Parents are one of the most important factors promoting children's educational success. Using parent survey data from rural primary and middle schools in one county, this study examines the implementation of the parent councils (PCs) policy in rural China, specifically, the relationships among PCs, school-based parent involvement (PI), and parent satisfaction. Our analysis shows that not all schools have established PCs and only a small proportion of parents were aware of the existence of PCs. Multi-level mixed effects logistic regression results indicate that having a PC and perceiving a PC in school were both associated with improved parent-teacher communication and parent participation in various school activities. However, the magnitudes of the relationships were substantially higher in the models using perceived existence of PCs. Because of more frequent parent-teacher communication and parent participation in school activities, parent satisfaction levels were higher in schools with PCs, and the satisfaction levels were even higher in schools where parents were aware of the PC's existence. The findings contribute to our understanding of the importance of PI and have implications for policymakers and school leaders to promote parent satisfaction by establishing PCs, raising the awareness of PCs among parents, and increasing meaningful school-based PI.

This study argues that the degree of linguistic abstraction in negative reviews can lead to varying levels of persuasiveness and that tailoring response strategies and voice according to the situation mitigates the impact of negative reviews. This study examines the impact of language abstraction in negative online reviews on potential customers’ attitudes and purchase intention, as well as the moderating effects of managerial responses, response strategies, and response voices on the persuasiveness of negative reviews. The data were collected through three studies that focused on reviews and managerial responses across both the service and functional product categories. The mediation analysis showed that, compared to abstract negative reviews, concrete negative reviews could significantly enhance potential customers’ attribution of responsibility toward service or product providers and their perception of stability and controllability of negative events, leading to a more negative perception of the company, but not lower purchase intention. Additionally, the moderation analysis showed that corporate responses could significantly reduce potential customers’ attribution of responsibility to the disputing company and improve their evaluation. Accommodative responses to concrete negative reviews and defensive responses to abstract negative reviews induced potential customers to make favorable judgments about the firm.

Osteosarcoma (OS) is the most common primary malignant bone tumor. In the clinic, usual strategies for OS treatment include surgery, chemotherapy, and radiation. However, all of these therapies have complications that cannot be ignored. Therefore, the search for better OS treatments is urgent. Black phosphorus (BP), a rising star of 2D inorganic nanoparticles, has shown excellent results in OS therapy due to its outstanding photothermal, photodynamic, biodegradable and biocompatible properties. This review aims to present current advances in the use of BP nanoparticles in OS therapy, including the synthesis of BP nanoparticles, properties of BP nanoparticles, types of BP nanoparticles, and modification strategies for BP nanoparticles. In addition, we have discussed comprehensively the application of BP in OS therapy, including single, dual, and multimodal synergistic OS therapies, as well as studies about bone regeneration and antibacterial properties. Finally, we have summarized the conclusions, limitations and perspectives of BP nanoparticles for OS therapy.

BackgroundThe angiography-derived index of microvascular resistance (A-IMR) is a novel tool for diagnosing coronary microvascular dysfunction (CMD) addressing limitation of unavailability. However, the clinical value of A-IMR remains controversial.MethodsA systematic review and meta-analysis was conducted. PubMed, EMBASE, Cochrane Library and Web of Science were searched for relevant studies. Studies that reported estimates of A-IMR's diagnostic accuracy (with thermodilution-based IMR as the reference test) and/or predictions of adverse cardiovascular events were selected. Pooled sensitivity, specificity, area under the summary receiver operating characteristic curve (sROC) were calculated to measure diagnostic performance; pooled hazard/risk ratio (HR/RR) and 95% confidence interval (95% CI) of major adverse cardiovascular events (MACE) or other independent adverse events were calculated to measure prognostic effect. This study was registered with PROSPERO (CRD42023451884).ResultsA total of 12 diagnostic studies pooling 1,642 vessels and 12 prognostic studies pooling 2,790 individuals were included. A-IMR yielded an area under sROC of 0.93 (95% CI: 0.91, 0.95), a pooled sensitivity of 0.85 (95% CI: 0.79, 0.89) and a pooled specificity of 0.89 (95% CI: 0.83, 0.93) for the diagnosis of CMD. CMD diagnosed using A-IMR was associated with higher risks of MACE (HR, 2.73, 95% CI: 2.16, 3.45), CV death (RR, 2.39, 95% CI: 1.49, 3.82) and heart failure hospitalization (HR, 2.30, 95% CI: 1.53, 3.45).ConclusionA-IMR demonstrated high diagnostic accuracy for CMD and showed a strong prognostic capability in predicting the risk of adverse CV outcomes.Systematic Review Registrationhttps://www.crd.york.ac.uk/prospero/display_record.php?ID=CRD42023451884, PROSPERO (CRD42023451884).

Objective: Individual differences were observed in the clinical efficacy of Botulinum toxin A (BoNT-A) in the treatment of the primary Meige syndrome. Our study aimed to explore the potential associations between the clinical efficacy of BoNT-A in the treatment of the primary Meige syndrome and variants of SNAP25, SV2C and ST3GAL2, which are involving in the translocation of the BoNT-A in vivo. Methods: Patients with the primary Meige syndrome treated with BoNT-A were enrolled. Clinical efficacy was evaluated by the maximum improvement rate of motor symptoms and the duration of efficacy. Variants of SNAP25, SV2C and ST3GAL2 were obtained by Sanger sequencing. Another cohort diagnosed with primary cervical dystonia was also enrolled in the replication stage. Results: Among the 104 primary Meige syndrome patients, 80 patients (76.9%) had a good efficacy (the maximum improvement rate of motor symptoms ≥30%) and 24 (23. 1%) had a poor (the maximum improvement rate of motor symptoms

This paper introduces the current situation of carbon trading market in China and the effect of carbon emission reduction in each region. Theoretically, it expounds the influence mechanism and spatial spillover way of carbon trading market on regional carbon emission. Next, we use the data of 30 provinces in China from 2006 to 2019 to build a continuous spatial difference in difference model (SDM-DID) to empirically study the spatial spillover effect of carbon trading market on regional emission reduction. The results are as follows: First, the implementation of the pilot policy of carbon emission trading has significantly promoted the carbon emission reduction in the pilot areas. From the perspective of impact mechanism, the implementation of carbon trading pilot policy promotes carbon emission reduction in pilot areas by promoting clean transformation of energy consumption structure, improving technology absorption capacity and stimulating development of low-carbon technologies. From the perspective of spatial spillover effect, China’s regional carbon emissions have significant spatial spillover effect, and carbon emissions trading has spatial spillover effect. From the perspective of spatial spillover, the carbon emission trading market promotes the carbon emission reduction in neighboring areas by promoting the clean transformation of energy structure in neighboring areas, improving technology absorption capacity and promoting technological progress.

Sonopiezoelectric therapy (SPT) has gained ever‐increasing prominence in tumor therapy as a combination of ultrasound (US) and piezoelectric nanomaterials to generate cytotoxic reactive oxygen species (ROS) for tumor suppression. In this study, the facile hydrothermal method is used to construct the distinct 2D piezocatalyst Bi2Fe4O9 nanosheets (BFO NSs). The engineered BFO NSs can create a piezoelectric field when exposed to US irradiation, facilitating the separation of electron–hole (e−–h+) pairs to play a crucial role in Fe3+/Fe2+ circulation, which catalyzes the activation of peroxymonosulfate (PMS), resulting in an amplified production of cytotoxic sulfate radical (SO4•−). The produced SO4•− combines with ROS generated by SPT including hydroxyl radicals (•OH), and singlet oxygen (1O2), leading to the augmented oxidative tumor suppression. The BFO NSs and PMS are further integrated into a thermal hydrogel (BFO/PMS‐Gel) for in vitro and in vivo investigations on their catalytic antitumor effects. In both in vitro and in vivo studies, it is validated that BFO/PMS‐Gel, under US irradiation, significantly enhances radicals generation efficiency, which achieves superior antitumor performance while maintaining high therapeutic biocompatibility and biosafety, providing a representative paradigm of US‐activated multiple radicals production as enabled by piezoelectric nanomedicine for antitumor treatment.

Background: A conclusive evidence regarding the optimal concentration and volume of local anesthetic for quadratus lumborum block is lacking. Methods: In this single-center, prospective, randomized, controlled study, 60 patients scheduled for laparoscopic colorectal surgery were randomly assigned to 3 different combinations of volume and concentration of ropivacaine (3 mg/kg) - Group 0.25%, Group 0.375% and Group 0.5%. All subjects received ultrasound-guided posterior quadratus lumborum block prior to the induction. The primary outcome was the complete sensory block rate of surgical site measured at 30 min after quadratus lumborum block, after extubation, at 12, 24, and 48 h after operation. Secondary outcomes were the changes in hemodynamic parameters before and after incision (ΔSBP, ΔDBP and ΔHR), postoperative pain score, the sufentanil consumption after surgery, length of stay and adverse reactions. Hyperspectral images (HSI) suffer from limited labeled data and the curse of dimensionality, which makes it difficult to classify imbalanced and small-sized HSI data. To address the mentioned issues, pixel pair features is designed to handle small-size problem. However, the augmented data inherits the class imbalance and even exhibits a larger imbalance ratio. Therefore, the ensemble-based Synthetic Minority Oversampling Technique (SMOTE) is integrated into pixel pair features to deal with imbalanced datasets and strengthen the quality of the augmented datasets. The proposed method is evaluated on five groups of HSI datasets and compared against seven state-of-the-art contrast algorithms, including ensemble-CNN (E-CNN), CNN with pixel pair features, ensemble CNN with pixel pair features, spatial–spectral E-ResNet, E-Transfering-VGGNet, ContextualNet, and CNN with K-means SMOTE. The proposed method demonstrates superior performance across three distinct difficulty levels, as determined by its average performance rank on five HSI datasets. This underscores its effectiveness in mitigating small size and class imbalance challenges.

Abstract Smart chiral liquid crystal elastomers are a class of soft photonic crystals with periodic nanostructures. There are two kinds of chiral liquid crystal elastomers with structural colors: cholesteric liquid crystal elastomers with a one‐dimensional helical nanostructure and blue‐phase liquid crystal elastomers with a three‐dimensional photonic crystal nanostructure. The self‐assembled nanostructure of chiral liquid crystal elastomers can be dynamically controlled under external stimulation, and the reflected color can be adjusted throughout the visible light range. Along with the development of innovative material systems and cutting‐edge manufacturing technologies, researchers have proposed diverse strategies to design and synthesize chiral liquid crystal elastomers and have thoroughly investigated their properties and potential applications. Here, we provide a systematic review of the progress in the design and fabrication of smart chiral liquid crystal elastomers, focusing on the cholesteric liquid crystal elastomers via surface‐enforced alignment, bar coating, 3D printing, anisotropic deswelling methods as well as the three‐dimensional self‐assembly of blue‐phase liquid crystal elastomers without additional alignment. Smart chiral liquid crystal elastomers are able to respond quickly to external stimuli and have a wide range of applications in areas such as adaptive optics, color‐changing camouflage, soft robotics, and information encryption. This review concludes with a perspective on the opportunities and challenges for the future development of smart chiral liquid crystal elastomers.

The online monitoring of metal particles in lubricating oil plays an important role in equipment fault diagnosis technology. This paper designs an online wear monitoring warning sensor based on the electromagnetic induction of metal particles in lubricating oil. First, different from the traditional three-coil sensing and detection method, the sensor consists of two excitation coils and one detection coil. In addition, the parallel capacitance and detection capacitance parameters are obtained by COMSOL’s scanning frequency, and the best detection capacitance is 7.53 nF. Second, according to the obtained optimal detection parallel capacitance value, the amplitude data of the particle signal, as well as the ferromagnetic particles above 60 µm and the non-ferromagnetic particles above 80 µm, are obtained. Third, according to the principle of three-coil induction, a mapping relationship model was established between the size range of metal wear particles in lubricating oil. Finally, we set up an oil wear particle platform to verify the sensor performance. The ferromagnetic and non-ferromagnetic particles are divided into multiple size ranges, and the peak-to-peak values of the output signal of different-sized wear particles are measured. We developed a prototype sensor and verified the signal consistency and repeatability of the sensor to wear metal particles. In addition, we used the sensor to test a large number of metal particles of different sizes (14 ferromagnetic particle standard samples with particle sizes ranging from 20.64 to 457.59 µm and ten non-ferromagnetic particle standard samples with particle sizes ranging from 40.52 to 348.07 µm). The experimental results show that the sensor can directly achieve monitoring sensitivity for ferromagnetic particles greater than 64.57 µm and non-ferromagnetic (copper) particles greater than 82.83 µm under the 4.2 mm aperture flow channel. The sensor can effectively realize the sensitivity on monitoring of small ferromagnetic particles and non-ferromagnetic particles, and it is particularly sensitive to small non-ferromagnetic particles.

Pollen and spore records in prodeltaic sediments hold significant potential for reconstructing paleoecologic and paleoclimatic evolution. However, uncertainties in these reconstructions arise from millennial-scale prodelta evolution, which dominates stratigraphic development and consequently influences sedimentary processes and/or pollen provenance. Here we explore the intricate relationship between pollen/spore records and prodelta stratigraphic evolution, using established seismic stratigraphy and ten sediment cores (five new, five from literature) within both the proximal and distal (mud belt) parts of the Huanghe (Yellow River) prodelta. In the proximal region, dominant lobate stratigraphic development, accompanied by shifts in river mouth and depocenter, leads to variations in pollen assemblages and contents within individual cores and differences in vertical pollen distribution across core sites. Transport distance appears to be a key factor, with arboreal pollens, particularly saccate ones (e.g., Pinus), positively correlating with the distance from the river mouth in their percentages within a single delta lobe, while non-arboreal and non-saccate arboreal pollens show higher percentages within shorter transport distances, despite longer distances leading to decreased total pollen concentrations. Likely due to the total pollen concentration after extended long-distance transport, this pattern is not observable in the distal mud belt. Subsurface stratigraphy in this mud belt reveals a complex pollen provenance characterized by Artemisia-Ulmus-Chenopodiaceae-Pinus, with non-arboreal pollens in dominance. Therein, non-arboreal pollens are not consistent with deposition from long-distance transport, and Ulmus pollens are uncommon in the western Bohai Sea. Interestingly, surface sediments in the mud belt display a different assemblage, characterized by Pinus-Artemisia-Quercus, consistent with the nearby Luanhe River prodelta, suggesting recent pollen supply from nearby sources, possibly due to the recent abandonment of the mud belt. Additionally, an energetic longshore transport/erosional regime reduces pollen content at the mud-belt margins and create pollen sinks (with the highest concentration) in the mud patch (accumulation area) within the erosion-dominated region adjacent to the mud belt. Our findings confirm that stratigraphic evolution, alongside hydrodynamic conditions and pollen provenance, governs pollen assemblages in deltaic/prodeltaic sediments. They can provide insights for palynological and pollen-based paleoclimatic and paleoecologic studies in other deltas.

BackgroundMosquito-borne diseases pose serious public health threats in Zhejiang Province, China, and vector control is believed to be the primary method for reducing transmission. Due to severe resistance problems, effective and sustainable methods without chemical insecticides are urgently required to control mosquito vectors. Attractive toxic sugar baits (ATSB) are newly developed methods to control mosquitoes in recent decades with the core element sugar bait, which was invented according to the sugar-feeding behavior of mosquitoes. In this study, we developed a Novel Sugar Bait Device (NSBD) trap by combining sugar bait and physical adhesive capture technology. The study aimed to evaluate the effect of the NSBD trap on controlling mosquitoes in residential environments and to identify the optimal sugar solution concentration in the sugar bait of the NSBD for real use.MethodsFour residential villages in Ningbo City with similar geographic environments and mosquito densities were selected for field trials in 2022. One village (site 1) was designated as the control group, and three villages (sites 2–4) served as the test groups to assess the effectiveness of NSBD traps with different sugar solution concentrations (6, 8, and 10%) in the sugar bait. Larval and adult mosquito densities were monitored monthly before and semi-monthly after the trials using the CDC light trap and larval pipette method.ResultsBefore the trials, we monitored mosquito density for 3 months to confirm the baseline mosquito density among the four sites, and no statistical differences in adult and larval mosquitoes were found (adult, F = 3.047, p > 0.05; larvae, F = 0.436, p > 0.05). After the trials, all NCBD traps effectively controlled larval and adult mosquito densities, with the highest standard decrease rates of larval and adult mosquito densities at 57.80 and 86.31%, respectively, observed in site 4. The most suitable sugar solution concentration in the sugar bait was 10%.ConclusionNSBD traps effectively controlled mosquitoes in residential environments during field trials. Without the use of insecticides, this may be a promising choice for mosquito vector control to prevent mosquito-borne diseases.

Background: Ovarian cancer, as a highly malignant tumor, features the critical involvement of tumor-associated fibroblasts in the ovarian cancer tissue microenvironment. However, due to the apparent heterogeneity within fibroblast subpopulations, the specific functions of these subpopulations in the ovarian cancer tissue microenvironment remain insufficiently elucidated. Methods: In this study, we integrated single-cell sequencing data from 32 ovarian cancer samples derived from four distinct cohorts and 3226 bulk RNA-seq data from GEO and TCGA-OV cohorts. Utilizing computational frameworks such as Seurat, Monocle 2, Cellchat, and others, we analyzed the characteristics of the ovarian cancer tissue microenvironment, focusing particularly on fibroblast subpopulations and their differentiation trajectories. Employing the CIBERSORTX computational framework, we assessed various cellular components within the ovarian cancer tissue microenvironment and evaluated their associations with ovarian cancer prognosis. Additionally, we conducted Mendelian randomization analysis based on cis-eQTL to investigate causal relationships between gene expression and ovarian cancer. Results: Through integrative analysis, we identified 13 major cell types present in ovarian cancer tissues, including CD8+ T cells, malignant cells, and fibroblasts. Analysis of the tumor microenvironment (TME) cell proportions revealed a significant increase in the proportion of CD8+ T cells and CD4+ T cells in tumor tissues compared to normal tissues, while fibroblasts predominated in normal tissues. Further subgroup analysis of fibroblasts identified seven subgroups, with the MMP11+Fib subgroup showing the highest activity in the TGFβ signaling pathway. Single-cell analysis suggested that oxidative phosphorylation could be a key pathway driving fibroblast differentiation, and the ATRNL1+KCN + Fib subgroup exhibited chromosomal copy number variations. Prognostic analysis using a large sample size indicated that high infiltration of MMP11+ fibroblasts was associated with poor prognosis in ovarian cancer. SMR analysis identified 132 fibroblast differentiation-related genes, which were linked to pathways such as platinum drug resistance. Conclusions: In the context of ovarian cancer, fibroblasts expressing MMP11 emerge as the primary drivers of the TGF-beta signaling pathway. Their presence correlates with an increased risk of adverse ovarian prognoses. Additionally, the genetic regulation governing the differentiation of fibroblasts associated with ovarian cancer correlates with the emergence of drug resistance.

Based on the usual Wigner-Weyl transformation theory we find that the Wigner hyperbolic rotation in phase space will map onto fractional squeezing operator in Hilbert space. The merit of Weyl ordering and the coherent state representation of Fresnel operator is used in our derivation.

Summary: Embodied emissions from the production of building materials account for 17% of China’s carbon dioxide (CO2) emissions and are important to focus on as China aims to achieve its carbon neutrality goals. However, there is a lack of systematic assessments on embodied emissions reduction potential of building materials that consider both the heterogeneous industrial characteristics as well as the Chinese buildings sector context. Here, we developed an integrated model that combines future demand of building materials in China with the strategies to reduce CO2 emissions associated with their production, using, and recycling. We found that measures to improve material efficiency in the value-chain has the largest CO2 mitigation potential before 2030 in both Low Carbon and Carbon Neutrality Scenarios, and continues to be significant through 2060. Policies to accelerate material efficiency practices, such as incorporating embodied emissions in building codes and conducting robust research, development, and demonstration (RD&D) in carbon removal are critical.

Detecting earthquake-induced bathymetric changes helps to understand the geomorphologic process of tufa lakes. Traditional field measurement methods are difficult for spatially complete and continuous bathymetric mapping. Multi-temporal high-resolution optical satellite images are cost-efficient data used for bathymetric change detection. However, for detecting bathymetric changes in tufa lakes, collecting high-density depth calibration data and constructing highly robust water depth inversion models pose certain challenges. This study takes Huohua Lake before and after the Jiuzhaigou Earthquake as the research object, and carries out the bathymetric change detection based on high-resolution remote sensing data. Initially, the WorldView-2 (WV-2) multispectral images obtained before and after the earthquake under the water-storage state of the lake were used as the data source, and the unmanned aerial vehicle (UAV)-based measurement under the water-free state of the lake after the earthquake was used as the bathymetric calibration and validation data. Then using satellite-derived image reflectance, we constructed two-phase bathymetric models with machine learning methods, namely random forest (RF), support vector machine (SVM), and multi-layer perceptron (MLP). The comparison results with classical regression models indicate that machine learning-based water depth inversion models are generally superior. Specifically, the R2 (coefficient of determination) of the optimal model RF reach 0.85 and 0.91, with RMSE (root mean square error) of 1.40 m and 1.08 m. The bathymetric difference maps generated from water depth inversion results reveal that during the period from October 2016 to January 2022, the core area of Huohua Lake experienced more erosion than accretion due to the earthquake-induced flooding. The spatial patterns of changes show that the erosion mainly located in the raised tufa mound area, while the accretion was concentrated in the shallow flat area. This study provides a remote sensing approach for quantifying bathymetric changes in tufa lakes after extreme geological disasters.

Periprosthetic hip infection caused by Brucella abortus is rare and only a few cases have been reported. This current case report presents a case of a man in his early 50s who developed periprosthetic hip infection 2 years after right hip arthroplasty. There was no fever or pain, the usual cardinal signs of infection, except for a sinus tract at the previous surgical incision. Laboratory and arthrocentesis culture examinations (done twice) confirmed infection with B. abortus . Accordingly, a two-stage revision surgery was performed accompanied by antibiotic treatment with doxycycline and rifampicin after each stage. There was no recurrence at the 2-year follow-up, with good functional recovery of the hip joint. Clinically, this case serves to highlight the fact that periprosthetic hip infections caused by B. abortus might not present with the typical symptoms such as fever or hip pain. Furthermore, this current case involved a chronic sinus tract, so the diagnostic and therapeutic course of this case offers useful insights for managing similar cases in the future. In addition, a review of the literature on the diagnosis and treatment of Brucella -caused periprosthetic hip infection is presented.

Abstract Background The main subtypes of idiopathic inflammatory myopathies (IIMs)—polymyositis (PM) and dermatomyositis (DM)—are often presented as interstitial lung disease (ILD) in clinical practice; therefore, many researchers have combined the three studies into PM/DM with ILD. Methods Using bibliometrics, the research status, progress, and hotspots of PM/DM with ILD between 2000 and 2022 were studied. Literature data on PM/DM with ILD were retrieved from the Web of Science (WoS) database for the research period. Visualization software, including VOSviewer, Pajek, CiteSpace, and Scimago Graphica were used for bibliometric analysis. Results A total of 1555 relevant articles were obtained, and the overall research in this field showed an increasing trend. Regarding contributing countries and venues, Japan published the most articles while Rheumatology was the most prolific journal. Regarding authors, the most published article was by Wang Guochun from Changchun University of Technology in China. Keyword analysis and cocited literature cluster analysis showed that diagnosis, classification, autoantibodies, antibodies, prognosis, complications, and treatment of PM/DM with ILD have been hot topics in this field recently. Moreover, our study shows that anti‐mda5 antibody, mortality, gene 5 antibody, IIMs, double‐blind, and prognostic factors, among others, may be new hot topics. Conclusion This study found that research on PM/DM with ILD has increased over time, and scholars are paying more attention to this field. The development of new drugs for the management, treatment, and prevention of PM/DM with ILD is the primary task of researchers and a direction for future research in this field.

BackgroundVulnerable plaque was associated with recurrent cardiovascular events. This study was designed to explore predictive biomarkers of vulnerable plaque in patients with coronary artery disease.MethodsTo reveal the phenotype-associated cell type in the development of vulnerable plaque and to identify hub gene for pathological process, we combined single-cell RNA and bulk RNA sequencing datasets of human atherosclerotic plaques using Single-Cell Identification of Subpopulations with Bulk Sample Phenotype Correlation (Scissor) and Weighted gene co-expression network analysis (WGCNA). We also validated our results in an independent cohort of patients by using intravascular ultrasound during coronary angiography.ResultsMacrophages were found to be strongly correlated with plaque vulnerability while vascular smooth muscle cell (VSMC), fibrochondrocyte (FC) and intermediate cell state (ICS) clusters were negatively associated with unstable plaque. Weighted gene co-expression network analysis showed that Secreted Phosphoprotein 1 (SPP1) in the turquoise module was highly correlated with both the gene module and the clinical traits. In a total of 593 patients, serum levels of SPP1 were significantly higher in patients with vulnerable plaques than those with stable plaque (113.21 [73.65 - 147.70] ng/ml versus 71.08 [20.64 - 135.68] ng/ml; P < 0.001). Adjusted multivariate regression analysis revealed that serum SPP1 was an independent determinant of the presence of vulnerable plaque. Receiver operating characteristic curve analysis indicated that the area under the curve was 0.737 (95% CI 0.697 - 0.773; P < 0.001) for adding serum SPP1 in predicting of vulnerable plaques.ConclusionElevated serum SPP1 levels confer an increased risk for plaque vulnerability in patients with coronary artery disease.

ABSTRACTReplication of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) genome is mediated by a complex of non-structural proteins (NSPs), of which NSP7 and NSP8 serve as subunits and play a key role in promoting the activity of RNA-dependent RNA polymerase (RdRp) of NSP12. However, the stability of subunits of the RdRp complex has rarely been reported. Here, we found that NSP8 was degraded by the proteasome in host cells, and identified tripartite motif containing 22 (TRIM22) as its E3 ligase. The interferon (IFN) signaling pathway was activated upon viral invasion into host cells, and TRIM22 expression increased. TRIM22 interacted with NSP8 and ubiquitinated it at Lys97 via K48-type ubiquitination. TRIM22 overexpression significantly reduced viral RNA and protein levels. Knockdown of TRIM22 enhanced viral replication. This study provides a new explanation for treating patients suffering from SARS-CoV-2 with IFNs and new possibilities for drug development targeting the interaction between NSP8 and TRIM22.IMPORTANCENon-structural proteins (NSPs) play a crucial role in the replication of severe acute respiratory syndrome coronavirus 2, facilitating virus amplification and propagation. In this study, we conducted a comprehensive investigation into the stability of all subunits comprising the RNA-dependent RNA polymerase complex. Notably, our results reveal for the first time that NSP8 is a relatively unstable protein, which is found to be readily recognized and degraded by the proteasome. This degradation process is mediated by the host E3 ligase tripartite motif containing 22 (TRIM22), which is also a member of the interferon stimulated gene (ISG) family. Our study elucidates a novel mechanism of antiviral effect of TRIM22, which utilizes its own E3 ubiquitin ligase activity to hinder viral replication by inducing ubiquitination and subsequent degradation of NSP8. These findings provide new ideas for the development of novel therapeutic strategies. In addition, the conserved property of NSP8 raises the possibility of developing broad antiviral drugs targeting the TRIM22-NSP8 interaction.

PurposeSteroid-induced osteonecrosis of the femoral head (SONFH) is a refractory orthopedic hip joint disease that primarily affects middle-aged and young individuals. SONFH may be caused by ischemia and hypoxia of the femoral head, where mitochondria play a crucial role in oxidative reactions. Currently, there is limited literature on whether mitochondria are involved in the progression of SONFH. Here, we aim to identify and validate key potential mitochondrial-related genes in SONFH through bioinformatics analysis. This study aims to provide initial evidence that mitochondria play a role in the progression of SONFH and further elucidate the mechanisms of mitochondria in SONFH.MethodsThe GSE123568 mRNA expression profile dataset includes 10 non-SONFH (non-steroid-induced osteonecrosis of the femoral head) samples and 30 SONFH samples. The GSE74089 mRNA expression profile dataset includes 4 healthy samples and 4 samples with ischemic necrosis of the femoral head. Both datasets were downloaded from the Gene Expression Omnibus (GEO) database. The mitochondrial-related genes are derived from MitoCarta3.0, which includes data for all 1136 human genes with high confidence in mitochondrial localization based on integrated proteomics, computational, and microscopy approaches. By intersecting the GSE123568 and GSE74089 datasets with a set of mitochondrial-related genes, we screened for mitochondrial-related genes involved in SONFH. Subsequently, we used the good Samples Genes method in R language to remove outlier genes and samples in the GSE123568 dataset. We further used WGCNA to construct a scale-free co-expression network and selected the hub gene set with the highest connectivity. We then intersected this gene set with the previously identified mitochondrial-related genes to select the genes with the highest correlation. A total of 7 mitochondrial-related genes were selected. Next, we performed Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis on the selected mitochondrial-related genes using R software. Furthermore, we performed protein network analysis on the differentially expressed proteins encoded by the mitochondrial genes using STRING. We used the GSEA software to group the genes within the gene set in the GSE123568 dataset based on their coordinated changes and evaluate their impact on phenotype changes. Subsequently, we grouped the samples based on the 7 selected mitochondrial-related genes using R software and observed the differences in immune cell infiltration between the groups. Finally, we evaluated the prognostic significance of these features in the two datasets, consisting of a total of 48 samples, by integrating disease status and the 7 gene features using the cox method in the survival R package. We performed ROC analysis using the roc function in the pROC package and evaluated the AUC and confidence intervals using the ci function to obtain the final AUC results.ResultsIdentification and analysis of 7 intersecting DEGs (differentially expressed genes) were obtained among peripheral blood, cartilage samples, hub genes, and mitochondrial-related genes. These 7 DEGs include FTH1, LACTB, PDK3, RAB5IF, SOD2, and SQOR, all of which are upregulated genes with no intersection in the downregulated gene set. Subsequently, GO and KEGG pathway enrichment analysis revealed that the upregulated DEGs are primarily involved in processes such as oxidative stress, release of cytochrome C from mitochondria, negative regulation of intrinsic apoptotic signaling pathway, cell apoptosis, mitochondrial metabolism, p53 signaling pathway, and NK cell-mediated cytotoxicity. GSEA also revealed enriched pathways associated with hub genes. Finally, the diagnostic value of these key genes for hormone-related ischemic necrosis of the femoral head (SONFH) was confirmed using ROC curves.ConclusionBID, FTH1, LACTB, PDK3, RAB5IF, SOD2, and SQOR may serve as potential diagnostic mitochondrial-related biomarkers for SONFH. Additionally, they hold research value in investigating the involvement of mitochondria in the pathogenesis of ischemic necrosis of the femoral head.

As high-energy cathode materials, conversion-type metal fluorides provide a prospective pathway for developing next-generation lithium-ion batteries. However, they suffer from severe performance decay owing to continuous structural destruction and active material dissolution upon cycling, which worsen at elevated temperatures. Here, we design a novel FeF2 cathode with in situ polymerized solid-state electrolyte systems to enhance the cycling ability of metal fluorides at 60 ​°C. Novel FeF2 with a mesoporous structure (meso-FeF2) improves Li+ diffusion and relieves the volume change that typically occurs during the alternating conversion reactions. The structural stability of the meso-FeF2 cathode is strengthened by an in situ polymerized solid-state electrolyte, which prevents the pulverization and ion dissolution that are inevitable for conventional liquid electrolytes. Under the double action of this in situ polymerized solid-state electrolyte and the meso-FeF2's mesoporous structure, the active material maintains an intact SEI layer and part of the mesoporous structure after long charge–discharge cycling, showing excellent cycling stability at high temperatures.
