A number of efforts have been launched to solve the global electronic waste (e-waste) problem. The efficiency of e-waste recycling is subject to variable national legislation, technical capacity, consumer participation, and even detoxification. E-waste management activities result in procedural irregularities and risk disparities across national boundaries. We review these variables to reveal opportunities for research and policy to reduce the risks from accumulating e-waste and ineffective recycling. Full regulation and consumer participation should be controlled and reinforced to improve local e-waste system. Aiming at standardizing best practice, we alter and identify modular recycling process and infrastructure in eco-industrial parks that will be expectantly effective in countries and regions to handle the similar e-waste stream. Toxicity can be deleted through material substitution and detoxification during the life cycle of electronics. Based on the idea of "Control-Alt-Delete", four patterns of the way forward for global e-waste recycling are proposed to meet a variety of local situations.

PurposeProgression-free survival (PFS) in metastatic castration-resistant prostate cancer (mCRPC) trials has been inconsistently defined and poorly associated with overall survival (OS). A reproducible quantitative definition of radiographic PFS (rPFS) was tested for association with a coprimary end point of OS in a randomized trial of abiraterone in patients with mCRPC.Patients and methodsrPFS was defined as ≥ two new lesions on an 8-week bone scan plus two additional lesions on a confirmatory scan, ≥ two new confirmed lesions on any scan ≥ 12 weeks after random assignment, and/or progression in nodes or viscera on cross-sectional imaging, or death. rPFS was assessed by independent review at 15% of deaths and by investigator review at 15% and 40% of deaths. rPFS and OS association was evaluated by Spearman's correlation.ResultsA total of 1,088 patients were randomly assigned to abiraterone plus prednisone or prednisone alone. At first interim analysis, the hazard ratio (HR) by independent review was 0.43 (95% CI, 0.35 to 0.52; P < .001; abiraterone plus prednisone: median rPFS, not estimable; prednisone: median rPFS, 8.3 months). Similar HRs were obtained by investigator review at the first two interim analyses (HR, 0.49; 95% CI, 0.41 to 0.60; P < .001 and HR, 0.53; 95% CI, 0.45 to 0.62; P < .001, respectively), validating the imaging data assay used. Spearman's correlation coefficient between rPFS and OS was 0.72.ConclusionrPFS was highly consistent and highly associated with OS, providing initial prospective evidence on further developing rPFS as an intermediate end point in mCRPC trials.

c-Jun N-terminal kinase (JNK) plays a key role in the regulation of neuronal apoptosis. Previous studies have revealed that forkhead transcription factor (FOXO3a) is a critical effector of JNK-mediated tumor suppression. However, it is not clear whether the JNK/FOXO3a pathway is involved in neuronal apoptosis in the developing rat brain after hypoxia-ischemia (HI). In this study, we generated an HI model using postnatal day 7 rats. Fluorescence immunolabeling and Western blot assays were used to detect the distribution and expression of total and phosphorylated JNK and FOXO3a and the pro-apoptotic proteins Bim and CC3. We found that JNK phosphorylation was accompanied by FOXO3a dephosphorylation, which induced FOXO3a translocation into the nucleus, resulting in the upregulation of levels of Bim and CC3 proteins. Furthermore, we found that JNK inhibition by AS601245, a specific JNK inhibitor, significantly increased FOXO3a phosphorylation, which attenuated FOXO3a translocation into the nucleus after HI. Moreover, JNK inhibition downregulated levels of Bim and CC3 proteins, attenuated neuronal apoptosis and reduced brain infarct volume in the developing rat brain. Our findings suggest that the JNK/FOXO3a/Bim pathway is involved in neuronal apoptosis in the developing rat brain after HI. Agents targeting JNK may offer promise for rescuing neurons from HI-induced damage.

In this paper, aiming at the problem of chrome-iron separation in electroplating sludge, the separation of ferrochrome by complexation and precipitation with benzoic acid as complexing agent is achieved. The optimal conditions consisted of a 1: 3 molar ratio of Fe3+: C6H5COOH, a reaction temperature of 30°C, a final pH of 2.5 and a reaction time of 2 min. The separation rate of the iron was 97.38% and the rate of loss of chromium was only 3.59%. The ferrochromium separation products were analyzed by XRD, fluorescence spectroscopy, infrared spectroscopy and H NMR Spectroscopy in order to study the mechanism of precipitation. The results showed that benzoic acid preferentially forms a complex with iron and iron benzoate precipitates with an increase pH. The iron benzoate crystals have a fine particle size, settle rapidly and are easy to filter. The separation of Cr 3+/Fe3+ was successful using our methodology.

BackgroundDynamin 2 (Dyn2) is a ~100kDa GTPase that assembles around the necks of nascent endocytic and Golgi vesicles and catalyzes membrane scission. Mutations in Dyn2 that cause centronuclear myopathy (CNM) have been shown to stabilize Dyn2 polymers against GTP-dependent disassembly in vitro. Precisely timed regulation of assembly and disassembly is believed to be critical for Dyn2 function in membrane vesiculation, and the CNM mutations interfere with this regulation by shifting the equilibrium toward the assembled state.MethodsIn this study we use two fluorescence fluctuation spectroscopy (FFS) approaches to show that a CNM mutant form of Dyn2 also has a greater propensity to self-assemble in the cytosol and on the plasma membrane of living cells.ResultsResults obtained using brightness analysis indicate that unassembled wild-type Dyn2 is predominantly tetrameric in the cytosol, although different oligomeric species are observed, depending on the concentration of expressed protein. In contrast, an R369W mutant identified in CNM patients forms higher-order oligomers at concentrations above 1μM. Investigation of Dyn2-R369W by Total Internal Reflection Fluorescence (TIRF) FFS reveals that this mutant forms larger and more stable clathrin-containing structures on the plasma membrane than wild-type Dyn2.Conclusions and general significanceThese observations may explain defects in membrane trafficking reported in CNM patient cells and in heterologous systems expressing CNM-associated Dyn2 mutants.

Metagenomic next‐generation sequencing (mNGS) is a valuable technique for identifying pathogens. However, conventional mNGS requires the separate processing of DNA and RNA genomes, which can be resource‐ and time‐intensive. To mitigate these impediments, we propose a novel method called DNA/RNA cosequencing that aims to enhance the efficiency of pathogen detection. DNA/RNA cosequencing uses reverse transcription of total nucleic acids extracted from samples by using random primers, without removing DNA, and then employs mNGS. We applied this method to 85 cases of severe acute respiratory infections (SARI). Influenza virus was identified in 13 cases (H1N1: seven cases, H3N2: three cases, unclassified influenza type: three cases) and was not detected in the remaining 72 samples. Bacteria were present in all samples. Pseudomonas aeruginosa, Klebsiella pneumoniae, and Acinetobacter baumannii were detected in four influenza‐positive samples, suggesting coinfections. The sensitivity and specificity for detecting influenza A virus were 73.33% and 95.92%, respectively. A κ value of 0.726 indicated a high level of concordance between the results of DNA/RNA cosequencing and SARI influenza virus monitoring. DNA/RNA cosequencing enhanced the efficiency of pathogen detection, providing a novel capability to strengthen surveillance and thereby prevent and control infectious disease outbreaks. As stringent demands for PI materials with low-temperature curable properties have increased in the high-frequency communication era, the introduction of flexible structures has gained prominence for enhancing molecular chain mobility. However, systematic studies on the effect of flexible structures on low-temperature curing ability remain limited. In this work, we designed a new dianhydride monomer with isopropylidene and ester groups named TABPP. Low-temperature curable polyimides with different molecular chain flexibility were prepared by choosing different diamines and anhydrides. Surprisingly, the degree of imidization was not straightforwardly aligned with molecular chain flexibility. An exemplar instance was PI-9-200, exhibiting the greatest flexibility among the samples, yet possessing the lowest degree of imidization (ID) of 68.59%. Based on the analysis of experimental results and front-line orbital energy levels, it could be seen that the mismatch between the ID and the flexibility may originate from the influence of electronic effects of the monomers. Notably, when enough flexible structures were introduced into the polyimide backbone, the effect of increasing the free volume appeared to outweigh the influence of incomplete imidization, thereby favoring the preparation of low-temperature curable PI films with outstanding dielectric properties. In particular, the dielectric constant of the prepared low-temperature curable PI films was as low as 2.50, which is the best performance among the low-temperature curable PI films. This work throws new light on the correlation between flexibility and low-temperature curing ability and offers fresh perspectives on the preparation of low-temperature curable PIs with excellent dielectric properties. In a previous study, we utilized saturated prewet high titanium heavy slag sand to produce UHPC (ST-UHPC). ST-UHPC has high impact and abrasion resistance. For better ST-UHPC applications, we investigate the mechanism of ST-UHPC under different curing systems from the microstructure and macroscopic perspective in this paper. We prepared ST-UHPC under four maintenance conditions: 20 °C standard curing, 90 °C steam curing, 90 °C dry curing and 210 °C 2 MPa pressure steam curing. Then, we analyzed the hydration product composition, the degree of cement hydration, the C-A-S-H gel microstructure and the substitution of Al3+ for Si4+ in relation to these prepared ST-UHPCs. Compared with standard curing, dry curing at 90 °C accelerated the water evaporation and reduced the hydration degree of ST-UHPC cementite. However, pressure steam curing significantly improved the hydration degree of ST-UHPC cementing material, and increased the MCL and Al[4]/Si of C-A-S-H gel. In addition, pressure steam curing reduced the Ca/Si and promoted the conversion of C-A-S-H cementing to tobermorite. Compared with dry curing at 90 °C, pressure steam curing significantly improved the macroscopic properties of ST-UHPC. The macro-performance difference of ST-UHPC under standard curing and 90 °C steam curing is small. The reason is that steam curing caused the water to be rapidly released in the internal aggregate of ST-UHPC. This resulted in the increase of the interface between the internal aggregate of ST-UHPC and the ST-UHPC cementate. The harmful pores in the ST-UHPC matrix under steam curing were also increased. To sum up, compared with standard curing, dry curing at 90 °C weakened the mechanical properties and microstructure of ST-UHPC, but steam pressure curing increased them. The single steam curing had no significant effect on the mechanical properties and microstructure of the ST-UHPC. Therefore, non-steam and room-temperature moisturizing maintenance should be adopted for anti-abrasive UHPC. [ABSTRACT FROM AUTHOR]
