Abstract

Many diseases are driven by proteins that are aberrantly ubiquitinated and degraded. These diseases would be therapeutically benefited by targeted protein stabilization (TPS). Here we present deubiquitinase-targeting chimeras (DUBTACs), heterobifunctional small molecules consisting of a deubiquitinase recruiter linked to a protein-targeting ligand, to stabilize the levels of specific proteins degraded in a ubiquitin-dependent manner. Using chemoproteomic approaches, we discovered the covalent ligand EN523 that targets a non-catalytic allosteric cysteine C23 in the K48-ubiquitin-specific deubiquitinase OTUB1. We showed that a DUBTAC consisting of our EN523 OTUB1 recruiter linked to lumacaftor, a drug used to treat cystic fibrosis that binds ΔF508-cystic fibrosis transmembrane conductance regulator (CFTR), robustly stabilized ΔF508-CFTR protein levels, leading to improved chloride channel conductance in human cystic fibrosis bronchial epithelial cells. We also demonstrated stabilization of the tumor suppressor kinase WEE1 in hepatoma cells. Our study showcases covalent chemoproteomic approaches to develop new induced proximity-based therapeutic modalities and introduces the DUBTAC platform for TPS.

Abstract

Axionlike particles (ALPs) are predicted in many extensions of the standard model, and their masses can naturally be well below the electroweak scale. In the presence of couplings to electroweak bosons, these particles could be emitted in flavor-changing B meson decays. We report herein a search for an ALP, a, in the reaction B^{±}→K^{±}a, a→γγ using data collected by the BABAR experiment at SLAC. No significant signal is observed, and 90% confidence level upper limits on the ALP coupling to electroweak bosons are derived as a function of ALP mass, improving current constraints by several orders of magnitude in the range 0.175  GeV

Abstract

Photoaffinity labeling (PAL) is a powerful tool for the identification of non-covalent small molecule-protein interactions that are critical to drug discovery and medicinal chemistry, but this approach is limited to only a small subset of robust photocrosslinkers. The identification of new photoreactive motifs capable of covalent target capture is therefore highly desirable. Herein, we report the design, synthesis, and evaluation of a new class of PAL warheads based on the UV-triggered 1,2-photo-Brook rearrangement of acyl silanes, which hitherto have not been explored for PAL workflows. Irradiation of a series of probes in cell lysate revealed an iPr-substituted acyl silane with superior photolabeling and minimal thermal background labeling compared to other substituted acyl silanes. Further, small molecule (+)-JQ1- and rapamycin-derived iPr acyl silanes were shown to selectively label recombinant BRD4-BD1 and FKBP12, respectively, with minimal background. Together, these data highlight the untapped potential of acyl silanes as a novel, tunable scaffold for photoaffinity labeling.

Abstract

We report on the first search for electron-muon lepton flavor violation (LFV) in the decay of a b quark and b antiquark bound state. We look for the LFV decay ϒ(3S)→e^{±}μ^{∓} in a sample of 118 million ϒ(3S) mesons from 27  fb^{-1} of data collected with the BABAR detector at the SLAC PEP-II e^{+}e^{-} collider operating with a 10.36 GeV center-of-mass energy. No evidence for a signal is found, and we set a limit on the branching fraction B[ϒ(3S)→e^{±}μ^{∓}]80  TeV on the energy scale Λ_{NP} divided by the coupling-squared g_{NP}^{2} of relevant new physics (NP).

Abstract

Observation research can shed light on the degree to which students have access to research-based instruction and intervention. In this systematic review of reading observation research for students with and at risk for emotional and behavioral disorders, we sought to identify trends in the settings and student populations investigated and research methods used, as well as to determine the degree to which this student population has access to research-based reading instruction. Eleven studies meeting selection criteria were identified and coded to extract information that was salient to research questions. Although the extant observation research is limited, findings suggest that concerns raised by (Vaughn et al., Journal of Special Education 36:2–13, 2002) approximately 18 years ago remain. Study limitations, implications for school practice, and areas for future research are discussed.

Abstract

Modern radio interferometers such as the LOw Frequency ARray (LOFAR) are capable of producing data at hundreds of gigabits to terabits per second. This high data rate makes the analysis of radio data cumbersome and computationally expensive. While high performance computing facilities exist for large national and international facilities, that may not be the case for instruments operated by a single institution or a small consortium. Data rates for next generation radio telescopes are set to eclipse those currently in operation, hence local processing of data will become all the more important. Here, we introduce the REAL-time Transient Acquisition backend (REALTA), a computing backend at the Irish LOFAR station (I-LOFAR) which facilitates the recording of data in near real-time and post-processing. 379. Winning the NIST Contest: A scalable and general approach to differentially private synthetic data

Abstract

We propose a general approach for differentially private synthetic data generation, that consists of three steps: (1) select a collection of low-dimensional marginals, (2) measure those marginals with a noise addition mechanism, and (3) generate synthetic data that preserves the measured marginals well. Central to this approach is Private-PGM, a post-processing method that is used to estimate a high-dimensional data distribution from noisy measurements of its marginals. We present two mechanisms, NIST-MST and MST, that are instances of this general approach. NIST-MST was the winning mechanism in the 2018 NIST differential privacy synthetic data competition, and MST is a new mechanism that can work in more general settings, while still performing comparably to NIST-MST. We believe our general approach should be of broad interest, and can be adopted in future mechanisms for synthetic data generation., Comment: 22 pages

Abstract

We report a measurement of the $D^0$ and $D^+$ lifetimes using $D^0\to K^-\pi^+$ and $D^+\to K^-\pi^+\pi^+$ decays reconstructed in $e^+e^-\to c\bar{c}$ data recorded by the Belle II experiment at the SuperKEKB asymmetric-energy $e^+e^-$ collider. The data, collected at center-of-mass energies at or near the $\Upsilon(4S)$ resonance, correspond to an integrated luminosity of $72\,{\rm fb}^{-1}$. The results, $\tau(D^0) = 410.5\pm1.1\,{\rm(stat)}\pm0.8\,{\rm(syst.)}\,{\rm fs}$ and $\tau(D^+) = 1030.4\pm4.7\,{\rm(stat)}\pm3.1\,{\rm(syst.)}\,{\rm fs}$, are the most precise to date and are consistent with previous determinations., Comment: 9 pages, 2 figures

We study "distance spheres": the set of points lying at constant distance from a fixed arbitrary subset $K$ of $[0,1]^d$. We show that, away from the regions where $K$ is "too dense" and a set of small volume, we can decompose $[0,1]^d$ into a finite number of sets on which the distance spheres can be "straightened" into subsets of parallel $(d-1)$-dimensional planes by a bi-Lipschitz map. Abstract

The ALPHA collaboration has successfully demonstrated the production and the confinement of cold antihydrogen, $\overline{\mathrm{H}}$. An analysis of trapping data allowed a stringent limit to be placed on the electric charge of the simplest antiatom. Charge neutrality of matter is known to a very high precision, hence a neutrality limit of $\overline{\mathrm{H}}$ provides a test of CPT invariance. The experimental technique is based on the measurement of the deflection of putatively charged $\overline{\mathrm{H}}$ in an electric field. The tendency for trapped $\overline{\mathrm{H}}$ atoms to be displaced by electrostatic fields is measured and compared to the results of a detailed simulation of $\overline{\mathrm{H}}$ dynamics in the trap. An extensive survey of the systematic errors is performed, with particular attention to those due to the silicon vertex detector, which is the device used to determine the $\overline{\mathrm{H}}$ annihilation position. The limit obtained on the charge of the $\overline{\mathrm{H}}$ atom is \mbox{$ Q = (-1.3\pm1.8\pm0.4)\times10^{-8}$}, representing the first precision measurement with $\overline{\mathrm{H}}$., Comment: 5 pages, 3 figures

Abstract

Onshore wind development has historically focused on cost-efficiency, which may lead to inequitable turbine distributions and public resistance due to landscape impacts. Using a multi-criteria planning approach, we show how onshore wind capacity targets can be achieved by 2050 in a cost-efficient, equitable and publicly acceptable way. For the case study of Germany, we build on the existing turbine stock and use open data on technically feasible turbine locations and scenicness of landscapes to plan the optimal expansion. The analysis shows that while the trade-off between cost-efficiency and public acceptance is rather weak with about 15% higher costs or scenicness, an equitable distribution has a large impact on these criteria. Although the onshore wind capacity per inhabitant could be distributed about 220% more equitably through the expansion, equity would severely limit planning flexibility by 2050. Our analysis assists stakeholders in resolving the onshore wind expansion trilemma.

Abstract

Decision support systems like computer-aided energy system analysis (ESA) are considered one of the main pillars for developing sustainable and reliable energy transformation strategies. Although today's diverse tools can already support decision-makers in a variety of research questions, further developments are still necessary. Intending to identify opportunities and challenges in the field, we classify modelling capabilities (32), methodologies (15) implementation issues (15) and management issues (7) from an extensive literature review. Based on a quantitative expert survey of energy system modellers (N=61) mainly working with simulation and optimisation models, the status of development and the complexity of realisation of those modelling topics are assessed. While the rated items are considered to be more complex than actually represented, no significant outliers are determinable, showing that there is no consensus about particular aspects of ESA that are lacking development. Nevertheless, a classification of the items in terms of a specially defined modelling strategy matrix identifies capabilities like land-use planning patterns, equity and distributional effects and endogenous technological learning as "low hanging fruits" for enhancement, as well as a large number of complex topics that are already well implemented. The remaining "tough nuts" regarding modelling capabilities include non-energy sector and social behaviour interaction effects. In general, the optimisation and simulation models differ in their respective strengths, justifying the existence of both. While methods were generally rated as quite well developed, combinatorial optimisation approaches, as well as machine learning, are identified as important research methods to be developed further for ESA.

Abstract

In this work we describe the High-Dimensional Matrix Mechanism (HDMM), a differentially private algorithm for answering a workload of predicate counting queries. HDMM represents query workloads using a compact implicit matrix representation and exploits this representation to efficiently optimize over (a subset of) the space of differentially private algorithms for one that is unbiased and answers the input query workload with low expected error. HDMM can be deployed for both $\epsilon$-differential privacy (with Laplace noise) and $(\epsilon, \delta)$-differential privacy (with Gaussian noise), although the core techniques are slightly different for each. We demonstrate empirically that HDMM can efficiently answer queries with lower expected error than state-of-the-art techniques, and in some cases, it nearly matches existing lower bounds for the particular class of mechanisms we consider., Comment: arXiv admin note: text overlap with arXiv:1808.03537

Abstract

End-to-end (E2E) spoken language understanding (SLU) systems predict utterance semantics directly from speech using a single model. Previous work in this area has focused on targeted tasks in fixed domains, where the output semantic structure is assumed a priori and the input speech is of limited complexity. In this work we present our approach to developing an E2E model for generalized SLU in commercial voice assistants (VAs). We propose a fully differentiable, transformer-based, hierarchical system that can be pretrained at both the ASR and NLU levels. This is then fine-tuned on both transcription and semantic classification losses to handle a diverse set of intent and argument combinations. This leads to an SLU system that achieves significant improvements over baselines on a complex internal generalized VA dataset with a 43% improvement in accuracy, while still meeting the 99% accuracy benchmark on the popular Fluent Speech Commands dataset. We further evaluate our model on a hard test set, exclusively containing slot arguments unseen in training, and demonstrate a nearly 20% improvement, showing the efficacy of our approach in truly demanding VA scenarios., Comment: Accepted to Interspeech 2021; 5 pages, 2 tables, 1 figure

Abstract

Little is known about the development of self-regulation processes during the preschool period in autism spectrum disorder (ASD). How parental characteristics such as the broader autism phenotype (BAP) relate to children's self-regulation is not well understood. Preschool-aged children with (n = 24) and without ASD (n = 21) completed an inhibitory control task and mothers reported on child emotion regulation and their own BAP traits. Children with ASD had lower emotion regulation, and emotion regulation was a protective factor in the association between ASD and internalizing behavioral concerns. Lability/negativity was highly overlapping with externalizing. Maternal BAP characteristics were differentially associated with all self-regulation outcomes across groups. Parental factors should be considered in emotion regulation interventions for young children with ASD.

388. Sex Differences in Training-Induced Activity of the Ubiquitin Proteasome System in the Dorsal Hippocampus and Medial Prefrontal Cortex of Male and Female Mice

Abstract

The ubiquitin proteasome system (UPS) is a primary mechanism through which proteins are degraded in cells. UPS activity in the dorsal hippocampus (DH) is necessary for multiple types of memory, including object memory, in male rodents. However, sex differences in DH UPS activation after fear conditioning suggest that other forms of learning may also differentially regulate DH UPS activity in males and females. Here, we examined markers of UPS activity in the synaptic and cytoplasmic fractions of DH and medial prefrontal cortex (mPFC) tissue collected 1 h following object training. In males, training increased phosphorylation of proteasomal subunit Rpt6, 20S proteasome activity, and the amount of PSD-95 in the DH synaptic fraction, as well as proteasome activity in the mPFC synaptic fraction. In females, training did not affect measures of UPS or synaptic activity in the DH synaptic fraction or in either mPFC fraction but increased Rpt6 phosphorylation in the DH cytoplasmic fraction. Overall, training-induced UPS activity was greater in males than in females, greater in the DH than in the mPFC, and greater in synaptic fractions than in cytosol. These data suggest that object training drives sex-specific alterations in UPS activity across brain regions and subcellular compartments important for memory.

Quality curriculum design and assessment processes are essential for promoting student competency development and demonstrating institutional effectiveness. As faculty have a key role in these efforts, it is important that they are equipped with adequate tools and support. A backward design model with embedded assessments aligned to industry standards is an approach that provides a clear process for faculty to design courses that engage students through formative and summative assessments with personalized feedback. Further, real-time analyses are possible via technology-enabled learning platforms which yields data to support the faculty-student dyad in the learning process. It also positions assessment specialists, administrators, and faculty to drive and systematically assess the impact of data-informed quality improvements to curricula. Administrative and institutional support is essential to establishing and sustaining a mature culture of assessment in which the relationships among teaching, learning, and assessment are recognized.
