Panax notoginseng possesses therapeutic potential for cardiovascular disorders. Endoplasmic reticulum (ER) stress plays a critical role in cardiovascular disease. Here we show that a rare ginsenoside-standardized extract (RGSE) from notoginseng exerts cardioprotection via attenuating ER stress-associated apoptosis. A total of 18 ginsenosides with a concentration of 816.75 mg/g were characterized and quantified in RGSE. Ginsenoside Rh4, Rg5, Rk3, and Rk1 were the most abundant. RGSE at 50 and 100 mg/kg significantly attenuated myocardial infarction and improved cardiac function in acute myocardial infarction (AMI). RGSE also prevented myocardial ischemia-induced oxidative stress and apoptosis in vivo and in vitro . In addition, RGSE decreased the overexpression of GRP78, GRP94 and CHOP, and inhibited the phosphorylation of PERK and IRE1α in ER stress. Further investigation revealed that RGSE effectually inactivated TXNIP/NLRP3 signaling pathway in AMI hearts. These observations suggest a novel role for notoginseng in regulating AMI. The pharmacokinetics of 5-fluorouracil (5-FU) in combination with or without American ginseng (seven-consecutive days oral dose) in rats were evaluated using liquid chromatography-electrospray ionization-mass spectrometry (LC-MS). Chromatographic separation was performed on a reverse LC column within a total run time of 6.5 min, which allowed for a relatively quick analysis. The limit of quantification for 5-FU was 15 ng/mL and this method was linear over 15-50,000 ng/mL. This method supported stabilizing determination of the plasma concentration of 5-FU over a period of 24 h. Precision both interday and intraday (coefficient of variation) was within 14% and accuracy (relative error) ranged from −5 to 14%. In view of the observed pharmacokinetic parameters, including maximum concentration, time to maximum concentration, area under the concentration-time curve (AUC), mean residence time, elimination half-life and clearance, our results showed no significant differences in all of the pharmacokinetic parameters between the ginseng co-treated group and 5-FU alone group. Some increase in AUC was observed in 5-FU plus ginseng group; however, the difference did not reach statistical significance compared with 5-FU alone. Compounds derived from natural products are becoming promising alternative drugs/tools in Alzheimer's disease (AD) therapeutics. From an in-house natural products library, seventeen hits were selected for their inhibitory effect on the production of amyloid-β (Aβ) with IC50 lower than 10 μM without causing obvious toxicity. Among these compounds, camptothecin (CPT) and its analogs showed inhibitory effects on amyloid-β 1-42 (Aβ42) with the IC50 value in the nanomolar range in HEKsw cells and SHSY5Ysw cells. Further studies showed that CPT and its analogs inhibited Aβ42 via a p53 dependent pathway. Meanwhile, CPT and its analogs could also inhibit Aβ42 induced IL-1β production in the THP-1 cells. Objective This study was to explore the antifibrotic effect and the possible mechanism of total polysaccharides of Y upingfeng ( YPF- P) on bleomycin ( BLM)-induced pulmonary fibrosis in rats. Methods Pulmonary fibrosis was induced in Sprague- Dawley rats by BLM (5 mg/kg), killed 14 and 28 days after BLM administration by abdominal aorta exsanguination and removed the lungs. Lung coefficient was counted at the same time. Besides, H&E and Masson's trichrome staining for histopathological changes of lung tissues were observed. Additionally, western blotting and immunohistochemical staining techniques were used to detect expression of transforming growth factor-β1 ( TGF-β1), type I collagen ( Col- I) and α-smooth muscle actin (α- SMA). Finally, the levels of Col- I and hydroxyproline ( HYP) in lung tissues were also utilized. Key findings YPF- P alleviated the increase of lung coefficient induced by BLM instillation in pulmonary fibrosis rat, pathologic changes and collagen distribution were obviously ameliorated, while the increase of α- SMA-positive cells and TGF-β1 expression was prevented after YPF- P treatment. Moreover, the contents of HYP and Col- I were decreased in YPF- P group. Conclusions YPF- P had antifibrotic effect in experiment, which may reduce the synthesis and promote the deposition of Col- I via suppressing the increase of TGF-β1-mediated activation of myofibroblasts. Purpose: To identify bioactive equivalent combinatorial components (BECCs) in herbal medicines. The exact composition of effective components in herbal medicines is often elusive due to the lack of adequate screening methodology. Herein, we propose a hypothesis that BECCs accounting for the whole efficacy of original herbal medicines could be discovered from a complex mixture of constituents. Methods: We developed a bioactive equivalence oriented feedback screening method and applied it to discover the BECCs from an herbal preparation Cardiotonic Pill (CP). The operations include chemical profiling of CP, followed by an iterative loop of determining, collecting and evaluating candidate BECCs. Results: A combination of 18 compounds was identified as BECCs from CP, which accounts for 15.0% (w/w) of original CP. We have demonstrated that the BECCs were as effective as CP in cell models and in a rat model of myocardial infarction. Conclusions: This work answers the key question of which are real bioactive components for CP that have been used in clinic for many years, and provides a promising approach for discovering BECCs from herbal medicines. Abstract: Aim: To reveal the profile of astragalosides for better quality evaluation of Radix Astragali, this study was aimed to investigate the transformation of astragalosides under different conditions. Method: Seven major astragalosides were selected for evaluation under acidic, neutral and alkaline conditions. The transformation in real plant samples was also examined and the products were characterized by LC-ESI-TOF/MS. Results: In weak acidic solution, all of the astragalosides are stable. In addition, the transformation ratios of the astragalosides under neutral and alkaline conditions were also obtained. Conclusion: In neutral solution, malonylastragaloside I was transformed to astragaloside I; and in alkaline solution, substituent group(s) in the xylose moiety of all the astragalosides were eliminated. Since astragalosdie IV is the basic skeleton structure of the astrgalosides, it is a common transformation product of other astragalosides. [Copyright &y& Elsevier]

Panaxadiol (PD) is a purified sapogenin of ginseng saponins, which exhibits anticancer activity. Epigallocatechin gallate (EGCG), a major catechin in green tea, is a strong botanical antioxidant. In this study, we investigated the possible synergistic anticancer effects of PD and EGCG on human colorectal cancer cells and explored the potential role of apoptosis in the synergistic activities. Effects of selected compounds on HCT-116 and SW-480 human colorectal cancer cells were evaluated by a modified trichrome stain cell proliferation analysis. Cell cycle distribution and apoptotic effects were analyzed by flow cytometry after staining with PI/RNase or annexin V/PI. Cell growth was suppressed after treatment with PD (10 and 20 µ m) for 48 h. When PD (10 and 20 µ m) was combined with EGCG (10, 20, and 30 µ m), significantly enhanced antiproliferative effects were observed in both cell lines. Combining 20 µ m of PD with 20 and 30 µ m of EGCG significantly decreased S-phase fractions of cells. In the apoptotic assay, the combination of PD and EGCG significantly increased the percentage of apoptotic cells compared with PD alone ( p < 0.01). The synergistic apoptotic effects were also supported by docking analysis, which demonstrated that PD and EGCG bound in two different sites of the annexin V protein. Data from this study suggested that apoptosis might play an important role in the EGCG-enhanced antiproliferative effects of PD on human colorectal cancer cells. Triptolide, a biologically active natural product from Tripterygium wilfordii, protects neurons from inflammation-mediated damage. Our results showed for the first time that triptolide inhibited the expression of CXCR2 and presenilin in a neuroblastoma cell line SHSY5Ysw. Moreover, triptolide potently inhibited amyloid-β1-42 production with IC50 value of 30 pM in HEK293sw cells or 2 nM in SHSY5Ysw cells, respectively. We also demonstrated that triptolide prevented primary cortical neurons from chemokine CXCL1-induced cytotoxicity. Therefore, our study indicates that the neural protective effect of triptolide is largely mediated by inhibiting CXCR2 activity. [ABSTRACT FROM AUTHOR]

Abstract: Radix Polygoni Multiflori, Rhizoma et Radix Polygoni Cuspidati, and Radix et Rhizoma Rhei are the most frequently used traditional Chinese medicines in the family Polygonaceae. The three herbal medicines (HMs) contain similar types of compounds. In Chinese Pharmacopoeia, five high-performance liquid chromatography (HPLC) coupled with ultraviolet (UV) detection methods have been employed for their quality control. The aim of this study was to develop a simple and conventional strategy, segmental monitoring based on variable wavelength detection (VWD), for simultaneous quantification of phenolic acids, flavonoids, stilbenes and anthraquinones in the three chemically analogous HMs. Compared with the commonly used HPLC-diode array detection (DAD), the proposed method afforded desirable performance on linearity, precision and sensitivity. Additionally, a quadrupole time-of-flight mass spectrometry (QTOF-MS) was applied to the structural confirmation of analytes from complex matrices. Based on the chemical profiles and contents of the analyses, 27 samples from three HMs were well classified using the principal component analysis. Abstract: As one of the most commonly used medicinal plants, ginseng has been an attractive model herb for study. A wide range of analytical methods has been used to characterize its constituents. However, less effort has been devoted to the rare ginseng saponins, especially their isomers and sugar linkages. In this study, we used segmental monitoring and diagnostic ion filtering to characterize ginseng saponins by rapid liquid chromatography with time-of-flight mass spectrometry (LC-TOF-MS). By using selected diagnostic ions, specific groups of ginseng saponins were readily extracted from the complicated matrix. 20(R) and 20(S) stereo-saponins were differentiated using the peak abundance ratio of [M-H2O+H]+ to [M-2H2O+H]+. The fragmentation behavior of ginsenosides was first reported in negative ion mode by MS/MS with high-energy collision-induced dissociation, producing rules to determine sugar numbers, positions and linkages. Using the rules, we identified and compared the nontarget ginseng saponin profiling of raw and steamed American ginseng roots and berries. We characterized 70 saponins in the samples. Abstract: Aim: To study the differentially-expressed proteins for malonylastragaloside I in human leukemia U937 cells. Methods: A suitable protein quantitative mode of label-free technique was established by LC Chip Q-TOF. It was found that the mode of peptide spectral intensity based on label-free quantification strategy of LC Chip Q-TOF showed high reproducibility, wide dynamic range, as well as simplicity and convenience. The optimized quantification mode of the peptide spectral intensity, normalization algorithm was applied to screen the differentially-expressed proteins for malonylastragaloside I (MA-I), a novel astragaloside from Astragalus sp., in human leukemia U937 cells. Results: Fifteen differentially-expressed proteins with up or down-regulation over 2.0 folds were identified, six of which including HSC70, RPLP2, PHB2, PCNA, TCTP and ANXA11 were confirmed by Western blotting. We also observed significant activation of PARP1 and cleavage of caspases-3 and −9 by MA-I in U937 cells, and the pan-caspase inhibitor Z-VAD-FMK could reduce MA-I inhibited cell proliferation, indicating that caspase activation may be involved in MA-I induced cell toxicity. Conclusion: This study has generated potentially functional proteins important for MA-I inhibited U937 cells proliferation by the optimized quantification mode of peptide spectral intensity. Abstract: Ginseng occupies a prominent position in the list of best-selling natural products in the world. Compared to the long history of use and widespread research on Asian ginseng, the study of American ginseng is relatively limited. In the past decade, some promising advances have been achieved in understanding the chemistry, pharmacology and structure–function relationship of American ginseng. To date, there is no systematic review of American ginseng. In this review, the different structures of the ginsenosides in American ginseng are described, including naturally occurring compounds and those resulting from steaming or biotransformation. Preclinical and clinical studies published in the past decade are also discussed. Highlighted are the chemical and pharmacological diversity and potential structural–activity relationship of ginsenosides. The goal is that this article is a useful reference to chemists and biologists researching American ginseng, and will open the door to agents in drug discovery. [Copyright &y& Elsevier]

Abstract: In this work, the metabolite profiles of Danggui Buxue Tang (DBT) in rat bile and plasma were qualitatively described, and the possible metabolic pathways of DBT were subsequently proposed. Emphasis was put on correlative analysis of metabolite profiling in different biological fluids. After oral administration of DBT, bile and plasma samples were collected and pretreated by solid phase extraction. Rapid resolution liquid chromatography coupled to time-of-flight mass spectrometry (RRLC–TOFMS) was used for characterization of DBT-related compounds (parent compounds and metabolites) in biological matrices. A total of 142 metabolites were detected and tentatively identified from the drug-containing bile and plasma samples. Metabolite profiling shows that rat bile contained relatively more glutathione-derived conjugates, more saponins compounds and more diverse forms of metabolites than urine. The metabolite profile in plasma revealed that glucuronide conjugates of isoflavonoids, dimmers, acetylcysteine conjugates and parent form of phthalides, as well as saponin aglycones were the major circulating forms of DBT. Collectively, the metabolite profile analysis of DBT in different biological matrices provided a comprehensive understanding of the in vivo metabolic fates of constituents in DBT. In recent studies of American ginseng, steaming or heating altered the ginsenoside profile and thereby increased anticancer effects. Yet the ginsenoside structures and their activities have not been systematically elucidated. In this commentary, we introduce the different ginsenosides in American ginseng, both the naturally occurring compounds and those resulting from steaming or biotransformation. We briefly review American ginseng''s reported anticancer effects and their mechanisms of action, and explore the possible structural-function relationship with a focus on sugar molecules, hydroxyl groups and stereoselectivity in ginsenosides. Understanding these relationships may produce insights into chemical and pharmacological approaches for enhancing the chemopreventive effects of ginsenoside and for developing novel anticancer agents. Asperosaponin VI (also named akebia saponin D) is a typical bioactive triterpenoid saponin isolated from the rhizome of Dipsacus asper Wall (Dipsacaceae). In this work, a sensitive high-performance liquid chromatography-electrospray ionization-mass spectrometry (HPLC-ESI-MS) assay has been established for determination of asperosaponin VI in rat plasma. With losartan as the internal standard (IS), plasma samples were prepared by protein precipitation with methanol. Chromatographic separation was performed on a C18 column with a mobile phase of 10 m m ammonium acetate buffer containing 0.05% formic acid-methanol (32 : 68, v/v). The analysis was performed on an ESI in the selected ion monitoring mode using target ions at m/z 951.4 for asperosaponin VI and m/z 423.2 for the IS. The calibration curve was linear over the range 3-1000 ng/mL and the lower limit of quantification was 3.0 ng/mL. The intra- and inter-assay variability values were less than 9.5 and 7.8%, respectively. The accuracies determined at the concentrations of 3.0, 100.0, 300.0 and 1000 ng/mL for asperosaponin VI were within ±15.0%. Buyang Huanwu Decoction (BYHYD) is a classical traditional Chinese medicinal prescription that has been widely used for treating cerebrovascular illnesses for hundreds of years. In this study, a comprehensive analytical method has been developed for quantitative analysis of the major constituents in BYHWD. This method was based on high-performance liquid chromatography coupled to a diode array and evaporative light scattering detectors (HPLC-DAD-ELSD) on a common reverse-phase C18 column. HPLC coupled with on-line time-of-flight mass spectrometry (HPLC-TOF/MS) was additionally adopted to provide further validation for the constituents. It was found that 0.3% aqueous formic acid and acetonitrile was the optimum mobile phase for gradient elution. This method, which showed excellent precision and accuracy, was successfully applied to quantify the bioactive constituents in six BYHWD products. The validated HPLC-DAD-ELSD method, together with the HPLC-TOF/MS analysis, provided a new basis for assessing the quality of traditional Chinese medicinal prescription consisting of many bioactive components. Abstract: The fact that the effects of herbal medicines (HMs) are brought about by their chemical constituents has created a critical demand for powerful analytical tools performing the chemical analysis to assure their efficacy, safety and quality. Liquid chromatography coupled to mass spectrometry (LC–MS) is an excellent technique to analyze multi-components in complex herbal matrices. Due to its inherent characteristics of accurate mass measurements and high resolution, time-of-flight (TOF) MS is well-suited to this field, especially for qualitative applications. The purpose of this article is to provide an overview on the potential of TOF, including the hybrid quadrupole- and ion trap-TOF (QTOF and IT-TOF), hyphenated to LC for chemical analysis in HMs or HM-treated biological samples. The peculiarities of LC–(Q/IT)TOF-MS for the analysis of HMs are discussed first, including applied stationary phase, mobile-phase selection, accurate mass measurements, fragmentation and selectivity. The final section is devoted to describing the applicability of LC–(Q/IT)TOF-MS to routine analysis of multi-components, including target and non-target (unknown) compounds, in herbal samples, emphasizing both the advantages and limitations of this approach for qualitative and quantitative purposes. The potential and future trends of fast high-performance liquid chromatography (HPLC) (e.g. rapid resolution LC and ultra-performance LC) coupled to (Q)TOF-MS for chemical analysis of HMs are highlighted. Abstract: A dynamic microdialysis sampling method with liquid chromatography–diode array detection and time-of-flight mass spectrometry (LC–DAD–TOF/MS) analysis was developed to investigate rat microsomal metabolisms of calycosin and formononetin, and their drug–drug interactions. Two hydroxylated metabolites from calycosin, and three hydroxylated or 4′-O-demethylated derivatives from formononetin were detected and identified after co-incubation with microsomes. Calibration curves offered linear ranges of two orders of magnitude with r 2 >0.999 for calycosin, formononetin and daidzein. The quantitative LC method provides a range of 0.028–0.034μg/mL for limits of detection, overall precision less than 5% and accuracy less than 3% by RSD. Besides, calycosin and formononetin were found to produce the depressive effect on the CYP450 enzyme reaction, and inhibit phase I enzyme reaction of each other when they are concurrent. Dynamic microdialysis sampling with LC–DAD–TOF/MS analysis developed in this work is a powerful tool for in vitro metabolism studies of drugs and metabolic interactions. [Copyright &y& Elsevier]

Abstract: A high-performance liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry (HPLC–ESI-TOF-MS) method has been developed for qualitative and quantitative analysis of isoflavonoids and saponins, as well as for the quality control of Radix Astragali and its preparations. The selectivity, reproducibility and sensitivity are compared with HPLC with diode array detection (DAD) and evaporative light scattering detection (ELSD). Limits of detection and quantification fell in ranges of 9–40 and 23–103ng/mL for 13 analytes with a injection of 10μL samples, and all calibration curves showed good linear regression (r 2 >0.9938) within the test range. The assay was successfully utilized to analyze the 13 marker components in 20 samples of Radix Astragali products. The quantitative results demonstrated that samples from different localities and manufacturers showed different quality. Advantages, in comparison with conventional HPLC–DAD–ELSD, are that reliable identification of target compounds could be achieved by accurate mass measurements (<3ppm) along with characteristic retention time, extracted ions chromatograms using a narrow mass window for quantification ensure that the chromatographic peaks are free from background or co-elutes interference, and the great enhancement in selectivity and sensitivity allows identification and quantification of low levels of constituents in complex Radix Astragali matrixes. Abstract: A quantitative method by ultra-fast HPLC coupled with time-of-flight mass spectrometry (TOF/MS) has been developed for analysis of 15 major constituents in Danggui Buxue Tang (DBT) preparations. Its performance was compared with a conventional HPLC method with diode array detection/evaporative light scattering detection (DAD/ESLD). Accurate mass measurements within 3ppm error were obtained for all the compounds. The analytical time by an ultra-fast system is 4 times faster than conventional HPLC, the limits of detection by TOF/MS are low to 0.004–0.08ng compared with 1–200ng for HPLC–DAD–ELSD, and acceptable linearity of response was demonstrated over two orders of magnitude (r 2 >0.99) for all analytes. Intra-day reproducibility was below 3% and inter-day values were below 5% R.S.D. Robustness and applicability of the method was validated for the analysis of market DBT real samples, proposing a rapid, sensitive and validated method for routine analysis and quality control of DBT. Abstract: A novel on-line technique for stacking and sweeping of long sample plugs with simultaneous determination of charged analytes in the plant (protocatechuic aldehyde, rosmarinic acid, danshensu, salvianolic acid B, and protocatechuic acid) by the nonionic microemulsion electrokinetic chromatography (MEEKC) is presented. The preconcentration efficiency provided about 9–28-fold for stacking and 7–14-fold for sweeping in the enhancements of LOD. The effects of oil phase, Brij-35 and buffer concentrations on stacking and sweeping efficiency were examined in order to optimize the two methods. In nonionic MEEKC, the effect of the type of oil and buffer contents on preconcentration mechanism is often sophisticated. This study had demonstrated that the oil type and buffer content in nonionic microemulsion indeed markedly altered the affinity of microemulsion with analytes. Finally, in comparison to the stacking method, the most apparent disadvantages of the sweeping method were the relatively high limits of detection and poor peak shapes. [Copyright &y& Elsevier]

Abstract: A liquid chromatography-electrospray ionization–mass spectrometry method has been developed and validated for identification and quantification of four major bioactive saponins in rat plasma after oral administration of extraction of saponins from Flos Lonicerae, i.e., macranthoidin B, macranthoidin A, dipsacoside B, and macranthoside B. Plasma samples were extracted with solid-phase extraction, separated on a Shim-pack CLC-ODS column and detected by MS in negative selective ion monitoring mode. Calibration curves offered linear ranges of two orders of magnitude with r 2 >0.999. The method showed the low limit quantification of 7.72, 6.06, 7.16, and 1.43ng/mL for macranthoidin B, macranthoidin A, dipsacoside B, and macranthoside B, respectively. The inter- and intra-CV precision (R.S.D.) were all within 10% and accuracy (% bias) ranged from −10 to 10%. The overall recovery was more than 70%. This developed method was subsequently successfully applied to pharmacokinetic profiles of the four saponins in rats. Rapid resolution liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry was developed for simultaneous determination of twelve saponins in Radix et Rhizoma Notoginseng (RRN) with macranthoidine A as internal standard. All calibration curves showed good linear regression ( r 2 > 0.9935) within test range. The limits of detection of the twelve compounds were in the range of 1.53–5.08 ng mL−1. A simple, sensitive, and validated liquid chromatographic method has been developed for the determination of tectorigenin in rat plasma and application to a pharmacokinetic study after oral administration of tectorigenin or its prodrug tectoridin. The analysis was performed on a Kromasil C18 analytical column using gradient elution with acetonitrile 0.1% phosphonic acid water at 0.8 mL min−1. The detection wavelength for UV detection was set at 264 nm. The established method was fully validated with parameters as follows: the intra- and inter-day assay precisions (CV) of three analytes were in the range of 4.2–13.3% and accuracies were between 98.0 and 107.5%; the calibration curve was linear with r 2 > 0.99 over a concentration range of 0.02–2 μg mL−1; the lower limit of quantification was 0.02 μg mL−1; tectorigenin showed stable in rat plasma after 12 h incubation at room temperature, 15 days storage at −80 °C and three freeze/thaw cycles, as well as in reconstitute buffer for 24 h at 25 °C; and the mean recoveries of tectorigenin were 92.3 ± 3.2, 95.5 ± 2.9 and 94.5 ± 3.0% with quality control levels of 0.02, 0.2 and 2 μg mL−1, respectively. Abstract: A novel fast high-performance liquid chromatography (HPLC) method coupled with diode array detection (DAD) and time-of-flight mass spectrometry (TOF/MS) was developed for qualitative and quantitative analysis of Radix Astragali products. The potential of fast HPLC on 1.8-μm particles was compared with the performance of HPLC on conventional 5-μm particles columns. Significant advantages of fast HPLC include high-speed chromatographic separation, four times faster than HPLC with conventional columns, and great enhancement in sensitivity with limits of detection low to 0.001ng. With dynamic adjustment of fragmentor voltage in TOF/MS, an efficient transmission of the ions was achieved to obtain the best sensitivity and abundant fragmentation. By accurate mass measurements within 5ppm error for each molecular ion and subsequent fragment ions, a reliable identification and differentiation of six major saponins including two groups of isomers and twelve main isoflavonoids was described here for the first time. For quantitative analysis by fast HPLC–TOF/MS, linearity of response over two orders of magnitude was demonstrated (r 2 >0.99) for all analytes. Intra-day reproducibility was below 3% RSD and inter-day values were below 5% RSD. A good correlation (slope=1.1108, r 2 =0.9853) was observed for accuracy test. It is concluded that the fast and sensitive HPLC-DAD–TOF/MS is powerful in qualitative and quantitative analysis of complex herbal medicines in terms of time savings, sensitivity, selectivity, precision, accuracy as well as increasing sample throughout and lowering solvent consumption. Column liquid chromatography with diode array and mass spectrometric detection was developed for the qualitative and quantitative analysis of the major constituents in Radix Angelicae sinensis. Sixteen compounds including phthalic acid, vanillic acid, ferulic acid, guaiacol, Z-6,7-epoxyligustilide, senkyunolide F, carvacrol, Z-ligustilide, Z-butylidenephthalide, E-6,7-dihydroxydihydroligustilide, senkyunolide I or senkyunolide H, coniferyl ferulate, sendanelolide, butylphthalide, E-ligustilide, E-butylidenephthalide except guaiacol and carvacrol were identified using online ESI–MS in comparisons with literature data and standard compounds. Nine compounds were quantified by LC–DAD simultaneously. For chromatographic analysis, a Merck C18 column (5 μm, 250 mm × 4.6 mm) with a compatible guard column (C18, 5 μm, 7.5 mm × 4.6 mm) was used. The mobile phase consisted of (A) 0.1% aqueous formic acid and (B) acetonitrile. Ten microliters of each sample solution were injected and eluted at a flow rate of 1 mL min-1. The column temperature was maintained at 30 °C. The validation of this method proved good linear regression ( r 2 > 0.9992) within the test ranges, desirable repeatability with overall intra- and inter-day variations of less than 4.91% and well acceptable recoveries varied between 90.91 and 96.73% while the RSDs were below 3.23% ( n = 3). The proposed method was successfully applied to the quantification of the nine components in sixteen samples from different localities in China. Abstract: A sensitive and reliable high-performance liquid chromatography–mass spectrometry (HPLC–MS) was developed and validated for simultaneous quantification of five main bioactive components, i.e., calycosin-7-O-β-d-glucoside, ononin, astragaloside IV, astragaloside I and ferulic acid in rat plasma after oral administration of Danggui Buxue Tang (DBT) extract. Plasma samples were extracted with solid-phase extraction (SPE) separated on an Inertsil ZORBAX C18 column and detected by MS with electrospray ionization (ESI) interface in negative selective ion monitoring (SIM) mode. Calibration curves offered linear ranges of two orders of magnitude with r 2 >0.99. The method had the lower limit quantification of 0.55, 0.46, 1.07, 1.12 and 4.6ng/mL for calycosin-7-O-β-d-glucoside, ononin, astragaloside IV, astragaloside I and ferulic acid, respectively, with precision less than 10%. The RSD of intra- and inter-day variations ranged from 2.10% to 6.19% and 2.37% to 6.72%. A cell extraction coupled with high-performance liquid chromatography (HPLC) analysis has been developed to screen the potential bioactive components in combined prescription of Danggui Buxue decoction. The method was validated by using HL-7702 cells, RAW 264.7 cells and Caco-2 cell extraction. According to the hypothesis that, when cells are incubated together with the extract of traditional Chinese medicines (TCMs), the potential bioactive compounds in the extract should selectively combine with the cells, cell-combining compounds were analyzed by HPLC-diode array-evaporative light scattering detectors. Their structures were elucidated in comparison with available reference compounds, and further confirmed by HPLC-electrospray ionization time-of-flight mass spectrometry with accurate mass measurement. The results demonstrated that nine compounds were combined with the HL-7702 cells, seven with RAW 264.7 cells, and thirteen with the Caco-2 cell line. In view of the two key steps of drugs action, absorption by intestinal epithelium cells and interaction with target cells, this rapid and reliable method could be utilized to predict the bioactive constituents in TCMs, and it was in agreement with the characteristics of combined prescriptions of TCMs as multi-components, multi-target sites and multi-channel actions. A novel method, using high-performance liquid chromatography combined with principal component analysis, was developed for the quality evaluation of danshen through simultaneous determination of seven components, namely danshensu, protocatechuic acid, protocatechuic aldehyde, salvianolic acid B, tanshinone I, cryptotanshinone and tanshinone IIA. These seven components were simultaneously separated on a Zorbax SB-C18 column. The mobile phase consisted of 0.05% phosphoric acid water and methanol:acetonitrile (1:1) with a gradient elution, and the detection wavelength was set at 280 nm. Thirty samples of danshen and its substitutes from different sources were investigated by the established method. The results showed that the content of each analyte varied considerably in different danshen samples. Among the seven components tested, salvianolic acid B, tanshinone IIA, cryptotanshinone, tanshinone I, danshensu and protocatechuic aldehyde were proved suitable and representative as chemical markers for the quality control of danshen except for protocatechuic acid. Moreover, principal component analysis was used for the similarity evaluation of different samples, and it could be straightforward and reliable to differentiate danshen samples of different origins. In conclusion, simultaneous quantification of multiple components by high-performance liquid chromatography combined with principal component analysis would be a better strategy for the quality evaluation of danshen. Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) is a label-free tissue-based throughput analytical technology for the qualitative and quantitative spatial analyses of biomolecules, such as proteins and peptides, lipids, drug molecules and/or their metabolites. It has proven to be a useful analytical platform in new drug discovery, disease diagnosis and prognosis, and evaluation of drug efficacy. We provide a brief and succinct overview of the role of these thematic areas in this review. We then draw the attention of the readgsers to the strengths and weaknesses of MALDI-MSI in global omics particularly, metabolomics, lipidomics and proteomics. By so doing, we throw light on the scientific areas that need redress in order to fully realize the potential of this analytical platform. • MALDI-MSI has gradually gained worldwide acceptance as credible analytical platform. • This review outlines the applicability of MALDI-MSI in drug discovery and development. • The review also highlights the pros and cons of MALDI-MSI in omics techniques. • MALDI-MSI as sole analytical platform might be inadequate to capture global omics data. • There is the need for multi-omics determinations to obtain thorough global omics data. Glucagon promotes hepatic gluconeogenesis and maintains whole-body glucose levels during fasting. The regulatory factors that are involved in fasting glucagon response are not well understood. Here we report a role of p52, a key activator of the noncanonical nuclear factor-kappaB signaling, in hepatic glucagon response. We show that p52 is activated in livers of HFD-fed and glucagon-challenged mice. Knockdown of p52 lowers glucagon-stimulated hyperglycemia, while p52 overexpression augments glucagon response. Mechanistically, p52 binds to phosphodiesterase 4B promoter to inhibit its transcription and promotes cAMP accumulation, thus augmenting the glucagon response through cAMP/PKA signaling. The anti-diabetic drug metformin and ginsenoside Rb1 lower blood glucose at least in part by inhibiting p52 activation. Our findings reveal that p52 mediates glucagon-triggered hepatic gluconeogenesis and suggests that pharmacological intervention to prevent p52 processing is a potential therapeutic strategy for diabetes. Elevated glucagon level in obesity and diabetes promotes hepatic glucose production and hyperglycemia. Nonalcoholic fatty liver disease (NAFLD) is a global epidemic, and there is no standard and efficient therapy for it. Chitosan oligosaccharide (COS) is widely known to have various biological effects, and in this study we aimed to evaluate the liver-protective effect in diet-induced obese mice for an enzymatically digested product of COS called COS23 which is mainly composed of dimers and trimers. An integrated analysis of the lipidome and gut microbiome were performed to assess the effects of COS23 on lipids in plasma and the liver as well as on intestinal microbiota. Our results revealed that COS23 obviously attenuated hepatic steatosis and ameliorated liver injury in diet-induced obese mice. The hepatic toxic lipids—especially triglycerides (TGs) and free fatty acids (FFAs)—were decreased dramatically after COS23 treatment. COS23 regulated lipid-related pathways, especially inhibiting the expressions of FFA-synthesis-related genes and inflammation-related genes. Furthermore, COS23 could alter lipid profiles in plasma. More importantly, COS23 also decreased the abundance of Mucispirillum and increased the abundance of Coprococcus in gut microbiota and protected the intestinal barrier by up-regulating the expression of tight-junction-related genes. Aside from its multiple medicinal uses, the fruit of Xylopia aethiopica is widely used in Africa as food. Herein, we characterize the protein profiles, mineral content and bioactive phytochemical composition of the seeds of this plant sourced in Ghana and Nigeria. Using label-free proteomics, a total of 677 proteins were identified, with 260 found in the Ghana-sourced samples while 608 proteins were detected in the samples from Nigeria. However, 114 proteins were common between the samples from the two countries, among which 48 were significantly changed. Bioinformatics and functional analyses revealed that the differential levels of the proteins were mainly linked to pathways involved amino acids metabolism and biosynthesis. The significantly changed proteins related mainly to catalytic activity and carbon metabolism. The samples from Nigeria also exhibited superior qualities in terms of their antioxidant effects, and total phenolic and flavonoid content. Finally, only the content of Na varied to a statistically significant level. This study lends support to its culinary use and hints towards the impact of location of cultivation on the quality of the seeds. Background and Purpose: Hepatic mitochondrial pyruvate carrier (MPC) transports pyruvate into mitochondria. This study investigated the involvement of MPC1 in hepatic glucagon response, in order to identify a possible pharmacological intervention.Experimental Approach: The correlation between hepatic glucagon response and MPC1 induction was investigated in fasted mice and primary hepatocytes. The effects of ginsenoside Rb1 on MPC1 function were observed.Key Results: Glucagon challenge raised blood glucose with hepatic MPC1 induction, and inhibition of MPC induction coincided with a reduced rise in blood glucose. cAMP-responsive element-binding protein (CREB) knockdown blocked glucagon-induced MPC1 expression, while CREB overexpression increased MPC1 expression. Luciferase reporter, chromatin immunoprecipitation assay, and promoter mutation confirmed that CREB increased MPC1 transcription through gene promoter induction. CREB regulated transcription co-activator 2 nuclear translocation was also required for CREB to promote MPC1 induction. Glucagon shifted mitochondrial pyruvate towards carboxylation for gluconeogenesis via the opposite regulation of pyruvate dehydrogenase and carboxylase with respect to MPC1 induction. MPC1 induction was necessary for glucagon to promote pyruvate-driven hepatic glucose production (HGP), but glucagon failed to influence HGP from other gluconeogenic substrates routed into the tricarboxylic acid cycle, independent of MPC. Rb1 blocked cAMP signalling by inhibiting AC activity and deactivated CREB by dephosphorylation, possibly contributing to inhibiting MPC1 induction to reduce HGP.Conclusions and Implications: CREB transcriptionally up-regulates MPC1 to provide pyruvate for gluconeogenesis. Rb1 reduced cAMP formation which consequently reduced CREB-mediated MPC1 induction and thereby might contribute to limiting pyruvate-dependent HGP. Metabolic dysregulations have emerged as a major mediator of cardiovascular disorders and fibrotic diseases. Metabolic reprogramming contributes a lot to cardiac fibroblast activation and cardiac fibrosis post-myocardial infarction (MI), yet the mechanism remains incompletely understood. Our work aimed to determine whether or not glycolytic reprogramming, regulated by phosphofructokinase-2/fructose-2,6-bisphosphatase 3 (PFKFB3), is a therapeutic target for alleviating post-MI cardiac fibrosis. Here, we showed that cardiac fibroblasts displayed cell energy phenotype toward augmented glycolysis in response to transforming growth factor-beta 1 (TGF-β1), evidenced by significant extracellular acidification rate (ECAR) increase and lactate accumulation. The expression of glycolytic enzyme PFKFB3, a master activator of glycolysis, was up-regulated in TGF-β1-treated cardiac fibroblasts and in cardiac fibroblasts of post-MI mice. Pharmacological inhibition of PFKFB3 by 3PO diminished TGF-β1-mediated profibrotic phenotypes, attenuated cardiac fibrosis, and preserved cardiac functions in post-MI mice. Meanwhile, the genetic inhibition of PFKFB3 decreased the cardiac fibroblast activation and reversed the differentiated phenotypes in vitro and in vivo. Mechanistically, we identified deubiquitinase OTUD4 as a new binding protein of PFKFB3, and their interaction blocked PFKFB3 degradation via OTUD4-mediated deubiquitylation. Taken together, this work characterized a key role for PFKFB3 in cardiac fibroblast activation and suggested that inhibiting PFKFB3-involved glycolysis is an alternative way to alleviate post-MI cardiac fibrosis. A simple, efficient and versatile biosensing platform capable of the multiplexed detection for glucose, lactate and choline was developed by the integration of bipolar electrochemistry and electrochemiluminescence (ECL) imaging. The sensing bipolar electrodes (BPEs) were simply modified via a one-step method adaptable to different enzymes. The biorecognition event happening between the substrate and the corresponding enzyme could be directly reported by the ECL emitted on the same pole from luminol and in situ generated H 2 O 2 . Under optimized conditions, the BPEs array was successfully applied for the determination of glucose, lactate and choline in the ranges of 0.01–1 mM, 0.01–1 mM and 0.02–5 mM, with the LOD of 7.57 μM, 8.25 μM and 43.19 μM, respectively. Owing to the improved stability of in situ generated H 2 O 2 , a whole series of analytes testing could be completed in the same BPE biochip. Subsequently, an array chip consisting of nine BPEs enabled the concomitant detection of glucose, lactate and choline, demonstrating the capability for multifunctional detection of biomolecules.
