Abstract

Nonalcoholic fatty liver disease (NAFLD) is common and partially heritable and has no effective treatments. We carried out a genome-wide association study (GWAS) meta-analysis of imaging (n= 66,814) and diagnostic code (3,584 cases versus 621,081 controls) measured NAFLD across diverse ancestries. We identified NAFLD-associated variants at torsin family 1 member B (TOR1B), fat mass and obesity associated (FTO), cordon-bleu WH2 repeat protein like 1 (COBLL1)/growth factor receptor-bound protein 14 (GRB14), insulin receptor (INSR), sterol regulatory element-binding transcription factor 1 (SREBF1) and patatin-like phospholipase domain-containing protein 2 (PNPLA2), as well as validated NAFLD-associated variants at patatin-like phospholipase domain-containing protein 3 (PNPLA3), transmembrane 6 superfamily 2 (TM6SF2), apolipoprotein E (APOE), glucokinase regulator (GCKR), tribbles homolog 1 (TRIB1), glycerol-3-phosphate acyltransferase (GPAM), mitochondrial amidoxime-reducing component 1 (MARC1), microsomal triglyceride transfer protein large subunit (MTTP),alcohol dehydrogenase 1B (ADH1B), transmembrane channel like 4 (TMC4)/membrane-bound O-acyltransferase domain containing 7 (MBOAT7) and receptor-type tyrosine-protein phosphatase δ (PTPRD). Implicated genes highlight mitochondrial, cholesterol and de novo lipogenesis as causally contributing to NAFLD predisposition. Phenome-wide association study (PheWAS) analyses suggest at least seven subtypes of NAFLD. Individuals in the top 10% and 1% of genetic risk have a 2.5-fold to 6-fold increased risk of NAFLD, cirrhosis and hepatocellular carcinoma. These genetic variants identify subtypes of NAFLD, improve estimates of disease risk and can guide the development of targeted therapeutics.

306. Ductile Ni-based bulk metallic glasses at room temperature

Abstract

In this work, a series of new Ni76-xCoxNb4P16B4(x = 5, 10, 15, in at.%) bulk metallic glasses (BMGs) with the maximum diameter in the range of 1.2–1.6 mm were prepared by combining fluxing and J-quenching technology. The effects of Co content on glass forming ability (GFA), thermal stability, mechanical properties, serrated-flow behaviors and corrosion performance of the present Ni-based BMGs were systematically investigated. The ranges of Tgand ΔTxare 664–668 K and 21–31 K, respectively. Uniaxial compressive tests revealed that the present Ni-based BMGs have the compressive plastic strain (εp), yield strength (δy), and fracture strength (δf) that range from 16% to 27%, 2.1–2.6 GPa, and 2.2–3.6 GPa, respectively. The unprecedented mechanical properties originate from more icosahedral-like clusters (ILCs) and crystal-like clusters (CLCs) in the present Ni-based BMGs. It is found that the serrated-flow behaviors of the present Ni-based BMGs transition self-organized critical state (SOCs) into chaotic state (Cs) as plasticity decreases. The stick-slip dynamics were used to calculate the average shear-band velocity (ASBV), which shows an increasing trend as the Co content rises from 5 at.% to 15 at.%. Electrochemical measurements in 3.5 wt.% NaCl solution showed that the Ni71Co5Nb4P16B4BMG has a self-corrosion current density of 3.30 × 10−8A/cm2and significantly stronger corrosion resistance than austenite 316L and 304L stainless steel. The present Ni-based BMGs have excellent comprehensive performance, which has great potential for industrial applications.

Abstract

Radioactive waste sorting often faces an unstructured and locally radioactive working environment. At present, remote operation sorting has problems such as low sorting efficiency, greater difficulty in operation, longer training periods for personnel, and poor autonomous control capabilities. Based on the premise of improving the adaptability and autonomous operation ability of robots in an unstructured environment, this paper uses the dual deep Q learning algorithm to optimize the classic deep Q learning algorithm to improve training speed and improve sorting efficiency and stability. Secondly, the sorting algorithm model of deep reinforcement learning is used to determine the optimal behavior in this state. Set up multiple sets of simulations and physical experiments to verify the sorting method. The results show that the robotic arm can autonomously complete sorting tasks under complex conditions and can significantly improve work efficiency when pushing and grasping collaborative operations and will preferentially grasp objects with high radioactivity in the radioactive area.
