Moisture content has an important influence on the stratification and screening of materials on the surface of screens, and materials with different external moisture concentrations present different screening characteristics. A vibrating flip-flow screen (VFFS) is a two-body vibration system with a high-vibrating-strength screen surface developed based on a circular vibrating screen (CVS), which offers significant advantages in terms of handling sticky fine particles. To better understand the difference in screening performance between a VFFS and CVS under different external moisture concentrations, it is necessary to conduct a comparative study on the distribution characteristics of materials on the screen’s surface. In this paper, a CVS dynamic model considering the stiffness force and position of the damping spring is proposed, and the influence of different arrangements of damping springs on the amplitude and angular displacement of the screen is analyzed. Under different external moisture concentrations, the screening percentages of the 3–1 mm sized fractions in sections I–IV of a VFFS and a CVS are greater than those of 1–0 mm sized fractions. The screening efficiencies of CVSs and VFFSs are 92.54% and 92.19%, respectively, at an external moisture content of 4.30%, so the screening effect of a low-vibration-intensity screen surface is better in the case of better material stratification. When the external moisture content of the material was increased from 4.30% to 8.19%, the screening efficiency of the VFFS and the CVS decreased by 33.85% and 41.32%, respectively. The screening efficiency of the CVS is more sensitive to the external moisture content of a material.

Ascorbic acid (AsA) is an antioxidant with significant functions in both plants and animals. Despite its importance, there has been limited research on the molecular basis of AsA production in the fruits of Capsicum annuum L. In this study, we used Illumina transcriptome sequencing (RNA-seq) technology to explore the candidate genes involved in AsA biosynthesis in Capsicum annuum L. A total of 8272 differentially expressed genes (DEGs) were identified by the comparative transcriptome analysis. Weighted gene co-expression network analysis identified two co-expressed modules related to the AsA content (purple and light-cyan modules), and eight interested DEGs related to AsA biosynthesis were selected according to gene annotations in the purple and light-cyan modules. Moreover, we found that the gene GDP-L-galactose phosphorylase (GGP) was related to AsA content, and silencing GGP led to a reduction in the AsA content in fruit. These results demonstrated that GGP is an important gene controlling AsA biosynthesis in the fruit of Capsicum annuum L. In addition, we developed capsanthin/capsorubin synthase as the reporter gene for visual analysis of gene function in mature fruit, enabling us to accurately select silenced tissues and analyze the results of silencing. The findings of this study provide the theoretical basis for future research to elucidate AsA biosynthesis in Capsicum annuum L.

The electrochemical carbon dioxide reduction reaction (CO2RR) has emerged as a promising approach to addressing global energy and environmental challenges. Alloys are of particular importance in these applications due to their unique chemical and physical properties. In this study, the possible mechanism of the C1 products from the electrochemical reduction of CO2 on four different surfaces of Pd3Au alloy bimetallic catalysts is predicted using the density functional theory. The differences in the number of d-band electrons and the charge distribution and morphology of the different surfaces result in differing catalytic activity and selectivity on the same surface. On different surfaces, Pd3Au alloy bimetallic catalysts have different potential limiting steps in CO2RR, resulting in differing selectivity. The Pd3Au (100) surface has a good selectivity for HER, indicating that the increase in the net charge on the surface of the alloy improves the selectivity for HER. The Pd3Au (211) surface, with a step structure, shows a good selectivity for methanol production from CO2RR. In addition, an electronic structure analysis shows that the selectivity of the reactions involved in the conversion of adsorbates is determined by the difference between the center of the d-band on the top of the catalyst, where the reactant and the product are located. The results of this study may provide some theoretical basis for designing and developing more efficient and selective CO2 reduction catalysts.

Coastal ecosystems provide important ecosystem services (ESs) and have been subject to conservation and restoration efforts in China for decades. However, ecosystem responses to coastal exploitation activity and their spatial determinants have not been sufficiently evaluated, which limits the efficacy of ecosystem restoration efforts. To fill these gaps, this study assessed the dominant change trends in ESs in southeastern China since the 1980s using an integrated biophysical model. Moreover, we explored the determinants of ESs, their spatial heterogeneity, and spillover effects via spatial econometrics and geospatial analysis approaches. The results indicate that coastal exploitation, particularly rapid urbanization and land reclamation, profoundly altered landscape composition and further affected ESs. In urbanization hotspots, rapid land use/cover conversion (i.e., wetland, woodland, and grassland losses) and increasingly intensive human activities have substantially lowered carbon stock, soil retention, and habitat quality services while increasing water yield and nitrogen export. Environmental, socioeconomic, and landscape variables were identified as important determinants of ES changes and exhibit significant spatial heterogeneity and spillover effects. Our findings indicate that such indicators are highly useful for ecosystem assessments, modeling, and forecasting for ES management and conservation efforts. The identified spatial determinants and their spillover effects demonstrate that regional landscape planning and ecosystem management must consider environmental, socioeconomic, and landscape indicators from a regionally integrated perspective and coordinate cross-border collaborations from neighboring areas to improve the efficacy of ecological projects. Our findings provide important references for scheme optimization and strategy adjustment for ES management, both in the study region and globally.

Photocatalytic oxidation (PCO) is a potential approach for air cleaning, especially when utilising titanium dioxide (TiO2). A MopFan is similar to a roller brush but is made of flexible fibres coated with TiO2. Unlike conventional filter/mesh UV systems, a MopFan provides a wide UV-TiO2 interaction surface area and airflow passage. This revolutionary technique can be low cost, efficient, and potentially effective against viruses, making it suitable for cleaning indoor air. It is easy to use but technically advanced. The system may be mounted on walls, floors, or placed on desktops. A photocatalytic air purification based on MopFan prototype was designed, constructed and tested. This study utilised copper wires (0.1 mm, 0.3 mm, 0.4 mm, and 0.5 mm), plastic fibres (0.5 mm and 1.1 mm), brass wire (0.4 mm), steel wire (0.38 mm), and organic “coco” fibres (0.4 mm). Copper wire (0.5 mm) and organic fibre (0.4 mm) were found to be effective against SARS-CoV-2, but brass (0.4 mm) and plastic (0.5 mm) fibres were found only partially effective. The purification performance was compared using MopFan with plastic (0.5 mm), brass (0.4 mm) and organic “coco” (0.4 mm) fibres but the other materials were rejected due to their poor qualities or difficulties in manufacturing. It was found that the system has a better effectiveness with organic fibres, around 21% of reduction consistently throughout the test. It was also found that by using the photocatalytic MopFan air cleaning system, the final concentration of pollutants in a room is determined by the rate and concentration of pollutant generation. Highlights Organic fibres do not require sanding prior to being coated with TiO2 solution. Copper and organic fibres are effective SARS-CoV-2 inhibitors. Organic fibres are the most efficient for air purification. The performance of purification is related to the concentration of pollutants.

ABSTRACT: Avian hepatitis E virus (HEV) is the major causative pathogen of the big liver and spleen disease, hepatitis-splenomegaly syndrome, and hepatic rupture hemorrhage syndrome. Until now, there are 6 different avian HEV genotypes that infect chickens have been reported worldwide. Epidemiologic investigations of the avian HEV demonstrated that avian HEV has been widely spread in China in recent years. In this study, an avian HEV named YT-aHEV was obtained from white-feathered broilers using LMH cells by virus isolation assay in Shandong province, China. The complete genome consists of 6656-nt excluding the poly(A) tail. The isolate was highly similar to the CaHEV strain and segregated into the same branch belonging to avian HEV genotype 3. Indirect immunofluorescence using capsid protein-specific polyclonal antibodies confirmed that YT-aHEV could establish productive infection and replicate stably in LMH cells. Furthermore, an in vivo avian HEV infection model was established successfully in specific pathogen-free chicken embryos by intravenous experiments. In the present study, we demonstrate an avian HEV infection associated with liver lesions of hemorrhage and swelling by LMH cells for the first time in a white-feather broiler flock in China. This research also provides a new diagnosis method for detection of avian HEV, which laid a foundation for the understanding of pathogenicity and molecular biology of this virus for further study.

Hepatocellular carcinoma (HCC) is one of the most common fatal malignancies and the main cause of cancer-related deaths. The multitarget tyrosine kinase inhibitors (TKIs) sorafenib and regorafenib are systemic therapeutic drugs approved for the treatment of HCC. Here, we found that sorafenib and regorafenib injured mitochondria by inducing mitochondrial Ca2+ (mtCa2+) overload and mitochondrial permeability transition pore (mPTP) opening, resulting in mitochondria-mediated cell death, which was alleviated by cyclosporin A (CsA), an inhibitor of mPTP. Meanwhile, mPTP opening caused PINK1 accumulation on damaged mitochondria, which recruited Parkin to mitochondria to induce mitophagy. Inhibition of autophagy by the lysosomal inhibitor chloroquine (CQ) or inhibition of mitochondrial fission by mdivi-1 aggravated sorafenib- and regorafenib-induced cell death. Moreover, knockdown of PINK1 also promotes sorafenib- and regorafenib-induced cell death. An in vivo study showed that sorafenib and regorafenib inhibited HepG2 cell growth more effectively in PINK1 knockdown cells than in shNTC cells in null mice. Thus, our data demonstrate that PINK1-Parkin-mediated mitophagy alleviates sorafenib and regorafenib antitumor effects in vitro and in vivo.

Bovine viral diarrhea virus (BVDV) is the causative agent of bovine viral diarrhea-mucosal disease (BVD-MD), an important viral disease in cattle that is responsible for extensive economic losses to the cattle industry worldwide. Currently, several underlying mechanisms involved in viral replication, pathogenesis, and evading host innate immunity of BVDV remain to be elucidated, particularly during the early stage of virus infection. To further explore the mechanisms of BVDV-host interactions, the transcriptomics and proteomics profiles of BVDV-infected MDBK cells were sequenced using RNA-seq and iTRAQ techniques, respectively, and followed by an integrative analysis. Compared with mock-infected MDBK cells, a total of 665 differentially expressed genes (DEGs) (391 down-regulated, 274 up-regulated) and 725 differentially expressed proteins (DEPs) (461 down-regulated, 264 up-regulated) were identified. Among these, several DEGs and DEPs were further verified using quantitative RT-PCR and western blot. Following gene ontology (GO) annotation and KEGG enrichment analysis, we determined that these DEGs and DEPs were significantly enriched in multiple important cellular signaling pathways including NOD-like receptor, Toll-like receptor, TNF, NF-κB, MAPK, cAMP, lysosome, protein processing in endoplasmic reticulum, lipid metabolism, and apoptosis signaling pathways. Significantly, the down-regulated DEGs and DEPs were predominantly associated with apoptosis-regulated elements, inflammatory factors, and antiviral elements that were involved in innate immunity, thus, indicating that BVDV could inhibit apoptosis and the expression of host antiviral genes to facilitate viral replication. Meanwhile, up-regulated DEGs and DEPs were primarily involved in metabolism and autophagy signaling pathways, indicating that BVDV could utilize the host metabolic resources and cell autophagy to promote replication. However, the potential mechanisms BVDV-host interactions required further experimental validation. Our data provide an overview of changes in transcriptomics and proteomics profiles of BVDV-infected MDBK cells, thus, providing an important basis for further exploring the mechanisms of BVDV-host interactions.

Chicken infectious anemia (CIA), caused by chicken anemia virus (CAV), is an immunosuppressive disease characterized by growth retardation, aplastic anemia, lymphoid depletion, and immunodepression in young chickens. In this study, 33 CAV strains were isolated from broilers in Shandong Province during 2020–2021. Phylogenetic analysis of full-length genome sequences showed that most CAV strains isolated in this study were scattered across different branches, but mainly clustered in two genotypes, indicating a certain regional characteristic. Analysis of VP1 protein identified several amino acid substitutions which were relevant with the virulence and virus spread efficiency. Interestingly, four putative DNA recombination events were detected in the genomes of novel isolated CAV strains. In summary, this study demonstrated a genomic diversity of CAV in broilers isolated in Shandong Province during 2020–2021, and provided information for the further study of CAV molecular epidemiology and viral evolution.

Abstract Background In Mainland China, advance directives (ADs) and end-of-life care for patients with tumours, especially patients with brain tumours who may have lost consciousness or the ability to speak at the early stage of their illness, have been poorly acknowledged. Thus, this study aimed to clarify the knowledge and preferences of ADs and end-of-life care in patients with brain tumours and to investigate predictors of patient preferences. Methods This was a population-based cross-sectional survey that was conducted via face-to-face interviews. Information on sociodemographic factors, brain tumour illness, knowledge and preferences of the advanced decisions and end-of-life care of the patients was collected. Results A total of 88.61% of participants had never heard of ADs, but 65.18% reported that they would like to make ADs. Knowledge of ADs, receiving surgical treatment or radiotherapy, being younger than 70 years old, being male, having educational qualifications of college or beyond, being childless, having medical insurance for nonworking or working urban residents and self-paying medical expenses were predictors of preference for making ADs. A total of 79.43% of participants wanted to discuss end-of-life arrangements with medical staff, and 63.29% of participants were willing to receive end-of-life care, even though it would not delay death. A total of 65.82% of patients with brain tumours wanted resuscitation, and as many as 45.45% of the patients thought that they did not need life support if they were in a persistent vegetative state. Brain primary tumours, being younger than 70 years old, male sex, educational qualification of junior middle school or below, having children, having new rural cooperative medical insurance and having medical expenses paid by children or spouses were predictors of choosing appropriate palliative care. Conclusions ADs and end-of-life care have been poorly acknowledged among patients with brain tumours in mainland China. Additional efforts should be encouraged amongst patients with primary brain tumours, those who are undergoing surgery and radiotherapy and those who have low socioeconomic status. A longitudinal and comprehensive study is encouraged to promote disease-specific ADs among Chinese patients with brain tumours.

Background and Aims: Besides prominent respiratory involvement, gastrointestinal manifestations are commonly reported in Coronavirus Disease 2019 (COVID-19) patients. We compared infection of ex vivo human intestinal tissues by SARS-CoV-2 and SARS-CoV with respect to their replication kinetics and immune activation profile. Methods: Human intestinal tissues were obtained from patients while undergoing surgical operations at Queen Mary Hospital, Hong Kong. Upon surgical removal, the tissues were immediately processed and infected with SARS-CoV-2 or SARS-CoV. Replication kinetics were determined with immunohistochemistry, qRT-PCR, and plaque assays. Immune activation in the infected intestinal tissues was assessed by detecting the gene expression of interferons and representative pro-inflammatory cytokines and chemokines. Results: SARS-CoV-2 could infect and productively replicate in the ex vivo human intestinal tissues with release of infectious virus particles, but not in ex vivo human liver and kidney tissues. Importantly, SARS-CoV-2 replicated less efficiently than SARS-CoV, induced less cytopathology in the human intestinal epithelium, and induced a more robust innate immune response including the activation of both type I and type III interferons, than SARS-CoV in human intestinal tissues. Conclusion: Using the ex vivo human intestinal tissues as a physiologically relevant model, our data indicated that SARS-CoV-2 could productively replicate in the human gut and suggested that the gastrointestinal tract might serve as an alternative route of virus dissemination. SARS-CoV-2 replicated less efficiently and induced less cytopathology than SARS-CoV in keeping with the clinical observations reported for COVID-19 and SARS, which might be the result of a more robust immune activation by SARS-CoV-2 than SARS-CoV in the human intestine.

Here, using lung epithelial cells and ex vivo tissue explants, the authors show that, in addition to ACE2, host heparan sulfate is directly involved in SARS-CoV-2 attachment and entry and provide data suggesting that host sialic acids may act as viral restriction factor in lung tissues.

Zika virus (ZIKV) is an emerging mosquito-borne flavivirus that poses significant threats to global public health. Macrophages and dendritic cells are both key sentinel cells in the host immune response and play critical roles in the pathogenesis of flavivirus infections. Recent studies showed that ZIKV could productively infect monocyte-derived dendritic cells (moDCs), but the role of macrophages in ZIKV infection remains incompletely understood. In this study, we first compared ZIKV infection in monocyte-derived macrophages (MDMs) and moDCs derived from the same donors. We demonstrated that while both MDMs and moDCs were susceptible to epidemic (Puerto Rico) and pre-epidemic (Uganda) strains of ZIKV, virus replication was largely restricted in MDMs but not in moDCs. ZIKV induced significant apoptosis in moDCs but not MDMs. The restricted virus replication in MDMs was not due to inefficient virus entry but was related to post-entry events in the viral replication cycle. In stark contrast with moDCs, ZIKV failed to inhibit STAT1 and STAT2 phosphorylation in MDMs. This resulted in the lack of efficient antagonism of the host type I interferon-mediated antiviral responses. Importantly, depletion of STAT2 but not STAT1 in MDMs significantly rescued the replication of ZIKV and the prototype flavivirus yellow fever virus. Overall, our findings revealed a differential interplay between macrophages and dendritic cells with ZIKV. While dendritic cells may be exploited by ZIKV to facilitate virus replication, macrophages restricted ZIKV infection.

It is critical that machine learning (ML) model predictions be trustworthy for high-throughput catalyst discovery approaches. Uncertainty quantification (UQ) methods allow estimation of the trustworthiness of an ML model, but these methods have not been well explored in the field of heterogeneous catalysis. Herein, we investigate different UQ methods applied to a crystal graph convolutional neural network to predict adsorption energies of molecules on alloys from the Open Catalyst 2020 dataset, the largest existing heterogeneous catalyst dataset. We apply three UQ methods to the adsorption energy predictions, namely k -fold ensembling, Monte Carlo dropout, and evidential regression. The effectiveness of each UQ method is assessed based on accuracy, sharpness, dispersion, calibration, and tightness. Evidential regression is demonstrated to be a powerful approach for rapidly obtaining tunable, competitively trustworthy UQ estimates for heterogeneous catalysis applications when using neural networks. Recalibration of model uncertainties is shown to be essential in practical screening applications of catalysts using uncertainties.

Electrocatalytic reduction of carbon dioxide to produce usable products and fuels such as alkanes, alkenes, and alcohols, is a very promising strategy. Recent experiments have witnessed great advances in precisely controlling the synthesis of single atom alloys (SAAs), which exhibit unique catalytic properties different from alloys and nanoparticles. However, only certain precious metals, such as Pd or Au, can achieve this transformation. Here, the density functional theory (DFT) calculations were performed to show that Zn-based SAAs are promising electrocatalysts for the reduction of CO2 to C1 hydrocarbons. We assume that CO2 reduction in Zn-based SAAs follows a two-step continuous reaction: first Zn reduces CO2 to CO, and then newly generated CO is captured by M and further reduced to C1 products such as methane or methanol. This work screens seven stable alloys from 16 SAAs (M = Fe, Ru, Os, Co, Rh, Ir, Ni, Pd, Pt, Cu, Ag, Au, V, Mo, Ti, Cr). Among them, Pd@Zn (101) and Cu@Zn (101) are promising catalysts for CO2 reduction. The reaction mechanisms of these two SAAs are discussed in detail. Both of them convert CO2 into methane via the same pathway. They are reduced by the pathway: *CO2 → *COOH → *CO + H2O; *CO → *CHO → *CH2O → *CH3O → *O + CH4 → *OH + CH4 → H2O + CH4. However, their potential determination steps are different, i.e., *CO2 → *COOH (ΔG = 0.70 eV) for Cu@Zn (101) and *CO → *CHO (ΔG = 0.72 eV) for Pd@Zn, respectively. This suggests that Zn-based SAAs can reduce CO2 to methane with a small overpotential. The solvation effect is simulated by the implicit solvation model, and it is found that H2O is beneficial to CO2 reduction. These computational results show an effective monatomic material to form hydrocarbons, which can stimulate experimental efforts to explore the use of SAAs to catalyze CO2 electrochemical reduction to hydrocarbons.

Avian hepatitis E virus (HEV) is the main etiologic pathogen of chicken big liver and spleen disease which is widely prevalent in China in recent years. However, due to the lack of a highly effective culture system in vitro, a genetically engineered subunit vaccine is the main direction of vaccine development. In this study, ORF3 genes of VaHEV strain from laying hens and YT-aHEV strain from broilers were amplified, respectively, and ORF3 protein was successfully expressed by Escherichia coli prokaryotic expression system. The serum samples were collected periodically to detect avian HEV antibodies by indirect immunofluorescence after specific pathogen free chickens immunized with the two proteins and their mixed proteins, the results showed that all serum samples were positive for antibodies to avian HEV. The antibody-positive chickens were artificially challenged with the cell-adapted strain YT-aHEV strain. The chickens from the immunized control group were infected successfully; no fecal detoxification was detected in the immunized group. In this study, two representative strains of ORF3 subunit vaccines of laying hens and broilers were prepared by prokaryotic expression, the immune effects of different proteins of these were evaluated through immunization and challenge studies in vivo, which provided a new technical possibility for prevention and control of avian HEV.
