Background Drought is the most significant factor limiting maize production, given that maize is a crop with a high water demand. Therefore, studies investigating the mechanisms underlying the drought tolerance of maize are of great importance. There are no studies comparing drought tolerance among economically important subspecies of maize. This study aimed to reveal the differences between the physio-biochemical, enzymatic, and molecular mechanisms of drought tolerance in dent (Zea mays indentata), popcorn (Zea mays everta), and sugar (Zea mays saccharata) maize under control (no-stress), moderate, and severe drought stress. Methods Three distinct irrigation regimes were employed to assess the impact of varying levels of drought stress on maize plants at the V14 growth stage. These included normal irrigation (80% field capacity), moderate drought (50% field capacity), and severe drought (30% field capacity). All plants were grown under controlled conditions. The following parameters were analyzed: leaf relative water content (RWC), loss of turgidity (LOT), proline (PRO) and soluble protein (SPR) contents, membrane durability index (MDI), malondialdehyde (MDA), and hydrogen peroxide (H2O2) content, the antioxidant enzyme activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), and catalase (CAT). Additionally, the expression of heat shock proteins (HSPs) was examined at the transcriptional and translational levels. Results The effects of severe drought were more pronounced in sugar maize, which had a relatively high loss of RWC and turgor, membrane damage, enzyme activities, and HSP90 gene expression. Dent maize, which is capable of maintaining its RWC and turgor in both moderate and severe droughts, and employs its defense mechanism effectively by maintaining antioxidant enzyme activities at a certain level despite less MDA and H2O2 accumulation, exhibited relatively high drought tolerance. Despite the high levels of MDA and H2O2 in popcorn maize, the up-regulation of antioxidant enzyme activities and HSP70 gene and protein expression indicated that the drought coping mechanism is activated. In particular, the positive correlation of HSP70 with PRO and HSP90 with enzyme activities is a significant result for studies examining the relationships between HSPs and other stress response systems. The discrepancies between the transcriptional and translational findings provide an opportunity for more comprehensive investigations into the role of HSPs in stress conditions.

Scylla paramamosain is an important mariculture animal. In the present study, we examined the physiological and biochemical characteristics of juvenile S. paramamosain under hypoosmotic stress and further verified the function of ABCC1, a key transport protein in gill tissue. The results showed that Na+/K+-ATPase, Ca2+-ATPase, and Na+/K+/2Cl- cotransporter in gill tissues increased first and then decreased. In addition, SOD and CAT activities also began to gradually increase, protecting gill tissues from oxidative damage. The hemolymph osmolarity decreased rapidly from 792 mOsm/kg to 560 mOsm/kg within 6 h, and then rose to 626.25 mOsm/kg at 48 h, and then remained stable. At the same time, Na+, Cl-, K+, and Ca2+ concentrations were decreased to varying degrees. There were significant changes in free amino acids such as aspartic acid, glutamic acid, and asparagine in hemolymph in response to acute salt depletion. By injection of ABCC1 inhibitor/activator, it was found that while ABCC1 transporter activity in the MK-571 group showed a decreasing trend at 9 h, Na+/K+ -ATPase activity began to increase and reached a peak at 48 h. Compared with the control group, the levels of aspartic acid, glutamic acid, asparagine, glycine, arginine, alanine, lysine, and proline in the MK-571 group showed significant differences at 6 h; Significant differences in glutamate, asparagine, arginine, alanine, lysine, and proline appeared at 12 h in the Thiethylperazine group. These results suggested that the dynamic changes of gill ion transport enzymes, antioxidant enzymes, inorganic ions, and free amino acids in hemolymph occurred in S. paramamosain under hypoosmotic stress, and the ABCC1 transporter played an important role in this regulatory process.

In some bacteria, the stress adaptation response, a defence mechanism against low pH, can also induce a number of physiological and genetic resistance mechanisms that provide advantages for bacteria to resist other environmental factors. This phenomenon is called cross-protection, which can potentially have serious consequences for food safety. In some fermented, acidified foods, low pH can provide a favourable environment for the growth of bacteria. Bacteria can adapt to acidic environments and become able to survive various factors that occur during storage and processing such as salt, antibiotics, or technological effects.

The objective of this study was to investigate the function of the aquaporin 4 gene (LvAQP4), which had previously been characterized in Pacific white shrimp, Litopenaeus vannamei. To achieve this, double-stranded RNA (dsRNA) targeting LvAQP4 was synthesized and injected into shrimp, resulting in a significant reduction in the expression of LvAQP4. Subsequent to the evaluation of the impact of LvAQP4 transcriptional silencing on shrimp under salinity challenges, we observed that the downregulation of LvAQP4 expression in the gills resulted in the downregulation of aquaglyceroporin (LvAQP3) and unorthodox aquaporin (LvAQP11) genes, as well as ion transporter genes, such as LvNKAα, LvNKAβ, LvCAc, and LvVHA-a. Notably, the inhibition of LvAQP4 had a significant impact on the expression patterns of these osmoregulatory genes, particularly during periods of high salinity stress. Furthermore, there was a notable increase in the mortality rate of shrimp under high-salinity stress. Moreover, the suppression of LvAQP4 through RNA interference led to a significant delay in the molting rate of shrimp. Furthermore, it resulted in an increase in the expression of LvMIH and a significant reduction in key genes involved in the ecdysone signaling pathway, including LvEcR, LvRXR, and LvE75. These findings suggest that LvAQP4 may play a regulatory role in the molting process of shrimp by modulating MIH and the ecdysone signaling pathway. Moreover, this study lays the groundwork for future investigations into the functions of aquaporins in shrimp osmoregulation and growth, contributing to an enhanced understanding of these essential regulatory mechanisms.

Sepia pharaonis, commonly known as the pharaoh cuttlefish, is a valued cephalopod in aquaculture. However, its sensitivity to salinity fluctuations has constrained its cultivation, especially in regions with varying salinity and during rainy seasons. To address this, our study implemented a graded salinity reduction approach, enabling S. pharaonis to acclimate to an environment with a salinity of 20. Despite a reduction in growth parameters such as mantle length and body weight in the Low-Salinity Acclimation group (LSA), the survival rates were largely consistent with the control group. After the acclimation process, the LSA group demonstrated a strikingly high survival rate of nearly 100% in salinity ranges from 18 to 24, a span previously thought to be deadly for non-acclimated S. pharaonis. Our evaluations indicate that the serum osmotic pressure and ion concentration of S. pharaonis decline with decreasing seawater salinity. During this process, there were significant alterations in the Na+/K+-ATPase activity and the content of free amino acids, suggesting their crucial role in low-salt osmoregulation. Overall, this study not only highlights the potential of the graded salinity reduction method to enhance the adaptability of S. pharaonis to fluctuating salinities, but also underscores the potential of graded salinity reduction as an innovative approach to enhance the resilience of aquaculture species.

Salt stress poses a significant challenge to crop productivity, and understanding the genetic basis of salt tolerance is paramount for breeding resilient soybean varieties. In this study, a soybean natural population was evaluated for salt tolerance during the germination stage, focusing on key germination traits, including germination rate (GR), germination energy (GE), and germination index (GI). It was seen that under salt stress, obvious inhibitions were found on these traits, with GR, GE, andGI diminishing by 32% to 54% when compared to normal conditions. These traits displayed a coefficient of variation (31.81% to 50.6%) and a substantial generalized heritability (63.87% to 86.48%). Through GWAS, a total of 1841 significant singlenucleotide polymorphisms (SNPs)were identified to be associated with these traits, distributed across chromosome 2, 5, 6, and 20. Leveraging these significant association loci, 12 candidate genes were identified to be associated with essential functions in coordinating cellular responses, regulating osmotic stress, mitigating oxidative stress, clearing reactive oxygen species (ROS), and facilitating heavymetal ion transport - all of which are pivotal for plant development and stress tolerance. To validate the candidate genes, quantitative real-time polymerase chain reaction (qRT-PCR) analysis was conducted, revealing three highly expressed genes (Glyma.02G067700, Glyma.02G068900, and Glyma.02G070000) that play pivotal roles in plant growth, development, and osmoregulation. In addition, based on these SNPs related with salt tolerance, KASP (Kompetitive Allele-Specific PCR)markers were successfully designed to genotype soybean accessions. Tomato (Solanum lycopersicum L.) is an important vegetable crop that grows easily under controlled conditions, such as in greenhouses and hydroponics. To overcome freshwater scarcity, researchers are searching for alternatives to groundwater sources such as desalinated water (saline water) for irrigation. High salinity in irrigation water alters physiological functions and crop development, thereby reducing the yield. Best management practices and the use of grafted tomato plants on salt-tolerant rootstocks can alleviate salinity stress. The present study was conducted to address the impact of salinity stress on leaf transpiration (Tr), stomatal conductance (gs), photosynthesis (Pn), leaf chlorophyll content, proline content, and yield of hydroponically cultivated tomato plants. Saline (NaCl) water was used for the preparation of nutrient solution with three salinity levels, electrical conductivity (EC, dS m−1) of 2.5 (control), 6.0, and 9.5. Three commercial tomato cultivars (Valouro-RZ, Ghandora-F1, and Feisty-Red) were used. Both self-rooted plants and plants grafted onto Maxifort rootstocks were transplanted onto a perlite substrate. The recorded data revealed that all studied cultivars were critically affected by higher salinity (≈ 9.5 dS m-1) compared to low (≈ 2.5 dS m-1) and medium (≈ 6.0 dS m-1) salinity levels. The Variations in Tr, Pn, gs, chlorophyll content of leaf, and yield between medium and high salinity trials were reported at 3%, 5%, 9%, 5%, and 7.1%, respectively, whereas no significant differences were observed between low and medium salinities. However, at medium salinity levels, grafted plants performed better in photosynthesis than non-grafted plants. This is due to the accumulation of leaf proline, which maintains osmotic regulation and photosynthetic activity by preventing cell damage at medium salinities. Hence, this study confirmed the use of saline water for growing tomatoes under hydroponic conditions up to an EC of 6.0 dS m-1 including the EC of nutrient fertilizers. [ABSTRACT FROM AUTHOR]

Simple Summary: Aquaculture produces fish that are then marketed to the population, but the type of production used today generates stress in fish. Constant stress, which occurs through the hormone cortisol, negatively affects the seawater adaptation of juvenile fish, which is detrimental to the aquaculture process. For many years, it was thought that only cortisol influenced physiological responses in fish. However, in recent years, the cortisol intermediary 11-deoxycorticosterone (DOC) has been considered a complementary stress-related hormone in fish. Given the above, this work aims to unravel whether DOC is involved in the regulation of early physiological and transcriptional responses in fish gills. To ascertain this, juvenile rainbow trout were injected with DOC and/or pharmacological inhibitors of corticosteroid receptors. Then, the physiological parameters and RNA sequencing of the gills were analyzed. The experiment concluded that DOC is important during stress and that it is vital for the balance between minerals and water in fish. The results of this work will help us understand the other effects of DOC in fish, as well as help improve the monitoring of animal welfare in the aquaculture industry through the incorporation of novel and potential stress molecular biomarkers. In aquaculture, stress can negatively affect fish growth. For years, the cortisol hormone has been thought to play both glucocorticoid and mineralocorticoid functions. Nevertheless, recent research has suggested that 11-deoxycorticosterone (DOC) released during stress could contribute to cortisol actions, though this process is still misunderstood. Here, we evaluated the DOC effects on physiological and early transcriptional responses by RNA-seq. Juvenile rainbow trout were treated with DOC and/or glucocorticoids (mifepristone) or mineralocorticoid (eplerenone) receptor antagonists. Subsequently, plasma was collected, and cDNA libraries were generated from the gills of vehicle (control), DOC, mifepristone, mifepristone with DOC, eplerenone, and eplerenone with DOC groups. Calcium and phosphate levels in plasma were changed. Results revealed 914 differentially expressed transcripts (DETs) induced by DOC compared with control, mainly associated with sodium ion transmembrane transport, gluconeogenesis, negative regulation of transmembrane transport, and activation of innate immune response. DOC versus eplerenone with DOC comparison displayed 444 DETs related to cell-cell junction organization, canonical glycolysis, positive regulation of immune response, and potassium ion transport. Conversely, no DETs were detected in DOC versus mifepristone with DOC comparison. These data suggest that DOC has a relevant role in gill stress response and ion transport, which is differentially regulated by mineralocorticoid receptors. [ABSTRACT FROM AUTHOR]

Whiteflies (Bemisia tabaci sensu lato) have a wide host range and are globally important agricultural pests. In Sub-Saharan Africa, they vector viruses that cause two ongoing disease epidemics: cassava brown streak disease and cassava mosaic virus disease. These two diseases threaten food security for more than 800 million people in Sub-Saharan Africa. Efforts are ongoing to identify target genes for the development of novel management options against the whitefly populations that vector these devastating viral diseases affecting cassava production in Sub-Saharan Africa. This study aimed to identify genes that mediate osmoregulation and symbiosis functions within cassava whitefly gut and bacteriocytes and evaluate their potential as key gene targets for novel whitefly control strategies. The gene expression profiles of dissected guts, bacteriocytes and whole bodies were compared by RNAseq analysis to identify genes with significantly enriched expression in the gut and bacteriocytes. Phylogenetic analyses identified three candidate osmoregulation gene targets: two a-glucosidases, SUC 1 and SUC 2 with predicted function in sugar transformations that reduce osmotic pressure in the gut; and a water-specific aquaporin (AQP1) mediating water cycling from the distal to the proximal end of the gut. Expression of the genes in the gut was enriched 23.67-, 26.54- and 22.30-fold, respectively. Genome-wide metabolic reconstruction coupled with constraint-based modeling revealed four genes (argH, lysA, BCAT & dapB) within the bacteriocytes as potential targets for the management of cassava whiteflies. These genes were selected based on their role and essentiality within the different essential amino acid biosynthesis pathways. A demonstration of candidate osmoregulation and symbiosis gene targets in other species of the Bemisia tabaci species complex that are orthologs of the empirically validated osmoregulation genes highlights the latter as promising gene targets for the control of cassava whitefly pests by in planta RNA interference. Introduction: With climate change, frequent exposure of bioenergy and food crops, specifically soybean (Glycine max L.), to low-temperature episodes is a major obstacle in maintaining sustainable plant growth at early growth stages. Silicon (Si) is a quasi-essential nutrient that can help to improve stress tolerance; however, how Si and a combination of cold stress episodes influence plant growth, plant physiology, and microbiome diversity has yet to be fully discovered. Methods: The soybean plants were exposed to cold stress (8-10°C) with or without applying Si, and the different plant organs (shoot and root) and rhizospheric soil were subjected to microbiome analysis. The plant growth, physiology, and gene expression analysis of plant defenses during stress and Si were investigated. Results and discussion: We showed that cold stress significantly retarded soybean plants' growth and biomass,whereas, Si-treated plants showed ameliorated negative impacts on plant growth at early seedling stages. The beneficial effects of Si were also evident from significantly reduced antioxidant activities -- suggesting lower cold-induced oxidative stress. Interestingly, Si also downregulated critical genes of the abscisic acid pathway and osmotic regulation (9-cis-epoxy carotenoid dioxygenase and dehydration-responsive element binding protein) during cold stress. Si positively influenced alpha and beta diversities of bacterial and fungal microbiomes with or without cold stress. Results showed significant variation in microbiome composition in the rhizosphere (root and soil) and phyllosphere (shoot) in Si-treated plants with or without cold stress exposures. Amongmicrobiome phyla, Proteobacteria, Bacteroidota, and Ascomycota were significantly more abundant in Si treatments in cold stress than in control conditions. For the core microbiome, we identified 179 taxa, including 88 unique bacterial genera in which Edaphobacter, Haliangium, and Streptomyces were highly abundant. Enhanced extracellular enzyme activities in the cold and Si+cold treatments, specifically phosphatase and glucosidases, also reflected the microbiome abundance. In conclusion, this work elucidates cold-mediated changes in microbiome diversity and plant growth, including the positive impact Si can have on cold tolerance at early soybean growth stages -- a step toward understanding crop productivity and stress tolerance. Abundant microbial biofilms inhabit underwater freshwater springs of the Dead Sea. Unlike the harsh (i.e., over 35% total dissolved salts) yet stable environment of the basin, the flow rate of the springs changes with random amplitude and duration, resulting in drastic shifts in salinity, pH, and oxygen concentrations. This requires the organisms to continuously adapt to new environmental conditions. Osmotic regulation is energetically expensive; therefore, the response of the biofilm organisms to rapid and drastic changes in salinity is interesting. For this purpose, we studied the metagenome of an enrichment culture obtained from a green biofilm-covered rock positioned in a spring. We obtained metagenomeassembled genomes (MAGs) of Prosthecochloris sp. (Chlorobiales), Flexistipes sp. (Deferribacterales), Izemoplasma (Izemoplasmatales), Halomonas sp. (Oceanospirillales), and Halanaerobium (Halanaerobiales). The MAGs contain genes for both the energetically cheaper "salt-in" and more expensive "saltout" strategies. We suggest that the dynamic response of these bacteria utilizes both osmoregulation strategies, similar to halophilic archaea. We hypothesize that the frequent, abrupt, and variable-in-intensity shifts in salinity, typical of the Dead Sea spring system, select for microorganisms with scalable adaptation strategies. Seldom found in saltmarshes, Linum maritimum is a halophyte of great conservation interest in the eastern Iberian Peninsula. Although the species has been reported in different plant communities, there is no information on its range of salinity tolerance or mechanisms of response to environmental stress factors. In this study, L. maritimum plants were subjected to increasing salt concentrations in controlled conditions in a greenhouse. After six months of watering with salt solutions, only plants from the control, 50 mM and 100 mM NaCl treatment groups survived, but seeds were produced only in the first two. Significant differences were found between the plants from the various treatment groups in terms of their growth parameters, such as plant height, fresh weight, and the quantity of flowers and fruits. The main mechanism of salt tolerance is probably related to the species' ability to activate K+ uptake and transport to shoots to partly counteract the accumulation of toxic Na+ ions. A biochemical analysis showed significant increases in glycine betaine, flavonoids and total phenolic compounds, highlighting the importance of osmotic regulation and antioxidant compounds in the salt tolerance of Linum maritimum. These findings have implications for the conservation of the species, especially under changing climatic conditions that may lead to increased soil salinity in its Mediterranean distribution area. Fruit cracking affects both the yield and economic efficiency of citrus; however, the underlying mechanism remains unclear. Therefore, this study focused on resistant and susceptible cultivars to identify the mechanisms underlying fruit cracking. The results showed that in 'Mingrijian', pectin morphological transformation and hemicellulose and lignin degradation in the pericarp were important contributing factors. During the critical fruit-cracking period (115–150 days after flowering), the water-soluble pectin, protopectin, and lignin contents in the pericarp of 'Daya' presented inverse changes relative to those in 'Mingrijian', thus enhancing the mechanical properties and resistance of pericarp. From 115 to 150 days after flowering, the soluble sugar content in the pulp of 'Mingrijian' increased rapidly by 97.35%, aiding in pulp water absorption and expansion. Moreover, the soluble protein content in the pericarp of 'Mingrijian' exhibited a declining trend and was lower than that of 'Daya', thus affecting the overall metabolism. The superoxide dismutase (SOD) activity in the pericarp of 'Mingrijian' gradually decreased from 115 to 180 days after flowering, while the peroxidase (POD) activity remained at a low level, resulting in weaker antioxidant capacity and lower environmental resistance. Streptococcus agalactiae is among the few pathogens that have not developed resistance to ß-lactam antibiotics despite decades of clinical use. The molecular basis of this long-lasting susceptibility has not been investigated, and it is not known whether specific mechanisms constrain the emergence of resistance. In this study, we first report ß-lactam tolerance due to the inactivation of the c-di-AMP phosphodiesterase GdpP. Mechanistically, tolerance depends on antagonistic regulation by the repressor BusR, which is activated by c-di-AMP and negatively regulates ß-lactam susceptibility through the BusAB osmolyte transporter and the AmaP/Asp23/GlsB cell envelope stress complex. The BusR transcriptional response is synergistic with the simultaneous allosteric inhibition of potassium and osmolyte transporters by c-di-AMP, which individually contribute to low-level ß-lactam tolerance. Genome-wide transposon mutagenesis confirms the role of GdpP and highlights functional interactions between a lysozyme-like hydrolase, the KhpAB RNA chaperone and the protein S immunomodulator in the response of GBS to ß-lactam. Overall, we demonstrate that c-di-AMP acts as a turgor pressure rheostat, coordinating an integrated response at the transcriptional and post-translational levels to cell wall weakening caused by ß-lactam activity, and reveal additional mechanisms that could foster resistance. In this study, with two almond strains(Xianfeng 1, Taobadan) offspring leaves as test material, five temperature level(4(CK), 2, 0, -2, and -4 ℃ ) and 4 processing time(1, 2, 3, and 4 h) were set to explore during the low temperature treatment, membrane structure stability, change of peroxidase activity and osmotic material content of the two almond strains, in order to screen out the strains with better cold resistance, provide help for selcection of almond offspring strains. The results showed that under the same low-temperature stress treatment of -4 ℃, the relative conductivity, malondialdehyde content, activity of peroxidase(POD), superoxide dismutase(SOD), and polyphenols oxidase(PPO) of Xianfeng 1 offspring all decreased first and then increased with the extension of treatment time. The relative conductivity and malondialdehyde content of the Taobadan offspring showed an increasing trend, reaching the maximum values under the treatment of -4 ℃ and 4 h. The content of soluble protein, soluble sugar, and proline, the activity of SOD and phenylalanine ammonia lyase(PAL) of Taobadan offspring decreased first and then increased with the extension of treatment time. At the same time of 4h treatment, the proline content of Xianfeng 1 offspring showed a trend of overall decrease and the decreasing range was the highest under the treatment of -2 ℃ and 4 h. The content of propanondialdehyde, soluble sugar, and proline, the SOD activity of Taobadan offspring showed a trend of overall incrase and the increasing range of the proline content and the SOD activity was the highest under the treatment of -4 ℃ and 4 h. The cold resistance of Taobadan offspring was significantly better than that of Xianfeng 1. The Taobadan offspring could be tolerate to lower temperature(-4 ℃ ) and longer duration. Ceroid lipofuscinosis neuronal 5 (CLN5) and cathepsin D (CTSD) are soluble lysosomal enzymes that also localize extracellularly. In humans, homozygous mutations in CLN5 and CTSD cause CLN5 disease and CLN10 disease, respectively, which are two subtypes of neuronal ceroid lipofuscinosis (commonly known as Batten disease). The mechanisms regulating the intracellular trafficking of CLN5 and CTSD and their release from cells are not well understood. Here, we used the social amoeba Dictyostelium discoideum as a model system to examine the pathways and cellular components that regulate the intracellular trafficking and release of the D. discoideum homologs of human CLN5 (Cln5) and CTSD (CtsD). We show that both Cln5 and CtsD contain signal peptides for secretion that facilitate their release from cells. Like Cln5, extracellular CtsD is glycosylated. In addition, Cln5 release is regulated by the amount of extracellular CtsD. Autophagy induction promotes the release of Cln5, and to a lesser extent CtsD. Release of Cln5 requires the autophagy proteins Atg1, Atg5, and Atg9, as well as autophagosomal‐lysosomal fusion. Atg1 and Atg5 are required for the release of CtsD. Together, these data support a model where Cln5 and CtsD are actively released from cells via their signal peptides for secretion and pathways linked to autophagy. The release of Cln5 and CtsD from cells also requires microfilaments and the D. discoideum homologs of human AP‐3 complex mu subunit, the lysosomal‐trafficking regulator LYST, mucopilin‐1, and the Wiskott–Aldrich syndrome‐associated protein WASH, which all regulate lysosomal exocytosis in this model organism. These findings suggest that lysosomal exocytosis also facilitates the release of Cln5 and CtsD from cells. In addition, we report the roles of ABC transporters, microtubules, osmotic stress, and the putative D. discoideum homologs of human sortilin and cation‐independent mannose‐6‐phosphate receptor in regulating the intracellular/extracellular distribution of Cln5 and CtsD. The mitochondrial retrograde signaling (RTG) pathway of communication from mitochondria to the nucleus was first studied in yeast Saccharomyces cerevisiae. It rewires cellular metabolism according to the mitochondrial state by reprogramming nuclear gene expression in response to mitochondrial triggers. The main players involved in retrograde signaling are the Rtg1 and Rtg3 transcription factors, and a set of positive and negative regulators, including the Rtg2, Mks1, Lst8, and Bmh1/2 proteins. Retrograde regulation is integrated with other processes, including stress response, osmoregulation, and nutrient sensing through functional crosstalk with cellular pathways such as high osmolarity glycerol or target of rapamycin signaling. In this review, we summarize metabolic changes observed upon retrograde stimulation and analyze the progress made to uncover the mechanisms underlying the integration of regulatory circuits. Mitochondrial‐rich cells (MRCs) are one of the most significant canceled type of epithelial cells. Morphologically these cells are totally different from other epithelial cells. These cells primarily implicated in sea‐water and fresh‐water adaptation, and acid–base regulation. However, in this review paper, we explored some of the most intriguing biological and immune‐related functional developmental networks of MRCs. The main pinpoint, MRCs perform a dynamic osmoregulatory and immunological functional role in the gut and male reproductive system. The Na+/K+_ATPase (NKA) and Na+/K+/2Cl cotransporter (NKCC) are key acidifying proteins of MRCs for the ion‐transporting function for intestinal homeostasis and maintenance of acidifying the luminal microenvironment in the male reproductive system. Further more importantly, MRCs play a novel immunological role through the exocrine secretion of nano‐scale exosomes and multivesicular bodies (MVBs) pathway, which is very essential for sperm maturation, motility, acrosome reaction, and male sex hormones, and these an essential events to produce male gametes with optimal fertilizing ability. This effort is expected to promote the novel immunological role of MRCs, which might be essential for nano‐scale exosome secretion. Leaf characteristics can reflect the adaptation of trees to drought stress. However, the effect of leaf maturity on drought stress has been neglected, leading to uncertainty in inferring individual tree responses to drought from leaves. The allocation strategy of photosynthetic carbon between leaf organs (fully expanded young and old leaves) under drought stress remains unclear. Poplar is a diverse and widespread tree species in arid and semi‐arid regions. Here, three poplar genotypes (Populus cathayana, P. × euramericana 'Nanlin 895', and P. alba × P. tremula var. glandulosa) were selected and exposed to different watering regimes. The responses and carbon allocation strategies of leaves with different maturity to drought were investigated using a combination of leaf traits and 13C pulse labelling technique. The results showed that (1) fully expanded young leaves had better osmotic regulation and antioxidant capacity than aged leaves under drought stress. (2) Aged leaves acted as a carbon source during water deficit, where their photosynthetic products were transferred and supplied to upper young leaves to promote stronger photosynthesis in young leaves to acquire resources for tree growth. This study highlights that the effect of leaf maturity should be considered in the future when investigating the effects of drought on woody plants, especially for continuously growing tree species. Therefore, our study not only demonstrates the existence of leaf‐age‐dependent responses to drought in poplar but also provides new insights into carbon allocation at the leaf level. Aquatic environments are an unusual habitat for most arthropods. Nevertheless, many arthropod species that were once terrestrial dwelling have transitioned back to marine and freshwater environments, either as semiaquatic or, more rarely, as fully aquatic inhabitants. Transition to water from land is exceptional, and without respiratory modifications to allow for extended submergence and the associated hypoxic conditions, survival is limited. In this article, we review marine-associated species that have made this rare transition in a generally terrestrial group, spiders. We include several freshwater spider species for comparative purposes. Marine-associated spiders comprise less than 0.3% of spider species worldwide but are found in over 14% of all spider families. As we discuss, these spiders live in environments that, with tidal action, hydraulic forces, and saltwater, are more extreme than freshwater habitats, often requiring physiological and behavioral adaptations to survive. Spiders employ many methods to survive inundation from encroaching tides, such as air bubble respiration, airtight nests, hypoxic comas, and fleeing incoming tides. While airway protection is the primary survival strategy, further survival adaptations include saltwater-induced osmotic regulation, dietary composition, predator avoidance, reproduction, locomotory responses, and adaptation to extreme temperatures and hydrostatic pressures that challenge existence in marine environments. The rapid advances in available transcriptomic and genomic data and our understanding of the physiology and biochemistry of whitefly–plant interactions have allowed us to gain new and significant insights into the biology of whiteflies and their successful adaptation to host plants. In this review, we provide a comprehensive overview of the mechanisms that whiteflies have evolved to overcome the challenges of feeding on phloem sap. We also highlight the evolution and functions of gene families involved in host perception, evaluation, and manipulation; primary metabolism; and metabolite detoxification. We discuss the emerging themes in plant immunity to whiteflies, focusing on whitefly effectors and their sites of action in plant defense–signaling pathways. We conclude with a discussion of advances in the genetic manipulation of whiteflies and the potential that they hold for exploring the interactions between whiteflies and their host plants, as well as the development of novel strategies for the genetic control of whiteflies. Water is essential to life. Terrestrial insects lose water by evaporation from the body surface and respiratory surfaces, as well as in the excretory products, posing a challenge made more acute by their high surface-to-volume ratio. These losses must be kept to a minimum and be offset by water gained from other sources. By contrast, insects such as the blood-sucking bug Rhodnius prolixus consume up to 10 times their body weight in a single blood meal, necessitating rapid expulsion of excess water and ions. How do insects manage their ion and water budgets? A century of study has revealed a great deal about the organ systems that insects use to maintain their ion and water balance and their regulation. Traditionally, a taxonomically wide range of species were studied, whereas more recent research has focused on model organisms to leverage the power of the molecular genetic approach. Key advances in new technologies have become available for a wider range of species in the past decade. We document how these approaches have already begun to inform our understanding of the diversity and conservation of insect systemic osmoregulation. We advocate that these technologies be combined with traditional approaches to study a broader range of nonmodel species to gain a comprehensive overview of the mechanism underpinning systemic osmoregulation in the most species-rich group of animals on earth, the insects. Animals can respond to extreme climates by behaviourally avoiding it or by physiologically coping with it. We understand behavioural and physiological thermoregulation, but water balance has largely been neglected. Climate change includes both global warming and changes in precipitation regimes, so improving our understanding of organismal water balance is increasingly urgent. We assessed the hydric physiology of US federally endangered blunt-nosed leopard lizards (Gambelia sila) by measuring cutaneous evaporative water loss (CEWL), plasma osmolality and body condition. Measurements were taken throughout their active season, the short period of year when these lizards can be found aboveground. Compared to a more mesic species, G. sila had low CEWL which is potentially desert-adaptive, and high plasma osmolality that could be indicative of dehydration. We hypothesized that throughout the G. sila active season, as their habitat got hotter and drier, G. sila would become more dehydrated and watertight. Instead, CEWL and plasma osmolality showed minimal change for females and non-linear change for males, which we hypothesize is connected to sex-specific reproductive behaviours and changes in food availability. We also measured thermoregulation and microhabitat use, expecting that more dehydrated lizards would have lower body temperature, poorer thermoregulatory accuracy and spend less time aboveground. However, we found no effect of CEWL, plasma osmolality or body condition on these thermal and behavioural metrics. Finally, G. sila spends considerable time belowground in burrows, and burrows may serve not only as essential thermal refugia but also hydric refugia. [ABSTRACT FROM AUTHOR]

The impact of ammonium (NH4+) stress on plant growth varies across species and cultivars, necessitating an in-depth exploration of the underlying response mechanisms. This study delves into elucidating the photosynthetic responses and differences in tolerance to NH4+ stress by investigating the effects on two wheat (Triticum aestivum L.) cultivars, Xumai25 (NH4+-less sensitive) and Yangmai20 (NH4+-sensitive). The cultivars were grown under hydroponic conditions with either sole ammonium nitrogen (NH4+, AN) or nitrate nitrogen (NO3−, NN) as the nitrogen source. NH4+ stress exerted a profound inhibitory effect on seedling growth and photosynthesis in wheat. However, these effects were less pronounced in Xumai25 than in Yangmai20. Dynamic photosynthetic analysis revealed that the suppression in photosynthesis was primarily attributed to stomatal limitation associated with a decrease in leaf water status and osmotic potential. Compared to Yangmai20, Xumai25 exhibited a significantly higher leaf K+ concentration and TaAKT1 upregulation, leading to a stronger stomatal opening and, consequently, a better photosynthetic performance under NH4+ stress. In conclusion, our study suggested stomatal limitation as the primary factor restricting photosynthesis under NH4+ stress. Excessive salt content in soil has adverse effects on cotton production, especially during the germination and seedling stages. γ-aminobutyric acid (GABA) is an important active substance that is expected to improve the resistance of plants to abiotic stresses. This study focused on two cotton cultivars (Gossypium hirsutum L.: Tahe 2 and Xinluzhong 62) and investigated the impact of exogenous GABA (0, 1, 2, 3, and 4 mM) on seed germination, seedling growth, and related morphological, physiological, and biochemical indicators under salt stress (150 mM NaCl). The results showed that salt stress significantly reduced the germination rate and germination index of cotton seeds (decreased by 20.34% and 32.14% for Tahe 2 and Xinluzhong 62, respectively), leading to decreased seedling height and biomass and causing leaf yellowing. Salt stress induced osmotic stress in seedlings, resulting in ion imbalance (marked reduction in K+/Na+ ratio) and oxidative damage. Under salt stress conditions, exogenous GABA increased the germination rate (increased by 10.64~23.40% and 2.63~31.58% for Tahe 2 and Xinluzhong 62, respectively) and germination index of cotton seeds, as well as plant height and biomass. GABA treatment improved leaf yellowing. Exogenous GABA treatment increased the content of proline and soluble sugars, with varying effects on betaine. Exogenous GABA treatment reduced the Na+ content in seedlings, increased the K+ content, and increased the K+/Na+ ratio (increased by 20.44~28.08% and 29.54~76.33% for Tahe 2 and Xinluzhong 62, respectively). Exogenous GABA treatment enhanced the activities of superoxide dismutase and peroxidase, and reduced the accumulation of hydrogen peroxide and malondialdehyde, but had a negative impact on catalase activity. In conclusion, exogenous GABA effectively improved cotton seed germination. By regulating osmoprotectant levels, maintaining ion homeostasis, and alleviating oxidative stress, GABA mitigated the adverse effects of salt stress on cotton seedling growth. Human myeloid leukemia cells (HL-60/S4) exposed to hyperosmotic stress with sucrose undergo dehydration and cell shrinkage. Interphase chromatin and mitotic chromosomes congeal, exhibiting altered phase separation (demixing) of chromatin proteins. To investigate changes in the transcriptome, we exposed HL-60/S4 cells to hyperosmotic sucrose stress (~600 milliOsmolar) for 30 and 60 minutes. We employed RNA-Seq of polyA mRNA to identify genes with increased or decreased transcript levels relative to untreated control cells (i.e., differential gene expression). These genes were examined for over-representation of Gene Ontology (GO) terms. In stressed cells, multiple GO terms associated with transcription, translation, mitochondrial function and proteosome activity, as well as "replication-dependent histones", were over-represented among genes with increased transcript levels; whereas, genes with decreased transcript levels were over-represented with transcription repressors. The transcriptome profiles of hyperosmotically-stressed cells suggest acquisition of cellular rebuilding, a futile homeostatic response, as these cells are ultimately doomed to a dehydrated death. The Scn7A gene encodes NaX, an atypical noninactivating Na+ channel, whose expression in sensory circumventricular organs is essential to maintain homeostatic responses for body fluid balance. However, NaX has also been detected in homeostatic effector neurons, such as vasopressin (VP)-releasing magnocellular neurosecretory cells (MNCVP) that secrete VP (antidiuretic hormone) into the bloodstream in response to hypertonicity and hypernatremia. Yet, the physiological relevance of NaX expression in these effector cells remains unclear. Here, we show that rat MNCVP in males and females is depolarized and excited in proportion with isosmotic increases in [Na+]. These responses were caused by an inward current resulting from a cell-autonomous increase in Na+ conductance. The Na+-evoked current was unaffected by blockers of other Na+-permeable ion channels but was significantly reduced by shRNA-mediated knockdown of Scn7A expression. Furthermore, reducing the density of NaX channels selectively impaired the activation of MNCVP by systemic hypernatremia without affecting their responsiveness to hypertonicity in vivo. These results identify NaX as a physiological Na+ sensor, whose expression in MNCVP contributes to the generation of homeostatic responses to hypernatremia. [ABSTRACT FROM AUTHOR]

The genetic basis underlying adaptive physiological mechanisms has been extensively explored in mammals after colonizing the seas. However, independent lineages of aquatic mammals exhibit complex patterns of secondary colonization in freshwater environments. This change in habitat represents new osmotic challenges, and additional changes in key systems, such as the osmoregulatory system, are expected. Here, we studied the selective regime on coding and regulatory regions of 20 genes related to the osmoregulation system in strict aquatic mammals from independent evolutionary lineages, cetaceans, and sirenians, with representatives in marine and freshwater aquatic environments. We identified positive selection signals in genes encoding the protein vasopressin (AVP) in mammalian lineages with secondary colonization in the fluvial environment and in aquaporins for lineages inhabiting the marine and fluvial environments. A greater number of sites with positive selection signals were found for the dolphin species compared to the Amazonian manatee. Only the AQP5 and AVP genes showed selection signals in more than one independent lineage of these mammals. Furthermore, the vasopressin gene tree indicates greater similarity in river dolphin sequences despite the independence of their lineages based on the species tree. Patterns of distribution and enrichment of Transcription Factors in the promoter regions of target genes were analyzed and appear to be phylogenetically conserved among sister species. We found accelerated evolution signs in genes ACE, AQP1, AQP5, AQP7, AVP, NPP4, and NPR1 for the fluvial mammals. Adaptation to environmental variation caused by global climate change is a significant aspect of fisheries management and ecology. A reduction in ocean salinity is visible in near-shore areas, especially in the Baltic Sea, where it is affecting the Atlantic cod population. Cod is one of the most significant teleost species, with high ecological and economical value worldwide. The population of cod in the Baltic Sea has been traditionally divided into two subpopulations (western and eastern) existing in higher- and lower-salinity waters, respectively. In recent decades, both Baltic cod subpopulations have declined massively. One of the reasons for the poor condition of cod in the Baltic Sea is environmental factors, including salinity. Thus, in this study, an oligonucleotide microarray was applied to explore differences between Baltic cod subpopulations in response to salinity fluctuations. For this purpose, an exposure experiment was conducted consisting of salinity elevation and reduction, and gene expression was measured in gill tissue. We found 400 differentially expressed genes (DEGs) involved in the immune response, metabolism, programmed cell death, cytoskeleton, and extracellular matrix that showed a subpopulation-dependent pattern. These findings indicate that osmoregulation in Baltic cod is a complex process, and that western and eastern Baltic cod subpopulations respond differently to salinity changes. [ABSTRACT FROM AUTHOR]

The Chinese mitten crab (Eriocheir sinensis) is an important aquatic economic species in China, which is sensitive to many toxicants that are likely to occur in the environment and is usually used as the bio-indicator of environmental pollution. As a common contaminant and heavy metal, lead (Pb) easily causes potentially irreversible toxic effects on aquatic organisms. Although increasing attention has been paid to the effect of heavy metals on E. sinensis, the molecular mechanism responding to Pb exposure in E. sinensis has not been comprehensively studied. Therefore, based on histopathology and transcriptome, this study attempted to explore the molecular mechanism of exposure to Pb in gill tissues. Histological analysis showed that the gills were damaged under the Pb stress, affecting their biological functions. Additionally, transcriptome data identified 2505 differentially expressed genes (DEGs), some of which were related to osmotic regulation, energy metabolism, immune defense, and apoptosis, playing vital roles in response to Pb stress. Our study revealed the molecular mechanism of E. sinensis responding to Pb exposure, providing an important foundation for understanding the harmfulness of Pb toxicity to E. sinensis during aquaculture. Parthenium weed poses significant threats to cropping systems, socioeconomic structures, and native ecosystems. The pronounced impact is primarily attributed to its rapid and efficient invasion mechanism. Despite that the detrimental effects of Parthenium weed are widely acknowledged, an in-depth scientific comprehension of its invasion mechanism, particularly regarding modifications in structural and functional attributes under natural conditions, is still lacking. To bridge this knowledge gap and formulate effective strategies for alleviating the adverse consequences of Parthenium weed, a study was conducted in the more cultivated and densely populated areas of Punjab, Pakistan. This study was focused on fifteen distinct populations of the star weed (Parthenium hysterophorus L.) to investigate the factors contributing to its widespread distribution in diverse environmental conditions. The results revealed significant variations in growth performance, physiological traits, and internal structures among populations from different habitats. The populations from wastelands exhibited superior growth, with higher accumulation of soluble proteins (TSP) and chlorophyll content (Chl a&b, TChl, Car, and Chl a/b). These populations displayed increased root and stem area, storage parenchyma, vascular bundle area, metaxylem area, and phloem area. Significant leaf modifications included thicker leaves, sclarification around vascular bundles, and widened metaxylem vessels. Roadside populations possessed larger leaf area, enhanced antioxidant activity, increased thickness of leaves in terms of midrib and lamina, and a higher cortical proportion. Populations found in agricultural fields depicted enhanced shoot biomass production, higher levels of chlorophyll b, and an increased total chlorophyll/carotenoid ratio. Additionally, they exhibited increased phloem area in their roots, stems, and leaves, with a thick epidermis only in the stem. All these outcomes of the study revealed explicit structural and functional modifications among P. hysterophorus populations collected from different habitats. These variations were attributed to the environmental variability and could contribute to the widespread distribution of this species. Notably, these findings hold practical significance for agronomists and ecologists, offering valuable insights for the future management of Parthenium weed in novel environments and contributing to the stability of ecosystems. Abstract Background The development of nanoscale secondary ion mass spectrometry (NanoSIMS) has revolutionized the study of biological tissues by enabling, e.g., the visualization and quantification of metabolic processes at subcellular length scales. However, the associated sample preparation methods all result in some degree of tissue morphology distortion and loss of soluble compounds. To overcome these limitations an entirely cryogenic sample preparation and imaging workflow is required. Results Here, we report the development of a CryoNanoSIMS instrument that can perform isotope imaging of both positive and negative secondary ions from flat block-face surfaces of vitrified biological tissues with a mass- and image resolution comparable to that of a conventional NanoSIMS. This capability is illustrated with nitrogen isotope as well as trace element mapping of freshwater hydrozoan Green Hydra tissue following uptake of 15N-enriched ammonium. Conclusion With a cryo-workflow that includes vitrification by high pressure freezing, cryo-planing of the sample surface, and cryo-SEM imaging, the CryoNanoSIMS enables correlative ultrastructure and isotopic or elemental imaging of biological tissues in their most pristine post-mortem state. The holly Ilex dabieshanensis K. Yao & M. B. Deng, a tree endemic to the Dabieshan Mountains region in China, is a commonly used landscaping plant. Like other crops, its growth is affected by salt stress. The molecular mechanism underlying salt tolerance in holly is still unclear. In this study, we used NaCl treatment and RNA sequencing (RNA-seq) at different times to identify the salt stress response genes of holly. A total of 4775 differentially expressed genes (DEGs) were identified. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis of the DEGs obtained at different salt treatment times (3, 6, 9, 12, and 24 h), as compared to control (ck, 0 h), showed that plant hormone signal transduction and carotenoid biosynthesis were highly enriched. The mechanism by which holly responds to salt stress involves many plant hormones, among which the accumulation of abscisic acid (ABA) and its signal transduction may play an important role. In addition, ion homeostasis, osmotic metabolism, accumulation of antioxidant enzymes and nonenzymatic antioxidant compounds, and transcription factors jointly regulate the physiological balance in holly, providing important guarantees for its growth and development under conditions of salt stress. These results lay the foundation for studying the molecular mechanisms of salt tolerance in holly and for the selection of salt-tolerant varieties.

Western diet (WD) has been shown to impair liver functions via endoplasmic reticulum (ER) stress and oxidative stress. Although osmolytes prevent liver dysfunction, little is known about the mechanisms by which they exert neuroprotective effects against WD-induced damage. We investigated neuroprotective effects of osmolytes and determined the involvement of inflammasome-mediated inflammation in liver and brain. Mice were fed a control diet, WD, or WD with taurine or betaine. Osmolyte supplementation attenuated serum lipid peroxidation and inflammatory cytokine levels in WD-fed mice. Oxidative stress, inflammasome-mediated inflammation, ER stress, and insulin resistance were lower in liver and brain of mice fed osmolyte-supplemented diet than in those fed WD. Moreover, they activated brain-derived neurotrophic factor signaling and decreased β-amyloid deposition and tau hyperphosphorylation in the brain. These data implicate that osmolytes might be promising neuroprotective dietary supplements for WD-induced brain damage, as well as for previously reported genetically and chemically induced brain damage.

Whiteflies (Bemisia tabaci sensu lato) have a wide host range and are globally important agricultural pests. In Sub-Saharan Africa, they vector viruses that cause two ongoing disease epidemics: cassava brown streak disease and cassava mosaic virus disease. These two diseases threaten food security for more than 800 million people in Sub-Saharan Africa. Efforts are ongoing to identify target genes for the development of novel management options against the whitefly populations that vector these devastating viral diseases affecting cassava production in Sub-Saharan Africa. This study aimed to identify genes that mediate osmoregulation and symbiosis functions within cassava whitefly gut and bacteriocytes and evaluate their potential as key gene targets for novel whitefly control strategies. The gene expression profiles of dissected guts, bacteriocytes and whole bodies were compared by RNAseq analysis to identify genes with significantly enriched expression in the gut and bacteriocytes. Phylogenetic analyses identified three candidate osmoregulation gene targets: two α-glucosidases, SUC 1 and SUC 2 with predicted function in sugar transformations that reduce osmotic pressure in the gut; and a water-specific aquaporin (AQP1) mediating water cycling from the distal to the proximal end of the gut. Expression of the genes in the gut was enriched 23.67-, 26.54- and 22.30-fold, respectively. Genome-wide metabolic reconstruction coupled with constraint-based modeling revealed four genes (argH, lysA, BCAT & dapB) within the bacteriocytes as potential targets for the management of cassava whiteflies. These genes were selected based on their role and essentiality within the different essential amino acid biosynthesis pathways. A demonstration of candidate osmoregulation and symbiosis gene targets in other species of the Bemisia tabaci species complex that are orthologs of the empirically validated osmoregulation genes highlights the latter as promising gene targets for the control of cassava whitefly pests by in planta RNA interference.

Despite that the detrimental effects of Parthenium weed are widely acknowledged, an in-depth scientific comprehension of its invasion mechanism, particularly regarding modifications in structural and functional attributes under natural conditions, is still lacking. To bridge this knowledge gap and formulate effective strategies for alleviating the adverse consequences of Parthenium weed, a study was conducted in the more cultivated and densely populated areas of Punjab, Pakistan. This study was focused on fifteen distinct populations of the star weed (Parthenium hysterophorus L.) to investigate the factors contributing to its widespread distribution in diverse environmental conditions. The results revealed significant variations in growth performance, physiological traits, and internal structures among populations from different habitats. The populations from wastelands exhibited superior growth, with higher accumulation of soluble proteins (TSP) and chlorophyll content (Chl a&b, TChl, Car, and Chl a/b). These populations displayed increased root and stem area, storage parenchyma, vascular bundle area, metaxylem area, and phloem area. Significant leaf modifications included thicker leaves, sclarification around vascular bundles, and widened metaxylem vessels. Roadside populations possessed larger leaf area, enhanced antioxidant activity, increased thickness of leaves in terms of midrib and lamina, and a higher cortical proportion. Populations found in agricultural fields depicted enhanced shoot biomass production, higher levels of chlorophyll b, and an increased total chlorophyll/carotenoid ratio. Additionally, they exhibited increased phloem area in their roots, stems, and leaves, with a thick epidermis only in the stem. All these outcomes of the study revealed explicit structural and functional modifications among P. hysterophorus populations collected from different habitats. These variations were attributed to the environmental variability and could contribute to the widespread distribution of this species. Notably, these findings hold practical significance for agronomists and ecologists, offering valuable insights for the future management of Parthenium weed in novel environments and contributing to the stability of ecosystems.

Chloride is one of the most abundant anions in the human body; it is implicated in several physiological processes such as the transmission of action potentials, transepithelial salt transport, maintenance of cellular homeostasis, regulation of osmotic pressure and intracellular pH, and synaptic transmission. The balance between the extracellular and intracellular chloride concentrations is controlled by the interplay of ion channels and transporters embedded in the cellular membranes. Vesicular members of the CLC chloride protein family (vCLCs) are chloride/proton exchangers expressed in the membrane of the intracellular organelles, where they control vesicular acidification and luminal chloride concentration. It is well known that mutations in CLCs cause bone, kidney, and lysosomal genetic diseases. However, the role of CLC exchangers in neurological disorders is only now emerging with the identification of pathogenic CLCN gene variants in patients with severe neuronal and intellectual dysfunctions. Nucleoid associated proteins (NAPs) maintain the architecture of bacterial chromosomes and regulate gene expression. Thus, their role as transcription factors may involve three-dimensional chromosome re-organisation. While this model is supported by in vitro studies, direct in vivo evidence is lacking. Here, we use RT-qPCR and 3C-qPCR to study the transcriptional and architectural profiles of the H-NS (histone-like nucleoid structuring protein)-regulated, osmoresponsive proVWX operon of Escherichia coli at different osmolarities and provide in vivo evidence for transcription regulation by NAP-mediated chromosome re-modelling in bacteria. By consolidating our in vivo investigations with earlier in vitro and in silico studies that provide mechanistic details of how H-NS re-models DNA in response to osmolarity, we report that activation of proVWX in response to a hyperosmotic shock involves the destabilization of H-NS-mediated bridges anchored between the proVWX downstream and upstream regulatory elements (DRE and URE), and between the DRE and ygaY that lies immediately downstream of proVWX. The re-establishment of these bridges upon adaptation to hyperosmolarity represses the operon. Our results also reveal additional structural features associated with changes in proVWX transcript levels such as the decompaction of local chromatin upstream of the operon, highlighting that further complexity underlies the regulation of this model operon. H-NS and H-NS-like proteins are wide-spread amongst bacteria, suggesting that chromosome re-modelling may be a typical feature of transcriptional control in bacteria. The contractile vacuole complex (CVC) is a dynamic and morphologically complex membrane organelle, comprising a large vesicle (bladder) linked with a tubular reticulum (spongiome). CVCs provide key osmoregulatory roles across diverse eukaryotic lineages, but probing the mechanisms underlying their structure and function is hampered by the limited tools available for in vivo analysis. In the experimentally tractable ciliate Tetrahymena thermophila, we describe four proteins that, as endogenously tagged constructs, localize specifically to distinct CVC zones. The DOPEY homolog Dop1p and the CORVET subunit Vps8Dp localize both to the bladder and spongiome but with different local distributions that are sensitive to osmotic perturbation, whereas the lipid scramblase Scr7p colocalizes with Vps8Dp. The H+-ATPase subunit Vma4 is spongiome specific. The live imaging permitted by these probes revealed dynamics at multiple scales including rapid exchange of CVC-localized and soluble protein pools versus lateral diffusion in the spongiome, spongiome extension and branching, and CVC formation during mitosis. Telocytes establish connections and communicate with various types of cells and structures. Few experimental studies have been performed on telocytes. In this study, we investigated the effect of salinity stress on telocytes in relation to osmoregulatory, immune, and stem cells. After exposing the common carp to 0.2 (control), 6, 10, or 14 ppt salinity, we extracted and fixed gill samples in glutaraldehyde, processed and embedded the samples in resin, and prepared semi-thin and ultrathin sections. Two types of telocytes were identified: intraepithelial and stromal telocytes. Intraepithelial telocytes were found to form part of the cellular lining of the lymphatic space and shed secretory vesicles into this space. Stromal telocytes were observed to shed their secretory vesicles into the secondary circulatory vessels. Both intraepithelial and stromal telocytes were enlarged and exhibited increased secretory activities as salinity increased. They exerted their effects via direct contact and paracrine signaling. The following changes were observed in samples from fish exposed to high salinity levels: chloride cells underwent hypertrophy, and their mitochondria became cigar-shaped; pavement cells were enlarged, and their micro-ridges became thin and elongated; stromal telocytes established contact with stem cells and skeletal myoblasts; skeletal muscle cells underwent hypertrophy; and macrophages and rodlet cells increased in number. In conclusion, our findings indicate that intraepithelial and stromal telocytes respond to salinity stress by activating cellular signaling and that they play major roles in osmoregulation, immunity, and regeneration. The river shad, hilsa (Tenualosa ilisha), is an anadromous fish that migrates from marine to freshwater for spawning. This transition/migration poses severe osmotic stress that hilsa needs to successfully minimize. The present study was conducted to evaluate the cellular (ultrastructure of gill and kidney), physiological (hemocyte counts, blood, and water osmolality), and biochemical (free amino acids, free fatty acids, blood glucose, and cortisol levels) parameters of hilsa collected from four different environmental salinity levels (0‰, 10‰, 20‰, and 30‰ salinity levels; using 10 fish samples from each salinity). Results indicate that increased amount of fatty/mucus cells in the gill and lower number of glomerular capsules in kidney were observed at low (0‰ and 10‰) salinities compared to high (20‰ and 30‰) salinities. Water and blood osmolality (total ionic content) showed declining trends with lowering salinities. Total number of blood cells was also found to vary significantly P < 0.05 among salinities. Salinity-specific blood cortisol and glucose levels of hilsa were observed as 0‰ > 10‰ > 30‰ > 20‰. Total essential and free amino acids of hilsa blood showed significantly declining trends P < 0.05 with salinity reductions. No significant differences were observed between 10‰, 20‰, and 30‰ salinities for different types of fatty acids, while hilsa collected from 0‰ showed significantly higher P < 0.05 levels of fatty acids compared to the remaining three salinities. Drought and high salinity are major environmental factors that reduce plant growth and development, leading to loss of plant productivity in agriculture. Under these stress conditions, photosynthesis is greatly suppressed despite the high cellular energy cost of stress response processes. Currently, the process that allows plants to secure the energy required for osmotic stress responses remains elusive. Here, we provide evidence that cytochrome b5 reductase 1 (CBR1), a cytochrome b5 reductase, plays an important role in ATP production in response to NaCl and dehydration stresses. Overexpression and loss of function of CBR1 led to enhanced resistance and sensitivity, respectively, to osmotic stress. Upon exposure to osmotic stress, CBR1 was localised to the endoplasmic reticulum (ER) instead of to mitochondria, where it was localised under normal conditions. Transgenic plants overexpressing ER‐targeted CBR1 showed enhanced resistance to osmotic stress. Moreover, CBR1‐ER and CBR1‐OX plants, had higher levels of ATP and unsaturated fatty acids under osmotic stress. However, these effects were abrogated by thioridazine and 2‐deoxy glucose, inhibitors of β‐oxidation and glycolysis, respectively. Based on these results, we propose that ER‐localised CBR1 triggers ATP production via the production and β‐oxidation of polyunsaturated fatty acids under osmotic stress. Climate change‐induced concurrent drought and salinity stresses significantly threaten global crop yields, yet the physio‐biochemical responses to combined stress in quinoa remain elusive. This study evaluated quinoa responses under four growth conditions: well‐watered, drought stress, salt stress, and drought + salt stress with (15 mM) or without (0 mM) exogenous hydrogen peroxide (H2O2) application. All examined stresses (alone or in combination) reduce quinoa growth and net photosynthesis, although salt stress was found to be less destructive than drought and combined stress. Strikingly, superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), stomatal conductance (gs), photosynthetic rate (PN), K+ uptake, shoot height, shoot fresh, and dry weight were increased by 46.1%, 22.2%, 101.6%, 12.9%, 12.1%, 22.4%, 7.1%, 14%, and 16.4%, respectively, under combined stress compared to drought alone. In addition, exogenous H2O2 effectively improved gaseous exchange, osmolytes' accumulation, and antioxidant activity, resulting in reduced lipid peroxidation, which eventually led to higher plant growth under all coercive conditions. The principle component analysis (PCA) indicated a strong positive correlation between antioxidant enzymes and inorganic ions, which contributed efficiently to osmotic adjustment, particularly under conditions of salinity followed by combined stress. In short, in combination, salt stress has the potential to mitigate drought‐induced injuries by promoting the absorption of inorganic solutes for osmoregulation in quinoa plants. Furthermore, exogenous application of H2O2 could be opted to enhance quinoa performance to increase its tolerance mechanism against drought and salinity, even under combined stress. Background: The Indian white shrimp, Penaeus indicus a native species of India, is important brackishwater aquaculture species. Shrimps are euryhaline in nature and they regulate osmotic and ionic concentrations by osmoregulatory process. However, variations in abiotic factors such as salinity result in stress to the shrimps during culture period affecting their growth and immunity. Methods and results: To understand the adaptive mechanism to stress in low salinity conditions, RNA-seq was used to compare the transcriptomic response of P. indicus upto 3 weeks. De novo assembly using Trinity assembler generated a total of 173,582 transcripts. The assembly had a mean length of 854 bp, N50 value of 1243 bp and GC content of 42.33%. Differential gene expression analysis, resulted in identification of 2130, 3090, and 5351 DEGs in 7 days, 14 days and 21 days respectively of salinity stress period. The pathway prediction of the assembled trinity transcripts using KEGG database showed total number of 329 pathways linking 12,430 transcripts. KEGG pathway enrichment analyses led to the identification of several enriched pathways related to lipid metabolism, amino acid metabolism, glycolysis, signalling pathways etc. Selected genes involved in osmoregulatory process and immune response in shrimps were validated and analysed for the gene expression levels by quantitative real-time PCR (qPCR). Conclusion: This study on the adaptive transcriptomic response of P. indicus to low salinity, will further help in our understanding of the molecular mechanisms underlying osmoregulation mechanism in shrimps. Organ cryopreservation would revolutionize transplantation by overcoming the shelf-life limitations of conventional organ storage. To prepare an organ for cryopreservation, it is first perfused with cryoprotectants (CPAs). These chemicals can enable vitrification during cooling, preventing ice damage. However, CPAs can also cause toxicity and osmotic damage. It is a major challenge to find the optimal balance between protecting the cells from ice and avoiding CPA-induced damage. In this study, we examined the organ perfusion process to shed light on phenomena relevant to cryopreservation protocol design, including changes in organ size and vascular resistance. In particular, we compared perfusion of kidneys (porcine and human) with CPA in either hypotonic or isotonic vehicle solution. Our results demonstrate that CPA perfusion causes kidney mass changes consistent with the shrink-swell response observed in cells. This response was observed when the kidneys were relatively fresh, but disappeared after prolonged warm and/or cold ischemia. Perfusion with CPA in a hypotonic vehicle solution led to a significant increase in vascular resistance, suggesting reduced capillary diameter due to cell swelling. This could be reversed by switching to perfusion with CPA in isotonic vehicle solution. Hypotonic vehicle solution did not cause notable osmotic damage, as evidenced by low levels of lactate dehydrogenase (LDH) in the effluent, and it did not have a statistically significant effect on the delivery of CPA into the kidney, as assessed by computed tomography (CT). Overall, our results show that CPA vehicle solution tonicity affects organ size and vascular resistance, which may have important implications for cryopreservation protocol design. Withania coagulans is a valuable medicinal plant with high demand, but its wild growth and local usage pose a threat to its natural habitat. This study aims to understand the plant’s growth, anatomy, and physiology in different environmental conditions to aid in conservation and re-vegetation efforts. Fifteen differently adapted populations of Withania coagulans were collected from diverse ecological regions, viz., (i) along the roadside, (ii) hilly areas, (iii) barren land, and (iv) wasteland to unravel the adaptive mechanisms that are responsible for their ecological success across heterogenic environments of Punjab, Pakistan. The roadside populations had high values of photosynthetic pigments, total soluble proteins, root endodermis thickness, stem and leaf cortical thickness, and its cell area. The populations growing in hilly areas showed better growth performance such as vigorous growth and biomass production. Additionally, there was enhanced accumulation of organic osmolytes (glycine betaine and proline), chlorophyll content (chl a/b), and enlarged epidermal cells, cortical cells, vascular bundles, metaxylem vessels, and phloem region in roots. In case of stem area, epidermal thickness, cortical thickness, vascular bundle, and pith area showed improved growth. However, the barren land population showed significant increase in carotenoid contents, vascular bundle area, and metaxylem area in roots, and xylem vessels and phloem area in stems and leaves. The wasteland population surpassed the rest of the populations in having greater root dry weight, higher shoot ionic contents, increased root area, thick cortical, and vascular bundle area in roots. Likewise, cortical thickness and its cell area, and pith area in stems, whereas large vascular bundles, phloem region, and high stomatal density were recorded in leaves. Subsequently, natural populations showed the utmost behavior related to tissue organization and physiology in response to varied environmental conditions that would increase the distribution and survival of species.
