Breast cancer (BC) is a very common cancer among women and one of the primary causes of death in women worldwide. Because BC has different molecular subtypes, the challenges associated with targeted therapy have increased significantly, and the identification of new therapeutic targets has become increasingly urgent. Blocking apoptosis and inhibiting cell death are important characteristics of malignant tumours, including BC. Under adverse conditions, including exposure to antitumour therapy, inhibition of cell death programmes can promote cancerous transformation and the survival of cancer cells. Therefore, inducing cell death in cancer cells is fundamentally important and provides new opportunities for potential therapeutic interventions. Lytic forms of cell death, primarily pyroptosis, necroptosis and ferroptosis, are different from apoptosis owing to their characteristic lysis, that is, the production of cellular components, to guide beneficial immune responses, and the application of lytic cell death (LCD) in the field of tumour therapy has attracted considerable interest from researchers. The latest clinical research results confirm that lytic death signalling cascades involve the BC cell immune response and resistance to therapies used in clinical practice. In this review, we discuss the current knowledge regarding the various forms of LCD, placing a special emphasis on signalling pathways and their implications in BC, which may facilitate the development of novel and optimal strategies for the clinical treatment of BC. Previous studies have pointed out that adding a single protrusion on the superhydrophobic surface can effectively reduce the contact time of droplets, which is of practical importance in applications like anti-icing, self-cleaning, and anti-frost. However, the droplet impact dynamics and the mechanism for contact time reduction are still far from completely understood. Therefore, in this paper, via a three-dimensional pseudopotential lattice Boltzmann model coupled with a modified curved boundary scheme (which satisfies mass conservation), the droplet impact dynamics is simulated in a wide range of Weber numbers (0 < We ≤ 67.7), protrusion size (10 ≤ W ≤ 30), and protrusion shape (triangle, square, and circle), with particular interest in understanding their effects on the contact time. We demonstrate that the variation of contact time depends on the bouncing type as well as the retraction way. Among three possible bouncing types observed in the simulation, including non-break bouncing, two droplets bouncing, and three droplets bouncing, two droplets bouncing has less contact time and could be facilitated by increasing We and using triangle protrusion. However, with increasing of W or using square protrusion, the bouncing type may change from two droplets bouncing to three droplets bouncing, and the deformation of fragments may become definitely different, leading to a long contact time. In addition, a transition from twice-retraction to once-retraction can be obtained by increasing We or making the protrusion shape sharp. When the fragments resulting from droplet splitting only retract once on the bottom wall, the contact time can be effectively reduced. [ABSTRACT FROM AUTHOR]

Owing to characteristics of single-step fabrication and rapid prototyping, 3D printing have potential applications in processing piezoelectric motors with complex structures. In this paper, a 3D printed resin-based bimodal piezoelectric motor is proposed. Four slots of the motor are designed to reduce the influence of large damping caused by polymer's viscoelastic and enhance mechanical properties. Considering the viscoelasticity and internal structure of the stator, a dynamic model based on the proposed motor is developed. Furthermore, parameters of these slots are optimized using the Taguchi method. Prototypes are fabricated and experimentally investigated. The results demonstrate that the first longitudinal mode and the second bending mode are 17 765 Hz and 18 006 Hz respectively, which are consistent with the analytical model results. Under the voltage of 300 Vpp, the maximum no-load speed and maximum driving load of the slotted motor are 200 r min−1 and 20 g with a speed of 6 r min−1, respectively. Compared with the motor without slots, the speed and load capacity are increased by 25% and 33%. Therefore, the motor with slots exhibits better mechanical output performance. [ABSTRACT FROM AUTHOR]

This research seeks to contribute to leadership theory by proposing an integrative framework to reveal how servant leadership and employee cognitive factors foster the conditions that promote employee innovative behavior. Building on social cognitive theory, we hypothesize that servant leadership improves employee innovative behavior through inspiring employee knowledge‐sharing. We predict that employee organizational identification strengthens the impact of servant leadership on knowledge‐sharing, and that employee learning goal orientation amplifies the relationship between employee knowledge‐sharing and innovative behavior. We also propose that the positive indirect effect of servant leadership on employee innovative behavior via knowledge‐sharing is strongest when both moderators are at a high level. Using data collected from 215 supervisor‐employee dyads from three private high‐tech firms in China and a multisource and multiphase research design, we find support for our proposed model. Theoretical and managerial implications for leadership and management practices are also discussed. Manufacturers face numerous product quality accidents and thus require an effective and general approach to rank the occurred quality accidents. Therefore, a universal risk-based quality accident ranking method emphasising the importance of the accident formation mechanism is proposed in this paper to rank, analyse, and prevent occurred quality accidents comprehensively. First, the risk formation mechanism of quality accident is proposed emphatically using two typical failure mechanisms and Axiomatic domain mapping, and the structure of big quality accident data is presented on the basis of big product lifecycle data in order to provide data support for the subsequent risk analysis. Second, the accident failure cost, probability, and detectivity are adopted as the critical risk factors, which are computed using the failure cost model, extended fault tree and Axiomatic relevance tree respectively. Third, a modified risk importance index considering the relative weight of risk factors is presented to measure and rank the risk priority of quality accidents. Finally, a case study on a computer control panel accident risk analysis is conducted to verify the effectiveness of the proposed approach. Purpose: Diabetic nephropathy (DN) is one of the most serious complications of diabetes mellitus and one of the most important causes of end-stage renal disease, but its pathogenesis has not been elucidated so far, and there is no effective treatment. Methods: DN models of rats and MPC-5 cells were established with streptozotocin (STZ) and high glucose (HG) in vivo and in vitro, respectively. Cell markers desmin and nephrin in foot kidney tissue were detected by Western blot. CCNG1 level in vitro was analyzed by Western blot and immunohistochemistry. CCK-8 assay and flow cytometry were conducted to analyze the effect of CCNG1 on HG-treated MPC-5 cells. Apoptosis-related proteins (Bcl-2, Bax and p53), CCNG1, and MDM2 were determined by RT-qPCR and Western blot. Results: The level of nephrin was decreased, while desmin was increased in STZ-induced DN rats and CCNG1 level was also enhanced by STZ. In vitro experiments indicated that MPC-5 cell viability was inhibited and apoptosis was induced by HG and we also found that CCNG1 expression was up-regulated by HG and negatively correlated with MDM2 level. The effects of HG on MPC-5 cell viability, apoptosis, and cell cycle were reversed by silencing CCNG1, but further deteriorated by overexpression of CCNG1. Furthermore, overexpression of MDM2 inhibited HG-induced MPC-5 cell injury and CCNG1 expression. Conclusions: These findings revealed that down-regulation of CCNG1 has protection effects in DN that is mechanistically linked to MDM2-p53 pathways. [ABSTRACT FROM AUTHOR]

Salmonella is an important zoonotic pathogen and is a major cause of gastrointestinal diseases worldwide. The current serious problem of antibiotic abuse has prompted the search for new substitutes for antibiotics. JH-3 is a small antimicrobial peptide with broad-spectrum bactericidal activity. In this study, we showed that JH-3 has good bactericidal activity towards the clinical isolate Salmonella enterica serovar Typhimurium strain CVCC541. The minimum inhibitory concentration (MIC) of JH-3 against this bacterium was determined to be 100 μg/mL, which could decrease the number of CVCC541 cells by 1000-fold in vitro within 5 h. The transmission electron microscopy (TEM) results showed that JH-3 can damage the cell wall and membrane of CVCC541, leading to the leakage of cell contents and subsequent cell death. To measure the bactericidal activity of CVCC541-infected mice were treated intraperitoneally 40 or 10 mg/kg JH-3 at 2 h or 3 days postinfection. Our results showed that treatment with 40 mg/kg JH-3 at 2 h postinfection had the best therapeutic effect and could significantly protect mice from a lethal dose of CVCC541. Furthermore, the clinical symptoms, bacterial burden in blood and organs, and intestinal pathological changes were all decreased and were close to normal. This study examined the therapeutic effect of the antimicrobial peptide JH-3 against S. enterica CVCC541 infection for the first time and determined the therapeutic effect of different JH-3 doses and treatment times, laying the foundation for studies of new antimicrobial agents.
