Despite their importance for understanding consumer‐resource dynamics, the dietary responses of large terrestrial predators to variations in prey richness and competition pressure are unclear. While a greater predator selectivity along with increasing prey abundance would be expected under an optimal foraging scenario, there is some evidence that predators may broaden their diet where there is a greater resource diversity. Furthermore, the use of large prey may be limited by increasing presence of competitors.We considered three widespread large carnivores (the grey wolf Canis lupus, the puma Puma concolor and the leopard Panthera pardus), whose distribution range encompasses different continents, with different communities of prey/competitors. We expected that the potential to modulate their use of large prey according to prey richness would vary according to different levels of potential competition.We collated data from more than 240 studies of the diets of wolf, puma and leopard to model whether the relationships between the diversity of used large prey (i.e. the Large Prey Index) and prey richness was modulated by carnivore richness, in different continents.The wolf showed an increase in the Large Prey Index with prey richness across its distribution range, where it is usually the apex predator in areas from which data are available. Conversely, the leopard showed this pattern in Asia, but not in Africa, where it often coexists with a greater array of potential competitors. For the puma, the Large Prey Index increased with prey richness throughout its distribution range, except in the areas where the larger and dominant jaguar also occurred.By emphasising the complex relationships between prey richness and predator diets, our results testify to the suppressive effects of larger competitors over the use of large prey by subordinate carnivores. Determining the abundance of cryptic carnivores is central to building successful conservation management to mitigate conflicts and support coexistence strategies. For these reasons, there is considerable investment in developing reliable, cost‐effective tools for estimating the abundance of wildlife. Nevertheless, field‐based comparisons of abundance methods remain uncommon, even while essential to refining methods and coming to consensus around best practices. Here, we compare two approaches still being tested in real‐world application for an emblematic puma (Puma concolor) population in the Torres del Paine UNESCO Biosphere Reserve in southern Chile: (1) the unmarked estimator, space‐to‐event model (STE), which utilizes photographs gathered with camera traps, and (2) the genotype spatial partial identity model (gSPIM), which is an adaptation of the more established spatially explicit genetic capture‐recapture method (SECR) based on genetic data extracted from scats collected in systematic surveys. We show the tremendous variation in resulting STE estimates depending upon the start time of the analysis and length of the sampling window, and showcase a refined iterative sampling approach in a Bayesian framework to both utilize the full camera data and to stabilize density estimates for a given sampling window. Across all sampling, estimates from the STE model ranged from 3.19 (1.6–5.1 representing 10th and 90th percentile of credible intervals) to 7.38 (3.3–11.6) independent pumas 100 km−2. By comparison, our gSPIM model estimated 5.1 independent pumas 100 km−2 (excluding kittens) (with credible intervals of 2.2–10.3). Neither method was compared with any known density to determine their accuracy. Nevertheless, we provide initial density estimates to guide conservation strategies for wildlife agencies and local communities overseeing and hosting nascent puma tourism and livestock ranching, as well as guidelines for the use of these methods for any wildlife species. The widespread eradication of large carnivores and subsequent expansion of top mesopredators has the potential to impact species and community interactions with ecosystem‐wide implications. An example of these trophic dynamics is the widespread establishment of coyotes following extirpation of wolves and mountain lions in eastern North America. Here, we examined occupancy of three carnivores in northern New York considering both environmental/habitat factors and interspecific interactions. We estimated the co‐occurrence of coyotes, fishers, and martens from a landscape‐scale winter camera trap survey repeated annually for three years. Martens occurred independently of both coyotes and fishers, while fishers and coyotes displayed positive intraguild interactions that were constant across the landscape. Both marten and fisher first‐order occupancy were driven by a combination of biotic and abiotic factors, with both species displaying positive associations with forest cover but antithetical responses to average snow depth. The integral and antithetical role of snow depth in driving the occurrence of martens (positive) and fishers (negative) in the landscape indicates that future climatic warming could reduce the availability of current spatial refuges for martens created by severe winter conditions. Climate‐driven alterations to established competitive interactions and co‐existence patterns between marten and fisher have critical implications for the species survival and conservation. We provide correlational evidence consistent with the potential for positive top–down effects of dominant mesocarnivores on subordinate species, with fisher occupancy increasing conditional on the presence of coyotes across the landscape. These findings align with the hypothesis that under certain conditions, coyotes may facilitate certain subordinate carnivores. The evidence produced here is consistent with hypotheses on the dynamic nature of trophic niches. We demonstrate the need to consider the interplay between climate, habitat, and interspecific interactions to understand wildlife occupancy patterns and inform wildlife management in a rapidly changing world. Modification of food webs is a frequent cause of shifts in ecosystem states that resist reversal when the food web is restored to its original condition. We used the restoration of the large carnivore guild including gray wolves (Canis lupis), cougars (Felis concolor), and grizzly bears (Ursus arctos horribilis) to the northern range of Yellowstone National Park as a model system to understand how ecosystems might resist reconfiguration after the restoration of apex predators to the food web. The absence of wolves, cougars, and grizzly bears for nearly a century from the northern range was the primary cause of dramatic changes in riparian plant communities. Willows (Salix spp.) were suppressed in height by intense browsing by the dominant herbivore, elk (Cervus canadensis). The loss of activity by beavers (Castor canadensis) coincided with the loss of tall willows. We hypothesized that intense elk browsing interrupted the mutualism between willow and beavers: ecosystem engineering by beavers was a critical component of willow habitat and tall willows were a critical component of habitat for beavers. This interruption made riparian communities resilient to the disturbance caused by the restoration of apex predators. We hypothesized further that reductions in elk browsing attributable to reductions in elk population size were not sufficient to prevent the suppression of willow growth. To test these hypotheses, we conducted a 20‐year, factorial experiment that crossed simulated beaver dams with the exclusion of browsing. We found that willows grew to heights expected for restored communities only in the presence of dams and reduced browsing. Willows experiencing ambient conditions remained well below this expectation. We found no difference in heights or growth rates of willows in experimental controls and willows in 21 randomly chosen sites, confirming that the results of the experiment were representative of range‐wide conditions. A reorganized community of large herbivores was implicated in the suppression of willow growth. We conclude that the restoration of large carnivores to the food web failed to restore riparian plant communities on Yellowstone's northern range, supporting the hypothesis that this ecosystem is in an alternative stable state caused primarily by the extirpation of apex predators during the early 20th century. Papillomaviruses (PV) infect epithelial cells and can cause hyperplastic or neoplastic lesions. In felids, most described PVs are from domestic cats (Felis catus; n = 7 types), with one type identified in each of the five wild felid species studied to date (Panthera uncia, Puma concolor, Leopardus wiedii, Panthera leo persica and Lynx rufus). PVs from domestic cats are highly diverse and are currently classified into three genera (Lambdapapillomavirus, Dyothetapapillomavirus, and Taupapillomavirus), whereas those from wild felids, although diverse, are all classified into the Lambdapapillomavirus genus. In this study, we used a metagenomic approach to identify ten novel PV genomes from rectal swabs of five deceased caracals (Caracal caracal) living in the greater Cape Town area, South Africa. These are the first PVs to be described from caracals, and represent six new PV types, i.e., Caracal caracal papillomavirus (CcarPV) 1–6. These CcarPV fall into two phylogenetically distinct genera: Lambdapapillomavirus, and Treisetapapillomavirus. Two or more PV types were identified in a single individual for three of the five caracals, and four caracals shared at least one of the same PV types with another caracal. After ~100 years of absence, beaver (Castor canadensis) were reintroduced to the San Pedro River in southeastern Arizona in 1999. Total streamflow in the San Pedro River has decreased during the past 90 years. We hypothesize that beaver under drought conditions may be limited in mobility due to low water and predation risk. Between 12 May and 16 June 2022, we used Reconyx Hyperfire 2 trail cameras to capture the behavior of beaver near a large dam on the San Pedro River. The site where the beaver built their dam has intermittent water flow and typically dries out every year during periods of low precipitation and high temperature. Beaver activity increased shortly before and after the pond dried out on 29 May 2022 (from 26 May to 31 May). We documented 7 separate occasions when beaver dragged large willow branches (~1 to 3 m long) with leaves away from the dam toward a presumed bank lodge. Beaver were only observed twice (both instances on 5 June) during the following 15 days of the study. This evidence suggests that beaver could stay in bank lodges during periods of low water flow, eating from their food caches. This unique caching behavior may be seen in beaver populations in desert climates or other drought-stressed environments. Mountain lions (Puma concolor) were observed at our study site before and after the pond dried, suggesting that this food-caching behavior has real value to the survival of beaver in the San Pedro River. The allosteric protein MCL-1 and its natural inhibitors, the BH3-only proteins PUMA, BIM, and NOXA regulate apoptosis by interacting promiscuously within an entangled binding network. Little is known about the transient processes and dynamic conformational fluctuations that are the basis for the formation and stability of the MCL-1/BH3-only complex. In this study, we designed photoswitchable versions of MCL-1/PUMA and MCL-1/NOXA, and investigated the protein response after an ultrafast photo-perturbation with transient infrared spectroscopy. We observed partial α-helical unfolding in all cases, albeit on strongly varying timescales (1.6 ns for PUMA, 9.7 ns for the previously studied BIM, and 85 ns for NOXA). These differences are interpreted as a BH3-only-specific "structural resilience" to defy the perturbation while remaining in MCL-1's binding pocket. Thus, the presented insights could help to better understand the differences between PUMA, BIM, and NOXA, the promiscuity of MCL-1, in general, and the role of the proteins in the apoptotic network. Many large carnivores, despite widespread habitat alteration, are rebounding in parts of their former ranges after decades of persecution and exploitation. Cougars (Puma concolor) are apex predator with their remaining northern core range constricted to mountain landscapes and areas of western North America; however, cougar populations have recently started rebounding in several locations across North America, including northward in boreal forest landscapes. A camera‐trap survey of multiple landscapes across Alberta, Canada, delineated a range edge; within this region, we deployed an array of 47 camera traps in a random stratified design across a landscape spanning a gradient of anthropogenic development relative to the predicted expansion front. We completed multiple hypotheses in an information‐theoretic framework to determine if cougar occurrence is best explained by natural land cover features, anthropogenic development features, or competitor and prey activity. We predicted that anthropogenic development features from resource extraction and invading white‐tailed deer (Odocoileus virgianius) explain cougar distribution at this boreal range edge. Counter to our predictions, the relative activity of native prey, predominantly snowshoe hare (Lepus americanus), was the best predictor of cougar occurrence at this range edge. Small‐bodied prey items are particularly important for female and sub‐adult cougars and may support breeding individuals in the northeast boreal forest. Also, counter to our predictions, there was not a strong relationship detected between cougar occurrence and gray wolf (Canis lupus) activity at this range edge. However, further investigation is recommended as the possibility of cougar expansion into areas of the multi‐prey boreal system, where wolves have recently been controlled, could have negative consequences for conservation goals in this region (e.g. the recovery of woodland caribou [Rangifer tarandus caribou]). Reintroduction is an important tool for the recovery of imperiled species. For threatened Pacific salmonids (Oncorhynchus spp.) species, hatchery‐origin (HOR) individuals from a nearby source are often used to reestablish populations in vacant, historically occupied habitat. However, this approach is challenged by the relatively low reproductive success that HOR Pacific salmonids experience when they spawn in the wild, relative to their natural‐origin (NOR) counterparts. In this study, we used genetic parentage analysis to compare the reproductive success of three groups of adult Chinook salmon (Oncorhynchus tshawytscha) reintroduced above Cougar Dam on the South Fork McKenzie River, Oregon: HOR Chinook salmon from an integrated stock; first‐generation, wild‐born descendants (hereafter F1s) of Chinook salmon produced at the same hatchery; and NOR Chinook salmon that are presumed to have been produced below the dam, on the mainstem McKenzie River, or elsewhere and volitionally entered a trap below Cougar Dam. We found that F1s produced nearly as many adult offspring as NORs, and 1.8‐fold more adult offspring than HORs. This result suggests that, for the South Fork McKenzie reintroduction program, a single generation in the wild increases fitness for the descendants of HOR Chinook salmon. Mesocarnivores face interspecific competition and risk intraguild predation when sharing resources with apex carnivores. Within a landscape, carnivores across trophic levels may use the same communication hubs, which provide a mix of risks (injury/death) and rewards (gaining information) for subordinate species. We predicted that mesocarnivores would employ different strategies to avoid apex carnivores at shared communication hubs, depending on their trophic position. To test our prediction, we examined how different subordinate carnivore species in the Santa Cruz Mountains of California, USA, manage spatial overlap with pumas (Puma concolor), both at communication hubs and across a landscape-level camera trap array. We estimated species-specific occurrence, visitation rates, temporal overlap, and Avoidance–Attraction Ratios from camera traps and tested for differences between the two types of sites. We found that mesocarnivores generally avoided pumas at communication hubs, and this became more pronounced when pumas scent-marked during their most recent visit. Coyotes (Canis latrans), the pumas' closest subordinate competitor in our system, exhibited the strongest avoidance at communication hubs. Gray foxes (Urocyon cinereoargenteus) avoided pumas the least, which may suggest possible benefits from pumas suppressing coyotes. Overall, mesocarnivores exhibited various spatiotemporal avoidance strategies at communication hubs rather than outright avoidance, likely because they benefit from information gained while 'eavesdropping' on puma activity. Variability in avoidance strategies may be due to differential predation risks, as apex carnivores often interact more aggressively with their closest competitors. Combined, our results show how apex carnivores trigger complex species interactions across the entire carnivore guild and how trophic position determines behavioral responses and subsequent space use of subordinate mesocarnivores across the landscape. The increasing frequency and severity of human‐caused fires likely have deleterious effects on species distribution and persistence. In 2020, megafires in the Brazilian Pantanal burned 43% of the biome's unburned area and resulted in mass mortality of wildlife. We investigated changes in habitat use or occupancy for an assemblage of eight mammal species in Serra do Amolar, Brazil, following the 2020 fires using a pre‐ and post‐fire camera trap dataset. Additionally, we estimated the density for two naturally marked species, jaguars Panthera onca and ocelots Leopardus pardalis. Of the eight species, six (ocelots, collared peccaries Dicotyles tajacu, giant armadillos Priodontes maximus, Azara's agouti Dasyprocta azarae, red brocket deer Mazama americana, and tapirs Tapirus terrestris) had declining occupancy following fires, and one had stable habitat use (pumas Puma concolor). Giant armadillo experienced the most precipitous decline in occupancy from 0.431 ± 0.171 to 0.077 ± 0.044 after the fires. Jaguars were the only species with increasing habitat use, from 0.393 ± 0.127 to 0.753 ± 0.085. Jaguar density remained stable across years (2.8 ± 1.3, 3.7 ± 1.3, 2.6 ± 0.85/100 km2), while ocelot density increased from 13.9 ± 3.2 to 16.1 ± 5.2/100 km2. However, the low number of both jaguars and ocelots recaptured after the fire period suggests that immigration may have sustained the population. Our results indicate that the megafires will have significant consequences for species occupancy and fitness in fire‐affected areas. Prey selection is a fundamental aspect of ecology that drives evolution and community structure, yet the impact of intraspecific variation on the selection for prey size remains largely unaccounted for in ecological theory. Here, we explored puma (Puma concolor) prey selection across six study sites in North and South America. Our results highlighted the strong influence of season and prey availability on puma prey selection, and the smaller influence of puma age. Pumas in all sites selected smaller prey in warmer seasons following the ungulate birth pulse. Our top models included interaction terms between sex and age, suggesting that males more than females select larger prey as they age, which may reflect experiential learning. When accounting for variable sampling across pumas in our six sites, male and female pumas killed prey of equivalent size, even though males are larger than females, challenging assumptions about this species. Nevertheless, pumas in different study sites selected prey of different sizes, emphasizing that the optimal prey size for pumas is likely context‐dependent and affected by prey availability. The mean prey weight across all sites averaged 1.18 times mean puma weight, which was less than predicted as the optimal prey size by energetics and ecological theory (optimal prey = 1.45 puma weight). Our results help refine our understanding of optimal prey for pumas and other solitary carnivores, as well as corroborate recent research emphasizing that carnivore prey selection is impacted not just by energetics but by the effects of diverse ecology. Large animals could be important drivers of spatial nutrient subsidies when they ingest resources in some habitats and release them in others, even moving nutrients against elevational gradients. In high Andean deserts, vicuñas (Vicugna vicugna) move daily between nutrient-rich wet meadows, where there is abundant water and forage but high risk of predation by pumas (Puma concolor), and nutrient-poor open plains with lower risk of predation. In all habitats, vicuñas defecate and urinate in communal latrines. We investigated how these latrines impacted soil and plant nutrient concentrations across three habitats in the Andean ecosystem (meadows, plains, and canyons) and used stable isotope analysis to explore the source of fecal nutrients in latrines. Latrine soils had higher concentrations of nitrogen, carbon, and other nutrients than did nonlatrine soils across all habitats. These inputs corresponded with an increase in plant quality (lower C:N) at latrine sites in plains and canyons, but not in meadows. Stable isotope mixing models suggest that ~7% of nutrients in plains latrines originated from vegetation in meadows, which is disproportionately higher than the relative proportion of meadow habitat (2.6%) in the study area. In contrast, ~68% of nutrients in meadow latrines appear to originate from plains and canyon vegetation, though these habitats made up nearly 98% of the study area. Vicuña diel movements thus appear to concentrate nutrients in latrines within habitats and to drive cross-habitat nutrient subsidies, with disproportionate transport from low-lying, nutrient-rich meadows to more elevated, nutrient-poor plains. When these results are scaled up to the landscape scale, the amount of nitrogen and phosphorus subsidized in soil at plains latrines was of the same order of magnitude as estimates of annual atmospheric nitrogen and phosphorus deposition for this region (albeit far more localized and patchy). Thus, vicuña-mediated nutrient redistribution and deposition appears to be an important process impacting ecosystem functioning in arid Andean environments, on par with other major inputs of nutrients to the system. Chronic wasting disease (CWD) is a highly contagious, fatal neurodegenerative disease caused by infectious prions (PrPCWD) affecting wild and captive cervids. Although experimental feeding studies have demonstrated prions in feces of crows (Corvus brachyrhynchos), coyotes (Canis latrans), and cougars (Puma concolor), the role of scavengers and predators in CWD epidemiology remains poorly understood. Here we applied the real-time quaking-induced conversion (RT-QuIC) assay to detect PrPCWD in feces from cervid consumers, to advance surveillance approaches, which could be used to improve disease research and adaptive management of CWD. We assessed recovery and detection of PrPCWD by experimental spiking of PrPCWD into carnivore feces from 9 species sourced from CWD-free populations or captive facilities. We then applied this technique to detect PrPCWD from feces of predators and scavengers in free-ranging populations. Our results demonstrate that spiked PrPCWD is detectable from feces of free-ranging mammalian and avian carnivores using RT-QuIC. Results show that PrPCWD acquired in natural settings is detectable in feces from free-ranging carnivores, and that PrPCWD rates of detection in carnivore feces reflect relative prevalence estimates observed in the corresponding cervid populations. Summary: Treating insomnia during pregnancy improves sleep and depressed mood. However, given well‐established links between poor sleep and a broad spectrum of adverse maternal outcomes, the benefits of insomnia care may reach beyond sleep and depression. The present study evaluated the preliminary efficacy of 'Perinatal Understanding of Mindful Awareness for Sleep' (PUMAS)—a mindfulness sleep programme tailored to pregnancy that combines behavioural sleep strategies and meditation—for enhancing everyday mindfulness and maternal–fetal attachment, as well as for alleviating anxiety, repetitive thinking, and sleep‐related daytime impairment. We conducted a secondary analysis of a single‐arm proof‐of‐concept trial of 11 pregnant women with fifth edition of the Diagnostic and Statistical Manual of Mental Disorders diagnosed insomnia disorder who completed PUMAS (six sessions), which was delivered in an individual format via telemedicine video. Pre‐ and post‐treatment outcomes included the Cognitive and Affective Mindfulness Scale‐Revised (CAMS‐R), Maternal–Fetal Attachment Scale (MFAS), Generalised Anxiety Disorder seven‐item survey (GAD‐7), Perseverative Thinking Questionnaire (PTQ), Daytime Insomnia Symptoms Response Scale (DISRS), and the Patient‐Reported Outcomes Measurement Information System Sleep‐Related Impairment Scale (PROMIS‐SRI). Symptom changes were evaluated with paired‐samples t tests. Results showed PUMAS patients reported large increases in CAMS‐R (Cohen's dz = 1.81) and medium‐large increases in MFAS scores (Cohen's dz = 0.73). Moreover, PUMAS patients reported large reductions in scores on the GAD‐7 (Cohen's dz = 1.09), PTQ (Cohen's dz = 1.26), DISRS (Cohen's dz = 1.38), and PROMIS‐SRI (Cohen's dz = 1.53). Preliminary evidence suggests that a mindfulness‐based perinatal sleep programme may benefit several domains of maternal wellbeing beyond sleep and depression. PUMAS substantially enhanced patient ratings of everyday mindfulness and maternal–fetal attachment, while reporting alleviations in anxiety, perseverative thinking, insomnia‐focused rumination, and sleep‐related daytime impairment. Artificial light at night (ALAN) is increasing in extent and intensity across the globe. It has been shown to interfere with animal sensory systems, orientation and distribution, with the potential to cause significant ecological impacts. We analysed the locations of 102 mountain lions (Puma concolor) in a light-polluted region in California. We modelled their distribution relative to environmental and human-disturbance variables, including upward radiance (nearby lights), zenith brightness (sky glow) and natural illumination from moonlight. We found that mountain lion probability of presence was highly related to upward radiance, that is, related to lights within approximately 500 m. Despite a general pattern of avoidance of locations with high upward radiance, there were large differences in degree of avoidance among individuals. The amount of light from artificial sky glow was not influential when included together with upward radiance in the models, and illumination from moonlight was not influential at all. Our results suggest that changes in visibility associated with lunar cycles and sky glow are less important for mountain lions in their selection of light landscapes than avoiding potential interactions with humans represented by the presence of nearby lights on the ground. There is an increasing interest in the use of noninvasive monitoring methods, such as the use of chemical profiles obtained from noninvasive samples. Here, we propose the use of carboxylic and sulfonic acids profiles obtained from scats as a noninvasive tool to identify mammalian carnivoran species and for sex determination. We used High-Performance Liquid Chromatography (HPLC) coupled to a Laser-Induced Fluorescence (LIF) detection system (HPLC-LIF) to retrieve carboxylic and sulfonic acids profiles from scats of five mammalian carnivores: Puma concolor, Herpailurus yagouaroundi, Leopardus pardalis and Felis catus (all Felidae) and Nasua nasua (Procyonidae). We were able to unambiguously identify the species and determinate the sex for all fecal samples through the pattern of presence-absence of carboxylic and sulfonic acids (digital coding). The method allowed the discrimination of the evaluated species of the Carnivora, even between closely related and ecologically similar felids. Moreover, our method is low cost and can be executed in a short period of time. Further investigation is necessary to evaluate if the method's discriminatory power is applicable to other mammalian species. Cognitive-behavioral therapy is effective for prenatal insomnia, but unresolved cognitive arousal limits patient outcomes. Therapies aimed at reducing cognitive arousal may benefit pregnant women with insomnia. This proof-of-concept trial evaluated Perinatal Understanding of Mindful Awareness for Sleep (PUMAS, which combines mindfulness with behavioral sleep strategies) on insomnia, depression, and cognitive arousal. A single-arm trial of 12 pregnant women with DSM-5 insomnia disorder (n = 5/12 with comorbid depression) who received six sessions of PUMAS delivered individually via telemedicine. Pretreatment and posttreatment outcomes included the insomnia severity index (ISI), Edinburgh postnatal depression scale (EPDS), pre-sleep arousal scale's cognitive factor (PSASC; nocturnal cognitive arousal), perinatal-focused rumination (appended to PSASC), and Glasgow sleep effort scale. Eleven of 12 patients completed all sessions. Intent-to-treat analyses revealed a 10.83-point reduction in ISI (Cohen's dz = 3.05), resulting in 83.3% insomnia remission. PUMAS produced large reductions in EPDS (Cohen's dz = 2.76 in depressed group), resulting in all five baseline depressed patients remitting from depression. PUMAS produced large reductions in nocturnal cognitive arousal, perinatal-focused rumination, and sleep effort (all Cohen's dzs>2.00). Patients were highly satisfied with PUMAS and identified the telemedicine format and meditation app as positive features of its delivery. Patients rated sleep restriction and guided meditations as the most helpful treatment components. Prenatal insomnia patients were highly engaged in PUMAS, which produced large acute reductions in insomnia, depression, and cognitive arousal. These findings support the concept and feasibility of PUMAS for pregnant women with insomnia who present with or without comorbid depression. We examined livestock producers' perceptions of wolves, grizzly bears, black bears, and mountain lions, as well as their experiences with using nonlethal methods to protect livestock from depredation. Tolerance levels for black bears and mountain lions were relatively higher, with no respondents preferring removal of mountain lions, and only one preferring removal of black bears, but only in the area surrounding his/her operation. Shrinking habitat, depleted prey sources, and hunting increase conflict between humans and jaguars in Latin America. Participant observation was used for 131 open-ended interviews in the Mesoamerican Biological Corridor of Costa Rica to describe and provide recommendations for Panthera's rancher outreach program. Changing husbandry practices is different than wanting to relocate jaguars or pumas, which suggests willingness to coexist is based on geo-physical proximities. Further, perceived attacks on livestock may be unreliable creating a perception of more predation, villainizing large felines, and challenging coexistence. This study urges wildlife managers to evaluate the effectiveness of relocation; suggests systematic recordkeeping of jaguar and puma attacks; encourages researchers to measure willingness to co-exist with large carnivores based on geo-physical distance; suggests strategies of coexistence may have both cultural and regional differences; and recommends a communication strategy through a citizen science approach, in order to educate ranchers and create social investment among communities. To enable long-term coexistence, species need to differentiate at least one of the three main dimensions of the ecological niche (temporal, spatial, or trophic dimension). Here, we investigated whether mammalian predators (Chrysocyon brachyurus, Cerdocyon thous, Lycalopex vetulus, and Puma concolor) follow the prediction of trophic niche partitioning, which is expected when partitioning of food resources represents an important mechanism for coexistence. We predicted low niche overlap in general and low between P. concolor and the other species. We analyzed 207 fecal samples collected at a landscape composed of forest remnants immersed in Eucalyptus plantations. Food items (animals and plants) were identified using exoskeletons, feathers, scales, teeth, hair, and seeds. We calculated the frequency and percentage of occurrence of food items, niche breadth, and niche overlap between pairs of species. Prey size was similar among all predators, consuming mainly small-sized prey (<1 kg). However, niche breadth was larger for smaller carnivores compared to larger ones. No species pair showed significantly lower niche overlap than expected by chance. Our study provided detailed information on trophic resource use of sympatric carnivores, showing that trophic niche partitioning seems not to be crucial for the coexistence of carnivores in the study area. Pumas (Puma concolor) are solitary large carnivores that exhibit high energetic investments while hunting prey that often take multiple days to consume. Therefore, pumas should behave in a way to maximize their energetic gains, including using caching, which is a behavior used by many mammal species to preserve and store food or to conceal it from conspecifics and scavengers to limit their losses. Yet pumas do not always cache their kills. In order to understand caching behavior, we used variables associated with the kills such as prey mass, search time, climate, and habitat to test 20 ecological models (representing four a priori hypotheses: food perishability, resource pulse, consumption time, and kleptoparasitism deterrence) in an information-theoretic approach of model selection to explore factors related to the caching behavior. Models were run with information from tracked radio-collared pumas in California over a 2.5-year period and identified a total of 352 kills. Overall, we documented pumas caching 61.5% of their kills, including 71.6% of Black-tailed Deer (Odocoileus hemionus columbianus), their primary prey in the study area. The model with a quadratic effect of adjusted mass of prey on puma caching probability had all of the empirical support (w = 1.00). Specifically, pumas were most likely to cache intermediate-sized prey, such as yearling and adult female deer, and also fed from cached kills for longer periods of time. Larger prey may be too large to easily cache, making it less energetically efficient—while small prey can often be consumed quickly enough to not require caching. This suggests that intermediate-sized prey may be the optimal size for caching, allowing a puma to feed for multiple days while not greatly increasing energetic output. The hypotheses we tested were not mutually exclusive and pumas caching their prey may occur for several reasons; nevertheless, our study demonstrated that pumas use caching to extend their foraging time and maximize energetic gains when preying on intermediate-sized prey. Wildlife species may shift towards more nocturnal behavior in areas of higher human influence, but it is unclear how consistent this shift might be. We investigated how humans impact large mammal diel activities in a heavily recreated protected area and an adjacent university-managed forest in southwest British Columbia, Canada. We used camera trap detections of humans and wildlife, along with data on land-use infrastructure (e.g., recreation trails and restricted-access roads), in Bayesian regression models to investigate impacts of human disturbance on wildlife nocturnality. We found moderate evidence that black bears (Ursus americanus) were more nocturnal in response to human detections (mean posterior estimate = 0.35, 90% credible interval = 0.04 to 0.65), but no other clear relationships between wildlife nocturnality and human detections. However, we found evidence that coyotes (Canis latrans) (estimates = 0.81, 95% CI = 0.46 to 1.17) were more nocturnal and snowshoe hares (Lepus americanus) (estimate = -0.87, 95% CI = -1.29 to -0.46) were less nocturnal in areas of higher trail density. We also found that coyotes (estimate = -0.87, 95% CI = -1.29 to -0.46) and cougars (Puma concolor) (estimate = -1.14, 90% CI = -2.16 to -0.12) were less nocturnal in areas of greater road density. Furthermore, coyotes, black-tailed deer (Odocoileus hemionus), and snowshoe hares were moderately more nocturnal in areas near urban-wildland boundaries (estimates and 90% CIs: coyote = -0.29, -0.55 to -0.04, black-tailed deer = -0.25, -0.45 to -0.04, snowshoe hare = -0.24, -0.46 to -0.01). Our findings imply anthropogenic landscape features may influence medium to large-sized mammal diel activities more than direct human presence. While increased nocturnality may be a promising mechanism for human-wildlife coexistence, shifts in temporal activity can also have negative repercussions for wildlife, warranting further research into the causes and consequences of wildlife responses to increasingly human-dominated landscapes. Cloud microphysics is one of the most time‐consuming components in a climate model. In this study, we port the cloud microphysics parameterization in the Community Atmosphere Model (CAM), known as Parameterization of Unified Microphysics Across Scales (PUMAS), from CPU to GPU to seek a computational speedup. The directive‐based methods (OpenACC and OpenMP target offload) are determined as the best fit specifically for our development practices, which enable a single version of source code to run either on the CPU or GPU, and yield a better portability and maintainability. Their performance is first examined in a PUMAS stand‐alone kernel and the directive‐based methods can outperform a CPU node as long as there is enough computational burden on the GPU. A consistent behavior is observed when we run PUMAS on the GPU in a practical CAM simulation. A 3.6× speedup of the PUMAS execution time, including data movement between CPU and GPU, is achieved at a coarse horizontal resolution (8 NVIDIA V100 GPUs against 36 Intel Skylake CPU cores). This speedup further increases up to 5.4× at a high resolution (24 NVIDIA V100 GPUs against 108 Intel Skylake CPU cores), which highlights the fact that GPU favors larger problem size. This study demonstrates that using GPU in a CAM simulation can save noticeable computational costs even with a small portion of code being GPU‐enabled. Therefore, we are encouraged to port more parameterizations to GPU to take advantage of its computational benefit. Plain Language Summary: Cloud microphysics is an important process in the atmosphere that describes the formation, growth, evaporation, and melting of cloud and precipitation particles. Therefore, it is a critical component for a weather or climate model to produce a reasonable prediction of cloud and precipitation. Due to its complexity, it is typically time‐consuming to compute the cloud microphysics. In this study, we enable the execution of cloud microphysics on the graphics processing unit (GPU), which is a processor that can run more computations simultaneously than a traditional CPU. There are multiple ways to program on GPU and we select the directive‐based method. It allows the same code to run on either the CPU or GPU without changing the original programming language. We demonstrate that by porting the cloud microphysics to GPU, we are able to the achieve noticeable speedup against CPU. Furthermore, the speedup is higher if we could take advantage of multiple GPUs at the same time or lay a heavier computation on each GPU. While Misiones, Argentina contains one of the largest remnants of Upper Paraná Atlantic Forest ecoregion, one of the world's biodiversity hotspots, only ~50% of this native forest is protected. Each protected area is at risk of becoming an island of native forest surrounded by a matrix of altered habitats due to ongoing land conversion. In an effort to maximize long-term connectivity between existing protected areas, DeMatteo [1] used a multifaceted cost analysis to determine the optimal location for the region's first multispecies corridor using noninvasive data on jaguars (Panthera onca), pumas (Puma concolor), ocelots (Leopardus pardalis), southern tiger cats (Leopardus guttulus), and bush dogs (Speothos venaticus). This work builds on this framework by integrating new field data that broadens the scope of species-specific data across the region's heterogeneous landscape, which varies in vegetation, disturbance, human proximity, and protective status. In addition, two different land use layers are compared across the distributions of the five carnivores, the overlap in their independent distributions, and their relationship to the multispecies corridor. Interpretation of these land use data to species-specific habitat suitability goes beyond DeMatteo [1], with a subdivision of suitability into marginal and optimal areas. This refined scale allows a reanalysis of key areas in the multispecies corridor, where connectivity was previously defined as at highly-at-risk, allowing for a more directed development of management strategies. These analyses and their interpretation extend beyond northern-central Misiones, as the threats are not unique to this region. The need to develop management strategies that balance human-wildlife needs will continue to grow as humans expand their footprint. The techniques applied in this analysis provide a way to identify key areas that require specific management strategies, either through restoration, protection, or a combination of both. The European rabbit (Oryctolagus cuniculus) is one of the main invasive species in Chile, where it became naturalized ca. 150 years ago. Their high reproductive capacity, lack of specialist predators, and great adaptability favored the settlement of rabbits in diverse mainland and island ecosystems of the country. Recently, rabbits have become central players in semi-arid ecosystems, such as those represented in Las Chinchillas National Reserve in north-central Chile. We undertook to analyze the place and role of rabbits in the food web of that Reserve, based on a bibliographic review and long-term annual data gathered from 1987 until 2022 (36 years). Results showed that the network comprised 77 species, where 69% were primary producers (plants), 18% were mid-level consumers (herbivores), and 13% were top-level consumers (predators). The most connected species in the food web was the rabbit, which positively or negatively affected the species interacting with it. Predators such as Galictis cuja, Geranoaetus polyosoma, Leopardus colocolo, and Puma concolor, and the scavenger Vultur gryphus, could be negatively affected by an eventual decrease (natural or human-caused) in the rabbit population of the Reserve. To the contrary, primary producers such as Oxalis perdicaria, Plantago hispidula, Schizanthus parvulus, Senna cumminggi, and Tropaeolum azureum could be positively affected by an increase in their biomass in response to a decrease in rabbits, favoring native rodents. We consider that analyzing the rabbit-centered food web and its impacts on native interacting species allows a better understanding of the relevance of invasive species in the local community, providing conceptual tools for rabbit management. Simple Summary: The use of enrichment in zoos and aquariums has a positive effect on the wellbeing of animals, and supports their reintroduction success; however, research concerning their use in rehabilitation centers is still limited. This study investigated three different enrichment devices and their ability to decrease undesired repetitive and/or anticipatory behaviors in California sea lions and northern elephant seals undergoing rehabilitation in Southern California. All three devices were found to be durable and safe throughout the study, and demonstrated a significant reduction in animals displaying undesirable stereotypical behaviors compared to observations when no enrichment devices were used. This study highlights the beneficial aspect of environmental enrichment devices for pinnipeds hospitalized in rehabilitation centers. Environmental enrichment devices (EEDs) have been proven to promote positive wellbeing in zoos and aquariums, and support animals' reintroduction success; however, their use in rehabilitation centers is still limited. This pilot study investigated the safety and efficacy of three EEDs, Artificial Kelp, Horse KONG™ and Wubba Kong™, and their ability to decrease and/or eliminate undesired stereotypic behaviors or looking at staff/staff areas in seven wild California sea lions (CSLs) and eight northern elephant seals (NESs) undergoing rehabilitation in Southern California. Observers conducted instantaneous sampling once a minute during a 30-min baseline, followed by a 30-min EED implementation on one focal animal at a time. The data were analyzed with generalized linear mixed models (GLMM). All three EEDs were found to be durable and safe throughout the study. Our results show a significant reduction in stereotypical behaviors compared to no EED treatments in CSLs, with the most significant effect being with the Horse KONG™. The Wubba KONG™ and Artificial Kelp provisions also reduced the undesired behavior in both species of being focused on human caretakers. Individual preferences for specific EEDs were found within species and between species, with the NESs using EEDs more than the CSLs. Hunting is a critical issue in wild felid conservation, contributing to the decline of these species worldwide. Here, we present 18 years of a community‐based monitoring program, quantifying and characterizing wild felid hunting in two sustainable development reserves in Central Amazonia. We investigated how felid hunting was affected by the flood pulse and whether local human population size, habitat type, and total hunting events influenced felid hunting. We recorded 71 adult felids being killed, most of them in opportunistic events (88.7%), usually during hunting expeditions aimed at other game species. Four felid species were recorded in the hunts (Panthera onca, Leopardus pardalis, Puma concolor, and Leopardus wiedii) with jaguars accounting for almost half of all hunting events (N = 35). We found that more felid hunting events occurred during the flooded season, in more populated communities, and in várzea habitats. Most felids were hunted opportunistically, demonstrating the complexity of human‐felid interactions. Stories of attacks on humans are widespread in the study area, which may strengthen an instinctive fear of felids, driving their persecution. The increase of hunting events during the flooded season seems to be related to a higher probability of encounters between humans and wild felids. High waters make fishing difficult, therefore, locals hunt more, entering deep into the forest through flooded habitats. Since jaguars have an arboreal lifestyle during the flooded season in várzea forests, they are easily spotted by locals and have less of a chance to escape since they are slower and more exposed when swimming, which increases the probability of being killed. It is generally acknowledged that involving local stakeholders in conservation projects is essential for their success. In our case, collaboration among researchers and community members was fundamental to access hunting information. The main objectives of this study are the assessment of Ceratitis capitata infestation level on Argan fruits, and monitoring of its seasonal dynamics, as well as the assessment of parasitism rate by its parasitoid Psyttalia concolor in Argan orchard. To study Argan fruit infestation level and the parasitism rate, infested Argan fruit were sampled and transferred immediately to laboratory. Sampled fruits were screened to determine the number of medfly punctures. Other sampled fruits were incubated in aerated plastic containers containing sterilized sand. C. capitata dynamics was monitored by using male attracted pheromones traps during 2018 and 2019. The obtained results show a current activity flight of C. capitata over the two studied years and dynamics variation between the two sampled years and between months. The peak of the C. capitata dynamics was recorded during May and June for 2018 and 2019 respectively. Low Argan infestation level was recorded during two periods: January and August-September. However, the peak of infestation was recorded during May (95%). P. concolor parasitism rate was relatively low. No significant effect of fruit origin on P.concolor parasitism rate (P>0.05) was found. Yet, the sampling period had a significant effect on parasitism rate (p<0.05). Argan tree is a suitable host plant promoting C. capitata development with a permanent flight dynamics year-round. Despite the mediocre and variable parasitism rate of P. concolor, this parasitoid can be a very important tool together with the other protective measures to reduce infestation rate of C. capitataand to decrease its population size. Mitigating human-caused mortality for large carnivores is a pressing global challenge for wildlife conservation. However, mortality is almost exclusively studied at local (within-population) scales creating a mismatch between our understanding of risk and the spatial extent most relevant to conservation and management of wide-ranging species. Here, we quantified mortality for 590 radio-collared mountain lions statewide across their distribution in California to identify drivers of human-caused mortality and investigate whether human-caused mortality is additive or compensatory. Human-caused mortality, primarily from conflict management and vehicles, exceeded natural mortality despite mountain lions being protected from hunting. Our data indicate that human-caused mortality is additive to natural mortality as population-level survival decreased as a function of increasing human-caused mortality and natural mortality did not decrease with increased human-caused mortality. Mortality risk increased for mountain lions closer to rural development and decreased in areas with higher proportions of citizens voting to support environmental initiatives. Thus, the presence of human infrastructure and variation in the mindset of humans sharing landscapes with mountain lions appear to be primary drivers of risk. The PumAB type-II toxin–antitoxin (TA) system is encoded by pumAB genes that are organized into an operon. This system is encoded by the pUM505 plasmid, isolated from a Pseudomonas aeruginosa clinical strain. The pumA gene encodes a putative RelE toxin protein (toxic component), whereas the pumB gene encodes a putative HTH antitoxin protein. The expression of the PumAB system in Escherichia coli confers plasmid stability. In addition, PumA toxin overexpression in P. aeruginosa possesses the capability to increase bacterial virulence, an effect that is neutralized by the PumB antitoxin. The aim of this study was to establish the mechanism of regulation of the PumAB toxin–antitoxin system from pUM505. By an in silico analysis of the putative regulatory elements, we identified two putative internal promoters, PpumB and PpumB-AlgU (in addition to the already reported PpumAB), located upstream of pumB. By RT-qPCR assays, we determined that the pumAB genes are transcribed differentially, in that the mRNA of pumB is more abundant than the pumA transcript. We also observed that pumB could be expressed individually and that its mRNA levels decreased under oxidative stress, during individual expression as well as co-expression of pumAB. However, under stressful conditions, the pumA mRNA levels were not affected. This suggests the negative regulation of pumB by stressful conditions. The PumB purified protein was found to bind to a DNA region located between the PpumAB and the pumA coding region, and PumA participates in PumB binding, suggesting that a PumA–PumB complex co-regulates the transcription of the pumAB operon. Interestingly, the pumA mRNA levels decreased after incubation in vitro with PumB protein. This effect was repressed by ribonuclease inhibitors, suggesting that PumB could function as an RNAse toward the mRNA of the toxin. Taken together, we conclude that the PumAB TA system possesses multiple mechanisms to regulate its expression, as well as that the PumB antitoxin generates a decrease in the mRNA toxin levels, suggesting an RNase function. Body size correlates with various biological attributes. Hence, its evolution has attracted considerable attention. The family Felidae (Mammalia: Carnivora) has extensive variation in body size among extant species, with a few species being strikingly larger. The body size of extremely large species compared with phylogenetically related species may have evolved through directional selection toward increased size in a particular lineage, although lineage-specific directional selection has not been explicitly incorporated into standard models of body size evolution on a phylogeny. We used simulation-based likelihood and approximate Bayesian computation to examine the effect of lineage-specific directional selection on body mass evolution in extant felids. Our analyses revealed the occurrence of lineage-specific directional selection that favors larger body mass in the genus Panthera (lion, Panthera leo; jaguar, Panthera onca; leopard, Panthera pardus; tiger, Panthera tigris; and snow leopard, Panthera uncia), as well as in the cheetah (Acinonyx jubatus) and puma (Puma concolor). Lineage-specific directional selection was not detected in the clouded leopard (Neofelis nebulosa), which, along with the genus Panthera, diverged early from all other extant felids. The cheetah, puma, and jaguarundi (Puma yagouarondi) compose a monophyletic group in which body size may have decreased during the evolution of the jaguarundi, although our analyses revealed no lineage-specific directional selection for smaller sizes in the species. Directional selection for increased body size shown herein seems to be associated with a lifestyle dependent on relatively large prey. Chameleons represent one of the most endangered groups of lizards, with most being characterized as threatened or near-threatened. Anthropogenic impacts on their native habitat are responsible for their declines. To protect these animals, we must develop functional assisted reproductive programs. However, a full understanding of their basic reproductive biology is required to develop such a program. This study represents the first attempt at characterizing the reproductive cycles of male chameleons. Sixteen veiled chameleons (Chamaeleo calyptratus) and 16 panther chameleons (Furcifer pardalis) being housed in captivity under static conditions were used for this study. Chameleons were anesthetized twice monthly for semen collection via electroejaculation, blood sampling to measure plasma testosterone concentrations, and ultrasonographic testicular measurements. Plasma testosterone concentrations were found to be significantly different between species (P= 0.02) and in the interaction of species and month (P= 0.04). In veiled chameleons, plasma testosterone concentrations were significantly elevated during October through December, while for panther chameleons plasma testosterone concentrations were elevated in July through September, and December. Electroejaculation success was found to be seasonal in both species. In veiled chameleons, semen collection was highest in April, May, and June, while in panther chameleons semen collection success was highest in March, April, May, and June. Median spermatozoa concentrations for veiled and panther chameleons were 6.66 and 7.52 × 107 sperm/ml, respectively. Motility was lower than expected for both veiled (median 23%) and panther chameleons (median 25%). Spermatozoa clumping was a common finding. The majority of the spermatozoa (mean 51%) were found to have normal morphology. The results of this study confirm that veiled chameleons have a prenuptial reproductive strategy, while panther chameleons have a postnuptial reproductive strategy. Additionally, this study demonstrated that chameleons can undergo repeated electroejaculations under anesthesia, twice monthly, for a year without significant mortality or clinical disease associated with the procedures. The Pronghorn 'antelope' (Antilocapra) of western North America have long been enigmatic because they are the second-fastest modern land mammal with no natural predators that come close to matching their speed. With the discovery of the now-extinct North American cheetah, Miracinonyx , the hypothesis was put forward that the incredible speed of pronghorns was related to being the primary prey of the fleet-footed Miracinonyx. However, there has been no direct means to test that relationship. Here, utilizing organic-molecule-rich bones from Natural Trap Cave, it is possible to test the predator-prey relationship between Antilocapra and Miracinonyx through isotopic analysis of bone collagen. Offsets in carbon and nitrogen isotopes between predator and prey are used to establish or discount predator-prey relationships between common predators found in Natural Trap Cave including wolves (Canis), cheetah-like cats (Miracinonyx), and North American lions (Panthera) with prey species like horses (Equus and Haringtonhippus), pronghorns (Antilocapra), sheep (Ovis), and bison (Bison). Results provide support the hypothesis that Miracinonyx preyed upon Antilocapra , but not exclusively. Antilocapra was also important prey for lions and wolves. Simple Summary: Tumors of the skin are one of the most commonly observed neoplasms in captive lizards. The current study characterizes keratoacanthoma, a previously undescribed skin tumor, in five male lizards (one bearded dragon, one veiled chameleon, and three panther chameleons) with an average to high age. In all lizards, keratoacanthomas presented as cystic nodules with a central keratin pearl that was predominantly located at the body wall. In all chameleons, a multicentric distribution was observed. Following surgical removal of the keratoacanthomas in all lizards, a follow-up period of one to two years was established. While the skin neoplasia reappeared in the bearded dragon and the veiled chameleon, no recurrence was seen in the panther chameleons. Keratoacanthoma constitutes a rather benign histologic variant of squamous cell carcinoma, representing a non-invasive but rapidly growing skin neoplasia that may be associated with the inappropriate use of ultraviolet lighting in the captive environment. In addition, panther chameleons may show a species predisposition as well as a tendency to develop multicentric keratoacanthomas. The present study delivers pertinent results for the diagnosis, prevention, and treatment of keratoacanthomas in lizards. The present study describes the clinical behavior as well as the histopathologic and immunohistochemical characteristics of keratoacanthomas (Kas) in three different saurian species. While Kas presented as two dermal lesions in a bearded dragon (Pogona vitticeps), multicentric Kas were observed in three panther chameleons (Furcifer pardalis) and a veiled chameleon (Chamaeleo calyptratus). Macroscopically, Kas presented as dome-shaped skin tumors with a centralized keratinous pearl and a diameter ranging from 0.1–1.5 cm. In all lizards, Kas were predominantly located at the dorsolateral body wall, and KA of the eyelid was additionally observed in three out of four chameleons. Histologically, KAs presented as relatively well-defined, circumscribed epidermal proliferations that consisted of a crateriform lesion containing a central keratinous pearl with minimally infiltrating borders. In all KAs, a consistent immunohistochemical pattern was observed, with the expression of cyclooxygenase-2, E-cadherin, and pan-cytokeratin. A follow-up period of one to two years was established in all lizards. While no recurrence was observed in the panther chameleons, recurrence of a single keratoacanthoma was observed in the bearded dragon after one year, and in the veiled chameleon, multicentric keratoacanthomas reappeared during a follow-up period of two years. We describe KA as a previously unrecognized neoplastic entity in lizards that constitutes a low-grade, non-invasive but rapidly growing skin tumor that may show a multicentric appearance, especially in chameleons. As previously postulated for dermal squamous cell carcinomas (SCC), artificial ultraviolet lighting may play an important role in the oncogenesis of KAs in lizards. Although dermal SCCs in lizards show similar predilection sites and gross pathologic features, our results suggest that KA should be considered as a histologic variant of SCC that represents a rather benign squamous proliferation in comparison to conventional SCCs. Fibroblast-like synoviocytes (FLSs), the main pathological cells in rheumatoid arthritis (RA), display tumor-like phenotype, including hyper-proliferation, apoptosis resistance, and aggressive phenotype. Excessive proliferation and insufficient apoptosis of RA-FLSs can lead to hyperplastic synovial pannus tissue, excess production of inflammatory mediators, and destruction of joints. In this article, we investigate the effect of PRIMA-1MET on the apoptosis induction and inhibition of pro-inflammatory cytokines in RA-FLSs. Synovial tissue samples were obtained from 10 patients with RA. The FLSs were treated with different concentrations of PRIMA-1MET. The rate of apoptosis and cell survival was assessed by flow cytometry and MTT assay and Real-time quantitative PCR was performed to evaluate the transcription of p53, IL-6, IL-1β, TNF-α, Noxa, p21, PUMA, Bax, Survivin, and XIAP in treated RA-FLSs. The protein level of p53, IκBα, and phospho-IκBα were measured using Western blotting. The results showed that PRIMA-1MET induced apoptosis in RA-FLSs and increased significantly the expression of Noxa, and decreased significantly IL-6, IL-1β, p53, and phospho-IκBα expression. PRIMA-1MET can induce apoptosis in RA-FLSs through induction of Noxa expression while p53 was downregulated. Furthermore, PRIMA-1MET treatment results in the suppression of pro-inflammatory cytokine production and NF-κB inhibition. Given the role of p53 and NF-κB in RA-FLSs, PRIMA-1MET can be considered as a new therapeutic strategy for rheumatoid arthritis. [ABSTRACT FROM AUTHOR]

The effects of predation risk on prey populations have been studied extensively; yet, how risk is manifested in a trophically linked guild—scavengers—has been overlooked. Risk could be particularly consequential for obligate scavengers that are vulnerable while foraging and rely on carrion provisioned by, and shared with, apex predators. We investigated whether Andean condors (Vultur gryphus) respond to predation risk in a landscape where the main source of carrion are camelids killed by pumas (Puma concolor). We hypothesized that condors would exhibit different behavioral responses to predation risk while they search, encounter, and exploit carrion. We explored condor habitat selection while flying by tracking nine birds with satellite transmitters and monitored via camera traps 41 natural carcasses and 25 experimental carrion stations. We found that condors searched for carrion in areas with a high probability of occurrence of puma kills. However, condors avoided exploiting carrion in areas featuring tall vegetation and steep slopes—selected by pumas to stalk prey—suggesting that condors manage risk primarily through the identification of safe foraging sites prior to landing. Our finding that condors avoided foraging near stalking cover for pumas highlights the importance of risk effects beyond predator–prey interactions, particularly for obligate scavengers. Jaguars and pumas are top-predator species in the Neotropics that are threatened by habitat destruction, illegal poaching of their body parts and their favored prey, and by the human–wildlife conflicts that arise when predators attack livestock. Much of the remaining felid habitat in the Americas is in protected nature reserves that are too small and isolated to support local populations. Surrounding forests therefore play a vital role in felid conservation. Successful long-term conservation of these two felids requires evidence-based knowledge of their biological and ecological requirements. We studied population distributions of jaguars and pumas and their prey in and between two small, private reserves of the Northern Yucatán Peninsula, Mexico, with areas of 25 and 43 km2. During 2 years of camera trapping (2015 and 2016), we detected 21 jaguars, from which we estimated an average space requirement of 28–45 km2/individual. Dietary niche overlap exceeded random expectation. The most frequently occurring prey items in jaguar and puma diets were collared peccary and deer. Jaguar also favored nine-banded armadillos and white-nosed coati, while puma favored canids. Both felids avoided ocellated turkey. Overall, diet of jaguars was less species-rich, but similar in niche breadth, to that of pumas. A fluid use of space by both species, in 2015 tending toward mutual attraction and in 2016 toward partial exclusion of pumas by jaguars, combined with the high dietary overlap, is consistent with a dominance hierarchy facilitating coexistence. Jaguars and pumas favor the same prey as the people in local communities who hunt, which likely will intensify human–wildlife impacts when prey become scarce. We conclude that even small reserves play an important role in increasing the continuity of habitat for prey and large felids, whose generalist habits suppress interspecific competition for increasingly limiting prey that are largely shared between them and humans.
