0 × 10−8) with PrCa and one locus significantly associated with early-onset PrCa (≤55 years). Our findings include missense variants rs1800057 (odds ratio (OR) = 1.16; P= 8.2 × 10−9; G>C, p.Pro1054Arg) in ATMand rs2066827 (OR = 1.06; P= 2.3 × 10−9; T>G, p.Val109Gly) in CDKN1B. The combination of all loci captured 28.4% of the PrCa familial relative risk, and a polygenic risk score conferred an elevated PrCa risk for men in the ninetieth to ninety-ninth percentiles (relative risk = 2.69; 95% confidence interval (CI): 2.55–2.82) and first percentile (relative risk = 5.71; 95% CI: 5.04–6.48) risk stratum compared with the population average. These findings improve risk prediction, enhance fine-mapping, and provide insight into the underlying biology of PrCa1. A large meta-analysis combining genome-wide and custom high-density genotyping array data identifies 63 new susceptibility loci for prostate cancer, enhancing fine-mapping efforts and providing insights into the underlying biology.

Abstract

Emergence of antibiotic‐resistance pathogens has caused serious health issues and if the current trend is to continue, treatment of the infection will become complicated and even unsuccessful due to new antimicrobial resistance (AMR). Therefore, there is a global drive to identify new methods to treat infection and develop better antibacterial materials and therapy. Although new and more potent antibiotics have aided the fight against microbes, they only offer a temporary solution because future bacteria strains may become resistant to these antibiotics and drugs. Recently, application of non‐biological methods such as, electrical currents and photothermal/dynamic therapies to kill bacteria, reveal new approaches to design antimicrobial biomaterials, as complications stemming from drug‐resistant bacteria can be obviated. Furthermore, recent research has focused on mimicking the surface patterns on plants and insects such as lotus leaves and dragonfly wings. Bio‐inspired micro/nano patterns have been replicated on a variety of biomaterials to improve the bacterial resistance and other properties with good success. This is an exciting research area with immense practical and clinical potentials. In this review, recent advances in the application of chemical/biological approaches to combat bacterial infection and AMR are summarized and the related mechanisms are discussed. Abstract

Background The lack of sufficient specificity and sensitivity among conventional cancer biomarkers, such as prostate specific antigen (PSA) for prostate cancer has been widely recognized after several decades of clinical implications. Autoantibodies (autoAb) among others are being extensively investigated as potential substitute markers, but remain elusive. One major obstacle is the lack of a sensitive and multiplex approach for quantifying autoAb against a large panel of clinically relevant tumor-associated antigens (TAA). Methods To circumvent preparation of phage lysates and purification of recombinant proteins, we identified B cell epitopes from a number of previously defined prostate cancer-associated antigens (PCAA). Peptide epitopes from cancer/testis antigen NY-ESO-1, XAGE-1b, SSX-2,4, as well as prostate cancer overexpressed antigen AMACR, p90 autoantigen, and LEDGF were then conjugated with seroMAP microspheres to allow multiplex measurement of autoAb present in serum samples. Moreover, simultaneous quantification of autoAb plus total PSA was achieved in one reaction, and termed the "A+PSA" assay. Results Peptide epitopes from the above 6 PCAA were identified and confirmed that autoAb against these peptide epitopes reacted specifically with the full-length protein. A pilot study was conducted with the A+PSA assay using pre-surgery sera from 131 biopsy-confirmed prostate cancer patients and 121 benign prostatic hyperplasia and/or prostatitis patients. A logistic regression-based A+PSA index was found to enhance sensitivities and specificities over PSA alone in distinguishing prostate cancer from nonmalignant cases. The A+PSA index also reduced false positive rate and improved the area under a receiver operating characteristic curve. Conclusions The A+PSA assay represents a novel platform that integrates autoAb signatures with a conventional cancer biomarker, which may aid in the diagnosis and prognosis of prostate cancer and others.
