Diabetic chronic wounds or amputation, which are complications of diabetes mellitus (DM), are a cause of great suffering for diabetics. In addition to the lack of oxygen, elevated reactive oxygen species (ROS) and reduced vascularization, microbial invasion is also a critical factor that induces non-healing chronic diabetic wounds, ie, wounds still remaining in the stage of inflammation, after which the wound tissue begins to age and becomes necrotic. To clear up the infection, alleviate the inflammation in the wound and prevent necrosis, many kinds of hydrogel have been fabricated to eliminate infections with pathogens. The unique properties of hydrogels make them ideally suited to wound dressings because they provide a moist environment for wound healing and act as a barrier against bacteria. This review article will mainly cover the recent developments and innovations of antibacterial hydrogels for diabetic chronic wound healing.

Thyroid cancer is considered to be one of the most common endocrine tumors worldwide. Cystathionine β-synthase (CBS) plays a crucial role in the occurrence of several types of malignancies. And yet, the mechanism of action of CBS in the growth of thyroid carcinoma cells is still unrevealed. We found that CBS level in thyroid carcinoma tissue was higher than that in adjacent normal tissue. The overexpression of CBS enhanced the proliferation, migration, and invasion of thyroid cancer cells, while the downregulation of CBS exerted reverse effects. CBS overexpression reduced the levels of cleaved caspase-3 and cleaved poly ADP-ribose polymerase in thyroid cancer cells, whereas CBS knockdown showed reverse trends. CBS overexpression decreased reactive oxygen species (ROS) levels but increased the levels of Wnt3a and phosphorylations of phosphatidylinositol 3-kinase (PI3K), protein kinase B (PKB/AKT), mammalian target of rapamycin (mTOR), β-catenin, and glycogen synthase kinase-3 beta, while CBS knockdown exerted opposite effects. In addition, CBS overexpression promoted the growth of xenografted thyroid carcinoma, whereas CBS knockdown decreased the tumor growth by modulating angiogenesis, cell cycle, and apoptosis. Furthermore, aminooxyacetic acid (an inhibitor of CBS) dose-dependently inhibited thyroid carcinoma cell growth. CBS can regulate the proliferation, migration, and invasion of human thyroid cancer cells via ROS-mediated PI3K/AKT/mTOR and Wnt/β-catenin pathways. CBS can be a potential biomarker for diagnosing or prognosing thyroid carcinoma. Novel donors that inhibit the expression of CBS can be developed in the treatment of thyroid carcinoma.

PEST-containing nuclear protein (PCNP) is a short-lived novel nuclear protein. It has been well evaluated that PCNP mediates the progression of several cancers, but the exact mechanisms are still under investigation. In this study, we provided an e-science view of PCNP protein from the aspects of protein structure, interactions, and bioinformatics-based analysis related to evolutionary features as well as proteomic profile. The phylogenetic relationship results reveal that PCNP is closely related to Pan troglodytes and the Bovidae family, while being distantly related to the Muridae family. The analysis of the physicochemical properties of PCNP demonstrated that it is a thermolabile protein which is slightly acidic and hydrophilic in nature. Further, coexpression and protein-protein interaction analyses were carried out, which demonstrated that the PCNP gene was remarkably expressed with MORF4LI and RSL24D1 genes and has close interactions with TRAM1, PSMC6, SRP9, PRKRIR, UHRF2, and BMI1 proteins. Gene ontology and pathway enrichment analyses showed that PCNP has a high tendency to work in cell cycle regulation. Moreover, among the four 3D structure generating tools, I-TASSER-generated structure had the highest quality factor score. The validation analysis revealed that the I-TASSER-generated structure exhibited the best quality factor score with maximum amino acids in the favored region. In addition, molecular dynamic simulation analysis approved the stable structure of the PCNP. This is the first study that highlights the usefulness of the understanding of the structural and functional analysis of the PCNP, which lays the groundwork for further experimental studies to validate the outcome.

Mpox (MPX), a zoonotic infection reported as early as 1970, is a double-stranded DNA virus, Mpox virus (MPXV), belonging to the genus Orthopoxvirus. MPX, a virus closely related to smallpox, was once considered a geographically restricted infection because it was predominantly endemic in West and Central Africa, except for the Democratic Republic of the Congo (DRC). However, in May 2022, a rare case of MPX was diagnosed in the UK. As of February 25, 2023, 86,127 confirmed cases and 97 deaths have been reported globally. It spreads through close contact with an infected person, infected animal, or an object which an infected person has touched. Although most people infected with MPXV may present with mild symptoms and will recover fully within a few weeks, people can have serious diseases and complications and may need to be hospitalized with it. Although smallpox vaccines and antiviral drugs are also effective against MPX, there is still no specific vaccine or drug for MPXV infection. Therefore, we need to take the right attitude and measures to stop the MPXV outbreak. In this review, we summarize the characteristics, mechanisms, immunology, and transmission of MPXV, as well as current vaccines, diagnostics, and treatments used for MPX, and provide clues for controlling MPX outbreaks and preventing such diseases.

Numerous studies have clarified the involvement of circular RNAs (circRNAs) in modulating malignant behavior of non-small cell lung cancer (NSCLC), while the concrete mechanism is not completely elucidated. The aim of the study was to figure out the latent functions and molecular mechanisms of circRNA Eps15-homology domain containing protein 2 (EHD2) on NSCLC proliferation, glycolysis and autophagy. The results clarified in NSCLC elevated expression of circEHD2 and declined expression of microRNA (miR)-3186-3p. Repressive circEHD2 or enhancive miR-3186-3p facilitated cell apoptosis rate and autophagy substrates LC3BII and Beclin-1, but curbed the colony-formation and DNA replication ability of NSCLC, glucose consumption, lactic acid production, glycolytic rate-limiting enzyme HK-2 and glutamine hydrolase GLS1 and P62, while overexpressed circEHD2 was adverse. Meanwhile, the impacts of repressive and elevated circEHD2 on NSCLC were turned around via reduced miR-3186-3p or forkhead box k1 (FOXK1) separately. Mechanically, FOXK1 was augmented via circEHD2's competitive integration of miR-3186-3p. Depressive circEHD2 refrained NSCLC tumor growth, which was accelerated via enhancive one. All in all, circEHD2 accelerates the proliferation and glycolysis of NSCLC, but refrains autophagy and apoptosis via strengthening FOXK1 via the adsorption of miR-3186-3p, which is supposed to be a latent molecular target for NSCLC therapy later.

Melanoma and non-melanoma skin cancers are now the most common types of skin cancer that have been reached to epidemic proportion. Based on the rapid prevalence of skin cancers, and lack of efficient drug delivery systems, it is essential to surge the possible ways to prevent or cure the disease. Aim of review Although surgical modalities and therapies have been made great progress in recent years, however, there is still an urgent need to alleviate its increased burden. Hence, understanding the precise pathophysiological signaling mechanisms and all other factors of such skin insults will be beneficial for the development of more efficient therapies. Key scientific concepts of review In this review, we explained new understandings about onset and development of skin cancer and described its management via polymeric micro/nano carriers-based therapies, highlighting the current key bottlenecks and future prospective in this field. In therapeutic drug/gene delivery approaches, polymeric carriers-based system is the most promising strategy. This review discusses that how polymers have successfully been exploited for development of micro/nanosized systems for efficient delivery of anticancer genes and drugs overcoming all the barriers and limitations associated with available conventional therapies. In addition to drug/gene delivery, intelligent polymeric nanocarriers platforms have also been established for combination anticancer therapies including photodynamic and photothermal, and for theranostic applications. This portfolio of latest approaches could promote the blooming growth of research and their clinical availability.

Thyroid cancer (TC) has increased globally, with a prominent increase in small, papillary thyroid cancers. PEST-containing nuclear protein (PCNP), a nuclear protein, has been found to be associated with human cancers in recent years. However, the role and molecular mechanism of PCNP in thyroid cancer remain underexplored. In the present study, the results showed that the expression levels of PCNP in human thyroid tissues were higher than those in adjacent non-tumor tissues. Overexpression of PCNP reduced the proliferation, migration, and invasion of human thyroid cancer cells and down-regulation of PCNP showed reverse effects. In addition, PCNP regulated cell cycle arrest through modifications in the expression of cell cycle regulating genes and PCNP affected apoptosis via activation of ERK/JNK/p38 pathway in thyroid cancer cells. Moreover, PCNP overexpression promoted autophagy by reducing the expression levels of Wnt/β-catenin pathway in TC cells, however, PCNP knockdown had opposite effects. Background The roles of Polypyrimidine tract-binding protein 3 (PTBP3) in regulating lung squamous cell carcinoma (LUSC) cells progression is unclear. The aim of this study was to investigate the role of PTBP3 in LUSC. Methods Expression and survival analysis of PTBP3 was firstly investigated using TCGA datasets. Quantitative reverse transcription PCR and Western blot were performed to detect PTBP3 expression in clinical samples. Moreover, cell counting kit 8 (CCK-8) assays, colony formation assays and in vivo tumor formation assays were used to examine the effects of PTBP3 on LUSC cell proliferation. RNA-sequence and analysis explores pathways regulated by PTBP3.Flow cytology was used analyzed cell cycle. Cell cycle-related markers were analyzed by Western blot. Results PTBP3 was found to be overexpressed in LUSC tissues compared with normal tissues. High PTBP3 expression was significantly correlated with poor prognosis. In vitro and vivo experiments demonstrated that PTBP3 knockdown caused a significant decrease in the proliferation rate of cells. Bioinformatics analysis showed that PTBP3 involved in cell cycle pathway regulation in LUSC. Furthermore, PTBP3 knockdown arrested cell cycle progression at S phase via decreasing CDK2/Cyclin A2 complex. In addition, downregulation of PTBP3 significantly decreased the expression of CDC25A. Conclusions Our results suggest that PTBP3 regulated LUSC cell proliferation via cell cycle and might be a potential target for molecular therapy of LUSC.

In practical multiple attribute decision making (MADM) problems, the interest groups or individuals intentionally set attribute weights to achieve their own benefits. In this case, the rankings of different alternatives are changed strategically, which is called the strategic weight manipulation in MADM. Sometimes, the attribute values are given with imprecise forms. Several theories and methods have been developed to deal with uncertainty, such as probability theory, interval values, intuitionistic fuzzy sets, hesitant fuzzy sets, etc. In this paper, we study the strategic weight manipulation based on the belief degree of uncertainty theory, with uncertain attribute values obeying linear uncertain distributions. It allows the attribute values to be considered as a whole in the operation process. A series of mixed 0-1 programming models are constructed to set a strategic weight vector for a desired ranking of a particular alternative. Finally, an example based on the assessment of the performance of COVID-19 vaccines illustrates the validity of the proposed models. Comparison analysis shows that, compared to the deterministic case, it is easier to manipulate attribute weights when the attribute values obey the linear uncertain distribution. And a further comparative analysis highlights the performance of different aggregation operators in defending against the strategic manipulation, and highlights the impacts on ranking range under different belief degrees.

Accurately evaluating minimal residual disease (MRD) could facilitate early intervention and personalized adjuvant therapies. Here, using ultradeep targeted next-generation sequencing (NGS), we evaluate the clinical utility of circulating tumor DNA (ctDNA) for dynamic recurrence risk and adjuvant chemotherapy (ACT) benefit prediction in resected non-small cell lung cancer (NSCLC). Both postsurgical and post-ACT ctDNA positivity are significantly associated with worse recurrence-free survival. In stage II-III patients, the postsurgical ctDNA positive group benefit from ACT, while ctDNA negative patients have a low risk of relapse regardless of whether or not ACT is administered. During disease surveillance, ctDNA positivity precedes radiological recurrence by a median of 88 days. Using joint modeling of longitudinal ctDNA analysis and time-to-recurrence, we accurately predict patients’ postsurgical 12-month and 15-month recurrence status. Our findings reveal longitudinal ctDNA analysis as a promising tool to detect MRD in NSCLC, and we show pioneering work of using postsurgical ctDNA status to guide ACT and applying joint modeling to dynamically predict recurrence risk, although the results need to be further confirmed in future studies., ctDNA has been shown to identify minimal residual disease (MRD) and is thus dynamically monitored in different types of tumours. Here, the authors show that serial longitudinal ctDNA analysis can be used as a tool to detect MRD, inform the use of adjuvant therapy, and predict recurrence risk in lung cancer.

Background: Most primary school students in China tend to show low health literacy and low adherence levels to healthy behaviors. Health education based on school has been shown to be effective in past studies, but separate health education curricula are rarely used in Chinese schools. This study evaluates the effects of a school-based health education curriculum on students’ knowledge, attitudes, and practices (or behaviors) (KAP) toward health. Methods: This study uses a cluster randomised controlled trail. Four primary schools were selected and randomly assigned into a control group (n=165) and intervention group (n=262). A one-semester Health Education Curriculum System (HECS) for senior grade pupils was implemented in the intervention group, and the control group was only given a 1-hour health education lecture. The evaluation was assessed from baseline, post-intervention, and 1 year after intervention by changes in students’ KAP on oral health, screen time, physical activity, and other lifestyle choices. Data were collected through questionnaires and analyzed with SPSS 20.0 using Chi-square and ACNOVA. Results: We found that the knowledge score increased immediately after intervention with a slight decrease in the follow-up stage, significant adjusted changes (95% CI) between the intervention and control groups in score was 5.83 (5.31 to 6.36) at post intervention and 2.59 (2.04 to 3.14) in 1 year follow-up. The prevalence of health-related behaviors were also enhanced to various extents from 13% to 38% between baseline and the post education stage in the intervention group (PConclusion:The current study shows that the health education curriculum can have positive effects on improving primary school students’ health KAP. Trial registration: The study registered with the Medical Ethics Board of Capital Medical University (Z2019SY063) on October 2019. At present, utilizing the insurance community is a common method to deal with investment risks along the Belt and Road; however, there is no clear method or mechanism to deal with the decision-making optimization and cost allocation of the insurance community participants. We propose a hybrid approach to solve this problem. First, we construct an underwriting decision optimization model for the insurance community using the multi-choice goal programming method, which generates the cost characteristic function based on a cooperative alliance. Second, we use the cooperative game method combined with the modified Shapley value method to take risk factors into consideration, which allows us to optimize the cost allocation among members of the insurance community. Finally, our simulation analysis results show that the multi-choice goal programming method can optimize the insurance community’s underwriting decisions. Specifically, the total underwriting cost is lower than the sum of the underwriting costs under the insurance company’s single-action strategy, and the total underwriting scale is as large as possible. Compared with the classical Shapley value method, the modified Shapley value method can better reflect differences in the underwriting risks of different regions, encouraging governments to take measures to reduce underwriting risks. To conclude, we propose some suggestions based on our research findings. The possible contributions of this paper are as follows: our research provides a hybrid optimization method based on multiple-choice objective programming and cooperative games to solve the cost allocation problem facing the insurance community, and it has some reference value for improving the cost-sharing system of the insurance community.
