Carvone is one of the naturally occurring monoterpenes, the largest class of secondary metabolites in plants, and exists in two enantiomers, R-carvone (R-car) and S-car. The objective of this study was to investigate the antimicrobial activity of R-car and S-car with gentamicin (GET) against methicillin-resistant Staphylococcus aureus (MRSA). MRSA is a major human pathogen that causes serious problems, including hospital-acquired pneumonia, abscesses and surgical wound infections. Nosocomial MRSA infections often exhibit multidrug resistance. In the present study, antimicrobial susceptibility testing was performed with R-car, S-car and GET using the broth microdilution method. Minimal inhibitory concentration values for R- and S-car against six different strains of S. aureus ranged between 500 and 1,000 µg/ml. Anti-MRSA activity was evaluated using the checkerboard and time-kill assays to investigate the potential synergistic effects of different combinations of the carvone enantiomers and GET. R-car plus S-car, R-car plus GET and S-car plus GET exhibited significant synergistic activity against MRSA. These findings suggest that the single-agent anti-MRSA activities of R-car, S-car and GET are effectively increased through combination therapy. The effect of the platinum and aluminum interface reaction on the capacitor properties were investigated in 64 k ferroelectric random access memory (RAM). Aluminum diffused into platinum and reacted to form an intermetallic compound during the post-anneal when aluminum had direct contact with the top platinum layer of the ferroelectric capacitor. The Al/Pt/PZT contact was changed into an Al–Pt and lead-zirconate-titanate contact by the reaction. As a result, ferroelectric properties degraded and the leakage current of the top electrode and lead-zirconate-tianate contact started to increase after annealing at 300°C. The capacitor structure was destroyed by the volume expansion due to the Al/Pt reaction at 400°C. To prevent this reaction, a TiN layer was introduced as the barrier layer. This contact scheme showed no breakdown of the ferroelectric capacitor up to 400°C, which indicated that TiN acts as a good diffusion barrier for the double-metal process.

Penetration depth of Pd/Ge/Ti/Au ohmic contact to high-low-doped n-GaAs has been investigated using cross-sectional transmission electron microscopy, and the results are used to interpret the electrical properties. The ohmic metals do not penetrate into the GaAs substrate at 300° C, but penetrate deeper than 1000 Å into GaAs at 380° C. This is due to fast indiffusion of Au toward the GaAs substrate through the grain boundary of the PdGe compound, followed by formation of AuGa through reaction with GaAs. The deep penetration into GaAs at 380° C results in the lowest contact resistance of 0.43 Ω mm due to direct contact of ohmic metals with the buried high-doped GaAs layer.

307. Polymer Electrolyte Membranes Derived from New Sulfone Monomers with Pendent Sulfonic Acid Groupsâ.

New monomers containing two or four pendent phenyl groups were synthesized by bromination of bis(4-fluorophenyl) sulfone, followed by Suzuki coupling with benzeneboronic acid. The resulting monomers were converted to the corresponding sulfonated monomers having two or four pendent sulfonic acid groups, predominately at the p-phenyl position. Aromatic nucleophilic substitution (SNAr) polycondensation using the di- and tetrasulfonated monomers provided sulfonated poly(arylene ether sulfone) copolymers S2-PAES-xxand S4-PAES-xx, respectively, where xxrefers to the molar ratio of the sulfonated to non-sulfonated pendent phenyl monomer. Copoly(arylene ether sulfone)s based on the corresponding non-sulfonated monomers were also synthesized for a parallel study on postpolymerization sulfonation of these copolymers. Postsulfonation occurred predominately at the para-pendent phenyl site, and the reactions were complete within a short time (about 30 min), without evidence of chain degradation. Flexible and tough membranes having high mechanical strength were obtained by solution casting of all four series of copolymers. The copolymers with two or four pendent sulfonic acid groups had high proton conductivities in the range of 44â142 mS/cm for S2-PAES-xxand 51â158 mS/cm for S4-PAES-xxat room temperature, respectively. The methanol permeabilities of these copolymers were in the range of 0.8 Ã 10â8â15.0 Ã 10â7cm2/s, which is lower than Nafion (16.7 Ã 10â7cm2/s). The S4-PAES-xxmembranes displayed better properties (lower water uptake and higher proton conductivities) than the S2-PAES-xxmembranes, which can be attributed to the more blocky architecture of the sulfonic acid groups in the S4 membranes. A combination of high proton conductivities, low water uptake, and low methanol permeabilities for some of the obtained copolymers indicated that they are good candidate materials for proton exchange membrane in fuel cell applications. Platycodon grandiflorum (Campanulaceae) is used as traditional medicine in Asian countries. In Korean traditional medicine, Platycodon root has been widely used since ancient times as a traditional drug to treat cold, cough and asthma. However, its effects on bone marrow-derived mast cell (BMMC)-mediated allergy and inflammation mechanisms remain unknown. In this study, the biological effect of Platycodon root ethanol extract (PE) was evaluated in BMMC after induction of allergic mediators by phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187) stimulation. The effect of PE on the production of several allergic mediators, such as interleukin-6 (IL-6), prostaglandin D
2 (PGD2 ), leukotriene C4 (LTC4 ), β-Hexosaminidase (β-Hex) and cyclooxygenase-2 (COX-2) protein, was investigated. The results demonstrate that PE inhibits PMA + A23187 induced production of IL-6, PGD2 , LTC4 , β-Hexosaminidase and COX-2 protein. The purpose of this study was to examine the relationship between the C(-1019)G polymorphism of the serotonin-1A receptor gene and eating behavior in female adolescents. A total of 204 post-menarche, adolescent women, aged 16–17 years, were recruited from two neighboring high schools in Seoul. Polymerase chain reaction (PCR) was used to isolate and examine the C(-1019)G polymorphism in the serotonin-1A receptor genes (rs6295) of all participants. The Bulimia Investigatory Test, Edinburgh (BITE) and the Eating Attitude Test-26 (EAT-26) were administered to all participants. The total score of the EAT-26 differed significantly among the three genotype groups [CC, CG, GG ( F = 4.844, p = 0.009)]. Both the EAT-26 ( F = 9.69, p = 0.002) and the BITE ( F = 5.22, p = 0.023) scores were higher in the participants who were G allele carriers than in the non-carrier group. The dieting subscale of the EAT-26 was higher among the G allele carriers ( F = 12.941, p < 0.001), and these results were maintained even after adjusting for depression and anxiety. These findings suggest that the C(-1019)G polymorphism in the 5-HT1A receptor gene is associated with disordered eating symptoms in Korean female adolescents. [ABSTRACT FROM AUTHOR]

310. Increased Cortisol after Stress is Associated with Variability in Response Time in ADHD Children.

Purpose: Children with attention-deficit/hyperactivity disorder (ADHD) often perform poorly during cognitive tests. We sought to evaluate cortisol as potential moderator of performance in mentally challenging tasks in children with ADHD. Materials and Methods: Ninety clinic-referred children with ADHD were studied. Cortisol contents in saliva were measured before and after administration of a continuous performance test (CPT). Results: Pre and post CPT cortisol levels were similar in 68 children. Children whose cortisol level increased after testing ( n = 22) displayed a significantly longer response time and increased response time variability scores as compared to children who did not display increase of cortisol after the CPT test. Even after controlling for the effects of response time and anxiety, the changes in cortisol levels were associated with effect on response time variability. Conclusion: The patients who showed an increased cortisol level after stress displayed a higher variability in response time than the patients who showed no change or a decreased cortisol level. The result of the current study suggests that stress- induced high norepinephrine (NE) release may accompany poorer attention performance in patients with ADHD. [ABSTRACT FROM AUTHOR]

Methyl gallate is a major component of Galla Rhois, as carvacrol is of oregano essential oils. Both have shown good antibacterial activity against intestinal bacteria. This study investigated the antibacterial activities of nalidixic acid in combination with methyl gallate and carvacrol against nalidixic acid resistant bacteria. The combined effect of nalidixic acid with methyl gallate and carvacrol was evaluated using the checkerboard method to obtain a fractional inhibitory concentration index. The results showed that the combinations of nalidixic acid + methyl gallate/carvacrol improved nalidixic acid resistant pathogenic bacteria inhibition with synergy or partial synergy activity. Thus, a strong bactericidal effect of the drug combinations was observed. In vitro data thus suggested that nalidixic acid combined with methyl gallate and carvacrol may be microbiologically beneficial, rather than antagonists. [ABSTRACT FROM AUTHOR]

312. Maternal Depression Predicts Maternal Use of Corporal Punishment in Children with Attention Deficit/Hyperactivity Disorder.

Purpose: We sought to determine if maternal depression contributed to the use of corporal punishment in children with attention-deficit/hyperactivity disorder (ADHD). Patients and Methods: The data were gathered through chart review of clinic-referred children with ADHD and their mothers who were evaluated at a psychiatric clinic located in a large academic medical center in Seoul, Korea. Daily records kept by parents and 13 items from the Physical Assault of the Parent-Child Conflict Tactics Scales (CTSPC) were used to assess corporal punishment. Ninety-one children with ADHD and their mothers were included in this study. Results: Mothers who used corporal punishment showed significantly higher scores on the Beck Depression Inventory (t = - 2.952, df = 89, p < 0.01) than mothers who did not. Moreover, maternal depression contributed to the use of corporal punishment in ADHD children (Nagelkerke R

Purpose: To examine the incidences and patterns of injuries that required medical attention among Asian football players. Methods: A total of 411 Asian football players at both senior and youth (U-20) elite levels were observed during 50 international matches. Independent injury observers and team doctors determined the occurrence of injuries and recorded the location, type, time, and circumstances of the injuries using a protocol sheet. Results: The overall injury frequency rate was 45.8 out of 1000 hours. As the tournaments progressed into the knockout stages, the incidence and severity of the injuries increased. The most common sites of injuries were the knees (18.5%), lower legs (17.3%), and ankles (14.2%). Although most injuries were diagnosed as contusions, the more serious injuries were those diagnosed as sprains (especially concerning the knee and ankle) or strains (thigh and back). Conclusions: The incidences of injuries to Asian football players were higher than those to European players, but the patterns of the injuries showed no major differences. Clinical Relevance: To develop an injury-prevention program, more solid and comprehensive data need to be collected to identify the risk factors and variables associated with higher incidences of injuries to Asian football players. Abstract Alcohol and cocaine (COC) are commonly co-used drugs that cause addiction and have harmful effects. Their abuse may threaten the health of the abuser and public safety by causing serious accidents or crimes. The recidivism rate of drug-related crimes closely correlates with alcoholism. Several incidences of alcohol consumption in combination with drug abuse have been reported. Here, liquid chromatography tandem mass spectrometric method was developed to simultaneously analyze ethyl glucuronide (EtG), a metabolite of ethanol; COC; cocaethylene (CE), an alcohol-derived metabolite of COC; and benzoylecgonine (BZE), a major metabolite of COC, to determine the concurrent use of alcohol with COC. For pre-treatment, ultracentrifugation (5 min, 50,000 g) and mixed-mode anion exchange solid-phase extraction were used to increase the recovery of target compounds and minimize the matrix effect of hair. The lower limits of quantification were: 7 pg/mg (EtG), 2 pg/mg (COC), 10 pg/mg (CE), and 1 pg/mg (BZE). The correlation coefficient (r) of the calibration curve within the quantified range of target compounds was ≥ 0.9978. The intra- and inter-day accuracies were − 6.1–9.7% and − 9.3–8.3%, and intra- and inter-day precisions were 0.5–10.3% and 0.6–14.4%, respectively. The recovery, matrix effect, process efficiency, and autosampler stability were 89.2–104.8%, 81.6–105.4%, 81.5–107.1%, and 96.6–109.7%, respectively. The novel analytical method was validated with hair samples from individuals suspected of alcohol and COC use, and the method could distinguish between independent and concurrent use. Based on the findings, the analytical approach developed in this study is anticipated to be valuable in drug and alcohol dependence tests that require the simultaneous detection of alcohol and COC abuse.

Estimating the measurement uncertainty (MU) is becoming increasingly mandatory in analytical toxicology. This study evaluates the uncertainty in the quantitative determination of urinary amphetamine (AP) and 4-hydroxyamphetamine (4HA) using a liquid chromatography–tandem mass spectrometry (LC–MS/MS) method based on the dilute-and-shoot approach. Urine sample dilution, preparation of calibrators, calibration curve, and method repeatability were identified as the sources of uncertainty. To evaluate the MU, the Guide to the Expression of Uncertainty in Measurement (GUM) approach and the Monte Carlo method (MCM) were compared using the R programming language. The MCM afforded a smaller coverage interval for both AP (94.83, 104.74) and 4HA (10.52, 12.14) than that produced by the GUM (AP (92.06, 107.41) and 4HA (10.21, 12.45)). The GUM approach offers an underestimated coverage interval for Type A evaluation, whereas the MCM provides an exact coverage interval under an abnormal probability distribution of the measurand. The MCM is useful in complex settings where the measurand is combined with numerous distributions because it is generated from the uncertainties of input quantities based on the propagation of the distribution. Therefore, the MCM is more practical than the GUM for evaluating the MU of urinary AP and 4HA concentrations using LC–MS/MS.

319. Association Between Working Hours and Depressive Symptoms Among Korean Employees.

Objectives Many studies have reported noticeable increases in the proportion of employees working either relatively short or relatively long hours. Such trends have been accompanied by an increasing concern that how much subjective mental well-being of employees would be influenced by their hours of work. The aim of this study was to investigate the association between work hours and clinically relevant depressive symptoms with demographic variables adjusted. Methods Participants were employees of a total of 56 private companies and local government organizations in Korea, aged 19 to 65 years. A self-report questionnaire that included items on working hour, job stress, levels of depression, and socio-demographic factors was administered to 15360 Korean employees, with 14477 valid responses. Hierarchical linear regression analyses, adjusted for sociodemographic factors, job related demographic factors, job stress, were used additionally to estimate the association between working hours and depressive scores. Results We found that working more than 40 hours per week correlated positively with the level of depressive symptoms after adjusting for demographic variables and the level of job stress. Furthermore, working 40 or fewer hours per week correlated negatively with the level of depressive symptoms. Being younger (β = -0.078, β = -0.099), being a female (β = 2.770, β = 1.268), and possessing a lower level of education (β = -0.315, β = -1.125) were significantly associated with higher level of depressive symptoms in all respondents. Conclusions Both of working excessively long or short hours is significantly associated with the prevalence of depressive symptoms. Establishing proper office hours for employees is critical to improving the quality of working conditions and maintaining good mental health in the workplace. [ABSTRACT FROM AUTHOR]

Abstract Numerous confidence estimation methods have been proposed for classification neural networks; however, this problem has not been well addressed for regression neural networks. That is, softmax layers are not available in regression networks and the interpretation of confidence becomes less clear. To alleviate these problems, a simple but effective method is proposed that computes the confidences of regression results. First, the confidence is considered as a scalar value representing relative error‐levels. Then, a mini‐batch based training method based on this interpretation is developed. Precisely, in each mini‐batch, desired outputs for confidence values are assigned by sorting current errors. Experimental results on the loose wheel nut detection problem as well as a simulated example have shown that the proposed method can be successfully applied to regression problems.

Prescriptions of Local Botanicals for Emergency Use (K. Hyang'yak Kugŭppang is the oldest medical text extant on the Korean Peninsula and known to have been compiled during the latter half of the Koryŏ dynasty (918--1392 ce). The key value of this work lies in the dissemination and praxis of medical knowledge. First, the author used annotations in order to record Koryŏ people's pronunciations of the names of medicinal ingredients and symptoms introduced in the main body of the text. In addition, he made use of actual empirical cases to enhance the persuasiveness of treatment methods and integrated medicine newly introduced from Song China (960--1279) into medicine familiarly used from before. Finally, he edited this text with a focus on important and simple yet efficacious treatment methods. The book continued to be used steadily following publication. It was additionally printed no fewer than twice by the government of the Chosŏn dynasty (1392--1910), which ousted Koryŏ, with its clinical usefulness heightened through the supplementation of explanations on medicinal ingredients use in these processes. In particular, the quotation of sentences from Prescriptions for Emergency Use in medical texts published by the Chosŏn government implies that the utility of the medical knowledge in this work was amply acknowledged. The intended readership of the medical information in Prescriptions for Emergency Use was the not the general populace who lived in the Korean Peninsula in the thirteenth-fourteenth centuries. They not only lacked the financial means to pay physicians but also were illiterate, so that they could not even read medical texts. In order for this work to be effective, it was necessary for it to address those who could read medical texts and put their contents into practice. In the end, the author of this book assumed scholar-gentry equipped with academic knowledge as its readers and sought to provide medical information tailored to their level and to realize medical service through them. Through this work, it is possible to see in a very concrete and vivid manner how medical knowledge was disseminated and, furthermore, how medical knowledge thus disseminated was put to use in an era when medical resources were insufficient. Curcumin, a natural polyphenolic flavonoid extracted from the rhizome of Curcuma longa L., was shown to possess superior potency to resensitize methicillin-resistant Staphylococcus aureus (MRSA) to antibiotics. Previous studies have shown the synergistic activity of curcumin with β-lactam and quinolone antibiotics. Further, to understand the anti-MRSA mechanism of curcumin, we investigated the potentiated effect of curcumin by its interaction in diverse conditions. The mechanism of anti-MRSA action of curcumin was analyzed by the viability assay in the presence of detergents, ATPase inhibitors and peptidoglycan (PGN) from S. aureus, and the PBP2a protein level was analyzed by western blotting. The morphological changes in the curcumin-treated MRSA strains were investigated by transmission electron microscopy (TEM). We analyzed increased susceptibility to MRSA isolates in the presence of curcumin. The optical densities at 600 nm (OD600) of the suspensions treated with the combinations of curcumin with triton X-100 and Tris were reduced to 63% and 59%, respectively, compared to curcumin without treatment. N,N'-dicyclohexylcarbodiimide (DCCD) and sodium azide (NaN3) were reduced to 94% and 55%, respectively. When peptidoglycan (PGN) from S. aureus was combined with curcumin, PGN (0-125 µg/mL) gradually blocked the antibacterial activity of curcumin (125 µg/mL); however, at a concentration of 125 µg/mL PGN, it did not completely block curcumin. Curcumin has a significant effect on the protein level of PBP2a. The TEM images of MRSA showed damage of the cell wall, disruption of the cytoplasmic contents, broken cell membrane and cell lysis after the treatment of curcumin. These data indicate a remarkable antibacterial effect of curcumin, with membrane permeability enhancers and ATPase inhibitors, and curcumin did not directly bind to PGN on the cell wall. Further, the antimicrobial action of curcumin involved in the PBP2a-mediated resistance mechanism was investigated. [ABSTRACT FROM AUTHOR]

Tectorigenin (TTR) is an O-methylated isoflavone derived from the rhizome of Belamacanda chinensis (L.) DC. It is known to perform a wide spectrum of biological activities such as antioxidant, anti-inflammatory, anti-tumor. The aim of this study is to examine the mechanism of antibacterial activity of TTR against methicillin-resistant Staphylococcus aureus (MRSA). The anti-MRSA activity of TTR was analyzed in combination assays with detergent, ATPase inhibitors, and peptidoglycan (PGN) derived from S. aureus. Transmission electron microscopy (TEM) was used to monitor survival characteristics and changes in S. aureus morphology. The MIC values of TTR against all the tested strains were 125 μg/mL. The OD(600) of each suspension treated with a combination of Triton x-100, DCCD, and NaN3 with TTR (1/10 × MIC) had been reduced from 68% to 80%, compared to the TTR alone. At a concentration of 125 μg/mL, PGN blocked antibacterial activity of TTR. This study indicates that anti-MRSA action of TTR is closely related to cytoplasmic membrane permeability and ABC transporter, and PGN at 125 μg/mL directly bind to and inhibit TTR at 62.5 μg/mL. Methicillin-resistant Staphylococcus aureus (MRSA) along with other resistant bacteria have become a significant social and clinical problem. Therefore, there is an urgent need to develop bioactive compounds from natural products as alternatives to the very few antibiotics that remain effective. Recently, the efflux mechanism has been identified as the main contributor to antibiotic resistance in bacteria. This study therefore aimed to evaluate tetrandrine (TET), an efflux pump inhibitor (EPI), as a potential antibiotic against MRSA. We investigated the antimicrobial activity of TET against 17 MRSA strains, of which 3 selected strains were studied in further detail using a time-kill assay. When these bacterial al strains (1 x 106 colony-forming units (cfu)/ml) were incubated with TET in a time-kill assay, log-scale bactericidal activity was observed, which lasted for 24 hr. In addition, TET exhibits a synergistic effect when combined with the multi-drug resistance (MDR)-efflux pump substrate ethidium bromide (EtBr). Structure-function studies of the antibiotic activity of TET in combination with EtBr may lead to the discovery of more effective efflux pump inhibitors. [ABSTRACT FROM AUTHOR]

Sanguinarine is a benzophenanthridine alkaloid derived from the root of Sanguinaria canadensis. It is known to perform a wide spectrum of biological activities. The aim of this study is to examine the antimicrobial actions of sanguinarine against methicillin-resistant Staphylococcus aureus (MRSA). Sanguinarine antimicrobial activity was assessed by broth dilution method; its mechanism of action was investigated by bacteriolysis, detergent or ATPase inhibitors and transmission electron microscopy were used to monitor the survival characteristics and the changes in bacteria morphology. The activity of sanguinarine against MRSA strains ranged from 3.12 to 6.25 μg/ml, while the minimum inhibitory concentrations of the two reference strains are 3.12 μg/ml and 1.56 μg/ml. The treatment of the cells with sanguinarine induced the release of membrane-bound cell wall autolytic enzymes, which eventually resulted in lysis of the cell. The OD600s of the suspensions treated with the combination of Tris-(hydroxymethyl) aminomethane and Triton X-100 with sanguinarine were reduced to 40% and 8%, respectively. Transmission electron microsco py of MRSA treated with sanguinarine showed alterations in septa formation. The predisposition of lysis and the altered morphology seen by transmission electron microscopy suggest that sanguinarine compromises the cytoplasmic membrane. [ABSTRACT FROM AUTHOR]
