Abstract: Biodefense vaccines are developed against a diverse group of pathogens. Vaccines were developed for some of these pathogens a long time ago but they are facing new challenges to move beyond the old manufacturing technologies. New vaccines to be developed against other pathogens have to determine whether to follow traditional vaccination strategies or to seek new approaches. Advances in basic immunology and recombinant DNA technology have fundamentally transformed the process of formulating a vaccine concept, optimizing protective antigens, and selecting the most effective vaccine delivery approach for candidate biodefense vaccines. As young novice drivers are inclined to getting involved in traffic accidents due to their improper emergency response under sorts of gender affective state, namely, mood, widely generated in fast-paced urban life, it is of great necessary to study the impact of mood state on responsive capacity for young novice drivers. Fourteen college students were recruited to take part in complex reaction experiments for this pilot study. Each subject's mood was collected through a simplified POMS scale, while their complex reaction time (CRT) and response error rate (RER) were acquired during the experiments. The study results showed that young novice drivers' RER was significantly positively correlated (pc=0.323 ∗ ∗ , "pc" omitted next) with their score of total mood disturbance (TMD), and a logarithmic regression model was feasible to describe the correlations with a good fitting effect. Further, their RER was also significantly positively correlated with score of negative mood state components such as nervousness (0.290 ∗ ∗ ), anger (0.300 ∗ ∗ ), fatigue (0.278 ∗ ∗ ), depression (0.287 ∗ ∗ ), and fluster (0.261 ∗ ), and a quadratic or cubic regression model was suitable to describe the correlations. Additionally, the young novice drivers' CRT was significantly positively correlated with score of nervousness (0.222 ∗ ∗ ), vigorousness (0.227 ∗ ), and fluster (0.273 ∗ ), and a quadratic or exponential regression model was suitable to describe the correlations. The results can provide theoretical support for developing targeted intervention to improve young novice drivers' emergency response capacity for driving training or traffic management authorities. Background: Hypoxia naturally happens in embryogenesis and thus serves as an important environmental factor affecting embryo development. Hif-1α, an essential hypoxia response factor, was mostly considered to mediate or synergistically regulate the effect of hypoxia on stem cells. However, the function and relationship of hypoxia and Hif-1α in regulating mesendoderm differentiation remains controversial. Results: We here discovered that hypoxia dramatically suppressed the mesendoderm differentiation and promoted the ectoderm differentiation of mouse embryonic stem cells (mESCs). However, hypoxia treatment after mesendoderm was established promoted the downstream differentiation of mesendoderm-derived lineages. These effects of hypoxia were mediated by the repression of the Wnt/β-Catenin pathway and the Wnt/β-Catenin pathway was at least partially regulated by the Akt/Gsk3β axis. Blocking the Wnt/β-Catenin pathway under normoxia using IWP2 mimicked the effects of hypoxia while activating the Wnt/β-Catenin pathway with CHIR99021 fully rescued the mesendoderm differentiation suppression caused by hypoxia. Unexpectedly, Hif-1α overexpression, in contrast to hypoxia, promoted mesendoderm differentiation and suppressed ectoderm differentiation. Knockdown of Hif-1α under normoxia and hypoxia both inhibited the mesendoderm differentiation. Moreover, hypoxia even suppressed the mesendoderm differentiation of Hif-1α knockdown mESCs, further implying that the effects of hypoxia on the mesendoderm differentiation were Hif-1α independent. Consistently, the Wnt/β-Catenin pathway was enhanced by Hif-1α overexpression and inhibited by Hif-1α knockdown. As shown by RNA-seq, unlike hypoxia, the effect of Hif-1α was relatively mild and selectively regulated part of hypoxia response genes, which fine-tuned the effect of hypoxia on mESC differentiation. Conclusions: This study revealed that hypoxia is fine-tuned by Hif-1α and regulates the mesendoderm and ectoderm differentiation by manipulating the Wnt/β-Catenin pathway, which contributed to the understanding of hypoxia-mediated regulation of development. The efficient conversion of lignite is important for environmental protection. Coal‐based fulvic acid (FA) was prepared by the oxidative degradation of lignite in a microwave field. The highest extraction rate of FA was 35.03 %. To reduce the heterogeneity of FA, it was divided into four fractions‐FAI, FAII, FAIII and FAIV‐with molecular weights of <500, 500–1000, 1000–3500, and >3500 Da, respectively. A variety of characterisation methods revealed that all the FA fractions contained carboxyl, phenolic hydroxyl, amino and sulfhydryl groups. The FAI contained the most oxygen‐containing functional groups. The benzene rings of the >1000 Da FA fractions were mostly tri‐substituted. FA could break dormancy and promote the germination of coreopsis seeds but this effect was weakened when the FA concentration was too high. FAI could effectively promote chlorophyll synthesis, FAII could effectively improve plant stress resistance and FAIII promoted seedling longitudinal growth. [ABSTRACT FROM AUTHOR]

Summary: After polyploidization originated from one interspecific hybridization event in Gossypium, Gossypium barbadense evolved to produce extra‐long staple fibres than Gossypium hirsutum (Upland cotton), which produces a higher fibre yield. The genomic diversity between G. barbadense and G. hirsutum thus provides a genetic basis for fibre trait variation. Recently, rapid accumulation of gene disruption or deleterious mutation was reported in allotetraploid cotton genomes, with unknown impacts on fibre traits. Here, we identified gene disruptions in allotetraploid G. hirsutum (18.14%) and G. barbadense (17.38%) through comparison with their presumed diploid progenitors. Relative to conserved genes, these disrupted genes exhibited faster evolution rate, lower expression level and altered gene co‐expression networks. Within a module regulating fibre elongation, a hub gene experienced gene disruption in G. hirsutum after polyploidization, with a 2‐bp deletion in the coding region of GhNPLA1D introducing early termination of translation. This deletion was observed in all of the 34 G. hirsutum landraces and 36 G. hirsutum cultivars, but not in 96% of 57 G. barbadense accessions. Retrieving the disrupted gene GhNPLA1D using its homoeolog GhNPLA1A achieved longer fibre length in G. hirsutum. Further enzyme activity and lipids analysis confirmed that GhNPLA1A encodes a typical phospholipase A and promotes cotton fibre elongation via elevating intracellular levels of linolenic acid and 34:3 phosphatidylinositol. Our work opens a strategy for identifying disrupted genes and retrieving their functions in ways that can provide valuable resources for accelerating fibre trait enhancement in cotton breeding. Gastric carcinoma (GC) heterogeneity represents a major barrier to accurate diagnosis and treatment. Here, we established a comprehensive single-cell transcriptional atlas to identify the cellular heterogeneity in malignant epithelial cells of GC using single-cell RNA sequencing (scRNA-seq). A total of 49,994 cells from nine patients with paired primary tumor and normal tissues were analyzed by multiple strategies. This study focused on the malignant epithelial cells, which were divided into three subtypes, including pit mucous cells, chief cells, and gastric and intestinal cells. The trajectory analysis results suggest that the differentiation of the three subtypes could be from the pit mucous cells to the chief cells and then to the gastric and intestinal cells. Lauren's histopathology of GC might originate from various subtypes of malignant epithelial cells. The functional enrichment analysis results show that the three subtypes focused on different biological processes (BP) and pathways related to tumor development. In addition, we generated and validated the prognostic signatures for predicting the OS in GC patients by combining the scRNA-seq and bulk RNA sequencing (bulk RNA-seq) datasets. Background: Alberta Stroke Program Early Computed Tomography Score (ASPECTS) is a grading system to assess the extent and distribution of early ischemic changes. Purpose: To assess inter-rater agreement for total and regional ASPECTS on non-contrast computed tomography (NCCT) images, CT angiography source images (CTA-SI), and CT-perfusion cerebral blood volume (CTP-CBV) maps, and their association with final infarction in patients with acute ischemic stroke (AIS). Material and Methods: A total of 96 consecutive patients with AIS who underwent pre-treatment NCCT and CTP were retrospectively enrolled. CTA-SI was reconstructed using the raw data of CTP. Total and regional ASPECTS were assessed on baseline NCCT, CTA-SI, and CTP-CBV, and on follow-up NCCT or diffusion-weighted imaging. Follow-up ASPECTS served as the reference standard for final infarction. Results: CTP-CBV demonstrated higher concordance for total ASPECTS (interclass correlation coefficient, 0.895 vs. 0.771 vs. 0.777) and regional ASPECTS in internal capsule, lentiform, caudate nuclei, M5 and M6, compared with NCCT and CTA-SI. CTP-CBV showed a trend of stronger correlation with final ASPECTS than NCCT and CTA-SI (0.717 vs. 0.711 vs. 0.565; P > 0.05). ASPECTS in the internal capsule (ρ, 0.756 vs. 0.556; P = 0.016) and caudate nucleus (ρ, 0.717 vs. 0.476; P = 0.010) on CTP-CBV were more strongly correlated with follow-up ASPECTS than NCCT. CTP-CBV showed higher accuracy for predicting final infarction in the internal capsule (92.5% vs. 90.3% and 87.1%; P > 1.000, P = 0.125, respectively) and caudate nucleus (87.1% vs. 79.6% and 77.4%; P = 0.453, P = 0.039, respectively) than CTA-SI and NCCT. A bionic tactile device is designed for object surface texture recognition, taking inspiration from the microstructure of human fingerprints. The sense of touch in humans is achieved through the frictional vibration and generation of electrical potential signals by subcutaneous receptors. Constructing a tactile sensing device involves generating distinct signals upon interacting with different materials. The piezoelectric film PVDF is particularly suitable as a sensitive material for sensors due to its excellent flexibility, strong mechanical strength, excellent dynamic response and cost-effectiveness. This paper presents the design of a PVDF-based fingerprint-inspired tactile sensor capable of differentiating various textures. By combining the collected signals with machine learning algorithms, diverse textures can be effectively identified. To demonstrate the sensor's superior performance, two experiments were conducted—one focused on recognizing different material textures, and the other on recognizing Braille characters. The accuracy achieved in these experiments was 97.4% and 96.5%, respectively, highlighting the technology's significant potential in intelligent robotics and human-computer interaction. Endometrial elasticity is a potential new marker for assessing endometrial receptivity and pregnancy outcomes based on endometrial thickness and type. Currently, little research has been conducted on the elasticity of the endometrium using shear wave elasticity imaging (SWEI). This study aimed to explore whether endometrial elasticity is an ultrasound marker for predicting clinical pregnancy outcomes after embryo transfer. A total of 245 infertile women underwent ultrasonography before embryo transfer at the Peking University Third Hospital. We compared the endometrial elasticity and sub-endometrial blood flow rate using SWEI in the groups with different pregnancy outcomes. Trends in clinical pregnancy outcomes across the quartiles of endometrial elasticity in the fundus of the uterus (E1) were assessed. Logistic regression analysis was performed to obtain odds ratios for clinical pregnancy outcomes based on the quartiles of E1, with or without adjusting for potential confounding variables. Women in the clinical pregnancy group had higher E1 values and sub-endometrial blood flow rates in the uterine fundus than those in the non-pregnancy group. Women in the highest quartile of E1 had the most favorable clinical pregnancy rates. SnO 2 double-shelled and triple-shelled hollow spheres were tailored by adjusting concentration of tin (IV) chloride solution during the process of the tin (IV) ions infused carbonaceous spheres. The structures of these SnO 2 multi-shelled hollow spheres were examined by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and their possible formation mechanism were also discussed. In virtue of triple-shelled hollow porous structure and higher specific surface area, SnO 2 triple-shelled hollow spheres exhibited enhanced photocatalytic properties compared to SnO 2 double-shelled hollow spheres. Amino acid substituted zinc phthalocyanines which are capable of reversing their charge from negative to positive in response to tumor microenvironment have been prepared by the isoelectric pHs regulation and charge reversal sensitivity enhancement. The charge reversal abilities of the prepared zinc phthalocyanines were judged by their isoelectric pHs and zeta potential values. The UV–Vis spectra, singlet oxygen generation ability, cellular uptake and in vitro anticancer ability of the prepared zinc phthalocyanines were investigated to study the influences of charge reversal on photodynamic activities. Charge reversible tetra- and octa-substituted glutamic acid ZnPcs showed enhanced singlet oxygen generation ability, cellular uptake and photocytotoxicity towards cancer cells, and were considered as promising photosensitizers for photodynamic therapy. There have been numerous studies concerning the associations of angiotensin-converting enzyme (ACE) gene insertion/deletion (I/D), angiotensin II receptor 1 (AT1R) gene A1166C, and endothelial nitric oxide synthase (eNOS) gene 4b/a polymorphisms with risk of pregnancy hypertensive disorders (PHDs). However, the results are inconsistent. A total of 83 eligible studies (10,354/18,446 cases/controls) were included in this meta-analysis. Pooled odds ratios with corresponding 95% confidence intervals were used to calculate these associations. The effects of ethnicity and types of PHDs were also considered. Results showed significant associations between the ACE gene polymorphism and PHDs in all of the populations except that in Africa. The associations also existed in AT1R, eNOS gene polymorphism and PHDs in part of the gene models in the overall population. These results indicated the ACE gene polymorphism was associated with an increased risk of PHDs, whereas the eNOS and AT1R gene polymorphism only have increased susceptibility to PHDs in part of the gene models. In this work, we develop a facile template method for the controlled synthesis of Mn 2 O 3 triple-shelled hollow spheres and porous spheres by simply adjusting the concentration of manganese nitrate aqueous solution during the process of the infusion of Mn(II) ions into carbonaceous spheres. The bidirectional reflectance model is commonly used to study surface structure and composition of atmosphereless celestial bodies basing on photometric measurements. We conducted a test of two bidirectional reflectance models, which are theoretically similar but with different form, to assess their ability for calculating the bidirectional reflectance of particulate surfaces and if the parameters could be confidently linked to the surface׳s property. Two types of natural particulate surfaces with controlled particle sizes vary from 300 μm to 900 μm have been measured in the visible and near-infrared wavelength with the NENULGS (Northeast Normal University Laboratory Goniospectrometer System), we only used these measurement results at 560 nm and 670 nm which are regarded to the evaluation standard of models. In this range of particle size, the bidirectional reflectance models were well match to the experimental data as the results shown in previous publications. Although some parameters of the models can be used to simulate the reflectance of particulate surface, they contain no reliable information on the physical property of our samples. Furthermore, the influence of the number of viewing angles on the precision of modeling results has been tested in this paper. It is clear that an increase of the number of viewing angles and the range of azimuth angles could allow us to improve precision on the estimation of parameters. Comparing with the best fitted model reflectance, we also found that if we used the parameters, which derived from measurements in the principal plane for individual incident zenith angle, to model the bidirectional reflectance may overestimate the computed results in the backward scattering direction and underestimate the computed results in the forward scattering direction. The difference between modeled results and measurements can reach up to 20% in the backward direction when using the parameters inverted in the principal plane. However, if we used the parameters, which derived from the combined measurements in the principal plane for two incident zenith angles, to model the bidirectional reflectance, the maximum difference reaches up to 50% in the backward direction. In the context of experimental measurement study, we suggest that there must be enough measured results at different viewing and azimuth angles when using the bidirectional reflectance models, which are considered as semi-empirical at best, to describe the scattering property or surface structure of particulate system as shown in this study. Background/Aims: Hepatocyte nuclear factor-4α (HNF4α), the liver enriched transcription factor (TF), is one of the major regulators of hepatocyte differentiation and proliferation. However, how HNF4α participate in liver regeneration after partial hepatectomy (PH) remains largely unknown. In order to identify the HNF4α-centered regulatory network, we applied an integrated analytic strategy, in which, TF array, mRNA microarray, bioinformatic analysis and ChIP-on-chip assays were integrated. Methods/Results: The TF signatures from MOUSE OATFA (TF-array) platform revealed that the activity of HNF4α was significantly reduced and 17 other TFs showed increased activity at 4 h post PH. Then the ChIP-on-chip analysis on HNF4α were combined with mRNA expression profiling to select the possible HNF4α target genes during early liver regeneration, which were then sub-grouped according to their signaling pathways. More specifically, the HNF4α target genes with the gene ontology (GO) terms of cytokine-cytokine receptor, Jak-STAT, MAPK, toll-like receptor and insulin signaling pathways were further analyzed with an advanced bioinformatics tool named oPOSSUM to identify TF binding sites occupancy and predict the co-regulatory relationship between TFs and targets. Furthermore, we identified that repressed HNF4α during the early phase of liver regeneration may contribute cooperatively to the induction of immediate early genes, such as, c-fos, c-jun and stat3. Conclusion: our data indicate that HNF4α may play an inhibitory role on the induction of specific promitogenic genes and liver regeneration initiation. The integrated approach of mRNA/OATFA/ChIP-DSL/oPOSSUM analysis may help us better characterize the target genes and co-regulatory network of HNF4α during the early stage of liver regeneration. Purpose: To assess the diagnostic accuracy of using Alberta Stroke Program Early Computed Tomography Score (ASPECTS) on CT perfusion (CTP) map to predict a volumetric target mismatch in patients with acute ischemic stroke (AIS). Methods: Three-hundred and seven AIS patients with an onset time within 24h or unclear onset time who underwent CTP evaluation for large vessel occlusion of anterior circulation were enrolled. CTP ASPECTS was evaluated on cerebral blood flow (CBF) and time-to-maximum (Tmax) colored maps, respectively. Automated perfusion analysis software was used to calculate the volumes of ischemic core (volumeCBF<30%) and tissue at risk (volumeTmax>6s). Target mismatch was defined as volumeCBF< 30%<70ml, volumemismatch≥15ml, and volumeTmax >6s/volume CBF< 30%≥1.8. Spearman correlation and receiver operating characteristic curves were used for statistical analyses. Results: Strong correlations were found between CBF ASPECTS and volumeCBF<30%, and between Tmax ASPECTS and volumeTmax>6s for overall population (ρ=−0.872, −0.757) and late-arriving patients (ρ=−0.900, −0.789). Mismatch ASPECTS moderately correlated with mismatch volume for overall population (ρ=0.498) and late-arriving patients (ρ=0.407). A CBF ASPECTS≥5 optimally predicted an ischemic core volume<70ml in overall population (sensitivity, 94.4%; specificity, 80.4%) and late-arriving patients (sensitivity, 89.5%; specificity, 90.5%). A CBF ASPECTS≥6 combined with a Mismatch ASPECTS≥1 optimally identified a target mismatch in overall population (sensitivity, 84.5%; specificity, 77.0%) and late-arriving patients (sensitivity, 83.7%; specificity, 90.0%). Conclusion: CTP ASPECTS might be useful in predicting target mismatch derived from automated perfusion analysis software, and assisting in patient selection for endovascular therapy. Coamorphous drug delivery systems have shown great potential in improving the solubility and bioavailability of poorly water-soluble drugs. However, the coamorphous formation mechanism remains unclear. In this study, coamorphous andrographolide (AP) - oxymatrine (OMT) was successfully prepared by solvent evaporation and characterized by PXRD and DSC. The potential groups of intermolecular interactions in coamorphous binary systems were predicted by the molecular electrostatic potential (ESP) in the density functional theory (DFT) and the HOMO and LUMO in the molecular frontier orbitals, which had been confirmed by FTIR, FT-NIR, and Raman spectroscopy. Molecular dynamics (MD) was used to explore the mechanism of coamorphous formation. The results of the solvent-state radial distribution function (RDF) showed that the hydrogen bonds between AP and OMT molecules were irregular and intermittent in the solvent system of AP-OMT-methanol (MeOH) until MeOH was completely volatilized to form a stable coamorphous AP-OMT system, which has been preliminarily verified by solvent-state FT-NIR and solvent-state Raman spectroscopy. In addition, the solubility of coamorphous AP-OMT was significantly higher than that of crystalline AP. The results of intrinsic dissolution experiments showed that the intrinsic dissolution rate (IDR) of coamorphous AP-OMT was 2.5 times that of crystalline AP, although recrystallization occurred during dissolution. In conclusion, the results of the experimental solvent-state RDF, solvent-state FT-NIR, and solvent-state Raman spectroscopy can be used to better understand the coamorphous formation process and to guide the preparation of coamorphous AP-OMT. The organic Rankine cycle (ORC) has been widely studied to recover waste heat from internal combustion engines in commercial on-road vehicles. To achieve a cost-effective ORC, a trade-off between factors such as costs, power outputs, back pressure, and weight needs to be carefully worked out. However, the trade-off is still a huge challenge in engine waste heat recovery. In this study, a thermoeconomic optimization study of a vehicle-mounted ORC unit is proposed to recover waste heat from various exhaust gas conditions of a light-duty vehicle. The optimization is carried out for four organic working fluids with different critical temperatures, respectively. Under the investigated working fluids, the lower specific investment cost (SIC) and higher mean net output power (MEOP) of ORC can be achieved using the organic working fluid with higher critical temperature. The maximum mean net output power is obtained by taking RC490 as working fluid and the payback period (PB) is 3.01 years when the petrol is EUR 1.5 per liter. The proposed strategy is compared with a thermodynamic optimization method with MEOP as an optimized objective. It shows that the proposed strategy reached SIC results more economically. The linear quadratic Gaussian (LQG) control for a quadrotor unmanned aerial vehicle (UAV) under false data injection attacks is studied. The LQG control depends on optimal state estimation, while this type of attacks makes the optimal state estimation unimplementable in practice. To address this problem, the authors prose a framework to detect attacks and then augment the information for controller design. Based on this framework, the LQG control is designed and a sufficient condition for the security of the closed‐loop control system is given. Stem cells maintain adult tissue homeostasis under physiological conditions. Uncontrolled stem cell proliferation will lead to tumorigenesis. How stem cell proliferation is precisely controlled is still not fully understood. Phosphorylation of Yun is essential for ISC proliferation. Yun is essential for the proliferation of normal and transformed intestinal stem cells. Our mass spectrometry and biochemical data suggest that Yun can be phosphorylated at multiple residues in vivo. Interestingly, we show that the phosphorylation among these residues is likely interdependent. Furthermore, phosphorylation of each residue in Yun is important for its function in ISC proliferation regulation. This paper concerns a fractional modeling and prediction method directly oriented toward an industrial time series with obvious non-Gaussian features. In the modern chemical industry process, soft sensing has been widely used. However, the lack of valid and sufficient data has made it difficult to apply advanced soft sensor modeling methods to realistic scenarios. In this paper, a novel soft sensing method based on deep learning is proposed to handle the problem of small data. Aiming at handling the issue of small sample size, a novel virtual sample generation method embedding a deep neural network as a regressor into conditional Wasserstein generative adversarial networks with gradient penalty (rCWGAN) is presented. In rCWGAN, conditional variables are introduced to make the training supervised and a dual training algorithm is specially designed. With the advanced structure and the designed training algorithm, rCWGAN has powerful sample generation capabilities and can well predict quality variables. Finally, an experiment on the purified terephthalic acid (PTA) solvent system is carried out for the validation of the presented rCWGAN. Polydimethylsiloxane (PDMS), as a polymer material with excellent mechanical properties and flexibility, is widely used in flexible wearable devices, soft robots and other fields. Commonly, the Mold-flipping is used to produce PDMS based samples, while it is time-consuming and impractical for the large-scale fabrication. With the additive manufacturing, printing pastes can be freely configured thanks to direct ink writing (DIW) technology, enabling rapid and high precision manufacturing. Herein, we have improved the rheology of Sylgard-184 by adding crosslinkers and silica nanoparticles to make it printable. By adjusting the printing path, flexible surfaces with textures were created. Ball-on-disc dry friction experiments were carried out on the specimens. Results show that the texture produced by the print path can effectively reduce the coefficient of friction (COF) compared to samples produced by the conventional molding. When the print path is at 60° to the sliding direction, COFs for samples are reduced by 23.2% to 32.9%. Sunscreen represents a large variety of creams which, in the analytical point of view, exhibit a similar matrix. Such matrix corresponds to a semi-solid emulsion of mixture of oil and water. The formulation of a cream can include metal and nonmetal elements in different contents in order to realize specific pharmaceutical or cosmetic functions designed for the product. The complex matrix of these materials makes their analysis challenging for classical elemental analytical techniques with specific and complicated sample pretreatment procedures needed for reliable quantification. In this work we demonstrate and assess direct determination, without any pretreatment, of elemental content, especially for metallic element such as titanium, in a sunscreen using laser-induced breakdown spectroscopy (LIBS). The used configuration corresponds to that of indirect ablation of a thin film of cream applied on the surface of a pure aluminum target. We especially focused, in this work, on the case of high concentration of TiO 2 nanoparticle in cream. Such choice was justified first by the fact that such concentration level is usually found in commercial sunscreens. On the other hand, titanium presents a large number of lines, neutral as well as singly ionized, in the spectral range from the near UV to the near IR. It provides therefore an ideal case to study line selection method to manage the effect of self-absorption, which becomes unavoidable at high concentration level, and to optimize measurement precision. Through such study, we try to deduce a quantifiable and generalizable line selection method for high performance LIBS measurements. More specifically, calibration curves were first established using 6 laboratory-prepared samples. The quadratic term of the curves was then studied as a function of the intensity of the used lines and their type (neutral or ion, resonant or non-resonant). The prediction performance of the lines was assessed with 2 validation samples with relatively low Ti content for one and higher Ti content for the second. The influences of the number of the ammonium groups and their arrangement manner on the photophysical properties of the quaternized zinc phthalocyanines were studied. The aggregation tendency, singlet oxygen generation ability and the fluorescence intensity properties of the quaternized zinc phthalocyanines were studied and compared. Results indicated that more quaternized ammonium groups and the straight chain arrangement manner for the quaternized zinc phthalocyanines can decrease aggregation degree and increase singlet oxygen generation and fluorescence intensity. To better understand the evolution of the South China Craton (SCC), we have determined the geochronological and geochemical compositions of newly recognized Grenville-aged metabasalts in the Shennongjia region of the northern Yangtze block. LA-ICP-MS U-Pb dating of zircons indicates that the metabasalts formed at 1063 ± 16 Ma. The rocks are calc-alkaline, are characterized by SiO2contents (50.50 to 55.62 wt.%), and have moderate-to-high MgO contents (7.25–9.60 wt.%). They display light rare earth element enrichment ((La/Yb)N = 7.0–9.8) with slightly negative Eu anomalies (Eu/Eu* = 0.82–0.90) and have pronounced depletion in high-field strength elements as well as positive Pb anomalies in the primitive mantle-normalized trace element pattern. They possess high initial Sr isotopic ratios of 0.7092–0.7107, large negativeεNd(t) values of −12.1 to −11.0, and a relatively narrow range of initial Pb isotope ratios (206Pb/204Pb = 16.503–17.019,207Pb/204Pb = 15.259–15.452, and208Pb/204Pb = 36.169–36.994). These isotopic characteristics are typical of basalts derived from an EM2 source region and suggest a subcontinental lithospheric mantle source that was metasomatized by subducted components (fluids and melts). Integrating our new data with documented igneous and metamorphic events during late Mesoproterozoic to early Neoproterozoic time in the region and western segments of the Yangtze block, we suggest that the Shennongjia area might be a microcontinent that was independent of the continental nucleus of the Yangtze block and was accreted to the Yangtze block at the end of Mesoproterozoic time. This study thus argues against the traditional view that the SCC was formed simply by Yangtze–Cathaysia collision and supports a hypothesis in which the Yangtze block was a collage of microcontinents accreted during the Grenvillian period accompanying the assembly of the Rodinia supercontinent. Carotenoids are mostly C 40 terpenoids, a class of hydrocarbons that participate in various biological processes in plants, such as photosynthesis, photomorphogenesis, photoprotection, and development. Carotenoids also serve as precursors for two plant hormones and a diverse set of apocarotenoids. They are colorants and critical components of the human diet as antioxidants and provitamin A. In this review, we summarize current knowledge of the genes and enzymes involved in carotenoid metabolism and describe recent progress in understanding the regulatory mechanisms underlying carotenoid accumulation. Under phosphate starvation, the root-associated activity of Arabidopsis acid phosphatase AtPAP10 is differentially regulated by local and systemic signalling at transcriptional, post-transcriptional, and post-translational levels.The induction and secretion of acid phosphatases (APases) is a universal response of plants to phosphate (Pi) starvation. AtPAP10 (Arabidopsis purple acid phosphatase 10) is a major Pi starvation-induced APase that is associated with the root surface in Arabidopsis. So far, the roles of local and systemic signalling in regulating root-associated AtPAP10 activity remain largely unknown. In this work, we show that a decrease of local, external Pi availability is sufficient to induce AtPAP10 transcription in roots in the presence of sucrose, a systemic signal from shoots, whereas the magnitude of the induction is affected by the Pi status of the whole plant. Once the AtPAP10 mRNAs are synthesized in roots, subsequent accumulation of AtPAP10 proteins in root cells and increase in AtPAP10 activity on the root surface are mainly controlled by local signalling. Previously, ethylene has been demonstrated to be a positive regulator of AtPAP10 activity. In this study, we provide evidence that under Pi deficiency ethylene mainly modulates enzymatic activity of AtPAP10 on the root surface, but not AtPAP10 transcription and protein accumulation, suggesting that it functions as a local signal. Furthermore, our work indicates that the effect of ethylene on the induction of root-associated AtPAP10 activity depends on sucrose, but that the effect of sucrose does not depend on ethylene. Background: Organ-specific metastatic context has not been incorporated into the clinical practice of guiding programmed death-(ligand) 1 [PD-(L)1] blockade, due to a lack of understanding of its predictive versus prognostic value. We aim at delineating and then incorporating both the predictive and prognostic effects of the metastatic-organ landscape to dissect PD-(L)1 blockade efficacy in non-small cell lung cancer (NSCLC).Methods: A total of 2062 NSCLC patients from a double-arm randomized trial (OAK), two immunotherapy trials (FIR, BIRCH), and a real-world cohort (NFyy) were included. The metastatic organs were stratified into two categories based on their treatment-dependent predictive significance versus treatment-independent prognosis. A metastasis-based scoring system (METscore) was developed and validated for guiding PD-(L)1 blockade in clinical trials and real-world practice.Results: Patients harboring various organ-specific metastases presented significantly different responses to immunotherapy, and those with brain and adrenal gland metastases survived longer than others [overall survival (OS), p = 0.0105; progression-free survival (PFS), p = 0.0167]. In contrast, survival outcomes were similar in chemotherapy-treated patients regardless of metastatic sites (OS, p = 0.3742; PFS, p = 0.8242). Intriguingly, the immunotherapeutic predictive significance of the metastatic-organ landscape was specifically presented in PD-L1-positive populations (PD-L1 > 1%). Among them, a paradoxical coexistence of a favorable predictive effect coupled with an unfavorable prognostic effect was observed in metastases to adrenal glands, brain, and liver (category I organs), whereas metastases to bone, pleura, pleural effusion, and mediastinum yielded consistent unfavorable predictive and prognostic effects (category II organs). METscore was capable of integrating both predictive and prognostic effects of the entire landscape and dissected OS outcome of NSCLC patients received PD-(L)1 blockade (p < 0.0001) but not chemotherapy (p = 0.0805) in the OAK training cohort. Meanwhile, general performance of METscore was first validated in FIR (p = 0.0350) and BIRCH (p < 0.0001), and then in the real-world NFyy cohort (p = 0.0181). Notably, METscore was also applicable to patients received PD-(L)1 blockade as first-line treatment both in the clinical trials (OS, p = 0.0087; PFS, p = 0.0290) and in the real-world practice (OS, p = 0.0182; PFS, p = 0.0045).Conclusions: Organ-specific metastatic landscape served as a potential predictor of immunotherapy, and METscore might enable noninvasive forecast of PD-(L)1 blockade efficacy using baseline radiologic assessments in advanced NSCLC. Orange (OR) is a DnaJ-like zinc finger protein with both nuclear and plastidial localizations. OR, and its orthologs, are highly conserved in flowering plants, sharing a characteristic C-terminal tandem 4× repeats of the CxxCxxxG signature. It was reported to trigger chromoplast biogenesis, promote carotenoid accumulation in plastids of non-pigmented tissues, and repress chlorophyll biosynthesis and chloroplast biogenesis in the nucleus of de-etiolating cotyledons cells. Its ectopic overexpression was found to enhance plant resistance to abiotic stresses. Here, we report that the expression of OR in Arabidopsis thaliana was upregulated by drought treatment, and seedlings of the OR-overexpressing (OE) lines showed improved growth performance and survival rate under drought stress. Compared with the wild-type (WT) and OR-silencing (or) lines, drought-stressed OE seedlings possessed lower contents of reactive oxygen species (such as H2O2 and O2−), higher activities of both superoxide dismutase and catalase, and a higher level of proline content. Our enzymatic assay revealed a relatively higher activity of Δ1-pyrroline-5-carboxylate synthase (P5CS), a rate-limiting enzyme for proline biosynthesis, in drought-stressed OE seedlings, compared with the WT and or lines. We further demonstrated that the P5CS activity could be enhanced by supplementing exogenous OR in our in vitro assays. Background: Dyslipidemia is a significant contributor to cardiovascular and cerebrovascular diseases. Research on the relationship between breakfast consumption frequency and dyslipidemia in the working population is lacking. Therefore, we aimed to investigate this relationship based on a retrospective cohort study of a large working population in China. Methods: This retrospective cohort study used data from the physical examinations and questionnaire survey of working participants at Nanfang Hospital from January 20, 2015 to October 16, 2020. Univariate and multivariate analyses were conducted to explore the relationship between breakfast consumption frequency and dyslipidemia in this working population (n = 7644). Results: The prevalence of dyslipidemia among the participants was 26.4%. The univariate logistic regression test showed that the breakfast consumption frequency was inversely correlated with dyslipidemia. After adjusting for multiple factors, such as sex, age, body mass index, hypertension, hyperuricaemia, diabetes, smoking status, alcohol consumption, education level, marital status, long-term exposure to kitchen oil fumes, attending business dinners, and sleep time, it was found that breakfast consumption remained inversely associated with dyslipidaemia. The odds ratio for daily breakfast consumption was 0.466 (95% confidence interval 0.283–0.770, P = 0.003). After adjusting for confounding factors, we found that the higher the frequency of breakfast consumption, the lower the odds ratios for hypertriglyceridaemia. Conclusions: This study demonstrated that breakfast consumption frequency was inversely correlated with dyslipidemia. The higher the frequency of breakfast, the lower the risk of hypertriglyceridaemia. Background. The Chinese patent drug Yinzhihuang granule (YZHG) is used to treat hepatitis B. This research is aimed at exploring the multicomponent synergistic mechanism of YZHG in the treatment of inflammation-cancer transformation of hepar and at providing new evidence and insights for its clinical application. Methods. To retrieve the components and targets of Yinzhihuang granules. The differentially expressed genes (DEGs) of hepar inflammation-cancer transformation were obtained from TTD, PharmGKB, and GEO databases. Construct the compound-prediction target network and the key module network using Cytoscape 3.7.1. Results. The results show that hepatitis B and hepatitis C shared a common target, MMP2. CDK1 and TOP2A may play an important role in the treatment with YZHG in hepatitis B inflammatory cancer transformation. KEGG pathway enrichment showed that key genes of modules 1, 2, and 4 were mainly enriched in the progesterone-mediated oocyte maturation signaling pathway and oocyte meiosis signaling pathway. The multicomponent, multitarget, and multichannel pharmacological benefits of YZHG in the therapy of inflammation-cancer transition of hepar are directly demonstrated by network pharmacology, providing a scientific basis for its mechanism. Attentional bias toward health-threat may theoretically contribute to the development and maintenance of health anxiety, but the empirical findings have been controversial. This study aimed to synthesize and explore the heterogeneity in a health-threat related attentional bias of health-anxious individuals, and to determine the theoretical model that better represents the pattern of attentional bias in health anxiety. Four databases (Web of Science, PubMed, PsycINFO, and Scopus) were searched for relevant studies, with 17 articles (N = 1546) included for a qualitative review and 16 articles (18 studies) for a three-level meta-analysis (N = 1490). The meta-analytic results indicated that the health anxiety group, compared to the control group, showed significantly greater attentional bias toward health-threat (g = 0.256). Further analyses revealed that attentional bias type, paradigm, and stimuli type were significant moderators. Additionally, compared to the controls, health-anxious individuals displayed significantly greater attention maintenance (g = 0.327) but nonsignificant attention vigilance to health-threat (g = −0.116). Our results provide evidence for the attention maintenance model in health-anxious individuals. Purpose: To evaluate whether Alberta Stroke Program Early CT Score (ASPECTS) could provide incremental value to collateral score, and their integration could be an effective surrogate of CTP in predicting target mismatch. Material and methods: One hundred and fifty-nine stroke patients (onset time 6–16 h or with unknown onset time) with MCA and/or ICA occlusion underwent non-contrast computed tomography (NCCT) and CT perfusion (CTP) scan for initial assessment. Simulated single-phase CT angiography (sCTA, peak arterial phase) and multiphase CTA (mCTA) were reconstructed from CTP. ASPECTS was assessed on NCCT and sCTA. Collateral score was evaluated on mCTA. Target mismatch was defined as infarct core volume < 70 mL, the mismatch ratio ≥ 1.8, and the absolute mismatch volume ≥ 15 mL. Pearson correlation analysis, Mann–Whitney U test, chi-square test, and receiver operating characteristic curve analyses were performed. Results: Median CTA source image (CTA-SI) ASPECTS was significantly lower than NCCT ASPECTS (p = 0.001). NCCT ASPECTS, CTA-SI ASPECTS, and mCTA collateral score correlated significantly with infarct core volume and mismatch ratio (all p < 0.05). Mismatch group showed significantly higher NCCT ASPECTS, CTA-SI ASPECTS, and mCTA collateral score than non-mismatch group (all p < 0.001). NCCT ASPECTS and CTA-SI ASPECTS showed comparable predicting performance with mCTA collateral score (p > 0.05). Adding CTA-SI ASPECTS to mCTA collateral score improved the performance of mCTA in predicting target mismatch (area under curve, 0.905 vs. 0.804, p = 0.003). Conclusion: ASPECTS can provide incremental information to collateral score in predicting target mismatch. Nuclear-encoded Atp23 was previously shown to have dual functions, including processing the yeast Atp6 precursor and assisting the assembly of yeast mitochondrial ATP synthase. However, it remains unknown whether there are genes functionally complementary to ATP23 to rescue atp23 null mutant. In the present paper, we screen and characterize three revertants of atp23 null mutant and reveal a T1121G point mutation in the mitochondrial gene COX1 coding sequence, which leads to Val374Gly mutation in Cox1, the suppressor in the revertants. This was verified further by the partial restoration of mitochondrial ATP synthase assembly in atp23 null mutant transformed with exogenous hybrid COX1 T1121G mutant plasmid. The predicted tertiary structure of the Cox1 p.Val374Gly mutation showed no obvious difference from wild-type Cox1. By further chase labeling with isotope [35S]-methionine, we found that the stability of Atp6 of ATP synthase increased in the revertants compared with the atp23 null mutant. Taking all the data together, we revealed that the T1121G point mutation of mitochondrial gene COX1 could partially restore the unassembly of mitochondrial ATP synthase in atp23 null mutant by increasing the stability of Atp6. Therefore, this study uncovers a gene that is partially functionally complementary to ATP23 to rescue ATP23 deficiency, broadening our understanding of the relationship between yeast the cytochrome c oxidase complex and mitochondrial ATP synthase complex. Food safety incidents and scandals, frequently breaking out around the world, are significantly reducing the confidence of consumers in food safety and quality. It is even more difficult to achieve the full transparency, visibility and traceability of dynamic cross-border and global food supply chains, which is critical to ensure food safety and quality, due to: the personalization and globalization of consumer demands; the diversities of food products, raw materials and ingredients; the loose relationships between food business partners; the technology and resource barriers in aligning business processes, coordinating and sharing information; and the changing government supervision mechanisms and standards. Social networking technologies and social software are promising enablers to facilitate end consumers, food companies and government agencies to participate in open discussion, comments and feedback on the quality and safety of cross-border foods. They are able to connect, interact, communicate and collaborate with each other in loose, open, effective and flexible ways for enhancing the transparency and visibility of cross-border and global food supply chains through collective wisdom and intelligence. In this paper a framework is proposed for developing social networks enabling food traceability systems, which not only leverages enterprise applications, public service platforms for food quality, safety and traceability operated by government or industrial associations, but utilizes such emerging technologies as Internet-of-Things, social analytic and mobile technologies to ease interactive communication and collaboration between various stakeholders of food chains. Guidelines, architectures, use scenarios, and technology alternatives for implementing the traceability systems are also discussed in this work. Abstract: The nature of doping anions in polymerization solution has great influences on structure and properties of electro-polymerized polypyrrole (PPy) films and, thereby, on the electrocatalytic activity for I−/I3 − redox reaction and accordingly the performance of dye-sensitized solar cells (DSSCs) based on PPy counter electrodes (CEs). The ions of chloride (Cl−), sulfate (SO4 2−), p-toluene sulfonate (TsO−), and dodecyl benzene sulfonate (DBS−) have been used as counter anions to prepare PPy films as CEs in the DSSCs. Compared with inorganic anions (Cl− and SO4 2−), the organic ones (TsO− and DBS−) afford more delocalized polarons. Moreover, PPy–DBS− films have the most fibrous or porous structure. Thus, PPy–DSB− has plenty of electrocatalytic active sites and high electric conductivity and, thereby, high electrocatalytic activity for I−/I3 − redox reaction. Due to the synergistic effects of fibrous/porous structure, high conductivity and low interfacial charge transfer impedance, PPy–DBS− CE based DSSCs show the best photovoltaic conversion efficiency up to 5.40%, reaching 88% of the DSSCs based on Pt CE. Non-alcoholic fatty liver disease (NAFLD) affects approximately 25 % of the global population, presenting a significant threat to human health and imposing a substantial economic burden on society. Cytokeratin 18 (CK18) has emerged as a promising non-invasive biomarker for early detection of NAFLD. However, current diagnostic methods face challenges in accurately identifying NAFLD in its early stages. To address this issue, our study aimed to generate new CK18 antibodies and utilize a highly sensitive single-molecule immunodetection system customized for NAFLD. We embarked on a process involving the production of novel CK18 monoclonal antibodies, which were subsequently engineered into recombinant forms. In comparison to commercial antibodies, our antibodies exhibited superior affinity. Grounded in the SMCxPRO platform, our assay underwent rigorous assessment, adhering to well-defined biomarker assay criteria. The assay demonstrated specificity to CK18 and achieved a lower limit of quantification of 1 pg/ml. Importantly, elevated serum CK18 levels were observed among NAFLD patients compared to healthy individuals. The single-molecule detection system, utilizing our novel antibodies, holds significant promise for the early-stage diagnosis, dynamic disease monitoring, and prognostic evaluation of NAFLD patients. In the "digital age", children have widespread access to electronic devices, potentially impacting inhibitory control (IC) development. We examined this association at both the behavioral and neural levels with two experiments. In Experiment 1, we assessed 188 participants (91 girls), averaging 37.74 months old, using maternal self-report questionnaire for screen time and Day-Night Task for IC measurement. In experiment 2, we evaluated 42 participants (24 girls), averaging 41.24 months old, using maternal self-report questionnaires for screen time, a modified version Flanker task for IC, and functional near-infrared spectroscopy (fNIRS) to measure neural activity during IC task. Results from Experiment 1 indicated a negative correlation between young children's screen media use time and IC, with high users performing worse IC performance than low users. Experiment 2 confirmed these findings and revealed marginally reduced activation in the left PFC among high users compared to low users, with a moderate effect size and weak supporting evidence. These preliminary results underscore a detrimental association between screen media use and IC, emphasizing the significance of restricting screen exposure in young children to prevent overexposure. Further studies with larger sample sizes are needed to validate these results and draw causal conclusions. • Child screen media use time was negatively associated with inhibitory control. • High screen media user group (≥1 h/day) performance worse during inhibitory control task than low user group (<1 h/day). • High screen media user group had marginally lower activation in the PFC than the low user group. Antibodies are principal immune components elicited by vaccines to induce protection from microbial pathogens. In the Thai RV144 HIV-1 vaccine trial, vaccine efficacy was 31% and the sole primary correlate of reduced risk was shown to be vigorous antibody response targeting the V1V2 region of HIV-1 envelope. Antibodies against V3 also were inversely correlated with infection risk in subsets of vaccinees. Antibodies recognizing these regions, however, do not exhibit potent neutralizing activity. Therefore, we examined the antiviral potential of poorly neutralizing monoclonal antibodies (mAbs) against immunodominant V1V2 and V3 sites by passive administration of human mAbs to humanized mice engrafted with CD34+ hematopoietic stem cells, followed by mucosal challenge with an HIV-1 infectious molecular clone expressing the envelope of a tier 2 resistant HIV-1 strain. Treatment with anti-V1V2 mAb 2158 or anti-V3 mAb 2219 did not prevent infection, but V3 mAb 2219 displayed a superior potency compared to V1V2 mAb 2158 in reducing virus burden. While these mAbs had no or weak neutralizing activity and elicited undetectable levels of antibody-dependent cellular cytotoxicity (ADCC), V3 mAb 2219 displayed a greater capacity to bind virus- and cell-associated HIV-1 envelope and to mediate antibody-dependent cellular phagocytosis (ADCP) and C1q complement binding as compared to V1V2 mAb 2158. Mutations in the Fc region of 2219 diminished these effector activities in vitro and lessened virus control in humanized mice. These results demonstrate the importance of Fc functions other than ADCC for antibodies without potent neutralizing activity. Author summary: In the past decade, HIV-1 has infected an estimated 1.5 to 2 million people every year, but vaccines needed to control this pandemic are unavailable. Among vaccines tested in the human efficacy trials, the RV144 vaccine regimen showed a modest efficacy and revealed non-neutralizing antibodies against the virus envelope glycoproteins as a correlate of reduced virus acquisition. To design more efficacious HIV-1 vaccines, a better understanding about antiviral mechanisms of these antibodies is needed. Here non-neutralizing monoclonal antibodies against two immunogenic sites on the virus envelope were evaluated for passive administration to humanized mice that were subsequently challenged with HIV-1. The antibodies did not block mucosal HIV-1 infection but reduced virus burden. The level of virus reduction correlated with the antibody binding potency and the effector functions mediated through their Fc fragments, which included antibody-dependent phagocytosis and complement activation, but not the commonly studied antibody-dependent cellular cytotoxicity. The importance of the Fc functions was further demonstrated by reduced virus control when mutations were introduced to decrease Fc activities. Background: Compound kushen injection (CKI), a Chinese patent drug, is widely used in the treatment of various cancers, especially neoplasms of the digestive system. However, the underlying mechanism of CKI in pancreatic cancer (PC) treatment has not been totally elucidated. Methods: Here, to overcome the limitation of conventional network pharmacology methods with a weak combination with clinical information, this study proposes a network pharmacology approach of integrated bioinformatics that applies a weighted gene co-expression network analysis (WGCNA) to conventional network pharmacology, and then integrates molecular docking technology and biological experiments to verify the results of this network pharmacology analysis. Results: The WGCNA analysis revealed 2 gene modules closely associated with classification, staging and survival status of PC. Further CytoHubba analysis revealed 10 hub genes (NCAPG, BUB1, CDK1, TPX2, DLGAP5, INAVA, MST1R, TMPRSS4, TMEM92 and SFN) associated with the development of PC, and survival analysis found 5 genes (TSPOAP1, ADGRG6, GPR87, FAM111B and MMP28) associated with the prognosis and survival of PC. By integrating these results into the conventional network pharmacology study of CKI treating PC, we found that the mechanism of CKI for PC treatment was related to cell cycle, JAK-STAT, ErbB, PI3K-Akt and mTOR signalling pathways. Finally, we found that CDK1, JAK1, EGFR, MAPK1 and MAPK3 served as core genes regulated by CKI in PC treatment, and were further verified by molecular docking, cell proliferation assay, RT-qPCR and western blot analysis. Conclusions: Overall, this study suggests that the optimized network pharmacology approach is suitable to explore the molecular mechanism of CKI in the treatment of PC, which provides a reference for further investigating biomarkers for diagnosis and prognosis of PC and even the clinical rational application of CKI. Abstract: This study uses an integrated model utilizing a small-world network and choice-based conjoint adoption model to examine the dynamics of consumer choice and diffusion in the hybrid electric vehicles market. It specifically compares the effectiveness of hybrid diffusion through the traditional word of mouth and via social media. The results show that without the advantage of increased gasoline prices, the growth of the hybrid vehicles market is insignificant, and that the Internet has a significant influence on the word of mouth effect in the purchasing process. Hybrid electric vehicles market shares decrease dramatically as a result of negative word of mouth communication via social media. The use of a higher fuel taxes is more effective than providing a subsidy for disposing of old vehicles and purchasing a hybrid. [Copyright &y& Elsevier]

Background: Omeprazole, a proton pump inhibitor (PPI), is widely used for the treatment of dyspepsia, peptic ulcer, gastroesophageal reflux disease, and functional dyspepsia. Polypharmacy is common in patients receiving omeprazole. Drug toxicity and treatment failure resulting from inappropriate combination therapy with omeprazole have been reported sporadically. Systematic review has not been available to address the pharmacokinetic drug-drug interaction (DDI) profile of omeprazole with adverse consequences, the factors determining the degree of DDI between omeprazole and comedication, and the corresponding clinical risk management. Methods: Literature was identified by performing a PubMed search covering the period from January 1988 to March 2013. The full text of each article was critically reviewed, and data interpretation was performed. Results: Omeprazole has actual adverse influences on the pharmacokinetics of medications such as diazepam, carbamazepine, clozapine, indinavir, nelfinavir, atazanavir, rilpivirine, methotrexate, tacrolimus, mycophenolate mofetil, clopidogrel, digoxin, itraconazole, posaconazole, and oral iron supplementation. Meanwhile, low efficacy of omeprazole treatment would be anticipated, as omeprazole elimination could be significantly induced by comedicated efavirenz and herb medicines such as St John's wort, Ginkgo biloba, and yin zhi huang. The mechanism for DDI involves induction or inhibition of cytochrome P450, inhibition of P-glycoprotein or breast cancer resistance protein-mediated drug transport, and inhibition of oral absorption by gastric acid suppression. Sometimes, DDIs of omeprazole do not exhibit a PPI class effect. Other suitable PPIs or histamine 2 antagonists may be therapeutic alternatives that can be used to avoid adverse consequences. The degree of DDIs associated with omeprazole and clinical outcomes depend on factors such as genotype status of CYP2C19 and CYP1A2, ethnicity, dose and treatment course of precipitant omeprazole, pharmaceutical formulation of object drug (eg, mycophenolate mofetil versus enteric-coated mycophenolate sodium), other concomitant medication (eg, omeprazole-indinavir versus omeprazole-indinavir-ritonavir), and administration schedule (eg, intensified dosing of mycophenolate mofetil versus standard dosing). Conclusion: Despite the fact that omeprazole is one of the most widely prescribed drugs internationally, clinical professionals should enhance clinical risk management on adverse DDIs associated with omeprazole and ensure safe combination use of omeprazole by rationally prescribing alternatives, checking the appropriateness of physician orders before dispensing, and performing therapeutic drug monitoring.
