As two central issues of global climate change, the continuous increase of both atmospheric CO2 concentrations and global temperature has profound effects on various terrestrial ecosystems. Microbial communities play pivotal roles in these ecosystems by responding to environmental changes through regulation of soil biogeochemical processes. However, little is known about the effect of elevated CO2 (eCO2) and global warming on soil microbial communities, especially in semiarid zones. We used a functional gene array (GeoChip 3.0) to measure the functional gene composition, structure, and metabolic potential of soil microbial communities under warming, eCO2, and eCO2 + warming conditions in a semiarid grassland. The results showed that the composition and structure of microbial communities was dramatically altered by multiple climate factors, including elevated CO2 and increased temperature. Key functional genes, those involved in carbon (C) degradation and fixation, methane metabolism, nitrogen (N) fixation, denitrification and N mineralization, were all stimulated under eCO2, while those genes involved in denitrification and ammonification were inhibited under warming alone. The interaction effects of eCO2 and warming on soil functional processes were similar to eCO2 alone, whereas some genes involved in recalcitrant C degradation showed no significant changes. In addition, canonical correspondence analysis and Mantel test results suggested that NO3-N and moisture significantly correlated with variations in microbial functional genes. Overall, this study revealed the possible feedback of soil microbial communities to multiple climate change factors by the suppression of N cycling under warming, and enhancement of C and N cycling processes under either eCO2 alone or in interaction with warming. These findings may enhance our understanding of semiarid grassland ecosystem responses to integrated factors of global climate change.

The integration of Si‐based anodes within sulfide‐based solid electrolyte (SSE) Li‐ion batteries (LIB) has emerged as a promising avenue of research and development, attracting increasing interest in recent years. This work comprehensively examines the latest research directions and major strides in this field. It covers the key advances in the design and engineering of nano‐ and micro‐structured Si anode architectures, and strategies of surface modification. Additionally, it explores the impacts of external pressure, the role of binders and conductive additives, and the implications of varying Si particle size. Beyond providing a detailed account of the evolution of Si anodes within SSE LIBs, this work also identifies critical research challenges that urgently need addressing. These include the electrochemical‐mechanical evolution behavior and failure mechanism of Si anodes in SSE LIBs, strategies for structural and interface modifications, methods for preparing Si electrodes, advancements in high‐performance SSEs, and the development of scalable technologies for SSE thin films. Moreover, it discusses high‐energy cathodes tailored for Si‐based SSE LIBs. With the widespread adoption of metagenomic sequencing, new perspectives have emerged for studying microbial ecological networks, yielding metabolic evidence of interspecies interactions that traditional co‐occurrence networks cannot infer. This protocol introduces the integrated Network Analysis Pipeline 2.0 (iNAP 2.0), which features an innovative metabolic complementarity network for microbial studies from metagenomics sequencing data. iNAP 2.0 sets up a four‐module process for metabolic interaction analysis, namely: (I) Prepare genome‐scale metabolic models; (II) Infer pairwise interactions of genome‐scale metabolic models; (III) Construct metabolic interaction networks; and (IV) Analyze metabolic interaction networks. Starting from metagenome‐assembled or complete genomes, iNAP 2.0 offers a variety of methods to quantify the potential and trends of metabolic complementarity between models, including the PhyloMint pipeline based on phylogenetic distance‐adjusted metabolic complementarity, the SMETANA (species metabolic interaction analysis) approach based on cross‐feeding substrate exchange prediction, and metabolic distance calculation based on parsimonious flux balance analysis (pFBA). Notably, iNAP 2.0 integrates the random matrix theory (RMT) approach to find the suitable threshold for metabolic interaction network construction. Finally, the metabolic interaction networks can proceed to analysis using topological feature analysis such as hub node determination. In addition, a key feature of iNAP 2.0 is the identification of potentially transferable metabolites between species, presented as intermediate nodes that connect microbial nodes in the metabolic complementarity network. To illustrate these new features, we use a set of metagenome‐assembled genomes as an example to comprehensively document the usage of the tools. iNAP 2.0 is available at https://inap.denglab.org.cn for all users to register and use for free. The aim of this study was to establish a HPLC method for the simultaneous determination of baicalin and chlorogenic acid in Gongyingqinglan Heji. After extraction with 20% methanol, Gongyingqinglan Heji was separated by C18 chromatographic column (4.6 mmx150 mm, 3.5 µm). The column temperature was set at 35 °C, the detection wavelength was 324 nm, the photo-diode array detector was used for analysis, and external standard method was used for quantification. The results showed that the linear relationship between baicalin and chlorogenic acid was good within the ranges of 20.91~209.12 mg/L and 5.12~51.20 mg/L, respectively (R²>0.999 0). The average recovery rate of baicalin was 101.09%, and the coefficient of variation was 1.15%. The average recovery rate of chlorogenic acid was 99.59%, and the coefficient of variation was 1.46%. The system adaptability test was good, and the chromatographic column had good tolerance, fully meeting the detection requirements. The study indicates that this method has strong operability, with good stability, repeatability and accuracy, and is suitable for the simultaneous determination of baicalin and chlorogenic acid content in Gongyingqinglan Heji. Stretchable electrodes based on liquid metals (LM) are widely used in human‐machine interfacing, wearable bioelectronics, and other emerging technologies. However, realizing the high‐precision patterning and mechanical stability remains challenging due to the poor wettability of LM. Herein, a method is reported to fabricate LM‐based multilayer solid–liquid electrodes (m‐SLE) utilizing electrohydrodynamic (EHD) printed confinement template. In these electrodes, LM self‐assembled onto these high‐resolution templates, assisted by selective wetting on the electrodeposited Cu layer. This study shows that a m‐SLE composed of PDMS/Ag/Cu/EGaIn exhibits line width of ≈20 µm, stretchability of ≈100%, mechanical stability ≈10 000 times (stretch/relaxation cycles), and recyclability. The multi‐layer structure of m‐SLE enables the adjustability of strain sensing, in which the strain‐sensitive Ag part can be used for non‐distributed detection in human health monitoring and the strain‐insensitive EGaIn part can be used as interconnects. In addition, this study demonstrates that near field communication (NFC) devices and multilayer displays integrated by m‐SLEs exhibit stable wireless signal transmission capability and stretchability, suggesting its applicability in creating highly‐integrated, large‐scale commercial, and recyclable wearable electronics. Inertial sensors act as inertial references in space gravitational wave detection missions, necessitating that their internal test mass (TM) maintains minimal residual acceleration noise. High-energy particles and cosmic rays in space, along with ion pumps in ground-based torsion pendulum experiments, can cause charge accumulation on the TM surface, leading to increased residual acceleration noise. Consequently, a charge management system was introduced to control the TM's charge. The complex light path propagation within the electrode housing (EH) makes the TM's charging and discharging process difficult to theoretically calculate and fully simulate. To address this issue, we propose a simulation method for charging and discharging inertial sensors within ultraviolet (UV) charge management systems. This method innovatively considers the impact of photoelectron emission angle and the TM's position offset from symmetry on performance. The method also simulates charging and discharging rates over time under conditions of symmetry and preliminarily examines factors affecting the TM's equilibrium potential. Exposure to Pb2+ in the environment, especially in water, poses a significant threat to human health and urgently necessitates the development of highly sensitive Pb2+ detection methods. In this study, we have integrated the high sensitivity of electrochemical techniques with allosteric transcription factors (aTFs) to develop an innovative electrochemical biosensing platform. This biosensors leverage the specific binding and dissociation of DNA to the aTFs (PbrR) on electrode surfaces to detect Pb2+. Under the optimal conditions, the platform has a broad linear detection range from 1 pM to 10 nM and an exceptionally low detection threshold of 1 pM, coupled with excellent selectivity for Pb2+. Notably, the biosensor demonstrates regenerative capabilities, enabling up to five effective Pb2+ measurements. After one week of storage at 4 °C, effective lead ion detection was still possible, demonstrating the biosensor's excellent stability, this can effectively save the cost of detection. The biosensor also achieves a recovery rate of 93.3% to 106.6% in real water samples. The biosensor shows its potential as a robust tool for the ultrasensitive detection of Pb2+ in environmental monitoring. Tubular injury and oxidative stress are involved in the pathogenesis of diabetic kidney disease (DKD). Astragaloside IV (ASIV) is a natural antioxidant. The effects and underlying molecular mechanisms of ASIV on DKD have not been elucidated. The db/db mice and high‐glucose‐stimulated HK2 cells were used to evaluate the beneficial effects of ASIV in vivo and in vitro. Succinylated proteomics was used to identify novel mechanisms of ASIV against DKD and experimentally further validated. ASIV alleviated renal dysfunction and proteinuria, downregulated fasting blood glucose, and upregulated insulin sensitivity in db/db mice. Meanwhile, ASIV alleviated tubular injury, oxidative stress, and mitochondrial dysfunction in vivo and in vitro. Mechanistically, ASIV reversed downregulated 17beta‐hydroxysteroid dehydrogenase type 10 (HSD17B10) lysine succinylation by restoring carnitine palmitoyl‐transferase1alpha (Cpt1a or CPT1A) activity in vivo and in vitro. Molecular docking and cell thermal shift assay revealed that ASIV may bind to CPT1A. Molecular dynamics simulations demonstrated K99 succinylation of HSD17B10 maintained mitochondrial RNA ribonuclease P (RNase P) stability. The K99R mutation of HSD17B10 induced oxidative stress and disrupted its binding to CPT1A or mitochondrial ribonuclease P protein 1 (MRPP1). Importantly, ASIV restored the interaction between HSD17B10 and MRPP1 in vivo and in vitro. We also demonstrated that ASIV reversed high‐glucose‐induced impaired RNase P activity in HK2 cells, which was suppressed upon K99R mutation of HSD17B10. These findings suggest that ASIV ameliorates oxidative stress‐associated proximal tubular injury by upregulating CPT1A‐mediated K99 succinylation of HSD17B10 to maintain RNase P activity. Background: Browning is the key problem hindering the industrialization of pear wine. The use of high‐yield glutathione Saccharomyces cerevisiae in the fermentation of pear wine can inhibit browning. Glutathione reductase (GR) can ensure the reduction of glutathione. Spore immobilization of enzymes is a new technology. It is a new attempt to apply spore‐immobilized GR in combination with high‐yield glutathione S. cerevisiae to inhibit browning of pear wine. Results: Saccharomyces cerevisiae spore immobilization enzyme technology was used to immobilize GR in the spores of mutant S. cerevisiae dit1∆, osw2∆ and chs3∆ and wild‐type S. cerevisiae. The enzyme activity of GR immobilized by chs3∆ spores was the highest of 3.08 U mg−1 min−1. The chs3∆ spore‐immobilized GR had certain resistance to ethanol, citric acid, sucrose, glucose and proteinase K. Electron microscopy analysis showed that the spore wall of chs3∆ had moderate size holes, which might be the main reason why it immobilized GR with the highest enzyme activity. And the GR was immobilized between the prespore membrane and mannoprotein layer of the spore wall. When chs3∆ spore‐immobilized GR (chs3∆‐GR) was added to Dangshan pear wine fermented by high‐yield glutathione S. cerevisiae JN32‐9, the presence of chs3∆‐GR could further protect amino acids, polyphenols and glucose from oxidation, thereby reducing the browning of the pear wine during storage by 47.32%. Conclusion: GR immobilized by S. cerevisiae spores was effective in inhibiting the browning of pear wine. Summary: The aim of this study was to create gluten‐free chiffon cakes by replacing wheat flour with rice flour (100%, 90% and 70% w/w) and adding yam (0%–30%). The effect of yam's water‐soluble fibre on cake batter properties was studied, including batter density, specific volume, colour, texture, moisture content, water activity and volatile compounds. Results revealed that yam addition decreased crumb lightness value, crust brown index and batter density. Batter viscosity and cake specific volume increased with yam levels, with the optimal amount being 10%. Specific volume decreased at a yam addition of 30%. Twenty‐four compounds were identified in gluten‐free chiffon cake with 10% yam addition (10Y). 1‐Octen‐3‐ol was the primary contributor, accounting for 89.9% odour activity value (OAV) of the aroma with a fruity, buttery, and mushroom odour, attributed to its low threshold. In addition, aldehyde compounds including hexanal, 3‐methylbutanal, 2‐methylbutanal, pentanal, 2‐methylpropanal, nonanal, octanal and heptanal, accounting for 9% of OAV, had high perception thresholds.
