Abstract

Previous studies on erythropoiesis revealed that micro RNAs (mi RNAs) play a critical role in erythroid differentiation. Given the abundance of identified mi RNAs and the limited understanding of erythroid mi RNAs, additional examination is required. Here, two sets of erythroid differentiation mi RNome data were analysed to screen for novel erythroid-inhibiting mi RNAs. MIR200A was selected based on its pattern of downregulated expression in the mi RNome datasets during induction of erythroid differentiation. Overexpression of MIR200A in K562 and TF-1 cells confirmed its inhibitory role in erythroid differentiation. Further in vivo study indicated that overexpression of mir200a inhibited primitive erythropoiesis of zebrafish. Transcriptome analyses after MIR200A overexpression in TF-1 cells indicated a significant role in regulating erythroid function and revealed potential regulation networks. Additionally, bioinformatics and experimental analyses confirmed that PDCD4 (programmed cell death 4) and THRB (thyroid hormone receptor, beta) are both targets of MIR200A-3p. Gain- and loss-of-function studies of PDCD4 and THRB revealed that the two targets were capable of promoting erythroid gene expression. 226. Aqueous Solution Growth of Millimeter-Sized Nongreen-Luminescent Wide Bandgap Cs4PbBr6Bulk Crystal

Abstract

Millimeter-sized Cs4PbBr6bulk single crystal that has no green luminescence was successfully grown from concentrated CsBr aqueous solution for the first time. Absorption spectrum indicates an optical absorption starting from 3.6 eV and a localized absorption in the range of 3.8–4.2 eV. Photoluminescence spectrum clearly shows that the crystal has no green emission. Along with the calculated electronic band structure, it is concluded that Cs4PbBr6has a wide bandgap of 3.6 eV. Through vacuum annealing treatment, the green luminescence of the original nongreen-luminescent Cs4PbBr6crystal was successfully activated, which is possibly due to the formation of CsPbBr3. This work gives the method to obtain millimeter-sized Cs4PbBr6crystals from aqueous solution and suggests a route to activate its green luminescence. The results are significant for the deep understanding of the intrinsic properties and exploring the potential applications of wide bandgap Cs4PbBr6.

227. High-Performance Graphene/β-Ga2O3Heterojunction Deep-Ultraviolet Photodetector with Hot-Electron Excited Carrier Multiplication

Abstract

Solar-blind ultraviolet (SBUV) detection has important applications in wireless secure communication, early warning, and so forth. However, the desired key device for SBUV detection and high-sensitivity and low-noise “sandwich” photodetector with large detective area is difficult to be fabricated because it is usually hard for traditional wide band gap semiconductors to boast both high conductivity and high SBUV transparency. Here, we proposed to use graphene as the transparent conductive layer to form graphene/β-Ga2O3heterojunction. With the help of large-area graphene and hot carrier multiplication, a SBUV photodetector with large detective area, low dark current, and high sensitivity was successfully assembled. Its photoresponsivity is 1–3 orders of magnitude higher than that of the conventional SBUV photodetectors, and its response speed can rival the best device ever reported.

Abstract

The neuronal Cdk5 activator p35 is involved in a multitude of neuronal activities, including cytoskeletal organization. We show here that p35 directly interacts with filamentous actin (F-actin) but not with monomeric actin (G-actin). Through binding, p35 induces the formation of actin bundles and stabilizes F-actin against dilution-induced depolymerization. p35 forms intermolecular self-associations, suggesting that p35 cross-links actin filaments into bundles via its intermolecular self-association. p35 dimerization and association with F-actin occur at the N-terminal region that is absent in the calpain-cleaved product p25, indicating that such p35 properties are lost by its truncation induced under neurotoxic conditions. Using p35 phosphorylated by Cdk5 and a mutational approach, we demonstrate that the phosphorylation of p35 promotes its homodimerization and p35-induced formation of F-actin bundles. In addition, the phosphorylation regulates p35 distribution to microtubule and actin cytoskeletons. Together, these observations define a novel function for p35 in cytoskeletal regulation.

Abstract

This study proposes a novel chaotic cuckoo search (CCS) optimization method by incorporating chaotic theory into cuckoo search (CS) algorithm. In CCS, chaos characteristics are combined with the CS with the intention of further enhancing its performance. Further, the elitism scheme is incorporated into CCS to preserve the best cuckoos. In CCS method, 12 chaotic maps are applied to tune the step size of the cuckoos used in the original CS method. Twenty-seven benchmark functions and an engineering case are utilized to investigate the efficiency of CCS. The results clearly demonstrate that the performance of CCS together with a suitable chaotic map is comparable as well as superior to that of the CS and other metaheuristic algorithms. Abstract

Studies on coding genes, miRNAs, and lncRNAs during erythroid development have been performed in recent years. However, analysis focusing on the integration of the three RNA types has yet to be done. In the present study, we compared the dynamics of coding genes, miRNA, and lncRNA expression profiles. To explore dynamic changes in erythropoiesis and potential mechanisms that control these changes in the transcriptome level, we took advantage of high throughput sequencing technologies to obtain transcriptome data from cord blood hematopoietic stem cells and the following four erythroid differentiation stages, as well as from mature red blood cells. Results indicated that lncRNAs were promising cell marker candidates for erythroid differentiation. Clustering analysis classified the differentially expressed genes into four subtypes that corresponded to dynamic changes during stemness maintenance, mid-differentiation, and maturation. Integrated analysis revealed that noncoding RNAs potentially participated in controlling blood cell maturation, and especially associated with heme metabolism and responses to oxygen species and DNA damage. These regulatory interactions were displayed in a comprehensive network, thereby inferring correlations between RNAs and their associated functions. These data provided a substantial resource for the study of normal erythropoiesis, which will permit further investigation and understanding of erythroid development and acquired erythroid disorders. Abstract

The mode-specific dynamics for the dissociative chemisorption of H2O on Cu(111) is first investigated by seven-dimensional quantum dynamics calculations, based on an accurately fitted potential energy surface (PES) recently developed by neural network fitting to DFT energy points. It is indicated that excitations in all three vibrational modes have a significant impact on reactivity, which are more efficacious than increasing the translational energy in promoting the reaction, with the largest enhancement for the excitation in the asymmetric stretching mode. There is large discrepancy between the six-dimensional reactivities with fixed azimuthal angles and seven-dimensional results, revealing that the 6D “flat surface” model cannot accurately characterize the reaction dynamics. The azimuthal angle-averaging approach is validated for vibrational excited states of the reactant, where the 7D mode-specific probability can be well reproduced by averaging the 6D azimuthal angle-fixed probabilities over 18 angles.
