PESTICIDE residues in food , *ROHU , *ORGANOCHLORINE compounds , *NITROGEN , *FISH hatcheries , *HIGH performance liquid chromatography - Abstract
Two different weight groups of farmed Labeo rohita were collected from fish hatchery, Satiana Road Faisalabad, Pakistan. Pesticide residues were determined by high-performance liquid chromatography (HPLC). HPLC analysis showed that seven pesticide residues (α-endosulfan, DDE, parathion methyl, isoproturon, atrazine, carbofuran, and deltamethrin) were detected in L. rohita in weight group W 1.Six pesticide residues (α-endosulfan, parathion methyl, isoproturon, atrazine, carbaryl, and carbofuran) were found in L. rohita weight group W 2. The overall comparison between two weight groups of L. rohita showed that maximal concentration of pesticide residues were observed in underweight group W 2 compared to W 1 weight group. - Abstract
Maternal oxidative stress during pregnancy may impair fetal growth and help in the development of diseases in adulthood. The aim of current study was to assess total oxidation status (TOS), related parameters and their relationship to DNA damage (%) and homocysteine level in normal pregnant women in low-income participants. In a cross-sectional study healthy women were grouped as normal, while age matched nulliparous and singleton pregnancies were included for first, second and third trimester groups. TOS ( P < 0.01), melanodialdehyde (MDA) ( P < 0.001), aspartate aminotransferase (AST) ( P < 0.01), triiodothyronine (T3) ( P < 0.01), thyroxine (T4) ( P < 0.01), and homocysteine ( P < 0.001), in pregnant women were significantly higher as compared to normal healthy women. While serum total proteins ( P < 0.01), albumin ( P < 0.01) and total antioxidant status (TAS) ( P < 0.001) decreased significantly as compared to normal healthy women. Women in third trimester showed a significantly high level of body temperature ( P < 0.01), triglyceride ( P < 0.01), LDL-cholesterol ( P < 0.05), AST ( P < 0.01), T3 ( P < 0.01), homocysteine ( P < 0.001), TOS ( P < 0.01) and MDA ( P < 0.001) but a lower concentration of serum proteins, albumin and TAS at the end of the pregnancy. Pearson correlation indicated a positive relationship of homocysteine with triglycerides ( P < 0.027), TOS ( P < 0.01), MDA ( P < 0.035) and had a negative relationship with total protein ( P < 0.026). DNA damage was strongly related with T3 ( P < 0.008), TOS ( P < 0.02), MDA ( P < 0.037) and MBI ( P < 0.048) profiles of pregnant women. These changes were considered normal for pregnant women having optimum blood pressure and normal child birth. Hormonal influences and hemodilution may contribute towards the observed changes in this study. [ABSTRACT FROM AUTHOR]

- Abstract
The article discusses a study which investigated the prevalence of Β-thalassemia disease in hospital population of Faisalabad, Pakistan related to age, consanguinity and anti-hepatitis C virus (HCV) antibody positivity. Results show that males were significantly more affected than females and that in total hospital population, Β-thalassemia major was more frequent than thalassemia intermedia. It concluded that higher total consanguinity rate and coefficient of inbreeding among thalassemic patients indicated genetic basis of the disease and higher anti-HCV positivity among patients reflects the poor facilities of transfusion.

- Abstract
α- and β-Galactosidases find application in food processing, health and nutrition. Aspergillus niger is one of the potent producer of these enzymes and was genotypically improved using gamma-ray induced mutagenesis. The mutant-derivative produced two-fold higher α- and β-galactosidases. For testing genetic variability and its relationship with phenotypic properties of the two organisms, DNA samples of the mutant and parental strains of A. niger were amplified with 28 deca-nucleotide synthetic primers. RAPD analysis showed significantly different pattern between parental and mutant cultures. The mutant derivative yielded homogeneous while parental strain formed heterogeneous amplification patterns. Seven primers identified 42.9% polymorphism in the amplification products, indicating that these primers determined some genetic variability between the two strains. Thus RAPD was found to be an efficient technique to determine genetic variability in the mutant and wild organisms. Both wild and mutant strains were analyzed for their potential to produce galactosidases. Comparison of different carbon sources on enzyme yield revealed that wheat bran is significant ( P < 0.01) effective producer and economical source followed by rice bran, rice polishing and lactose. The mutant was significantly better enzyme producer and could be considered for its prospective application in food, nutrition and health and that RAPD can be effectively used to differentiate mutant strain from the parental strain based on the RAPD patterns. The primary objective of this study was to investigate the seasonal fish and crustacean variations concerning taxonomic composition, species richness, and diversity in sandy beach habitat. For this purpose, we investigated the Sonmiani Hor lagoon area during four distinct seasons, i.e., northeast (NE) monsoon, pre-monsoon, south-west (SW) monsoon, and post-monsoon for one year. During each haul, the net was pulled about 100m along the beach in 0.5m depth. The results showed a strong linear correlation between the diversity index and equitability in fishes (r = 0.978). The diversity index was strong negatively correlated with the abundance and biomass (r = -0.978, -0.972, respectively). The physical attributes like sea surface water temperature and salinity showed a strong negative effect on species assemblages (r = -0.981 and -0.943, respectively). The mean air and water temperature illustrated approximately 3°C difference during NE and pre-monsoon seasons. However, salinity, pH, and electrical conductivity did not show any significant seasonal variabilities. Under the ecological indices, the fish species displayed higher diversity (H’ = 3.19) during SW monsoon, whereas the lowest diversity was observed during pre-monsoon (H’ = 1.58). The equitability and species richness, however, remained more noticeable during SW monsoon (J’ = 0.81). The total number of individuals of fish and crustaceans reached 4799 with 3813 fish individuals and 986 individuals of crustaceans. A total of 27 families of fish while five crustacean families comprising of 30 genera and 38 fish species while ten genera and 17 species of crustaceans were recorded. Liza subviridis displayed the highest abundance among the sampled fish species. In conclusion, fish species constituted a significant part of the coastal fauna in the study area. The seasonal variations displayed distinct variations in fish species composition and diversity.

Untreated sewage and industrial wastes from Faisalabad city are disposed to River Chenab through Chakbandi Main Drain (CMD). The present project is planned to investigate the effects of this freshwater pollution on the body of fish Ictalurus punctatus. The specimens of this fish species were collected upstream and downstream of the entrance of CMD into River Chenab. Fish gills, liver, kidney and muscles from dorsolateral regions of fish were subjected to histopathology. Farmed fish and fish from upstream areas were used as control. Fish collected from polluted experimental sites showed significant damage in selected organs. Gill tissues showed an abnormality in the form of an uplifting of the primary epithelium, fusion, vacuolation, hypertrophy, and necrosis. While liver tissues subjected to hepatocytes degeneration, necrosis, mitochondrial granular hepatocyte, and sinusoids dilation. Kidney tissues indicated increased bowmen space and constricted glomerulus and degenerated nephrons. Edema, necrosis, and atrophy were observed in muscle tissues of fish from polluted areas. Fish from the upstream area showed fused gill lamellae, inflammatory cell infiltration, hypertrophy and vacuolation in hepatocytes. Kidney tissues indicated the presence of nuclear tubular cells, destructive renal tubules, hemorrhage, and necrosis at tubular epithelium. Intra myofibril spaces were also observed in muscles. Specimens of control fish indicated no variation in gills, liver, kidney, and muscles. The present study revealed a strong correlation between the degree of tissue damage and environmental contamination. Present findings also compel global warnings to protect our water bodies and fish to rescue the human population.
