Abstract The magnetic microrobots actuated by an external magnetic field can access distant, enclosed, and small spaces under fuel‐free conditions, which is apromising technology for manipulation and delivery under microenvironment; however, the complicated fabrication method limits their applications. Herein, three techniques including melt electrospinning writing (MEW), micromolding, and skiving process are combined to successfully mass‐produce tadpole‐like magnetic polycaprolactone/Fe3O4 (PCL/Fe3O4) microrobot. Importantly, the tadpole‐like microrobots under an external magnetic field can achieve two locomotions: rolling mode and propulsion mode. The rolling motion can approach the working destination quickly with a speed of ≈2 mm s−1. The propulsion motion (0−340 µm s−1) can handle a microcargo. Such a simple and cost‐effective production method shows a great potential for scale‐up fabrication of advanced shape‐design, mass‐production, and multifunctionality microrobot.

Objective: This study aimed to investigate the molecular mechanism of tumor necrosis factor (TNF) superfamily-related genes and potential therapeutic drugs for glioblastoma multiforme (GBM) patients based on transcriptome and epigenome.Methods: Gene expression data, corresponding clinical data, and methylation data of GBM samples and normal samples in the TCGA-GBM and GTEx datasets were downloaded. The TNF-related genes were obtained, respectively, from two groups in the TCGA dataset. Then, the TNF-related differentially expressed genes (DEGs) were investigated between two groups, followed by enrichment analysis. Moreover, TNF superfamily-related gene expression and upstream methylation regulation were investigated to explore candidate genes and the prognostic model. Finally, the protein expression level of candidate genes was performed, followed by drug prediction analysis.Results: A total of 41 DEGs including 4 ligands, 18 receptors, and 19 downstream signaling molecules were revealed between two groups. These DEGs were mainly enriched in pathways like TNF signaling and functions like response to TNF. A total of 5 methylation site-regulated prognosis-related genes including TNF Receptor Superfamily Member (TNFRSF) 12A, TNFRSF11B, and CD40 were explored. The prognosis model constructed by 5 genes showed a well-prediction effect on the current dataset and verification dataset. Finally, drug prediction analysis showed that zoledronic acid (ZA)-TNFRSF11B was the unique drug–gene relation in both two databases.Conclusion: Methylation-driven gene TNFRSF12A might participate in the development of GBM via response to the TNF biological process and TNF signaling pathway and significantly associated with prognosis. ZA that targets TNFRSF11B expression might be a potential effective drug for clinical treatment of GBM.

The combination of Global Positioning System-Real Time Kinematic (GPS-RTK) and depth sounder is an important technical method of modern underwater topographic survey. In this paper, the combined technology was integrated with unmanned surface vehicle (USV) technology to construct an unmanned survey system suitable for underwater topographic survey in stilling basin, and it was applied to survey the underwater topography of the stilling basin of Tingzikou hydrojunction project. Based on the surveying and mapping data and 3D model, the trend of elevation change of the stilling basin including apron, end sill, antiscour section, and river convergence section was analyzed. The results show that, for the surface outlet stilling basin of Tingzikou, after flood, the structure is complete, the boundaries are clear, the water quality is good, and there is little sediment on the surface of apron from overflow dam section to end sill. Furthermore, no obvious sediment or structural abnormality has been found in the still basin. Affected by the bedrock and cofferdam cobble gravel sediment, the elevation of the antiscour section and the downstream convergence section of the surface outlet stilling basin increased significantly. For the bottom outlet stilling basin of Tingzikou, there is large sediment within a range of 20 m∼40 m from the bottom sill and the maximum height of it reaches 2.04 m, while there is no obvious sediment or structural abnormality in the remaining region of basin. The critical sedimentation height can effectively judge the safety grade of the sedimentation height in stilling basin and provide a reliable support for assessing the overall safety of stilling basin. The application of unmanned survey technology improves the accuracy and timeliness of underwater topography and sedimentation distribution of stilling basin, which has significant application research value and promotion significance.

Introduction Cases of the novel coronavirus disease (COVID-19) continue to spread around the world even one year after the declaration of a global pandemic. Those with weakened immune systems, due to immunosuppressive medications or disease, may be at higher risk of COVID-19. This includes individuals with autoimmune diseases, cancer, transplants, and dialysis patients. Assessing the risk and outcomes of COVID-19 in this population has been challenging. While administrative databases provide data with minimal selection and recall bias, clinical and behavioral data is lacking. To address this, we are collecting self-reported survey data from a randomly selected subsample with and without COVID-19, which will be linked to administrative health data, to better quantify the risk of COVID-19 infection associated with immunosuppression. Methods and analysis Using administrative and laboratory data from British Columbia (BC), Canada, we established a population-based case-control study of all individuals who tested positive for SARS-CoV-2. Each case was matched to 40 randomly selected individuals from two control groups: individuals who tested negative for SARS-CoV-2 (i.e., negative controls) and untested individuals from the general population (i.e., untested controls). We will contact 1000 individuals from each group to complete a survey co-designed with patient partners. A conditional logistic regression model will adjust for potential confounders and effect modifiers. We will examine the odds of COVID-19 infection according to immunosuppressive medication or disease type. To adjust for relevant confounders and effect modifiers not available in administrative data, the survey will include questions on behavioural variables that influence probability of being tested, acquiring COVID-19, and experiencing severe outcomes. Ethics and dissemination This study has received approval from the University of British Columbia Clinical Research Ethics Board [H20-01914]. Findings will be disseminated through scientific conferences, open access peer-reviewed journals, COVID-19 research repositories and dissemination channels used by our patient partners.

Osteosarcoma is a malignant primary bone tumor commonly occurring in children and adolescents. The treatment of local osteosarcoma is mainly based on surgical resection and chemotherapy, whereas the improvement of overall survival remains stagnant, especially in recurrent or metastatic cases. Tumor microenvironment (TME) is closely related to the occurrence and development of tumors, and macrophages are among the most abundant immune cells in the TME. Due to their vital roles in tumor progression, macrophages have gained increasing attention as the new target of tumor immunotherapy. In this review, we present a brief overview of macrophages in the TME and highlight the clinical significance of macrophages and their roles in the initiation and progression of osteosarcoma. Finally, we summarize the therapeutic approaches targeting macrophage, which represent a promising strategy in osteosarcoma therapies.

Objective To evaluate the association between having arthritis and the perceived need for mental healthcare and use of mental health support among individuals with mental disorders.Design A cross-sectional analysis using data from Canadian Community Health Survey—Mental Health (2012).Setting The survey was administered across Canada’s 10 provinces using multistage cluster sampling.Participants The study sample consisted of individuals reporting depression, anxiety or bipolar disorder.Study variables and analysis The explanatory variable was self-reported doctor-diagnosed arthritis, and outcomes were perceived need for mental healthcare and use of mental health support. We computed overall and gender-stratified multivariable binomial logistic regression models adjusted for age, gender, race/ethnicity, income and geographical region.Results Among 1774 individuals with a mental disorder in the study sample, 436 (20.4%) reported having arthritis. Arthritis was associated with increased odds of having a perceived need for mental healthcare (adjusted OR (aOR) 1.71, 95% CI 1.06 to 2.77). In the gender-stratified models, this association was increased among men (aOR 2.69, 95% CI 1.32 to 5.49) but not women (aOR 1.48, 95% CI 0.78 to 2.82). Evaluation of the association between arthritis and use of mental health support resulted in an aOR of 1.50 (95% CI 0.89 to 2.51). Individuals with arthritis tended to use medications and professional services as opposed to non-professional support.Conclusion Comorbid arthritis among individuals with a mental disorder was associated with an increased perceived need for mental healthcare, especially in men, underscoring the importance of understanding the role of masculinity in health seeking. Assessing the mental health of patients with arthritis continues to be essential for clinical care.

Chondrocytes are the only cell type in normal cartilage. The pathological changes of osteoarthritis (OA) mostly revolve around the apoptosis and dysfunction of chondrocytes. Autophagy, as an intracellular degradation system that maintains the steady state of energy metabolism in cells, has been shown to restore the function of damaged chondrocytes, alleviating the occurrence and progression of OA. In this review, we explored the relationship between autophagy and OA and the key molecules of autophagy pathway that regulate the progression of OA, providing new ideas for OA treatment by targeting autophagy.

Improving revascularization is one of the major measures in fracture treatment. Moderate local inflammation triggers angiogenesis, whereas systemic inflammation hampers angiogenesis. Previous studies showed that Akkermansia muciniphila, a gut probiotic, ameliorates systemic inflammation by tightening the intestinal barrier. In this study, fractured mice intragastrically administrated with A. muciniphila were found to display better fracture healing than mice treated with vehicle. Notably, more preosteclasts positive for platelet-derived growth factor-BB (PDGF-BB) were induced by A. muciniphila at 2 weeks post fracture, coinciding with increased formation of type H vessels, a specific vessel subtype that couples angiogenesis and osteogenesis, and can be stimulated by PDGF-BB. Moreover, A. muciniphila treatment significantly reduced gut permeability and inflammation at the early stage. Dextran sulfate sodium (DSS) was used to disrupt the gut barrier to determine its role in fracture healing and whether A. muciniphila still can stimulate bone fracture healing. As expected, A. muciniphila evidently improved gut barrier, reduced inflammation and restored the impaired bone healing and angiogenesis in DSS-treated mice. Our results suggest that A. muciniphila reduces intestinal permeability and alleviates inflammation, which probably induces more PDGF-BB+ preosteoclasts and type H vessel formation in callus, thereby promoting fracture healing. This study provides the evidence for the involvement of type H vessels in fracture healing and suggests the potential of A. muciniphila as a promising strategy for bone healing. The development of flexible transparent conductive electrodes has been considered as a key issue in realizing flexible functional electronics. Inkjet printing provides a new opportunity for the manufacture of FFE due to simple process, cost-effective, environmental friendliness, and digital method to circuit pattern. However, obtaining high concentration of inkjet- printed silver nanowires (AgNWs) conductive ink is a great challenge because the high aspect ratio of AgNWs makes it easy to block the jetting nozzle. This study provides an inkjet printing AgNWs conductive ink with low viscosity and high concentration of AgNWs and good printing applicability, especially without nozzle blockage after printing for more than 4 h. We discussed the effects of the components of the ink on surface tension, viscosity, contact angle as well as droplet spreading behavior. Under the optimized process and formulation of ink, flexible transparent conductive electrode with a sheet resistance of 32 Ω·sq−1–291 nm·sq−1 and a transmittancy at 550 nm of 72.5–86.3% is achieved. We investigated the relationship between the printing layer and the sheet resistance and the stability of the sheet resistance under a bending test as well as the infrared thermal response of the AgNWs–based flexible transparent conductive electrode. We successfully printed the coupling electrodes and demonstrated the excellent potential of inkjet-printed AgNWs—based flexible transparent conductive electrode for developing flexible functional electronics.

Traffic noise and solid waste pollution are two major problems that restrict urban development and affect urban environments. In this study, a new kind of cement-based material for sound barriers was prepared using industrial waste fly ash cenosphere to explore the material ratio of the sound absorption, sound insulation, and composite layers and to optimize the material’s properties. The research findings showed that the compressive strength had significant effects on the material properties of the sound absorption layer, with the optimal compressive strength range being 0.2–0.4 MPa. At 0.4 MPa, the material with an aggregate-to-binder ratio of 1.0 had the best comprehensive properties. The sound insulation layer had the best compressive strength of 29.00 MPa at a 45% fiber admixture. The composite had the best sound insulation when the thickness ratio of the sound absorption and insulation layers was 60:40, and the sound transmission loss was 38 dB. The embodied carbon (EC) and embodied energy (EE) of the new fly ash cenosphere across the whole life cycle were 57.57 kgCO2e and 477.08 MJ, respectively, which were 4.8−52.9% and 53.2−82.3% lower than other traditional sound barriers, respectively. Thus, they were environmentally friendly and had satisfactory energy-saving and environmental protection values.

Escherichia coli is classified into four major phylogenetic groups (A, B1, B2, and D) that are associated with antibiotic resistance genes. Although antibiotic-resistant E. coli is commonly detected in municipal wastewater, little is known about the relationship between the phylogenetic groups and antibiotic-resistant E. coli in wastewater. In this study, the survival of E. coli in wastewater and the changes to the relationships between each phylogroup and the antibiotic-resistant profiles of E. coli isolates from wastewater were investigated under aerobic conditions for 14 days. The isolates were classified into the phylogroups A, B1, B2, and D or others by multiplex PCR. In addition, the susceptibility of the isolates to 11 antibiotics was assessed with the minimum inhibitory concentration (MIC) assay. While E. coli counts decreased in the wastewater with time under aerobic conditions, the prevalence of phylogroup B2 had increased to 73% on day 14. Furthermore, the MIC assay revealed that the abundance of antibiotic-resistant E. coli also increased on day 14. After batch-mixing the experiments under aerobic conditions, the surviving antibiotic-resistant E. coli included mainly multidrug-resistant and beta-lactamase-producing isolates belonging to phylogroup B2. These results suggest that the phylogroup B2 isolates that have acquired antibiotic resistance had a high survivability in the treated wastewater.

Fannia scalaris (Fabricius, 1794) is closely related to human life in ecological habits, which can lead to health concerns since they feed on various contamination sources. In this study, we first present the mitochondrial genome (mitogenome) of F. scalaris (GenBank No. MT017706). The length of this mitogenome was composed of 15,040 base pairs, including 13 protein-coding genes, two ribosomal RNA, 22 transfer RNA, and an AT-rich region. It consisted of A 39.3%, G 9.1%, C 13.0%, and T 38.6%. The arrangement of the genes was consistent with that of the ancestral metazoan. Furthermore, phylogenetic relationship indicated that F. scalaris was obviously separated from the muscid flies. This study provides useful genetic data in order to further understand the evolutionary relationship of the Muscidae.

“Ivy sign” is a special imaging manifestation of moyamoya disease (MMD), which shows continuous linear or punctate high intensity along the cortical sulci and subarachnoid space on magnetic resonance images. Ivy sign was reported to reflect the development of compensatory collaterals, and to be closely related to hemodynamic changes and clinical symptoms, and to indicate the postoperative prognosis, in MMD patients. It is a unique and critical marker for MMD. However, due to the lack of consistent criteria, such as definition, grading, and identification standards, ivy sign has not received much attention. We undertook a comprehensive literature search and summarized the current situation regarding ivy sign in MMD in terms of baseline characteristics, detection methods, definition, regional division and distribution patterns, grading criterions, incidence, related factors, the mechanism of ivy sign, and the effects of treatments. We also provided related concerns raised and future prospects relevant to studies about ivy sign in MMD.

Background The highest rate of cancer-related deaths worldwide is from lung adenocarcinoma (LUAD) annually. Metabolism was associated with tumorigenesis and cancer development. Metabolic-related genes may be important biomarkers and metabolic therapeutic targets for LUAD. Materials and Methods In this study, the gleaned cohort included LUAD RNA-SEQ data from the Cancer Genome Atlas (TCGA) and corresponding clinical data (n = 445). The training cohort was utilized to model construction, and data from the Gene Expression Omnibus (GEO, GSE30219 cohort, n = 83; GEO, GSE72094, n = 393) were regarded as a testing cohort and utilized for validation. First, we used a lasso-penalized Cox regression analysis to build a new metabolic-related signature for predicting the prognosis of LUAD patients. Next, we verified the metabolic gene model by survival analysis, C-index, receiver operating characteristic (ROC) analysis. Univariate and multivariate Cox regression analyses were utilized to verify the gene signature as an independent prognostic factor. Finally, we constructed a nomogram and performed gene set enrichment analysis to facilitate subsequent clinical applications and molecular mechanism analysis. Result Patients with higher risk scores showed significantly associated with poorer survival. We also verified the signature can work as an independent prognostic factor for LUAD survival. The nomogram showed better clinical application performance for LUAD patient prognostic prediction. Finally, KEGG and GO pathways enrichment analyses suggested several especially enriched pathways, which may be helpful for us investigative the underlying mechanisms.

Differential expressions of genes are widely evaluated for the diagnosis and prognosis correlations with diseases. But limited studies investigate how transcriptional regulations are quantitatively altered in diseases. This study proposes a novel model-based quantitative measurement of transcriptional regulatory relationships between mRNA genes and Transcription Factor (TF) genes (mqTrans features). This study didn't consider the regulatory relationships between TF genes, so the mRNA genes were the protein-coding genes excluding the TF genes. The models are trained in the control samples in a lung cancer dataset and evaluated in two independent datasets and the hold-out testing samples from the third dataset. Twenty-nine mRNA genes are detected with transcriptional regulations quantitatively altered in lung cancers. The transcriptional modification technologies like RNA interference (RNAi) may be utilized to restore the altered transcriptional regulations in lung cancers.

Aims and hypothesisEpidermal growth factor (EGF) has been shown to induce the migration of various cancer cells. However, the underlying signaling mechanisms for EGF-induced migration of oral squamous cell carcinoma (OSCC) remain to be elucidated. WNT7A, a member of the family of 19 Wnt secreted glycoproteins, is commonly associated with tumor development. It is mostly unknown whether and, if so, how EGF modulates WNT7A in OSCC cells. The role of WNT7A in OSCC was thus investigated to explore the underlying signaling mechanisms for EGF-induced migration of OSCC.MethodsCell migration was measured by Wound healing assay and Transwell assay. Western blotting was carried out to detect the expression of WNT7A, MMP9, β-catenin, p-AKT, and p-ERK. The cells were transfected with plasmids or siRNA to upregulate or downregulate the expression of WNT7A. The location of β-catenin was displayed by immunofluorescence microscopy. Immunohistochemistry was carried out to confirm the relation between WNT7A expression and OSCC progression.ResultsThe present study showed that the levels of WNT7A mRNA and protein were increased by EGF stimulation in OSCC cells. Besides, it was proved that p-AKT, but not p-ERK, mediated the expression of WNT7A protein induced by EGF. Furthermore, the inhibition of AKT activation prevented the EGF-induced increase of WNT7A and matrix metallopeptidase 9 (MMP9) expression and translocation of β-catenin from the cytoplasm to the nucleus. Moreover, histological analysis of OSCC specimens revealed an association between WNT7A expression and poor clinical prognosis of the disease.ConclusionsThe data in this paper indicated that WNT7A could be a potential oncogene in OSCC and identified a novel PI3K/AKT/WNT7A/β-catenin/MMP9 signaling for EGF-induced migration of OSCC cells.

Drought and heat stresses can influence the expressions of genes, and thereby affect the growth and development of plants. Alternative splicing (AS) of genes plays crucial roles through increasing transcriptome diversity in plant stress responses. Tea plants, widely cultivated in the tropics and subtropics, are often simultaneously exposed to drought and heat stresses. In the present study, we performed a global transcriptome of tea leaves treated with drought, heat or their combination. In total, 19,019, 20,025 and 20,253 genes underwent AS in response to drought (DT), heat (HT) and their combined stress (HD), respectively, of which 12,178, 11,912 and 14,413 genes differentially spliced in response to DT, HT and HD, respectively. Also, 2,447 specific differentially spliced genes (DSGs) were found only in response to HD. All DSGs accounted for 48% of the annotated genes in tea tree genome. Comparison of DSGs and differentially expressive genes (DEGs) showed that the proportions of HT and HD-induced DSGs were 13.4% and 9.2%, while the proportion of DT increased to 28.1%. Moreover, the DEG-DSG overlapped genes tended to be enriched in a wide large of pathways in response to DT. The results indicated that the AS of genes in tea leaves was extensively triggered by drought, heat and their combined stresses. In addition, the AS enhanced the transcriptome adaption in response to drought and heat stresses, and the AS also provoked specific molecular functions in response to drought and heat synergy stress. The study might have practical significance for molecular genetic breeding of tea plants with stress resistance.

Zika virus (ZIKV) exhibits a tropism for brain tumor cells and has been used as an oncolytic virus to target brain tumors in mice with modest effects on extending median survival. Recent studies have highlighted the potential for combining virotherapy and immunotherapy to target cancer. We postulated that ZIKV could be used as an adjuvant to enhance the long-term survival of mice with malignant glioblastoma and generate memory T-cells capable of providing long-term immunity against cancer remission. To test this hypothesis mice bearing malignant intracranial GL261 tumors were subcutaneously vaccinated with irradiated GL261 cells previously infected with the ZIKV. Mice also received intracranial injections of live ZIKV, irradiation attenuated ZIKV, or irradiated GL261 cells previously infected with ZIKV. Long-term survivors were rechallenged with a second intracranial tumor to examine their immune response and look for the establishment of protective memory T-cells. Mice with subcutaneous vaccination plus intracranial irradiation attenuated ZIKV or intracranial irradiated GL261 cells previously infected with ZIKV exhibited the greatest extensions to overall survival. Flow cytometry analysis of immune cells within the brains of long-term surviving mice after tumor rechallenge revealed an increase in the number of T-cells, including CD4+ and tissue-resident effector/ effector memory CD4+ T-cells, in comparison to long-term survivors that were mock-rechallenged, and in comparison to naïve untreated mice challenged with intracranial gliomas. These results suggest that ZIKV can serve as an adjuvant to subcutaneous tumor vaccines that enhance long-term survival and generate protective tissue-resident memory CD4+ T-cells.

Cu-Ni-Si alloys have been widely used in electronics and electrical industries.The preparation method of alloy was not limited to smelting, powder metallurgy method has also attracted the attention of many researchers. In this study, Cu-Ni-Si alloys were prepared using hot-pressed sintering and elemental copper powders, in which nickel and silicon powders were used as raw materials. The results show that, when the Cu-Ni-Si alloys were prepared using hot-pressed sintering, there were no Ni-Si intermetallic compounds except the δ -Ni _2 Si phase in the microstructure of alloys. After the ageing treatment, when the mass ratios of Ni/Si were 2:1 and 3:1, the precipitation of δ -Ni _2 Si phase was significantly less. However, when the mass ratios of Ni/Si were 4:1 and 5:1, the precipitation of δ -Ni _2 Si phase particles increased significantly. The results from electrical conductivity and Vickers hardness show that, after the ageing treatment, both the electrical conductivity and Vickers hardness of the alloys greatly improved.

As popularly consumed fruit berries, grapes are widely planted and processed into products, such as raisins and wine. In order to identify the influences of different climatic conditions on grape coloring and quality formation, we selected two common varieties of grape berries, ‘Red Globe’ and ‘Xin Yu’, for investigation. Grapes were separately grown in different climates, such as a temperate continental arid climate and a temperate continental desert climate, in Urumqi and Turpan, China, for five developmental stages. As measured, the average daily temperature and light intensity were lower in Urumqi. Urumqi grape berries had a lower brightness value (L*) and a higher red-green value (a*) when compared to Turpan’s. A RT-qPCR analysis revealed higher transcriptions of key genes related to anthocyanin biosynthesis in Urumqi grape berries, which was consistent with the more abundant phenolic substances, especially anthocyanins. The maximum antioxidant activity in vitro and cellular antioxidant activity of grape berries were also observed in Urumqi grape berries. These findings enclosed the influence of climate on anthocyanin accumulation and the antioxidant capacity of grapes, which might enlarge our knowledge on the quality formation of grape berries and might also be helpful for cultivating grapes with higher nutritional value.

A new species, Tylencholaimus helanensis sp. n., extracted from the rhizosphere soil of unidentified grasses from Helan Mountain, Inner Mongolia, China was identified. The new species is characterized by having a body length of 0.93–1.07 mm with the lip region approximately one-quarter of the body diameter at the posterior end of the neck region wide; female didelphic-amphidelphic; pars proximalis vaginae violin-shaped. Males were not found. SEM observations of the new species were made and a phylogenetic analysis of both the 18S rDNA and the D2-D3 region of 28S rDNA is presented.

Background/Aims: Hepatocellular carcinoma (HCC) has a high morbidity as well as mortality and is believed to be one of the most prevalent cancers worldwide. The long non-coding RNA metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) is involved in numerous cancers, including HCC. This study aimed to explore the role of MALAT1 in HCC progression. Methods: The expression levels of MALAT1 and Vimentin in HCC tissues and relative pair-matched adjacent normal liver tissues were analyzed by RT-PCR, and immunohistochemistry. Using bioinformatics analysis and dual-luciferase assay, we examined the correlation between MALAT1 and miR-30a-5p. Dual-luciferase assay and western blotting suggested that Vimentin was a target of miR-30a-5p. A wound healing assay and transwell assays were employed to determine the effect of MALAT1 and miR-30a-5p on cell migration and invasion in HCC. Results: Our data demonstrated that the levels of MALAT1 and Vimentin were upregulated in HCC tissues and that miR-30a-5p was a direct target of MALAT1. Silenced MALAT1 and overexpressed miR-30a-5p each inhibited cell migration and invasion. Additionally, dual-luciferase assay and western blotting demonstrated that MALAT1 could competitively sponge miR-30a-5p and thereby regulate Vimentin. Conclusion: Our data suggest that MALAT1 acts as an oncogenic lncRNA that promotes HCC migration and invasion. Therefore, the MALAT1-miR-30a-5p-Vimentin axis is a potential therapeutic target and molecular biomarker in HCC.

Background/Aims: Recently, we and others showed that the relative abundance of a specific vessel subtype, strongly positive for CD31 and Endomucin (CD31hiEmcnhi), is associated with bone formation and bone loss, and platelet-derived growth factor-BB (PDGF-BB) secreted by preosteoclasts induces the formation of the specific vessels and thereby stimulates osteogenesis. Inhibition of Src homology 2 domain-containing protein tyrosine phosphatase-2 (SHP-2) has been shown to block the fusion of preosteoclasts into mature osteoclasts. However, it is unclear whether inhibition of SHP-2 could promote preosteoclast-induced angiogenesis and then enhance bone formation. This study aimed to determine the effects of a specific SHP-2 inhibitor (NSC-87877) on CD31 hiEmcnhi vessel and bone formation. Methods: 3-month-old C57BL/6 mice were subjected to either ovariectomy (OVX) or sham operation. OVX mice were intraperitoneally injected with NSC-87877 and the control (sham) mice were treated with an equal volume of diluents (PBS). Two months later, bone samples from mice were collected for µCT, histological, immunohistochemical and immunofluorescent analyses to assess bone mass, osteogenic and osteoclastic acitivities, as well as the densities of CD31hiEmcnhi vessels. A series of angiogenesis- related assays were performed to test the effects of NSC-87877 on the pro-angiogenic activities of preosteoclasts in vitro. Results: We found that NSC-87877 is sufficient to induce bone-sparing effects in OVX-induced osteoporotic mouse model. We also found that NSC-87877 induces higher numbers of preosteoclasts and CD31hiEmcnhi vessels and higher levels of PDGF-BB in bone marrow of osteoporotic mice. In vitro assays showed that NSC-87877 prevents preosteoclast fusion, increases PDGF-BB production, and augments the pro-angiogenic abilities of preosteoclasts. Conclusion: Our results suggest that NSC-87877 can be used as a promising therapeutic agent for osteoporosis by inhibiting osteoclast formation and promoting preosteoclast-induced angiogenesis.

Severe plastic deformation (SPD) was carried out on AZ80 alloy by high-pass multi-directional forging (MDF). Results showed that AZ80 alloy was effectively refined through MDF, and average grain size was refined to 0.73 µm after 24 passes of forging. Its tensile strength, yield strength and elongation reached 333.8 MPa, 233.7 MPa and 17.8%, respectively, which were 1.22 times, 1.35 times and 2.54 times of those before the forging. When the MDF was less than 6 passes, the coarse grains were mechanically divided and refined, and a small amount of recrystallized grains appeared in the deformation band and its cross zone. When the MDF was more than 6 passes, grain division, recrystallization nucleation and grain growth occurred simultaneously, the microstructure refinement was slowed, and the uniformity was improved continuously. The heating between forging passes mainly caused restoration in the early forging, and the static recrystallization refinement in the middle of forging. In the later stage of forging, the grain growth caused by static recrystallization and the grain refinement caused by the deformation canceled out, and the microstructure no longer continued to refine with the increase of the forging pass. Hot compression tests of Cu-Ni-Si alloy with high Ni and Si contents were performed at the deformation temperatures of 750–950 °C, and the strain rates of 0.01–10 s−1 on a Gleeble-3800 thermo-mechanical simulator. The critical conditions for dynamic recrystallization (DRX) of Cu-Ni-Si alloy were obtained by processing the collected data and combining with the inflection point of θ-σ curve with the minimum value criterion of −∂θ/∂σ-σ curve. There was a certain correlation between the critical strain (εc) and the peak strain (εp), that was, εc/εp = 0.723, and the functional relationship between εc and Z parameter was εc = 1.05 × 10−3Z0.059. The average recrystallized grain size under various deformation conditions was measured, and the DRX grain growth model of Cu-Ni-Si alloy was established as dDRX = 5650.4ε̇−0.12891exp(−73142/RT). Porous silicon nitride (Si₃N₄) ceramics with highly ordered and unidirectionally aligned channels were fabricated by the extrusion method using nylon fibers as the pore forming agent. The microstructure, mechanical properties and gas permeability were investigated. The results showed that the added nylon fibers were mostly converted to pores, the average observed pore diameter was approximately 19.5 μm and shrank about 22% from the size of original pore former (25 μm) during sintering. The porosity and gas permeability increased and the 3-point bending strength decreased proportionally with increasing the nylon fiber content. When the nylon fiber content increased to 20 wt. %, the porosity, bending strength and gas permeability of the extrusion samples with uni-directionally aligned pores was higher than that of non-extruded sample with random pores. For the extruded samples, the resulting porous silicon nitride ceramics showed a high gas permeability owing to their uni-directionally pore oriented structure.

Abstract Background White matter hyperintensities (WMHs) are frequently detected in migraine patients. However, their significance and correlation to migraine disease burden remain unclear. This study aims to examine the correlation of WMHs with migraine features and explore the relationship between WMHs and migraine prognosis. Methods A total of 69 migraineurs underwent MRI scans to evaluate WMHs. Migraine features were compared between patients with and without WMHs. After an average follow-up period of 3 years, these patients were divided into two groups, according to the reduction of headache frequency: improved and non-improved groups. The percentage and degree of WMHs were compared between these two groups. Results A total of 24 patients (34.8%) had WMHs. Abstract Background Aspergillus niger is a highly important industrial microorganism because of its amazing capacity to produce citric acid (CA). To explore the metabolic mechanism and physiological phenotype associated with high CA productivity, the transcriptomes of high CA-producing A. niger YX-1217 and degenerative strain YX-1217G were investigated using A. niger ATCC1015 as a control. Results These strains showed distinct transcriptional differences in CA production. By contrast, the genes encoding glycoside hydrolases, aspartyl endoproteases, and carboxypeptidases were unusually upregulated in CA-producing strain YX-1217, which involved the carbohydrate hydrolysis and polypeptide degradation pathways, and should be related to its powerful capacity to utilize cornmeal fluidified liquid as raw material for the production of CA. In central metabolism of YX-1217, gene 9.735.1, which encodes glyceraldehyde 3-phosphate dehydrogenase, and two transcriptionally outstanding genes, 6000119 (An15g01920) and 3.2152.1 (An08g10920) that encode citrate synthase, were upregulated, thereby ensuring CA accumulation. In addition, a relatively strong electron transport chain, a regeneration system for NAD+/NADP+, and an efficient resistance mechanism may have contributed to the high CA production rate of YX-1217. Conclusions These comparisons have shed light on the mechanism underlying high CA yield in A. niger YX-1217 as well as provide insights into the development of novel strains that produce other organic acids.

The Klobuchar model has been widely used to correct the ionospheric delay in applications. However, the NVTEC (Nighttime Vertical Total Electron Content) of the Klobuchar model employs an empirical constant of 9 TECU (Total Electron Content Unit) at L1 frequency. In this paper, the rationality and reliability of the nighttime constant setting are investigated using the GIM (Global Ionosphere Map) product of the IGS (International GNSS Service) from 1998 to 2015. Our study indicates that the suitable time span of NVTEC average in nighttime should be between 20:00 and 06:00 LT (local time). The NVTEC is highly correlated with seasons, having positive extremes in spring and autumn and negative extremes in summer through the mean values in all latitudes. In addition to seasonal dependence, solar activity in the solar cycle 23 strongly influences NVTEC as well and leads to its variation within a range between 25 and 30 TECU in spring and autumn at solar maximum, which is about 1.5 times greater than that in summer and winter. The NVTEC also has a dependence on the latitude at solar maximum, with the mean value from 30 TECU in low latitudinal regions to 15 TECU in high latitudinal regions. Therefore, these results demonstrate that the nighttime VTEC has much greater deviations from the imperial constant in the Klobuchar model, and the newly estimated constant is expected to bring improvement to the predictability of the Klobuchar ionospheric delay model in nighttime. Objective: Nicotinamide phosphoribosyl transferase (NAMPT) is the rate-limiting enzyme in the salvage pathway that produces nicotinamide adenine dinucleotide (NAD+), an essential co-substrate regulating a myriad of signaling pathways. We produced a mouse that overexpressed NAMPT in skeletal muscle (NamptTg) and hypothesized that NamptTg mice would have increased oxidative capacity, endurance performance, and mitochondrial gene expression, and would be rescued from metabolic abnormalities that developed with high fat diet (HFD) feeding. Methods: Insulin sensitivity (hyperinsulinemic-euglycemic clamp) was assessed in NamptTg and WT mice fed very high fat diet (VHFD, 60% by kcal) or chow diet (CD). The aerobic capacity (VO2max) and endurance performance of NamptTg and WT mice before and after 7 weeks of voluntary exercise training (running wheel in home cage) or sedentary conditions (no running wheel) were measured. Skeletal muscle mitochondrial gene expression was also measured in exercised and sedentary mice and in mice fed HFD (45% by kcal) or low fat diet (LFD, 10% by kcal). Results: NAMPT enzyme activity in skeletal muscle was 7-fold higher in NamptTg mice versus WT mice. There was a concomitant 1.6-fold elevation of skeletal muscle NAD+. NamptTg mice fed VHFD were partially protected against body weight gain, but not against insulin resistance. Notably, voluntary exercise training elicited a 3-fold higher exercise endurance in NamptTg versus WT mice. Mitochondrial gene expression was higher in NamptTg mice compared to WT mice, especially when fed HFD. Mitochondrial gene expression was higher in exercised NamptTg mice than in sedentary WT mice. Conclusions: Our studies have unveiled a fascinating interaction between elevated NAMPT activity in skeletal muscle and voluntary exercise that was manifest as a striking improvement in exercise endurance. Karst rocky desertification (KRD) is a global environmental degradation problem caused by human activities and vegetation deterioration. Vegetation restoration in KRD is very difficult due to severe water loss, soil erosion and extensive bare bedrock. For achieving vegetation restoration on bare bedrock areas of KRD, the nutrient medium technique was developed, which is based on the special hydro-geological conditions that is seasonal rainfall is abundant in Karst region of Southwest China and limestone fissures are extremely developed and rich in fissure water. The objective of this research is to investigate the feasibility of using nutrient medium technique to achieve vegetation restoration in KRD region. Through the indoor and field experiments, some properties of the nutrient medium mixture were tested, including water retention, water absorption and nutrient retention. The results showed that the moisture content on the 75th day and water absorption rate of No.2 mixture was 19.9% and 1.67 mm/d, respectively, which could be used as optimum mix ratio for producing nutrient medium. The field results showed that the nutrient medium technique could effectively guarantee the survival of sapling at early age and sustainable growth at later age in the field, and the nutrient medium had less nutrient loss. This novel approach can retain and absorb moisture, and saplings do not need artificially supply water and fertiliser in the dry season, and the use of nutrient medium technique to achieve vegetation restoration in KRD region seems to be a feasible option.

Background/Aims: Prostate cancer (PCa) is the second most frequently diagnosed cancer in men worldwide. Currently available therapies for hormone-refractory PCa are only marginally effective. Plumbagin (PLB), a natural naphthoquinone isolated from the traditional folk medicine Plumbago zeylanica, is known to selectively kill tumor cells. Nevertheless, antitumor mechanisms initiated by PLB in cancer cells have not been fully defined. Methods: MTT assay was used to evaluate the effect of PLB on the viability of cancer cells. Cell apoptosis and reactive oxygen species (ROS) production were determined by flow cytometry. Protein expression was detected by western blotting. In vivo anti-tumor effect was measured by using tumor xenoqraft model in nude mice. Results: In the present study, we found that PLB decreases cancer cell growth and induces apoptosis in DU145 and PC-3 cells. In addition, by increasing intracellular ROS levels, PLB induced a lethal endoplasmic reticulum stress response in PCa cells. Importantly, blockage of ROS production significantly reversed PLB-induced ER stress activation and cell apoptosis. In vivo, we found that PLB inhibits the growth of PCa xenografts without exhibiting toxicity Treatment of mice bearing human PCa xenografts with PLB was also associated with induction of ER stress activation. Conclusion: Inducing ER stress by PLB thus discloses a previously unrecognized mechanism underlying the biological activity of PLB and provides an in-depth insight into the action of PLB in the treatment of hormone-refractory PCa.

Background: Tat-interacting protein 30 (TIP30) has been reported to be a tumor suppressor, with reduced or absent expression in various tumors. However, its role in bladder urothelial cancer (BUC) has not been investigated. Therefore, herein, we investigated the expression of TIP30 protein in BUC and normal bladder mucosa and the clinical significance of TIP30 expression in the prognosis of BUC. Methods: We reviewed data from 79 cases of BUC and 15 adjacent tissue samples from 79 patients treated at our institution between 2004 and 2007. TIP30 expression was examined by immunohistochemistry. The relationship between TIP30 expression and tumor stage, histological grade, and survival was analyzed. Differences between groups were evaluated using the t-test or matched-pairs test, and differences in the survival rates were analyzed with the log-rank test. Results: TIP30 protein expression was significantly reduced in BUC tissue (t = −6.91, P < 0.05) compared with normal tissue samples, and in invasive bladder cancer (t = 10.89, P < 0.05) compared with superficial bladder cancer. TIP30 protein expression differed significantly among different differentiated groups classified either according to the World Health Organization (2004, F = 17.48, P < 0.01) or World Health Organization (1973, F = 10.68, P < 0.01). TIP30 protein expression was significantly reduced in high-grade papillary urothelial carcinoma compared with papillary urothelial neoplasm of low malignant potential (P < 0.05) and low-grade papillary urothelial carcinoma (P < 0.05). Meanwhile, TIP30 protein expression was significantly reduced in Grade III BUC, compared with Grade I (P < 0.05) and Grade II (P < 0.05). Patients with low TIP30 expression showed a higher incidence of disease progression than those with high TIP30 expression (t = 2.63, P < 0.05). Kaplan-Meier survival analysis showed a strong positive relationship between TIP30 expression and overall survival (OS) (χ2 = 17.29, P < 0.05). Conclusions: TIP30 expression was associated with clinical tumor stage in BUC, suggesting that it might play an important role in disease progression. Furthermore, TIP30 might predict postoperative OS. Thus, its evaluation might be useful for predicting prognosis.

Fritillaria taipaiensis is a valuable traditional Chinese medicinal plant that has experienced continuous decline over its cropping area. The present study aims to explore the reasons for the quality and growth decline. The fungal diversities and biochemical factors in its rhizospheric soils with cultivation duration from 1 up to 5 years were analyzed and compared. The results showed that rhizospheric fungi of F. taipaiensis belong to six phyla, including Neocallimastigomycota, Glomeromycota, Basidiomycota, Chytridiomycota, Zygomycota and Ascomycota. Thirteen genera (Pseudogymnoascus, Fusarium, Mortierella, Colletotrichum, Laetinaevia, Gibberella, Synchytrium, Lysurus, Trichocladium, Volutella, Monoblepharis, Aquamyces and Trichoderma) constituted the “core community” in the rhizosphere of F. taipaiensis. The dominant fungal genera varied significantly in rhizospheric soils with different cultivation years. The abundance of fungal species in the soil declined with the cultivation year generally. The pH, available P, organic matter and urease activity were the primary factors determining the fungal community composition in the rhizosphere. The content of organic matter, available N, P and K and the activities of urease and alkaline phosphatase decreased with cultivation years. The soil pH increased with cultivation years and was unsuitable for F. taipaiensis growth. These features suggested that long-term single planting altered the fungal community structure, fertility conditions and soil enzyme activities in F. taipaiensis rhizospheric soils, which could be detrimental for plant growth and quality.

The path tracking control system is a crucial component for autonomous vehicles; it is challenging to realize accurate tracking control when approaching a wide range of uncertain situations and dynamic environments, particularly when such control must perform as well as, or better than, human drivers. While many methods provide state-of-the-art tracking performance, they tend to emphasize constant PID control parameters, calibrated by human experience, to improve tracking accuracy. A detailed analysis shows that PID controllers inefficiently reduce the lateral error under various conditions, such as complex trajectories and variable speed. In addition, intelligent driving vehicles are highly non-linear objects, and high-fidelity models are unavailable in most autonomous systems. As for the model-based controller (MPC or LQR), the complex modeling process may increase the computational burden. With that in mind, a self-optimizing, path tracking controller structure, based on reinforcement learning, is proposed. For the lateral control of the vehicle, a steering method based on the fusion of the reinforcement learning and traditional PID controllers is designed to adapt to various tracking scenarios. According to the pre-defined path geometry and the real-time status of the vehicle, the interactive learning mechanism, based on an RL framework (actor–critic—a symmetric network structure), can realize the online optimization of PID control parameters in order to better deal with the tracking error under complex trajectories and dynamic changes of vehicle model parameters. The adaptive performance of velocity changes was also considered in the tracking process. The proposed controlling approach was tested in different path tracking scenarios, both the driving simulator platforms and on-site vehicle experiments have verified the effects of our proposed self-optimizing controller. The results show that the approach can adaptively change the weights of PID to maintain a tracking error (simulation: within ±0.071 m; realistic vehicle: within ±0.272 m) and steering wheel vibration standard deviations (simulation: within ±0.04°; realistic vehicle: within ±80.69°); additionally, it can adapt to high-speed simulation scenarios (the maximum speed is above 100 km/h and the average speed through curves is 63–76 km/h).

Force-distance curve-based atomic force microscopy can measure material nanomechanics, but only if the probe and material stiffness match, which limits the measurement range. Abstract Background Being physically active is an essential component of successful self-management for people with inflammatory arthritis; however, the vast majority of patients are inactive. This study aims to determine whether a technology-enabled counselling intervention can improve physical activity participation and patient outcomes. Methods The Effectiveness of Online Physical Activity Monitoring in Inflammatory Arthritis (OPAM-IA) project is a community-based randomized controlled trial with a delayed control design. We will recruit 130 people with rheumatoid arthritis or systemic lupus erythematosus, who can be physically active without health professional supervision. Randomization will be stratified by diagnosis. In Weeks 1–8, participants in the Immediate Group will: 1) receive education and counselling by a physical therapist (PT), 2) use a Fitbit and a new web-based application, FitViz, to track and obtain feedback about their physical activity, 3) receive 4 biweekly follow-up calls from the PT. Those in the Delayed Group will receive the same program in Week 10. We will interview a sample of participants about their experiences with the intervention. Participants will be assessed at baseline, and Weeks 9, 18 and 27. The primary outcome measure is time spent in moderate/vigorous physical activity in bouts of ≥ 10 min, measured with a portable multi-sensor device in the free-living environment. Secondary outcomes include step count, time in sedentary behaviour, pain, fatigue, mood, self-management capacity, and habitual behaviour. Discussion A limitation of this study is that participants, who also administer the outcome measures, will not be blinded. Nonetheless, by customizing existing self-monitoring technologies in a patient-centred manner, individuals can be coached to engage in an active lifestyle and monitor their performance. The results will determine if this intervention improves physical activity participation. The qualitative interviews will also provide insight into a paradigm to integrate this program to support self-management. This paper addresses the problem of instance-level 6DoF pose estimation from a single RGBD image in an indoor scene. Many recent works have shown that a two-stage network, which first detects the keypoints and then regresses the keypoints for 6d pose estimation, achieves remarkable performance. However, the previous methods concern little about channel-wise attention and the keypoints are not selected by comprehensive use of RGBD information, which limits the performance of the network. To enhance RGB feature representation ability, a modular Split-Attention block that enables attention across feature-map groups is proposed. In addition, by combining the Oriented FAST and Rotated BRIEF (ORB) keypoints and the Farthest Point Sample (FPS) algorithm, a simple but effective keypoint selection method named ORB-FPS is presented to avoid the keypoints appear on the non-salient regions. The proposed algorithm is tested on the Linemod and the YCB-Video dataset, the experimental results demonstrate that our method outperforms the current approaches, achieves ADD(S) accuracy of 94.5% on the Linemod dataset and 91.4% on the YCB-Video dataset.

Laccases can catalyze the remediation of hazardous synthetic dyes in an eco-friendly manner, and thermostable laccases are advantageous to treat high-temperature dyeing wastewater. A novel laccase from Geothermobacter hydrogeniphilus (Ghlac) was cloned and expressed in Escherichia coli. Ghlac containing 263 residues was characterized as a functional laccase of the DUF152 family. By structural and biochemical analyses, the conserved residues H78, C119, and H136 were identified to bind with one copper atom to fulfill the laccase activity. In order to make it more suitable for industrial use, Ghlac variant Mut2 with enhanced thermostability was designed. The half-lives of Mut2 at 50 °C and 60 °C were 80.6 h and 9.8 h, respectively. Mut2 was stable at pH values ranging from 4.0 to 8.0 and showed a high tolerance for organic solvents such as ethanol, acetone, and dimethyl sulfoxide. In addition, Mut2 decolorized approximately 100% of 100 mg/L of malachite green dye in 3 h at 70 °C. Furthermore, Mut2 eliminated the toxicity of malachite green to bacteria and Zea mays. In summary, the thermostable laccase Ghlac Mut2 could effectively decolorize and detoxify malachite green at high temperatures, showing great potential to remediate the dyeing wastewater.

This study aims to explore the effect of intermittent sequential pneumatic compression (ISPC) intervention on the coupling relationship between arterial blood pressure (ABP) and changes in oxyhaemoglobin (Δ [O2Hb]). The coupling strength between the two physiological systems was estimated using a coupling function based on dynamic Bayesian inference. The participants were 22 cerebral infarction patients and 20 age- and sex-matched healthy controls. Compared with resting state, the coupling strength from ABP to Δ [O2Hb] oscillations was significantly lower in the bilateral prefrontal cortex (PFC), sensorimotor cortex (SMC), and temporal lobe cortex (TLC) during the ISPC intervention in cerebral infarction patients in interval II. Additionally, the coupling strength was significantly lower in the bilateral SMC in both groups in interval III. These findings indicate that ISPC intervention may facilitate cerebral circulation in the bilateral PFC, SMC, and TLC in cerebral infarction patients. ISPC may promote motor function recovery through its positive influences on motor-related networks. Furthermore, the coupling between Δ [O2Hb] and ABP allows non-invasive assessments of autoregulatory function to quantitatively assess the effect of rehabilitation tasks and to guide therapy in clinical situations.

RNA interference (RNAi) is a powerful tool for the analysis of gene function in nematodes. Fatty acid and retinol binding protein (FAR) is a protein that only exists in nematodes and plays an important role in their life activities. The rice white-tip nematode (RWTN), Aphelenchoides besseyi, is a migratory endoparasitic plant nematode that causes serious damage in agricultural production. In this study, the expression levels of eight RWTN genes were effectively decreased when RWTN was fed Ab-far-n (n: 1–8) hairpin RNA transgenic Botrytis cinerea (ARTBn). These functions of the far gene family were identified to be consistent and diverse through phenotypic changes after any gene was silenced. Such consistency indicates that the body lengths of the females were significantly shortened after silencing any of the eight Ab-far genes. The diversities were mainly manifested as follows: (1) Reproduction of nematodes was clearly inhibited after Ab-far-1 to Ab-far-4 were silenced. In addition, silencing Ab-far-2 could inhibit the pathogenicity of nematodes to Arabidopsis; (2) gonad length of female nematodes was significantly shortened after Ab-far-2 and Ab-far-4 were silenced; (3) proportion of male nematodes significantly increased in the adult population after Ab-far-1, Ab-far-3, and Ab-far-5 were silenced, whereas the proportion of adult nematodes significantly decreased in the nematode population after Ab-far-4 were silenced. (4) Fat storage of nematodes significantly decreased after Ab-far-3, Ab-far-4, and Ab-far-7 were silenced. To our knowledge, this is the first study to demonstrate that Ab-far genes affect sex formation and lipid metabolism in nematodes, which provides valuable data for further study and control of RWTNs.

Silver nanowire (AgNWs) inks for inkjet printing were prepared and the effects of the solvent system, wetting agent, AgNWs suspension on the viscosity, surface tension, contact angle between ink droplet and poly(ethylene) terephthalate (PET) surface, and pH value of AgNWs ink were discussed. Further, AgNWs flexible transparent conductive films were fabricated by using inkjet printing process on the PET substrate, and the effects of the number printing layer, heat treatment temperature, drop frequency, and number of nozzle on the microstructures and photoelectric properties of AgNWs films were investigated in detail. The experimental results demonstrated that the 14-layer AgNWs printed film heated at 60 °C and 70 °C had an average sheet resistance of 13 Ω∙sq−1 and 23 Ω∙sq−1 and average transparency of 81.9% and 83.1%, respectively, and displayed good photoelectric performance when the inkjet printing parameters were set to the voltage of 20 V, number of nozzles of 16, drop frequency of 7000 Hz, droplet spacing of 15 μm, PET substrate temperatures of 40 °C and nozzles of 35 °C during printing, and heat treatment at 60 °C for 20 min. The accumulation and overflow of AgNWs at the edges of the linear pattern were observed, which resulted in a decrease in printing accuracy. We successfully printed the heart-shaped pattern and then demonstrated that it could work well. This showed that the well-defined pattern with good photoelectric properties can be obtained by using an inkjet printing process with silver nanowires ink as inkjet material.

(1) Background: There are currently limited treatments for castration-resistant prostate cancer. Immunotherapy involving Sipuleucel-T has increasingly drawn attention for prostate cancer management. BCG plays a vital role in treating bladder cancer, mainly by inducing immune activation, but is rarely used for prostate cancer. (2) Methods: The TCGA database, PCR, and Western blotting were used to analyze the expression of STEAP1 in mouse and human tissues. Then, we constructed a fusion protein vaccine with Mycobacterium tuberculosis Ag85B and three repeated octapeptide epitopes of a six-transmembrane epithelial antigen of the prostate 1 (STEAP1186-193), Ag85B-3×STEAP1186-193. The uptake of the fusion protein vaccine by DCs was evaluated by confocal microscopy, and DC markers were detected using flow cytometry after incubation with the fusion protein. The immune response against prostate cancer was evaluated by the LDH assay and xenografts in vitro and in vivo. Then, the tumor microenvironment was determined using IHC and ELISA. In addition, the epitope was mutated using CRISPR-Cas9 to illustrate that the fusion protein elicited immunization against STEAP1. (3) Results: The TCGA database analysis, PCR, and Western blotting showed that STEAP1 was highly expressed in human and murine prostate cancer. After the uptake of the purified fusion protein vaccine by DCs, CD11c, CD80, CD86, and MHC II were upregulated and triggered a cytotoxic T lymphocyte (CTL) response against TRAMP-C1 and RM1 cells in vitro. Furthermore, the fusion protein vaccine inhibited tumor growth and improved the tumor microenvironment in vivo, with more CD3+ cells and fewer FOXP3+ cells in the tumor. Serum IFN-γ and IL-2 were significantly higher than in the control group, while IL-4 expression was lower, indicating that the fusion protein vaccine activated Th1 immunity. The immune response against prostate cancer was greatly suppressed when the antigen targets were knocked out using CRISPR-Cas9. (4) Conclusion: In summary, our results provide the first evidence that a vaccine based on a fusion protein consisting of Ag85B and a prostate cancer octapeptide epitope with complete Freund’s adjuvant (CFA), triggers a robust immune response and inhibits tumor growth in murine prostate cancer.

The inkjet printing process is a promising electronic printing technique for large-scale, printed, flexible and stretchable electronics because of features such as its high manufacturing speed, environmental friendliness, simple process, low cost, accurate positioning, and so on. As the base material of printed conductive patterns, conductive ink is the foundation of the development of printed electronics technology, and directly affects the performance and the quality of electronic products. In this paper, conductive ink with silver nanowires (AgNWs) was prepared, with AgNWs of lengths of 2–5 µm and diameters of 20 nm or so, isopropyl alcohol and ethylene glycol as the mixed solvents, and modified polysilane as the wetting agent. We discussed the relationship between the formula of the AgNWs ink and the surface tension, viscosity, contact angle between ink droplet and poly(ethylene) terephthalate (PET) surface, as well as the film-forming properties of the ink. Further, we analyzed the effects of the number of printed layers and the ink concentration of the AgNWs on the microstructures, photoelectric properties and accuracy of the printed patterns, as well as the change in the sheet resistance of the film during different bending cycles. The experimental results show that flexible transparent conductive patterns with a light transmittance of 550 nm of 83.1–88.4% and a sheet resistance of 34.0 Ω∙sq−1–78.3 nm∙sq−1 can be obtained by using AgNWs ink of 0.38 mg∙mL−1 to 0.57 mg∙mL−1, a poly (ethylene terephthalate) (PET) substrate temperature of 40 °C, a nozzle temperature of 35 °C, and heat treated at 60 °C for 10 min. These performances indicate the excellent potential of the inkjet printing of AgNWs networks for developing flexible transparent conductive film.

Synthesizing functional nanomaterials from naturally abundant clay has always been of vital importance for resource utilization, however, the lack of new methods to effectively utilize low-grade clay presents a significant challenge. Herein, a calcium oxalate/sepiolite nanocomposite (SMN-x) was prepared by using the water bath heating method to convert the associated calcium carbonate in low-grade sepiolite into calcium oxalate. The developed composite was subsequently used to remove Pb(II) from the aqueous solutions. The SMN-3 adsorbent prepared by heating in a water bath at 90 °C for 3 h (with a high specific surface area of 234.14 m2·g−1) revealed the maximum Pb(II) adsorption capacity of 504.07 mg·g−1 at pH 5, which was about five times higher than that of sepiolite (105.57 mg·g−1). Further, the SMN-3 adsorbent possessed a much higher selectivity for Pb(II) as compared to the other metal ions. Moreover, the residue was noted to be stable and safe. The adsorption kinetics and isotherms conformed to the quasi-second-order kinetic and Langmuir models. During the adsorption process, ion exchange was noted to the main mechanism, however, it was also accompanied by electrostatic attraction. This study provides a novel strategy for the sustainable development of simple and efficient adsorbents by utilizing low-grade clay minerals.

Sulforaphane (SFN) is a natural glucosinolate found in cruciferous vegetables that acts as a chemopreventive agent, but its mechanism of action is not clear. Due to antioxidative mechanisms being thought central in preventing cancer progression, SFN could play a role in oxidative processes. Since redox imbalance with increased levels of reactive oxygen species (ROS) is involved in the initiation and progression of bladder cancer, this mechanism might be involved when chemoresistance occurs. This review summarizes current understanding regarding the influence of SFN on ROS and ROS-related pathways and appraises a possible role of SFN in bladder cancer treatment.

Plant structures and chemicals, which are developed from the shoot apical meristem (SAM), form the main barriers to insect feeding. A plant chimera containing cells of different genetic origins in the SAM will be morphologically and chemically different compared with the parents and thus may result in differential resistance to herbivores. In this study, we explore if particular elements of plant resistance are localized in one of the layers of SAM; the replacement of one cell layer in a chimera may be linked to change of a single resistance trait to herbivores. The morphology and glucosinolate profiles of two periclinal chimeras (labeled as TTC and TCC, respectively) and grafted parents tuber mustard (labeled as TTT) and red cabbage (labeled as CCC) were compared and the performance of whitefly (Bemisia tabaci) in host selection, oviposition preference and development were assessed under controlled conditions. Both chimeras possessed leaf trichomes as parent tuber mustard TTT, however, TTC had significantly more trichomes than TCC and parent TTT. Leaf wax content of both chimeras was intermediate between the two parents. Five aliphatic and two indole glucosinolates were detected in both chimeras, whereas three aliphatic glucosinolates (3-methyl-sulfinylpropyl, 4-methyl-sulfinylbutyl and 2-hydroxy-3-butenyl) were not detected in tuber mustard, and one aliphatic glucosinolate (3-butenyl) was not detected in red cabbage. Unexpectedly for a chimera, the quantities of two aliphatic glucosinolates (3-methyl-sulfinylpropyl and 4-methyl-sulfinylbutyl) in both TTC and TCC were 3- to 5-fold higher than parents. In olfactory preference assays, B. tabaci showed preference to CCC, followed by TCC, TTC and TTT, and number of eggs laid showed the same pattern: CCC>TCC>TTC>TTT. Interestingly, more whiteflies landed on TTT plants than the other three types in a free choice experiment and the developmental duration from egg to adult was the shortest on TTT and increased in the order TTT

Abstract This study reports an acupuncture needle modified with an iron-porphyrin functionalized graphene composite (FGPC) for real-time monitoring of nitric oxide (NO) release in acupoints of rats. A gold film was first deposited to the needle surface to enhance the conductivity. The FGPC was prepared via hydrothermal synthesis, and subsequently applied to the tip surface of acupuncture needle by electrochemical deposition method. The functionalized needle enabled a specific and sensitive detection of NO based on the favorably catalytic properties of iron-porphyrin and the excellent conductivity of graphene. Amperometric data showed that the needle achieved not only a low detection limit down to 3.2 nM in PBS solution, but also a satisfactory selectivity. Interestingly, the functionalized needle could be inserted into the acupoints of rats for real-time monitoring of NO in vivo. It was found that a remarkable response to NO was respectively obtained in different acupoints when stimulated by L-arginine (L-Arg), revealing that the release of NO was detectable in acupoints. We expect this work would showcase the applications of acupuncture needle in detecting some important signaling molecules in vivo, and exploring the mechanism of acupuncture treatment.

Abstract Radopholus similis is an important parasitic nematode of plants. Serine carboxypeptidases (SCPs) are peptidases that hydrolyse peptides and proteins and play critical roles in the development, invasion, and pathogenesis of certain parasitic nematodes and other animal pathogens. In this study, we obtained the full-length sequence of the SCP gene from R. similis (Rs-scp-1), which is 1665 bp long and includes a 1461-bp open reading frames encoding 486 amino acids with an 18-aa signal peptide. This gene is a double-copy gene in R. similis. Rs-scp-1 was expressed in the procorpus, esophageal glands and intestines of females and in the esophageal glands and intestines of juveniles. Rs-scp-1 expression levels were highest in females, followed by juveniles and males, and lowest in eggs. Rs-scp-1 expression levels were significantly suppressed after R. similis was soaked in Rs-scp-1 dsRNA for 12 h. Nematodes were then inoculated into Anthurium andraeanum after RNAi treatment. Compared with water treatment, R. similis treated with RNAi were reduced in number and pathogenicity. In summary, we obtained the first SCP gene from a plant parasitic nematode and confirmed its role in the parasitic process.
