Abstract: Intracontinental orogens remain less understood than accretionary or collisional orogens that are related to plate margin interactions. In the center of the South China block, the Xuefengshan Belt provides a well-exposed example of such an intracontinental orogen of Early Mesozoic age. Detailed field tectonic observations indicate that the Xuefengshan Belt can be divided into a Western Outer Zone characterized by km-scale box-fold structures, and an Eastern Zone, separated from the Western Outer Zone by the SE-dipping Main Xuefengshan Thrust. In the Eastern Zone, NW verging folds coeval with a pervasive slaty cleavage and a NW–SE trending lineation are the dominant structures. From west to east, the dip of the cleavage surface exhibits a fan-like pattern. The bulk architecture of the Xuefengshan Belt results from polyphase deformation: D1 is characterized by a top-to-the-NW ductile shearing; D2 corresponds to SE-directed back thrusting and folding; D3 consists of upright folds with vertical cleavage and lineation. At depth, a high strain zone characterized by greenschist facies metamorphic rocks and a top-to-the-NW ductile shearing corresponds to a ductile décollement zone that accommodated the deformation of the Neoproterozoic to Early Triassic sedimentary series. Kinematic compatibility suggests that the synmetamorphic ductile shearing was coeval with the D1 event in the sedimentary cover. The Xuefengshan Belt is interpreted as an Early Mesozoic intracontinental orogen, which possibly originated from the SE-directed continental subduction of a piece of the South China block in response to northwestwards subduction of the Pacific plate. In orogenic belts, a basal décollement zone often develops at depth to accommodate the shortening due to folding and thrusting of the sedimentary cover. In the Early Mesozoic intracontinental Xuefengshan Belt of South China, such a décollement zone is exposed in the core of anticlines formed by the emplacement of the late-orogenic granitic plutons. Our detailed, multi-scale structural analysis documents a synmetamorphic ductile deformation. In the basal décollement, the Neoproterozoic pelite and sandstone, and the intruding Early Paleozoic granites were deformed and metamorphosed into mylonites and orthogneiss, respectively. The metamorphic foliation contains a NW-SE stretching lineation associated with top-to-the-NW kinematic indicators. The ductile shearing of these high-strained rocks can be correlated with NW-verging folds and thrusts recognized in the Neoproterozoic to Early Triassic sedimentary cover. Monazite U-Th-Pb chemical dating, and zircon SIMS U-Pb dating provide age constraints of the ductile shearing between 243 and 226 Ma, and late-orogenic granite emplacement around 235-215 Ma. In agreement with recent geochronological data, these new results show that the Xuefengshan Belt is an Early Mesozoic orogen dominated by the NW-directed shearing and thrusting. At the southeastern boundary of the Xuefengshan Belt, the Chenzhou-Linwu fault separates the Early Mesozoic domain to the NW from the Early Paleozoic domain to the SE. The tectonic architecture of this belt was possibly originated from the continental underthrusting to the SE of the South China block in response to northwest-directed subduction of the Paleo-Pacific plate. Abstract: The Xuefengshan Belt, characterized by large-scale fold and thrust structures and widespread granites, is a key area to decipher the tectonic evolution of the South China block. In this belt, two magmatic episodes are recorded by Early Paleozoic and Early Mesozoic granites. In this paper, we carried out precise SIMS zircon U and in situ Lues measurements on these granitic plutons. Our study indicates that the Early Paleozoic and the Early Mesozoic granites are late-orogenic products of the Early Paleozoic orogen and the Middle Triassic Xuefengshan orogen, respectively. In the Xuefengshan Belt, the Early Paleozoic event is poorly registered, since this area corresponds to the outer zone of the early Paleozoic orogen. The Silurian–Early Devonian granites are late-orogenic plutons emplaced after the main tectonic and metamorphic stage in the Wuyi–Baiyun–Yunkai belt, coeval with a widespread, subsequent extensional event in eastern and southern South China. On the other hand, Triassic granites are formed in an intracontinental environment with weakly to strongly peraluminous signatures. Zircon Uresented here, associated with a summary of newly acquired data in the same region, suggest that the emplacement of anatectic granites occurred around 225–215Ma, not in a rather wide range of ca. 245–200Ma. In situ zircon εHf(t) values indicate a crust-derived source without a mantle-derived input for the two generations of granites. Combining our data with recent studies, we infer that the central area of the South China block has experienced two tectonothermal events: the Early Paleozoic magmatism developed as a result of the collapse of the Early Paleozoic orogen, while these Early Mesozoic granites can be the late-orogenic products of the intracontinental Xuefengshan orogen, most likely manifesting the far field effect by the subduction of the Paleo-Pacific ocean plate at the southeastern margin of the South China block. The Ediacaran-Cambrian transition was one of the most critical intervals in Earth history. During this interval, widespread chert was precipitated, commonly as a stratal wedge in carbonates, along the southern margin of the Yangtze Platform, South China. The chert wedge passes into a full chert succession further basinward to the south-east. Four lithotypes of chert are identified across the marginal zone in western Hunan: mounded, vein, brecciated and bedded chert. The mounded chert is characterized by irregular to digitiform internal fabrics, generally with abundant original vesicles and/or channels that mostly are lined by botryoidal chalcedony cements with minor quartz and barite crystals. The host chert (or matrix) of these mounds is dominated by amorphous cryptocrystalline silica, commonly disseminated with pyrite. The vein chert, with minor quartz locally, generally cross-cuts the overlying dolostone and chert horizons and terminates under the mounded and/or bedded chert bodies. The brecciated chert commonly occurs as splayed 'intrusions' or funnel-shaped wedges and cross-cuts the topmost dolostones. The bedded chert, the most common type, generally is thin to medium-bedded and laminated locally; it is composed of amorphous silica with minor amounts of black lumps. Microthermometry of fluid inclusions from vein and void-lining minerals (mainly quartz and barite) revealed homogenization temperatures from 120 to 180°C for the trapped primary fluids. Compositionally, these chert deposits generally are pure, with SiO2 > 92 wt%, and only minor Fe2O3 and Al2O3 contents, most of which show positive Europium anomalies in rare earth element patterns, especially for the mounded chert. All these data suggest that the marginal zone chert deposits resulted from a low-temperature, silica-rich hydrothermal system, in which the mounded chert was precipitated around the releasing vents, i.e. as silica chimneys. The vein and splayed brecciated chert, however, was formed along the syndepositional fault/fracture conduits that linked downward, while the bedded chert was precipitated in the quieter water column from the fallout of hydrothermal plumes onto the sea floor. These petrological and geochemical data provide compelling evidence and a new clue to the understanding of the extensive silica precipitation; rapid tectono-depositional and oceanic changes during the Ediacaran-Cambrian transition in South China. Abstract: The south-Liaodong peninsula massif is the easternmost Mesozoic metamorphic core complex, recognized in Eastern China. It provides a good example of the combination of ductile shearing, syn-kinematic plutonism and polyphase exhumation. The Jurassic granodioritic plutons, located at the footwall of the detachment normal fault, and dated here at ca 160Ma, recorded two different phases of cooling. A slow cooling regime of about 3–10°C/my prevailing before 122Ma, was followed by a significant increase in cooling rate of about 40–55°C/my after that time. By contrast, a single fast cooling path was recorded by the Cretaceous monzogranite situated in the footwall of the detachment normal fault. This result indicates that the Jurassic and Cretaceous plutons recorded different exhumation processes: a Jurassic slow or negligible exhumation and a Cretaceous fast one assisted by normal faulting. These two cooling stages correspond to distinct geodynamic processes during the Jurassic and Cretaceous. Extensional tectonics seems not significant before Early Cretaceous. The second stage, dominated by an extensional regime which develops after ca 120Ma, is tentatively correlated to the lithosphere removal of the North China Craton. [Copyright &y& Elsevier]

Antiphospholipid syndrome (APS) is an autoimmune disease with arteriovenous thrombosis and recurrent miscarriages as the main clinical manifestations. Due to the complexity of its mechanisms and the diversity of its manifestations, its diagnosis and treatment remain challenging issues. Antiphospholipid antibodies (aPL) not only serve as crucial "biomarkers" in diagnosing APS but also act as the "culprits" of the disease. Endothelial cells (ECs), as one of the core target cells of aPL, bridge the gap between the molecular level of these antibodies and the tissue and organ level of pathological changes. A more in-depth exploration of the relationship between ECs and the pathogenesis of APS holds the potential for significant advancements in the precise diagnosis, classification, and therapy of APS. Many researchers have highlighted the vital involvement of ECs in APS and the underlying mechanisms governing their functionality. Through extensive in vitro and in vivo experiments, they have identified multiple aPL receptors on the EC membrane and various intracellular pathways. The heterogeneity of the T cell receptor (TCR) repertoire critically influences the autoimmune response in obstetric antiphospholipid syndrome (OAPS) and is intimately associated with the prophylaxis of autoimmune disorders. Investigating the TCR diversity patterns in patients with OAPS is thus of paramount clinical importance. This investigation procured peripheral blood specimens from 31 individuals with OAPS, 21 patients diagnosed with systemic lupus erythematosus (SLE), and 22 healthy controls (HC), proceeding with TCR repertoire sequencing. Concurrently, adverse pregnancy outcomes in the OAPS cohort were monitored and documented over an 18-month timeframe. We paid particular attention to disparities in V/J gene utilisation and the prevalence of shared clonotypes amongst OAPS patients and the comparative groups. When juxtaposed with observations from healthy controls and SLE patients, immune repertoire sequencing disclosed irregular T- and B-cell profiles and a contraction of diversity within the OAPS group. Marked variances were found in the genomic rearrangements of the V gene, J gene, and V/J combinations. Utilising a specialised TCRβ repertoire, we crafted a predictive model for OAPS classification with robust discriminative capability (AUC = 0.852). Our research unveils alterations in the TCR repertoire among OAPS patients for the first time, positing potential covert autoimmune underpinnings. These findings nominate the TCR repertoire as a prospective peripheral blood biomarker for the clinical diagnosis of OAPS and may offer valuable insights for advancing the understanding of OAPS immunologic mechanisms and prognostic outcomes.

The gas chromatography retention index (RI) is an important parameter for the identification of different types of compounds in the field of chromatographic analysis; however, the experimental collection of RI values is a extremely cumbersome process. Thus, there is an urgent need for the establishment of a simple, efficient, and accurate model for the prediction of the RI values of compounds. In this study, first, the experimental RI values for 60 plant essential oil constituents were obtained. Next, a model describing the hologram quantitative structure-activity relationship (HQSAR) between the structural properties of the essential oil constituents and their RI values was investigated and constructed. The optimal HQSAR model was established by setting the model parameters "fragment size", "fragment distinction", "hologram length" and "principal components" to "1-4", "C, Ch", "199", and "4", respectively. Finally, the predictive ability of the model was verified using external test set validation and leave-one-out cross-validation (LOO-CV). The experimental results were as follows, the root mean square error of prediction (RMSEP), predictive determination coefficient ([Formula: see text]), concordance correlation coefficient (CCC), and mean relative error (MRE) for external test set validation were 40.45, 0.984, 0.968, and 2.20%, respectively. Meanwhile, the root mean square error of cross validation (RMSECV) and MRE for LOO-CV were 72.56 and 4.17%, respectively. These findings demonstrate that the established HQSAR model has a good predictive ability and can accurately predict the RI values of plant essential oil constituents. In addition, the molecular contribution maps of the HQSAR model revealed that the RI values of aromatic compounds increase when hydroxyl groups are connected to their alkyl chains. Aliphatic compounds feature long chain alkyl groups, which can lead to an increase in RI values. The above phenomena highlight the promising application prospects of HQSAR for studying the RI values of plant essential oil constituents. Therefore, this study provides a reliable theoretical basis for predicting the RI values of other essential oil constituents.

With the rapid development of the lithium ion battery industry, emerging lithium (Li) enrichment in nature has attracted ever-growing attention due to the biotoxicity of high Li levels. To date, fast lithium ion (Li + ) detection remains urgent but is limited by the selectivity, sensitivity, and stability of conventional technologies based on passive response processes. In nature, archaeal plasma membrane ion exchangers (NCLX_Mj) exhibit Li + -gated multi/monovalent ion transport behavior, activated by different stimuli. Inspired by NCLX_Mj, we design a pH-controlled biomimetic Li + -responsive solid-state nanochannel system for on-demand Li + detection using 2-(2-hydroxyphenyl)benzoxazole (HPBO) units as Li + recognition groups. Pristine HPBO is not reactive to Li + , whereas negatively charged HPBO enables specific Li + coordination under alkaline conditions to decrease the ion exchange capacity of nanochannels. On-demand Li + detection is achieved by monitoring the decline in currents, thereby ensuring precise and stable Li + recognition (>0.1 mM) in the toxic range of Li + concentration (>1.5 mM) for human beings. Pesticides have caused concerns about food safety due to their residual effects in vegetables and fruits. Imidacloprid, as the frequently used neonicotinoid pesticide, could harm cardiovascular and respiratory function and cause reproductive toxicity in humans. Therefore, reliable methods for portable, selective, and rapid detection are desirable to develop. Herein, we report a neuron-inspired nanofluidic biosensor based on a tyrosine-modified artificial nanochannel for sensitively detecting imidacloprid. The functional tyrosine is modified on the outer surface of porous anodic aluminum oxide to rapidly capture imidacloprid through π-π interactions and hydrogen bonds. The integrated nanofluidic biosensor has a wide concentration range from 10 -8 to 10 -4 g/mL with an ultralow detection limit of 6.28 × 10 -9 g/mL, which outperforms the state-of-the-art sensors. This work provides a new perspective on detecting imidacloprid residues as well as other hazardous pesticide residues in environmental and food samples.

In this paper, a series of porous hierarchical Mg/Al layered double hydroxides (named as LDH, TTAC-MgAl-LDH, CTAC-MgAl-LDH, and OTAC-MgAl-LDH) was synthesized by a simple green hydrothermal method using wormlike micelles formed by salicylic acid and surfactants with different carbon chain lengths (0, 14, 16, and 18) as soft templates. BET, XRD, FTIR, TG, and SEM characterizations were carried out in order to investigate the structure and properties of the prepared materials. The results showed that the porous hierarchical CTAC-MgAl-LDH had a large specific surface area and multiple pore size distributions which could effectively increase the reaction area and allow better absorption capability. Benefiting from the unique architecture, CTAC-MgAl-LDH exhibited a large adsorption capacity for sulfonated lignite (231.70 mg/g) at 25 °C and a pH of 7, which outperformed the traditional LDH (86.05 mg/g), TTAC-MgAl-LDH (108.15 mg/g), and OTAC-MgAl-LDH (110.51 mg/g). The adsorption process of sulfonated lignite followed the pseudo-second-order kinetics model and conformed the Freundlich isotherm model with spontaneous heat absorption, which revealed that electrostatic adsorption and ion exchange were the main mechanisms of action for the adsorption. In addition, CTAC-MgAl-LDH showed a satisfactory long-time stability and its adsorption capacities were still as high as 198.64 mg/g after two adsorption cycles.

Background: Detection of minimal residual disease (MRD) by multiparameter flow cytometry (MFC) is a well-established risk stratification factor and therapeutic modification strategy in B acute lymphoblastic leukemia (B-ALL). However, current 8 color (8c)-MFC for MRD detection had the sensitivity of 0.01% with false negative or positive. Hence, a more sensitive and applicable MFC-MRD method is urgently needed. The aim of this study is to establish a single-tube 21c-MFC method to detect B-ALL MRD, evaluate its performance, and to investigate its preliminary clinical application., Methods: We selected 21 markers to establish a single-tube 21c-MFC method. The repeatability and sensitivity of this method was validated by adding Nalm-6 cells to normal bone marrow. Samples from control group (n = 6), B-ALL group (n = 7) and complete remission (CR) group (n = 26) were detected by 21c- and 8c-MFC separately. The expression characteristics of these markers was analyzed in control and B-ALL group, and the consistency of 21c- and 8c-MFC in detecting MRD was compared., Results: Repeatability of this method was 1.91% of CV and sensitivity was up to 0.005%. In control group, the expression of CD81, CD97, and CD200 gradually decreased and CD44, HLA-DR, CD73, and CD72 gradually increased with the maturation of normal B cells. In B-ALL group, CD73stro, CD81low, CD44stro, CD123stro, and CD58stro showed high-frequency expression. The consistency rate of 21c- and 8c-MFC in detecting MRD was 96%., Conclusions: A single-tube 21c-MFC method was established for MRD detection in B-ALL and had higher sensitivity than the 8c-MFC method.

A biomimetic hierarchical membrane consisting of ZIF-8 and MXene with controllable morphology could be fabricated by the facile electrochemical deposition method, well-realizing Li + /Mg 2+ sieving. This membrane could work stably in real brine with perm-selectivity of Li + /Mg 2+ up to 47.4.

In order to solve the problem of poor dispersion and stability of mixed metal hydroxide (MMH), a kind of mixed metal hydroxide-like compound (MMHlc) gel was synthesized for use as the base mud in drilling fluid instead of bentonite gel. Na 2 CO 3 , Na 2 SiO 3 , and C 17 H 33 CO 2 Na were used as precipitants to form MMHlc with larger interlayer spacing and smaller particle size. MMHlc was synthesized by the coprecipitation method at 25 °C with a metal molar ratio of Mg:Al:Fe = 3:1:1. The performance evaluation of the treated drilling fluid showed that MMHlc (S2) synthesized using Na 2 SiO 3 as the precipitant had the characteristics of low viscosity, low filtration, and a high dynamic plastic ratio at 25 °C, which fully met the requirements of oil field application, and it maintained its excellent properties after being aged at 250 °C for 16 h. Linear expansion and rolling recovery experiments showed that the S2 sample had excellent rheological properties and good inhibition. X-ray diffraction and FT-IR experiments showed that S2 had the most complete crystal structure, its interlayer distance was large, and its ion exchange capacity was strong. The thermogravimetric experiment showed that the S2 crystal was stable and the temperature resistance of the crystal could reach 340 °C. Zeta potential, particle size analysis, SEM, and TEM results showed that S2 is a nanomaterial with a complete morphology and uniform distribution. The drilling fluid of this formula had the characteristics of low viscosity, low filtration loss, and a high dynamic plastic ratio, and it met the conditions for oil field application. Considering these results, the new MMH prepared by our research institute is a drilling fluid material that can be used at ultra-high temperatures and can provide important support for drilling ultra-deep wells.

Sequence variation resulting from the evolution of IGH clones and immunophenotypic drift makes it difficult to track abnormal B cells in children with precursor B cell acute lymphoblastic leukemia (pre-B-ALL) by flow cytometry, qPCR, or next-generation sequencing (NGS). The V-(D)-J regions of immunoglobulin and T cell receptor of 47 pre-B-ALL samples were sequenced using the Illumina NovaSeq platform. The IGH rod-like tracer consensus sequence was extracted based on its rod-like alpha-helices structural similarity predicted by AlphaFold2. Additional data from published 203 pre-B-ALL samples were applied for validation. NGS-IGH (+) patients with pre-B-ALL had a poor prognosis. Consistent CDR3-coded protein structures in NGS-IGH (+) samples could be extracted as a potential follow-up marker for pre-B-ALL children during treatment. Nanofluidic reverse electrodialysis provides an attractive way to harvest osmotic energy. However, most attention was paid to monotonous membrane structure optimization to promote selective ion transport, while the role of external fields and relevant mechanisms are rarely explored. Here, we demonstrate a Kevlar-toughened tungsten disulfide (WS 2 ) composite membrane with bioinspired serosa-mimetic structures as an efficient osmotic energy generator coupling light. As a result, the output power could be up to 16.43 W m -2 under irradiation, outperforming traditional two-dimensional (2D) membranes. Both the experiment and simulation uncover that the generated photothermal and photoelectronic effects could synergistically promote the confined ion transport process. In addition, this membrane also possesses great anti-fouling properties, endowing its practical application. The transcription factor nuclear factor erythroid 2 like 1 (NFE2L1 or NRF1) regulates constitutive and inducible expression of proteasome subunits and assembly chaperones. The precursor of NRF1 is integrated into the endoplasmic reticulum (ER) and can be retrotranslocated from the ER to the cytosol where it is processed by ubiquitin-directed endoprotease DDI2. DDI2 cleaves and activates NRF1 only when NRF1 is highly polyubiquitinated. It remains unclear how retrotranslocated NRF1 is primed with large amount of ubiquitin and/or very long polyubiquitin chain for subsequent processing. Here, we report that E3 ligase UBE4A catalyzes ubiquitination of retrotranslocated NRF1 and promotes its cleavage. Depletion of UBE4A reduces the amount of ubiquitin modified on NRF1, shortens the average length of polyubiquitin chain, decreases NRF1 cleavage efficiency and causes accumulation of non-cleaved, inactivated NRF1. Expression of a UBE4A mutant lacking ligase activity impairs the cleavage, likely due to a dominant negative effect. UBE4A interacts with NRF1 and the recombinant UBE4A can promote ubiquitination of retrotranslocated NRF1 in vitro. In addition, knocking out UBE4A reduces transcription of proteasomal subunits in cells. Iron overloads osteoporosis mainly occurs to postmenopausal women and people requiring repeated blood transfusions. Iron overload increases the activity of osteoclasts and decreases the activity of osteoblasts, leading to the occurrence of osteoporosis. Conventional treatment options include calcium supplements and iron chelators. However, simple calcium supplementation is not effective, and it does not have a good therapeutic effect. Oxidative stress is one of the triggers for osteoporosis. Therefore, the study focuses on the antioxidant aspect of osteoporosis treatment. The present work revealed that antioxidant carboxymethyl chitosan-based carbon dots (AOCDs) can effectively treat iron overload osteoporosis. More interestingly, the functional modification of AOCDs by doping calcium gluconate (AOCDs:Ca) is superior to the use of any single component. AOCDs:Ca have the dual function of antioxidant and calcium supplement. AOCDs:Ca effectively improve the bioavailability of calcium and achieve ultra-low concentration calcium supplement for the treatment of iron-induced osteoporosis in zebrafish.

Solid-free drilling fluid has more advantages as a new type of drilling fluid compared with traditional drilling fluid, such as improving drilling efficiency, protecting oil and not having clay particles clog the oil and gas layer. In this study, Zn/Cu/Fe-doped magnesium-aluminum hydroxide (Mg-Al MMH) was prepared using the co-precipitation method and evaluated in solid-free drilling fluid. The inhibition mechanism of synthesized hydroxide was analyzed by X-ray diffraction, laser particle-size analysis and thermogravimetric analysis. The samples were directly used as drilling fluid base muds for performance evaluation. The results showed that the linear expansion rate of 4% M6-Fe was only 12.32% at room temperature within 2 h, that the linear expansion rate was 20.28% at 90 °C and that the anti-swelling rate was 81.16% at room temperature, indicating that it has a strong inhibition ability at both room temperature and at high temperatures. Meanwhile, the possibility of multi-mixed metal hydroxide as a drilling fluid base mud is discussed in this study. We found that 4% M6-Fe exhibited low viscosity, a high YP/PV ratio and high temperature resistance, and its apparent viscosity retention rate reached 100% rolled at 200 °C for 16 h, with a YP/PV ratio of 2.33.

Fyn, a non-receptor tyrosine kinase, plays an important role in microglial-mediated neuroinflammation and may serve as a candidate therapeutic target for neuropsychiatric diseases. In this study, we discovered that chrysin, a natural flavonoid compound, suppressed the activation of Fyn kinase and further alleviated neuroinflammation-induced neuron damage and behavior deficits. Functionally, chrysin improved lipopolysaccharide (LPS)-induced memory impairment and depressive behaviors in mice, it also protected against LPS-induced neuronal degeneration and loss and synaptic defects in mice. Our study demonstrated that chrysin inhibited the activation of microglia and reduced the expression of NLRP3 and IL-1β. Furthermore, our data indicated that chrysin blocked phosphorylation of Fyn and activation of NF-κB. Transfection with siRNA-Fyn validated that knockdown of Fyn partly abolished the inhibitory effect of chrysin on the expression of the NLRP3 inflammasome and NF-κB activation. We demonstrate a compact and efficient Ho:YLF slab laser that is pumped by a self-made Tm:YAP slab laser. The maximum output power of 125 W at 2064 nm was obtained with the incident pump power of 245 W. To the best of our knowledge, this is the first report for an Ho:YLF laser oscillator to reach hundred-watts-level output power. The slope efficiency with respect to the incident pump power was 62.5%, and the optical-to-optical conversion efficiency was 51%. The RMS instability of the maximum output power was measured to be 0.24% in 30 min. The beam quality factors M 2 were 475 and 1.59 in the x and y directions, respectively.

Oyster is rich in plasmalogens that are ether phospholipids with biological functions to human body. Air-frying is a popular technique for preparing delicious oyster but makes the plasmalogens vulnerable to oxidation. In this study, the effect of air-frying processing on plasmalogens oxidation was studied by lipidomic approach. Plasmalogens were always mixed with normal phospholipids, thus the lipid extract was treated with mild acid hydrolysis to rapidly degrade plasmalogens owing to the acid lability of vinyl ether linkage at sn-1 position. After hydrophilic interaction chromatography MS/MS analysis, there were three plasmalogen classes, plasmanylcholine, plasmanylethanolamine, and plasmanylinositol, completely separated, and each plasmalogen molecular species was identified and quantified. It indicated that the content of plasmalogens underwent an obvious decrease during the air-frying process. To weaken such effect, the influence of air-frying temperature was further inspected by multivariate statistical analyses. The main variables, including the ions of m/z 756.4927, 784.5486, 828.5812, etc., were revealed by unsupervised principle component analysis, supervised orthogonal partial least-square analysis, and variable importance in projection plot. As a conclusion, air-frying has health benefits in reducing fat content but destructive to plasmalogens, thus interventions are recommended to prevent the degradation of plasmalogens., (© 2021 Wiley-VCH GmbH.)

Objective: We expected to demonstrate a practical framework for oral squamous cell carcinoma (OSCC) candidate biomarker analysis at the pathway level based on the attract method, so as to give great insights to reveal the pathological mechanism underlying this disease at its early stage., Methods: First, gene expression profile of OSCC was recruited and preprocessed. Then, Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analysis was conducted. Next, attract method, an approach that begins its analysis from the "foundation knowledge sets" to discriminate the cell-phenotypes by those well-annotated gene-sets, then expands the syn-expression groups via decomposing each significant pathway into correlated subsets and extends the analysis to the entire expression was applied to identify core pathways. Finally, gene ontology (GO) functional enrichment analysis was performed on each of the correlated set groups to discover any potentially shared biological themes., Results: A total of 226 pathways were obtained. Then, 39 core KEGG pathways was identified via attract. After removing the uninformative genes, a total of 1, 2, and 3 clusters were separately identified for the three discriminative pathways extracellular matrix (ECM)-receptor interaction, neuroactive ligand-receptor interaction, and cell adhesion molecules (CAMs) pathway based on the correlation coefficient < 0.85. GO functional enrichment analysis for the correlated partners groups indicated that there were 40, 11, 78 significant GO terms for ECM-receptor interaction, neuroactive ligand-receptor interaction, and CAMs pathway, respectively., Conclusions: We predict that pathways such as ECM-receptor interaction, neuroactive ligand-receptor interaction, and CAMs may play significant roles in OSCC and targeting these pathways may provide an effective avenue to combat the complicated illness., Competing Interests: None

In the early summer of 2012 and 2013, mass mortalities of blood ark shell (Scapharca [Anadara] broughtonii), broodstocks were reported in several hatcheries on the coast of northern China. Clinical signs including slow response, gaping valves and pale visceral mass were observed in diseased individuals. In response to these reported mortalities, 238 samples were collected from hatcheries at 6 sites. Microscopic changes including lysed connective tissue, dilation of the digestive tubules, eosinophilic inclusion bodies, nuclear chromatin margination and pyknosis were found in affected animals. Transmission electron microscopy (TEM) revealed herpes-like viral particles within the connective tissue of the mantle. Quantative PCR (qPCR) and nested PCR (nPCR) analysis using primers specific for ostreid herpesvirus 1 (OsHV-1) indicated significant higher prevalence of OsHV-1 DNA in cases associated with mass mortalities than those without mass mortalities (p = 0.0012 for qPCR, p < 0.0001 for nPCR). qPCR also indicated that samples associated with mass mortalities carried high viral DNA loads, while the loads in apparently healthy samples were significantly lower (t = 3.15, df = 92, p = 0.002). Sequence analysis of the C2/C6 region of nPCR products revealed 5 newly described variants, which were closely related to each other. Phylogenetic analysis of the 5 virus variants and 48 virus variants reported in previous studies identified 2 main phylogenetic groups, and the 5 virus variants identified here were allocated to a separate subclade. To our knowledge, this is the first report of mass mortalities of bivalve broodstocks associated with OsHV-1 infection.
