Abstract

• Rapid solidification of Ti-Ni-Al alloy was investigated by ESL technology. • ESL plus quenching process led to formation of multiple nanostructures. • Solute cluster, distortion phase and other nanoprecipitate were achieved. • Ti-Ni-Al alloy with metastable nanostructures was conspicuous hardened. We prepared a Ti-Ni-Al alloy comprised of metastable equiaxed (βTi) dendrites and interdendritic Ti 2 Ni phase by means of electrostatic levitation plus quenching. It exhibited an impressive improvement in hardness compared with the sample through DTA processing. By employing the double Cs corrector transmission electron microscope characterization, the coherent solute cluster and semi-coherent nano-distortion (βTi) were found to form within the metastable (βTi) dendrites, and (αTi) nanoprecipitates generated inside interdendritic Ti 2 Ni phase matrix. These dispersive metastable nanostructures and the novel (αTi) + Ti 2 Ni biphase boundary contributed cooperatively to pinning dislocation. The harmonic cooperation of precipitation strengthening, solution strengthening and fine-grained strengthening were realized to enhance the alloy. [ABSTRACT FROM AUTHOR]

374. Mic60/Mitofilin determines MICOS assembly essential for mitochondrial dynamics and mtDNA nucleoid organization

Abstract

The MICOS complex (mitochondrial contact site and cristae organizing system) is essential for mitochondrial inner membrane organization and mitochondrial membrane contacts, however, the molecular regulation of MICOS assembly and the physiological functions of MICOS in mammals remain obscure. Here, we report that Mic60/Mitofilin has a critical role in the MICOS assembly, which determines the mitochondrial morphology and mitochondrial DNA (mtDNA) organization. The downregulation of Mic60/Mitofilin or Mic19/CHCHD3 results in instability of other MICOS components, disassembly of MICOS complex and disorganized mitochondrial cristae. We show that there exists direct interaction between Mic60/Mitofilin and Mic19/CHCHD3, which is crucial for their stabilization in mammals. Importantly, we identified that the mitochondrial i-AAA protease Yme1L regulates Mic60/Mitofilin homeostasis. Impaired MICOS assembly causes the formation of 'giant mitochondria' because of dysregulated mitochondrial fusion and fission. Also, mtDNA nucleoids are disorganized and clustered in these giant mitochondria in which mtDNA transcription is attenuated because of remarkable downregulation of some key mtDNA nucleoid-associated proteins. Together, these findings demonstrate that Mic60/Mitofilin homeostasis regulated by Yme1L is central to the MICOS assembly, which is required for maintenance of mitochondrial morphology and organization of mtDNA nucleoids.

Abstract

This paper presents an analytical simulation of the dynamics of a gear transmission system compared to experimental results from a gear noise test rig at the NASA Lewis Research Center. The analytical procedure developed couples the dynamic behaviors of the rotor-bearing-gear system with the response of the gearbox structure. The modal synthesis method is used in solving the overall dynamics of the system. Locally each rotor-gear stage is modeled as an individual rotor-bearing system using the matrix transfer technique. The dynamics of each individual rotor are coupled with other rotor stages through the nonlinear gear mesh forces and with the gearbox structure through bearing support systems. The modal characteristics of the gearbox structure are evaluated using the finite element procedure. A variable time stepping integration scheme is used to calculate the overall time transient behavior of the system in modal coordinates. The global dynamic behavior of the system is expressed in a generalized coordinate system. Transient and steady state vibrations of the gearbox system are presented int he time and frequency domains. The vibration characteristics of a simple single mesh gear noise test rig is modeled. The numerical simulations are compared to experimental data measured under typical operation conditions. The comparison of system natural frequencies, peak vibration amplitudes, and gear mesh frequencies are generally in good agreement.

Abstract

Blockade of extracellular high mobility group box 1 (HMGB1) can significantly prolong murine cardiac allograft survival. Here, we determined the role of HMGB1 in xenotransplantation. Sprague‐Dawley rat hearts were transplanted heterotopically into BALB/c mice. Xenografts without any treatment developed predominant acute vascular rejection within 6 days. Both passively released HMGB1 from xenografts and actively secreted HMGB1 from infiltrated immune cells were significantly increased after xenotransplantation. HMGB1‐neutralizing antibody treatment significantly prolonged xenograft survival and attenuated pathologic damage, immune cell infiltration, and HMGB1 expression and release in the xenografts. Compared to control IgG treatment evaluated at study endpoint, treatment with HMGB1‐neutralizing antibody markedly suppressed xenoreactive B cell responses, as evidenced by the significant inhibition of anti‐rat antibody production and deposition in xenografts at Day 6 posttransplant. Furthermore, treatment with anti‐HMGB1 antibody suppressed B cell activation and reduced IFN‐γ and IL‐17A production after xenotransplantation. These results demonstrate for the first time that HMGB1 plays an important role in mediating acute xenograft rejection. Thus, we have shown that neutralization of extracellular HMGB1 can significantly inhibit xenoreactive B cell responses and delay xenograft rejection in a rat‐to‐mouse model of xenotransplantation, uncovering new insights in the role of HMGB1 in transplantation. Using a rat‐to‐mouse cardiac xenotransplantation model, the authors find that blockade of extracellular high‐mobility group box 1 significantly inhibits xenoreactive B cell responses and delays acute vascular xenogeneic rejection.

Abstract

Abstract: Pseudobinary eutectic generated by pseudobinary eutectic transition or peri-eutectic transition is a crucial structure in ternary alloy systems. Its formation mechanism strongly influences mechanical properties of these metallic materials. However, it was customarily neglected. In this paper, two pseudobinary eutectics, i.e. (Ag+ ζ) and (Ag+ ε 2), were investigated during the rapid solidification of Cu–Ag–Ge ternary alloy in a 3m-drop tube. The sharp temperature variations and dramatic kinetic activities of the falling alloy droplets before solidification cause special microstructural characteristics. (Ag) dendrite is the heterogeneous nucleus for anomalous (Ag+ ζ) pseudobinary eutectic in large droplets. Lamellar (Ag+ ζ) pseudobinary eutectic grain forms independently on condition that primary (Ag) dendrite cannot form and its eutectic morphology becomes anomalous with the decrease of droplet size. Nanoscaled (Ag+ ε 2) pseudobinary eutectic generating at the last stage of solidification is the product of both peri-eutectic and pseudobinary eutectic transitions. It distributes in the gaps of (Ag+ ζ) pseudobinary eutectic grains and its morphology remains lamellar regardless of droplet size. [Copyright &y& Elsevier]

Abstract

The establishment of reliable materials genome databases involving the thermophysical properties of liquid metals and alloys promotes the progress of materials research and development, whereas acquiring these properties imposes great challenges on experimental investigation. Here, we proposed a deep learning method and achieved a deep neural network (DNN) interatomic potential for the entire Ti–Ni–Cr–Al system from pure metals to quaternary alloys. This DNN potential exhibited sufficient temperature and compositional transformability which extended beyond the training and provided the prediction of the liquid structure and thermophysical properties for metallic materials with both density functional theory accuracy and classic molecular dynamics efficiency. The predicted results agreed well with the reported experimental data. This work opens a feasible way to address the challenges of rapidly and accurately acquiring thermophysical properties data for liquid pure metals and multicomponent alloys, covering a broad temperature range from superheated to undercooled state.
