Force-feeding was considered as a traditional high-efficiency approach to improve growth performance and accelerate fat deposition of Pekin ducks. However, force-feeding is a serious violation of international advocacy on animal welfare, because it can induce serious injuries to animals, such as damages to the digestive tract, effects on immunity and even severe oxidative stress. Therefore, it is urgent to stop force-feeding. The aim of this study was to determine the effects of force feeding on immune function, digestive function and oxidative stress in the mucosa of duodenum and jejunum of Pekin ducks. A total of 500 ducks were randomly divided into two groups. The control group was allowed to feed freely on a basal diet. The experimental group was force-fed by inserting a plastic feeding tube 8 to 10 inches long down the esophagus for 6 days. Compared with the control group, there was a significant (P <0.05) increase in serum diamine oxidase, d -lactic acid, endotoxin and corticosterone levels in the force-feeding group. The crypt depth in duodenum and jejunum showed significant differences (P <0.05) between the two groups and the intestinal villus epithelium cell was severely damaged in force-feeding group. Similarly, the activities of digestive enzymes as well as the levels of immune function in the duodenal and jejunal mucosa in the force-feeding group were significantly higher than the control group (P <0.05). However, there was a significant decrease in the superoxide dismutase, glutathione peroxidase and catalase levels with a marked increase in malondialdehyde level in duodenal and jejunal mucosa (P <0.05). In summary, at the end of the fattening period with force-feeding for 6 days, Pekin ducks experienced an adverse effect on the integrity of their duodenal and jejunal mucosa epithelium cell as well as their immune function and antioxidant capacity of Pekin ducks but also had improvement in digestive enzyme activities. Hydrogen sulfide (H2S) is regarded as gaseous signaling molecule in plant response to various biotic and abiotic stresses. In this study, we investigated potential role of H2S in modulating photosynthesis in low light (LL) stress-induced tall fescue (Festuca arundinacea Schreb.) seedlings. Results demonstrated that LL significantly reduced the plant height, leaf width, tiller number, dry mass, turf quality, chlorophyll content, net photosynthetic rate, stomatal conductance, intercellular CO2 concentration, transpiration rate, maximal quantum yield of PSII photochemistry, photochemical efficiency of PSII, photochemical quenching, electron transport rate, and enzymatic activity of ribulose-1,5-bisphosphate carboxylase. Then, these parameters were found to be considerably alleviated by H2S donor application. Moreover, exogenous application of NaHS decreased the concentration of malondialdehyde, increased activities of peroxidase, superoxide dismutase, ascorbate peroxidase, total soluble sugar, soluble protein, and endogenous concentration of H2S. In addition, these responses could be reversed by treatment with hypotaurine (H2S scavenger) and aminooxy acetic acid (H2S biosynthesis inhibitor). These results suggested that H2S was possibly involved in the regulation of photosynthesis to strengthen LL tolerance by maintaining a high level of photochemical efficiency and improving antioxidant enzyme activities in tall fescue. OBJECTIVE: Allergic rhinitis (AR) is a chronic inflammatory disease. This study aimed to investigate the role of microRNA-155 (miR-155) in type-2 innate lymphoid cells (ILC2s) on AR. PATIENTS AND METHODS: Nasal mucosa tissues and peripheral blood samples were collected. mRNA expression of miR-155, interleukin- 25 (IL-25), and interleukin-33 (IL-33) in nasal mucosa tissues was determined using quantitative Real Time-Polymerase Chain Reaction (qRTPCR). The AR model was established by injecting with murine IL-33. The frequency of ILC2s was quantified using flow cytometry. MiR-155 agomir or antagomir was intranasally administrated to mice. MiR-155 and helper T cell 2 (Th2) cytokines were measured with quantitative Real-time PCR (qRT-PCR), enzyme-linked immunosorbent assay (ELISA) or Western blotting, respectively. Hematoxylin and eosin (HE) staining was used for the histopathological examination. RESULTS: Compared with controls, mRNA levels miR-155 (p<0.001), IL-25 (p<0.05), and IL-33 (p<0.001) were increased in nasal mucosa tissues of AR patients and AR mice, and ILC2s ratios were enhanced in human peripheral blood (p<0.0001), which were much higher after intranasal administration with miR-155 agomir (p<0.0001). MiR-155 expression of AR mice was significantly reduced after intranasal administration with miR-155 antagomir (p<0.05). Frequencies of ILC2s in human peripheral blood significantly correlated with miR- 155 (r=0.4803, p=0.0130). MiR-155 up-regulation markedly increased frequencies of nasal rubbing/sneezing and levels of IL-4, IL-5, IL-9, and IL-13. Pathological changes were worsened after miR- 155 agomir and ameliorated after miR-155 antagomir administration. MiR-155 agomir mice (p<0.001) showed higher ILC2s, whereas lower in miR-155 antagomir mice compared to AR mice (p<0.05). OBJECTIVE: To elucidate whether microRNA-142 could regulate the development of nasopharyngeal carcinoma (NPC) by mediating gene of phosphate and tension homology deleted on chromosome ten (PTEN) expression. PATIENTS AND METHODS: The microRNA- 142 expression in NPC tissues and paracancerous tissues was detected by the quantitative Real Time-Polymerase Chain Reaction (qRTPCR). Correlation between the microRNA-142 expression and the prognosis of NPC patients was analyzed. MicroRNA-142 expression in NPC cell lines was determined as well. By transfection of microRNA-142 inhibitor or negative control, biological performances of NPC cells were accessed through cell counting kit-8 (CCK-8), colony formation, wound healing, and transwell assay. Dual-luciferase reporter gene assay was conducted to verify the binding condition between microRNA-142 and its target gene PTEN. Rescue experiments were carried out by co-transfection of microRNA-142 inhibitor and si-PTEN, followed by detecting the invasive capacity of NPC cells. Protein expressions of relative genes in the PI3K/AKT pathway after the microRNA-142 knockdown in NPC cells were determined by Western blot. RESULTS: MicroRNA-142 was highly expressed in NPC tissues than that of paracancerous tissues, which was correlated with poor prognosis of NPC patients. MicroRNA-142 was also highly expressed in NPC cells. Downregulated microRNA-142 inhibited proliferative, migratory, and invasive capacities of NPC cells. Dual-luciferase reporter gene assay verified that microRNA-142 could directly bind to PTEN. Knockdown of PTEN could reverse the inhibitory effect of microRNA-142 on invasive capacity of NPC cells. Finally, Western blot results demonstrated that the microRNA-142 knockdown inhibited the PI3K/AKT pathway in NPC cells. CONCLUSIONS: MicroRNA-142 is highly expressed in NPC. MicroRNA-142 enhances the proliferative and invasive capacities of NPC cells by inhibiting PTEN expression, thus promoting NPC development. [ABSTRACT FROM AUTHOR]

N6-methyladenosine (m6A) is the most abundant—internal modification of eukaryotic mRNA. m6A can be installed and removed by specific enzymes. The "writer," "eraser," and "reader" of m6A modification have been reported. These discoveries facilitate our understanding of the functional significance of m6A. m6A plays an essential role in diverse biological processes by recruiting the corresponding YTH domain-containing proteins, as well as recruiting additional translation initiation factors. Here, we provide an update on the various aspects of YTH domain-containing proteins, including an introduction to the YTH domain, the categories, distribution in cells, and biological roles of YTH proteins. OBJECTIVE: Mounting evidence has suggested that microRNAs (miRNAs) play crucial roles in the progression of nasopharyngeal carcinoma (NPC). However, the molecular mechanism remains not fully understood. We aimed to examine the expression and biological function of miR-122-5p in NPC. MATERIALS AND METHODS: Quantitative Real Time-Polymerase Chain Reaction was conducted to examine the expression of miR- 122-5p. Cell Counting Kit-8 assay, colony formation assay, wound healing assay and transwell assay were performed to measure cell proliferation, colony formation, migration and invasion. Luciferase reporter assay and Western blot assay were used to confirm the target gene of miR-122-5p. RESULTS: The results showed that miR-122- 5p was significantly downregulated in NPC cell lines. Additionally, it was demonstrated that special AT-rich sequence-binding protein 1 (SATB1) was targeted by miR-122-5p. Furthermore, our results revealed that miR-122-5p inhibits cell proliferation, colony formation, cell migration and cell invasion by targeting SATB1.
