Abstract

Malus sieversii (Ldb.) Roem. is the original species of modern cultivated apple and a key national essential conservation plant in China. In recent years, degradation and death of wild apple has been exacerbated by imbalances in the rhizosphere micro-ecosystems of wild apple forests due to soil nutrient loss, grazing, climate change and pest and disease outbreaks. However, the structure, diversity and response to environmental factors of wild apple rhizosphere microbial communities are so far unclear. In this study, the rhizosphere bacterial and eukaryotic communities of M. sieversii (Ldb.) Roem. in eight regions of the Yili River were analyzed using 16S/18S rDNA high-throughput sequencing technology. The results indicated that the bacterial operational taxonomic units (OTUs), Shannon index, and community composition were significantly lower in regions A, E, and F than in other regions. By contrast, the dominant eukaryotic communities in all regions were relatively similar in composition and differed less than the relative abundance of bacterial communities. Geographical and climatic distance were found to be key factors influencing the composition and diversity of wild apple rhizosphere microbial communities through mantel analysis. Moreover, these factors above were more correlated with bacterial diversity than with eukaryotes. This study identified the structure of wild apple rhizosphere microbial communities in Xinjiang and their interaction mechanisms under geographical and environmental gradients. 311. Selective brain hypothermia attenuates focal cerebral ischemic injury and improves long‐term neurological outcome in aged female mice.

Abstract

Aims: This study aimed to investigate the effects of mild selective brain hypothermia on aged female ischemic mice. Methods: A distal middle cerebral artery occlusion (dMCAO) model was established in aged female mice, who were then subjected to mild selective brain hypothermia immediately after the dMCAO procedure. Neurological behavioral examinations were conducted prior to and up to 35 days post‐ischemia. Infarct volume, brain atrophy, pro‐inflammation, and anti‐inflammation microglia/macrophages phenotype and white matter injury were evaluated by immunofluorescence staining. Correlations between neurological behaviors and histological parameters were evaluated by Pearson product linear regression analysis. Results: Sensorimotor and cognitive function tests confirmed the protective effect of mild selective brain hypothermia in elderly female ischemic mice. In addition, hypothermia decreased the infarct volume and brain atrophy induced by focal cerebral ischemia. Furthermore, hypothermia alleviated ischemia‐induced short‐term and long‐term white matter injury, which was correlated with behavioral deficits. Finally, hypothermia suppressed the harmful immunological response by promoting the transformation of pro‐inflammatory microglia/macrophages to anti‐inflammatory phenotype. This polarization was negatively correlated with neuronal loss and white matter injury. Conclusion: Mild selective brain hypothermia promoted long‐term functional recovery by alleviating white matter damage in an aged female mouse model of ischemia. [ABSTRACT FROM AUTHOR]

Abstract

Antibiotic residues, generated by the irrational use of drugs and environmental pollution, have always been a great challenge to aquaculture safety. Therefore, a quick, convenient, and performance-excellent way to detect antibiotic residues in aquaculture fish is urgently required. In this study, a multiplex immunochromatographic strip biosensor based on gold nanoparticles was developed for the simultaneous detection of five classes of antibiotic residues (24 β-lactam antibiotics, 26 sulfonamides, five tetracyclines, 24 quinolones, and four amphenicols) in aquaculture fish within 10 min. The detection ranges of five representative antibiotics, penicillin G, sulfamethazine, tetracycline, enrofloxacin, and chloramphenicol, were 2.33–38.4, 0.688–17.1, 1.4–48.1, 1.45–32.9, and 0.537–9.06 µg/kg, respectively. The accuracy and stability of these measurements were demonstrated by analyzing spiked fish samples, with recovery rates of 87.5%–115.2% and a coefficient of variation < 9.5%. The cross-reaction rates, based on the five representative antibiotics, were 3.77%–202% for β-lactams, 3.95%–137% for sulfonamides, 9.19%–100% for tetracyclines, 4.9%–145% for quinolones, and 3.2%–100% for amphenicols. The excellent testing performance of the biosensor strip to most of antibiotic residues in aquaculture fish ensures they meet the maximum residue limits required by countries or organizations. Abstract

Aiming at the problems that the gearbox composite fault vibration signals were susceptible to severe interference from background noise, which made it difficult to accurately separate the impact characteristics of composite faults with traditional methods, an adaptive CYCBD and 1.5-dimensional derivative spectrum were proposed for composite fault diagnosis. First, the cyclic frequency vomponents associated with the fault characteristics in the composite fault vibration signals were detected by using cyclic spectrum analysis to construct cyclic frequency sets of different target types. After that, an improved algorithm for adaptively selecting the optimal filter length of CYCBD with the third-order cumulative sparsity(TCS) as the index was proposed according to different types of cyclic frequency sets, so as to better obtain the optimal filter length containing different fault shock components. Finally, a new 1.5-dimensional derivative spectrum was proposed for feature enhancement to improve the signal-to-noise ratio and to analyze the prominent fault feature frequencies in the spectrum o discriminate the fault types. The algorithm was validated by simulated signals and fault experimental platform data, and the results show that the method may achieve the accurate separation and diagnosis of compound faults in gearboxes. Abstract

Millimeter sized hydrothermal carbon beads (HCB) with excellent acidic resistance and enhanced adsorption performance for Cr(VI) was obtained via hydrothermal treatment of chitosan beads (CSB). Their physicochemical properties were comparatively characterized by Raman spectra, FT-IR, elemental composition analysis, SEM, N2 adsorption-desorption and XPS. The static adsorption shows that the HCB-180 obtained at 180 °C can reduce Cr(VI) concentration from 20 to 0.3 mg L−1 (less than the Chinese standards for industrial wastewater discharge), while CSB can only reduce it to 4.7 mg L−1. Furthermore, removal efficiency of HCB-180 was hardly affected by the co-existing anions including Cl-, NO3-, HPO42- and SO42-. Its breakthrough curve in the dynamic adsorption process could be well fitted by Yoon-Nelson model. It was found that the hydrothermal treatment of CSB is beneficial to expose its active sites. After five cycle experiments, its removal efficiency can still reach 95.85%, while CSB severely dissolved after the second cycle, indicating that the highly stable HCB-180 with easy separation characteristic has great application prospect in the treatment of chromium-containing industrial wastewater.
