Ferroptosis is primarily triggered by a failure of the glutathione (GSH)-glutathione peroxidase 4 (GPX4) reductive system and associated overwhelming lipid peroxidation, in which enzymes regulating polyunsaturated fatty acid (PUFA) metabolism, and in particular acyl-CoA synthetase long chain family member 4 (ACSL4), are central. Here, we found that exogenous oxygen radicals generated by photodynamic therapy (PDT) can directly peroxidize PUFAs and initiate lipid autoxidation, coinciding with cellular GSH depletion. Different from canonical ferroptosis induced by RSL3 or erastin, PDT-initiated lipid peroxidation and ferroptotis-like cell death is independent of lipoxygenase (ALOXs) and ACSL4. Especially, this form of cell death modality can be triggered in malignant cells insensitive to or acquired resistance to canonical ferroptosis inducers. We also observed a distinct iron metabolism pathway in this PDT-triggered cell death modality, in which cytosolic labile iron is decreased probably due to its relocation to mitochondria. Inhibition of the mitochondrial Ca2+ and Fe2+ uniporter (MCU) effectively prevented PDT-triggered lipid peroxidation and subsequent cell death. Therefore, we tentatively term this distinct ferroptosis-like cell death as liperoptosis. Moreover, using the clinically approved photosensitizer Verteporfin, PDT inhibited tumor growth through inducing prevailing ferroptosis-like cell death in a mouse xenograft model. With its site-specific advantages, these findings highlight the value of using PDT to trigger lipid peroxidation and ferroptosis-like cell death in vivo, and will benefit exploring the exact molecular mechanism of immunological effects of PDT in cancer treatment.

As the first line of defense against intestinal bacteria and toxins, intestinal epithelial cells are always exposed to bacteria or lipopolysaccharide (LPS), whereas pathogenic bacteria or LPS can cause intestinal epithelial cell damage. Previous studies have shown that konjac mannan oligosaccharides (KMOS) have a positive effect on maintaining intestinal integrity, and Bacillus subtilis (BS) can promote the barrier effect of the intestine. However, it is still unknown whether KMOS and BS have a synergistic protective effect on the intestines. In this study, we used the LPS-induced Caco-2 cell injury model and mouse intestinal injury model to study the synergistic effects of KMOS and BS. Compared with KMOS or BS alone, co-treatment with KMOS and BS significantly enhanced the activity and antioxidant capacity of Caco-2 cell, protected mouse liver and ileum from LPS-induced oxidative damage, and repaired tight junction and mucus barrier damage by up-regulating the expression of Claudin-1, ZO-1 and MUC-2. Our results demonstrate that the combination of KMOS and BS has a synergistic repair effect on inflammatory and oxidative damage of Caco-2 cells and aIIeviates LPS-induced acute intestinal injury in mice.

Significant progress has been made on sorghum transformation in the last decades; however, the transformation process has been constrained by the availability of immature embryos because most of the researchers have utilized immature embryos as favorable explants. Although immature embryos have been proven to be optimal for tissue culture and transformation, isolation of immature embryos is time-consuming, labor-intensive, and limited by warm weather. In this study, we developed an efficient genetic transformation system using mature seeds as explants. The nptII and gus gene, used as the selective marker and report gene respectively, have been co-transformed by particle bombardment. After optimization of tissue culture, the G418 concentration, and transgenic, the average transformation frequency at 13.33% was achieved routinely. The transgenic events and transgene copy numbers were determined by PCR and RT-PCR, respectively. The geneticin selection and GUS staining on T1 seedlings confirmed that the transgenic plants were heritable. Our results demonstrated that the efficient sorghum transformation system has been established using mature seeds as explants. This transformation system will promote sorghum research on genetic engineering and genome editing without seasonal weather conditions restriction and explant resources restriction.

Keynote paper presented at the International Leucaena Conference, 1‒3 November 2018, Brisbane, Queensland, Australia. A research program to develop sterile leucaena has commenced to enhance red-meat production in additional regions of Australia including Western Australia, Northern Territory and New South Wales, where growing seeded leucaena is not currently permitted or encouraged. In this study we report on the development of methodology using a mutagenizing agent, EMS (ethyl methanesulfonate), to cause mutations in the self-fertile commercial leucaena cultivar, Redlands. Several experiments to determine the optimum rate of EMS have been completed and first generation mutagenized plants (M0) established in the field at Redlands Research Station, Cleveland, Queensland, Australia. An EMS concentration of 0.35% applied to germination paper proved the best method to achieve a target emergence percentage of 50%. To date, 27 of 179 mutagenized M0 seedless plants are considered to be putatively sterile. A further 1,200 M0 plants have been established in the field providing an even greater chance of identifying sterile leucaena plants with the desired forage quality and psyllid-resistance attributes.

Because the Duffing equation is very sensitive to weak signals and noise immunity, the modified Duffing detection system is applied to detect the weak nonlinear signal in nonlinear ultrasonic experiment. Firstly, the Duffing equation is improved to construct detection models to detect the nonlinear ultrasonic signal wave mixing in experiment. Then, the maximum Lyapunov exponent of detection model with state change is calculated to evaluate the fatigue damage. When the collected nonlinear receiving signal in wave mixing experiment is substituted into four detection models, if the phase portraits of Duffing equation changes from the chaotic state into periodic state, the difference frequency signal at 3 MHz can be detected and this states the specimen has fatigue damage. In addition, the maximum Lyapunov exponent of detection model with state change is calculated and less than 0. The damage index is induced and it has a good corresponding relation with fatigue damage of metallic specimen, which the correspondence would be used to quantitatively evaluate the fatigue damage of specimen within a certain range. Therefore this research work is beneficial to extract the weak nonlinear signals for fatigue damage evaluation of metallic components.

In this study, 111 Cryptosporidium parvum IId isolates from several species of animals in China, Sweden, and Egypt were subtyped by multilocus sequence typing (MLST). One to eleven subtypes were detected at each of the 12 microsatellite, minisatellite, and single nucleotide polymorphism (SNP) loci, forming 25 MLST subtypes. Host-adaptation and significant geographical segregation were both observed in the MLST subtypes. A clonal population structure was seen in C. parvum IId isolates from China and Sweden. Three ancestral lineages and the same RPGR sequence were shared by these isolates examined. Therefore, the present genetic observations including the higher nucleotide diversity of C. parvum IId GP60 sequences in Western Asia, as well as the unique distribution of IId subtypes (almost exclusively found in Asia, Europe, and Egypt) and in combination with the domestication history of cattle, sheep, and goats, indicated that C. parvum IId subtypes were probably dispersed from Western Asia to other geographical regions. More population genetic structure studies involving various C. parvum subtype families using high-resolution tools are needed to better elucidate the origin and dissemination of C. parvum in the world. Background/Aims: Autophagy is a process of evolutionarily conservative degradation, which could maintain cellular homeostasis and cope with various types of stress. LncRNAs are considered as competing endogenous RNAs (ceRNAs) contributing to autophagy. GAS5 has been suggested as a new potential factor to mediate autophagy pathway and the underlying mechanism remains to be further confirmed. This study was taken to identify the effect of GAS5/miR-23a/ATG3 axis on autophagy and cell viability. Methods: The western blotting assay was used to detecte the protein levels of LC3, mTOR, Beclin-1, ATG3, ATG5-ATG12 complex and p62. The mRNA level of Pre-miR-23a, Pri-miR-23a, miR-23a, GAS5, LC3, mTOR and ATG3 were quantified by real-time RT-PCR. Dual-luciferase reporter assays were performed to confirm the direct binding of miR-23a and ATG3 or GAS5. Cell viability was evaluated by CCK-8 and flow cytometry. Results: We showed that miR-23a could directly suppress ATG3 expression in 293T cells, which suggested that ATG3 was identified as a target of miR-23a. MiR-23a mimics could restrain LC3 II, Beclin1 levles and ATG5-ATG12 complex formation. Meanwhile, miR-23a also increased the expression of mTOR and p62. Notably, there was a putative miR-23a-binding site in GAS5. MiR-23a overexpression might suppress the GAS5 expression, but the repressive effect was abolished by mutation of binding sites. Importantly, overexpression of GAS5 could inhibit the mature miR-23a and has no effect on miR-23a precursors. Knockdown of GAS5 suppressed the expression of LC3 II, ATG3 and ATG5-ATG12 complex formation, whereas p62 and mTOR levels were promoted. The further results showed that miR-23a overexpression and GAS5 inhibition both significantly suppressed cell viability and promoted the apoptosis rate following LPS stimulation, and knockdown of miR-23a exhibited the opposite effects. Conclusions: Our study revealed that down-regulation GAS5 attenuated cell viability and inhibited autophagy through ATG3-dependent autophagy by regulating miR-23a expression. The results suggested that GAS5/miR-23a/ATG3 axis might be a novel regulatory network contributing to a better understanding of regulation on autophagy program and cell viability.

This review article introduces the formation and development of stereology in China under the background of the development of international stereology. In the early 1970s, some stereological monographs and collections were introduced into China, and Chinese scholars began to understand, study and promote stereology knowledge. Meanwhile, the widespread use of image analysis systems has contributed to the spread of stereology in China. On the other hand, academic exchanges and personnel training have played a catalytic role in the formation of stereology in China. According to China National Knowledge Infrastructure (CNKI) statistics, the number and impact of Chinese papers in stereology continues to grow during the past 30 years. After in-depth discussion, Chinese scholars have adopted a broader definition of stereology. With economic development and technological progress, China has great potential to develop, promote and apply the stereological methods and the related technologies.

Objective To examine the real-world patterns of oral anticoagulant (OAC) therapy in patients with acute coronary syndrome (ACS) and atrial fibrillation (AF) in Southern China undergoing percutaneous coronary intervention (PCI) and determine the clinical characteristics associated with OAC prescription.Design A retrospective cohort study.Setting This study was conducted in the Shunde Hospital, Southern Medical University and the second hospital of Zhaoqing, China, from January 2013 to 31 December 2018.Participants Patients were aged ≥18 years, hospitalised for ACS and received PCI treatment.Outcome measures AF was diagnosed based on an ECG recording or a Holter monitor. Prescription of OACs and antiplatelets were determined from the discharge medication list.Results A total of 3612 patients with ACS were included: 286 (7.9%) were diagnosed with AF, including 45 (1.2%) with paroxysmal AF, 227 (6.3%) with persistent/permanent AF and 14 (0.4%) with unclassified AF. Although 95.5% of patients with AF were at high risk (CHA2DS2-VASc score ≥2) of stroke, only 21.7% of them were discharged on OACs (10.5% received warfarin and 11.2% received non-vitamin K antagonist OACs). Lepus yarkandensis specifically lives in arid climate with rare precipitation of Tarim Basin in western China. Aquaporins (AQPs) are a family of channel proteins that facilitate water transportation across cell membranes. Kidney AQPs play vital roles in renal tubule water permeability and maintenance of body water homeostasis. This study aimed to investigate whether kidney AQPs exhibit higher expression in arid-desert living animals. Immunohistochemistry results revealed localization of AQP1 to the capillary endothelial cells in glomerulus and epithelial cells in proximal tubule and descending thin limbs, AQP2 to the apical plasma membrane of principal cells in the cortical collecting duct (CCD), outer medullary collecting duct (OMCD), and IMCD cells in the initial inner medullary collecting duct (IMCD1) and middle IMCD (IMCD2), and AQP3 and AQP4 to the basolateral plasma membrane of principal cells and IMCD cells in CCD, OMCD, IMCD1, and IMCD2 in L. yarkandensis kidneys. Quantitative real-time PCR analysis showed higher mRNA levels of AQP1, AQP2, AQP3, and AQP4 in L. yarkandensis kidneys compared with Oryctolagus cuniculus. Similar results were obtained by western blotting. Our results suggested that higher expression levels of AQP1, AQP2, AQP3, and AQP4 in L. yarkandensis kidneys favored for drawing more water from the tubular fluid.

Abstract Metal–salen complexes are widely used as catalysts in numerous fundamental organic transformation reactions. Here, CoFe hydroxide/carbon nanohybrid is reported as an efficient oxygen evolution electrocatalyst derived from the in situ formed molecular Fe–salen complexes and Co2+ ions at a low temperature of 160 °C. It has been evidenced that Fe–salen as a molecular precursor facilitates the confined‐growth of metal hydroxides, while Co2+ plays a critical role in catalyzing the transformation of organic ligand into nanocarbons and constitutes an essential component for CoFe hydroxide. The resulting Co1.2Fe/C hybrid material requires an overpotential of 260 mV at a current density of 10 mA cm−2 with high durability. The high activity is contributed to uniform distribution of CoFe hydroxides on carbon layer and excellent electron conductivity caused by intimate contact between metal and nanocarbon. Given the diversity of molecular precursors, these results represent a promising approach to high‐performance carbon‐based water splitting catalysts.

Plants are sessile organisms requiring mechanisms that enable them to balance water supply and demand in dry environments. Demand (D) is largely driven by canopy size (transpirational leaf area), although differences in transpiration per unit leaf area also occur. Supply (S) is primarily driven by water capture via the root system. Drought stress can be defined as the situation where supply of water cannot meet demand of the crop, such that water availability is the limiting factor for biomass accumulation. Under such conditions, plants will need to reduce D in order to meet the limited S, access more water to increase S, or increase the efficiency with which water is utilised. We used sorghum, a model C4 crop species, to demonstrate how the stay-green trait can modulate canopy development and root architecture to enhance adaptation. We show how stay-green positively impacts the balance between S and D under post-flowering drought, including insights at the molecular level. We provide examples of how canopy and root traits impact the S/D balance in other cereals under water limitation. For example, on the supply side, the extent of genetic variation for root angle (RA) has been evaluated in sorghum, wheat and barley, and genomic regions associated with RA have been mapped. Furthermore, the relationship between RA and grain yield has been explored in barley and sorghum field trials. The capacity to manipulate components of S and D to optimise the S/D balance should assist crop improvement programs to develop enhanced ideotypes for dry environments.

This study evaluated the transformation of a Carrière forest into a near-natural forest after 60 years of natural development. The structure and soil characteristics of planted forest, the near-natural forest (coniferous-broadleaved mixed forest), and secondary forest ( Fisch. ex Ledeb. forest) were compared. Tree, shrub and herb species diversity of the mixed and forests was higher than that of the planted forest. Examination of soil characteristics revealed that compared to the pure pine forest, nitrogen (N) and phosphorus (P) concentrations of the mixed and forests increased in the forest floor and soil, but total carbon (C) concentration decreased in the forest floor, countered by increases in the soil. Furthermore, soil cation exchange capacity (CEC) and pH in the forest increased when deciduous broadleaved species were present. Total microbial biomass and bacterial biomass in the soils were greatest in the forest, followed by the mixed, and then the forests. However, fungal biomass did not significantly differ among the three forests. Overall, the findings of this study suggest that different forest types can affect soil microbial biomass and community structure. To dynamically monitor the horizontal well, we studied the oil−water two-phase water holdup detection method based on transmission lines, and designed a micro-sensor and a sensor-array water holdup detection tool. We modeled the relationship of the dielectric constant of the transmission line filling medium and the amplitude and phase shift of the electromagnetic wave signal on the transmission line by using the time-domain analysis. We proposed a novel method to measure the water holdup of oil−water mixtures based on the phase shift of signals on the conical spiral transmission line. Furthermore, we simulated and optimized the structural parameters by software simulation, and developed a small conical spiral water holdup sensor suitable for arraying. The single sensor with detection circuits can achieve the full scale (water holdup from 0% to 100%) measurement with resolution better than 3%. On this basis, 12 sensors are used to develop a clock-like sensor-array water holdup detection tool, realizing the array detection of the distribution of the cross-section medium in horizontal wells.

Serial sectioning technique provides plenty of quantitative geometric information of the microstructure analyzed, including those unavailable from stereology with one- and two-dimensional probes. This may be why it used to be and is being continuously served as one of the most common and invaluable methods to study the size and the size distribution, the topology and the distribution of topology parameters, and even the shape of three-dimensional space filling grains or cells. On the other hand, requiring tedious lab work, the method is also very time and energy consuming, most often only less than one hundred grains per sample were sampled and measured in almost all reported practice. Thus, a question is often asked: for typical microstructures in engineering materials, are so many grains or cells sampled adequate to obtain reliable results from this technique? To answer this question, experimental data of 1292 contiguous austenite grains in a low-carbon steel specimen obtained from the serial sectioning analysis are presented in this paper, which demonstrates the effect of sampling on the measurement of various parameters of grain size distribution and of the grain topology distribution. The result provides one of rules of thumb for grain stereology of similar microstructures.

The conventional serial sectioning analysis and a set of modern stereological methods, including disector, selector, point-sampled intercepts, point-sampled area, and their combinations, have been used in this paper to measure the grain size, grain size distribution, topological parameters and their distributions in a spacefilling single-phase grain structure of steel. The results from different methods are compared and used to evaluate the methods quantitatively, based on which some suggestions will be given for selection of experimental methods in materials stereology research.

A 3D Monte Carlo technique has been used to simulate a complex process of recrystallization in a material containing precipitates after graded deformation. By the simulation model, the inhomogeneous deformed microstructure and its evolution during subsequent annealing were directly displayed; moreover, the locally varying recrystallized microstructure was quantitatively analyzed. The simulation results were well verified by experiments with an aluminum alloy containing Al3Zr particles that had been subjected to deformation and annealing for recrystallization. In the stereological analysis of a non-random microstructure and its evolution kinetics, the visual simulation technique showed apparent advantages for sampling, detection and measuring.

The stability analysis of global asymptotic stability of neural networks of neutral type with both discrete interval delays and general activation functions is discussed. New delay-dependent conditions are obtained by using more general Lyapunov-Krasovskii functionals. Meanwhile, these conditions are expressed in terms of a linear matrix inequality (LMI) and can be verified using the MATLAB LMI toolbox. Numerical examples are used to illustrate the effectiveness of the proposed approach.
