Background: The conservation of aquatic and semiaquatic turtles requires knowledge of the area and vegetation structure of habitat used for nesting, and nesting migration route. We aimed to survey the effects of habitat features to the nest site selection, nesting success, and test the possibility of nest site fidelity. Our study was carried out at 10 different nesting areas, with special emphasis on data from returning females in a pond system in Hungary between 2014 and 2017. Results: Most nesting attempts were found in closed sand steppes, uncharacteristic dry and semi-dry grasslands habitat patches. The principal component analysis (PCA) showed that increased sandy soil cover, sunlight and slope were important variables in nest site choice. The increasing PCA first axis score significantly increased the chance of an emergence. The degradation of open steppe vegetation, occurrence of weeds, invasive and disturbance tolerant species have a negative effect on the selection of nest sites. We observed that 96.55% of nests were located within 20 m south of a pine forest at preferred nest site at pond 5, which provided the right incubation temperature through partial shading. The returning females nested significantly closer to the northern pine forest than the single clutch females. Most probably the returning females already has the necessary experience to select the right nesting site. The individually marked females did not choose new nesting areas during the monitored years which suggests nesting area fidelity, but we did not find nest site fidelity. Conclusion: The maintenance of mosaic habitat structure, slowing down the succession process at the nesting area should be basic priorities in European pond turtle conservation programs. We suggested a spatial and temporal scheduling of land management and agricultural work to the local farmers. If the actual nest site is in an agricultural area, all work should be avoided throughout the year. Agricultural machinery should avoid the migration routes of adult turtles and emerged hatchlings during the concerned period. Simple Summary: The Chinese three-keeled pond turtle is an endangered species in the wild but with large-scale farming. However, it is difficult to distinguish individuals belonging to different habitats from their appearance. The current study investigates intestinal microbial diversity, metabolic pathways, and resistance to various types of antibiotics among wild, farm, and released turtles. The results showed that at the phylum level, Bacteroidota, Firmicutes, Fusobacteriota, and Actinobacteriota were abundant in the wild group, but Chloroflexi was only dominant in the farm and released groups. In addition, most genera increased in the groups of farm and released turtles, while few were abundant in the wild group. A metabolic pathway analysis revealed heightened activity in the associated pathways in the wild turtles. The antibiotic resistance gene macB was higher in the farm turtle group, while tetA was more abundant in the wild turtles, and srpYmcr was abundant in the released group. These outcomes were deemed helpful for a better understanding of the relationship between the intestinal microbiota of Mauremys reevesii and its habitats and could be useful for tracking habitats to protect, manage, and conserve this endangered species. The intestine of living organisms harbors different microbiota associated with the biological functioning and health of the host and influences the process of ecological adaptation. Here, we studied the intestinal microbiota's composition and functional differences using 16S rRNA and metagenomic analysis in the wild, farm, and released Chinese three-keeled pond turtle (Mauremys reevesii). At the phylum level, Bacteroidota dominated, followed by Firmicutes, Fusobacteriota, and Actinobacteriota in the wild group, but Chloroflexi was more abundant in the farm and released groups. Moreover, Chryseobacterium, Acinetobacter, Comamonas, Sphingobacterium, and Rhodobacter were abundant in the released and farm cohorts, respectively. Cetobacterium, Paraclostridium, Lysobacter, and Leucobacter showed an abundance in the wild group. The Kyoto Encyclopedia of Genes and Genomes (KEGG) database revealed that the relative abundance of most pathways was significantly higher in the wild turtles (carbohydrate metabolism, lipid metabolism, metabolism of cofactors, and vitamins). The comprehensive antibiotic resistance database (CARD) showed that the antibiotic resistance gene (ARG) subtype macB was the most abundant in the farm turtle group, while tetA was higher in the wild turtles, and srpYmcr was higher in the released group. Our findings shed light on the association between the intestinal microbiota of M. reevesii and its habitats and could be useful for tracking habitats to protect and conserve this endangered species. In the first range-wide population viability model for the northwestern and southwestern pond turtles (Actinemys marmorata and Actinemys pallida, respectively), a stage-based population projection matrix was assembled with 3 life stages: hatchling, juvenile, and adult. Vital rates were defined using biologically appropriate statistical distributions, with additional parametric uncertainty included for the adult survival parameter. A triple-loop stochastic simulation model was built around a population viability analysis to project pond turtle populations into the future. Initial abundance was calculated using available historical presence data and remotely sensed landscape condition metrics. A negative binomial regression was used to predict the relationship between abundance, habitat area, and human modification. Populations of pond turtles are dominated by adult individuals, so we applied a nonstable stage distribution to initial abundance values. Initial abundances of analysis units were variable across the species' ranges, but all populations declined precipitously in the population projections. By the end of the century, the mean range-wide probability of extinction was 44.3% for the northwestern species and 57.8% for the southwestern species. Consistent with other long-lived chelonian species, population growth rate was most sensitive to adult survival, indicating that where possible, conservation efforts focusing on increasing or maintaining adult survival would benefit the species. Elasticity analysis indicated a bet-hedging life history strategy where long-term reproductive output is maximized through longevity, small clutches, and frequent reproductive bouts in the face of highly variable juvenile survival. The population dynamics presented here indicate that efforts to bolster adult survival would be most beneficial in terms of long-term population viability, which can inform targeted research and management. Knowledge about the distributions of species and the variables influencing their occurrence is important for their management and conservation, but factors affecting occurrence can vary across the range of a species. Northwestern pond turtles (Actinemys marmorata) are widespread generalist turtles, but are nonetheless of conservation concern throughout their range. To better understand the distribution of northwestern pond turtles and introduced American bullfrogs (Lithobates catesbeianus), we surveyed streams on private timberlands of the interior foothills of northern California using visual encounter surveys and collecting samples of environmental DNA. We found that northwestern pond turtle occurrence was negatively related to elevation in our sampling frame. Detection probabilities with environmental DNA were approximately twice those of visual encounter surveys, but both methods were effective for detecting turtles in streams. American bullfrogs were detected in a single sample at each of 2 sites (one by environmental DNA, one by visual encounter surveys). Management for northwestern pond turtles in forest streams within our sample area will likely have the largest effect at lower elevation sites where turtles are most likely to occur. The present study aimed to report the morphometric and hematological indices and genotoxicity of a free-life population of D'Orbigny's slider turtles (Trachemys dorbigni) living in an urban area in Southern Brazil. For that, 16 specimens were randomly captured in an urban canal that receives irregular releases of wastewater. Biometrics and external visual changes were analyzed, such as turtle shell deformities, and the presence of parasites. Blood samples were collected to evaluate the hematological profile and the presence of micronuclei and other erythrocyte nuclear abnormalities as potential mutagenic and genotoxic effects. Water physicochemical parameters were also measured. Organisms with ectoparasites (31.25%) and small carapace deformations (56.25%) were observed, but maximum carapace length and weight were considered normal for the species according to the literature. The blood profile indicated low hemoglobin and hematocrit and a high number of total leukocytes, particularly eosinophils which characterize parasitic infections. A frequency of 0.12% for the micronucleus was considered basal, but the frequency of other erythrocyte abnormalities was evident, mainly of blebbed nuclei (63.79%), indicating chromosomal damage in the early stage. The results of this study suggest that natural populations of chelonian inhabiting urbanized areas are impacted by anthropogenic activities in the surrounding environment. Knowledge of the spatial distribution of reptiles is essential for decision-making in conservation under future climate change scenarios. We present a new compilation of reptile records for Odesa Oblast (i.e. province), south-west Ukraine. We compiled 662 records: 200 from our own research during 2012–2022, 362 from the published literature, 73 from public databases and 27 from museum collections. Fourteen native species of reptile (one species of Emydidae, four of Lacertidae, one of Anguidae, six of Colubridae and two of Viperidae) have been recorded in Odesa Oblast but the distribution of several are poorly known and/or records have rarely been published. We also report four introduced reptile species (one each of Emydidae, Gekkonidae, Lacertidae and Anguidae). We present the data in a grid of 462 10 × 10 km cells covering the oblast. In this compilation we did not record any new species, but our records include previously unreported localities for some species. Species richness was highest in the areas along the Black Sea, in protected areas. The main threats to the reptiles in Odesa Oblast are the alteration and degradation of habitats, military action, uncontrolled pressure from infrastructure projects and the presence of invasive species. The lower Rio Grande and Pecos River of the southwest United States have been heavily modified by human activities, profoundly impacting the integrity of their aquatic wildlife. In this context, we focused our study on the population genomics of the Rio Grande Cooter (Pseudemys gorzugi), a freshwater turtle of increasing conservation concern, residing in these two rivers and their tributaries. The genetic data revealed two distinct populations: one in the Pecos and Black Rivers of New Mexico and another in the Rio Grande and Devils River of Texas, with admixed individuals identified at the confluence of the Rio Grande and Pecos River. In addition to having a smaller geographic range, we found lower observed heterozygosity, reduced nucleotide diversity, and a smaller effective population size (N e) in New Mexico population. Our results depict a significant isolation-by-distance pattern across their distribution, with migration being notably infrequent at river confluences. Graptemys ouachitensis (CAGLE, 1953) belongs to the Graptemys genus, the Emydidae family, and the Testudines order. This study involved sequencing the complete mitochondrial genome (mitogenome) of G. ouachitensis using next-generation sequencing, and analyzing the essential characteristics, and phylogenetic relationship. The results revealed that the G. ouachitensis mitogenome was 16,674 bp in length (A: 34.1%, C: 26.0%, G: 13.0%, T: 26.9%) and included 22 tRNAs, 13 proteincoding genes, two ribosomal RNA genes, and a non-coding control region (GenBank accession: NC071766). The genome composition of G. ouachitensis presented a slight AþT bias (61.0%) and exhibited a positive AT skew (0.118) and a negative GC skew (-0.333). A phylogenetic analysis based on the complete mitogenome indicated that the G. ouachitensis was more closely associated with Malaclemys terrapin than the other eight known Emydidae species. Thus, our findings present a novel mitogenome at the species level. This study introduces the first complete mitogenome of G. ouachitensis, providing valuable molecular information for phylogenetic and conservation genetics analyses of G. ouachitensis. Background: The Asian yellow pond turtle (Mauremys mutica) is an important commercial freshwater aquaculture species in China. This species is a highly sexually dimorphic species, with males growing at a faster rate than females and exhibits temperature-dependent sex determination (TSD), in which the incubation temperature during embryonic development determines the sexual fate. However, the mechanisms of the sex determination or sex differentiation in the Asian yellow pond turtle are remain a mystery. Results: Temperature-specific gonadal transcriptomics of the Asian yellow pond turtle were performed during the thermosensitive period (stage 15) using RNA-seq technology to identify candidate genes that initiate gonadal differentiation. We uncovered candidates that were the first to respond to temperature. These candidates were sexually dimorphic in expression, reflecting differences in gonadal (Cirbp, Runx1) and germline differentiation (Vasa, Nanos1, Piwil2), gametogenesis (Hmgb3, Zar1, Ovoinhibitor-like, Kif4), steroid hormone biosynthesis (Hsd17b5, Hsd17b6), heat shock (Dnajb6, Hsp90b1, Hsp90aa1) and transient receptor potential channel genes (Trpm1, Trpm4, Trpm6, Trpv1). Conclusions: Our work will provide important genetic information to elucidate the mechanisms of sex control in the Asian yellow pond turtles, and will contribute important genetic resources for further studies of temperature-dependent sex determination in turtles. The objective of this study is to assess the occurrence of intestinal parasites in Mediterranean pond turtle Mauremys leprosa leprosa collected from three contrasting environments in Morocco. Stool samples from 92 turtles were examined for parasite detection and enumeration. The identified intestinal parasites belong to helminths (oxyurids and ascarid) and protozoa (Entamoebidae). A total of 25 turtles (27.17%) were found to be infected by helminths and/or protozoan parasites. No adult form of these parasites was detected. Eggs of oxyurid and ascarid were detected in individuals of populations studied from Oued Ksob (23.07% and 30.76% of n = 13 turtles) and Oued Zat (34.14% and 24.39% of n = 41 turtles), respectively. For protozoa, Entamoeba cysts were present in turtles in Oued Ksob (15.38% of n = 13 turtles), Oued Zat (12.19% of n = 41 turtles), and Oued Tensift (5.26% of n = 38 turtles) localities. Oxyurid eggs showed the highest intensity at Oued Zat reaching 29.30 ± 59.59 eggs per gram (EPG), versus 12 ± 0.38 EPG for ascaris eggs in Oued Ksob. Entamoeba cysts were detected in lower levels with a maximum of 1.66 ± 1.50 cysts per gram (CPG), in Oued Zat. The prevalence of turtles eliminating eggs was statistically significant between localities for different parasite groups. This study reports for the first time a parasitological characterization of gastrointestinal parasites in wild populations of M. leprosa leprosa from contrasting environments, suggesting a relationship between turtles' infestation and the quality of their habitat. Globally, many pond turtles (Family Emydidae) are of conservation concern due to contracting ranges, increasingly fragmented habitats, and declining populations. One of these turtles, the diamondback terrapin (Malaclemys terrapin), is of concern within the United States, but surveying potential estuarine habitat for the presence of the species requires high levels of time and effort. Here a species-specific, probe-based qPCR assay for identifying terrapin DNA from environmental DNA (eDNA) is described. To expand the utility of this tool, the assay was designed to allow for potential detection of at least 19 other pond turtles taxa and the utility was tested by designing a second species-specific probe for red-eared slider (Trachemys scripta elegans) with a different fluorophore, which can be used simultaneously with the terrapin probe to detect either species. Probes were found to be species-specific and effective at detecting low levels of DNA for the target species, the diamondback terrapin (10 fg/µL) and the red-eared slider (100 fg/µL) in laboratory conditions. The assay also showed species-specific detection in environmental samples taken from field locations where one species or the other were known to reside. Through citizen science approaches, this assay could be used for discovering novel terrapin habitats, monitoring invasive red-eared slider populations or surveying other emydids, with species-specific probes, at a fraction of the cost of traditional surveys. We evaluated and compared the effects of different environmental pollutants on haematological, microbiological and genotoxic characteristics of populations of the freshwater turtle Emys orbicularis from the Kavak Delta and Dardanos in Çanakkale (NW Türkiye). The haematocrit values of samples of E. orbicularis differed considerably between the two localities: they were higher in the Kavak Delta samples. The mean cell haemoglobin concentration was also significantly different between the two localities (higher in the samples of the Dardanos). The water samples of the two localities were within the limit (Class I-II) for all parameters, except Dardanos temperature (Class III). Although it was not statistically significant, the nuclear abnormality formation in the erythrocytes of the sample from Dardanos was numerically higher than that from the Kavak Delta. Phylogenetic analysis reveals that C. dorsalis forms a monophyletic group with C. picta turtles but is distinctly separated from them, aligning with previous findings. In Korea, C. dorsalis forms a discrete clade, separate from both native and invasive turtle species. No evidence of genetic disturbance or intermingling is observed. This is the first case of a complete mitochondrial genome from C. dorsalis and provides crucial data for understanding C. dorsalis and managing invasive species effectively, emphasizing the need for continued mitochondrial genome data accumulation.

AbstractSternotherus carinatus has been considered as a potential invasive species in Korea. However, the mitochondrial genome information of S. carinatus which can be used to control its effect on ecosystem is lacking. In this study, the complete mitochondrial genome of S. carinatus in Korea was sequenced and characterized. The mitochondrial genome consists of 37 genes (13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes) and a noncoding region. Phylogenetic analysis based on the mitochondrial genome sequences showed that S. carinatus from Korea is separated from other turtles which are the invasive species in Korea. Sequence divergence calculations indicated near-zero divergence between S. carinatus populations in Korea, the USA, and China, suggesting limited genetic differentiation. In the context of the broader issue of invasive species disrupting ecosystems, this research contributes to the identification of mitochondrial genomes for various freshwater turtle species, emphasizing the need for extended data collection to discern genetic mixing trends between native and non-native species. This study is a significant step toward managing S. carinatus as a potential invasive species in Korea.

AbstractThe complete mitochondrial genome of Chrysemys picta bellii in Korea was sequenced and characterized. The mitochondrial genome consists of 37 genes (13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes) and a noncoding region. Phylogenetic analysis based on the mitochondrial genome sequences revealed that C. p. bellii from Korea formed a cluster with C. p. bellii from China and C. picta from the USA, while showing clear separation from other turtle species within the C. picta cluster. This study presented the first complete mitochondrial genome from C. p. bellii in Korea, offering crucial information for managing invasive species and protecting the local ecosystem.

Blood cell counts are valuable diagnostic tools for assessing the health status of chelonians, however, reference standards for healthy blood parameters in various turtle species are lacking. In this study, forty wild female Malayemys subtrijuga were captured from ponds in Kasetsart University, and transported to laboratory for recuperating in captivity. All turtles were infected with a single leech species, Placobdelloides siamensis, with a mean of 513.7 ± 164.9 individuals per turtle, and exhibited penetrating and lesion wounds from leech infestations on both their skin and shell. Subsequently, they were cleaned and treated to eliminate ecto- and endoparasites before the recuperation period began. The turtles did not exhibit significantly differences in weight, carapace length (CL), red blood cell count (RCC), and white blood cell count (WCC) with a mean of 654.2 ± 199.9 g, 15.0 ± 2.5 cm, 327,080 ± 70,156 cells/mm3, and 73,340 ± 15,859 cells/mm3, respectively, during the initial records (week 0). However, after being maintained for 17 weeks, their health significantly improved in term of their blood parameters (RCC and WCC) and weight, except CL which remained unchanged, with a mean of 491,470 ± 16,169 cells/mm3, 18,790 ± 1496 cells/mm3, and 738.9 ± 191.5 g, respectively. Therefore, the health status obtained in this study can be used as a reference for blood parameters, weight, and recuperation period for the treatment of ill wild M. subtrijuga in captivity or as part of conservation management programs for turtles.

Dispersal has major impacts on population dynamics, population genetics and evolution, and also is critical for population management and conservation. Dispersal is frequently sex‐ and age‐specific, but current knowledge is strongly taxonomically biased toward birds and mammals.Here, we provide estimates of dispersal in a threatened freshwater turtle species, the European pond turtle Emys orbicularis. Based on 15 years of capture‐mark‐recapture (CMR) monitoring and DNA samples from 194 individuals, we quantified both demographic and genetic dispersal between three sites separated by 1.5–3.5 km. We also investigated the effect of age and sex on dispersal.Direct (CMR) and indirect (genetic) approaches provided consistent results, showing that the studied sites are connected with a flow of about one to three dozen migrants per generation. Dispersal was both age‐ and sex‐biased in this species, with frequent dispersal of adult males and a strong philopatry of juveniles (of both sexes) and adult females.The network of canals and marshes allow effective dispersal to occur among the three study sites despite the relatively large distance separating them (≤3.5 km).The strong philopatry of juveniles contrasts with the frequently higher dispersal rate in young birds and mammals and shows the relevance of investigating dispersal in various taxonomic groups. Fossils of Cretaceous sea turtles adapted to an open marine lifestyle remain rare finds to date. Furthermore, the relationships between extant sea turtles, chelonioids, and other Mesozoic marine turtles are still contested, with one key species being Santanachelysgaffneyi Hirayama, 1998, long considered the earliest true sea turtle. The species is an Early Cretaceous member of Protostegidae, a controversial clade either placed within or closely related to Chelonioidea or, alternatively, along the stem lineage of hidden-neck turtles (Cryptodira) and representing an independent open marine radiation. Santanachelysgaffneyi is one of the most completely preserved early protostegids and is therefore critical for establishing the global phylogenetic position of the group. However, the single known specimen of this taxon is yet to be described in detail. Here we describe a second specimen of Santanachelysgaffneyi from its type horizon, the Romualdo Formation (late Aptian) of the Santana Group of the Araripe basin, NE Brazil. The skeletal elements preserved include the posterior part of the skull, neck vertebrae, shoulder girdle, anterior-most and left/central part of the carapace with few peripherals, and plastron lacking most of the hyoplastra. The remaining part of the carapace was apparently completed by fossil dealers using an anterior part of the pleurodiran Araripemydidae, tentatively identified as a shell portion of cf. Araripemysbarretoi, a more common Santana fossil turtle, among other indeterminate turtle shell fragments. We identified species and approximate size of prey items in 109 digital photographs of North American River Otters (Lontra canadensis) hunting in a 2-km stretch of South Fork Eel River, California, from January 2017 through December 2021. Over the 5-y study, 403 observations of River Otter social-group types and sizes were recorded, including singles, family groups with pups, and groups of up to 8 adults. An average of 3.0 River Otters per week (sx = 0.2, range 1 to 13 individuals) were observed in the study area. Eleven species of prey were taken by River Otters, including primarily, Sacramento Pikeminnow (Ptychocheilus grandis; 54.1% of 109), followed by crayfish spp. (24.8%), Pacific Lamprey (Entosphenus tridentatus; 10.1%), herpetofauna (6.4%), and less-frequently taken fish species (4.6%). Contingency table frequencies indicated Sacramento Pikeminnow and crayfish were taken more in months of low water flows in the river. Herpetofauna prey included 1 American Bullfrog (Lithobates catesbeinus), 2 Foothill Yellow-legged Frogs (Rana boylii), 1 Northwestern Pond Turtle (Actinemys marmorata), and 3 Rough-skinned Newts (Taricha granulosa), apparently with no ill effects to the otters. Sacramento Pikeminnows were captured by social groups of 2 and 3 or more River Otters more often than expected, and by single otters and family groups less often than expected; whereas crayfish were captured by single otters and family groups more often than expected, and by social groups of 2 and 3 or more, less often than expected. The Central American river turtle (Dermatemys mawii) is one of the of the most critically endangered turtle species worldwide, as a result of habitat loss and heavy hunting for consumption. Research on D. mawii becomes more challenging as wild populations are disappearing, while basic ecological data are still required to inform conservation efforts. In this study, our aim was to analyze the diet of D. mawii based on sex, and compare the information provided by stomach flushing and fecal samples. We examined both types of samples from individuals captured in 2017 and 2018 in a lentic system in the Lacantun River Basin, Chiapas, Mexico. We identified 3 broad categories (aquatic macrophytes, riparian resources and invertebrates) and 12 different food items, of which four are new reports of vascular plants for the diet of this turtle. The principal items consumed by D. mawii were Pistia stratiotes (a free-floating macrophyte) and Ludwigia sp. (a riparian plant). A low index of relative importance (≤ 0.5) for invertebrates suggests that they may be ingested accidentally when feeding on plants. We did not detect significant sex differences in the relative contribution of food items between the two sampling methods; additionally, we found a large dietary overlap (Cλ ≥ 0.89) between sexes that indicates a similar use of resources in this lentic system. We found all 3 broad food categories in stomach flushing samples and only one (riparian resources) in fecal samples; however, we did not find significant differences in the number of food items. With populations declining across their geographic range, the spotted turtle (Clemmys guttata) is currently a species of conservation concern. Though broadly distributed, the species is particularly enigmatic at its southern periphery, and many aspects of its ecology and population biology in this portion of the range have either just recently been described or are currently unknown. One of the current knowledge gaps is a robust assessment of the population genetics of the southern populations. We collected tissue samples from 204 spotted turtles from 5 sites across South Carolina, Georgia, and Florida and used 11 microsatellite loci to investigate the genetic diversity and population structure in these populations. We found that southern populations exhibited low, but significant, population differentiation (mean FST = 0.062) and each site clustered as its own genetic group. Genetic diversity across sites was comparable to estimates reported for northern populations. Net effective population sizes were generally robust and no populations showed indication of recent bottlenecks. Our results suggest that populations inhabiting relatively intact environments do not appear to face immediate threats from past loss of genetic diversity. However, continued monitoring, both demographic and genetic, of this long-lived species is an important management goal to insure that continued global changes do not threaten population viability. Sternotherus carinatus has been considered as a potential invasive species in Korea. However, the mitochondrial genome information of S. carinatus which can be used to control its effect on ecosystem is lacking. In this study, the complete mitochondrial genome of S. carinatus in Korea was sequenced and characterized. The mitochondrial genome consists of 37 genes (13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes) and a noncoding region. Phylogenetic analysis based on the mitochondrial genome sequences showed that S. carinatus from Korea is separated from other turtles which are the invasive species in Korea. Sequence divergence calculations indicated near-zero divergence between S. carinatus populations in Korea, the USA, and China, suggesting limited genetic differentiation. In the context of the broader issue of invasive species disrupting ecosystems, this research contributes to the identification of mitochondrial genomes for various freshwater turtle species, emphasizing the need for extended data collection to discern genetic mixing trends between native and non-native species. The complete mitochondrial genome of Chrysemys picta bellii in Korea was sequenced and characterized. The mitochondrial genome consists of 37 genes (13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes) and a noncoding region. Phylogenetic analysis based on the mitochondrial genome sequences revealed that C. p. bellii from Korea formed a cluster with C. p. bellii from China and C. picta from the USA, while showing clear separation from other turtle species within the C. picta cluster. This study presented the first complete mitochondrial genome from C. p. bellii in Korea, offering crucial information for managing invasive species and protecting the local ecosystem. The Mediterranean pond turtle Mauremys leprosa is widely distributed in Morocco. Significant variations could be observed in this species due to the fact that it occupies a vast and environmentally diverse area. Thus, the analysis of population parameters is crucial in elucidating the differences between populations. Differences between individuals may be linked to environmental factors, including many habitat characteristics. In the present study, variation in population structure, body condition, morphology and sexual dimorphism, were examined for the Sahara Desert blue-eyed turtle M. l. saharica in southern Morocco from seven distinct localities distributed in four different drainages in southern Morocco. A total of 224 turtles were captured, sexed, weighed and then measured for their carapace and tail dimensions before being released. Among the studied populations, four were dominated by adult individuals (Taakilt: 96%, Oued Guir: 87%, Oued Ziz: 75%, Sidi ElMehdaoui: 70%), two comprised 100% of adults (Oued Noun and Tarmigt) and one population consisted of 50% adults (Lower Draa). The sex ratio was significantly different between populations, being balanced in two populations (Tarmigt and Sidi El Mehdaoui), male-biased in three populations (Oued Zizi, Oued Noun, Lower Draa) and female-biased in the two remaining populations (Oued Guir and Taakilt). Body Condition Index (BCI, g/cm3) was significantly different among populations but not between sexes, or for the Sex × Locality interaction. The Sexual Size Dimorphism (SSD), females being larger than males, varies among population with the greatest degree observed in Oued Guir population (SSD = 0.41) and the lowest in Sidi El Mehdaoui population (SSD = -0.14). Length-Weight Relation (LWR) shows that M. l. saharica has isometric growth for both male and female of Oued Guir and Tarmigt, for female of Oued Ziz, Taakilt, Oued Noun and lower Draa and hypo-allometric for male of Oued Ziz, Taakilt, Oued Noun, Lower Draa and for both male and female of Sidi El Mehdaoui. Morphological variation of carapace is shown in this species associated with population variation among basins. As semi-aquatic species that use both terrestrial and aquatic habitats, freshwater turtles and their microbial communities are especially sensitive to the impacts of habitat disturbance. In this study, we use 16S rRNA amplicon sequencing to characterize the shell and cloacal bacterial communities of turtles in the San Francisco Bay Area. We captured western pond turtles (Actinemys/Emys marmorata) across eight sites located in urban and rural environments, along with invasive red-eared sliders (Trachemys scripta elegans). We assessed differences in western pond turtle bacterial communities diversity/composition between shell and cloacal samples and evaluated how alpha/beta diversity metrics were influenced by habitat quality. We found phylum-level bacterial taxonomic turnover in the bacterial communities of western pond turtles relative to the host tissue substrate samples. Our findings indicate that location identity elicits a high degree of lower-level (i.e. species/genus) bacterial taxonomic turnover. Further, we found that samples originating from good quality habitat had poorer shell bacterial communities but more diverse cloacal ones. The shell bacterial communities of red-eared sliders overlapped with those western pond turtles suggesting the existence of microbial dispersal between these two species. Our results add to our current understanding of turtle symbiont microbial ecology by establishing patterns of bacterial symbiont variation in an urban to rural gradient. [ABSTRACT FROM AUTHOR]

Bothremydidae was one of the most diverse and widespread group of side-necked turtles (pleurodirans) during the Cretaceous and part of the Paleogene. In South America, the Paleogene record of bothremydids is restricted to Puentemys mushaisaensis from the middle–late Paleocene Cerrejón Formation of Colombia, Inaechelys pernambucensis from the Paleocene of Brazil, and Motelomama olssoni from the early Eocene of Perú. Here, we describe two shells of P. mushaisaensis and several other isolated bones conferred to this taxon from the upper Paleocene and lower Eocene Arcillolitas de Socha Formation found in the Socha Region, Boyacá Department of Colombia. U-Pb dating of detrital zircon from two levels from this formation indicates maximum depositional ages of 56.83±0.04 Ma and 57.2±0.5 Ma for the areniscas guía interval of the formation. The new occurrence of P. mushaisaensis in the Socha region, at least 500 km south from Cerrejón, indicates a wider biogeographical distribution of northern South America Paleocene herpetofauna, possibly helped by low topography and ecosystems connectivity via a faunistic corridor.

The Asian box turtles of the Cuora galbinifrons complex (C. galbinifrons, C. bourreti, and C. picturata) rank among the most endangered chelonian species in the world. Despite several previous studies, the phylogenetic relationships and species boundaries of this complex remain a matter for dispute due to a shortage of field-collected samples for genetic validation, observed discordance between mitochondrial and nuclear markers, and reported intergradation zones combined with a strong tendency of hybridization in the genus. Here, we re-investigate the relationships and potential hybridization between the species of the C. galbinifrons complex based on the most comprehensive dataset to date consisting of 394 morphologically identified specimens (136 C. galbinifrons, 200 C. bourreti, 49 C. picturata, and nine individuals allegedly from Hainan). The turtles mainly came from assurance colonies as well as from zoological and private collections across the USA and Europe. Bayesian and Maximum Likelihood analyses of a concatenated mitochondrial dataset (COI and ND4 plus adjacent tRNA genes) yielded almost identical topologies, supporting three major clades corresponding to C. galbinifrons, C. bourreti, and C. picturata, respectively. In accordance with previous studies, C. bourreti represented the sister clade to C. galbinifrons and these two clades together were sister to C. picturata. Haplotype networks revealed pronounced mitochondrial divergences between the taxa. STRUCTURE and PCA analyses using 12 microsatellite loci also confirmed three distinct clusters that are in agreement with the recognized species. Only a few specimens with admixed ancestry (hybrids) or mismatched mitochondrial identity were revealed, suggesting extremely limited gene flow among the three species. Herpesviruses can be significant reptile pathogens. Herpesviral infection in a wild-caught, male spider tortoise (Pyxis arachnoides) under human care was detected during a routine wellness examination prior to transition between zoologic organizations. The tortoise had no clinical signs of illness. Oral swabs obtained during a physical examination as part of pre-shipment risk mitigation for infectious disease were submitted for consensus herpesvirus PCR assay and sequencing. Based on comparative sequence analysis, the novel herpesvirus identified is a member of the subfamily Alphaherpesvirinae. Studies of herpesviral phylogeny in chelonian species support branching patterns of turtle herpesviruses that closely mirror those of their hosts. The symmetry of these patterns is suggestive of close codivergence of turtle herpesviruses with their host species. The distribution of these viruses in both tortoises and emydids suggests a phylogenetic duplication event in the herpesviruses after host divergence of the Pleurodira and basal to the divergence of Americhelydia. Herpesviral infections have been documented to cause higher morbidity when introduced to aberrant host species, and significant consideration must be given to the presence of herpesviruses in the management of tortoise collections, particularly collections that include various species of testudines. The Maghreb population of Emys orbicularis extends over a narrow strip of northern Morocco, Algeria, and Tunisia. Our understanding of the presence of this turtle in Algeria, in contrast to Morocco and Tunisia, is quite limited and frequently rests on a few small-scale studies or even reports from a century ago. The present study provides the first complete data on the distribution and level of threat of this species in Algeria. A field survey was carried out based on the data obtained from scattered local reports and available studies that suggested the presence of the species in Algerian wetlands. Data on climatic and habitat preferences, the geographic elevation of inhabited localities, and their conservation status were collected. A total of 45 wetlands were inhabited by E. orbicularis, of which 40 were part of a well-interconnected eastern metapopulation, joined by an extensive hydrographic network. In some places, the species has persisted for over 130 years in the same wetland complex. However, the five remaining populations living in central North Algeria are fragmented, highly isolated, and therefore more threatened. With a few exceptions, turtles inhabit lowlands below 100 m above sea level. Detected sub-populations inhabit mostly stagnant waters (60%), sometimes dams and lakes (24%), and sporadically also slow-moving water bodies (16%). Altogether, the Algerian distribution of E. orbicularis extends to a total area of 2900 km2, a 40 km wide strip with a length of 490 km west of the Tunisian border. Although restricted to this narrow strip, the persistence of numerous populations in Algeria, especially in Numidia, testifies to their relatively better conservation status. In order to ensure the sustainability of these efforts, follow-up and monitoring activities should be suggested, with special attention paid to highly isolated populations to maintain their long-term viability. The spotted pond turtle (Geoclemys hamiltonii) is a threatened and less explored species endemic to Bangladesh, India, Nepal, and Pakistan. To infer structural variation and matrilineal phylogenetic interpretation, the present research decoded the mitogenome of G. hamiltonii (16,509 bp) using next-generation sequencing technology. The mitogenome comprises 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), two ribosomal RNAs (rRNAs), and one AT-rich control region (CR) with similar strand symmetry in vertebrates. The ATG was identified as a start codon in most of the PCGs except Cytochrome oxidase subunit 1 (cox1), which started with the GTG codon. The non-coding CR of G. hamiltonii was determined to have a unique structure and variation in different domains and stem-loop secondary structure as compared with other Batagurinae species. The PCGs-based Bayesian phylogeny inferred strong monophyletic support for all Batagurinae species and confirmed the sister relationship of G. hamiltonii with Pangshura and Batagur taxa. We recommend generating more mitogenomic data for other Batagurinae species to confirm their population structure and evolutionary relationships. In addition, the present study aims to infer the habitat suitability and habitat quality of G. hamiltonii in its global distribution, both in the present and future climatic scenarios. We identify that only 58,542 km2 (7.16%) of the total range extent (817,341 km2) is suitable for this species, along with the fragmented habitats in both the eastern and western ranges. Comparative habitat quality assessment suggests the level of patch shape in the western range is higher (71.3%) compared to the eastern range. Our results suggest a massive decline of approximately 65.73% to 70.31% and 70.53% to 75.30% under ssp245 and ssp585 future scenarios, respectively, for the years between 2021-2040 and 2061-2080 compared with the current distribution. The present study indicates that proper conservation management requires greater attention to the causes and solutions to the fragmented distribution and safeguarding of this endangered species in the Indus, Ganges, and Brahmaputra (IGB) river basins. Reeves' Turtles (also known as Chinese Three-Keeled Pond Turtles, Mauremys reevesii) exhibit temperature-dependent sex determination (TSD). Recent studies reported genome sequencing and described normal embryonic and gonadal developmental patterns in M. reevesii, making them a promising model for studying TSD. Nevertheless, development of other reproductive organs, such as the Müllerian duct, has not been described in this species. The Müllerian duct is a tubular structure that primarily forms in mesonephros during embryogenesis in both sexes. While the Müllerian duct further develops into the reproductive tract in females, it typically regresses in males. In this study, we investigated the development and regression of the Müllerian duct in M. reevesii during the latter half of embryogenesis under female- (FPT; 31°C) and male-producing temperatures (MPT; 26°C). Histological analysis revealed that the Müllerian duct showed the first sign of regression at the end of the temperature-sensitive period under MPT and significantly regressed at the prehatching stage (stage 24). Development and regression of the Müllerian duct in M. reevesii followed general trajectories reported in mouse and chicken studies. Timing of regression was consistent with reports in another TSD turtle, Pond Sliders (Trachemys scripta). We review in detail the published fossil record of turtles from Sardinia and, in addition, we document previously undescribed specimens for the first time. Among these undescribed specimens, is the oldest occurrence of Testudo hermanni on the island, from the Early Pleistocene of Monte Tuttavista. The turtle fossil record in Sardinia goes back to the Eocene and comprises 18 different taxa, pertaining to 6 lineages: Podocnemididae, Cheloniidae, Emydidae, Geoemydidae, Testudinidae, and Trionychidae. Remarkable is the occurrence of Eocene pleurodires, whose presence is in agreement with the Oligo-Miocene rifting of the Corso-Sardinian block. Interestingly, the fossil record provides evidence for the presence in the island of both Testudo hermanni and Emys orbicularis during the Pleistocene although according to molecular data the extant populations of these two taxa were introduced in recent times. Finally, a large ungual phalanx from the Middle–Late Pleistocene Monte San Giovanni bone breccia testifies the occurrence of a giant tortoise in the Quaternary terrestrial ecosystem of Sardinia. Simple Summary: The concept of this investigation emerged when a rapidly increased number of poached European pond turtles in poor health condition, with shell necrosis and massive skin hemorrhages, were temporarily put into a pond that belongs to a quarantine section at Belgrade Zoo. Using cytology as an initial tool, different parasitic stages of hemogregarines have been noticed in the blood smears of the examined animals. Cytological and microscopic examination of the samples proved to be sufficient for establishing the infection, but molecular analyses were used in examining the phylogeny of the blood parasites. Over the eight years (2015–2023) of monitoring, the number of diseased and dead turtles has decreased, which could be hypothetically attributed to the elimination of leeches as the definitive host. The eight-year study (2015–2023) was performed on a large sample of poached European pond turtles infected with Haemogregarina stepanowi and held in a pond that belongs to a quarantine section of Belgrade Zoo. The protected species of European pond turtles have been found in poor health, with general weakness, anorexia, and low motility. Comprehensive cytological, hematological, molecular, and postmortem evaluations have been performed. Initially, Diff Quick staining of the blood smears revealed rounded or elongated erythrocytes, often bearing premeront or U-shaped gamont of the hemogregarines inside. The reduced erythrocyte numbers, hemoglobin, and hematocrit values found in the examined population of infected turtles indicated anemia. Macroscopically, shell necrosis and massive skin hemorrhages were the most prominent findings observed in diseased turtles. Microscopically, the lungs, liver, kidneys, and spleen revealed hyperemia, hemorrhages, and the presence of parasitic stages in tissue samples in 31 of 40 necropsied turtles. Simple Summary: We explored the effect of maternal size on offspring size and number using a 4-year dataset based on paternity analyses in Asian yellow pond turtle, Mauremys mutica, and found that the maternal size significantly affected the offspring number but not offspring size. The offspring size was significantly correlated with maternal age. Our results suggest that the offspring size-number trade-off theory does not apply to cultured M. mutica and also provides basic data for the efficient artificial breeding management of M. mutica in captivity. Offspring size-number trade-off is a critical component of life-history theory and is important for further understanding the reproductive strategies of animals. The relationship between this trade-off and maternal size has been explored in several turtle species, except for the Asian yellow pond turtle, Mauremys mutica. To investigate how the maternal condition affects offspring size and number, we explored the relationships among the maternal body size and the number and size of cultured M. mutica hatchlings using a 4-year dataset. Our results showed that different females not only produced different sizes of offspring but also produced different numbers of offspring. No trade-off in egg size number was detected. According to regression analysis, we did not find that the maternal body size significantly influenced the offspring mass; however, we detected that the offspring size was significantly correlated with the clutch size and maternal age. The mean body mass of offspring increased with maternal age, and the clutch size varied significantly over four years, which was correlated with offspring size, maternal body size and age. However, the number of offspring per female increased with the maternal plastron length rather than age. Our results were inconsistent with the optimal offspring size theory in that females did not increase their offspring size but rather increased the offspring number to increase their fitness, which will also provide a basis for the efficient cultivation management of turtles. Large-scale mortality due to Aeromonas hydrophila (A. hydrophila) infection has considerably decreased the yield of the Chinese pond turtle (Mauremys reevesii). Purslane is a naturally active substance with a wide range of pharmacological functions, but its antibacterial effect on Chinese pond turtles infected by A. hydrophila infection is still unknown. In this study, we investigated the effect of purslane on intestinal morphology, digestion activity, and microbiome of Chinese pond turtles during A. hydrophila infection. The results showed that purslane promoted epidermal neogenesis of the limbs and increased the survival and feeding rates of Chinese pond turtles during A. hydrophila infection. Histopathological observation and enzyme activity assay indicated that purslane improved the intestinal morphology and digestive enzyme (α-amylase, lipase and pepsin) activities of Chinese pond turtle during A. hydrophila infection. Microbiome analysis revealed that purslane increased the diversity of intestinal microbiota with a significant decrease in the proportion of potentially pathogenic bacteria (such as Citrobacter freundii, Eimeria praecox, and Salmonella enterica) and an increase in the abundance of probiotics (such as uncultured Lactobacillus). Haemogregarines are blood parasites with a life-cycle involving a vertebrate as the intermediate host and an invertebrate as the definitive host and vector. Extensive phylogenetic investigations based on 18S-rRNA gene sequences have shown that Haemogregarina stepanowi (Apicomplexa: Haemogregarinidae) is able to infest a large diversity of freshwater turtle species, including the European pond turtle Emys orbicularis, the Sicilian pond turtle Emys trinacris, the Caspian turtle Mauremys caspica, the Mediterranean pond turtle Mauremys leprosa, and the Western Caspian turtle Mauremys rivulata, among others. From the same molecular markers, H. stepanowi is further considered to be a complex of cryptic species predisposed to infect the same host species. While Placobdella costata is known to be the unique vector of H. stepanowi, it is only recently that independent lineages within P. costata have been illustrated—suggesting the presence of at least five unique leech species across Western Europe. The aims of our study were therefore to investigate from mitochondrial markers (COI) the genetic diversity within haemogregarines and leeches infecting freshwater turtles of the Maghreb, in order to identify processes of parasite speciation. We showed that H. stepanowi consists of at least five cryptic species in the Maghreb, while two Placobella species were identified in the same area. Although an Eastern–Western speciation pattern was apparent for both leeches and haemogregarines, we cannot make definitive conclusions regarding co-speciation patterns between parasites and vectors. Chrysemys , commonly known as painted turtles, have the largest native biogeographic range of all North American turtles. The presence of a new species, Chrysemys corniculata sp. nov. , in the Late Hemphillian-Early Blancan North American Land Mammal Age (latest Miocene-Early Pliocene) of Tennessee provides further data on the evolution of Chrysemys , deirochelyines and emydids. The new fossil species lies basally in Deirochelyinae and suggests that either Chrysemys represents a basal deirochelyine morphology and is one of the oldest genera in the family, or that similar basal morphologies have evolved multiple times throughout deirochelyine evolution. Its occurrence at the same time as Chrysemys picta , during the Hemphillian-Early Blancan, a time of high biodiversity in emydid turtles, suggests either multiple species of Chrysemys during the Late Hemphillian-Early Blancan (at least one in the mid-west and one farther east), or multiple lineages with basal morphologies during this time. Early fossil deirochelyines occur after the greenhouse conditions of the Eocene and the Mid-Miocene Climatic Optimum. Vicariance led to deirochelyines becoming more speciose, including the occurrence of C. corniculata , after the Mid-Miocene Climatic Optimum, potentially suggesting cooler temperatures aided in the evolution of the subfamily and their speciation during the Hemphillian and into the Early Blancan. Aquatic habitats in the Great Plains of North America have increased risk of droughts under climate forecasts. Droughts have the potential to influence the population dynamics of pond turtles, and long-term studies are useful to assess the impact of climatic variation on turtles. We compiled twelve years of mark-recapture data for painted turtles (Chrysemys picta) captured in a pond in Keith County, Nebraska during 2005–2016 that included two periods of drought. We used a robust design analysis to investigate influences on population size, annual survival, temporary immigration, and capture probability. Estimates of the annual population size ranged from 92 (CI: 90–94) to 180 (CI: 175–186) but did not vary with drought conditions. Despite a relatively stable depth of water in our study pond, the probability of annual survival was reduced by 0.07 in females and 0.10 in males during drought years. Approximately one-fifth (temporary emigration probability: 0.19, CI = 0.16–0.23) of the population was outside the study pond at any given time. Our long-term research provides insights into the potential challenges to turtles in aquatic habitats undergoing prolonged changes in long-term climate conditions. [ABSTRACT FROM AUTHOR]
