The genus Sidera (Hymenochaetales, Basidiomycota) comprises white-rot, mono- or dimitic fungi with poroid or hydnoid hymenophore. It has a worldwide distribution albeit with fewer species present in the Southern Hemisphere. Although recent studies revealed the existence of several new Sidera species, there are still taxonomic inconsistencies and obscure phylogenetic relationships amongst certain taxa of the genus. In this work, a large number of Sidera collections were used to obtain an updated phylogeny, based on ITS and 28S rDNA sequences by including new material from Mediterranean Europe. The monophyly of the genus was strongly supported and all species with poroid hymenophore formed a highly-supported lineage with two major subclades. In total, 23 putative species were recognised. Amongst those, five are considered to possibly represent entities new to science, but further work is required since they are represented by single specimens or environmental sequences. Examined collections originally named S. lenis from southern Europe were grouped within S. vulgaris. Similarly, several collections under various names were hereby identified as S. vulgaris, including those of the recently described species S. tibetica. Furthermore, a critical discussion (based on morphoanatomical findings) is made on the key features that could be used to distinguish S. lenis from S. vulgaris. [ABSTRACT FROM AUTHOR]

Phenol-formaldehyde (PF) resins can be impregnated and cured in situ to improve the woods dimensional stability and decay resistance. In search of renewable alternatives, the substitution of phenol by lignin cleavage products (LCP) has been discussed. However, the different chemical nature may affect the performance of the resin against fungal decay, formaldehyde emission, and equilibrium moisture content. In this study, 30 % (w/w) of the phenol in PF resins were substituted by LCP obtained from microwave-assisted pyrolysis. Scots pine sapwood was modified with the resin. The decay resistance against Rhodonia placenta, Gloeophyllum trabeum, and Trametes versicolor was determined. Additionally, effects of specimen organisation within the Petri dish, different substrates, length of leaching, and type of inoculum were studied. Further, the materials water vapor sorption properties and formaldehyde emission were determined. All modifications effectively reduced fungal decay. With 10 % weight percent gain (WPG), initial decay was detected, while 20 % WPG and 30 % WPG provided efficient protection. The substitution of phenol increases the formaldehyde emission. While further reduction in formaldehyde in the resin admixture or formaldehyde scavengers may be required, the method described herein can be used to partly replace fossil-based phenol, while maintaining good fungal resistance. Basidiomycetes found in forest ecosystems have been used traditionally as food and medicine for centuries, and various studies have validated many of their health benefits. In recent years, the focus in medicinal basidiomycetes has increased, and several notable trends in their application and research have been observed. In light of the emerging literatures, this review attempts to summarise and discuss evidences from the past decade on the importance of exploring basidiomycetes for their pharmacological properties. It also aims to serve as a critical reference for research works focused on the potentials of wild basidiomycetes in Malaysia, as well as from worldwide, to produce a diverse range of bioactive metabolites. Special attention is given to the low-molecular weight bioactive compounds produced in cultured mycelium and culture broth of these macrofungi. It further highlights the unique attributes of wild bioluminescent basidiomycetes species which hold great potential to be harnessed for mycopharmaceuticals with commercial applications in healthcare industries. China has a huge area of diverse landscapes and is believed to conceive incredibly high fungal diversity. To systematically and promptly report Chinese fungal species, we initiate the series of Catalogue of fungi in China here. In the first paper of this series, we focus on plant-inhabiting fungi. A total of 33 new taxa are described all over China. These taxa include two new genera, viz. Cremeoefibula and Nothopucciniastrum, 18 new species, viz. Annulohypoxylon lancangensis, Ascotaiwania coffeae, Clitocella neofallax, Coleopuccinia yunnanensis, Cremeoefibula hengduanensis, Crepidotus furcaticystidiosus, C. tomentellus, Diachea macroverrucosa, Helicogloea hangzhouensis, Hyalopsora caprearum, Nemania polymorpha, Phanerochaetella austrosinensis, Physalacria tianzhongshanensis, Setophaeosphaeria panlongensis, Subulicystidium boreale, Trechispora subaraneosa, Vikalpa dujuanhuensis, and Xylaria pteridicola, and 13 new combinations, viz. Nothopucciniastrum actinidiae, N. boehmeriae, N. coriariae, N. corni, N. coryli, N. fagi, N. kusanoi, N. hikosanense, N. hydrangeae-petiolaris, N. miyabeanum, N. styracinum, N. tiliae, and N. yoshinagae. The morphological characteristics and phylogenetic evidence are used to support the establishment of these new taxa and the accuracy of their taxonomic placements. Heterotrophic soil microorganisms are responsible for ~50% of the carbon dioxide released by respiration from the terrestrial biosphere each year. The respiratory response of soil microbial communities to warming, and the control mechanisms, remains uncertain, yet is critical to understanding the future land carbon (C)‐climate feedback. Individuals of nine species of fungi decomposing wood were exposed to 90 days of cooling to evaluate the medium‐term effect of temperature on respiration. Overall, the effect of temperature on respiration increased in the medium term, with no evidence of compensation. However, the increasing effect of temperature on respiration was lost after correcting for changes in biomass. These results indicate that C loss through respiration of wood‐decomposing fungi will increase beyond the direct effects of temperature on respiration, potentially promoting greater C losses from terrestrial ecosystems and a positive feedback to climate change. This paper presents an annotated checklist of the Basidiomycota taxa (including lichenicolous fungi and the subdivision Pucciniomycotina) from the Azores archipelago and reviews the published records to account for their taxonomic status. The number of Basidiomycota species recorded in the Azores has increased considerably during the 20th century and now stands at 544 species. This study provides distribution data and includes changes in the nomenclature of the listed taxa. Sampling campaigns contributed to 116 new records of Basidiomycota for the Azores archipelago. In addition, there were new records for eight islands: 162 species found for the first time on São Miguel Island, 55 species new to Santa Maria Island, 33 species new to Flores Island, 15 species new to Terceira Island, 9 species new to Pico Island, 17 species new to São Jorge Island, 4 species new to Graciosa Island, and 2 species new to Corvo Island. The transformation of vegetation cover in the archipelago has been very drastic, and this is reflected in the presence of many foreign fungal species on the islands. From these data, we conclude that within Macaronesia, the diversity of Basidiomycota in the Azores is more similar to that in Madeira than in the Canary Islands. [ABSTRACT FROM AUTHOR]

ctive: As a result of the study, branched polysaccharides were isolated from the basidiomycete raw materials of Ganoderma lucidum. It has been established that the isolated fractions contain branched polysaccharides in the form of complexes with melanin. Methods: After purification of polysaccharides by ion-exchange chromatography, two fractions were obtained from basidial raw materials: neutral polysaccharides GW-1 with a yield of 25.71% and anionic polysaccharides GW-2, the yield of which was 5.26%, respectively. The physicochemical properties of the obtained samples were studied by IR and UV spectroscopy. The degree of purity of the obtained fractions of branched polysaccharides was established. Using gas chromatography, one-dimensional (13C NMR, 1H NMR) and two-dimensional (COSY, TOCSY, HSQC, HMBC, NOESY) NMR spectroscopy, the composition and molecular structure of the obtained polysaccharide samples were determined. Results and Discussion: The results showed that the isolated and purified polysaccharides are branched glucans with 1,4,6- and 1,3,6-bonds between glucopyranose units. Pharmacotoxicological studies were carried out on white outbred mice and it was found that the resulting polysaccharides belong to class V, practically non-toxic compounds (LD50³ 2000 mg/kg). Conclusions: Isolated polysaccharides (GW) are promising biologically active components, on the basis of which it is possible to create drugs with hepatoprotective and antitumor activity. The emergence of antibiotic resistance in pathogens is one of the major health concerns facing mankind as different bacterial strains have developed resistance to antibiotics over the period of time due to overuse and misuse of antibiotics. Besides this, ability to form biofilms is another major factor contributing to antibiotic resistance, which has necessitated the need for exploration for novel and effective compounds with ability to inhibit biofilm formation. Endophytic fungi are reported to exhibit antibacterial and anti-biofilm potential and could serve as a potent source of novel antibacterial compounds. Majority of the bioactivities have been reported from fungi belonging to phylum Ascomycota. Endophytic basidiomycetes, inspite of their profound ability to serve as a source of bioactive compounds have not been exploited extensively. In present study, an attempt was made to assess the antibacterial, anti-biofilm and biofilm dispersion potential of an endophytic basidiomycetous fungus Schizophyllum commune procured from the culture collection of our lab. Ethyl acetate extract of S. commune showed good antibacterial activity against Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, Salmonella enterica and Vibrio cholerae. Minimum inhibitory concentration and minimum bactericidal concentration of the extract were in the range of 1.25-10 mg/ml against the tested bacterial pathogens. The mode of action was determined to be bactericidal which was further confirmed by time kill studies. Good anti-biofilm activity of S. commune extract was recorded against K. pneumoniae and S. enterica, which was further validated by fluorescence microscopy. Substituted benzaldehydes are the most commonly used natural-occurring flavours in the world. The consumer's preference for 'natural or organic' aromas has increased the request for flavours possessing the 'natural' status. The resulting shortage of aromatic aldehydes of extractive origin, such as vanillin, veratraldehyde and piperonal, can be offset by developing a new biotechnological synthesis method. Here, we report a study on the microbiological reduction of five natural benzoic acid derivatives, namely p-anisic, vanillic, veratric, piperonylic and eudesmic acids, to produce the corresponding fragrant aldehydes. We found that different Basidiomycota strains can efficiently perform this transformation, with good chemical selectivity and tolerance to the toxicity of substrates and products. Besides confirming the carboxylic acid reductase activity of the already studied fungi Pycnoporus cinnabarinus, we discovered that other species such as Pleurotus eryngii, Pleurotus sapidus and Laetiporus sulphureus as well as the non-ligninolytic fungi Lepista nuda are valuable microorganisms for the synthesis of anisaldehyde, vanillin, veratraldehyde, piperonal and 3,4,5-trimethoxybenzaldehyde from the corresponding acids. According to our findings, we propose a reliable process for the preparation of the above-mentioned aldehydes, in natural form. A review of selected studies on fungal endophytes confirms the paucity of Basidiomycota and basal fungi, with almost 90% attributed to Ascomycota. Reasons for the low number of Basidiomycota and basal fungi, including the Chytridiomycota, Mucoromycota, and Mortierellomycota, are advanced, including isolation procedure and media, incubation period and the slow growth of basidiomycetes, the identification of non-sporulating isolates, endophyte competition, and fungus–host interactions. We compare the detection of endophytes through culture-dependent methods and culture-independent methods, the role of fungi on senescence of the host plant, and next-generation studies. Culture collections (CCs) play an important role in the ex situ conservation of biological material and maintaining species and strains, which can be used for scientific and practical purposes. The Komarov Botanical Institute Basidiomycetes Culture Collection (LE-BIN) preserves a large number of original dikaryon strains of various taxonomical and ecological groups of fungi from different geographical regions. Started in the late 1950s for the investigation of Basidiomycetes' biological activity, today, in Russia, it has become a unique specialized macromycetes collection, preserving 3680 strains from 776 species of fungi. The Collection's development is aimed at ex situ conservation of fungal diversity, with an emphasis on preserving rare and endangered species, ectomycorrhizal fungi, and strains useful for biotechnology and medicine. The main methods applied in the collection for maintaining and working with cultures are described, and the results are presented. Some problems for the isolation and cultivation of species are discussed. The taxonomical structure and variety of the strains in the collection fund are analyzed, and they show that the taxonomical diversity of fungi in the LE-BIN is commensurable with the largest CCs in the world. The achievements from the ex situ conservation of the diversity of macromycetes and the main results from the screening and investigation of the collection's strains demonstrate that a number of strains can be prospective producers of enzymes (oxidoreductases and proteases), lipids, and biologically active compounds (terpenoids, phthalides, etc.) for biotechnology and medicine. Russula subgenus Compactae is a group of ectomycorrhizal basidiomycetes, usually with large pileate fruitbodies. European members of the group are characterised by the absence of bright colours on the surfaces of their pilei, the context turning grey to black after cutting, the abundance of short lamellulae in the hymenophore, and spores with an inamyloid suprahilar spot and with low reticulate ornamentation. Our multi-locus phylogenetic study confirmed that this morphological delimitation corresponds to a well-supported clade. Within this clade, 16 species are recognised in Europe, of which five belong to the R. albonigra lineage and were described in a previous study, while eleven are fully described in this study. The application of the names R. acrifolia, R. adusta, R. anthracina, R. atramentosa, R. densissima, R. nigricans and R. roseonigra is based on the position of sequences retrieved from types or authentic material. Based on type sequences, R. fuliginosa is synonymised with R. anthracina and two varieties of R. anthracina are considered synonyms of R. atramentosa. The application of the name R. densifolia is based on a morphological match with the traditional species interpretation and the neotype specimen. Three species are described as new, R. marxmuelleriana sp. nov., R. picrophylla sp. nov. and R. thuringiaca sp. nov. This study recognises three major lineages and two species with isolated positions within the European Compactae and a morphological barcode was assigned to the species using an analysis of 23 selected characters. A search of publicly available sequences from the UNITE database revealed that the majority of species are host tree generalists and widely distributed in temperate and Mediterranean areas of Europe. The basidiomycete Serpula lacrymans is responsible for major timber devastation in houses. Basidiomycetes are known to harbour a diverse but poorly understood microbial community of bacteria, archaea, yeasts and filamentous fungi. In this study, we used amplicon‐sequencing to analyse the abundance and composition of prokaryotic communities associated with fruiting bodies of S. lacrymans and compared them to communities of surrounding material to access the 'background' community structure. Our findings indicate that bacterial genera cluster depended on sample type and that the main driver for microbial diversity is specimen, followed by sample origin. The most abundant bacterial phylum identified in the fruiting bodies was Pseudomonadota, followed by Actinomycetota and Bacteroidota. The prokaryote community of the mycelium was dominated by Actinomycetota, Halobacterota and Pseudomonadota. Actinomycetota was the most abundant phylum in both environment samples (infested timber and underground scree), followed by Bacillota in wood and Pseudomonadota in underground samples. Nocardioides, Pseudomonas, Pseudonochardia, Streptomyces and Rubrobacter spp. were among others found to comprise the core microbiome of S. lacrymans basidiocarps. Bioconversion of plant material with white-rot fungi has demonstrated improvements in in vitro digestibility, however, effects on rumen fermentation parameters and methane (CH4) production, when included as a dietary ingredient, are not clear. The aim of this study was to assess changes in rumen fermentation parameters and in vitro CH4 production by dietary inclusion of ryegrass-fescue hay biodegraded with P. ostreatus (H-PO) during 14 days of solid fermentation. The experiment used 2 diets with contrasting acid detergent fibre (aADFom) content which were added with 0, 3 and 6% H-PO. Substrates were incubated 3 times with sampling at 24 h. Results were analysed by ANOVA, and compared with Tukey's test and polynomial contrasts. Inclusion of H-PO at concentrations of 3 and 6% resulted in increased in vitro dry matter (DM) disappearance (IVDMD) by 10 and 12% respectively, whereas high concentrations of aADFom reduced IVDMD by 10% as compared to low aADFom. Methane production (mg/g DM incubated) and yield (mg/g DM degraded) were reduced by increasing aADFom and P. ostreatus biodegraded hay concentrations in the diets (p < 0.001) whereas concentrations of total short-chain fatty acids were not affected by the inclusion of biodegraded hay (p = 0.080) or aADFom concentration (p = 0.076). Increasing concentrations of biodegraded hay resulted in increased acetate (p < 0.001), and reduced propionate proportions (p = 0.021) but did not affect butyrate (p = 0.233). In conclusion the use of biodegraded hay as a pre-treated ingredient improved the utilisation of low and high fibre diets by increasing the in vitro disappearance, and decreasing CH4 emissions, without increasing the propionate proportion. The use of hay biodegraded with P. ostreatus (H-PO) improves the digestibility of ruminant diets. The use of H-PO decreases methane, and increases acetate production in in vitro conditions. We produced amazake (a fermented, sweet, non-alcoholic Japanese beverage) using mushrooms instead of Aspergillus oryzae. Fourteen strains were screened from 55 strains of edible mushroom by amylase activity at 50℃. Thermal stability of amylase was investigated for these strains. Amylase of Grifola frondosa SKB003 and Fuscoporia obliqua NBRC 8681 showed high thermal stability. The optimal medium composition for amylase production in G. frondosa SKB003 and F. obliqua NBRC 8681 was investigated. G. frondosa SKB003 showed extremely high amylase activity, 526 mU/g (50℃) and 565 mU/g (60℃) on brown rice medium. We selected G. frondosa SKB003, and produced amazake with this strain. The total sugar content and reducing sugar content of amazake produced by G. frondosa SKB003 (349.1 g/L and 232.9 g/L) were higher than those for commercial amazake (252.3 g/L and 161.6 g/L). The taste and aroma of amazake produced by G. frondosa SKB003 were good, and a significant difference was not observed for commercial amazake on sensory evaluation. [ABSTRACT FROM AUTHOR]

During nocturnal field expeditions in the Brazilian Atlantic Rainforest, an unexpected bioluminescent fungus with reduced form was found. Based on morphological data, the taxon was first identified as belonging to the cyphelloid genus Maireina, but in our phylogenetic analyses, Maireina was recovered and confirmed as a paraphyletic group related to genera Merismodes and Cyphellopsis. Maireina filipendula, Ma. monacha, and Ma. subsphaerospora are herein transferred to Merismodes. Based upon morphological and molecular characters, the bioluminescent cyphelloid taxon is described as the new genus Eoscyphella, characterized by a vasiform to urceolate basidiomata, subglobose to broadly ellipsoid basidiospores, being pigmented, weakly to densely encrusted external hyphae, regularly bi-spored basidia, unclamped hyphae, and an absence of both conspicuous long external hairs and hymenial cystidia. Phylogenetic analyses based on ITS rDNA and LSU rDNA support the proposal of the new genus and confirm its position in Cyphellopsidaceae. The poroid species of Auriculariales have been placed in Aporpium, Elmerina, Protodaedalea and Protomerulius and include resupinate, effused-reflexed or pileate basidiomata, 4-celled basidia and globose, ovoid to allantoid basidiospores. According to recent DNA-based phylogenetic studies, those genera have good support in Auriculariales, but many species without phylogenetic treatment remain in uncertain taxonomic position, especially from the Neotropics. In the present paper, specimens with poroid hymenophore and septate basidia were collected in Brazil and morphological and DNA analyses, based in ITS and LSU regions, evidenced three new species (Elmerina brasiliensis, E. santanaensis and Protomerulis pedratalhadensis) and three new combinations (E. minipora, E. strigosa and E. obtusispora). Simple Summary: The Antarctic Polar Plateau is one of the most extreme environments on Earth and our knowledge on the microbial diversity inhabiting this region is still limited. The BacFinder project investigated microbial diversity on the snow surface of the Polar Plateau, focusing on the vicinity of the Concordia Antarctic Research Station, to assess the microbial diversity and the potential impact of human presence on such a pristine environment. We found that seasonality was the main driver for both bacterial and fungal assemblages, while biodiversity appeared unaffected by distance from the base. Amplicon sequencing revealed a predominance of Basidiomycota (49%) and Ascomycota (42%) for the fungal component. Bacteroidota (65.8%) is the main representative of the bacterial component. Basidiomycetes are almost exclusively represented by yeast-like fungi. Overall, the study highlighted the impact of human activity on the microbial composition in this environment and may provide critical information on the habitability of extra-terrestrial analogs on our planet and on the possibility to explore the surfaces of icy worlds. The French–Italian Concordia Research Station, situated on the Antarctic Polar Plateau at an elevation of 3233 m above sea level, offers a unique opportunity to study the presence and variation of microbes introduced by abiotic or biotic vectors and, consequently, appraise the amplitude of human impact in such a pristine environment. This research built upon a previous work, which explored microbial diversity in the surface snow surrounding the Concordia Research Station. While that study successfully characterized the bacterial assemblage, detecting fungal diversity was hampered by the low DNA content. To address this knowledge gap, in the present study, we optimized the sampling by increasing ice/snow collected to leverage the final DNA yield. The V4 variable region of the 16S rDNA and Internal Transcribed Spacer (ITS1) rDNA was used to evaluate bacterial and fungal diversity. From the sequencing, we obtained 3,352,661 and 4,433,595 reads clustered in 930 and 3182 amplicon sequence variants (ASVs) for fungi and bacteria, respectively. Amplicon sequencing revealed a predominance of Basidiomycota (49%) and Ascomycota (42%) in the fungal component; Bacteroidota (65.8%) is the main representative among the bacterial phyla. Basidiomycetes are almost exclusively represented by yeast-like fungi. Our findings provide the first comprehensive overview of both fungal and bacterial diversity in the Antarctic Polar Plateau's surface snow/ice near Concordia Station and to identify seasonality as the main driver of microbial diversity; we also detected the most sensitive microorganisms to these factors, which could serve as indicators of human impact in this pristine environment and aid in planetary protection for future exploration missions. Interest in edible and medicinal macrofungi is millennial in terms of their uses in health and food products in Central Asia, while interest in inedible and medicinal macrofungi has grown in popularity in recent years. Edible and inedible medicinal basidiomycetes were collected during field surveys from different regions of Uzbekistan. The morphological characters and similarity assessment of rDNA-Internal Transcribed Spacer sequence data were used to measure diversity and habitat associations. A number of 17 species of medicinal macrofungi of ethnomycological and medicinal interest was found associated with 23 species of trees and shrubs belonging to 11 families and 14 genera. Polyporaceae and Hymenochaetaceae were represented by the highest number of species followed by Ganodermataceae, Fomitopsidaceae, Auriculariaceae, Cerrenaceae, Grifolaceae, Phanerochaetaceae, Laetiporaceae, Schizophyllaceae, and Stereaceae. The highest number of medicinal basidiomycete species was reported in the following host genera: Acer, Betula, Celtis, Crataegus, Juglans, Juniperus, Lonicera, Malus, Morus, Platanus, Populus, Prunus, Quercus, and Salix. An updated list of edible and inedible medicinal mushrooms identified in Uzbekistan, their morphological characteristics, and phylogenetic placement are given for the first time. Information is provided on their uses in traditional and modern medicine. Their bioactive compounds and extracts can be applied as medicines, as well as food and cosmetic ingredients. Tremella caloplacae (Zahlbr.) Diederich is a species complex including at least nine different species. Here, we formally describe the new species Tremella elegantis , T. nimisiana , T. parietinae , T. pusillae and T. sorediatae. Tremella elegantis induces galls in the hymenium of Rusavskia elegans and forms 2-celled basidia, where cells rarely elongate and sometimes give the appearance of two immature, independent basidia. Tremella nimisiana has small basidiomata (less than 1 mm diam.), narrowly ellipsoid to pyriform 2-celled, occasionally clavate to subcylindrical 3-celled basidia, and grows in the hymenium of Xanthocarpia species. Tremella parietinae is characterized by the exclusive growth in the hymenium of Xanthoria parietina , the broadly fusiform to ellipsoid probasidia, and the subspherical, pyriform or ellipsoid 2(–3)-celled basidia. Tremella pusillae has ellipsoidal probasidia, 2(–3)-celled pyriform or ellipsoidal basidia that sometimes are constricted at the septum, and grows only on Calogaya pusilla. Tremella sorediatae is characterized by inducing galls on the thallus of Rusavskia sorediata and by pyriform to ellipsoid basidia that sometimes are constricted at the septum. Three species are not formally described and are left unnamed as Tremella sp. 13 on Calogaya biatorina , Tremella sp. 14 on Calogaya decipiens and Tremella sp. 15 on Polycauliona sp. This study investigates the mycelial biomass production and chitosan extraction potential of various Basidiomycota strains, including Heterobasidion annosum, Phanerochaete chrysosporium, Pleurotus ostreatus, Trametes versicolor, and Lentinus lepideus. Both submerged fermentation (SF) and solid-state fermentation (SSF) methods were employed. The chitosan yield in basidiocarps of Pleurotus ostreatus, Agaricus bisporus, and Ganoderma applanatum was also evaluated as a reference material. The chitosan extracted from fungal cells was characterized using elemental analyses and FTIR spectroscopy. Among the cultivated strains, P. chrysosporium exhibited the highest mycelial biomass concentration in SF (1.03 g 100 mL–1) after 14 days, while T. versicolor achieved the highest biomass concentration in SSF (3.65 g 100 mL–1). The highest chitosan yield was obtained from the mycelium of P. chrysosporium (0.38%) and T. versicolor (0.37%) in shaken SF. Additionally, commercially cultivated A. bisporus demonstrated the highest chitosan yield in fungal fruiting bodies (1.7%). The extracted chitosan holds potential as a functional biopolymer additive for eco-friendly materials, serving as an alternative to synthetic wet and dry strength agents in packaging materials. Development of new biological preparations to control Heterobasidion root rot is a complex process, but when a potential antagonist is identified, cultivation of the fungus is required. In this study, five different substrates (deciduous sawdust, coniferous sawdust, rye bran, straw and corn kernels) were tested as substrates for the cultivation of three fungal species: Bjerkandera adusta, Phlebiopsis gigantea, and Sistotrema brinkmannii, which could be potentially used against Heterobasidion spp. Mycelial growth was evaluated visually, and oidia production was estimated microscopically. In the straw substrate, P. gigantea produced significantly more (p < 0.05) oidia compared to the other substrates. In addition, oidia production at two different incubation temperatures were compared. As a result, the best substrate for cultivation of all three fungal species was coniferous sawdust. The edible oyster mushroom Pleurotus ostreatus is one of the most cultivated species worldwide. Morphogenesis associated with the maturation of fruit bodies is controlled by two unlinked loci of sexual compatibility matA and matB with multiple alleles (tetrapolar system of sexual compatibility). Quantitative analysis of the alleles of mating compatibility loci in 17 natural isolates collected in the Moscow region was performed in mon–mon (monokaryons–monokaryon) and di–mon (dikaryon–monokaryon) crossings. Four monokaryotic testers strains which were heteroallelic at both mating type loci were obtained for each of the five natural mushroom isolates by using original technique of sterile spore prints on Petri dishes and mon–mon crossing. Twelve natural isolates were crossed via di-mon mating with the four monokaryotic testers M-38. Genetic analysis of the alleles of sexual compatibility loci in 17 natural isolates revealed multiple alleles at both loci: at least ten alleles at matA locus and eight alleles at matB locus. Structural organization analysis of the matA locus was performed in silico for homokaryotic strains PC9 and PC15 based on the whole-genome sequencing data available at DOE Joint Genome Institute. The matA locus has an extremely divergent structure: there are one copy of the homeodomain gene hd1 and one copy of the hd2 gene in the PC9 strain, whereas the matA locus of the PC15 strain is composed by two copies of hd1.1 and hd1.2 genes (class HD1 homeodomain proteins) and one copy of hd2 gene (class HD2 proteins). Comprehensive analysis of amino acid sequences of HD1 and HD2 homeodomain proteins demonstrated that the proteins have a globular structure with the nuclear localization and contain a variable N-terminus and a more conserved DNA-binding domain with a specific conserved motif WFXNXR in the third ɑ-helix. The results suggest that multiple alleles of the matA locus of sexual compatibility in basidiomycete fungi is achieved due to both different copy number of the coding hd genes within the locus and the variability of the coding gene sequences. The fungus Moniliophthora perniciosa secretes protein effectors that manipulate the physiology of the host plant, but few effectors of this fungus have had their functions confirmed. We performed functional characterization of a promising candidate effector of M. perniciosa. The inoculation of rBASIDIN at 4 µmol L−1 in the mesophyll of leaflets of Solanum lycopersicum caused symptoms of shriveling within 6 h without the presence of necrosis. However, when sprayed on the plant at a concentration of 11 µmol L−1, it caused wilting symptoms only 2 h after application, followed by necrosis and cell death at 48 h. rBASIDIN applied to Theobroma cacao leaves at the same concentration caused milder symptoms. rBASIDIN caused hydrogen peroxide production in leaf tissue, damaging the leaf membrane and negatively affecting the photosynthetic rate of Solanum lycopersicum plants. Phylogenetic analysis indicated that BASIDIN has orthologs in other phytopathogenic basidiomycetes. Analysis of the transcripts revealed that BASIDIN and its orthologs are expressed in different fungal species, suggesting that this protein is differentially regulated in these basidiomycetes. Therefore, the results of applying BASIDIN allow the inference that it is an effector of the fungus M. perniciosa, with a strong potential to interfere in the defense system of the host plant. Fungal diseases are a leading threat to human health, especially in individuals with compromised immunity. Although there have been recent important advances in antifungal drug development, antifungal resistance, drug–drug interactions and difficulties in delivery remain major challenges. Among its pleiotropic actions, nitric oxide (NO) is a key molecule in host defense. We have developed a flexible nanoparticle platform that delivers sustained release of NO and have demonstrated the platform's efficacy against diverse bacteria as well as some fungal species. In this work, we investigate the effects of two NO-releasing particles against a panel of important human yeast. Wood decomposition is regulated by multiple controls, including climate and wood traits, that vary at local to regional scales. Yet decomposition rates differ dramatically when these controls do not. Fungal community dynamics are often invoked to explain these differences, suggesting that knowledge of ecosystem properties that influence fungal communities will improve understanding and projection of wood decomposition. We hypothesize that deadwood inputs decompose faster in forests with higher stocks of downed coarse woody material (CWM) because CWM is a resource from which lignocellulolytic fungi rapidly colonize new inputs. To test this hypothesis, we measure decomposition of 1,116 pieces of fine woody material (FWM) of five species, incubated for 13 to 49 months at five locations spanning 10°-latitude in eastern U.S. forest. We place FWM pieces near and far from CWM across observational transects and experimental common gardens. Soil temperature positively affects location-level mean decomposition rates, but these among-location differences are smaller than within-location variation in decomposition. Some of this variability is caused by CWM, where FWM pieces next to CWM decompose more rapidly. These effects are greater with time of incubation and lower initial wood density of FWM. The effect size of CWM is of the same relative magnitude as for the known controls of temperature, deadwood density and diameter. Abundance data for CWM is available for many forests and hence may be an ecosystem variable amenable for inclusion in decomposition models. Our findings suggest that conservation efforts to rebuild depleted CWM stocks in temperate forests may accelerate decomposition of fresh deadwood inputs. [ABSTRACT FROM AUTHOR]

The newly discovered cyphelloid species Chromocyphella meloana is here described and illustrated based on specimens collected in continental France. Hypothetic reproductive isolation of this species is supported by the morpholological characteristics of fruit bodies, which initially develop as corticioid basidiomes, and mature into folded, torn structures producing ellipsoid brown rugose basidiospores. A combination of ITS, 28S rDNA and rpb2 sequences is used to classify the collection as a new species within the genus Chromocyphella. A key to the species of Chromocyphella is provided. The genus Scleroderma contains gasteroid basidiomycetes, which form globose spores with echinulate to reticulate ornamentation on the surface. Based on the morphological observations in combination with molecular data, four new species, S. erubescens, S. separatum, S. squamulosum, and S. vinaceum, were described from Yunnan, southwestern China. Images of fresh basidiomata and scanning electron microscope (SEM) images of basidiospores are provided. The potential of wood-rotting and litter-deconstructing basidiomycetes to convert lignocellulose into a wide variety of products has been extensively studied. In particular, wood-rotting basidiomycete secretomes are attracting much attention from researchers and biotechnology companies due to their ability to produce extracellular hydrolytic and oxidative enzymes that effectively degrade cellulose, hemicellulose, and lignin of plant biomass. An analysis of the available literature data shows that Basidiomycota fungi, which are most adapted to the depolymerization of plant polysaccharides, are promising but so far unexploited sources of new hydrolytic enzymes. The review summarizes the latest data on the great variety, common features, and unique properties of individual fungi and the production of cellulases and xylanases by various physiological and ecological groups of basidiomycetes. The most important microbial cellulase-producing strains for submerged and solid-phase fermentation, as well as the main substrates, including the use of agro-industrial waste, are considered. It highlights ways to increase both cellulase and xylanase expression levels and the cost-effectiveness of producing these enzymes for various biotechnological applications. It is anticipated that this review will be particularly useful to novice scientists working in the lignocellulose biorefinery, as it describes current knowledge and issues related to the production and regulation of polysaccharide hydrolyzing enzyme synthesis. Background: The Trichosporonaceae family comprises a large number of basidiomycetes widely distributed in nature. Some of its members, especially Trichosporon asahii, have the ability to cause human infections. This ability is related to a series of virulence factors, which include lytic enzymes production, biofilm formation, resistance to oxidising agents, melanin and glucuronoxylomannan in the cell wall, metabolic plasticity and phenotypic switching. The last two are poorly addressed within human pathogenic Trichosporonaceae. Objective: These factors were herein studied to contribute with the knowledge of these emerging pathogens and to uncover mechanisms that would explain the higher frequency of T. asahii in human infections. Methods: We included 79 clinical isolates phenotypically identified as Trichosporon spp. and performed their molecular identification. Lactate and N‐acetyl glucosamine were the carbon sources of metabolic plasticity studies. Morphologically altered colonies after subcultures and incubation at 37°C indicated phenotypic switching. Results and Conclusion: The predominant species was T. asahii (n = 65), followed by Trichosporon inkin (n = 4), Apiotrichum montevideense (n = 3), Trichosporon japonicum (n = 2), Trichosporon faecale (n = 2), Cutaneotrichosporon debeurmannianum (n = 1), Trichosporon ovoides (n = 1) and Cutaneotrichosporon arboriforme (n = 1). T. asahii isolates had statistically higher growth on lactate and N‐acetylglucosamine and on glucose during the first 72 h of culture. T. asahii, T. inkin and T. japonicum isolates were able to perform phenotypic switching. These results expand the virulence knowledge of Trichosporonaceae members and point for a role for metabolic plasticity and phenotypic switching on the trichosporonosis pathogenesis. Transcriptional regulation is vital for organismal survival, with many layers and mechanisms collaborating to balance gene expression. One layer of this regulation is genome organization, specifically the clustering of functionally related, co-expressed genes along the chromosomes. Spatial organization allows for position effects to stabilize RNA expression and balance transcription, which can be advantageous for a number of reasons, including reductions in stochastic influences between the gene products. The organization of co-regulated gene families into functional clusters occurs extensively in Ascomycota fungi. However, this is less characterized within the related Basidiomycota fungi despite the many uses and applications for the species within this clade. This review will provide insight into the prevalence, purpose, and significance of the clustering of functionally related genes across Dikarya, including foundational studies from Ascomycetes and the current state of our understanding throughout representative Basidiomycete species. For more than a century, lichens have been used as an example of dual-partner symbiosis. Recently, this has been challenged by the discovery of various basidiomycetous yeasts that coexist in multiple lichen species, among which Cladonia lichens from Europe and the United States were discovered to be highly specifically associated with the basidiomycetous yeast of the family Microsporomycetaceae. To verify this highly specific relationship, we investigated the diversity of basidiomycetous yeasts associated with Cladonia rei, a widely distributed lichen in Japan, by applying two approaches: yeast isolation from the lichen thalli and meta-barcoding analysis. We obtained 42 cultures of Cystobasidiomycetous yeast which were grouped into six lineages within the family Microsporomycetaceae. Unexpectedly, although the cystobasidiomycetes-specific primer was used, not only the cystobasidiomycetous yeasts but species from other classes were also detected via the meta-barcoding dataset; in particular, pucciniomycetous yeasts were found at a high frequency in some samples. Further, Halobasidium xiangyangense, which was detected in every sample with high abundance, is highly likely a generalist epiphytic fungus that has the ability to associate with C. rei. In the pucciniomycetous group, most of the detected species belong to the scale insect-associated yeast Septobasidium genus. In conclusion, even though Microsporomyces species are not the only yeast group associated with Cladonia lichen, our study demonstrated that the thalli of Cladonia rei lichen could be a suitable habit for them. Mature tropical urban trees are susceptible to root and trunk rot caused by pathogenic fungi. A metagenomic survey of such fungi was carried out on 210 soil and tissue samples collected from 134 trees of 14 common species in Singapore. Furthermore, 121 fruiting bodies were collected and barcoded. Out of the 22,067 OTUs (operational taxonomic units) identified, 10,646 OTUs had annotation information, and most were either ascomycetes (63.4%) or basidiomycetes (22.5%). Based on their detection in the diseased tissues and surrounding soils and/or the presence of fruiting bodies, fourteen basidiomycetes (nine Polyporales, four Hymenochaetales, one Boletales) and three ascomycetes (three species of Scytalidium) were strongly associated with the diseased trees. Fulvifomes siamensis affected the largest number of tree species surveyed. The association of three fungi was further supported by in vitro wood decay studies. Genetic heterogeneity was common in the diseased tissues and fruiting bodies (Ganoderma species especially). This survey identified the common pathogenic fungi of tropical urban trees and laid the foundation for early diagnosis and targeted mitigation efforts. Ballistospory has been a governing factor in mushroom diversification. Modifications to fruit body morphology are subject to a series of fundamental constraints imposed by this uniquely fungal mechanism. Gill spacing in lamellate mushrooms, tube width in poroid species, and other configurations of the hymenium must comply with the distance that spores shoot themselves from their basidia. This reciprocal relationship between the development of fruit bodies and spores may have been maintained by a form of evolutionary seesaw proposed in this article. The necessity of the accurate gravitropic orientation of gills and tubes is another constraint on mushroom development and physiology, along with the importance of evaporative cooling of the hymenium for successful spore discharge and the aerodynamic shaping of the fruit body to aid dispersal. Ballistospory has been lost in secotioid and gasteroid basidiomycetes whose spores are dispersed by animal vectors and has been replaced by alterative mechanisms of active spore discharge in some species. Background: Orsellinic acid (2,4-dihydroxy-6-methylbenzoic acid, OA) and its structural analog o-Orsellinaldehyde, have become widely used intermediates in clinical drugs synthesis. Although the research on the biosynthesis of such compounds has made significant progress, due to the lack of suitable hosts, there is still far from the industrial production of such compounds based on synthetic biology. Results: With the help of genome mining, we found a polyketide synthase (PKS, HerA) in the genome of the Hericium erinaceus, which shares 60% amino acid sequence homology with ArmB from Armillaria mellea, an identified PKS capable of synthesizing OA. To characterize the function of HerA, we cloned herA and heterologously expressed it in Aspergillus oryzae, and successfully detected the production of OA. Subsequently, the introduction of an incomplete PKS (Pks5) from Ustilago maydis containing only three domains (AMP-ACP-R), which was into herA-containing A. oryzae, the resulted in the production of o-Orsellinaldehyde. Considering the economic value of OA and o-Orsellinaldehyde, we then optimized the yield of these compounds in A. oryzae. The screening showed that when maltose was used as carbon source, the yields of OA and o-Orsellinaldehyde were 57.68 mg/L and 15.71 mg/L respectively, while the yields were 340.41 mg/Kg and 84.79 mg/Kg respectively in rice medium for 10 days. Conclusions: Herein, we successfully expressed the genes of basidiomycetes using A. oryzae heterologous host. As a fungus of ascomycetes, which not only correctly splices genes of basidiomycetes containing multiple introns, but also efficiently produces their metabolites. This study highlights that A. oryzae is an excellent host for the heterologous production of fungal natural products, and has the potential to become an efficient chassis for the production of basidiomycete secondary metabolites in synthetic biology. Gene fusion is a process through which two or more distinct genes are fused into a single chimeric gene. Unlike most harmful fusion genes in cancer cells, in this study, we first found that spermidine synthetase- (SPDS, catalyst of spermidine biosynthesis) and saccharopine reductase- (SR, catalyst of the penultimate step of lysine biosynthesis) encoding genes form a natural chimeric gene, FfSpdsSr, in Flammulina filiformis. Through the cloning of full-length ORFs in different strains and the analysis of alternative splicing in developmental stages, FfSpdsSr has only one copy and unique transcript encoding chimeric SPDS-SR in F. filiformis. By an orthologous gene search of SpdsSr in more than 80 fungi, we found that the chimeric SpdsSr exists in basidiomycetes, while the two separate Spds and Sr independently exist in ascomycetes, chytridiomycetes, and oomycetes. Further, the transcript level of FfSpdsSr was investigated in different developmental stages and under some common environmental factors and stresses by RT-qPCR. The results showed that FfSpdsSr mainly up-regulated in the elongation stage and pileus development of F. filiformis, as well as under blue light, high temperature, H2O2, and MeJA treatments. Moreover, a total of 15 sets of RNA-Seq data, including 218 samples of Neurospora crassa, were downloaded from the GEO database and used to analyze the expression correlation of NcSpds and NcSr. The results showed that the separate NcSpds and NcSr shared highly similar co-expression patterns in the samples with different strains and different nutritional and environmental condition treatments. The chimeric SpdsSr in basidiomycetes and the co-expression pattern of the Spds and Sr in N. crassa indicate the special link of spermidine and lysine in fungi, which may play an important role in the growth and development of fruiting body and in response to the multiple environmental factors and abiotic stresses. The Auriculariaceae accounts for most of the species in the Auriculariales, and all species in the family are wood-decaying fungi with gelatinous, crustaceous, or woody basidiomes. Many new taxa were published recently, but the taxonomy and phylogeny of the corticioid species in the Auriculariaceae are far from resolved. We undertook a comprehensive taxonomic and phylogenetic study of the family with emphasis on corticioid specimens collected from East and Southeast Asia. Phylogenetic analyses on concatenated ITS and 28S rDNA sequences of representative taxa of the Auriculariaceae and the genera Eichleriella and Heteroradulum were carried out that resolved five new lineages. Heterocorticium gen. nov. is established for two species with resupinate coriaceous basidiomes with smooth, pigmented hymenophores. Five new species, H. bambusicola (generic type), H. latisporum, Eichleriella alpina, E. bambusicola, and Heteroradulum maolanense, are described and illustrated. In addition, Heterochaete delicata, H. discolor, and H. sinensis are transferred to Eichleriella, whereas H. roseola is regarded as a synonym of Kneiffia discolor (= H. discolor). Eichleriella aculeobasidiata is treated as a synonym of Heterochaete sinensis (= E. sinensis). Heterochaete mussooriensis is transferred to Heteroradulum with Heteroradulum semis as a heterotypic synonym. Traits related to reproduction and dispersal drive the assembly and dynamics of species communities and can explain and predict how species respond to habitat loss and fragmentation and to the changing climate. For fungi, such links remain poorly known.We examine how spore production rate, a key demographic trait, is influenced by the interaction between environmental conditions and species traits. We monitored the spore production of 97 wood‐inhabiting fungal species on 107 decaying logs for 2 years and analysed the data with a hierarchical community model.Our analysis demonstrates clear species differences in seasonal patterns, with spring and summer release dominating in perennial species, contrary to the commonly held view of autumn as the primary "mushroom season". Many species follow a diurnal pattern with a higher spore release rate during the night. Such patterns in release timing have important implications for dispersal, as shown by recent model simulations.The overall level of spore release was negatively correlated with spore size, providing new evidence that fungi face the classic trade‐off of investing either in the number or size of offspring.We found that different species within the functional group of wood‐inhabiting fungi display alternative strategies in spore release timing and along the trade‐off between offspring size and number. Fungal secondary metabolites are often pathogenicity or virulence factors synthesized by genes contained in secondary metabolite gene clusters (SMGCs). Nonribosomal polypeptide synthetase (NRPS) clusters are SMGCs which produce peptides such as siderophores, the high affinity ferric iron chelating compounds required for iron uptake under aerobic conditions. Armillaria spp. are mostly facultative necrotrophs of woody plants. NRPS-dependent siderophore synthetase (NDSS) clusters of Armillaria spp. and selected Physalacriaceae were investigated using a comparative genomics approach. Siderophore biosynthesis by strains of selected Armillaria spp. was evaluated using CAS and split-CAS assays. At least one NRPS cluster and other clusters were detected in the genomes studied. No correlation was observed between the number and types of SMGCs and reported pathogenicity of the species studied. The genomes contained one NDSS cluster each. All NDSSs were multi-modular with the domain architecture (ATC)3(TC)2. NDSS clusters of the Armillaria spp. showed a high degree of microsynteny. In the genomes of Desarmillaria spp. and Guyanagaster necrorhizus, NDSS clusters were more syntenic with NDSS clusters of Armillaria spp. than to those of the other Physalacriaceae species studied. Three A-domain orthologous groups were identified in the NDSSs, and atypical Stachelhaus codes were predicted for the A3 orthologous group. In vitro biosynthesis of mainly hydroxamate and some catecholate siderophores was observed. Hence, Armillaria spp. generally contain one highly conserved, NDSS cluster although some interspecific variations in the products of these clusters is expected. Under natural conditions in Spain, the fruiting bodies of Pleurotus eryngii var. eryngii, a basidomycete of great economic and gastronomic importance, appears at the base of the stem of (mainly) Eryngium campestre, surrounded by the plant's dried-out basal leaves. Traditionally collected in the wild of Mediterranean areas, this fungus is now cultivated all over the world for its culinary and even medicinal properties. However, controversy exists regarding its lifestyle. Some authors indicate it is a saprotroph, while others suggest it is a weak or even virulent parasite that causes male sterility in E. campestre. The present work aims to clarify whether P. eryngii behaves as a saprotroph, a weak or virulent parasite to E. campestre. The soil beside the roots of naturally growing E. campestre plants was inoculated with a commercial strain of P. eryngii, and the trophic mode of the fungus recorded. Soil and root samples were subjected to molecular analysis to confirm the absence/presence of the fungus. The death of the plants and the molecular analysis showed P. eryngii is a facultative necrotrophic parasite of E. campestre in natural conditions and a virulent parasite in greenhouse. The coalescence of fluid droplets on the surface of ballistospores powers their launch into the air at a speed of up to one meter per second with an acceleration of thousands of g 's. This mechanism has been studied for more than a century and its solution is an emblem of mycological progress. Because the spores move too fast for the launch to be watched with a light microscope, early advances were made by inferences about what must be happening when the spores disappeared rather than direct observations. These investigations were followed by ingenious experiments that led to a satisfying explanation of ballistospory by the 1990s. Ultra-high-speed video recordings of spore discharge verified this model in the 2000s and subsequent research has shown how the mechanism has been adapted to launch spores over different distances. The available evidence suggests that many of these adaptations have been achieved by changes in spore morphology. Fungi contain species with a plethora of ways of adapting to life in nature. Consequently, they produce large amounts of diverse biomolecules that can be generated on a large scale and in an affordable manner. This makes fungi an attractive alternative for many biotechnological processes. Ascomycetes and basidiomycetes are the most commonly used fungi for synthesis of metal-containing nanoparticles (NPs). The advantages of NPs created by fungi include the use of non-toxic fungus-produced biochemicals, energy efficiency, ambient temperature, pressure conditions, and the ability to control and tune the crystallinity, shape, and size of the NPs. Furthermore, the presence of biomolecules might serve a dual function as agents in NP formation and also capping that can tailor the (bio)activity of subsequent NPs. This review summarizes and reviews the synthesis of different metal, metal oxide, metal sulfide, and other metal-based NPs mediated by reactive media derived from various species. The phyla ascomycetes and basidiomycetes are presented separately. Moreover, the practical application of NP mycosynthesis, particularly in the fields of biomedicine, catalysis, biosensing, mosquito control, and precision agriculture as nanofertilizers and nanopesticides, has been studied so far. Finally, an outlook is provided, and future recommendations are proposed with an emphasis on the areas where mycosynthesized NPs have greater potential than NPs synthesized using physicochemical approaches. A deeper investigation of the mechanisms of NP formation in fungi-based media is needed, as is a focus on the transfer of NP mycosynthesis from the laboratory to large-scale production and application. Competition between 31 macromycete species and two harmful micromycetes Aspergillus niger and Penicillium polonicum was evaluated using dual-culture plate assay. All investigated fungi, except for Inonotus obliquus and Lepista luscina, possessed different levels of antagonistic activity against the tested micromycetes. Hypsizygus marmoreus and Lyophyllum shimeji were inactive against A. niger. Morchella esculenta and Oxyporus obducens were passive in the case of co-growth with P. polonicum. The study of fungal interactions showed variability in types of reactions and level of their visualisation. Co-cultivation of the studied species resulted in the following reactions: deadlock after mycelial contact and at a distance, partial or complete replacement after initial deadlock on contact. In general, the studied macromycetes showed moderate activity against the two micromycetes, as 11 species showed active and 7 species moderate antagonism. Results revealed higher antagonistic activity of macromycetes against P. polonicum than in A. niger co-cultures. Wood-decaying species Ganoderma lucidum and Trametes versicolor were the most active fungi against the two tested micromycetes with a maximal antagonism index. These findings provide valuable insights which can be further explored by means of in vivo assays to find a suitable agent for the biocontrol of diseases or spoilage caused by A. niger and P. polonicum. Polyhydroxyalkanoates (PHAs) including poly-3-hydroxybutyrate (P3HB) as secondary metabolisms were in vitro produced by the edible basidiomycetous mushroom Astraeus odoratus during its growth on malt agar extract. Various carbon and nitrogen sources containing cellulose, glucose, glycerol, rice straw powder, soybean meal and peptone were investigated for the growth of basidiomycetous mushrooms. During cultivation, the A. odoratus culture exudated the considerably extracellular fluid up to approx. 2.3 ml on 2% malt extract agar plate within 7 days. The chemical compounds of the exudated fluid were further investigated by Fourier-transform infrared spectroscopy (FTIR) and gas chromatography/mass spectrometry (GC/MS); and its morphology of the lyophilized sample was observed by scanning electron microscope (SEM). FTIR results showed the characteristic bands of OH at 3445 cm−1, CH/CH2/symmetric CH3 (stretch) at 2923 and 2852 cm−1, C=O at 1730 cm−1, asymmetric CH3 (bend) at 1454 and 1414 cm−1, C−O of COO– at 1396 cm−1 and C–O–C at 1223, 1160, 1116, 1058 and 1019 cm−1 which were similar to the absorptive characteristics of P3HB. Methyl ester derivatives of GC/MS results identified 7 compounds including: 3-hydroxybutanoic (monomer of PHB), aminobenzoic, salicylic, hexadecenoic, octadecadienoic, octadecenoic and octadecanoic acids. SEM images revealed a fibriform and porous materials. Hence, the occurrence of PHAs was first described in a basidiomycetous mushroom A. odoratus. Guar gum is a polymer that is widely used as a gelling agent for technological liquids in the petroleum industry. In this work, we have studied the potential for the environmentally friendly biodegradation of guar gum by enzymes of basidiomycetes for efficient disposal of oil industry wastes. We compared the enzymatic activity against guar gum of seven basidiomycete strains for the first time, namely Trametes hirsute MT-24.24, Lactarius necator, T. hirsute MT-17.24, Schizophyllum commune MT-33.01, Fomes fomentarius MT-4.05, Fomitopsis pinicola MT-5.21, and T. versicolor It-1. This comparison showed that the preparation based on the Fomitopsis pinicola MT-5.21 fungal mycelium at a concentration of 0.05% provides the most efficient decomposition of a hydraulic fracturing fluid containing guar gum. By varying the enzyme concentration in this fluid, it is possible to control the decrease in its viscosity over time. The developed enzyme preparation is an efficient and environmentally friendly guar gum biodegrader and can serve for processing waste fracturing fluids based on polysaccharides in order to reuse water resources. Background: Oxidative stress is responsible for the onset of several chronic and degenerative diseases. Exogenous supply of antioxidants is reported to neutralize the effects of oxidative stress. Several synthetic antioxidants suffer from various side effects which necessitates the exploration of antioxidant compounds from natural sources. Endophytic fungi residing in the plants are gaining the attention of researchers as a source of novel antioxidants. Majority of the research conducted so far on endophytic fungi has been restricted to the members of phylum ascomycota. Basidiomycota, inspite of their immense bioactive potential remain relatively unexploited. This study aimed to assess the ameliorative effects of an endophytic Schizophyllum commune (basidiomycetous fungus) against oxidative stress associated altered antioxidant levels, genotoxicity and cellular damage to different organs in bisphenol A exposed fresh water fish Channa punctatus. Results: Good antioxidant and genoprotective potential was exhibited by S. commune extract in in vitro studies conducted using different antioxidant, DNA damage protection, and cytokinesis blocked micronuclei assays. In vivo studies were performed in fresh water fish Channa punctatus exposed to bisphenol A. A significant decrease in the considered parameters for DNA damage (% micronuclei and comet assay) were recorded in fish treated with S. commune extract on comparison with untreated bisphenol A exposed group. The S. commune extract treated fish also exhibited an increase in the level of antioxidant enzymes viz. catalase, superoxide dismutase and glutathione reductase as well as histoprotective effect on various organs. GC-MS analysis revealed the presence of 3-n-propyl-2,4-pentanedione, n-heptadecanol-1, trans-geranylgeraniol, 3-ethyl-2-pentadecanone, 1-heneicosanol and squalene as some of the compounds in S. commune extract. Knowledge about the resistance of wood–polymer composites (WPCs) to biological attack is of high importance for purpose-oriented use in outdoor applications. To gain this knowledge, uniform test methods are essential. EN 15534-1 (2018) provides a general framework, including the recommendation of applying a pre-weathering procedure before the biological laboratory tests. However, the procedure's manner is not specified, and its necessity assumes that a durability test without such pre-weathering will not produce the structural changes that occur during outdoor use. To verify this assumption, this study examined the influence of natural, ground-level pre-weathering on the material properties of different WPC variants, which were tested at intervals of six months in four durability tests under laboratory conditions in accordance with EN 15534-1 (2018). Weathering factors were calculated from determined characteristic values such as mass loss, and loss in moduli of elasticity (MOE) and rupture (MOR). The weathering factors based on mechanical properties tended to decrease with increasing weathering duration. The expected negative influence of pre-weathering on these material properties was thus not confirmed. The weathering factors based on mass loss were subject to high variation. No significant effect of pre-weathering on mass loss due to fungal attack became evident. Overall, the necessity of a pre-weathering step in biological durability tests shall be questioned based on the presented results. [ABSTRACT FROM AUTHOR]
