Brown carbon (BrC) is a type of organic carbon with light-absorbing abilities, especially in ultraviolet (UV) radiation, which could significantly contribute to global warming. Observations have shown high BrC concentrations and absorption in China, suggesting potentially large BrC emissions. The potential contribution of fossil fuel combustion to BrC emission has been ignored in most previous studies. Here, we use GEOS-Chem to simulate BrC distribution and absorption in China, accounting for three major primary BrC sources: residential coal and biofuel combustion, vehicle exhausts, and open biomass burning. Based on the literature and related energy consumption data, we estimate the specific emission ratio of BrC versus BC, and BrC mass absorption efficiency (MAE) for each source. Combined with BC emission, total BrC emission in China is then estimated to be 3.42 Tg yr−1 in 2018, of which 71% is from residential combustion, 14% is from vehicle exhaust, and 15% is from open biomass burning. Residential combustion is the main source of surface BrC in China, accounting for 60% on average, followed by open biomass burning (23%) and vehicle exhaust emissions (17%). There is a clear seasonality in surface BrC concentrations with the maximum in winter (5.1 µg m−3), followed by spring (2.8 µg m−3), autumn (2.3 µg m−3), and summer (1.3 µg m−3). BrC AAOD at 365 nm ranges from 0.0017 to 0.060 in China, mainly dominated by residential combustion (73%), followed by open biomass burning (16%), and vehicle exhaust emissions (11%). It is also estimated that BrC accounts for 45–67% (52% on average) of total carbonaceous aerosol AAOD at 365 nm, implying an equal importance of BrC and BC regarding the absorption in UV radiation.

Mass concrete is usually poured in layers. To ensure the interlayer bonding quality of concrete, the lower layer should be kept in a plastic state before the upper layer is added. Ultimately, it will lead to the prediction of concrete setting time as a critical task in concrete pouring. In this experiment, the setting time of concrete in laboratory and field environments was investigated. The equivalent age of concrete at the initial setting was also analyzed based on the maturity theory. Meanwhile, factors affecting the setting time in the field environment were studied by means of multiple stepwise regression analysis. Besides, the interlayer splitting tensile strength of concrete subjected to different temperatures and wind speeds was determined. The results of laboratory tests show that both setting time and interlayer splitting tensile strength of concrete decrease significantly with the increase of air temperature and wind speed. In addition, the equivalent age of concrete at initial setting remains the same when subjected to different temperatures, while it decreases obviously with the increase of wind speed. In the field environment, the equivalent age of concrete at initial setting is greatly different, which is related to the variability of relative humidity and wind speed. The average air temperature and maximum wind speed are the main factors affecting the initial setting time of concrete. Furthermore, a prediction model is established based on the stepwise regression analysis results, which can predict the actual setting state in real-time, and hence controlling the interlayer bonding quality of dam concrete.

A long-gauge fiber Bragg grating (FBG) strain sensor with enhanced strain sensitivity is proposed, which is encapsulated with two T-shaped metal blocks. Its fabrication method is described briefly, and the strain sensitivity can be flexibly adjusted through changing its packaging method. A series of experiments are carried out to study the packaging and its sensing properties. The experimental results show that the strain and temperature sensitivity coefficient of the sensor are three times larger than the common FBG sensors. The linearity coefficients of the FBG sensor are larger than 0.999, and the relative error of the repeatability of all sensor samples is less than 1%. Through the stability test on the actual bridge, it is revealed that the long-term stability of the sensor is excellent, and the maximum error is less than 1.5%. In addition, the proposed FBG strain sensors are used to conduct a shear strengthening experiment on a reinforced concrete (RC) beam to verify its working performance. The experimental results show that the strain change and crack propagation of the RC beam are well monitored by the sensors during the loading process.

Abstract Ecosystem activation system (EAS) was developed to create beneficial conditions for microbiome recovery and then restore and maintain the ecological integrity (microbial community, phytoplankton, zooplankton) for eutrophic freshwater rehabilitation. A 30 day’s filed test of EAS indicated that over 50% of contaminant was removed and the algae visibly disappeared. EAS treatment 2.5-fold increased the diversity of microbial community and changed the microbial community structure (e.g., two and three-fold decrease in the amount of Flavobacterium and Pseudomonas, typical abundant species of eutrophic freshwater, respectively). Further, the diversity of phytoplankton and zooplankton of treated water suggested that these species were diverse. Representative phytoplankton of eutrophic freshwater, Chlorella and Chlamydomonas were undetectable. The possible mechanism of EAS is restoring the trophic levels of the water body via bottom-up approach by microbial community.

Abstract Marek’s disease (MD), caused by Marek’s disease virus (MDV), is a lymphotropic neoplastic disease. Previous miRNAome analysis showed gga-miR-219b was significantly downregulated in MDV-induced lymphoma, and one of its potential target genes, B-cell chronic lymphocytic /lymphoma 11B (BCL11B) was predicted. In this study, we further investigated the function of gga-miR-219b, and the gain/loss of function assay showed gga-miR-219b inhibited cell migration and reduced cell proliferation by promoting apoptosis not by cell cycle arrest. Gga-miR-219b also suppressed expression of two cell invasion-related genes MMP2 and MMP9. The results indicated suppressive effect of gga-miR-219b on MD tumorigenesis. The gene BCL11B was verified as a direct target gene of gga-miR-219b. RNA interference was performed to block BCL11B. As expected, the effects triggered by BCL11B downregulation were in accordance with that triggered by gga-miR-219b overexpression, suggesting that BCL11B was a stimulative regulator of MD transformation. Moreover, both gga-miR-219b and BCL11B influenced the expression of Meq gene, the most important oncogene in MDV. Additionally, gene expression level of anti-apoptotic genes BCL2 and BCL2L1 was downregulated and pro-apoptotic gene TNFSF10 was upregulated in MSB1 cells with gga-miR-219b overexpression or BCL11B knockdown, which suggested gga-miR-219b promoted cell apoptosis via regulating gene expression in the apoptosis pathways.

Background/Aims: This study aimed to investigate the role of microRNA (miR)-122a in regulating zonulin during the modulation of intestinal barrier. Methods: Zonulin proteins and their target gene expression were analyzed in miR-122a-overexpressing cell lines and in the target gene of epidermal growth factor receptor (EGFR). An mmu-miR-122a intestinal epithelial conditional transgenic (miR-122a-TG) mouse model was established to investigate EGFR and zonulin expression. MiR-122a was also detected in the clinical specimens of inflammatory bowel disease. Results: EGFR was identified as a target gene of miR-122a. The expression level of miR-122a was positively correlated with that of zonulin. The expression level of zonulin was significantly increased, whereas the expression level of EGFR was significantly decreased in the miR-122a-TG mice and in the corresponding primary epithelial culture (P < 0.05). These results were consistent with the data of the clinical specimens. Conclusions: miR-122a could be a positive factor of zonulin by targeting EGFR, which increased the intestinal epithelial permeability in vivo and in vitro.

Blind Source Separation techniques are widely used in the field of wireless communication for a very long time to extract signals of interest from a set of multiple signals without training data. In this paper, we investigate the problem of separation of the human voice from a mixture of human voice and sounds from different musical instruments. The human voice may be a singing voice in a song or may be a part of some news, broadcast by a channel with background music. This paper proposes a generalized Short Time Fourier Transform (STFT)-based technique, combined with filter bank to extract vocals from background music. The main purpose is to design a filter bank and to eliminate background aliasing errors with best reconstruction conditions, having approximated scaling factors. Stereo signals in time-frequency domain are used in experiments. The input stereo signals are processed in the form of frames and passed through the proposed STFT-based technique. The output of the STFT-based technique is passed through the filter bank to minimize the background aliasing errors. For reconstruction, first an inverse STFT is applied and then the signals are reconstructed by the OverLap-Add method to get the final output, containing vocals only. The experiments show that the proposed approach performs better than the other state-of-the-art approaches, in terms of Signal-to-Interference Ratio (SIR) and Signal-to-Distortion Ratio (SDR), respectively.

Objective Irisin is a myokine that greatly affects energy expenditure and systemic metabolism. While thyroid hormone is likely associated with irisin, a direct relationship remains to be fully elucidated. This study aimed to investigate plasma irisin levels in Chinese patients with hypothyroidism. Methods A total of 155 subjects were divided into the hypothyroidism group or the control group. Fifty-seven patients in the hypothyroidism group received levothyroxine treatment. Baseline irisin levels were measured in the two groups and post-treatment levels were measured in the hypothyroidism group. Results Irisin levels were significantly lower in the hypothyroidism group than in the control group. In the hypothyroidism group, irisin levels were positively associated with free triiodothyronine and free thyroxine levels, and negatively associated with thyrotropin levels. In the hypothyroidism group, irisin levels were significantly increased after levothyroxine treatment. Multiple linear regression models showed that total cholesterol and free thyroxine levels were the only significant predictors of serum irisin levels. Conclusions Irisin levels are decreased in patients with hypothyroidism. Our results suggest that decreased irisin levels are directly associated with reduced thyroid hormone levels. These values may be restored after levothyroxine treatment in Chinese patients with hypothyroidism.

Retinal ganglion cells (RGCs) are the only output neurons that conduct visual signals from the eyes to the brain. RGC degeneration occurs in many retinal diseases leading to blindness and increasing evidence suggests that RGCs are susceptible to various injuries in a type-specific manner. Glutamate excitotoxicity is the pathological process by which neurons are damaged and killed by excessive stimulation of glutamate receptors and it plays a central role in the death of neurons in many CNS and retinal diseases. The purpose of this study is to characterize the susceptibility of genetically identified RGC types to the excitotoxicity induced by N-methyl-D-aspartate (NMDA). We show that the susceptibility of different types of RGCs to NMDA excitotoxicity varies significantly, in which the αRGCs are the most resistant type of RGCs to NMDA excitotoxicity while the J-RGCs are the most sensitive cells to NMDA excitotoxicity. These results strongly suggest that the differences in the genetic background of RGC types might provide valuable insights for understanding the selective susceptibility of RGCs to pathological insults and the development of a strategy to protect RGCs from death in disease conditions. In addition, our results show that RGCs lose dendrites before death and the sequence of the morphological and molecular events during RGC death suggests that the initial insult of NMDA excitotoxicity might set off a cascade of events independent of the primary insults. However, the kinetics of dendritic retraction in RGCs does not directly correlate to the susceptibility of type-specific RGC death.

In this study, we compared the antibacterial effectivity of the eggs of six precocial and four altricial bird species using Escherichia coli, based on their eggshell traits. The ultrastructure of eggshell was observed using a scanning electron microscope (SEM). According to SEM results, eggs from precocial birds (chicken, turkey, quail, duck, ostrich, and goose) had cuticle on the eggshells, while eggs from altricial birds (pigeon, budgerigar, munia, and canary) did not. The environment/selection pressure may induce the divergent evolution process in eggs of precocial and altricial birds. The E. coli experiment results showed that chicken, turkey, quail, duck, and goose eggs, with a high cuticle opacity, exhibited a much lower E. coli penetration rate. In contrast, the eggs with poor (ostrich) or without (pigeon, budgerigar, munia, and canary) cuticle exhibited a higher penetration rate. It is suggested that cuticle is a main barrier against bacterial penetration in precocial birds' eggs. Turkey and quail eggs showed the lowest E. coli contamination rate (3.33% and 2.22%, respectively), probably because of the tightly connected nanosphere structure on their cuticle. As for altricial birds' eggs, the eggs of budgerigar, munia, and canary with small pore diameter (0.57 to 1.22 μm) had a lower E. coli penetration rate than pigeon eggs (45.56%, 66.67%, 50%, and 97.78%, respectively, P < 0.05), indicating that pore diameter played a significant role in defending against bacterial trans-shell invasion. We found that eggshell thickness and pore area decreased with egg size. The cuticle quality had no relationship with egg size, but was closely related to the bird species. The E. coli penetration rate of altricial birds' eggs was significantly higher than that of precocial birds' eggs, mainly because the pores are exposed on the eggshell surface and cuticle protection is absent. This study provides detailed information on the eggshell cuticle, which gives insight into the cuticle evolution process that occurred in precocial and altricial bird species. Moreover, the results of E. coli penetration may help understanding the antibacterial behavior in birds.

During the stringent response, bacteria synthesize guanosine-3',5'-bis(diphosphate) (ppGpp) and guanosine-5'-triphosphate 3'-diphosphate (pppGpp), which act as secondary messengers to promote cellular survival and adaptation. (p)ppGpp 'alarmones' are synthesized and/or hydrolyzed by proteins belonging to the RelA/SpoT Homologue (RSH) family. Many bacteria also encode 'small alarmone synthetase' (SAS) proteins (e.g. RelP, RelQ) which may also be capable of synthesizing a third alarmone: guanosine-5'-phosphate 3'-diphosphate (pGpp). Here, we report the biochemical properties of the Rel (RSH), RelP and RelQ proteins from Staphylococcus aureus (Sa-Rel, Sa-RelP, Sa-RelQ, respectively). Sa-Rel synthesized pppGpp more efficiently than ppGpp, but lacked the ability to produce pGpp. Sa-Rel efficiently hydrolyzed all three alarmones in a Mn(II) ion-dependent manner. The removal of the C-terminal regulatory domain of Sa-Rel increased its rate of (p)ppGpp synthesis ca. 10-fold, but had negligible effects on its rate of (pp)pGpp hydrolysis. Sa-RelP and Sa-RelQ efficiently synthesized pGpp in addition to pppGpp and ppGpp. The alarmone-synthesizing abilities of Sa-RelQ, but not Sa-RelP, were allosterically-stimulated by the addition of pppGpp, ppGpp or pGpp. The respective (pp)pGpp-synthesizing activities of Sa-RelP/Sa-RelQ were compared and contrasted with SAS homologues from Enterococcus faecalis (Ef-RelQ) and Streptococcus mutans (Sm-RelQ, Sm-RelP). Results indicated that EF-RelQ, Sm-RelQ and Sa-RelQ were functionally equivalent; but exhibited considerable variations in their respective biochemical properties, and the degrees to which alarmones and single-stranded RNA molecules allosterically modulated their respective alarmone-synthesizing activities. The respective (pp)pGpp-synthesizing capabilities of Sa-RelP and Sm-RelP proteins were inhibited by pGpp, ppGpp and pppGpp. Our results support the premise that RelP and RelQ proteins may synthesize pGpp in addition to (p)ppGpp within S. aureus and other Gram-positive bacterial species.

AimsStress granules (SGs) are transient cytoplasmic mRNA and protein complexes that form in eukaryotic cells under stress. SGs are related to multiple diseases, but there are no reports of the existence of SGs in atrial fibrillation (AF).Methods and resultsCell models of AF were established by field stimulation at 600 times per minute. HL-1 cells, cardiomyocytes and cardiac fibroblasts were transfected with G3BP1-cDNA plasmid by Lipofectamine 2000. The presence of SGs was detected by immunofluorescence analysis against GTPase-activating protein SH3 domain binding protein 1 (G3BP1) and poly(A)-binding protein 1 (PABP-1) and electron microscopy. Stable HL-1 cell lines transfected with lentivirus overexpressing G3BP1were constructed to further induce the formation of SGs in AF. Reactive oxygen species (ROS) and calcium overload in tachypaced HL-1 cells were studied by flow cytometry. The effects of G3BP1 overexpression on cardiac fibroblast proliferation and the protein expression level of collagen I/III and fibronectin-1 were also studied. Additionally, we detected protein synthesis in general and in single cells by puromycin incorporation in paced HL-1 cells. Here, we first showed that SGs are present in both tachypaced mouse cardiomyocytes and HL-1 atrial cells, although the presence is partial and at a low level. G3BP1 overexpression promoted SG formation, inhibited the rapid pacing-induced increase in ROS level, and attenuated calcium overload in HL-1 cells (all PConclusionsSGs are rapidly induced and present partially in AF, and G3BP1 overexpression promotes SG formation and confers cytoprotection against oxidative stress, calcium overload and atrial fibrosis in AF.

Cilia-related proteins are believed to be involved in a broad range of cellular processes. Retinitis pigmentosa GTPase regulator interacting protein 1-like (RPGRIP1L) is a ciliary protein required for ciliogenesis in many cell types, including epidermal keratinocytes. Here we report that RPGRIP1L is also involved in the maintenance of desmosomal junctions between keratinocytes. Genetically disrupting the Rpgrip1l gene in mice caused intraepidermal blistering, primarily between basal and suprabasal keratinocytes. This blistering phenotype was associated with aberrant expression patterns of desmosomal proteins, impaired desmosome ultrastructure, and compromised cell-cell adhesion in vivo and in vitro. We found that disrupting the RPGRIP1L gene in HaCaT cells, which do not form primary cilia, resulted in mislocalization of desmosomal proteins to the cytoplasm, suggesting a cilia-independent function of RPGRIP1L. Mechanistically, we found that RPGRIP1L regulates the endocytosis of desmogleins such that RPGRIP1L-knockdown not only induced spontaneous desmoglein endocytosis, as determined by AK23 labeling and biotinylation assays, but also exacerbated EGTA- or pemphigus vulgaris IgG-induced desmoglein endocytosis. Accordingly, inhibiting endocytosis with dynasore or sucrose rescued these desmosomal phenotypes. Biotinylation assays on cell surface proteins not only reinforced the role of RPGRIP1L in desmoglein endocytosis, but also suggested that RPGRIP1L may be more broadly involved in endocytosis. Thus, data obtained from this study advanced our understanding of the biological functions of RPGRIP1L by identifying its role in the cellular endocytic pathway.

MicroRNAs (miRNAs) are small non-coding RNAs that typically inhibit the translation and stability of messenger RNAs (mRNAs). They are ~22 nucleotides long and control both physiological and pathological processes. Altered expression of miRNAs is often associated with human diseases. Thus, miRNAs have become important therapeutic targets, and some clinical trials investigating the effect of miRNA-based therapeutics in different types of diseases have already been conducted. The tumor microenvironment (TME) comprises cells such as infiltrated immune cells, cancer-associated endothelial cells (CAEs) and cancer-associated fibroblasts (CAFs), and all the components participate in the complicated crosstalk with tumor cells to affect tumor progression. Altered miRNAs expression in both these stromal and tumor cells could drive tumorigenesis. Thus, in this review, we discuss how aberrantly expressed miRNAs influence tumor progression; summarize the crosstalk between infiltrated immune cells, CAEs, CAFs, and tumor cells through miRNAs, and clarify the important roles of miRNAs in the tumor microenvironment, which may facilitate the clinical application of miRNA-based therapies.

Abstract Background: Restenosis after percutaneous coronary intervention in coronary heart disease remains an unsolved problem. Clusterin (CLU) (or Apolipoprotein [Apo] J) levels have been reported to be elevated during the progression of postangioplasty restenosis and atherosclerosis. However, its role in neointimal hyperplasia is still controversial. Objective: To elucidate the role Apo J in neointimal hyperplasia in a rat carotid artery model in vivo with or without rosuvastatin administration. Methods: Male Wistar rats were randomly divided into three groups: the control group (n = 20), the model group (n = 20) and the statin intervention group (n = 32). The rats in the intervention group were given 10mg /kg dose of rosuvastatin. A 2F Fogarty catheter was introduced to induce vascular injury. Neointima formation was analyzed 1, 2, 3 and 4 weeks after balloon injury. The level of Apo J was measured by real-time PCR, immunohistochemistry and western blotting. Results: Intimal/medial area ratio (intimal/medial, I/M) was increased after balloon-injury and reached the maximum value at 4weeks in the model group; I/M was slightly increased at 2 weeks and stopped increasing after rosuvastatin administration. The mRNA and protein levels of Apo J in carotid arteries were significantly upregulated after rosuvastatin administration as compared with the model group, and reached maximum values at 2 weeks, which was earlier than in the model group (3 weeks). Conclusion: Apo J served as an acute phase reactant after balloon injury in rat carotid arteries. Rosuvastatin may reduce the neointima formation through up-regulation of Apo J. Our results suggest that Apo J exerts a protective role in the restenosis after balloon-injury in rats.

Today, the rapid increase in phenotypic and genotypic information is leading to larger mixed model equations (MMEs) and rendering genetic evaluation more time-consuming. It has been demonstrated that a preconditioned conjugate gradient (PCG) algorithm via an iteration on data (IOD) technique is the most efficient method of solving MME at a low computing cost. Commonly used software applications implementing PCG by IOD merely employ functions from the Intel Math Kernel Library (MKL) to accelerate numerical computations and have not taken full advantage of the multicores or multiprocessors of computer systems to reduce the execution time. Making the most of multicore/multiprocessor systems, we propose PIBLUP, a parallel, shared memory implementation of PCG by IOD to minimize the execution time of genetic evaluation. In addition to functions in MKL, PIBLUP uses Message Passing Interface (MPI) shared memory programming to parallelize code in the entire workflow where possible. Results from the analysis of the two datasets show that the execution time was reduced by more than 80% when solving MME using PIBLUP with 16 processes in parallel, compared to a serial program using a single process. Retinal ganglion cells (RGCs) have a wide variety of dendritic architectures, which are critical for the formation of their function-specific synaptic circuitry. The developmental regulation of the dendrites of RGCs is thought to be subtype dependent. The purpose of this study is to characterize the dendritic development of a genetically identified RGC subtype, JamB RGCs (J-RGCs), and the roles of glutamate receptor activity on the dendritic development of these cells. We show that the dendrites of J-RGCs are strictly ramified in the outer portion of the inner plexiform layer (IPL) of the retina at the age of postnatal day 8 (P8), mimicking the ramification pattern of adults. However, several other important features of dendrites undergo substantial developmental refinement after P8. From P8 to P13, the dendritic development of J-RGCs is characterized by a dramatic increase of dendritic length and the size of the dendritic field. After eye opening, the dendritic development of J-RGCs is characterized by a tremendous decrease of the number of dendritic protrusions (spine-like structures) and a consolidation of the size of the dendritic field. To determine whether the dendritic development of J-RGCs is regulated by glutamatergic activity, we conditionally knocked out the expression of an obligatory subunit of N-methyl-D-aspartate receptors (NMDARs), NR1 (Grin1), in J-RGCs. We found that J-RGCs with the NMDAR mutation have decreased dendrite outgrowth and dendritic field expansion but increased number of dendritic protrusions before eye opening. To determine if visual experience regulates the development of J-RGC dendrites, we raised the mice in complete darkness after birth. Light deprivation prevented the decrease in the number of dendritic protrusions and the consolidation of the dendritic field of wild type (WT) mice after eye opening. However, light deprivation has no additional effect on the number of dendritic protrusions or the size of the dendritic field of J-RGCs with NMDAR mutation. Together, these results revealed the roles of light stimulation and NMDAR activity on the dendritic development of J-RGCs.

A new method was proposed for the damage identification of box girder bridges under moving load, wherein the difference of strain influence line (DSIL) was taken as an index to represent the long-gauge strain difference before and after damage. The damage identification theory based on long-gauge strain influence lines was derived for box girder bridges with shear lag effect under consideration, and a regularized index DSIL was proposed for the quantitative identifications of damage location and extent. A series of experiments were carried out to study the influences of speed, vehicle type, and vehicle weight on the damage identification, and the experimental data were obtained by long-gauge fiber Bragg grating strain sensors. Moreover, numerical simulations were performed to confirm the method. The experimental and numerical results show that the method can locate the damage accurately, and quantitatively identify the damage extent under different working conditions. The experimental damage extent is generally slightly higher than the theoretical, with an average identification error smaller than 5%. Additionally, the relative error of damage extent is smaller than 3% under different working conditions. Thus, the effectiveness of this method was verified.

In vitro models of the liver have a good simulation of the micro-liquid environment inside the human liver and the communication between cell tissues, which provides an important research tool for drug research and liver disease treatment. In this paper, we designed a 3D liver chip and real-time monitoring system based on microfluidic technology. The in vitro model of the liver on the chip was established by the three-dimensional microfluidic chip pipeline and the corresponding microwell array. Meanwhile, the culture medium is continuously injected on the chip, and the electrochemical impedance spectroscopy and near-infrared spectroscopy of the liver chip are recorded and analyzed from day one to day five. When the 3D cultured liver chip in vitro model reached a certain period and stabilized, paracetamol with varying gradients of concentration was applied to the cultured cells for drug resistance testing. The experimental results show that the liver chip and its monitoring system designed in this paper can maintain 100% cell viability of hepatocytes in vitro for a long time. Furthermore, it can meet the requirements of measurement technologies such as electrical impedance measurement and near-infrared spectroscopy in real-time, providing a stable culture platform for the further study of organ chips.

Textured surface anomaly detection is a significant task in industrial scenarios. In order to further improve the detection performance, we proposed a novel two-stage approach with an attention mechanism. Firstly, in the segmentation network, the feature extraction and anomaly attention modules are designed to capture the detail information as much as possible and focus on the anomalies, respectively. To strike dynamic balances between these two parts, an adaptive scheme where learnable parameters are gradually optimized is introduced. Subsequently, the weights of the segmentation network are frozen, and the outputs are fed into the classification network, which is trained independently in this stage. Finally, we evaluate the proposed approach on DAGM 2007 dataset which consists of diverse textured surfaces with weakly-labeled anomalies, and the experiments demonstrate that our method can achieve 100% detection rates in terms of TPR (True Positive Rate) and TNR (True Negative Rate).

This paper investigates the detection of the transmitted power violation (TPV) in the satellite-terrestrial integrated network, where the terrestrial base station may break the spectrum policies so that severe damages are made to the satellite systems. Due to the lack of prior information on specific abnormal behaviors, this problem is complex and challenging. To tackle it, we first turn to the geolocation spectrum database based detecting framework, where not only the tasks of each segment but also the spectrum policies are specified. Then, the ternary hypothesis test and the generalized Neyman–Pearson (GMNP) test criterion are applied to maximize the detection probability under the false-alarm constraint. What is more, the Abnormal after Normal (AaN) detector is developed to simplify the analysis. Finally, simulations are conducted to demonstrate that the proposed detector can realize the detection of TPV in most cases at the expense of less than 10% detection probability.

Panax ginseng C.A. Meyer is extensively used as a food additive because of its medicinal and nutritional properties. This study investigated the inhibitory activity of eight ginsenosides against α-glycosidase and protein tyrosine phosphatase 1B (PTP1B). Results showed the anti-hyperglycaemic activities of the eight ginsenosides were in the order of 20(R)-dammarane-3β, 6α, 12β, 20, 25-pentol (25-OH-PPT) > 20(R)-25-methoxydammarane-3β, 12β, 20-tetrol (25-OCH3-PPT) > 20(R)-protopanaxatriol (PPT) > 20(S)-panaxatriol (PT) > 20(R)-dammarane-3β, 12β, 20, 25-tetrol (25-OH-PPD) > 20(R)-25-methoxydammarane-3β, 12β, 20-triol (25-OCH3-PPD) > 20(R)-protopanaxadiol (PPD) > 20(S)-panaxadiol (PD), and 25-OH-PPT exerted stronger inhibitory activity than acarbose and Na3VO4. Meanwhile, simultaneous quantitative estimation results of eight ginsenosides suggested good linear regression within test ranges, precision, accuracy. 25-OH-PPT of the stems (leaves), flowers, and fruits contained 2.69, 3.31 and 7.20%, respectively. High performance liquid chromatography-evaporative light-scattering detector (HPLC-ELSD) method was proven to be simple, fast, and effective.

Marek’s disease (MD) is a highly contagious viral neoplastic disease caused by Marek’s disease virus (MDV), which can lead to huge economic losses in the poultry industry. Recently, microRNAs (miRNAs) have been found in various cancers and tumors. In recent years, 994 mature miRNAs have been identified through deep sequencing in chickens, but only a few miRNAs have been investigated further in terms of their function. Previously, gga-miR-103-3p was found downregulated in MDV-infected samples by using Solexa deep sequencing. In this study, we further verified the expression of gga-miR-103-3p among MDV-infected spleen, MD lymphoma from liver, noninfected spleen, and noninfected liver, by qPCR. The results showed that the expression of gga-miR-103-3p was decreased in MDV-infected tissues, which was consistent with our previous study. Furthermore, two target genes of gga-miR-103-3p, cyclin E1 (CCNE1) and transcription factor Dp-2 (E2F dimerization partner 2) (TFDP2), were predicted and validated by luciferase reporter assay, qPCR, and western blot analysis. The results suggested that CCNE1 and TFDP2 are direct targets of gga-miR-103-3p in chickens. Subsequent cell proliferation and migration assay showed that gga-miR-103-3p suppressed MDCC-MSB1 migration, but did not obviously modulate MDCC-MSB1 cell proliferation. In conclusion, gga-miR-103-3p targets the CCNE1 and TFDP2 genes, and suppresses cell migration, which indicates that it might play an important role in MD tumor transformation.

Abstract: Damaged eggshells result in losses of eggs. The genetic mechanism of variable eggshell strength is still unclear. The current study was conducted to verify whether the eggshell calcification related genes, CALB1, SPP1, DMP4, BMP2 and SLIT2, were associated with eggshell mechanical property. For this purpose quantitative PCR (q-PCR) analysis was performed to detect gene expression between two groups of hens laying strong and weak eggs. The hens were selected from 360 White Leghorn layers at 60 wk to ensure that the strong and weak eggs differed significantly in breaking strength but not in eggshell thickness and weight. Purpose: Social influence has a decisive role in shaping a person"s cognition and behavior. Chinese face consciousness, including moral component, is an important part of Chinese traditional culture, which influences people to implement moral behavior. With both eye-tracking technology and traditional questionnaire, this research aims to explore people"s moral psychology and the psychological processing mechanisms of Chinese face consciousness, as well as the impact of Chinese face consciousness on the preference for the ecological product. Method and Data: 75 college and MBA students" eye movement data were collected when they read different kinds of moral materials, as well as data from the subsequent questionnaires. To test the hypothesis, ANOVA analysis and Heat Map analysis were performed. Besides, the PROCESS of bootstrap was used to test mediation effect. Findings: The results reveal that: 1. Compared to the moral-situation reading, when subjects read immoral situations, they need more processing time due to the moral dissonance and cognitive load. 2. Compared to the control condition, when threatened moral self is primed, subjects prefer to choose ecological product. 3. Protective face orientation is the mediator between threatened moral self and preference to ecological product. Key Contributions: First, this study broadens the use of eye-tracking technology in marketing and demonstrates a better understanding of the relationship between morality and consumer behavior in a more scientific way. Second, this study not only distinguishes the meanings between “protective face orientation" and “acquisitive face orientation", but also innovatively validates that when moral self is threatened, consumers tend to choose ecological product as moral compensation in order to protect their face. It can shed light on the promotion of ecological product in practical applications.

Background: Bispectral index (BIS) is considered very useful to guide anesthesia care in elderly patients, but its use is controversial for the evaluation of the adequacy of analgesia. This study compared the BIS changes in response to loss of consciousness (LOC) and loss of somatic response (LOS) to nociceptive stimuli between elderly and young patients receiving intravenous target-controlled infusion (TCI) of propofol and remifentanil. Methods: This study was performed on 52 elderly patients (aged 65–78 years) and 52 young patients (aged 25–58 years), American Society of Anesthesiologists physical status I or II. Anesthesia was induced with propofol administered by TCI. A standardized noxious electrical stimulus (transcutaneous electrical nerve stimulation, [TENS]) was applied (50 Hz, 80 mA, 0.25 ms pulses for 4 s) to the ulnar nerve at increasing remifentanil predicted effective-site concentration (Ce) until patients lost somatic response to TENS. Changes in awake, prestimulus, poststimulus BIS, heart rate, mean arterial pressure, pulse oxygen saturation, predicted plasma concentration, Ce of propofol, and remifentanil at both LOC and LOS clinical points were investigated. Results: BISLOCin elderly group was higher than that in young patient group (65.4 ± 9.7 vs. 57.6 ± 12.3) (t = 21.58, P < 0.0001) after TCI propofol, and the propofol Ce at LOC was 1.6 ± 0.3 μg/ml in elderly patients, which was significantly lower than that in young patients (2.3 ± 0.5 μg/ml) (t = 7.474, P < 0.0001). As nociceptive stimulation induced BIS to increase, the mean of BIS maximum values after TENS was significantly higher than that before TENS in both age groups (t = 8.902 and t = 8.019, P < 0.0001). With increasing Ce of remifentanil until patients lost somatic response to TENS, BISLOSwas the same as the BISLOCin elderly patients (65.6 ± 10.7 vs. 65.4 ± 9.7), and there were no marked differences between elderly and young patient groups in BISawake, BISLOS, and Ce of remifentanil required for LOS. Conclusion: In elderly patients, BIS can be used as an indicator for hypnotic-analgesic balance and be helpful to guide the optimal administration of propofol and remifentanil individually. This paper reports the mineralogy and geochemistry of the Early Permian No. 5 coal from the Chuancaogedan Mine, Junger Coalfield, China, using optical microscopy, scanning electron microscopy (SEM), Low-temperature ashing X-ray diffraction (LTA-XRD) in combination with Siroquant software, X-ray fluorescence (XRF), and inductively coupled plasma mass spectrometry (ICP-MS). The minerals in the No. 5 coal from the Chuancaogedan Mine dominantly consist of kaolinite, with minor amounts of quartz, pyrite, magnetite, gypsum, calcite, jarosite and mixed-layer illite/smectite (I/S). The most abundant species within high-temperature plasma-derived coals were SiO2 (averaging 16.90%), Al2O3 (13.87%), TiO2 (0.55%) and P2O5 (0.05%). Notable minor and trace elements of the coal include Zr (245.89 mg/kg), Li (78.54 mg/kg), Hg (65.42 mg/kg), Pb (38.95 mg/kg), U (7.85 mg/kg) and Se (6.69 mg/kg). The coal has an ultra-low sulfur content (0.40%). Lithium, Ga, Se, Zr and Hf present strongly positive correlation with ash yield, Si and Al, suggesting they are associated with aluminosilicate minerals in the No. 5 coal. Arsenic is only weakly associated with mineral matter and Ge in the No. 5 coals might be of organic and/or sulfide affinity.

According to the practice of temperature control in dam concrete, within a few days after a concrete block is poured, the temperature at the core of the concrete rises rapidly. The maximum temperature may still exceed the standard even under a relatively perfect post-cooling system, which is mostly caused by failure to quickly and correctly judge the development of the early-age temperature. This study investigates concrete temperature at an early age via in situ monitoring data collected from Baihetan arch dam and Wudongde arch dam. A simplified algorithm of temperature prediction is formed, which only considers the heat released by cement hydration and the cooling effect of cooling pipes. The influence of a cooling pipe on the measuring point of the thermometer is investigated, and a simple empirical formula to calculate the cooling effect is obtained. An equation for the rate of hydration temperature rise is achieved by combining measured data and the formula used to calculate the cooling effect. Furthermore, through the explorations of the related data, it is determined that the cement hydration ratio of the two dams is quite low during concreting. On the basis of the data collected from the field, the method to predict temperature proposed in this study is tested and proven.

Retinal ganglion cell (RGC) death occurs in many eye diseases, such as glaucoma and traumatic optic neuropathy (TON). Increasing evidence suggests that the susceptibility of RGCs varies to different diseases in an RGC type-dependent manner. We previously showed that the susceptibility of several genetically identified RGC types to N-methyl-D-aspartate (NMDA) excitotoxicity differs significantly. In this study, we characterize the susceptibility of the same RGC types to optic nerve crush (ONC). We show that the susceptibility of these RGC types to ONC varies significantly, in which BD-RGCs are the most resistant RGC type while W3-RGCs are the most sensitive cells to ONC. We also show that the survival rates of BD-RGCs and J-RGCs after ONC are significantly higher than their survival rates after NMDA excitotoxicity. These results are consistent with the conclusion that the susceptibility of RGCs to ONC varies in an RGC type-dependent manner. Further, the susceptibilities of the same types of RGCs to ONC and NMDA excitotoxicity are significantly different. These are valuable insights for understanding of the selective susceptibility of RGCs to various pathological insults and the development of a strategy to protect RGCs from death in disease conditions.

We explore a one-stage method for surface anomaly detection in industrial scenarios. On one side, encoder-decoder segmentation network is constructed to capture small targets as much as possible, and then dual background suppression mechanisms are designed to reduce noise patterns in coarse and fine manners. On the other hand, a classification module without learning parameters is built to reduce information loss in small targets due to the inexistence of successive down-sampling processes. Experimental results demonstrate that our one-stage detector achieves state-of-the-art performance in terms of precision, recall and f-score.

Graphene has huge application potential in the field of micro-nano devices, and due to the micro/nano-level application size, the thermal expansion coefficient of graphene has a certain impact on device performance. In this paper, a comprehensive study was carried out by molecular dynamics on the in-plane and through-plane thermal expansion effect of graphene. First of all, the in-plane thermal expansion coefficients of zigzag and armchair graphene were analyzed; they were similar and relatively close to the experiment value in the temperature range of 223–428 K. Secondly, the through-plane thermal expansion effect of double-layered graphene was investigated; the thermal expansion coefficient of through-plane was an order of magnitude larger than that of in-plane. Finally, the influence of graphene stacking on the thermal expansion effect was studied; Order-stacked graphene has a greater thermal expansion effect than that of AB- and ABC-stacked graphene. These discoveries in this article are of great significance to the development and application of graphene-based micro-nano devices.

Heterosis, a phenomenon characterized by the superior performance of hybrid individuals relative to their parents, has been widely utilized in livestock and crop breeding, while the underlying genetic basis remains elusive in chickens. Here, we performed a reciprocal crossing experiment with broiler and layer chickens and conducted RNA sequencing on liver tissues for reciprocal crosses and their parental lines to identify inheritance patterns of gene expression. Our results showed that heterosis of the abdominal fat percentage was 69.28%−154.71% in reciprocal crosses. Over-dominant genes of reciprocal crosses were significantly enriched in three biological pathways, namely, butanoate metabolism, the synthesis and degradation of ketone bodies, and valine, leucine, and isoleucine degradation. Among these shared over-dominant genes, we found that a lipid-related gene, HMGCL, was enriched in these pathways. Furthermore, we validated this gene as over-dominant using qRT-PCR. Although no shared significant pathway was detected in the high-parent dominant genes of reciprocal crosses, high-parent dominant gene expression was the major gene inheritance pattern in reciprocal crosses and we could not exclude the effect of high-parent dominant genes. These findings suggest that non-additive genes play important roles in the heterosis of important traits in chickens and have important implications regarding our understanding of heterosis.

To study the effect of redispersible polymer emulsion powder on the mechanical properties of carbon fiber-reinforced polymer concrete (CFRPC), the compressive, flexural, and splitting tests of CFRPC specimens with different polymer−cement ratios (polymer−cement mass ratios) were performed in this study. The modification effect of emulsion powder on CFRPC was analyzed from the perspectives of the strength and deformation properties of the specimens. The results show that the static properties of CFRPC increased first and then decreased with the increase of the polymer−cement ratio, in which the splitting tensile strength had the most significant increase; the flexural strength took second place and the compressive strength had a slight increase. When the polymer−cement ratio was 8%, the flexural and splitting tensile strength of the CFRPC specimens increased significantly by 36% and 61%, respectively. According to electron microscopy images, adding emulsion powder can effectively improve the structure of fiber−matrix transition zones and enhance the bond property between fibers and the matrix.

Combining research areas of biomechanics and pedestrian dead reckoning (PDR) provides a very promising way for pedestrian positioning in environments where Global Positioning System (GPS) signals are degraded or unavailable. In recent years, the PDR systems based on a smartphone’s built-in inertial sensors have attracted much attention in such environments. However, smartphone-based PDR systems are facing various challenges, especially the heading drift, which leads to the phenomenon of estimated walking path passing through walls. In this paper, the 2D PDR system is implemented by using a pocket-worn smartphone, and then enhanced by introducing a map-matching algorithm that employs a particle filter to prevent the wall-crossing problem. In addition, to extend the PDR system for 3D applications, the smartphone’s built-in barometer is used to measure the pressure variation associated to the pedestrian’s vertical displacement. Experimental results show that the map-matching algorithm based on a particle filter can effectively solve the wall-crossing problem and improve the accuracy of indoor PDR. By fusing the barometer readings, the vertical displacement can be calculated to derive the floor transition information. Despite the inherent sensor noises and complex pedestrian movements, smartphone-based 3D pedestrian positioning systems have considerable potential for indoor location-based services (LBS).

Cytochrome P450 (CYP) superfamily enzymes are broadly involved in a variety of physiological and toxicological processes. However, genome-wide analysis of this superfamily has never been investigated in the chicken genome. In this study, genome-wide analyses identified 45 chicken CYPs (cCYPs) from the chicken genome, and their classification and evolutionary relationships were investigated by phylogenetic, conserved protein motif, and gene structure analyses. The comprehensive evolutionary data revealed several remarkable characteristics of cCYPs, including the highly divergent and rapid evolution of the cCYPs, and the loss of cCYP2AF in the chicken genome. Furthermore, the cCYP expression profile was investigated by RNA-sequencing. The differential expression of cCYPs in developing embryos revealed the involvement of cCYPs in embryonic development. The significantly regulated cCYPs suggested its potential role in hepatic metabolism. Additionally, 11 cCYPs, including cCYP2AC1, cCYP2C23a, and cCYP2C23b, were identified as estrogen-responsive genes, which indicates that these cCYPs are involved in the estrogen-signaling pathway. Meanwhile, an expression profile analysis highlights the divergent role of different cCYPs. These data expand our view of the phylogeny and evolution of cCYPs, provide evolutionary insight, and can help elucidate the roles of cCYPs in physiological and toxicological processes in chicken.

Egg weight (EW) is an economically-important trait and displays a consecutive increase with the hen's age. Because extremely large eggs cause a range of problems in the poultry industry, we performed a genome-wide association study (GWAS) in order to identify genomic variations that are associated with EW. We utilized the Affymetrix 600 K high density SNP array in a population of 1,078 hens at seven time points from day at first egg to 80 weeks age (EW80). Results reveal that a 90 Kb genomic region (169.42 Mb ~ 169.51 Mb) in GGA1 is significantly associated with EW36 and is also potentially associated with egg weight at 28, 56, and 66 week of age. The leading SNP could account for 3.66% of the phenotypic variation, while two promising genes (DLEU7 and MIR15A) can be mapped to this narrow significant region and may affect EW in a pleiotropic manner. In addition, one gene (CECR2 on GGA1) and two genes (MEIS1 and SPRED2 on GGA3), which involved in the processes of embryogenesis and organogenesis, were also considered to be candidates related to first egg weight (FEW) and EW56, respectively. Findings in our study could provide worthy theoretical basis to generate eggs of ideal size based on marker assisted breeding selection.

Selection for cold tolerance in chickens is important for improving production performance and animal welfare. The identification of chicken breeds with higher cold tolerance and production performance will help to target candidates for the selection. The thyroid gland plays important roles in thermal adaptation, and its function is influenced by breed differences and transcriptional plasticity, both of which remain largely unknown in the chicken thyroid transcriptome. In this study, we subjected Bashang Long-tail (BS) and Rhode Island Red (RIR) chickens to either cold or warm environments for 21 weeks and investigated egg production performance, body weight changes, serum thyroid hormone concentrations, and thyroid gland transcriptome profiles. RIR chickens had higher egg production than BS chickens under warm conditions, but BS chickens produced more eggs than RIRs under cold conditions. Furthermore, BS chickens showed stable body weight gain under cold conditions while RIRs did not. These results suggested that BS breed is a preferable candidate for cold-tolerance selection and that the cold adaptability of RIRs should be improved in the future. BS chickens had higher serum thyroid hormone concentrations than RIRs under both environments. RNA-Seq generated 344.3 million paired-end reads from 16 sequencing libraries, and about 90% of the processed reads were concordantly mapped to the chicken reference genome. Differential expression analysis identified 46-1,211 genes in the respective comparisons. With regard to breed differences in the thyroid transcriptome, BS chickens showed higher cell replication and development, and immune response-related activity, while RIR chickens showed higher carbohydrate and protein metabolism activity. The cold environment reduced breed differences in the thyroid transcriptome compared with the warm environment. Transcriptional plasticity analysis revealed different adaptive responses in BS and RIR chickens to cope with the cold, and showed higher responsiveness in BS compared with RIR chickens, suggesting greater adaptability of the thyroid in BS chickens. Moreover, 10,053 differential splicing events were revealed among the groups, with RNA splicing and processing, gene expression, transport, and metabolism being the main affected biological processes, identifying a valuable alternative splicing repertoire for the chicken thyroid. A short isoform of TPO (encoding thyroid peroxidase) containing multiple open reading frames was generated in both breeds by skipping exons 4 and 5 in the cold environment. These findings provide novel clues for future studies of the molecular mechanisms underlying cold adaptation and/or acclimation in chickens.

A desktop NMR spectrometer was used to qualitatively analyze samples in drug-related cases in order to enhance the accuracy of the results and identify new drugs. Twelve known drugs and their derivatives were used to establish the parameters, conditions, and procedures for the methods and validate the feasibility and reliability of the methods. First, 1-D and 2-D NMR data for these 12 drugs and their derivatives were obtained in detail using a 600-MHz NMR spectrometer to create a data library. Next, some of these 12 drugs were analyzed using a Picospin 80 MHz desktop NMR spectrometer to set up the analytical procedure and method. With the procedure and method established, real case samples were analyzed and the data were compared to those obtained by a standard method. The results indicate that the desktop NMR spectrometer is a reliable and promising approach that can be used in criminology to quickly identify whether or not samples contain illegal drugs.

BackgroundThe prevalence of atrial fibrillation (AF) is significantly higher in rheumatoid arthritis (RA) patients, but the underlying mechanisms remain poorly understood. The goal of this study was to assess the effects of RA on AF susceptibility and atrial arrhythmogenic remodeling in a rat model of RA. Methods and ResultsCollagen‐induced arthritis was induced in rats by immunization with type II collagen in Freund's incomplete adjuvant. Among the rats that developed arthritis, AF susceptibility and atrial remodeling were examined 8 weeks after the primary immunization. AF inducibility and duration were substantially increased in collagen‐induced arthritis rats, and AF duration was significantly and positively correlated with the serum IL‐6 and TNF‐α levels. Rats with collagen‐induced arthritis showed prolonged atrial conduction time with no changes in the atrial effective refractory period. Atrial conduction delay was accompanied by significantly increased atrial fibrosis. In addition, atrial structural and autonomic remodeling, including left atrial dilation, apoptosis and autophagy of atrial myocytes, and atrial heterogeneous sympathetic hyperinnervation, was observed. Interestingly, we found that collagen‐induced arthritis had no significant effects on connexins, Nav1.5, and the main ion channels' protein expressions in atria. ConclusionsWe demonstrated that RA increased AF susceptibility by inducing AF‐promoting atrial remodeling. This study may provide insights into mechanisms underlying RA‐induced AF and validate a model that is suitable for further mechanistic and therapeutic exploration.

Cerebral ischemia-reperfusion injury (IRI) has a complex pathogenesis, and interleukin-17 (IL-17) is a newly identified class of the cytokine family that plays an important role in ischemic inflammation. An oxygen-glucose deprivation (OGD) model showed that IL-17A expression was significantly up-regulated in microglial cells. After IL-17A siRNA transfection, the inhibition of proliferation, and the increased apoptosis in microglial cells, induced by OGD/reperfusion, was improved, and the elevation of Caspase-3, Caspase-8, Caspase-9, and poly ADP ribose polymerase (PARP) activities was inhibited. Mass spectrometry demonstrated that IL-17A functioned through a series of factors associated with oxidative stress and apoptosis and regulated Caspase-3 activity and apoptosis in microglial cells via the p53 and PI3K/Akt signaling pathways. IL-17A, HMGB1, and ROS were regulated mutually to exhibit a synergistic effect in the OGD model of microglial cells, but the down-regulation of IL-17A or HMGB1 expression did not completely inhibit the production of ROS. These findings demonstrated that ROS might be located upstream of IL-17A and HMGB1 so that ROS can regulate HMGB1/IL-17A expression to affect the p53 and PI3K/Akt signaling pathways and therefore promote the occurrence of apoptosis in microglial cells. These findings provide a novel evidence for the role of IL-17A in ischemic cerebral diseases.

With better understanding of the quality and physico-mechanical properties of rocks of dam foundation, and the physico-mechanical properties and structure design of arch dam in association with the foundation excavation of Xiluodu arch dam, the excavation optimization design was proposed for the foundation surface on the basis of feasibility study. Common analysis and numerical analysis results demonstrated the feasibility of using the weakly weathered rocks III1 and III2 as the foundation surface of super-high arch dam. In view of changes in the geological conditions at the dam foundation along the riverbed direction, the design of extending foundation surface excavation area and using consolidating grouting and optimizing structure of dam bottom was introduced, allowing for harmonization of the arch dam and foundation. Three-dimensional (3D) geomechanics model test and finite element analysis results indicated that the dam body and foundation have good overload stability and high bearing capacity. The monitoring data showed that the behaviors of dam and foundation correspond with the designed patterns in the construction period and the initial operation period.

Objective: To review the functional mechanism of apolipoprotein J (apoJ) in the process of atherosclerosis and the feasibility of apoJ as a therapeutic endpoint. Data Sources: Relevant articles published in English from 1983 to present were selected from PubMed. The terms of "atherosclerosis, apolipoprotein J, clusterin (CLU), oxidative stress, and inflammation" were used for searching. Study Selection: Articles studying the role of apoJ with atherosclerosis and restenosis after injury were reviewed. Articles focusing on the intrinsic determinants of atherosclerosis were selected. The exclusion criteria of articles were that the studies on immunologic vasculitis. Results: ApoJ, involved in numerous physiological process important for lipid transportation and vascular smooth muscle cell differentiation, including apoptotic cell death, cell-cycle regulation, cell adhesion, tissue remodeling, immune system regulation, and oxidative stress, plays a role in the development of clinical atherosclerosis. In the process of relieving atherosclerosis, apoJ can promote cholesterol and phospholipid export from macrophage-foam cells, and exhibit cytoprotective and anti-inflammatory actions by interacting with lots of known inflammatory proteins which may predict the onset of clinical cardiovascular events and may actually play a causal role in mediating atherosclerotic disease such as C-reactive protein, paraoxonase, and leptin. As known as CLU, apoJ has been identified to play central roles in the process of vascular smooth cells migration, adhesion, and proliferation, which can contribute significantly to restenosis after vascular injury. Conclusions: Intense effort and substantial progress have been made to identify the apoJ that relieves atherosclerosis and vascular restenosis after percutaneous coronary intervention. More work is needed to elucidate the exact mechanisms of and the interrelationship between the actions of apoJ and to successfully achieve regression of atherosclerosis by regarding it as a therapeutic endpoint.

Background: In this prospective randomized study, we compared the predicted blood and effect-site C 50 for propofol and remifentanil target-controlled infusion (TCI) and the bispectral index (BIS) values at loss of consciousness (LOC) and response to a standard noxious painful stimulus (LOS) in elderly and young patients, respectively. We hypothesized that the elderly patients will require lower target concentration of both propofol and remifentanil at above two clinical end-points. Methods: There were 80 American Society of Anesthesiologists (ASA) physical status I-II unpremedicated patients enrolled in this study, they were divided into elderly group (age ≥65 years, n = 40) and young group (aged 18-64 years, n = 40). Propofol was initially given to a predicted blood concentration of 1.2 μg/ml and thereafter increased by 0.3 μg/ml every 30 s until Observer′s Assessment of Alertness and Sedation score was 1. The propofol level was kept constant, and remifentanil was given to provide a predict blood concentration of 2.0 ng/ml, and then increased by 0.3 ng/ml every 30 s until loss of response to a tetanic stimulus. BIS (version 3.22, BIS Quattro sensor) was also recorded. Results: In elderly group, the propofol effect-site C 50 at LOC of was 1.5 (1.4-1.6) μg/ml, was significantly lower than that of young group, which was 2.2 (2.1-2.3) μg/ml, the remifentanil effect-site C 50 at LOS was 3.5 (3.3-3.7) ng/ml in elderly patients, was similar with 3.7 (3.6-3.8) ng/ml in young patients. Fifty percent of patients lost consciousness at a BIS value of 57.3 (56.4-58.1), was similar with that of young group, which was 55.2 (54.0-56.3). Conclusion: In elderly patients, the predicted blood and effect-site concentrations of propofol at LOC were lower than that of young patients. At same sedation status, predicted blood and effect-site concentrations of remifentanil required at LOS were similar in elderly and young patients. BIS were not affected by age. Low-propofol/high-opioid may be optional TCI strategy for elderly patients.

This protocol described an efficient aqueous polymer two-phase system followed by alkaline solution washing to prepare Lilium davidii or Oryza sativa plasma membrane with high purity.

Plasma membrane (PM) proteins play crucial roles in diverse biological processes. But their low abundance, alkalinity and hydrophobicity make their isolation a difficult task. This protocol describes an efficient method for PM proteins isolation, digestion and fractionation so that they can be well prepared for mass spectrometry analysis.
