Although increasingly used in animal feeding, the black soldier fly (Hermetia illucens - HI) and mealworm (Tenebrio molitor - TM) larvae and pupae require the characterization and modulation of their fat profile for use in the feed industry. In this study, we provide a comprehensive fatty acid profile of the total lipid triglycerides and phospholipids, and sterol composition. For a more detailed characterization of the nutritional properties, we defined the profile of esterified fatty acids (FAs) in the sn-2 position of the triglycerides, which represents the most adsorbed fraction by animals during digestion. Twenty-four batches (12 batches per species divided into 6 batches of larvae and 6 batches of pupae) of 70 subjects were used for the analysis. For each of the developmental stages, individuals in the intermediate stage were considered (2 months and 8 days for TM larvae and pupae respectively; 9 days and 7 days for HI larvae and pupae respectively). Some fatty acids (decanoic, lauric, or myristic acid) were more abundant in HI than in TM. These FAs were produced de novo , indicating that HI produce them from carbohydrates as a source of acetyl-CoA. Hermetia illucens showed a higher level of conjugated linoleic acid (CLA) isomers, a class of fatty acids with positive health effects. In contrast, TM showed a similar fatty acid profile to the plant lipid source (such as soy and sunflower) due to the high level of linoleic acid. Analysis of the triglyceride fraction showed a different affinity for the three glyceride positions. In particular, lauric acid, polyunsaturated fatty acids n-3 and CLA isomers esterified preferentially in sn-2, and therefore were the most adsorbed by the animal during digestion. As for the phospholipids, HI showed a higher level, above all in terms of the phosphatidylethanolamine fraction. Interestingly there was no sphingomyelin in the pupae of both species. The high level of phospholipids is a useful feature for the emulsion of fats in animal feed diets. Lastly, we observed a considerable amount of phytosterol in the sterol profile of both species, which makes these insects a valid alternative to other food sources of plant origin for animal nutrition. The leaf area index (LAI) is a key biophysical variable for agroecosystem monitoring, as well as a relevant state variable in crop modelling. For this reason, temporal and spatial determination of LAI are required to improve the understanding of several land surface processes related to vegetation dynamics and crop growth. Despite the large number of retrieved LAI products and the efforts to develop new and updated algorithms for LAI estimation, the available products are not yet capable of capturing site-specific variability, as requested in many agricultural applications. The objective of this study was to evaluate the potential of non-parametric approaches for multi-temporal LAI retrieval by Sentinel-2 multispectral data, in comparison with a VI-based parametric approach. For this purpose, we built a large database combining a multispectral satellite data set and ground LAI measurements collected over two growing seasons (2018 and 2019), including three crops (i.e., winter wheat, maize, and alfalfa) characterized by different growing cycles and canopy structures, and considering different agronomic conditions (i.e., at three farms in three different sites). The accuracy of parametric and non-parametric methods for LAI estimation was assessed by cross-validation (CV) at both the pixel and field levels over mixed-crop (MC) and crop-specific (CS) data sets. Overall, the non-parametric approach showed a higher accuracy of prediction at pixel level than parametric methods, and it was also observed that Gaussian Process Regression (GPR) did not provide any significant difference (p-value > 0.05) between the predicted values of LAI in the MC and CS data sets, regardless of the crop. Indeed, GPR at the field level showed a cross-validated coefficient of determination (R2CV) higher than 0.80 for all three crops. [ABSTRACT FROM AUTHOR]

Poplar (Populus L. spp.) Short Rotation Coppice systems (SRCs) for bioenergy production are being converted back to arable land. Transitioning to Alley Cropping Systems (ACSs) could be a suitable strategy for integrating former tree rows and arable crops. A field trial (Pisa, Central Italy) was set up with the aim of assessing the C storage of an ACS system based on hybrid poplar and sorghum (Sorghum bicolor L. Moench) and comparing it with that of an SRC cultivation system. The carbon budget at the agroecosystem scale was assessed in the first year of the transition using the net biome production (NBP) approach with a simplified method. The overall NBP for the SRC was positive (96 ± 40 g C m−2 year−1), highlighting that the system was a net carbon sink (i.e., NBP > 0). However, the ACS registered a net C loss (i.e., NBP < 0), since the NBP was −93 ± 56 g C m−2 year−1. In the first year of the transition, converting the SRC into an ACS counteracted the potential beneficial effect of C storage in tree belowground biomass due to the high heterotrophic respiration rate recorded in the ACS, which was fostered by the incorporation of residues and tillage disturbance in the alley. Additional years of heterotrophic respiration measurements could allow for an estimate of the speed and extent of C losses. [ABSTRACT FROM AUTHOR]

Background: Dietary supplements based on tannin extracts or essential oil compounds (EOC) have been repeatedly reported as a promising feeding strategy to reduce the environmental impact of ruminant husbandry. A previous batch culture screening of various supplements identified selected mixtures with an enhanced potential to mitigate ruminal methane and ammonia formation. Among these, Q-2 (named after quebracho extract and EOC blend 2, composed of carvacrol, thymol, and eugenol) and C-10 (chestnut extract and EOC blend 10, consisting of oregano and thyme essential oils and limonene) have been investigated in detail in the present study with the semi-continuous rumen simulation technique (Rusitec) in three independent runs. For this purpose, Q-2 and C-10, dosed according to the previous study, were compared with a non-supplemented diet (negative control, NC) and with one supplemented with the commercial EOC-based Agolin ® Ruminant (positive control, PC)., Results: From d 5 to 10 of fermentation incubation liquid was collected and analysed for pH, ammonia, protozoa count, and gas composition. Feed residues were collected for the determination of ruminal degradability. On d 10, samples of incubation liquid were also characterised for bacterial, archaeal and fungal communities by high-throughput sequencing of 16S rRNA and 26S ribosomal large subunit gene amplicons. Regardless of the duration of the fermentation period, Q-2 and C-10 were similarly efficient as PC in mitigating either ammonia (-37% by Q-2, -34% by PC) or methane formation (-12% by C-10, -12% by PC). The PC was also responsible for lower feed degradability and bacterial and fungal richness, whereas Q-2 and C-10 effects, particularly on microbiome diversities, were limited compared to NC., Conclusions: All additives showed the potential to mitigate methane or ammonia formation, or both, in vitro over a period of 10 d. However, several differences occurred between PC and Q-2/C-10, indicating different mechanisms of action. The pronounced defaunation caused by PC and its suggested consequences apparently determined at least part of the mitigant effects. Although the depressive effect on NDF degradability caused by Q-2 and C-10 might partially explain their mitigation properties, their mechanisms of action remain mostly to be elucidated., (© 2024. The use of alternative feed ingredients from the Agro-industry could be an efficient tool to improve the sustainability of dairy cow production. Since the richness in polyphenols, olive oil pomace (OOP), produced during olive oil milling, seems a promising by-product to ameliorate milk's nutritional value. The aim of this study was to test the use of OOP produced by means of a new technology (biphasic with stone deprivation) in dairy cow feeding strategy to evaluate the effect on animal performances, rumen microbiota, biohydrogenation processes and milk quality by a multidisciplinary approach. Forty multiparous Italian-Friesian dairy cows, at middle lactation, were randomly allotted into two homogenous groups and fed respectively a commercial diet (CON) and the experimental diet (OOPD) obtained by adding OOP to CON as partial replacement of maize silage. The two diets were formulated to be isoproteic and isoenergetic. The same diets were tested also in an in vitro trial aimed to evaluate their rumen degradability (% DEG). The dietary supplementation with OOP did not affect DM intake, rumen % DEG and milk production. The milk's nutritional quality was improved by increasing several important functional fatty acids (FAs; i.e., linoleic acid, conjugated linoleic acid, oleic acid, vaccenic acid). This finding was related to a decrease in rumen liquor biohydrogenation rate of unsaturated FAs. The stochiometric relation between volatile FA production in the rumen and methanogenesis suggested that OOP lowers the methane potential production (CON = 0.050 mol/L vs OOPD = 0.024 mol/L, SEM = 0.005, P = 0.0011). Rumen microbiota and fungi community did not be strongly altered by OOP dietary inclusion because few bacteria were affected at the genus level only. Particularly, Acetobacter, Prevotellaceae&#95;UCG-004, Prevotellaceae&#95;UCG-001, Eubacterium coprostanoligenes, Lachnospira, Acetitomaulatum, Lachnospiraceae&#95;NK3A20 group were more abundant with OOPD condition (P < 0.05). We compared the effect of two different kinds of steam oven-a standard (SO) and a superheated steam (SHS) oven-on four different kinds of samples: hamburgers, bovine steaks, pork steaks, and salmon fillets. Ten samples of each meat/fish were divided into three parts. Then samples were analyzed as (i) raw, (ii) cooked with SO, and (iii) cooked with SHS. For each sample, we determined the proximate composition, fatty acid composition and thiobarbituric acid reactive substances (TBARS). The results of fatty acid composition were processed both with a linear model and with a multivariate approach by using three complementary discriminant analysis techniques: canonical (CAN), stepwise (St) and discriminant (DA). SHS was effective in degreasing hamburgers but not the other kinds of samples. Cooking methods selectively affected the fatty acid profile of samples, SHS being higher in MUFA and lower in PUFA n-3 than SO. This result was also confirmed by the discriminant analysis. Finally, samples cooked with SHS showed a lower fatty acid oxidation extent than SO, as the TBARS value was significantly lower in the SHS than in the SO, irrespective of the type of meat/fish cooked.

The aims of the present study were to evaluate the preventive and the therapeutic effect of Stodi ® as phytogenic feed additive rich in phenolic substances on the calf diarrhea, during the first 24 days of life. A total of 40 calves were included and randomly divided into Group C (control group) and Group T (treated group) with placebo or treatment administration started from the third day of life (T0). Calves belonged to group C received 2 L of warm water, while the calves assigned to group T received 2L of warm water plus 30 g of Stodi ® . Solutions administration was maintained until day 21 (T21) that was the end of the experimental period. Calves were weighed at T0 and T21 to assess the average daily gain (ADG). Physical examination and fecal score evaluation were performed daily. The duration of a diarrheic episode, the age of the first diarrhea outbreak (TDE) and the frequency of diarrheic episodes were recorded. Complete blood count, methemoglobin and liver enzymes were evaluated at T0 and at T21 in all the calves by spectrophotometer and clinical chemistry analysis, respectively. Data were analyzed using a mixed model. A Chi-square and a Mann-Whitney test were also performed. No difference was found for ADG between the groups. The difference of mean age at TDE was not statistically significant between C and T group. The number of calves with diarrhea in the C group tended to be higher than that of T group ( p = 0.13). Calves in group C spent more days with clinical sign of diarrhea compared to group T ( p = 0.016). Complete blood count, methemoglobin and liver enzymes were within the reference ranges. The feed additive Stodi ® seemed to be effective in shortening neonatal diarrhea episodes in calves thanks to the administration of 30 g per day of product. The rumen is characterised by a complex microbial ecosystem, which is particularly active in lipid metabolism. Several studies demonstrated a role of diet and breed on bacterial community profile, with the effect on metabolic pathways. Despite the knowledge achieved on metabolism and the bacterial profile, little is known about the relationship between individual bacteria and metabolic pathways. Therefore, a multivariate approach was used to search for possible relationships between bacteria and products of several pathways. The correlation between rumen bacterial community composition and rumen lipid metabolism was assessed in 40 beef steers (20 Maremmana and 20 Aubrac) reared with the same system and fed the same diet. A canonical discriminant analysis combined with a canonical correlation analysis (CCA) was performed to explore this correlation. The variables showing a Pearson correlation higher than 0.6 as absolute value and significant were retained for CCA considering the relationship of bacterial composition with several metabolic pathways. The results indicated that some bacterial genera could have significant impacts on the presence of several fatty acids. However, the relationship between genera and fatty acid changes according to the breed, demonstrating that the metabolic pathways change according to the host genetic background, related to breed evolution, although there is also an intra-breed genetic background which should not be ignored. In Maremmana, Succiniclasticum and Rikenellaceae&#95;RC9&#95;gut&#95;group showed a high positive correlation with dimethylacetals (DMAs) DMA C13:0 , DMA C14:0 , DMA C14:0iso , DMA C15:0 , DMA C15:0iso , and DMA C18:0 . Prevotellaceae&#95;UCG-003 correlates with C18:3c9c12c15 and C18:1t11, while Fibrobacter and Succiniclasticum correlate with C18:2c9t11 and Lachnospiraceae&#95;NK3A20&#95;group correlates with C18:1c12. Prevotellaceae&#95;UCG-003, Ruminococcaceae&#95;UCG-010, and Oribacterium showed a positive correlation with C13:0iso, and C17:0. Conversely, in Aubrac, Treponema&#95;2 and Rikenellaceae&#95;RC9&#95;gut&#95;group correlated with DMA C14:0iso , DMA C16:0iso , DMA C17:0iso , while Ruminococcaceae&#95;UCG-010, Christensenellaceae&#95;R-7&#95;group and Ruminococcaceae&#95;NK4A214&#95;group correlated with DMA C18:1t11 , DMA C14:0 , DMA C18:1c12 . Acetitomaculum correlated with C18:2c9c12, C18:1c12, C18:1c13, C18:1t12 and Lachnospiraceae&#95;NK3A20&#95;group with C18:1t6-8 and C18:1t9. Saccharofermentas, Ruminococcaceae&#95;UCG-010 and Rikenellaceae&#95;RC9&#95;gut&#95;group correlated with C18:2c9t11 while, Prevotellaceae&#95;UCG-001 and Ruminococcus&#95;1 correlated with C14:0iso, C15:0, C15:0iso, C17:0. Saccharofermentans, Rikenellaceae&#95;RC9&#95;gut&#95;group, Ruminococcaceae&#95;NK4A214&#95;group, and Ruminococcaceae&#95;UCG-010 correlated with C13:1c12 and C16:0iso. These results lead to hypothesise a possible association between several metabolic pathways and one or a few bacterial genera. If these associations are confirmed by further investigations that verify the causality of a bacterial genus with a particular metabolic process, it will be possible to deepen the knowledge on the activity of the rumen population in lipid metabolism.
