Due to the diversity and limitation of determination methods, published data on the fatty acid (FA) compositions of different milk samples have contributed to inaccurate comparisons. In this study, we developed a high-throughput gas chromatography-mass spectrometry method to determinate milk FA, and the proposed method had satisfactory linearity, sensitivity, accuracy, and precision. We also analyzed the FA compositions of 237 milk samples from Holstein cows, Jersey cows, buffalos, yaks, humans, goats, donkeys, and camels. Holstein, Jersey, goat, and buffalo milks contained high content of even-chain saturated FA, whereas goat milk had higher content of medium- and short-chain FA (MSCFA). Yak and camel milk are potential functional foods due to their high levels of odd- and branched-chain FA and low ratios of n-6 to n-3 polyunsaturated FA (PUFA). Human milk contained lower levels of saturated FA, MSCFA, and conjugated linoleic acid, and higher levels of monounsaturated FA and PUFA. As a special nonruminant milk, donkey milk contained low levels of monounsaturated FA and high levels of PUFA and MSCFA. Based on the FA profiles of 8 types of milk, nonruminant milk was distinct from ruminant milk, whereas camel and yak milk were different from other ruminant milks and considered as potential functional foods for balanced human diet.

OBJECTIVE To investigate the role of omega-3 polyunsaturated fatty acids (Ω-3)–derived proresolving lipid mediators (PRLM) in the resolution of mild airway inflammation in horses. ANIMALS 20 horses with mild airway inflammation. PROCEDURES Horses previously eating hay were fed hay pellets (low Ω-3 content; n = 10) or haylage (high Ω-3 content; 9) for 6 weeks. Dust exposure was measured in the breathing zone with a real-time particulate monitor. Bronchoalveolar lavage (BAL) was performed at baseline, week 3, and week 6. The effect of PRLM on neutrophil apoptosis and efferocytosis was examined in vitro. BAL fluid inflammatory cell proportions, apoptosis of circulating neutrophils, efferocytosis displayed by alveolar macrophages, and plasma lipid concentrations were compared between groups fed low and high amounts of Ω-3 by use of repeated measures of generalized linear models. RESULTS Dust exposure was significantly higher with hay feeding, compared to haylage and pellets, and equivalent between haylage and pellets. BAL fluid neutrophil proportions decreased significantly in horses fed haylage (baseline, 11.8 ± 2.4%; week 6, 2.5 ± 1.1%) but not pellets (baseline, 12.1 ± 2.3%; week 6, 8.5% ± 1.7%). At week 6, horses eating haylage had significantly lower BAL neutrophil proportions than those eating pellets, and a significantly lower concentration of stearic acid than at baseline. PRLM treatments did not affect neutrophil apoptosis or efferocytosis. CLINICAL RELEVANCE Despite similar reduction in dust exposure, horses fed haylage displayed greater resolution of airway inflammation than those fed pellets. This improvement was not associated with increased plasma Ω-3 concentrations. Feeding haylage improves airway inflammation beyond that due to reduced dust exposure, though the mechanism remains unclear.

Low in dietary ω3 polyunsaturated fatty acid (PUFA) consumption has been associated with increased incidence of cancers. Many basic and clinical studies have been conducted over the last several decades. We previously reviewed multi-targeted therapy of cancer by omega-3 fatty acids in 2008, and since hundreds of new clinical trials are being conducted to validate the effectiveness of ω3 PUFA in cancer therapy. Because of the availability of such large amount of clinical trial data, in this update we summarize clinical data, sort out trials that show promising results, and discuss potential mechanism(s) responsible for the clinical outcomes. It appears that ω3 PUFA mainly affects cancer-associated symptoms, namely cachexia, inflammation, neuropathy, post operative complications and quality of life. Mechanisms responsible for these effects are possible regulation of skeletal muscle protein turnover, inflammatory response and neuron cell survive by ω3 PUFA.

Cardiac autonomic neuropathy (CAN) is a severe and common complication of diabetes mellitus. Despite its correlation with the increased risk of cardiovascular mortality and with various symptoms and impairments, the significance of CAN cannot be overestimated. Lifestyle modification, intensive glycemic control might prevent development or progression of CAN. Pathogenetic treatment of CAN includes: balanced diet and physical activity; optimization of glycemic control; treatment of dyslipoproteinemia; correction of metabolic abnormalities in myocardium; prevention and treatment of thrombosis; use of aldose reductase inhibitors; application of dihomo-gamma-linolenic acid (DGLA); use of acetyl-L-carnitine, antioxidants, first of all α-lipoic acid (α-LA), administration of ω-3 and ω-6 polyunsaturated fatty acids (PUFA), vasodilators, fat-soluble vitamin B1, aminoguanidine; substitutive therapy with growth factors, in severe cases — treatment of orthostatic hypotension. The promising methods is the use of agents that increase blood flow in vasa vasorum, including analogs of prostacyclin, thromboxane A2 blockers, and the drugs that contribute to the strengthening and/or normalization in the function of Na+-,K+-ATPase (phosphodiesterase inhibitor), α-LA, DGLA, ω-3 PUFAs, as well as simultaneous administration of α-LA, ω-3 PUFA and DGLA., Кардиальная автономная нейропатия (КАН) является серьезным и частым осложнением сахарного диабета. Довголанцюгові поліненасичені жирні кислоти (ДЛПНЖК) є основними функціональними жирами материнського молока. Переважаючими та функціонально найважливішими ДЛПНЖК є арахідонова (АК) і докозогексаєнова кислота (ДГК). Крім імітації складу грудного молока, існують ще дві важливі причини, щоб додавати ДЛПНЖК у молочні суміші для немовлят. По-перше, немовлята не можуть синтезувати достатню кількість ДЛПНЖК, щоб компенсувати відсутність цих речовин у продуктах харчування. По-друге, тканина мозку, сітківка очей та імунна система потребують великої кількості ДЛПНЖК як будівельного матеріалу та функціональних складових, щоб підтримати інтенсивний ріст і розвиток у ранньому дитинстві. Цей період є критичним для формування та розвитку цих органів, тому оптимальне постачання субстратів є необхідним для їх оптимального функціонування. Ці ідеї в поєднанні з результатами понад 20 рандомізованих контрольованих клінічних випробувань і численних інших досліджень, що доводять позитивний вплив харчових ДЛПНЖК на здоров’я й розвиток немовлят, свідчать про необхідність збагачення продуктів дитячого харчування ДГК і АК. Найбільш вивченими позитивними ефектами такого збагачення є поліпшення зорової функції і когнітивного розвитку дітей раннього віку. Нові дані пов’язують підвищене споживання ДЛПНЖК із корисним впливом на імунні функції і серцево-судинну систему малюків.

The objective of this study was to evaluate the quality of life in patients with ulcerative colitis (UC) of moderate severity and concomitant subclinical atherosclerosis before and after combination treatment with statin and omega-3 polyunsaturated fatty acids (PUFAs). In patients with UC and concomitant atherosclerosis we have detected deterioration in quality of life mostly by the components of physical health (SF-36 questionnaire), which is directly proportional to the degree of atherosclerosis severity (p < 0.05). During combined treatment with statins and omega-3 PUFAs, there was a significant improvement in parameters of not only physical, but also mental health (p < 0.05). Analyzing data by specialized GSRS questionnaires, we have noted improvement in quality of life by three of the five scales — abdominal pain, dyspeptic and diarrheal syndromes (p < 0.05).

Consuming fat results in postprandial lipemia, which is defined as an increase in blood triglyceride (TG) concentration. According to current knowledge, an excessively elevated postprandial TG concentration increases the risk of cardiovascular disease (CVD). It is well known that meal-dependent (e.g., nutrient composition) as well as meal-independent factors (e.g., age) determine the magnitude of the lipemic response. However, there is conflicting evidence concerning the influence of fatty acid (FA) composition on postprandial TG concentration. The FA composition of a meal depends on the fat source used; for example, butter and coconut oil are rich in SFAs, while olive oil and canola oil have a high content of unsaturated FAs. To investigate the influence of meals prepared with fat sources rich in either SFAs or unsaturated FAs on postprandial lipemia, we carried out a systematic literature search in PubMed, Scopus, and the Cochrane Library. Randomized crossover studies were analyzed and the AUC of postprandial TG concentration served as the primary outcome measure. To examine the influence of health status, we differentiated between metabolically healthy individuals and those with CVD risk factors. In total, 23 studies were included. The results show that, in metabolically healthy adults, the FA composition of a meal is not a relevant determinant of postprandial lipemia. However, in individuals with CVD risk factors, SFA-rich meals (>32 g SFA/meal) often elicited a stronger lipemic response than meals rich in unsaturated FAs. The results suggest that adults with hypertriglyceridemia, an elevated BMI (≥30 kg/m(2)), and/or who are older (>40 y) may benefit from replacing SFA sources with unsaturated FAs. These hypotheses need to be verified by further studies in people with CVD risk factors using standardized postprandial protocols. The family of Brassicaceae accounts for one of the earliest cataloged annual oilseed referred to as Mustard. Brassica nigra and Brassica rapa var. toria are two of the commercially important varieties of mustard seed which are endowed with biomedical properties. The present study was designed for the comparative scrutiny of quality and physicochemical parameters of both oilseed mustard varieties. Oil content, near infrared reflectance analysis, moisture content, color, pungency, free fatty acid value, acid value, presence of argemone test and gas chromatography were studied. B. nigra and B. rapa toria showed oil content corresponding to 37.68% and 43.06% respectively. Less moisture content was observed in B. rapa toria (5.33 %) in comparison to B. nigra. Higher color units were investigated in B. nigra in comparison to B. rapa toria. B. rapa toria showed lesser pungency with 0.034 % allyl isothiocynate content. B. nigra illustrated free fatty acid (FFA) content of 0.79% and high acid value (1.58%). While B.rapa toria showed less FFA and acid value. Argemone oil contamination was not seen in oil of both varieties. B. nigra showed 6.45%, 69.25%, 24.29% of saturated, monounsaturated and polyunsaturated fatty acids respectively. B. rapa toria demonstrated valuable gas chromatographic analysis with lesser saturated fatty acid and higher monounsaturated fatty acid. In comparison to B. nigra, higher oil content, lesser moisture, low FFA value, less acid value and favorable fatty acid composition analysis has been shown by B. rapa toria there upon, indicating superior quality of B. rapa toria oilseed.

Fatty acids are an essential class of raw materials for the chemical industry. Fatty acids find widespread application in food, cosmetics, soaps, surfactants, pharmaceutical, and nutraceutical industry. A specific type of derivative- fatty acid methyl esters are used as a bio based alternative to fossil fuels. A lot of research has undergone to develop novel efficient processes to purify fatty acids from reaction mixture or a mixture of fatty acids. Membrane performance is usually dependent on size of the molecule to be separated in the membrane and pore size which decides the rate of permeation of molecule through the membrane. This paper discusses about the performance of two polymer nanofiltration membranes developed recently in purifying a mixture of fatty acids and exhibits critical area being the determinant factor for molecules to permeate through the membrane. Epoxy nanofiltration membranes have separated EPA-EE and DHA-EE from each other based on molecular weight and critical area, whereas these membranes separated, small organic molecules in the molecular weight range 100–300 gmol−1 based on critical area. Epoxy nanofiltration membranes have also successfully separated saturated fatty acids from each other with selectivity of 10:1. Polydicyclopentadiene based nanofiltration membrane have shown to separate saturated fatty acids from mono and polyunsaturated fatty acids with good selectivity based on critical area. This paper emphasizes critical area is a better representation of molecular size as compared to molecular weight.

The growth and reproductive potential of composting earthworm is influenced by the chemical composition of initial feed mixture. Thus the GC-MS technique is used to determine the biochemical foot prints of the vermibed substrate before composting process. The present study conducted to analyze the chemical composition of precomposted animal dung material of Elephus maximus and Bos taurus by GC-MS analysis. The dung materials were precomposted up to 14 days and used for the analysis. The GC-MS spectrogram of Elephus maximus and Bos taurus showed 20 and 15 different compounds respectively. Predominant levels of esters were observed in elephant dung, which were dominated by the esterified polyunsaturated fatty acids with maximum peak area for elaidinsaeure methyl ester (26.6%). In cow dung the maximum peak area was obtained for 1-Hydroxy-1-methyl) −2-Methyl tetrahydro −2 furanyl] − 4, 4, 14-Trimethylandrost- 9 (11) – ene −3, 6, 16 (17%) and alcohol were the dominant functional group followed by carboxylic acids and esters. Thus the results indicated that the presence of higher percentage of esterified poly unsaturated fatty acids in elephant dung reflects its higher quality suitable for vermicomposting, since the dietary unsaturated fatty acids have the positive impacts on the reproduction of earthworm by enhancing the functionality in sperm production.

Background Maternal nutrition is associated with epigenetic and cardiometabolic risk factors in offspring. Research in humans has primarily focused on assessing the impact of individual nutrients. Objective We sought to assess the collective impact of maternal dietary monounsaturated (MUFA), polyunsaturated (PUFA), and saturated fat (SFA) on epigenetic aging and cardiometabolic risk markers in healthy newborn infants using a geometric framework approach. Design Body fatness (n = 162), aortic intima-media thickness (n = 131), heart rate variability (n = 118), and epigenetic age acceleration (n = 124) were assessed in newborn infants. Maternal dietary intake was cross-sectionally assessed in the immediate postpartum period via a validated 80-item self-administered food-frequency questionnaire. Generalized additive models were used to explore interactive associations of nutrient intake, with results visualized as response surfaces. Results After adjustment for total energy intake, maternal age, gestational age, and sex there was a 3-way interactive association of MUFA, PUFA, and SFA (P = 0.001) with newborn epigenetic aging. This suggests that the nature of each fat class association depends upon one another. Response surfaces revealed MUFA was positively associated with newborn epigenetic age acceleration only at proportionately lower intakes of SFA or PUFA. We also demonstrate a potential beneficial association of omega-3 PUFA with newborn epigenetic age acceleration (P = 0.008). There was no significant association of fat class with newborn aortic intima-media thickness, heart rate variability, or body fatness. Conclusions In this study, we demonstrate an association between maternal dietary fat class composition and epigenetic aging in newborns. Future research should consider other characteristics such as the source of maternal dietary fatty acids.

Fatty acid desaturase (FADS) generates double bond at a certain position of the corresponding polyunsaturated fatty acids (PUFAs) with high selectivity, the enzyme activity and PUFAs products of which are essential to biological systems and are associated with a variety of physiological diseases. Little is known about the structure of FADSs and their amino acid residues related to catalytic activities. Identifying key residues of Micromonas pusilla delta 6 desaturase (MpFADS6) provides a point of departure for a better understanding of desaturation. In this study, conserved amino acids were anchored through gene consensus analysis, thereby generating corresponding variants by site-directed mutagenesis. To achieve stable and high-efficiency expression of MpFADS6 and its variants in Saccharomyces cerevisiae, the key points of induced expression were optimized. The contribution of conserved residues to the function of enzyme was determined by analyzing enzyme activity of the variants. Molecular modeling indicated that these residues are essential to catalytic activities, or substrate binding. Mutants MpFADS6[Q409R] and MpFADS6[M242P] abolished desaturation, while MpFADS6[F419V] and MpFADS6[A374Q] significantly reduced catalytic activities. Given that certain residues have been identified to have a significant impact on MpFADS6 activities, it is put forward that histidine-conserved region III of FADS6 is related to electronic transfer during desaturation, while histidine-conserved regions I and II are related to desaturation. These findings provide new insights and methods to determine the structure, mechanism and directed transformation of membrane-bound desaturases.

Background There is increasing evidence that dysregulation of polyunsaturated fatty acids (FAs) mediated membrane function plays a role in the pathophysiology of schizophrenia. Even though preclinical findings have supported the anti-inflammatory properties of omega-3 FAs on brain health, their biological roles as anti-inflammatory agents and their therapeutic role on clinical symptoms of psychosis risk are not well understood. In the current study, we investigated the relationship of erythrocyte omega-3 FAs with plasma immune markers in a clinical high risk for psychosis (CHR) sample. In addition, a mediation analysis was performed to examine whether previously reported associations between omega-3 FAs and clinical outcomes were mediated via plasma immune markers. Clinical outcomes for CHR participants in the NEURAPRO clinical trial were measured using the Brief Psychiatric Rating Scale (BPRS), Schedule for the Scale of Assessment of Negative Symptoms (SANS) and Social and Occupational Functioning Assessment Scale (SOFAS) scales. The erythrocyte omega-3 index [eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA)] and plasma concentrations of inflammatory markers were quantified at baseline (n = 268) and 6 month follow-up (n = 146) by gas chromatography and multiplex immunoassay, respectively. In linear regression models, the baseline plasma concentrations of Interleukin (IL)-15, Intercellular adhesion molecule (ICAM)-1 and Vascular cell adhesion molecule (VCAM)-1 were negatively associated with baseline omega-3 index. In addition, 6-month change in IL-12p40 and TNF-α showed a negative association with change in omega-3 index. In longitudinal analyses, the baseline and 6 month change in omega-3 index was negatively associated with VCAM-1 and TNF-α respectively at follow-up. Mediation analyses provided little evidence for mediating effects of plasma immune markers on the relationship between omega-3 FAs and clinical outcomes (psychotic symptoms and functioning) in CHR participants. Our results indicate a predominantly anti-inflammatory relationship of omega-3 FAs on plasma inflammatory status in CHR individuals, but this did not appear to convey clinical benefits at 6 month and 12 month follow-up. Both immune and non-immune biological effects of omega-3 FAs would be resourceful in understanding the clinical benefits of omega-3 FAs in CHR papulation.

Background Population growth, climate change, and food shortage have led to increased demand for alternate sources of lipids to meet our food and energy needs. Oleaginous microorganisms such as yeasts, bacteria, microalgae, fungi, and thraustochytrids are good resources for producing lipids. Scope and approach Many factors affect the amount and composition of lipid accumulation in microorganisms, such as genetic makeup and environmental conditions during growth, such as carbon and nitrogen sources, pH, temperature, and exposure to natural light in growing media. Food biotechnology advances such as improvements in fermentation technology, genetic and metabolic engineering of oleaginous microorganisms, and the use of low-cost carbon sources, such as food and agricultural waste materials, make lipid production using microbial sources more feasible. Key findings and conclusions Microbial lipid production has some unique benefits for the production of specialty fats and oils, including cocoa butter equivalents, polyunsaturated fatty acid oils and biosurfactants. Moreover, microbial oil production has advantages over plant and marine oils, including lower cultivation costs and the ability to engineer oil structure and thus functionality using genetic modification. Microbial oils production allows for the production of more sustainable and safer oils, without pesticides, herbicides or heavy metals, year-round with a small footprint and without the need for land.

Nonalcoholic fatty liver disease (NAFLD) is a common but complex chronic liver disease, driven by environmental and genetic factors. We assessed metabolic and dietary risk factor associations with NAFLD liver mortality using the Global Burden of Disease (GBD) 2017 data. NAFLD liver deaths were calculated (per 100,000) as age-standardized rates (ASRs) from 195 countries and territories (21 GBD regions; 7 GBD superregions). Dietary risks included low intake of fruits, vegetables, legumes, whole grains, nuts/seeds, milk, fiber, calcium, seafood omega-3 fatty acids, and polyunsaturated fatty acids, and high intake of red meat, processed meat, sugar-sweetened beverages, trans fatty acids, and sodium. Metabolic risks included high low-density lipoprotein cholesterol, systolic blood pressure (BP), fasting glucose (FG), body mass index (BMI), as well as low bone mineral density and impaired kidney function (IKF). Socio-demographic index (SDI)-adjusted partial Spearman correlation coefficients and multivariable generalized linear regression models/bidirectional stepwise selection (significance level for entry, 0.2; for stay, 0.05) determined the associations. The ASR for NAFLD liver deaths was 2.3 per 100,000 (2017) and correlated with dietary risk factors (0.131, -0.010-0.267) and metabolic risk factors (SDI-adjusted = 0.225, 95% CI 0.086-0.354). High intake of sugar-sweetened beverages and red meat (0.358, 0.229-0.475; 0.162, 0.022-0.296), and low intake of nuts/seed and milk (0.154, 0.014-0.289; 0.145, 0.004-0.280) was significant for NAFLD liver deaths. Other risk factors for liver death included IKF (0.402, 0.276-0.514), increased BMI (0.353, 0.223-0.407), FG (0.248, 0.111-0.376), and BP (0.163, 0.022-0.297). High intake of trans fatty acids (2.84% increase [1.65%-4.03%]) was the largest associated risk of NAFLD liver deaths. In addition to metabolic risks, dietary risks independently drive the global burden of NAFLD-related liver mortality. Conclusion: These data provide additional support for policies to improve dietary environment for NAFLD burden reduction.

The incidence of infertility is increasing world wild. Among the diet factors, are the quality and quantity of the fat that contributes to spermatogenesis. Canola oil is known to contain oleic acid which is an omega-9 monounsaturated fatty acid, linoleic acid which is an essential omega-6 polyunsaturated fatty acid and alpha-linolenic acid, a plant-based omega-3. Canola oil has the lowest saturated fatty acid content among cooking oil. This study has been designed to investigate the possible effect of Canola oil, the recently used cooking oil, on the testicular structure and ultra-structure. Twelve adult male albino rats (200-250g) were obtained from the animal house in Faculty of Pharmacy, Mansoura University. The animals were housed two in a cage at a constant temperature of 18°C and humidity 45%. Rats were divided into control and treated; treated rats received a diet contains 7ml/100gm canola oil for 39 days. Six rats in each group were weighed and sacrificed at the end of the experiment; testis was dissected and weighed. The specimens were used for paraffin sections and electron microscope examination. The Canola oil-treated group showed irregular seminiferous tubules, germ cells with vacuolated cytoplasm and deeply stained nuclei compared with the control group. The treated testis ultrastructure showed degenerated Sertoli cell with an irregular nucleus, degenerated spermatocytes with the cytoplasm showing phagosomes, lysosomes and increased autophagic vacuoles. Interstitial cells appeared degenerated with vacuolated cytoplasm an. Canola oil is low in saturated fat and high in monounsaturated fat content, making it an ideal healthy cooking oil. This study revealed that Canola oil usage resulted in distorted seminiferous tubules, degenerated germ cells and ultrastructural changes in the treated testis. Canola oil should be used with caution, especially in males, to avoid its hazardous effect on the testis.

The study was conducted to investigate the effect of feeding algae biomass rich in polyunsaturated fatty acids at 0%, 1.5%, 3.0% and 4.5%, respectively, using Panicum maximum as basal feed, on rumen fermentative properties of pregnant West African Dwarf (WAD) does. A total of twenty (20) pregnant WAD does were grouped into four treatments containing five (5) animals per treatment and randomly allotted to the experimental diets. Rumen fluid were collected prior to mating and at the last trimester of pregnancy for determination of rumen fermentative properties. Data obtained were subjected to one way Analysis of Variance in a Completely Randomized Design. Inclusion of algae biomass up to 4.5% had no significant (p>0.05) effect on bacteria population in the rumen of the experimental does. The inclusion levels of algae biomass gave rise to the predominance of Eischeria coli (a gram negative bacterium) in the rumen of pregnant does fed the algae biomass diet. Results obtained showed a significant (p0.05) influenced by the inclusion of algae biomass.

Purpose There is an ongoing debate whether vegetarian (VG) and especially vegan (VN) diets are nutritionally adequate in early childhood. Hence, the Vegetarian and Vegan Children Study (VeChi Diet Study) aimed to assess the food and nutrient intake of VG and VN infants. Methods The study examined the diets of 1–3-year-old VG, VN, and omnivorous (OM) children (n = 430). Dietary intake was assessed via a 3-day weighed dietary record and compared between groups using ANCOVA. Lifestyle data were collected using a questionnaire. Here, the results of micronutrient and fatty acid intakes are presented. Results Most nutrient intakes (with and without supplements) differed significantly between VN children and the two other groups, with a more favourable overall micronutrient intake in VN, followed by VG children, [e.g., the highest intake of vitamin E (8.3 mg/d vs. VG 7.4 mg/d and OM 5.1 mg/d), vitamin B1 (569 µg/d vs. VG 513 µg/d and OM 481 µg/d), folate (143 µg/d vs. VG 116 µg/d and OM 108 µg/d), magnesium (241 mg/d vs. VG 188 mg/d and OM 164 mg/d), and iron (8.9 mg/d vs. VG 7.3 mg/d and OM 6.0 mg/d)] as well as fat quality [highest intake of polyunsaturated fatty acids (8.7 E% vs. VG 6.9 E% and OM 4.5 E%) and lowest intake of saturated fatty acids (9.1 E% vs. VG 11.9 E% and OM 14.0 E%)]. In contrast, OM children had the highest intake of vitamin B2 (639 µg/d vs. VG 461 µg/d and VN 429 µg/d), calcium (445 mg/d vs. VG 399 mg/d and VN 320 mg/d), iodine (47 µg/d vs. VG 33 µg/d and VN 31 µg/d), and DHA (35.4 mg/d vs. VG 16.6 mg/d and VN 18.4 mg/d). Without supplementation, OM children had the highest average vitamin B12 intake (1.5 µg/d vs. VG 0.6 µg/d and VN 0.2 µg/d), whereas VN children had the highest average vitamin B12 intake with supplementation (73.8 µg/d vs. VG 1.3 µg/d and OM 1.7 µg/d). Without supplementation, none of the groups’ median intakes met the harmonised Average Requirement (h-AR) for vitamin D and iodine. Moreover, VG and VN children did not achieve h-ARs for vitamin B2, vitamin B12, and iron—if a low absorption of iron is anticipated; VN children also did not do so for calcium. Conclusion In early childhood, VN and VG diets can provide most micronutrients in desirable amounts and a preferable fat quality compared to an OM diet. Special focus should be paid to (potentially) critical nutrients, particularly vitamin D, iodine, and DHA for all children regardless of diet, as well as vitamin B2, vitamin B12, calcium, and iron for VG and VN children. The efficacy of n-3 polyunsaturated fatty acids (PUFAs) in improving outcomes in a renal ischemia-reperfusion injury (IRI) model has previously been reported. However, the underlying mechanisms remain poorly understood and few reports demonstrate how dietary n-3 PUFAs influence the composition of membrane phospholipids in the kidney. Additionally, it has not been elucidated whether perilla oil (PO), which is mainly composed of the n-3 alpha-linolenic acid, mitigates renal IRI. In this study, we investigated the effect of dietary n-3 PUFAs (PO), compared with an n-6 PUFA-rich soybean oil (SO) diet, on IRI-induced renal insufficiency in a rat model. Levels of membrane phospholipids containing n-3 PUFAs were higher in the kidney of PO-rich diet-fed rats than the SO-rich diet-fed rats. Levels of blood urea nitrogen and serum creatinine were significantly higher in the ischemia-reperfusion group than the sham group under both dietary conditions. However, no significant differences were observed in blood urea nitrogen, serum creatinine, or histological damage between PO-rich diet-fed rats and SO-rich diet-fed rats. In the kidney of PO-rich diet-fed rats, levels of arachidonic acid and arachidonic acid-derived pro-inflammatory lipid mediators were lower than SO-rich diet-fed rats. Eicosapentaenoic acid and eicosapentaenoic acid-derived lipid mediators were significantly higher in the kidney of PO-rich than SO-rich diet-fed rats. These results suggest that dietary n-3 PUFAs alter the fatty acid composition of membrane phospholipids and lipid mediators in the kidney; however, this does not attenuate renal insufficiency or histological damage in a renal IRI model.

Dexamethasone (DEX) is a synthetic glucocorticoid with anti-inflammatory properties. We evaluated a potentially protective dexamethasone influence on hepatocellular lipid metabolism and fatty acid (FA) transporters expression. The HepG2 cells were incubated with palmitic acid (PA) and/or dexamethasone in two different time expositions (16 h and 40 h). Intracellular and extracellular lipid and sphingolipid concentrations were estimated by the gas-liquid chromatography and high-performance liquid chromatography, respectively. The protein expression involved in FA uptake and lipid metabolism was determined by immunoblotting. The treatment of HepG2 with dexamethasone and palmitate enhanced lipid transport to the cell via increased especially FABPpm expression and resulted in the increased triacylglycerol (TAG), diacylglycerol (DAG) and ceramide deposition. Dexamethasone with palmitate treatment altered FA composition resulting in the elevated n-3 polyunsaturated fatty acid (PUFA) activity in DAG and TAG and the diminished n-6 PUFA activity in DAG after prolonged exposure. We may speculate that although protective lipid secretion into media and decrease in inflammatory FA precursors dexamethasone treatment exacerbated lipotoxicity in HepG2 cells.

Summary Background & aims Sarcopenia is characterized by the progressive loss of skeletal muscle mass and function, which reduces mobility and quality of life. Risk factors for sarcopenia include advanced age, physical inactivity, obesity, and chronic diseases such as cancer or rheumatoid arthritis. Omega-3 long chain polyunsaturated fatty acids (LC PUFAs) might be associated with a reduction in risk of sarcopenia due to their anti-inflammatory effects. Methods We conducted a systematic review and meta-analysis to quantify the effects of omega-3 LC PUFAs on muscle mass, volume and function parameters. The National Library of Medicine's MEDLINE/PubMed database was searched on 9th October 2020 for randomized controlled trials that used omega-3 LC PUFAs as an intervention with muscle-related endpoints. A snowballing search to identify additional studies was completed on 23rd April 2021. The meta-analysis was conducted using meta-essentials worksheet 3. Bias was assessed using the Jadad scale. Results 123 studies were identified with the systematic searches. Most studies were performed in disease populations, such as cancer or chronic obstructive pulmonary disease (COPD), or in healthy individuals after a fatiguing exercise bout. The endpoints lean body mass, skeletal muscle mass, mid-arm muscle circumference, handgrip strength, quadriceps maximal voluntary capacity (MVC), and 1-repetition maximum chest press were selected for meta-analysis based on the number of available studies; thus 66 studies were included in the quantitative synthesis. Using a random effects model and 2-tailed p-value, there was a significant relationship in favor of omega-3 LC PUFA supplementation for lean body mass (effect size 0.27, 95%CI 0.04 to 0.51), skeletal muscle mass (effect size 0.31, 95%CI 0.01 to 0.60) and quadriceps MVC (effect size 0.47, 95%CI 0.02 to 0.93). Conclusion The results indicate that there is a positive effect of omega-3 LC PUFA supplementation on overall body muscle mass and strength. Small study size and heterogeneity limit the applicability of these findings for sarcopenia prevention. Larger trials in populations at risk of sarcopenia would strengthen the evidence base.

Background Monoglycerides (MAG) and Diglycerides (DAG) belong to the category of naturally-occurring glycerolipids. They have wide applications in the food, pharmaceutical, and cosmetic industries, with commercial demand supplied by the consolidated industrial catalytic route of vegetable oil glycerolysis. Despite the economic competitiveness of producing these types of emulsifiers from vegetable oils, the increasing demand for products with high nutrition value makes some kinds of microalgae oil potential feedstock of high-quality fatty acids to serve this growing market. Scope and approach An alternative to the use of vegetable oils is the production of triacylglycerols from microalgae. Usually, microalgal oils have a high content of Polyunsaturated Fatty Acids (PUFA) and the cultivation of microalgae may present fewer environmental impacts, considering reduced use of arable land, efficient CO2 biofixation, and high productivity. Microalgae lipids are mostly studied for biodiesel production, but this work shows the potential to explore more valuable applications due to their composition, discussing the possibility of producing MAGs and DAGs from microalgae lipids. Key findings and conclusions While biodiesel B99-B100 costs USD3.56/gallon according to the U.S. Energy Dept. (April 2021), a food emulsifier (soybean lecithin) is sold for USD147/kg (Alfa Aesar, August 2021). Hence, it is imperative to consider high-value bioproducts from an economic point of view. Microalgal oil can be rich in ω-3 and ω-6 fatty acids, being a promising source of MAGs and DAGs with higher nutritional value. Glycerolysis studies of this feedstock are restricted to the enzymatic route, but different alternatives are shown in this work.

Background & aims: Circulating microvesicles (cMV) are both effectors and biomarkers of cardiovascular disease (CVD), and the effects of omega 3 polyunsaturated fatty acids (n3 PUFA) in MV shedding are not yet well known. Therefore, we aimed to investigate the effects of long-term n3 PUFA supplementation on cMV release from cells of the vascular compartment in elderly subjects at very high risk of CVD. Methods: We included 156 elderly patients 2-8 weeks after suffering an acute myocardial infarction from the OMEMI cohort. Subjects were randomly allocated to receive 930 mg EPA + 660 mg DHA (n3 PUFA intervention) or corn oil (56% linoleic acid, 32% oleic acid, 10% palmitic acid) used as placebo daily for two years. At inclusion and after one-year follow-up, prothrombotic [annexin V (AV)] cMV derived from blood and vascular cells were phenotyped by flow cytometry. Results: No differences were observed in the levels of cMV between the randomized groups at inclusion in the study. After one-year follow-up, total AV, platelet-derived CD61/AV, and endothelial-derived CD31/AV and CD31/CD42b/AV cMV increased significantly in both groups. In the n3 PUFA supplemented group, platelet-derived CD62P/AV, CD42b/AV and CD31/CD42b/AV; leukocyte-derived CD62L/AV, CD45/AV, and CD11b/AV, as well as endothelial derived CD146/AV, CD62E/AV, and CD309/AV cMV also increased significantly. No significant differences were however, observed in the changes of cMV levels between groups. Conclusion: In elderly Norwegians who have suffered a recent acute myocardial infarction and treated as per guidelines, long-term supplementation with 1.8 g/day n3 PUFA does not modulate prothrombotic MV release from blood and vascular cells. Summary Background & aims Epidemiologic studies are inconsistent regarding the association of dietary omega-3 polyunsaturated fatty acids (PUFA) and/or fish intake with risk of age-related macular degeneration (AMD) incidence and progression. The objective was to determine these associations by conducting a meta-analysis of available studies. Methods Three electronic databases were searched for studies that quantified dietary omega-3 PUFA and/or fish intake from inception to December 2020 without language restriction. Three investigators independently assessed for inclusion and extracted data. Study-specific risk estimates were combined using random-effects model. Potential dose–response associations were explored with the use of generalized least-squares trend estimation. Results 21 studies were included in the meta-analysis. Higher dietary intakes of omega-3 PUFA was significantly associated with 14% (relative risk [RR]: 0.86, 95% confidence interval [CI]: 0.77, 0.96) and 29% (RR: 0.71, 95% CI: 0.55, 0.91) lower risk of early and late AMD, respectively. The dose–response analysis showed a 6% and 22% decrease in the risk of early and late AMD for each additional 1 g/d omega-3 PUFA intake. For individual omega-3 PUFA, the intake of eicosapentaenoic acid and docosahexaenoic acid was inversely associated with lower AMD risk, whereas no association was found for the alpha-linolenic acid. Consistent inverse associations were also found between fish intake and AMD. The pooled RRs comparing extreme categories of fish intake were 0.79 (95% CI: 0.70, 0.90) and 0.71 (95% CI: 0.60, 0.85) for early and late AMD risk, respectively. Every 15 g/d of fish consumption was associated with 13% and 14% lower early and late AMD. In addition, fish intake was associated with a significantly reduced risk of AMD progression (RR: 0.73, 95% CI: 0.53, 1.00). Conclusions A high intake of dietary omega-3 PUFA or fish was associated with a reduced risk of developing of AMD, which further supports that consumption of omega-3 PUFA-rich foods may be a new avenue nutritional approach to preventing AMD.

Summary Background Although studies suggest that concentrations of omega-3 and omega-6 fatty acids are lower in individuals with schizophrenia, evidence for beneficial effects of fatty acid supplementation is scarce. Therefore, in this study, we aimed to determine whether omega-3 and omega-6 fatty acid concentrations are causally related to schizophrenia. Methods We did a two-sample Mendelian randomisation study, using deidentified summary-level data that were publicly available. Exposure-outcome relationships were evaluated using the inverse variance weighted two-sample Mendelian randomisation method using results from genome-wide association studies (GWASs) of fatty acid concentrations and schizophrenia. GWAS results were available for European (fatty acids) and European and Asian (schizophrenia) ancestry samples. Overall age and gender information were not calculable from the summary-level GWAS results. Weighted median, weighted mode, and Mendelian randomisation Egger regression methods were used as sensitivity analyses. To address underlying mechanisms, further analyses were done using single instruments within the FADS gene cluster and ELOVL2 gene locus. FADS gene cluster and ELOVL2 gene causal effects on schizophrenia were calculated by dividing the single nucleotide polymorphism (SNP)-schizophrenia effect estimate by the SNP-fatty acid effect estimate with standard errors derived using the first term from a delta method expansion for the ratio estimate. Multivariable Mendelian randomisation was used to estimate direct effects of omega-3 fatty acids on schizophrenia, independent of omega-6 fatty acids, lipoproteins (ie, HDL and LDL), and triglycerides. Findings Mendelian randomisation analyses indicated that long-chain omega-3 and long-chain omega-6 fatty acid concentrations were associated with a lower risk of schizophrenia (eg, inverse variance weighted odds ratio [OR] 0·83 [95% CI 0·75–0·92] for docosahexaenoic acid). By contrast, there was weak evidence that short-chain omega-3 and short-chain omega-6 fatty acids were associated with an increased risk of schizophrenia (eg, inverse variance weighted OR 1·07 [95% CI 0·98–1·18] for α-linolenic acid). Effects were consistent across the sensitivity analyses and the FADS single-SNP analyses, suggesting that long-chain omega-3 and long-chain omega-6 fatty acid concentrations were associated with lower risk of schizophrenia (eg, OR 0·74 [95% CI 0·58–0·96] for docosahexaenoic acid) whereas short-chain omega-3 and short-chain omega-6 fatty acid concentrations were associated with an increased risk of schizophrenia (eg, OR 1·08 [95% CI 1·02–1·15] for α-linolenic acid). By contrast, estimates from the ELOVL2 single-SNP analyses were more imprecise and compatible with both risk-increasing and protective effects for each of the fatty acid measures. Multivariable Mendelian randomisation indicated that the protective effect of docosahexaenoic acid on schizophrenia persisted after conditioning on other lipids, although evidence was slightly weaker (multivariable inverse variance weighted OR 0·84 [95% CI 0·71–1·01]). Interpretation Our results are compatible with the protective effects of long-chain omega-3 and long-chain omega-6 fatty acids on schizophrenia, suggesting that people with schizophrenia might have difficulty converting short-chain polyunsaturated fatty acids to long-chain polyunsaturated fatty acids. Further studies are required to determine whether long-chain polyunsaturated fatty acid supplementation or diet enrichment might help prevent onset of schizophrenia. In response to infection or tissue damage, resident peritoneal macrophages (rpMACs) produce inflammatory lipid mediators from the polyunsaturated fatty acid (PUFA), arachidonic acid (AA). Long-chain acyl-CoA synthetase 4 (ACSL4) catalyzes the covalent addition of a CoA moiety to fatty acids, with a strong preference for AA and other PUFAs containing three or more double bonds. PUFA-CoA can be incorporated into phospholipids, which is the source of PUFA for lipid mediator synthesis. In this study, we demonstrated that deficiency of Acsl4 in mouse rpMACs resulted in a significant reduction of AA incorporated into all phospholipid classes and a reciprocal increase in incorporation of oleic acid and linoleic acid. After stimulation with opsonized zymosan (opZym), a diverse array of AA-derived lipid mediators, including leukotrienes, PGs, hydroxyeicosatetraenoic acids, and lipoxins, were produced and were significantly reduced in Acsl4-deficient rpMACs. The Acsl4-deficient rpMACs stimulated with opZym also demonstrated an acute reduction in mRNA expression of the inflammatory cytokines, Il6, Ccl2, Nos2, and Ccl5. When Acsl4-deficient rpMACs were incubated in vitro with the TLR4 agonist, LPS, the levels of leukotriene B4 and PGE2 were also significantly decreased. In LPS-induced peritonitis, mice with myeloid-specific Acsl4 deficiency had a significant reduction in leukotriene B4 and PGE2 levels in peritoneal exudates, which was coupled with reduced infiltration of neutrophils in the peritoneal cavity as compared with wild-type mice. Our data demonstrate that chronic deficiency of Acsl4 in rpMACs reduces the incorporation of AA into phospholipids, which reduces lipid mediator synthesis and inflammation.

Introduction n-3 long-chain polyunsaturated fatty acids (LCPUFA) have anti-inflammatory effects and may reduce colorectal cancer risk. The purpose of this study was to evaluate the effects of n-3 LCPUFA supplementation on markers of rectal cell proliferation and apoptosis and examine how genetic variation in desaturase enzymes might modify this effect. Methods We conducted a randomized, double-blind, control six-month trial of 2.5 grams of n-3 LCPUFA per day compared to olive oil. Study participants had a history of colorectal adenomas. Randomization was stratified based on the gene variant rs174535 in the fatty acid desaturase 1 enzyme (FADS1). Our primary outcome was change in markers of rectal epithelial proliferation and apoptosis. Results A total of 141 subjects were randomized. We found no difference in apoptosis markers between participants randomized to n-3 LCPUFA compared to olive oil (P = 0.41). N-3 LCPUFA supplementation increased cell proliferation in the lower colonic crypt compared to olive oil (P = 0.03) however baseline indexes of proliferation were different between the groups at randomization. We found no evidence that genotype modified the effect.
