Background: The clinical application of robotic-assisted gastrectomy remains controversial, especially as clinical studies of this operation navigated by carbon nanoparticle suspension injection (CNSI) have not been conducted. This study aims to assess the perioperative safety and efficacy of CNSI-guided robotic-assisted gastrectomy in patients with gastric cancer by focusing on short-term outcomes., Methods: A retrospective analysis of patients who underwent CNSI-guided laparoscopic or robotic-assisted gastrectomy with a pathological diagnosis of gastric cancer was conducted. Data on demographics, surgical management, clinical-pathological results and short-term outcomes were compared among the groups., Results: A total of 126 eligible patients were separated into the robotic-assisted gastrectomy (RAG) group (n = 16) and the laparoscopic gastrectomy (LG) group (n = 110) in total. The operation time of the RAG group is longer than the LG group (p = 0.0000). When it comes to perioperative and short-term complications, there exists no statistical difference between the two groups., Conclusion: The time required for CNSI-guided robotic-assisted gastrectomy is longer than that for CNSI-guided laparoscopic gastrectomy. Background: T-cell malignancies (TCMs), including T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoma (TCL), are highly aggressive and have a poor prognosis. To further understand prognostic stratifications and to design targeted therapies, this study aims to explore novel, potential biomarkers based on alterations in immune costimulatory molecules (CMs) for TCMs., Methods: Peripheral blood from 25 de novo T-ALL patients in our clinical center and transcriptome data from 131 to 162 patients with peripheral TCL (PTCL) from the GSE19069 and GSE58445 dataset, respectively, were obtained to assess the expression levels of CMs and their prognostic significance., Results: Seven CMs were associated with overall survival (OS). Among these CMs, CD5 and CD6 had the highest pairwise positive correlation ( R  = 0.69). CD5 and CD6 were significantly down-regulated in TCM patients compared with healthy individuals (HIs), and lower CD5 and CD6 expression was associated with poor OS for both T-ALL and TCL patients, particularly for patients greater than 60 years old. Furthermore, CD5 was positively correlated with CD6 in TCM patients. Compared with patients who were CD5 high CD6 high , T-ALL and TCL patients who were CD5 low CD6 low had poor OS. Importantly, CD5 high CD6 high was an independent prognostic predictor for OS in T-ALL (HR = 0.39, 95% CI: 0.23-0.65, P < 0.001) and TCL (HR = 0.35, 95% CI: 0.19-0.62, P < 0.001) patients., Conclusions: Low expression of CD5 and CD6 was associated with poor OS for TCM patients, and this may be a potential immune biomarker panel for prognostic stratification of TCM patients., Competing Interests: The authors declare that there are no conflicts of interest., (Copyright © 2022 Songnan Sui et al.)

Background and Aims: Chronic hepatitis B virus (CHB) infection remains a major global health burden and the non-invasive and accurate diagnosis of significant liver fibrosis (≥ F2) in CHB patients is clinically very important. This study aimed to assess the potential of the joint use of ultrasound images of liver parenchyma, liver stiffness values, and patients' clinical parameters in a deep learning model to improve the diagnosis of ≥ F2 in CHB patients., Methods: Of 527 CHB patients who underwent US examination, liver elastography and biopsy, 284 eligible patients were included. We developed a deep learning-based data integration network (DI-Net) to fuse the information of ultrasound images of liver parenchyma, liver stiffness values and patients' clinical parameters for diagnosing ≥ F2 in CHB patients. The performance of DI-Net was cross-validated in a main cohort (n = 155) of the included patients and externally validated in an independent cohort (n = 129), with comparisons against single-source data-based models and other non-invasive methods in terms of the area under the receiver-operating-characteristic curve (AUC)., Results: DI-Net achieved an AUC of 0.943 (95% confidence interval [CI] 0.893-0.973) in the cross-validation, and an AUC of 0.901 (95% CI 0.834-0.945) in the external validation, which were significantly greater than those of the comparative methods (AUC ranges: 0.774-0.877 and 0.741-0.848 for cross- and external validations, respectively, p s  < 0.01)., Conclusion: The joint use of ultrasound images of liver parenchyma, liver stiffness values, and patients' clinical parameters in a deep learning model could significantly improve the diagnosis of ≥ F2 in CHB patients., (© 2022. The nitrate radical (NO 3 ) and dinitrogen pentoxide (N 2 O 5 ) play an important role in the nocturnal atmosphere chemistry. Observations of NO 3 radicals and N 2 O 5 were performed in a semirural ground site at Tai'Zhou in polluted southern China using cavity ring down spectroscopy (CRDS) from 23 May to 15 June 2018. The observed NO 3 and N 2 O 5 concentrations were relatively low, with 1 min average value of 4.4 ± 2.2 and 26.0 ± 35.7 pptV, respectively. The N 2 O 5 uptake coefficient was determined to be from 0.027 to 0.107 based on steady state lifetime method. Fast N 2 O 5 hydrolysis was the largest contributor to the loss of NO 3 and contributed to substantial nitrate formation, with an average value of 14.83 ± 6.01 µg/m 3 . A series of new peptidomimetics targeting the polo-box domain (PBD) of polo-like kinase 1 (Plk1) was identified based on the potent and selective pentapeptide Plk1 PBD inhibitor PLHSpT. Unnatural amino acid residues were introduced to the newly designed compound and the N-terminal substituent of the peptidomimetic was investigated. The optimized compound 9 inhibited the Plk1 PBD with IC 50 of 0.267 μM and showed almost no inhibition to Plk2 PBD or Plk3 PBD at 100 μM. Biolayer interferometry studies demonstrated that compound 9 showed potent binding affinity to Plk1 with a K d value of 0.164 μM, while no K d were detected against Plk2 and Plk3. Compound 9 showed improved stability in rat plasma compared to PLHSpT. Binding mode analysis was performed and in agreement with the observed experimental results. A series of d-amino acid-containing peptidomimetics were designed, synthesized as novel polo-like kinase 1 (Plk1) polo-box domain (PBD) inhibitors based on the reported peptide Plk1 PBD inhibitor. Their inhibitory activity to Plk1, Plk2, and Plk3 PBD were evaluated using our fluorescence polarization (FP) assay. Compound 18 bound to Plk1 PBD with IC 50 of 0.80 μM and showed nearly no inhibition to Plk2 PBD or Plk3 PBD at 100 μM. Compound 18 induced Hela cells to undergo apoptosis by increasing the ratio of the cells at the G2/M phase by decreasing the neosynthesized proteins in a dose-dependent manner from 50 to 150 μM. Compound 18 showed improved stability in rat plasma compared to l-peptide inhibitor LHSpTA. A series of non-peptide inhibitors targeting the polo-box domain (PBD) of polo-like kinase 1 (Plk1) was designed based on the potent and selective minimal tripeptide Plk1 PBD inhibitor. Seven compounds were designed, synthesized and evaluated for fluorescence polarization (FP) assay. The most promising compound 10 bound to Plk1 PBD with IC 50 of 3.37 μM and had no binding to Plk2 PBD or Plk3 PBD at 100 μM. Molecular docking study was performed and possible binding mode was proposed. MM/GBSA binding free energy calculation were in agreement with the observed experimental results. An inexpensive, compact instrument for sensitive measurement of nocturnal nitrogen oxides NO 3 and N 2 O 5 in ambient air at high time resolution has been described. The instrument measures NO 3 and N 2 O 5 which is converted into the NO 3 radical through thermal decomposition by optical extinction using a diode laser at 662.08 nm in two separate detection channels. The minimum detection limits (1σ) for the NO 3 radical and N 2 O 5 are estimated to be 2.3 pptv and 3.1 pptv in an average time of 2.5 s, with the accessible effective absorption path length generally exceeding 30 km, which is sufficient for quantifying NO 3 radical and N 2 O 5 concentrations under moderately polluted conditions. The total uncertainties of the NO 3 and N 2 O 5 measurements are 8% and 15% respectively, which are mainly dominated by the uncertainty of NO 3 across section calculated for 353 K in this system. In addition, the dependence of the instrument's sensitivity and accuracy on a variety of conditions was presented in winter of 2016 and in summer of 2017 during two China-UK joint campaigns. Distinct N 2 O 5 vertical profiles were observed at night in winter. The equilibrium among observed NO 2 , NO 3 and N 2 O 5 based on the equilibrium constants during summer time also provides confirmation of the measurement accuracy of the instrument.

Dinitrogen pentoxide (N 2 O 5 ) is one of the basic trace gases which plays a key role in nighttime atmosphere. An intercomparison and validation of different N 2 O 5 measurement methods is important for determining the true accuracy of these methods. Cavity ring down spectroscopy (CRDS) and cavity enhanced absorption spectrometer (CEAS) were used to measure N 2 O 5 at the campus of the University of Chinese Academy of Sciences (UCAS) from February 21, 2016 to March 4, 2016. The detection limits were 1.6ppt (1σ) at 30s intervals for the CEAS instrument and 3.9ppt (1σ) at 10s time resolution for the CRDS instrument respectively. In this study, a comparison of the 1min observations from the two instruments was presented. The two data sets showed a good agreement within their uncertainties, with an absolute shift of 15.6ppt, slope of 0.94 and a correlation coefficient R 2 =0.97. In general, the difference between the CRDS and CEAS instruments for N 2 O 5 measurement can be explained by their combined measurement uncertainties. However, high relative humidity (>60%) and high PM2.5 concentration (>200μg/m 3 ) may contribute to the discrepancies. The excellent agreement between the measurement by the CRDS and CEAS instruments demonstrates the capability of the two instruments for accurately measuring N 2 O 5 with high sensitivity., (Copyright © 2017 Elsevier B.V. Aim: The aim of this study was to investigate the relationship between the expression of neutral endopeptidase (NEP) and dipeptidyl peptidase IV (DPP IV) proteins, and the clinical significance of the two proteins in patients with intrahepatic cholangiocarcinomas (IHCC)., Methods: Expression patterns and subcellular localizations of NEP and DPP IV proteins in 186 primary IHCC and 60 noncancerous liver tissue specimens were detected by immunohistochemistry., Results: Both the expression of NEP and DPP IV proteins in IHCC tissues were significantly higher than those in noncancerous liver tissues (both P<0.001). Of 186 patients with IHCC, 128 (68.82%) highly expressed both NEP and DPP IV proteins. In addition, the coexpression of NEP and DPP IV proteins was significantly associated with advanced tumor stage (P=0.009), positive lymph node metastasis (P=0.016) and distant metastasis (P=0.013), and the presence of recurrence (P=0.027). Moreover, Kaplan-Meier analysis showed that IHCC patients with high NEP expression, high DPP IV expression, and combined overexpression of NEP and DPP IV proteins all had poorer overall survival and early recurrence after surgery. Furthermore, Cox analysis suggested that NEP expression, DPP IV expression, and combined expression of NEP and DPP IV proteins were all independent prognostic markers for overall survival and recurrence-free survival in patients with IHCC., Conclusion: Our data suggest, for the first time, that both the expression of NEP and DPP IV proteins may be upregulated in human IHCC tissues and the combined expression of NEP and DPP IV proteins may play important roles in progression and prognosis of patients with IHCC.
