Objective: Lupus pericarditis, a common manifestation of systemic lupus erythematosus (SLE), can be fatal. We examined the prevalence of lupus pericarditis and its associated factors in a Taiwanese SLE cohort. Methods: Patients with SLE treated at Change Gung Memorial Hospital between January 2005 and December 2012 were included, and their age, sex, SLE disease duration, SLE disease activity index (SLEDAI) score, laboratory test results, comorbidities, and treatment regimen were noted. Factors related to lupus pericarditis were examined using univariate and multivariate logistic regression analyses. Results: Of the 689 patients, 88.7% were women; age at diagnosis (± standard deviation (SD)) was 40.78 ± 15.59 years, and disease duration at study entry was 11.93 ± 8.21 years. The prevalence of lupus pericarditis was 16.4% (n = 113). Notably, older age at diagnosis (p = 0.0165), longer disease duration (p = 0.009), higher SLEDAI score (p < 0.0001), renal disorder (p = 0.003), lymphocytopenia (p < 0.0001), thrombocytopenia (p = 0.004), and anti-phospholipid antibody (aPL) seropositivity (p = 0.002) were significantly associated with lupus pericarditis. In multivariate analysis, adjusted for sex, SLE disease duration, age, and SLEDAI score, patients with lymphocytopenia and aPL seropositivity were related to a twofold (odds ratio (OR) 2.015, 95% confidence interval (CI) 1.091–3.858) and 1.5-fold (OR 1.569, 95% CI 1.017–2.421) greater prevalence of lupus pericarditis, respectively. Conclusions: Lupus pericarditis occurred in approximately one fifth of patients in this cohort. Patients with SLE with lymphocytopenia or anti-phospholipid antibody seropositivity were associated with a higher rate of lupus pericarditis. Key Points • Lupus pericarditis is a common manifestation of SLE that occurred in one-fifth patients in this study. • Lymphocytopenia and aPL antibody seropositivity are associated with a higher likelihood of developing lupus pericarditis. • Patients with lupus pericarditis should be identify early and treated with caution to prevent further morbidity and mortality. A novel DNA vaccine vector encoding the Mycobacterium tuberculosis secreted antigen Ag85A fused with the influenza A virus (IAV) HA2 protein epitopes, pEGFP/Ag85A-sHA2 (pAg85A-sHA2), was designed to provide protection against influenza. The antigen encoded by the DNA vaccine vector was efficiently expressed in mammalian cells, as determined by reverse transcription polymerase chain reaction (RT-PCR) and fluorescence analyses. Mice were immunized with the vaccine vector by intramuscular injection before challenge with A/Puerto Rico/8/34 virus (PR8 virus). Sera and the splenocyte culture IFN-γ levels were significantly higher in immunized mice compared with the control mice. The novel vaccine group showed a high neutralization antibody titer in vitro. The novel vaccine vector also reduced the viral loads, increased the survival rates in mice after the PR8 virus challenge and reduced the alveolar inflammatory cell numbers. Sera IL-4 concentrations were significantly increased in mice immunized with the novel vaccine vector on Day 12 after challenge with the PR8 virus. These results demonstrated that short HA2 (sHA2) protein epitopes may provide protection against the PR8 virus and that Ag85A could strengthen the immune response to HA2 epitopes, thus, Ag85A may be developed as a new adjuvant for influenza vaccines.

Epichloë fungal endophytes are broadly found in cool-season grasses. The symbiosis between these grasses and Epichloë may improve the abiotic and biotic resistance of the grass plant, but some Epichloë species produce alkaloids that are toxic for livestock. Therefore, it is important to understand the characteristics of the grass-Epichloë s symbiosis so that the beneficial aspects can be preserved and the toxic effects to livestock can be avoided. Since the 1990s, Chinese researchers have conducted a series of studies on grass-Epichloë symbiosis. In this review, we describe the current state of Epichloë endophyte research in Chinese native grasses. We found that more than 77 species of native grasses in China are associated with Epichloë endophytes. In addition, we review the effects of various Epichloë species on native grass responses to abiotic and biotic stress, phylogeny, and alkaloid production. We provide an overview of the study of Epichloë species on native grasses in China and directions for future research.

Objective. Systemic lupus erythematosus (SLE) is an autoimmune disease identified by a plethora of production of autoantibodies. Autoreactive T cells may play an important role in the process. Attenuated T cell vaccination (TCV) has proven to benefit some autoimmune diseases by deleting or suppressing pathogenic T cells. However, clinical evidence for TCV in SLE is still limited. Therefore, this self-controlled study concentrates on the clinical effects of TCV on SLE patients. Methods. 16 patients were enrolled in the study; they accepted TCV regularly. SLEDAI, clinical symptoms, blood parameters including complements 3 and 4 levels, ANA, and anti-ds-DNA antibodies were tested. In addition, the side effects and drug usage were observed during the patients’ treatment and follow-up. Results. Remissions in clinical symptoms such as facial rash, vasculitis, and proteinuria were noted in most patients. There are also evident reductions in SLEDAI, anti-ds-DNA antibodies, and GC dose and increases in C3 and C4 levels, with no pathogenic side effects during treatment and follow-up. Conclusions. T cell vaccination is helpful in alleviating and regulating systemic lupus erythematosus manifestation.

Background Emodin has been proved to have an anti-ischemic effect on the brain, however little research has been done on its effect on vision-threatening retinal ischemia. Thus, an investigation was carried out into the hypothetical efficacy of emodin against retinal ischemia and the role of β-catenin/VEGF in its therapeutic mechanism. Methods Retinal ischemia, followed by reperfusion (IR), was inducted by raising the intraocular pressure of a Wistar rat’s eye to 120 mmHg for 60 min. Additionally, pre-ischemic/post-ischemic intravitreous injections of emodin (4, 10 and 20 μM) or vehicle were carried out on the eye with retinal ischemia. MTT assay, electroretinograms, cresyl violet staining retinal thickness measurements, and fluorogold retrograde labelling of retinal ganglion cells (RGCs) as well as Western blotting were carried out. Results Cultured RGC-5 cells subjected to oxygen glucose deprivation (OGD) were used to confirm the effective concentrations of emodin (administered 1 h pre-OGD, pre-OGD emodin). The most effective and significant (P = 0.04) dose of pre-OGD emodin was observed at 0.5 μM (cell viability: 47.52 ± 3.99%) as compared to pre-OGD vehicle treatment group (38.30 ± 2.51%). Furthermore, pre-ischemic intravitreous injection of 20 μM emodin (Emo20 + IR = 0.99 ± 0.18, P , as compared to pre-ischemic intravitreous vehicle (Vehicle+IR = 0.04 ± 0.02). Post-ischemic intravitreous 20 μM emodin also significantly (P 2), the significant (P 2). As compared to the normal control (the ratio of β-catenin/VEGF to β-actin was set as 1 in the Sham group), the β-catenin/VEGF protein level significantly (P P = 0.02) attenuated by treatment with emodin 20 μM (Emo20 + IR = 1.00 ± 0.19/1.23 ± 0.44). Conclusions The present results suggest that emodin might protect against retinal ischemia insulted neurons such as RGCs by significantly downregulating the upregulation of β-catenin/VEGF protein that occurs during ischemia.

Transition metal-doped electronic deficiency boron clusters have led to a vast variety of electronic bonding properties in chemistry and materials science. We have determined the ground state structures of PdBn0/- (n = 10-20) clusters by performing CALYPSO search and density functional theory (DFT) optimization. The identified lowest energy structures for both neutral and anionic Pd-doped boron clusters follow the structure evolution from two dimensional (2D) planar configurations to 3D distorted Pd-centered drum-like or tubular structures. Photoelectron spectra are simulated by time-dependent DFT theoretical calculations, which is a powerful method to validate our obtained ground-state structures. More interestingly, two "magic" number clusters, PdB12 and PdB16, are found with enhanced stability in the middle size regime studied. Subsequently, molecular orbital and adaptive natural density partitioning analyses reveal that the high stability of the PdB16 cluster originates from doubly σ π aromatic and bonding interactions of d-type atomic orbitals of the Pd atom with tubular B16 units. The tubular C8v PdB16 cluster, with robust relative stability, is an ideal embryo for forming finite and infinite nanotube nanomaterials.

Background X-linked Alport syndrome (XLAS) is the most common form of Alport syndrome (AS), involves mutations in the COL4A5 gene encoding the type IV collagen a5 chain. In this research, we will report the analysis of the COL4A5 gene in a Chinese family with XLAS, and investigate the effect of the missense mutation of this family on type IV collagen. Methods Targeted sequencing using next-generation sequencing (NGS) was conducted for genes (COL4A3/4/5). Normal and mutation COL4A5 plasmids were constructed and then transfected into human podocytes, none plasmid and empty plasmid transfection as control. And then real-time PCR, western blot and indirect immunofluorescence were used to detect the COL4A1/3/5 mRNA, protein, and immunofluorescence expression of each group. Results In this study, we found an Alport family, and the whole exon sequencing found a new missense mutation c.1844G>C in exon 25. The results of real-time PCR, western blot and immunofluorescence showed that in the mutation group, both the mRNA and protein levels of COL4A5 were significantly reduced. Conclusions c.1844G>C is a functional variation of COL4A5, which might play a very important role in the occurrence and development of AS.

Importance A high prevalence of retinal abnormalities have been reported in congenital heart disease (CHD), but quantitative analysis of retinal vasculature is scarce. Optical coherence tomography angiography (OCTA) is a noninvasive method to quantitatively assess the retinal microvasculature. Background To investigate the retinal microvasculature changes in CHD patients by using OCTA. Design Cross‐sectional study. Participants A total of 158 participants including 57 cyanotic CHD (CCHD) patients, 60 acyanotic CHD (ACHD) patients and 41 control subjects were included. This systematic review and meta-analysis was conducted to identify if long-term bosentan is an effective and safe treatment for pulmonary arterial hypertension (PAH) regardless of type, including idiopathic PAH (IPAH), and PAH associated with congenital heart disease (APAH-CHD), connective tissue disease (APAH-CTD), and human immunodeficiency virus (APAH-HIV). All relevant observations were systematically searched by two independent investigators and obtained from three databases, including PubMed, EMBASE and the Cochrane Library, from the inception of each database to February 2020. Currently, long-term administration was defined as no less than 12 months. A random-effects or fixed-effects model was selected according to outcomes of the heterogeneity test for meta-analysis, where standardized mean difference (SMD) with 95% confidence intervals (CIs) was used for continuous outcomes, in addition to the estimated effect (ES; 95% CI) for the synthesized survival rate. Furthermore, subgroup analysis was applied to analyze the differences of efficacy and survivals in each type of PAH cohort. Fifteen studies including a total of 659 subjects undergoing oral bosentan administration for at least 12 months were pooled in this quantitative review. Background Pulmonary complications, including infections, are highly prevalent in patients after hematopoietic cell transplantation with chronic graft-vs-host disease. These comorbid diseases can make the diagnosis of early lung graft-vs-host disease (bronchiolitis obliterans syndrome) challenging. A quantitative method to differentiate among these pulmonary diseases can address diagnostic challenges and facilitate earlier and more targeted therapy. Study Design and Methods We conducted a single-center study of 66 patients with CT chest scans analyzed with a quantitative imaging tool known as parametric response mapping. Parametric response mapping results were correlated with pulmonary function tests and clinical characteristics. Five parametric response mapping metrics were applied to K-means clustering and support vector machine models to distinguish among posttransplantation lung complications solely from quantitative output. Results Compared with parametric response mapping, spirometry showed a moderate correlation with radiographic air trapping, and total lung capacity and residual volume showed a strong correlation with radiographic lung volumes. K-means clustering analysis distinguished four unique clusters. Clusters 2 and 3 represented obstructive physiology (encompassing 81% of patients with bronchiolitis obliterans syndrome) in increasing severity (percentage air trapping 15.6% and 43.0%, respectively). Cluster 1 was dominated by normal lung, and cluster 4 was characterized by patients with parenchymal opacities. A support vector machine algorithm differentiated bronchiolitis obliterans syndrome with a specificity of 88%, sensitivity of 83%, accuracy of 86%, and an area under the receiver operating characteristic curve of 0.85. Interpretation Our machine learning models offer a quantitative approach for the identification of bronchiolitis obliterans syndrome vs other lung diseases, including late pulmonary complications after hematopoietic cell transplantation.

Allograft rejection after renal transplantation remains a challenge to overcome. Interleukin (IL)-21, a cytokine with pleiotropic effects, maintains immune homeostasis post-transplantation. Here, we report higher levels of IL-21 in kidney transplant recipients with non-rejection (NR) than in recipients with T cell-mediated rejection (TCMR, P < 0.001) and antibody-mediated rejection (ABMR, P = 0.005). We observed a negative correlation between IL-21 and creatinine (Cr) levels (P = 0.016). The receiving operating characteristic (ROC) curve showed a promising diagnostic value of IL-21 to identify acute rejection with an area under the curve (AUC) of 0.822 (P < 0.001). In contrast, exogenous administration of IL-21 accelerated acute rejection in a comparative translational kidney transplant (KT) mouse model. Reduced IL-21 levels in the peripheral blood were observed in KT mice after IL-21 injection. Further analysis revealed that increased IL-21 levels in the spleen induced proliferation of CD4+ T cells and CD19+ B cells after IL-21 treatment. Our findings suggest a critical function of IL-21 in kidney transplantation and the potential involvement of the IL-21/IL-21R pathway in acute rejection management.

Background: Hydroxyapatite, which is naturally and synthetically available, is often used as a biomaterial because of its similarity to bone. Aim: In this study, Natural hydroxyapatite powder, synthesized from sheep bone, and synthetic hydroxyapatite were used as matrix. Materials and Methods: Hybrid bio-ceramic composites were obtained by adding 5 wt. % expanded perlite-TiO2-ZrO2-MgO-P2O5 to both matrixes. The bio-ceramic materials which were mixed with mechanical mixer for 30 minutes were pressed with hydraulic press under 25 MPa pressure and sintered at 900°C for 1 hour. Density, micro-hardness, X-Ray Diffraction (XRD), Scanning Electron Microscope (SEM) and Energy Dispersive Spectroscopy (EDS) analysis were performed to determine characteristics of the samples. Results: As a result, it was identified that the micro-hardness of natural hydroxyapatite was higher. In addition, the increase in micro-hardness values of ZrO2-reinforced samples was higher than the TiO2-reinforced samples. Conclusion: Hydroxyapatite, calcium silicate, calcium phosphate structures were observed in XRD analysis. Micro-pores were observed in TiO2-reinforced samples while more dense structures were observed in ZrO2-reinforced samples.

Purpose The on-board flat-panel cone-beam computed tomography (CBCT) lacks molecular/functional information for current online image-guided radiation therapy (IGRT). It might not be adequate for adaptive radiation therapy (ART), particularly for biologically guided tumor delineation and targeting which might be shifted and/or distorted during the course of RT. A linear accelerator (Linac) gantry-mounted on-board imager (OBI) was proposed using a single photon counting detector (PCD) panel to achieve single photon emission computed tomography (SPECT), energy-resolved spectral CT, and conventional CBCT triple on-board imaging, which might facilitate online ART with an addition of volumetric molecular/functional imaging information. Methods The system was designed and evaluated in the GATE Monte Carlo platform. The OBI system including a kV-beam source and a pixelated cadmium zinc telluride (CZT) detector panel mounted on a medical Linac orthogonally to the MV beam direction was designed to obtain online CBCT, spectral CT, and SPECT tri-modal imaging of patients in the treatment room. The spatial resolutions of the OBI system were determined by imaging simulated phantoms. The CBCT imaging was evaluated by a simulated contrast phantom. A PMMA phantom containing gadolinium was imaged to demonstrate quantitative imaging of spectral-CT/CBCT of the system. The capability of tri-modal imaging of the OBI was demonstrated using three different spectral CT imaging methods to differentiate gadolinium, gold, calcium within simulated PMMA and the SPECT to image radioactive 99m Tc distribution. The dual-isotope SPECT imaging of the system was also evaluated by imaging a phantom containing 99m Tc and 123 I. The radiotherapy-related parameters of iodine contrast fraction and virtual non-contrast (VNC) tissue electron density in the Kidney1 inserts of a simulated phantom were decomposed using the Bayesian eigentissue decomposition method for contrast-enhanced CBCT/spectral-CT of the OBI in a single scan. Results The spatial resolutions of CBCT and SPECT of the OBI were determined to be 15.1 lp/cm at 10% MTF and 4.8-12 mm for radii of rotation of 10-40 cm, respectively. In CBCT image of the contrast phantom, most of the soft-tissue inserts were visible with sufficient spatial structure details. As compared to the CBCT image of gadolinium, the spectral CT image provided higher image contrasts. Calcium, gadolinium, and gold were separated well by using the spectral CT material imaging methods. The reconstructed distribution of 99m Tc agreed with the spatial position within the phantom. The two isotopes were separated from each other in dual-isotope SPECT imaging of the OBI. The iodine fractions and the VNC electron densities were estimated in the iodine-enhanced Kidney1 tissue inserts with reasonable RMS errors. The main procedures of the tri-modal imaging guided online ART workflow were presented with new functional features included. Conclusions Using a single photon counting CZT detector panel, an on-board SPECT, spectral CT, and CBCT tri-modal imaging could be realized in Linacs. With the added online molecular/functional imaging obtained from the new OBI for the online ART proposed, the accuracy of radiation treatment delivery could be further improved.

Introduction Gastric cancer (GC), one of the most prevalent malignancies, is the third-leading cause of cancer-related deaths globally. The aim of this study is to investigate the involvement of non-structural maintenance of chromosomes condensin I complex subunit G (NCAPG) in the prognosis of GC. Methods Western blotting and immunostaining were employed to measure the NCAPG level in gastric tissues and cells. Kaplan-Meier analysis was applied to analyze the prognostic value of NCAPG in GC. RNA interference was applied to investigate the influence of the NCAPG silencing on GC cell growth and spread. Results NCAPG overexpression was associated with several clinicopathologic characteristics, including nodal status (P = 0.0378), distant metastasis (P = 0.0088), staging (P = 0.0230), vascular invasion (P = 0.0012), and disease-free survival (P = 0.004). Kaplan-Meier analysis revealed that NCAPG overexpression was positively correlated to poor GC patients disease-free and overall survival (P = 0.004 and P < 0.001, respectively). Univariate Cox regression analysis showed that the overexpression of NCAPG was a prognostic biomarker of GC (P = 0.005). In cultured GC cells, the knockdown of NCAPG suppressed cell proliferation, migration and invasion. Meanwhile, further studies revealed that the NCAPG silencing induces the G0/G1 cell cycle arrest and accordingly represses cell division. Finally, Western blotting showed that NCPAG knockdown dysregulated cell cycle- and epithelial-mesenchymal transition-related molecules. Conclusion Overall, the results reveal that NCAPG overexpression is a candidate prognostic biomarker and potential therapeutic target in GC.

OBJECTIVEThe authors sought to compare the dosimetric quality of hypofractionated stereotactic radiosurgery in treating sizeable brain tumors across the following treatment platforms: GammaKnife (GK) Icon, CyberKnife (CK) G4, volumetric modulated arc therapy (VMAT) on the Varian TrueBeam STx, double scattering proton therapy (DSPT) on the Mevion S250, and intensity modulated proton therapy (IMPT) on the Varian ProBeam.METHODSIn this retrospective study, stereotactic radiotherapy treatment plans were generated for 10 patients with sizeable brain tumors (> 3 cm in longest diameter) who had been treated with VMAT. Six treatment plans, 20–30 Gy in 5 fractions, were generated for each patient using the same constraints for each of the following radiosurgical methods: 1) GK, 2) CK, 3) coplanar arc VMAT (VMAT-C), 4) noncoplanar arc VMAT (VMAT-NC), 5) DSPT, and 6) IMPT. The coverage; conformity index; gradient index (GI); homogeneity index; mean and maximum point dose of organs at risk; total dose volume (V) in Gy to the normal brain for 2 Gy (V2), 5 Gy (V5), and 12 Gy (V12); and integral dose were compared across all platforms.RESULTSAmong the 6 techniques, GK consistently produced a sharper dose falloff despite a greater central target dose. GK gave the lowest GI, with a mean of 2.7 ± 0.1, followed by CK (2.9 ± 0.1), VMAT-NC (3.1 ± 0.3), and VMAT-C (3.5 ± 0.3). The highest mean GIs for the proton beam treatments were 3.8 ± 0.4 for DSPT and 3.9 ± 0.4 for IMPT. The GK consistently targeted the lowest normal brain volume, delivering 5 to 12 Gy when treating relatively smaller- to intermediate-sized lesions (less than 15–20 cm3). Yet, the differences across the 6 modalities relative to GK decreased with the increase of target volume. In particular, the proton treatments delivered the lowest V5 to the normal brain when the target size was over 15–20 cm3 and also produced the lowest integral dose to the normal brain regardless of the target size.CONCLUSIONSThis study provides an insightful understanding of dosimetric quality from both photon and proton treatment across the most advanced stereotactic radiotherapy platforms.

Objective We aim to non-invasively facilitate activation of spared neural circuits after cervical spinal cord injury (SCI) and amyotrophic lateral sclerosis (ALS). We developed and tested a novel configuration for cervical transcutaneous spinal stimulation (cTSS). Methods cTSS was delivered via electrodes placed over the midline at ~T2-T4 levels posteriorly and ~C4-C5 levels anteriorly. Electromyographic responses were measured in arm and hand muscles across a range of stimulus intensities. Double-pulse experiments were performed to assess homosynaptic post-activation depression (PAD). Safety was closely monitored. Results More than 170 cTSS sessions were conducted without major safety or tolerability issues. A cathode-posterior, 2 ms biphasic waveform provided optimal stimulation characteristics. Bilateral upper extremity muscle responses were easily obtained in subjects with SCI and ALS. Resting motor threshold at the abductor pollicis brevis muscle ranged from 5.5 to 51.0 mA. As stimulus intensity increased, response latencies to all muscles decreased. PAD was incomplete at lower stimulus intensities, and decreased at higher stimulus intensities. Conclusions Posteroanterior cTSS has the capability to target motor neurons both trans-synaptically via large-diameter afferents and non-synaptically via efferent motor axons. Significance Posteroanterior cTSS is well tolerated and easily activates upper extremity muscles in individuals with SCI and ALS.

Neodymium ion (Nd3+)-doped yttrium lithium fluoride (LiYF4, YLF) laser crystals have shown significant prospects as excellent laser materials in many kinds of solid-state laser systems. However, the origins of the detailed information of their local structure and luminescence evolution are still poorly understood. Herein, we use an unbiased CALYPSO structure searching technique and density functional theory to study the local structure of Nd3+-doped YLF. Our results reveal a new stable phase with the P4[combining macron] (No. 81) space group for Nd3+-doped YLF, indicating that the host Y3+ ion site was naturally occupied by the Nd3+ ion impurity. On the basis of our newly developed WEPMD method, we adopt a specific type of orthogonal correlation crystal field to obtain a new set of crystal-field parameters as well as 182 complete Stark energy levels. Many absorption and emission lines for Nd3+-doped YLF are calculated and discussed based on Judd-Ofelt theory, and our results indicate that some of the observed absorption and emission lines are perfectly reproduced by our theoretical calculations. Additionally, we predict several promising transition lines in the visible and near-infrared spectral regions, including the electronic dipole emission lines 4F5/2 → 4I9/2 at 808 nm and 2H9/2 → 4I9/2 at 799 nm, as well as the magnetic dipole emission lines 4F3/2(27) → 4I11/2(6) at 1047 nm and 4F3/2(27) → 4I11/2(8) at 1052 nm. These transition channels indicate that Nd3+-doped YLF laser crystals have greatly promising laser actions for serving as a solid-state laser material.

Summary A series of readily accessible and scalable benzo[1,2-b:4,5-b′]dithiophene (BDT)-2,5-dithienyl-thieno[3,4-c]pyrrole-4,6-dione (TPD-T2)-based donor polymers are utilized in organic photovoltaic (OPV) cells blended with the non-fullerene acceptor IT-4F. All polymers readily dissolve in chlorine-free solvents such as xylene, and the corresponding photoactive blend films can be processed in ambient from this solvent to fabricate cells with power conversion efficiencies (PCEs) >12%–14%. Furthermore, the blend processing and OPV metrics are remarkably insensitive to the processing methodology (spin coating versus blade coating), processing solvent, polymer molecular mass and dispersity index, and the results were rationalized by UV-vis, PL, fsTA, AFM, TEM, GIWAXS, and SCLC measurements. These properties enable the first OPV modules, processed in ambient from a benign solvent, with a certified PCE of 10.1% for an area of 20.4 cm2 and >7% after light soaking. The same module also delivers a power of ∼40 μW/cm2 (PCE ∼22%) under indoor lighting.

Anti-perovskite Ca3PN is investigated comprehensively under hydrostatic pressure by combining density functional theory (DFT) with crystal structure predictions. The post-post-perovskite phase is revealed to be part of the anti-perovskite “family” using a special reconstructive (with the same Pnma space groups) phase transition from the perovskite phase under pressure. Ca3PN further undergoes another two reconstructive phase transitions under higher pressure, where one is a semiconductor-to-metal transition and the other is a transformation between another two orthorhombic states – homogeneously with the same space group, i.e., Fmmm. Pressure-induced dynamical stability and mechanical properties of those stable phases are also predicted and explained. Most exciting of all, we even capture the superconducting critical temperature Tc (above 4 K), of the two metal phases in Ca3PN. These findings not only are an insight into an important and alluring process of reconstructive phase transitions in the anti-perovskite “family”, but also uncover unusual properties, thus stimulating experimental interest.

Purpose: This paper investigated a framework to efficiently compute and merge Monte Carlo simulations with new technique development in cancer treatment. A newly designed 3-D prompt gamma (PG) camera system was developed using Monte Carlo computer simulation method to validate the utility of the camera simulated for in-beam proton range verification during cancer treatment. Materials and Methods: We used the TOPAS Monte Carlo code to model a 3-D PG slit-camera system based on pixelated cadmium zinc telluride (CZT) semiconductor detectors. The hierarchy structure of the system modeling includes a top level parameter file and a set of low level parameter files needed to specify particle source, physics setting, geometry, scoring, time-dependent motion, data and graphical output of the system. A high-performance computer cluster to perform the time-consuming computing was used. The effectiveness of the Monte Carlo computer simulation for the system modeling was assessed through statistical simulation experiments. The energy spectra detected by the CZT camera were simulated by irradiating a polymethyl methacrylate (PMMA) phantom with a proton beam of 160 MeV. The axial beam shifting within the PMMA phantom and ranges of the distal beam spots of C-Shape dose distribution in the TG-119 phantom were measured by the modeled camera, respectively. Through simulating a double-head slit camera system, the transverse positions of the proton beam spots were determined based on the measurement of time of flights of photons from positron annihilation events induced by protons within patient. Results: The results from the Monte Carlo computer simulation showed that the energy spectra detected by the CZT camera consisted of main characteristic peaks, including the 4.44 MeV peak from de-excitation of 12C and the 511 KeV peak from positron annihilation interaction. The gamma emission between 4.0 to 5.0 MeV contributed to the majority of the detected profiles. In the PMMA phantom experiments with 2.2 × 108 protons at 160 MeV simulated, a 1 mm standard deviation on range estimation was achieved. With the collimator at 25 cm away from the beam axis, a less than 3 mm standard deviation on range estimation in TG-119 phantom for the individual distal beam spots was obtained. Using the time-of-flight method, the transverse position accuracy of the beam spot measured was less than 1 cm using the simulated double-head camera. Conclusions: The study demonstrated the effectiveness of Monte Carlo computer simulation framework to model the proton beam range monitoring process. Moreover, the computationally modeled proton range verification capability using the 3-D PG camera system, with millimeter accuracy, can be readily translated from computer modeling to medical practice.

The structural phase transition, ferroelectric polarization, and electric properties have been investigated for photovoltaic films CsMI3 (M = Pb, Sn) epitaxially grown along (001) direction based on the density functional theory. The calculated results indicate that the phase diagrams of two epitaxial CsPbI3 and CsSnI3 films are almost identical, except critical transition strains varying slightly. The epitaxial tensile strains induce two ferroelectric phases Pmc21, and Pmn21, while the compressive strains drive two paraelectric phases P212121, P21212. The larger compressive strain enhances the ferroelectric instability in these two films, eventually rendering them another ferroelectric state Pc. Whether CsPbI3 or CsSnI3, the total polarization of Pmn21 phase comes from the main contribution of B-position cations (Pb or Sn), whereas, for Pmc21 phase, the main contributor is the I ion. Moreover, the epitaxial strain effects on antiferrodistortive vector, polarization and band gap of CsMI3 (M = Pb, Sn) are further discussed. Unusual electronic properties under epitaxial strains are also revealed and interpreted.

- Abstract
Cadmium (Cd) contamination in rice is a potential health hazard when ingested through the food chain worldwide. Reducing the Cd content in rice through agronomic measures is an effective way to reduce the risk of Cd contamination to human health. In order to clarify the correlation between temperature and light conditions and Cd accumulation (Cd-A) and Cd content of brown rice (CdBR) during the field growth period (FGP) of rice, consequently provide a theoretical basis for the selection of sowing date (SD) for “Low-Cd-Rice” production, field experiment with different SDs was carried out by using two rice varieties with different Cd accumulation characteristics (Luliangyou 996, V1, a high Cd accumulation variety; Zhuliangyou 819, V2, a low Cd accumulation variety). The results showed that the temperature and light factors such as mean soil temperature (ST), mean air temperature (AT), soil accumulation temperature (SAT), air accumulation temperature (AAT), ultraviolet radiation accumulation (UR), photosynthetic radiation accumulation (PR), light intensity accumulation (I) and sunshine hours accumulation (SH) varied to different degrees under different SDs; The difference in CdBR in two varieties could be up to 2.82 and 8.48 times respectively among SDs, with the CdBR of S4 and S5 of V2 being lower than the national standard of 0.2 mg/kg. The relative expression of OsIRT1 in the root system was significantly positively correlated with ST, SAT, AT, AAT, and SH, while OsNramp5, OsNramp1, and OsHMA3 showed significant negative correlations with ST, SAT, AT, AAT, and SH in relative expression in the root system; OsIRT1 expressed in the roots of V1 was significantly negatively correlated with CdBR, while OsHMA3 expression was significantly positively correlated with CdBR; OsLCD, OsNramp1, and OsHMA3 expression in the roots of V2 were significantly positively correlated with Cd-A and CdBR, while OsIRT1 in the roots of V2 and OsLCT1 in the leaves were significantly negatively correlated with Cd-A; The expression of OsNramp5 in roots was significantly negatively correlated with Cd-A and CdBR in both V1 and V2. Bias correlation analysis showed that ST, SAT, AT, and AAT were significantly negatively correlated with both Cd-A and CdBR; SH was significantly negatively correlated with CdBR in V1. Summarily, the temperature and light conditions during the FGP of rice and their regulation of the expression levels of related genes could be changed by sowing selection, so as to achieve safe production of rice under Cd-contaminated fields.

340. Changes in the content of Puerarin-PLGA nanoparticles in mice under the influence of alcohol and analysis of their antialcoholism

To observe the metabolic changes and antialcoholic effect of Puerarin-PLGA nanoparticles (PUE-NP) in mice. PUE-NP was prepared and characterized by particle size distribution and morphology. The mouse models with acute alcoholism were established to observe their behavioral changes after alcohol poisoning. The expressions of biologically active enzymes such as CRE, BUN, AST, ALT in serum and SOD and TLR4 in liver of mice in each group were detected, and the pathological changes in liver and kidney tissues were observed by HE staining. The PUE-NP metabolism in mice was determined by in vitro release assay and HPLC. PUE-NP nanoparticles had good morphology and structure, and the mouse models with alcohol poisoning were established successfully. Compared with alcohol group, puerarin and PUE-NP increased the disappearance latency time of righting reflex, and the recovery time of righting reflex was significantly shortened. Water maze results showed that Puerarin and PUE-NP had inhibitory effect on impaired memory. HPLC results showed that PUE-NP reached its peak in mice after 1 h, and the content percentage was twice that of puerarin preparation alone, and the distribution time of puerarin concentration in vivo was prolonged, indicating that PLGA nanoparticles had a loading and slow-release effect on puerarin and increased the bioavailability of puerarin in mice. In addition, compared with the alcohol group, Puerarin and PUE-NP improved serum ALT, AST, CRE, and BUN levels in mice, enhanced SOD activity in liver, and inhibited TLR4 expression. The effect was better in the PUE-NP group than in the Puerarin group. PUE-NP delayed the release and metabolism of Puerarin and had better effect in the treatment of the alcoholic liver and kidney injury.

Graphene oxide (GO) is commonly used in photocatalytic reactions but mainly as a cocatalyst. Limited information is available on the intrinsic ability of GO to photocatalytically reduce CO2 as a sole photocatalyst and the activation of light irradiation. In this study, simulated sunlight (SS) and UV-irradiated GO (GOSS and GOUV, respectively) showed enhanced efficiency of photocatalytic reduction of CO2 compared to pristine GO, with a CO yield (4 h) ratio of GOSS/GOUV/GO of 2.7:2.1:1. Here, irradiation plays two important roles: (1) irradiating GO to eliminate CO released under photolysis from photocatalytic reactions and (2) activating GO to create defects and restore the large π-conjugated network, obtaining photolysis-saturated and photoactivated GO for photocatalytic reduction of CO2. The increasing defect density and π conjugation of irradiated GO, as supported by X-ray photoelectron and Raman spectroscopy, improve the yield of photoelectrons and prolong the lifetime of photogenerated charge carriers, as supported by electron spin resonance and transient absorption spectroscopy. This results in an enhanced photocatalytic efficiency of irradiated GO. The higher CO yield of GOSS compared to GOUV indicates that simulated sunlight irradiation is more favorable for GO activation. Our results show that activating GO under irradiation enhances the photocatalytic reduction of CO2.

The absorption and emission transitions of Tm3+-doped LiYF4 have been extensively investigated due to the excellent properties and enormous applications of these materials as laser materials. However, the challenging issues regarding the local structure and luminescence mechanism have not been conclusively established to date. To address these challenges, the CALYPSO structure search method is employed, and the results first reveal the ground-state structure of Tm3+-doped LiYF4, which crystallizes in the space group P4 (No. 81) of the tetragonal system. The Y3+ ions are replaced by Tm3+ ions, forming a local configuration of [TmF8]5-. Furthermore, the complete Stark energy levels of Tm3+-doped LiYF4 are predicted by using our newly developed WEPMD method, which provides preliminary preparation for further spectral exploration. Judd-Ofelt analysis is performed to evaluate the electric dipole transition intensities. Two prominent transitions, 3H5 → 3H6 (1223 nm) and 3H4 → 3H6 (801 nm), are predicted to be good candidates for near-infrared lasers. This study not only is useful for determining the luminescence properties of Tm3+-doped LiYF4 but also offers an effective way to search for other rare-earth-doped lasing crystals for the future design of lasing materials.

343. Retinal Microvasculature Changes in Patients With Coronary Total Occlusion on Optical Coherence Tomography Angiography

Background: Retinal microvasculature has been associated with coronary artery disease (CAD), but the exact contributory role in coronary total occlusion (CTO) is unclear. We aimed to investigate whether retinal vasculature is associated with CTO and could provide incremental value in the assessment of CTO.Methods: A total of 218 CAD patients including 102 CTO and 116 non-CTO were enrolled. Retinal vasculature was measured by optical coherence tomography angiography (OCTA) for all patients. Receiver operating characteristic (ROC) curve was used to assess the performance of retinal vasculature in differentiating CTO from non-CTO patients.Results: In non-CTO CAD patients, vessel density (VD) of mean superficial capillary plexus (SCP) and parafovea SCP were 49.85 and 52.56%, respectively; in CTO patients, VD of mean SCP and parafovea SCP were 47.77, and 49.58%, respectively. After multiple adjustment, VD in the SCP was significantly lower in CTO patients compared to non-CTO patients. VD of superior hemi in the parafovea SCP combined with the clinical variates showed the best ability to predict CTO from CAD with an area under the curve (AUC) of 0.812 (specificity of 89.0% and sensitivity of 65.9%).Conclusions: In CTO patients, retinal VD was significantly decreased, and microvascular damage might specifically target to arterioles than capillaries. Retinal vasculature could thus be a surrogate for detecting the microvascular damage and assist in the assessment of CTO patients. OCTA examination could be suggested to monitor the process of coronary arteries lesions.

Fibroblast-like synoviocytes (FLSs) play a key role in controlling synovial inflammation and joint destruction in rheumatoid arthritis (RA). The contribution of long noncoding RNAs (lncRNAs) to RA is largely unknown. Here, we show that the lncRNA LINK-A, located mainly in cytoplasm, has higher-than-normal expression in synovial tissues and FLSs from patients with RA. Synovial LINK-A expression was positively correlated with the severity of synovitis in patients with RA. LINK-A knockdown decreased migration, invasion, and expression and secretion of matrix metalloproteinases and proinflammatory cytokines in RA FLSs. Mechanistically, LINK-A controlled RA FLS inflammation and invasion through regulation of tyrosine protein kinase 6–mediated and leucine-rich repeat kinase 2–mediated HIF-1α. On the other hand, we also demonstrate that LINK-A could bind with microRNA 1262 as a sponge to control RA FLS aggression but not inflammation. Our findings suggest that increased level of LINK-A may contribute to FLS-mediated rheumatoid synovial inflammation and aggression. LINK-A might be a potential therapeutic target for RA.

We study the observation of polarized stochastic gravitational-wave background (SGWB) in pulsar-timing-array experiments. The time residual for an observed pulsar is formulated as a line-of-sight integral that incorporates the effects of the pulsar term, from which we construct the correlation function of the time residual between a pair of pulsars in terms of the overlap reduction functions (ORFs) for the SGWB intensity and polarization anisotropies.
