Abstract

Operative joint replacement to treat disabling joint conditions secondary to degenerative and inflammatory arthritides has become one of the most efficacious and cost-effective procedures to relieve pain and restore joint function. However, prosthetic implants are not built to last forever and osteolysis and aseptic loosening has been associated with prosthetic arthroplasties since their introduction. The functional life of a synthetic joint is influenced by many factors including the material of the implant, operation procedures and the surgeon involved, as well as patient-related factors. Although promising developments have been achieved in this field, more than 10% of all implants still have to undergo operative revision within 15 years after the initial operation. Failure due to sepsis, fractures and dislocations has become rare; premature loosening of implants on the other hand is becoming much more important. Prosthetic loosening without concurrent infection or trauma is called aseptic loosening. It is generally accepted that small particles ("wear debris") and activated macrophages play a key role in aseptic loosening. The pathophysiology of this condition, however, is still not very well characterized. In this article, we review the molecular mechanisms and signal pathways that were unravelled as responsible factors for loosening orthopaedic implants. Finally, we discuss possible novel strategies for future therapeutic approaches.

353. Peripheral blood B lymphocytes derived from patients with idiopathic pulmonary arterial hypertension express a different RNA pattern compared with healthy controls: a cross sectional study

Abstract

BACKGROUND: Idiopathic pulmonary arterial hypertension (IPAH) is a progressive and still incurable disease. Research of IPAH-pathogenesis is complicated by the lack of a direct access to the involved tissue, the human pulmonary vasculature. Various auto-antibodies have been described in the blood of patients with IPAH. The purpose of the present work was therefore to comparatively analyze peripheral blood B lymphocyte RNA expression characteristics in IPAH and healthy controls. METHODS: Patients were diagnosed having IPAH according to WHO (mean pulmonary arterial pressure > or = 25 mmHg, pulmonary capillary occlusion pressure < or = 15 mmHg, absence of another explaining disease). Peripheral blood B-lymphocytes of patients and controls were immediately separated by density gradient centrifugation and magnetic beads for CD19. RNA was thereafter extracted and analyzed by the use of a high sensitivity gene chip (Affymetrix HG-U133-Plus2) able to analyze 47000 transcripts and variants of human genes. The array data were analyzed by two different softwares, and up-and down-regulations were defined as at least 1.3 fold with standard deviations smaller than fold-changes. RESULTS: Highly purified B-cells of 5 patients with IPAH (mean pulmonary artery pressure 51 +/- 13 mmHg) and 5 controls were analyzed. Using the two different analyzing methods we found 225 respectively 128 transcripts which were up-regulated (1.3-30.7 fold) in IPAH compared with healthy controls. Combining both methods, there were 33 overlapping up-regulated transcripts and no down-regulated B-cell transcripts. CONCLUSION: Patients with IPAH have a distinct RNA expression profile of their peripheral blood B-lymphocytes compared to healthy controls with some clearly up-regulated genes. Our finding suggests that in IPAH patients B cells are activated.

Abstract

Pigeons are known to be able to categorize a wide variety of visual stimulus classes. However, it remains unclear which are the characteristics of the perceptually relevant features employed to reach such good performance. Here, we investigate the relative contributions of texture and shape information to categorization decisions about complex natural classes. We trained three groups of pigeons to discriminate between sets of photorealistic frontal images of human faces according to sex and subsequently tested them on different stimulus sets. Only the pigeons that were presented with texture information were successful at the discrimination task. Pigeons seem to possess a sophisticated texture processing system but are less capable in discriminating shapes. The results are discussed in terms of the possible evolutionary advantages of utilizing texture as a very general and potent perceptual dimension in the birds' visual environment.

Abstract

Background and Aims Daktulosphaira vitifoliae Fitch (grape phylloxera, Phylloxeridae) is one of the most devastating pests in viticulture, mainly because of their root feeding activity. Up to today fundamental relations between belowground habitat and population dynamics remain unclear. In this 2-year study, we investigated the relations between grape phylloxera population and phenotypic traits of roots and feedings sites. Methods and Results We extracted root and soil samples frequently of two closely related mature rootstocks [ Vitis berlandieri × V. riparia (cvs 5C and 125AA)]. We quantified and characterised larval population and crowding and assessed root gall pigmentation, root morphology and soil parameters. We identified and described three stages of seasonal population dynamics: reproduction, overwintering and declining. Further, we demonstrated a significant impact of root gall pigmentation and crowding on population dynamics. Conclusions In temperate climates, grape phylloxera is able to overwinter at high density on roots of American rootstocks. Population dynamics are highly dependent on root gall development and the ability to crowd. Significance of the Study The results of our study are considered to have a significant impact on the development of management strategies for grape phylloxera. [ABSTRACT FROM AUTHOR]

Abstract

Rheumatoid arthritis (RA) is a chronic autoimmune-disease of unknown origin that primarily affects the joints and ultimately leads to their destruction. The involvement of immune cells is a general hallmark of autoimmune-related disorders. In this regard, macrophages, T cells and their respective cytokines play a pivotal role in RA. However, the notion that RA is a primarily T-cell-dependent disease has been strongly challenged during recent years. Rather, it has been understood that resident, fibroblast-like cells contribute significantly to the perpetuation of disease, and that they may even play a role in its initiation. These rheumatoid arthritis synovial fibroblasts (RASFs) constitute a quite unique cell type that distinguishes RA from other inflammatory conditions of the joints. A number of studies have demonstrated that RASFs show alterations in morphology and behaviour, including molecular changes in signalling cascades, apoptosis responses and in the expression of adhesion molecules as well as matrix-degrading enzymes. These changes appear to reflect a stable activation of RASFs, which occurs independently of continuous exogenous stimulation. As a consequence, RASFs are no longer considered passive bystanders but active players in the complex intercellular network of RA

Abstract

It is the ambition of evolutionary psychology to explain how the basic features of human mental life came to be selected because of their contribution to biological survival. Counted among the most basic must be the subjective qualities of conscious sensory experience: the felt redness we experience on looking at a ripe tomato, the felt saltiness on tasting an anchovy, the felt pain on being pricked by a thorn. But, as many theorists acknowledge, with these qualia, the ambition of evolutionary psychology may have met its match. Everyone agrees that a trait can only contribute to an organism's biological survival in so far as it operates in the public domain. Yet almost everyone also agrees that the subjective quality of sensory experience is (at least for all practical purposes) private and without external influence. Then, maybe we must either concede that the subjective quality of sensations cannot after all have been determined by selection (even if this is theoretically depressing) or else demonstrate that the quality of sensations is not as private as it seems to be (even if this is intuitively unconvincing). No. I believe neither of these solutions to the puzzle is in fact the right one. I argue instead that the truth is that the quality of sensations has indeed been shaped by selection in the past, despite the fact that it is today effectively private. And this situation has come about as a result of a remarkable evolutionary progression, whereby the primitive activity of sensing slowly became "privatized" - that is to say, removed from the domain of overt public behavior and transformed into a mental activity that is now, in humans, largely if not exclusively internal to the subject's mind.
