207. Mesenchymal Stem Cell-Derived Exosomes are Effective for Radiation Enteritis and Essential for the Proliferation and Differentiation of Lgr5+Intestinal Epithelial Stem Cells by Regulating Mir-195/Akt/β-Catenin Pathway

Abstract

Background:: Radiation enteritis (RE) is a common complication of abdominal or pelvic radiotherapy, which when severe, could be life-threatening. Currently, there are no effective treatments. Studies have shown that mesenchymal stem cells (MSCs)-derived exosomes (MSC-exos) exhibit promising therapeutic effects in inflammatory diseases. However, the specific role of MSC-exos in RE and the regulatory mechanisms remain elusive. Methods:: In vivoassay was carried out by injecting MSC-exos into the total abdominal irradiation (TAI)-induced RE mouse model. Forin vitroassay, Lgr5-positive intestinal epithelial stem cells (Lgr5+IESC) were extracted from mice, followed by irradiation along with MSC-exos treatment. HE staining was performed to measure histopathological changes. mRNA expression of inflammatory factors TNF-α and IL-6 and stem cell markers LGR5, and OCT4 were quantified by RT-qPCR. EdU and TUNEL staining was performed to estimate cell proliferation and apoptosis. MiR-195 expression in TAI mice and radiation-induced Lgr5+IESC was tested. Results:: We found that the injection of MSC-exos inhibited inflammatory reaction, increased stem cell marker expression, and maintained intestinal epithelial integrity in TAI mice. Furthermore, MSC-exos treatment increased the proliferation and simultaneously suppressed apoptosis in radiation-stimulated Lgr5+IESC. MiR-195 expression increased by radiation exposure was decreased by MSC-exos therapy. MiR-195 overexpression facilitated the progress of RE by counteracting the effect of MSC-exos. Mechanistically, the Akt and Wnt/β-catenin pathways inhibited by MSC-exos were activated by miR-195 upregulation. Conclusion:: MSC-Exos are effective in treating RE and are essential for the proliferation and differentiation of Lgr5+IESCs. Moreover, MSC-exos mediates its function by regulating miR-195 Akt β-catenin pathways.

Abstract

The abundant extracellular matrix (ECM) in solid tumors causes limited drug penetration and hypoxia‐mediated chemoresistance, which lead to poor chemotherapy outcomes. To solve this problem, a biomimetic metal–organic framework nanodrug (ZIF‐8‐DOX‐LY‐RM) is developed with red blood cell membrane (RM) camouflage and encapsulation of type 1 transforming growth factor β receptor (TGFBR1) inhibitor and doxorubicin (DOX) for efficient chemotherapy. Based on the biomimetic properties of the erythrocyte membrane, ZIF‐8‐DOX‐LY‐RM can effectively accumulate in tumor tissues with immune escape and prolonged blood circulation. Then, the enriched nanodrug ZIF‐8‐DOX‐LY‐RM releases the TGFBR1 to remove collagen, subsequently leading to enhanced nanodrug penetration and increased oxygen supply. The abundant oxygen supply then relieves hypoxia‐mediated chemoresistance of DOX through increased cellular uptake and elevated reactive oxygen species production. The in vivo studies demonstrate the outstanding performance of ZIF‐8‐DOX‐LY‐RM nanoparticles in chemotherapy of cancer. This work presents an ECM normalization strategy for the synergistic collagen depletion and hypoxia alleviation and opens a promising avenue for effective chemotherapy of solid tumors. Abstract

High stability and extended circulation time in vivo are quite favorable for practical biomedical applications of nanomaterials, because they greatly facilitate the preferential tumor accumulation of nanomaterials, resulting in enhanced signal fidelity for imaging and improved therapeutic effect for treatment. Although many surface modification approaches have been employed to improve the stability and circulating behavior of nanomaterials, it still remains challenging in acquiring stable and long-lasting nanomaterials for in vivo bioimaging and therapy, especially for nanoscale metal-organic frameworks (NMOFs) due to their intrinsic instability in physiological conditions. Herein, a facile, one-step strategy is reported to encapsulate the zirconium (Zr)-based NMOF UiO-66 within 1,2-dioleoyl-sn-glycero-3-phosphate (DOPA) lipid bilayer (DOPA-LB). Contrary to UiO-66 NMOFs functionalized with polyethylene glycol, the obtained UiO-66@DOPA-LB presents significantly enhanced stability and impressive blood circulation time, allowing a higher accumulation of UiO-66@DOPA-LB in the tumor tissue. Benefited from these meritorious features, UiO-66@DOPA-LB labeled with near-infrared dye, IRDye 800CW, can not only achieve highly sensitive imaging of breast cancer tumor (5 mm), but also exhibits superior capability for early tumor (1-2 mm) detection. This study enriches the surface modification approach of NMOFs, and is of great importance for practical application of NMOFs in biomedical areas.

Abstract

Understanding and controlling the growth morphology of two-dimensional crystals of transition metal dichalcogenides (TMDs) is essential in developing high-quality crystalline material for spintronics, valleytronics, electronics, and optics. Here we report our studies on the evolution of crystallite morphology of MoS2observed in chemical vapor deposition. It is shown that as time goes on, the growth morphology of MoS2flakes undergoes a transition from a triangle to a star shape, then back to a triangle, and finally to a bulgy irregular morphology. By tuning the temperature of the element sources, the atomic ratio of S and Mo on the growing interface can be adjusted. The variation of the S/Mo ratio affects the edge diffusion length and nucleation behavior on the edge of crystallite, leading to different growth morphologies. Our observations provide clues on how to engineer the morphology and control the quality of TMD crystalline sheets.

250. The promoting effect of health education on medical treatment seeking behavior in the context of healthy China: based on the comparison of migrants and registered local residents

Abstract

The migrants are the important participant and contributor in China's economic and social construction, but they still have many restrictions on their access to health services. This paper analyzes the differences between the migrants and the registered local residents in looking for medical treatment and explores the relationship between health education and medical treatment seeking behavior, so as to find an appropriate way to improve the health level of the migrants. By using the comparative survey data of the floating population and registered population in 2017 and establishing the binary Logit model, this paper finds that the health education has a promoting effect on medical treatment seeking behaviors. In terms of the influence mechanism of health education on medical treatment seeking behavior, the mediating effect of social participation factors is stronger in the migrants’ group because making new friends and learning about local public health resources through health education is of great significance to the migrants. It is proposed to expand the coverage of health education among migrant groups and improve the accessibility of health education. At the same time, public medical and health services should be used as a window to guide the migrants to seek medical treatment in an orderly manner while assisting their social integration.
