The effectiveness of a community-based parent education and support program in ameliorating risk factors associated with physical child abuse was tested. Data were collected on 34 families at three points in time with a combination of questionnaires, interviews, and telephone contacts. Parental depression and stress were significantly reduced, but social support and child misbehaviors were not. The authors offer suggestions for tailoring programs, including strategies to treat depression and stress, the addition of social-skills training to facilitate involvement in healthy support networks, and the development of a cohesive child program. Parent--child programs that are part of a comprehensive service-delivery system may have the best chance for meeting the needs of violent families. [ABSTRACT FROM AUTHOR]

Current theories of message production typically assume that speakers design messages to accomplish goals, but provide limited detail about how speakers form goals. The cognitive rules (CR) model presents one explicit set of assumptions about how interaction goals are formed. To date, predictions from the model have fared well for individuals high in interpersonal construct differentiation, but have received little support for less differentiated individuals. This chapter offers two suggestions for how the CR model might be elaborated to accommodate individual differences in goal formation. One possibility is that highly differentiated individuals possess more complex schemata for forming goals than do less differentiated individuals, which allow highly differentiated persons to process evaluatively inconsistent information but also make them prone to perceptual biases. A second possibility is that highly and less differentiated individuals rely on different heuristic principles when forming interaction goals, especially in situations where they lack the ability or motivation to process goal-relevant knowledge systematically. Negotiation is a strategic process, carried out through maneuvers designed to accomplish goals. Most negotiation theorists, however, treat goals as global, predetermined, and static task-oriented objectives. This chapter focuses on the role of interaction goals in negotiation. Bargainers' interaction goals are organized within a scheme that varies in two respects: type (instrumental, relational, and identity) and level of abstraction (global, regional, local). Following the discuasion of goal types, the chapter explores the dynamic and conflictual nature of negotiators' interaction goals, and then suggests that goal analyses could shed insight into how bargaining contexts influence strategies and out- comes, and how negotiators develop expertise. A goals focus emphasizes that negotiators act purposefully but with "bounded rationality," constantly manage conflicting objectives, and enact bargaining behaviors that are guided by interaction goals but often refrsrne those goals. The OCCI moves from a stylistic to a strategic conceptualization of conflict by assessing communicative tactics that individuals use in organizational situations. The OCCI consists of items that refer to verbal and nonverbal behaviors and to situational factors that shape choice of conflict behaviors. It also employs three rather than five modes of conflict. Although the article acclaims the internal reliability of the scale, it quibbles with the decision to combine collaborating and compromise and with the apparent item desirability bias inherent in the OCCI and other measures of interpersonal conflict. popular method for assessing compliance-gaming behavior involves having people rate lists of preformulated message strategies for likelihood of use. This 'selection procedure' has been employed much more frequently than the alternative 'construction procedure,' a method that requires people to generate their own message strategies. The present article argues that the selection procedure is much less sensitive than the construction procedure to the effects of situational and individual-difference variables on compliance-gaining behavior. The article further suggests that the insensitivity of the selection procedure is due to a type of social desirability bias known as the item desirability effect. Seven studies were carried out testing the Marwell and Schmitt (1967) and Wiseman and Schenk-Hamlin (1 981) strategy checklists for the item desirability effect. These studies found that (a) the likelihood of use ratings prouided for the strategies on both checklists could be accurately predicted by the rated social appropriateness of the strategies, (b) likelihood of use ratingsfor preforrnulated strategies haue relatively poor reliability, and (c) the construction procedure is much less susceptible to social desirability biases than the selection procedure. This study examined the association between social media use and first-year college students' academic self-efficacy in two large, research intensive universities in Flanders (N = 513) and the United States (N = 431). Given cultivation and social cognitive theories' premises that consistent media messages can shape attitudes and beliefs about self and other, perceptions of others' academic ease (or the perceptions of how difficult it is for peers to do well in college) was included as a mediator. For the U.S., Twitter was directly and indirectly associated with self-efficacy. In the Flanders sample, both Facebook and Twitter had significant direct and indirect effects on self-efficacy. The results' opposite directionality (e.g., Twitter's positive direct effect, Facebook's negative direct effect) suggest the two media operate differently. Contextual differences and theoretical implications for media effects research are discussed along with practical implications for the first-year transition. [ABSTRACT FROM AUTHOR]

The post-positivist articulation of the theory of communicative disenfranchisement (TCD) asserts that people subjected to disenfranchising talk (DT; talk that discredits, silences, and stereotypes) experience proximal consequences (PCs; reduced agency, perceived credibility, and ability to exercise rights and privileges) and make negative inferences about others' future interactional goals (negative goal inferences [NGIs]). Through the mediators of PCs and NGIs, DT is theorized to lead to more distal health and well-being outcomes. This article developed measures of DT, PCs, and NGIs to test the TCD's post-positivist model in the context of chronic pain. Items were developed and refined in a pilot study. Then, in two validation studies, exploratory and confirmatory factor analyses were conducted and health and well-being outcomes were assessed. Findings offer initial empirical support for the reliability and convergent, divergent, and predictive validity of the DT, PCs, and NGIs measures, as well as the model proposed in the post-positivist articulation of the TCD.

Perinatal mortality is a common problem in mouse breeding colonies. Few studies have examined the influence of environmental changes on mouse pup survival. In this study, monogamous breeding cages of C57BL/6J mice were set up and randomized into 3 cage change groups: 1) cage change at 8 d after parturition, 2) cage change at 3 d after parturition, or 3) cage change at 3 d after parturition with the addition of a polycarbonate hut in the cage. Pairs were bred to produce a minimum of 4 litters. Pup survival to weaning relative to experimental cage change date, and survival rates after cage change were evaluated. The results revealed no significant differences between experimental groups. The majority of pup loss occurred within the first 24 h after birth for those pups that were alive at birth. Overall, the postpartum day of cage change did not affect the perinatal survival of mouse pups.

Oxysterols are involved in a plethora of biological processes, including a wide variety of diseases. Therefore, monitoring oxysterols is important for obtaining a deeper understanding of their biological roles and utilizing them as, for example, biomarkers. However, oxysterols can be challenging compounds to study, as they can be very similar in chemical structure but still have distinct biological roles. In addition, oxysterols may be difficult to detect, even with advanced analytical instrumentation. We here focus on the use of liquid chromatography-mass spectrometry (LC-MS) for the analysis of oxysterols, with an additional focus on the steps needed to prepare oxysterols for LC-MS. Steps can include chemical modification of the oxysterols for improving LC-MS sensitivity and adding chemicals that can reveal if the oxysterol levels have been perturbed during preparation. We then round off with descriptions and applications of various sample preparations for different biological matrices, from blood to cells, and biosamples with emerging attention, for example, exosomes and organoids. Taken together, oxysterol analysis is highly compatible with a wide variety of biosamples, allowing for a deeper understanding of these challenging analytes., (© 2024. Integrating concepts from the communication theory of resilience (CTR) with theories of health information management and media-stimulated interpersonal conversation, this study investigates associations between worry about COVID-19, acquiring health information from government, news, and personal sources, and enactment of resilience processes. Findings from a representative sample ( N  = 600) of Floridians show that the extent to which people worried that they or their loves ones might contract COVID-19 during the first wave of the pandemic predicted the extent to which they reported getting health information from a range of news (e.g. national and local newspapers) and personal (e.g. family, friends/coworkers) sources. A serial multiple mediator model found that worry about COVID-19 was associated with acquiring information about COVID from news sources, which predicted getting information about COVID from personal sources, which in turn predicted enactment of resilience processes. Discussion centers on the role that emotions as well as information management at multiple levels (e.g. media, close relationships) play in people's attempts to enact resilience in the context of a public health emergency.

The lack of physiological parity between 2D cell culture and in vivo culture has led to the development of more organotypic models, such as organoids. Organoid models have been developed for a number of tissues, including the liver. Current organoid protocols are characterized by a reliance on extracellular matrices (ECMs), patterning in 2D culture, costly growth factors and a lack of cellular diversity, structure, and organization. Current hepatic organoid models are generally simplistic and composed of hepatocytes or cholangiocytes, rendering them less physiologically relevant compared to native tissue. We have developed an approach that does not require 2D patterning, is ECM independent, and employs small molecules to mimic embryonic liver development that produces large quantities of liver-like organoids. Using single-cell RNA sequencing and immunofluorescence, we demonstrate a liver-like cellular repertoire, a higher order cellular complexity, presenting with vascular luminal structures, and a population of resident macrophages: Kupffer cells. The organoids exhibit key liver functions, including drug metabolism, serum protein production, urea synthesis and coagulation factor production, with preserved post-translational modifications such as N-glycosylation and functionality. The organoids can be transplanted and maintained long term in mice producing human albumin. The organoids exhibit a complex cellular repertoire reflective of the organ and have de novo vascularization and liver-like function. Memorable messages oftentimes are associated with hope, can foster anticipatory resilience prior to the experience of disruptive events, and can buffer the negative impact of adverse experiences on mental health. However, associations among memorable messages, engagement in resilience processes, and mental health have not been examined. Drawing from the communication theory of resilience (CTR), analyses of data from 262 Chinese adults suggested that memorable messages provided a sense of agency and pathways - two interrelated but distinct components of dispositional hope - and functioned as a form of anticipatory resilience that was positively associated with outcomes of coping with disruptive life events including mental health, posttraumatic growth, and meaning in life. These associations were mediated by the extent to which individuals engaged in change- and continuity-related resilience processes as outlined in CTR. Findings offer theoretical implications for memorable message and resilience research as well as practical implications for communicative strategies that can cultivate anticipatory resilience, promote positive adaptation, learning, and reflection, and mitigate the negative effects that disruptive events can have on mental health.

Maintaining compliance with cage density recommendations in The Guide for the Care and Use of Laboratory Animals precludes continuous trio breeding in standard-sized mouse cages. This study evaluated and compared several parameters of reproductive performance, intracage ammonia concentration, and fecal corticosterone levels in 2 strains of mice, C57BL/6J (B6) and B6.129S(Cg)- Stat1 tm1Dlv /J (STAT1 -/- ), housed as continuous breeding pairs or trios in standard-sized mouse cages, and continuous breeding trios in standard-sized rat cages. Reproductive performance data indicated that STAT1 -/- trios raised in rat cages weaned significantly more pups per litter than did STAT1 -/- trios raised in mouse cages, and B6 mice had higher pup survival rates at weaning than did STAT1 -/- mice in mouse cages housing continuous breeding trios. In addition, the Production Index was significantly higher for B6 breeding trios in rat cages than for B6 trios in mouse cages. Intracage ammonia concentration increased with cage density, with significantly higher ammonia concentrations in mouse cages housing trios compared with rat cages housing trios. However, fecal corticosterone levels did not differ significantly regardless of genotype, breeding configuration, or cage size, and daily health checks revealed no clinical abnormalities under any of the conditions evaluated. These results suggest that, although continuous trio breeding in standard-sized mouse cages does not seem to compromise mouse welfare, it offers no advantage in reproductive performance compared with pair breeding, and in some cases, it might be disadvantageous in this regard. Further, high intracage ammonia in mouse cages containing breeding trios might necessitate more frequent cage changes.

Anesthesia in rhesus macaques is required for many procedures. Although ketamine is the backbone of most anesthetic protocols, tolerance to the drug can develop, resulting in the need for higher doses to provide sufficient restraint. Combination with other drugs, such as α-agonists, can be ketamine-sparing, providing for sufficient restraint at lower ketamine doses. In addition, because α-agonists are reversible, recovery from anesthesia has the potential to be much shorter. We hypothesized that use of a low dose of ketamine with a high dose of dexmedetomidine, an α2 receptor selective agonist, in male and female rhesus macaques less than 15 y of age would provide adequate anesthesia for short procedures and that recovery would be faster than in macaques given a higher dose of ketamine (10 mg/kg) alone. We found that the combination, in conjunction with atipamezole for reversal, provided smooth induction of anesthesia and significantly shorter recovery time than did ketamine alone, with no significant effects of sex. The combination of low dose ketamine and high dose dexmedetomidine also provided a 30-min window of anesthesia with analgesia sufficient for mild to moderately painful procedures.

The laryngeal mask airway, the LMA® Gastro™ (Teleflex Medical, Athlone, Ireland), is a novel airway device which permits upper gastrointestinal (GI) endoscopy to be performed via a dedicated large calibre oesophageal lumen. It has been validated in adult studies, but to our knowledge, there have been no data published regarding its use in a paediatric population. Following a brief education programme, the LMA Gastro was introduced on a trial basis at our institution and made freely available. Over a four-month period, our pre-existing endoscopy audit framework captured data on a total of 55 patients who had the LMA Gastro device chosen as the primary airway for elective upper GI endoscopy. These data were collected prospectively by the treating anaesthetist and included a range of airway and endoscopy outcomes. Of the 55 patients, the LMA Gastro provided an adequate airway in 52 (94.5%). Forty-six (88.5%) were sited on first attempt, and 50 (96.2%) insertions were rated 'easy' by the anaesthetist. Aside from three insertion failures, there were no airway events. The endoscopy success rate was 100% in the 52 patients who had an LMA Gastro airway successfully inserted. First-pass oesophageal access was achieved in 51 (98%) cases, and 100% of insertions were rated 'easy' by the gastroenterologist. Despite our relative inexperience with it, the device had a high success rate for airway maintenance and oesophageal access in our paediatric patient population. In our institution, the LMA Gastro provided a useful airway option in older children undergoing elective upper GI endoscopy.

Oxytocin is a pleiotropic, peptide hormone with broad implications for general health, adaptation, development, reproduction, and social behavior. Endogenous oxytocin and stimulation of the oxytocin receptor support patterns of growth, resilience, and healing. Oxytocin can function as a stress-coping molecule, an anti-inflammatory, and an antioxidant, with protective effects especially in the face of adversity or trauma. Oxytocin influences the autonomic nervous system and the immune system. These properties of oxytocin may help explain the benefits of positive social experiences and have drawn attention to this molecule as a possible therapeutic in a host of disorders. However, as detailed here, the unique chemical properties of oxytocin, including active disulfide bonds, and its capacity to shift chemical forms and bind to other molecules make this molecule difficult to work with and to measure. The effects of oxytocin also are context-dependent, sexually dimorphic, and altered by experience. In part, this is because many of the actions of oxytocin rely on its capacity to interact with the more ancient peptide molecule, vasopressin, and the vasopressin receptors. In addition, oxytocin receptor(s) are epigenetically tuned by experience, especially in early life. Stimulation of G-protein-coupled receptors triggers subcellular cascades allowing these neuropeptides to have multiple functions. The adaptive properties of oxytocin make this ancient molecule of special importance to human evolution as well as modern medicine and health; these same characteristics also present challenges to the use of oxytocin-like molecules as drugs that are only now being recognized. SIGNIFICANCE STATEMENT: Oxytocin is an ancient molecule with a major role in mammalian behavior and health. An adult male rhesus macaque ( Macaca mulatta ) that was enrolled in a study evaluating cognition and memory presented with suppurative exudate along the margins of a long-standing cranial implant that included a stainless-steel head post, plastic left-sided recording cylinder, and acrylic over a previously placed right-sided recording cylinder. Both cylinders were located at the level of the prefrontal cortex. After treatment comprising systemic antibiotics and daily cleaning with povidone-iodine for several months, the macaque underwent single-photon emission computed tomography-computed tomography (SPECT-CT) in which his neutrophils were labeled with 99m Tc-hexamethylpropylene amine oxime ( 99m Tc-HMPAO) to evalu- ate for active infection below the implant. Soft tissue inflammation and osteomyelitis were found at the site of the previous right-sided recording cylinder. Cephalosporin and tetracycline antibiotics were administered for 12 wk. Follow-up SPECT-CT imaging was then performed to evaluate response to medical treatment. Results indicated no change in the degrees of soft tissue inflammation and osteomyelitis associated with the right-sided recording cylinder site. SPECT-CT imaging was used to guide the surgical removal of the implant and debridement of the infected tissue. On removal of the entire cranial implant, the osteomyelitis and soft tissue inflammation observed on the pre- and posttreatment SPECT-CT scans were confirmed. In addition, a large cavitary defect through the calvarium with suppurative exudate was discovered below the base of the head post. Infection in this defect was not apparent on SPECT; however, the bony defect was confirmed on reevaluation of the CT images. We concluded that the infection in this defect was silent on SPECT due to the limited vascularization of the sur-rounding bone and the chronicity of the infection. This case study is the first to describe the use of SPECT-CT for evaluating soft tissue inflammation and osteomyelitis beneath a cranial implant in a NHP.

Providing social housing for adult male macaques can be challenging. One successful strategy for long-term social housing of adult male macaques is to pair them with adult females; however, unwanted breeding must be prevented by sterilization of the male or female. Vasectomy is a simple, highly effective, and minimally invasive method of contraception that is used at our institution to facilitate social housing. We performed a retrospective review to analyze the surgical outcomes and rate of postoperative complications after vasectomy of adult rhesus macaques at our research facility. In addition, we evaluated the success rate of pairing vasectomized macaques with female partners. Over 10 y, 16 macaques were vasectomized, of which 5 developed postoperative complications such as orchitis, epididymitis, or surgical site infection. These complications resolved completely and without incident after antibiotic and analgesic therapy; an additional male had postoperative incisional swelling that resolved quickly after NSAID treatment. This complication rate is consistent with that in humans by surgeons who perform open vasectomies relatively infrequently. In addition, 5 of the vasectomized macaques (31%) developed sperm granulomas, which are a common and generally benign complication in humans and have been reported to develop in 40% of macaques after vasectomy. Successful pair housing with a female partner was achieved for 13 of 16 (81%) of the vasectomized macaques. We conclude that surgical vasectomy is a safe and simple procedure that can be used as a highly effective method to facilitate social housing of adult male rhesus macaques in research facilities.

In this study, 87 partners of deployed National Guard service members completed daily diaries in which they recorded for up to 7 consecutive days the channels (e.g., phone) by which they communicated with their service member, the communication activities (e.g., support provision) they and their service member engaged in, and how connected they felt to their service member. Multilevel modeling was used to explore two types of associations between couples' communication activities and partners' feelings of connection for partners who communicated with their service member via phone and/or video during the week. Findings indicated that, across the week, partners who reported that their service member provided them with higher levels of support and who made decisions together more often as a couple felt more connected to their service member (between-person associations). Additionally, on days when partners reported they provided support during phone calls more than they did on average, or their service member provided them support during video calls more than their service member did on average, they reported greater feelings of connection (within-person associations). Future research should explore how daily fluctuations in deployment communication may reinforce or challenge existing relationship processes, thus impacting how couples maintain their relationships after, as well as during, deployment. An adult female beagle (Canis lupus familiaris) used in a model of doxorubicin-induced cardiomyopathy presented with epithelial desquamation on the shoulders and ventrum after receiving the 8th weekly intravenous dose of the free form of doxorubicin (20 mg/m2; total accumulation, 160 mg/m2). The lesions were empirically treated with topical disinfectants and topical and systemic antibiotics. Despite treatment, the lesions progressed and ulcerated. Bacterial culture revealed Staphylococcus aureus, but trichogram, skin scraping, and fungal culture were negative for microorganisms. Skin biopsies revealed epidermal and apocrine gland hyperplasia, apocrine gland dilation, abnormal maturation of epithelial keratinocytes, and perivascular lymphocytic infiltration. These histopathologic findings resemble those in humans and canines after chronic administration of doxorubicin-containing pegylated liposomes. Here we report a clinical presentation after chronic administration of the free form of doxorubicin. In dogs, cutaneous toxicity after administration of pegylated liposomal doxorubicin is most often localized to the footpads, limbs, and axillary and urogenital regions. In the current case, lesions affected the ventrum and trunk but did not involve the footpads or axillary or urogenital regions.

Gray short-tailed opossums are used in a wide variety of research in the areas of developmental biology, oncology, immunology, and comparative biology. Despite many frequent experimental manipulations of these animals under anesthesia, few studies to date have characterized the effects of anesthesia in this species. Our aim was to identify safe and effective injectable anesthetic combinations using ketamine and xylazine or ketamine and dexmedetomidine at doses of 40 mg/kg to 100 mg/kg for ketamine, 5 mg/kg to 10 mg/kg for xylazine, and 0.05 mg/kg to 0.1 mg/kg for dexmedetomidine. Effects of the proposed regimens ranged from light sedation to surgical anesthesia, but only 100 mg/kg ketamine + 0.1 mg/kg dexmedetomidine induced surgical anesthesia in all opossums, with a mean duration of 25.4 min. The 2 lowest doses of ketamine and xylazine (40 mg/kg ketamine + 5 mg/kg xylazine and 40 mg/kg ketamine + 10 mg/kg xylazine) achieved sedation to light anesthesia in all animals but did not produce a surgical plane of anesthesia in any animal. All regimens that induced a surgical plane of anesthesia caused bradycardia and bradypnea, and 75 mg/kg ketamine + 10 mg/kg xylazine and 100 mg/kg ketamine + 0.1 mg/kg dexmedetomidine caused the greatest decreases in SpO2. Except for one opossum that died of unknown causes, all animals remained healthy and apparently free of anesthetic complications. Among all treatments, isoflurane delivered by a precision vaporizer provided the most consistent and reliable anesthesia; therefore, we recommend inhalant anesthesia over the injectable combinations used in this study.

Two aged female rhesus macaques (Macaca mulatta) presented with weight loss and intermittent inappetence. The signalment and constellation of clinical signs led clinicians to suspect the presence of intestinal adenocarcinoma. Because of each animal's advanced age and inconclusive radiographic findings, a noninvasive diagnostic tool was preferred over exploratory laparotomy to assist in determining a diagnosis. Consequently, 2-[(18)F]fluoro-2-deoxy-d-glucose (FDG) positron emission tomography-CT (FDG-PET-CT) was chosen to aid in confirming a suspicion of gastrointestinal adenocarcinoma in both animals. FDG is a glucose analogue labeled with fluorine-18 and is taken up by highly metabolically active cells, as observed in many cancers. Tomography revealed an annular constriction of the small intestine with focal FDG uptake in one animal, and an FDG avid transmural mass in the ascending colon of the second animal. Necropsy later confirmed both sites to be adenocarcinomas. This report supports the use of FDG-PET-CT as an adjunct to conventional radiography in the diagnosis of intestinal adenocarcinoma in nonhuman primates.

The Extended Parallel Process Model (EPPM) posits that an effective fear appeal includes both threat and efficacy components; however, research has not addressed whether there is an optimal threat-to-efficacy ratio. It is possible that varying levels of threat and efficacy in a persuasive message could yield different effects on attitudes, beliefs, and behaviors. In a laboratory experiment, women (n = 442) were exposed to human papilloma virus (HPV) prevention messages containing one of six threat-to-efficacy ratios and one of two message frames (messages emphasizing the connection between HPV and cervical cancer or HPV and genital warts). Multiple mediation analysis revealed that a 1-to-1 ratio of threat to efficacy was most effective at increasing prevention intentions, primarily because it caused more fear and risk susceptibility than other message ratios. Response efficacy significantly mediated the relationship between message framing and intentions, such that participants exposed to a genital warts message reported significantly higher intentions, and this association can be explained in part through response efficacy. Implications for future theoretical research as well as campaigns and intervention research are discussed.

Murine ulcerative dermatitis (UD) is a common progressive condition of mice with a C57BL/6 background. Typically, mice present with scabs and crusts on the skin of the dorsal neck and ears, and are often severely pruritic. Animals tend to scratch the lesions, causing additional trauma to the already ulcerated and inflamed skin. Therapeutic intervention largely has been unsuccessful, in part due to the lack of a known cause for the disease. Though the exact etiology of UD has not been elucidated, substance P (SP) has recently been demonstrated as an important neuropeptide linked to the itch-scratch cycle. SP functions at the tachykinin neurokinin 1 (NK1) receptor. We hypothesized that inhibition of SP binding to the NK1 receptor would decrease the itch sensation, thus decreasing scratching behavior and subsequent skin trauma. The purpose of this study was to evaluate the effectiveness of an NK1 receptor antagonist, maropitant citrate, as a treatment for murine UD. Treatment with 1 mg/kg maropitant citrate significantly reduced the size of UD lesions in mice.

Objective: To clarify the nature and extent of differences in the ways that physically abusive, neglectful, and non-maltreating parents communicate during interactions with their children., Method: A meta-analysis was conducted of 33 observational studies comparing parent-child interactions in families where parents have a documented history of physical abuse or neglect vs. where parents have no history of child maltreatment. Parental behaviors were grouped into three clusters (positivity, aversiveness, and involvement) for comparison across studies., Results: When comparing maltreating (physically abusive or neglectful) vs. non-maltreating parents, mean weighted effect sizes for the three behavioral clusters range from d=.46 to .62. Physically abusive parents are distinguished from non-maltreating parents more so than neglectful parents in terms of aversive behavior, whereas the reverse is true for involvement. Publication date, parent and child age, and task structure moderate the magnitude, though not direction, of differences., Conclusion: Parents with a documented history of child physical abuse or child neglect also are distinguished from non-maltreating parents by the levels of aversiveness, positivity, and involvement they display during interactions that constitute the parent-child relationship., Practice Implications: Researchers and practitioners need to carefully consider sample size, length and setting of observation, and interaction tasks when using observational methods.

Cytomegalovirus is a leading cause of morbidity and mortality among neonatal and immunocompromised patients. The use of vaccine prophylaxis continues to be an effective approach to reducing viral infections and their associated diseases. Murine cytomegalovirus (mCMV) has proven to be a valuable animal model in determining the efficacy of newly developed vaccine strategies in vivo. Live recombinant vesicular stomatitis viruses (rVSV) have successfully been used as vaccine vectors for several viruses to induce strong humoral and cellular immunity. We tested the ability of intranasal immunization with an rVSV expressing the major envelope protein of mCMV, glycoprotein B (gB), to protect against challenge with mCMV in a mouse model. rVSV-gB-infected cells showed strong cytoplasmic and cell surface expression of gB, and neutralizing antibodies to gB were present in mice after a single intranasal vaccination of VSV-gB. After challenge with mCMV, recovery of live virus and viral DNA was significantly reduced in immunized mice. In addition, primed splenocytes produced a CD8+ IFNgamma response to gB. The ability to induce an immune response to a gene product through mucosal vaccination with rVSV-gB represents a potentially effective approach to limiting CMV-induced disease.

In the present work, an orthogonal two-dimensional (2D) capillary liquid chromatography (LC) method for fractionation and separation of proteins using wide range pH gradient ion exchange chromatography (IEC) in the first dimension and reversed phase (RP) in the second dimension, is demonstrated. In the first dimension a strong anion exchange (SAX) column subjected to a wide range (10.5-3.5) descending pH gradient was employed, while in the second dimension, a large pore (4,000 A) polystyrene-divinylbenzene (PS-DVB) RP analytical column was used for separation of the protein pH-fractions from the first dimension. The separation power of the off-line 2D method was demonstrated by fractionation and separation of human plasma proteins. Seventeen pH-fractions were manually collected and immediately separated in the second dimension using a column switching capillary RP-LC system. Totally, more than 200 protein peaks were observed in the RP chromatograms of the pH-fractions. On-line 2D analysis was performed for fractionation and separation of ten standard proteins. Two pH-fractions (basic and acidic) from the first dimension were trapped on PS-DVB RP trap columns prior to back-flushed elution onto the analytical RP column for fast separation of the proteins with UV/MS detection.

A juvenile (1 year old ) female rhesus macaque (Macaca mulatta) developed a chronic active skin condition characterized by pruritus, erythema, alopecia, scaling, exfoliation, and lichenification. Lesions were limited to the ventrum, specifically rostral mandible and neck, axilla and inguinal regions, distal extremities, and interdigital regions. Differential diagnoses included infection, dietary deficiency, metabolic abnormality, endocrinopathy, and immunological injury. Diagnostic tests included complete hemogram, serum chemistry, skin scrapes for ectoparasite detection, hair plucks for dermatophyte culture, and a serum-based hypersensitivity panel. All results were within normal limits. Dermal biopsies revealed lesions consistent with active allergic dermatitis, and a diagnosis of atopic dermatitis was made. Oral cyclosporine (5 mg/kg daily) rapidly eliminated clinical evidence of dermatitis. Histologically, lesions resolved after 12 months of treatment. Atopic dermatitis is an inflammatory skin condition for which there are neither pathognomonic clinical or diagnostic features nor a single successful therapy. Basic criteria such as pruritus, lichenification, a chronic course, and history of allergies strongly support the diagnosis. One successful therapeutic agent is a macrolide calcineurin inhibitor, cyclosporine. It represents a safer class of immunomodulatory drugs than corticosteroids and provides targeted alteration of lymphocyte function. To our knowledge this case represents the first reported successful treatment of atopic dermatitis in a nonhuman primate utilizing cyclosporine.
