Vector control remains one of the best strategies to prevent the transmission of trypanosome infections in humans and livestock and, thus, a good way to achieve the elimination of human African trypanosomiasis and animal African trypanosomiasis. A key prerequisite for the success of any vector control strategy is the accurate identification and correct mapping of tsetse species. In this work, we updated the tsetse fly species identification and distribution in many geographical areas in Cameroon. Tsetse flies were captured from six localities in Cameroon, and their species were morphologically identified. Thereafter, DNA was extracted from legs of each tsetse fly and the length polymorphism of internal transcribed spacer‐1 (ITS1) region of each fly was investigated using PCR. ITS1 DNA fragments of each tsetse species were sequenced. The sequences obtained were analysed and compared to those available in GenBank. This enabled to confirm/infirm results of the morphologic identification and then, to establish the phylogenetic relationships between tsetse species. Morphologic features allowed to clearly distinguish all the tsetse species captured in the South Region of Cameroon, that is, Glossina palpalis palpalis, G. pallicera, G. caliginea and G. nigrofusca. In the northern area, G. morsitans submorsitans could also be distinguished from G. palpalis palpalis, G. tachinoides and G. fuscipes, but these three later could not be distinguished with routine morphological characters. The ITS1 length polymorphism was high among most of the studied species and allowed to identify the following similar species with a single PCR, that is, G. palpalis palpalis with 241 or 242 bp and G. tachinoides with 221 or 222 bp, G. fuscipes with 236 or 237 bp. We also updated the old distribution of tsetse species in the areas assessed, highlighting the presence of G. palpalis palpalis instead of G. fuscipes in Mbakaou, or in sympatry with G. morsitans submorsitans in Dodeo (northern Cameroon). This study confirms the presence of G. palpalis palpalis in the Adamawa Region of Cameroon. It highlights the limits of using morphological criteria to differentiate some tsetse species. Molecular tools based on the polymorphism of ITS1 of tsetse flies can differentiate tsetse species through a simple PCR before downstream analyses or vector control planning. [ABSTRACT FROM AUTHOR]

Burkina Faso relies on its substantial bovine population for meat and milk production, ensuring food security. The country hosts three primary taurine cattle populations: Lobi, Gourounsi Nahouri, and Gourounsi Ténado. These cattle are adapted to local conditions and exhibit valuable trypan tolerant traits, playing a crucial role in sustaining local communities and holding cultural and socioeconomic significance. This study aimed to assess the genetic diversity and structure of Burkina's primary taurine cattle populations using 27 microsatellite markers. Blood samples from 143 cattle representing these populations were genotyped. The analysis included assessing genetic diversity, deviations from Hardy-Weinberg equilibrium, calculating genetic distances, and population structure. The results revealed that all loci were polymorphic, indicating high allelic diversity. The overall mean FIS was moderate (0.028), ranging from -0.36 (CSRM60) to 0.73 (INRA035). Genetic differentiation between populations was moderate, accounting for 4% of the total differences. The highest pairwise FST was observed between Lobi and Gourounsi Ténado. The neighbor-joining tree displayed high admixture levels between Gourounsi populations, while Lobi cattle clustered as a distinct population. The population structure analysis indicated significant zebu gene introgression in Burkina taurine populations with relatively higher levels of admixtures in Gourounsi cattle compared to Lobi. The study provided a thorough genetic analysis of Burkina Faso's taurine cattle populations, uncovering the diversity and population structure. The study also revealed the differences in the prevalence of tsetse flies and associated trypanosomosis across the native tracts of Burkinabe taurine cattle populations had shaped the level of zebu introgression in them. Background: Insect cell lines play a vital role in many aspects of research on disease vectors and agricultural pests. The tsetse fly Glossina morsitans morsitans is an important vector of salivarian trypanosomes in sub-Saharan Africa and, as such, is a major constraint on human health and agricultural development in the region. Methods: Here, we report establishment and partial characterisation of a cell line, GMA/LULS61, derived from tissues of adult female G. m. morsitans. GMA/LULS61 cells, grown at 28 °C in L-15 (Leibovitz) medium supplemented with foetal bovine serum and tryptose phosphate broth, have been taken through 23 passages to date and can be split 1:1 at 2-week intervals. Karyotyping at passage 17 revealed a predominantly haploid chromosome complement. Species origin and absence of contaminating bacteria were confirmed by PCR amplification and sequencing of fragments of the COI gene and pan-bacterial 16S rRNA gene respectively. However, PCR screening of RNA extracted from GMA/LULS61 cells confirmed presence of the recently described Glossina morsitans morsitans iflavirus and Glossina morsitans morsitans negevirus, but absence of Glossina pallipides salivary gland hypertrophy virus. GMA/LULS61 cells supported infection and growth of 6/7 different insect-derived strains of the intracellular bacterial symbiont Wolbachia. Conclusions: The GMA/LULS61 cell line has potential for application in a variety of studies investigating the biology of G. m. morsitans and its associated pathogenic and symbiotic microorganisms. Background: Human African trypanosomiasis (HAT) is a neglected tropical disease that usually occurs in rural areas in sub-Saharan Africa. It caused devastating epidemics during the 20th century. Sustained, coordinated efforts by different stakeholders working with national sleeping sickness control programmes (NSSCPs) succeeded in controlling the disease and reducing the number of cases to historically low levels. In 2012, WHO targeted the elimination of the disease as a public health problem by 2020. This goal has been reached and a new ambitious target was stated in the WHO road map for NTDs 2021–2030 and endorsed by the 73rd World Health Assembly: the elimination of gambiense HAT transmission (i.e. reducing the number of reported cases to zero). The interruption of transmission was not considered as an achievable goal for rhodesiense HAT, as it would require vast veterinary interventions rather than actions at the public health level. Methodology/principal findings: Data reported to WHO by NSSCPs were harmonized, verified, georeferenced and included in the atlas of HAT. A total of 802 cases were reported in 2021 and 837 in 2022. This is below the target for elimination as a public health problem at the global level (< 2000 HAT cases/year); 94% of the cases were caused by infection with T. b. gambiense. The areas reporting ≥ 1 HAT case/10 000 inhabitants/year in 2018–2022 cover a surface of 73 134 km2, with only 3013 km2 at very high or high risk. This represents a reduction of 90% from the baseline figure for 2000–2004, the target set for the elimination of HAT as a public health problem. For the surveillance of the disease, 4.5 million people were screened for gambiense HAT with serological tests in 2021–2022, 3.6 million through active screening and 0.9 million by passive screening. In 2021 and 2022 the elimination of HAT as a public health problem was validated in Benin, Uganda, Equatorial Guinea and Ghana for gambiense HAT and in Rwanda for rhodesiense HAT. To reach the next goal of elimination of transmission of gambiense HAT, countries have to report zero cases of human infection with T. b. gambiense for a period of at least 5 consecutive years. The criteria and procedures to verify elimination of transmission have been recently published by WHO. Conclusions/significance: HAT elimination as a public health problem has been reached at global level, with seven countries already validated as having reached this goal. This achievement was made possible by the work of NSSCPs, supported by different public and private partners, and coordinated by WHO. The new challenging goal now is to reach zero cases by 2030. To reach this goal is crucial to maintain the engagement and support of donors and stakeholders and to keep the involvement and coordination of all partners. Along with the focus on elimination of transmission of gambiense HAT, it is important not to neglect rhodesiense HAT, which is targeted for elimination as a public health problem in the WHO road map for NTDs 2021–2030. HAT surveillance is maintained through active screening in endemic areas but also within a network of health facilities able to detect the disease. Background: The insecticide-treated baits known as Tiny Targets are one of the cheapest means of controlling riverine species of tsetse flies, the vectors of the trypanosomes that cause sleeping sickness in humans. Models of the efficacy of these targets deployed near rivers are potentially useful in planning control campaigns and highlighting the principals involved. Methods and principal findings: To evaluate the potential of models, we produced a simple non-seasonal model of the births, deaths, mobility and aging of tsetse, and we programmed it to simulate the impact of seven years of target use against the tsetse, Glossina fuscipes fuscipes, in the riverine habitats of NW Uganda. Particular attention was given to demonstrating that the model could explain three matters of interest: (i) good control can be achieved despite the d egradation of targets, (ii) local elimination of tsetse is impossible if invasion sources are not tackled, and (iii) with invasion and target degradation it is difficult to detect any effect of control on the age structure of the tsetse population. Conclusions: Despite its simplifications, the model can assist planning and teaching, but allowance should be made for any complications due to seasonality and management challenges associated with greater scale. Author summary: We produced a simple model of the population dynamics of the riverine tsetse fly, Glossina fuscipes fuscipes, to simulate the field results of controlling this insect for seven years using Tiny Targets, i.e., artificial insecticide-treated baits, in NW Uganda. Puparia are commonly found in tsetse fly larviposition sites during studies on larval ecology. This chitinous shell is representative of past or ongoing exploitation of these sites by tsetse flies. The morphological characteristics of the puparium are not sufficiently distinctive to allow identification of the species. This study explores the applicability of biomolecular techniques on empty puparia for tsetse fly species identification. Five techniques were compared for DNA extraction from tsetse fly puparia, 1/Chelex® 100 Resin, 2/CTAB, 3/Livak's protocol, 4/DEB + proteinase K and 5/QIAamp® DNA Mini kit, using two homogenisation methods (manual and automated). Using a combination of two primer pairs, Chelex, CTAB, and DEB + K proved the most efficient on fresh puparia with 90, 85, and 70% samples identified, respectively. Shifting from fresh to one- to nine-month-old puparia, the Chelex method gave the best result allowing species identification on puparia up to seven months old. The subsequent testing of the Chelex extraction protocol identified 152 (60%) of 252 field-collected puparia samples at species level. The results show that reliable genetic identification of tsetse flies species can be performed from empty puparia, what can prove of great interest for future ecological studies on larviposition sites. The Chelex technique was the most efficient for DNA extraction, though the age-limit of the samples stood at seven months, beyond which DNA degradation probably compromises the genetic analysis. Several species of horse flies (Diptera: Tabanidae) are known as vectors of Trypanosoma (Megatrypanum) theileri and T. theileri-like trypanosomes; these host–parasite relationships were established based on the developmental stages of these parasites discovered in the hindgut of horse flies. T. theileri and T. theileri-like trypanosomes have been detected in cattle and wild deer in Japan; however, the vector horse fly species remains unidentified. Therefore, in this study, we aimed to identify the potential horse fly species serving as vectors of T. theileri in Japan. A total of 176 horse flies were collected between June to September 2020 and 2021 in Tokachi, Hokkaido, Japan. The T. theileri infection in the captured horse flies was determined by PCR and microscopic analyses of their midgut and hindgut. Additionally, the trypanosome, microscopically detected in a horse fly, was molecularly characterized and phylogenetically analyzed using 18S rRNA and partial cathepsin L-like protein gene (CATL) sequence of the trypanosome. The microscopy and PCR analyses revealed 0.57% and 35.8% prevalence of T. theileri in horse flies, respectively. Epimastigote stages of T. theileri, adhered to the hindgut epithelial cells of Tabanus chrysurus via flagella or actively moving in the lumen of the gut, were detected. Phylogenetic analysis revealed the connection of isolated trypanosomes with T. theileri in the TthI clade. These results suggest that Ta. chrysurus is a potential vector of T. theileri. Heatwaves are increasing in frequency and intensity due to climate change, pushing animals beyond physiological limits. While most studies focus on survival limits, sublethal effects on fertility tend to occur below lethal thresholds, and consequently can be as important for population viability. Typically, male fertility is more heat-sensitive than female fertility, yet direct comparisons are limited. Here, we measured the effect of experimental heatwaves on tsetse flies, Glossina pallidipes, disease vectors and unusual live-bearing insects of sub-Saharan Africa. We exposed males or females to a 3-day heatwave peaking at 36, 38 or 40°C for 2 h, and a 25°C control, monitoring mortality and reproduction over six weeks. For a heatwave peaking at 40°C, mortality was 100%, while a 38°C peak resulted in only 8% acute mortality. Females exposed to the 38°C heatwave experienced a one-week delay in producing offspring, whereas no such delay occurred in males. Over six weeks, heatwaves resulted in equivalent fertility loss in both sexes. Combined with mortality, this lead to a 10% population decline over six weeks compared to the control. Furthermore, parental heatwave exposure gave rise to a female-biased offspring sex ratio. Tsetse flies, the sole biological vectors of trypanosomiasis, are predominantly controlled using visual traps and targets baited with attractant lures. Formulation of the lures is informed by compositions of odors from vertebrate hosts preferred by specific tsetse species. However, there are no effective lures for Glossina austeni, a major vector of trypanosomiasis along eastern-coastal region of Africa. Formulation of the lure can be informed by knowledge of G. austeni, preferred vertebrate hosts. We thus sought to understand these hosts by assessment of putative bloodmeal sources of this tsetse fly in Arabuko Sokoke National Reserve where this species is naturally present. We sampled tsetse flies using NGU traps, isolated non-teneral G. austeni flies based on their feeding status, and identified vertebrate source of bloodmeals in their midgut contents using vertebrate 16S rRNA-PCR High-Resolution Melting analysis. We analyzed the relative vertebrate species frequencies in the bloodmeals using Fisher's exact tests. Overall, we trapped 122 flies, most of which (66.39%) were non-teneral, among which we successfully identified the vertebrate bloodmeals in 30 samples. Specifically, we detected putative suni antelope (Neotragus moschatus), harnessed bushbuck (Tragelaphus scriptus), buffalo (Syncerus caffer) and cattle (Bos taurus) derived bloodmeals. Putative suni antelope bloodmeals were significantly more frequent (63.22%), than those of the harnessed bushbuck (23.33%), buffalo (10.00%) or cattle (3.33%) (p < 0.05 Fisher's exact tests) among the samples analyzed. Suni antelope thus appears to predominate vertebrate bloodmeal source for G. austeni in the reserve, coincident with findings reported elsewhere, and is therefore a viable candidate for bioprospecting for G. austeni responsive attractants. Simple Summary: Artificial feeding systems have emerged as a vital tool in research on arthropods like mosquitoes, ticks, blackflies, sandflies, tsetse flies, fleas, and triatomine bugs, aiding in the understanding of pathogen transmission. This review explores various artificial feeding systems used to study human–vector relationships and pathogen transmission, detailing their roles in insect-related research. We discuss the advantages and disadvantages of these systems, their practical applications, and speculate on future directions in vector-borne disease research. Recognizing the strengths and weaknesses of different artificial feeding systems will help researchers to choose the right tools for developing effective pathogen transmission and disease control strategies. This review examines the advancements and methodologies of artificial feeding systems for the study of vector-borne diseases, offering a critical assessment of their development, advantages, and limitations relative to traditional live host models. It underscores the ethical considerations and practical benefits of such systems, including minimizing the use of live animals and enhancing experimental consistency. Various artificial feeding techniques are detailed, including membrane feeding, capillary feeding, and the utilization of engineered biocompatible materials, with their respective applications, efficacy, and the challenges encountered with their use also being outlined. This review also forecasts the integration of cutting-edge technologies like biomimicry, microfluidics, nanotechnology, and artificial intelligence to refine and expand the capabilities of artificial feeding systems. These innovations aim to more accurately simulate natural feeding conditions, thereby improving the reliability of studies on the transmission dynamics of vector-borne diseases. This comprehensive review serves as a foundational reference for researchers in the field, proposing a forward-looking perspective on the potential of artificial feeding systems to revolutionize vector-borne disease research. The success of any tsetse control program depends on the knowledge of their behaviour. This study assessed the host choice and feeding behaviours of Glossina morsitans siblings whose parents were bloodfed on rabbits, guinea pigs, rodents, and squirrels. Each individual host was placed in a screen cage, which allowed flies to enter through openings on each side. The groups of flies (20 per replicate), which were colour-marked differently based on their parents' blood meal hosts, were released from the centre of large semi-field cage. The released flies were aspirated after 24 h and then sorted based on their location, feeding status, and parents' blood meal. A total of 213 flies (72.95% of those recovered) were attracted to the hosts. The numbers of flies attracted to different hosts varied significantly (χ24 = 33.685, p = 0.0001): rodents (n = 80, p = 0.006), rabbits (n = 59, p = 0.331), guinea pigs (n = 49, p = 0.057), and squirrels (n = 25, p = 0.005). The numbers of flies attracted to their parent's blood meal source varied significantly (χ212 = 56.476, p < 0.001): rabbits (n = 35, 59.32%, p < 0.001), rodents (n = 25, 31.25%, p = 0.043), and guinea pigs (n = 19, 38.78%, p = 0.45). But only 39 flies (18.31% of the total attracted) bloodfed on the hosts, including guinea pigs (n = 10, 25.64%), rodents (n = 23, 58.97%), rabbits (n = 6, 15.38%), and squirrels (n = 0, 0.0%). There was significant variation in the number of flies that fed successively across hosts (χ24 = 49.478, p < 0.001). The findings from this study confirm the presence of differential attractiveness of the hosts to flies and the so-called "Hopkins host selection principle" or "pre-imaginal conditioning". Therefore, the study attracts the need for detailed investigation on the influence of blood meal sources on tsetse fly siblings' behaviours across filial generations using small mammals or other large mammal species. This study was conducted to determine the prevalence of Trypanosoma species in cattle due to the increasing movement of animals within the metropolis. Blood specimens were collected from 1200 cattle during slaughter at four abattoirs (Bogobiri, Nasarawa, Ibesikpo, and Ikot Eneobong) between February and May 2022. Dry universal containers were positioned in the stream of blood oozing from the cut neck of each cow. From these containers, 4 mL of blood was aspirated into an ethylenediamine-tetra-acetic acid (EDTA) bottle using a 5 mL syringe. Blood specimens were processed for microscopy at the Parasitology Laboratory of the University of Calabar Teaching Hospital. Wet preparation of each specimen was performed and examined for motile haemoparasites. Triple centrifugation was performed on 2 mL of each blood specimen at increasing speeds of 1000, 1500, and 3000 revolutions per minute for 5 min. During each centrifugation process, the resulting plasma was harvested and re-centrifuged. Finally, the resulting buffy coat layer and deposit were resuspended and used to prepare smears on clean microscope slides. After drying in air, blood smears were stained with 3% Giemsa solution for 30 min. The microscopy results showed that 570 stained blood specimens were positive for Trypanosoma species, which morphologically resembled Trypanosoma brucei because of their long free flagellum. The overall prevalence of trypanosomes in cattle was 47.5%. Of the total number of infected cattle, 330 were males and 240 were females. The prevalence rates in male and female cattle were 27.5% and 20 %. There was no statistical difference in the occurrence of trypanosomes in the male and female cattle (X² = 9.41; p > 0.05). However, the findings suggest that male cattle tend to forage more into the bush and are thus more likely to be exposed to bites of infected Glossina flies than their female counterparts. The high prevalence of trypanosomes in cattle, as shown in this study, may have major epidemiologic significance, considering the increasing rate of open grazing by cattle within residential areas in major cities all over Nigeria. Background: Tsetse flies (Glossina spp.) are the definitive biological vectors of African trypanosomes in humans and animals. Controlling this vector is the most promising method of preventing trypanosome transmission. This requires a comprehensive understanding of tsetse biology and host preference to inform targeted design and management strategies, such as the use of olfaction and visual cues in tsetse traps. No current review exists on host preference and blood meal analyses of tsetse flies. Methods: This review presents a meta-analysis of tsetse fly blood meal sources and the methodologies used to identify animal hosts from 1956 to August 2022. The Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews (PRIMA-ScR) was applied. This focused on tsetse-endemic countries, blood meal analysis methodologies and the blood meal hosts identified. The articles were retrieved and screened from databases using predetermined eligibility criteria. Results: Only 49/393 of the articles retrieved matched the inclusion criteria. Glossina's main hosts in the wild included the bushbuck, buffalo, elephant, warthog, bushpig and hippopotamus. Pigs, livestock and humans were key hosts at the domestic interface. The least studied species included Glossina fuscipleuris, G. fusca, G. medicorum, G. tabaniformis and G. austeni. In the absence of preferred hosts, Glossina fed opportunistically on a variety of hosts. Precipitin, haemagglutination, disc diffusion, complement fixation, ELISA and PCR-based assays were used to evaluate blood meals. Cytochrome b (Cyt b) was the main target gene in PCR to identify the vertebrate hosts. Conclusions: Tsetse blood meal sources have likely expanded because of ecological changes that could have rendered preferred hosts unavailable. The major approaches for analysing tsetse fly blood meal hosts targeted Cyt b gene for species identification by Sanger sequencing. However, small-fragment DNAs, such as the mammalian 12S and 16S rRNA genes, along with second- and third-generation sequencing techniques, could increase sensitivity for host identification in multiple host feeders that Sanger sequencing may misidentify as "noise". Background: In tropical Africa animal trypanosomiasis is a disease that has severe impacts on the health and productivity of livestock in tsetse fly-infested regions. Trypanosoma congolense savannah (TCS) is one of the main causative agents and is widely distributed across the sub-Saharan tsetse belt. Population genetics analysis has shown that TCS is genetically heterogeneous and there is evidence for genetic exchange, but to date Trypanosoma brucei is the only tsetse-transmitted trypanosome with experimentally proven capability to undergo sexual reproduction, with meiosis and production of haploid gametes. In T. brucei sex occurs in the fly salivary glands, so by analogy, sex in TCS should occur in the proboscis, where the corresponding portion of the developmental cycle takes place. Here we test this prediction using genetically modified red and green fluorescent clones of TCS. Methods: Three fly-transmissible strains of TCS were transfected with genes for red or green fluorescent protein, linked to a gene for resistance to the antibiotic hygromycin, and experimental crosses were set up by co-transmitting red and green fluorescent lines in different combinations via tsetse flies, Glossina pallidipes. To test whether sex occurred in vitro, co-cultures of attached epimastigotes of one red and one green fluorescent TCS strain were set up and sampled at intervals for 28 days. Results: All interclonal crosses of genetically modified trypanosomes produced hybrids containing both red and green fluorescent proteins, but yellow fluorescent hybrids were only present among trypanosomes from the fly proboscis, not from the midgut or proventriculus. It was not possible to identify the precise life cycle stage that undergoes mating, but it is probably attached epimastigotes in the food canal of the proboscis. Yellow hybrids were seen as early as 14 days post-infection. One intraclonal cross in tsetse and in vitro co-cultures of epimastigotes also produced yellow hybrids in small numbers. The hybrid nature of the yellow fluorescent trypanosomes observed was not confirmed by genetic analysis. Conclusions: Despite absence of genetic characterisation of hybrid trypanosomes, the fact that these were produced only in the proboscis and in several independent crosses suggests that they are products of mating rather than cell fusion. The three-way strain compatibility observed is similar to that demonstrated previously for T. brucei, indicating that a simple two mating type system does not apply for either trypanosome species. Trypanosoma brucei is the causative agent of African trypanosomiasis and is transmitted by the tsetse fly (Glossina spp.). All stages of this extracellular parasite possess a single flagellum that is attached to the cell body and confers a high degree of motility. While several stages are amenable to culture in vitro, longitudinal high-resolution imaging of free-swimming parasites has been challenging, mostly due to the rapid flagellar beating that constantly twists the cell body. Here, using microfabrication, we generated various microfluidic devices with traps of different geometrical properties. Investigation of trap topology allowed us to define the one most suitable for single T. brucei confinement within the field of view of an inverted microscope while allowing the parasite to remain motile. Chips populated with V-shaped traps allowed us to investigate various phenomena in cultured procyclic stage wild-type parasites, and to compare them with parasites whose motility was altered upon knockdown of a paraflagellar rod component. Among the properties that we investigated were trap invasion, parasite motility, and the visualization of organelles labelled with fluorescent dyes. We envisage that this tool we have named "Tryp-Chip" will be a useful tool for the scientific community, as it could allow high-throughput, high-temporal and high-spatial resolution imaging of free-swimming T. brucei parasites. In the mammalian host, the biology of tissue-dwelling Trypanosoma brucei parasites is not completely understood, especially the mechanisms involved in their extravascular colonization. The trypanosome flagellum is an essential organelle in multiple aspects of the parasites' development. The flagellar protein termed FLAgellar Member 8 (FLAM8) acts as a docking platform for a pool of Cyclic AMP response protein 3 (CARP3) that is involved in signaling. FLAM8 exhibits a stage-specific distribution suggesting specific functions in the mammalian and vector stages of the parasite. Analyses of knockdown and knockout trypanosomes in their mammalian forms demonstrated that FLAM8 is not essential in vitro for survival, growth, motility and stumpy differentiation. Functional investigations in experimental infections showed that FLAM8-deprived trypanosomes can establish and maintain an infection in the blood circulation and differentiate into insect transmissible forms. However, quantitative bioluminescence imaging and gene expression analysis revealed that FLAM8-null parasites exhibit a significantly impaired dissemination in the extravascular compartment, that is restored by the addition of a single rescue copy of FLAM8. In vitro trans-endothelial migration assays revealed significant defects in trypanosomes lacking FLAM8. FLAM8 is the first flagellar component shown to modulate T. brucei distribution in the host tissues, possibly through sensing functions, contributing to the maintenance of extravascular parasite populations in mammalian anatomical niches, especially in the skin. Transmitted by the bite of an infected tsetse fly, upon deposition in the skin of a mammalian host, these parasites occupy both the vasculature and extravascular tissues. Using quantitative in vivo bioluminescence imaging and ex vivo gene expression quantification in host infected tissues and blood, we reveal that the flagellar parasite protein FLAM8 modulates the extravascular dissemination of trypanosomes in the mammalian host. FLAM8 is known to act as a docking platform for signaling complexes in the flagellum, but we observe that it does not influence parasite differentiation into transmissible stages. However, we show that the absence of FLAM8 results in the loss of a key component of the flagellar adenylate cyclase signaling complexes, and reduces parasite migration through endothelial cell monolayers, suggesting that FLAM8 is important for parasite exchanges between the intravascular and the extravascular compartments. Vector-borne pathogens (VBPs) causing vector-borne diseases (VBDs) can circulate among humans, domestic animals, and wildlife, with cattle in particular serving as an important source of exposure risk to humans. The close associations between humans and cattle can facilitate the transmission of numerous VBPs, impacting public health and economic security. Published studies demonstrate that cattle can influence human exposure risk positively, negatively, or have no effect. There is a critical need to synthesize the information in the scientific literature on this subject, in order to illuminate the various ecological mechanisms that can affect VBP exposure risk in humans. Therefore, the aim of this systematic review was to review the scientific literature, provide a synthesis of the possible effects of cattle on VBP risk to humans, and propose future directions for research. This study was performed according to the PRISMA 2020 extension guidelines for systematic review. After screening 470 peer-reviewed articles published between 1999–2019 using the databases Web of Science Core Collection, PubMed Central, CABI Global Health, and Google Scholar, and utilizing forward and backward search techniques, we identified 127 papers that met inclusion criteria. Results of the systematic review indicate that cattle can be beneficial or harmful to human health with respect to VBDs depending on vector and pathogen ecology and livestock management practices. Cattle can increase risk of exposure to infections spread by tsetse flies and ticks, followed by sandflies and mosquitoes, through a variety of mechanisms. However, cattle can have a protective effect when the vector prefers to feed on cattle instead of humans and when chemical control measures (e.g., acaricides/insecticides), semio-chemicals, and other integrated vector control measures are utilized in the community. We highlight that further research is needed to determine ways in which these mechanisms may be exploited to reduce VBD risk in humans. Author summary: Vector-borne diseases (VBDs) are caused by pathogens spread by blood-feeding arthropods from an infected to an uninfected organism. These infections may be caused by pathogenic bacteria, viruses, or protozoans and arthropod vectors may spread these pathogens to humans, domestic animals, and wildlife. Humans and cattle spend a significant amount of time in close proximity with each other through various activities such as agriculture, animal husbandry, trading, and animal farming, which can potentially increase risk to human health. Previously published studies indicated cattle can impact vector-borne pathogen (VBP) transmission both positively and negatively, however, there has not been a recent synthesis of the scientific literature on this subject. Through this global systematic review of the scientific literature, we found that cattle could have either harmful or beneficial impacts on human health when it comes to VBDs, but most often increase exposure risk to VBPs in humans. We identified various mechanisms from the scientific literature by which cattle can impact VBP risk in humans. Further research is needed to better understand specific ecological mechanisms by which cattle impact human health and develop measures that will prevent and reduce VBP exposure risk in humans. Tsetse flies (Glossina spp.) are major vectors of African trypanosomes, causing either Human or Animal African Trypanosomiasis (HAT or AAT). Several approaches have been developed to control the disease, among which is the anti‐vector Sterile Insect Technique. Another approach to anti‐vector strategies could consist of controlling the fly's vector competence through hitherto unidentified regulatory factors (genes, proteins, biological pathways, etc.). The present work aims to evaluate the protein abundance in the midgut of wild tsetse flies (Glossina palpalis palpalis) naturally infected by Trypanosoma congolense s.l. Infected and non‐infected flies were sampled in two HAT/AAT foci in Southern Cameroon. After dissection, the proteomes from the guts of parasite‐infected flies were compared to that of uninfected flies to identify quantitative and/or qualitative changes associated with infection. Among the proteins with increased abundance were fructose‐1,6‐biphosphatase, membrane trafficking proteins, death proteins (or apoptosis proteins) and SERPINs (inhibitor of serine proteases, enzymes considered as trypanosome virulence factors) that displayed the highest increased abundance. The present study, together with previous proteomic and transcriptomic studies on the secretome of trypanosomes from tsetse fly gut extracts, provides data to be explored in further investigations on, for example, mammal host immunisation or on fly vector competence modification via para‐transgenic approaches. Background: Significant progress has been made towards African sleeping sickness elimination in the last decade. Indeed, the World Health Organization (WHO) global goal of eliminating the chronic form of the disease as a public health problem was achieved in 2020 (i.e., < 2,000 new cases per year). Vector control has played an important role in achieving this goal. In this study, we evaluated the impact of the insecticide impregnated Tiny Targets on tsetse fly densities and their infection rates with Trypanosoma spp in the Campo sleeping sickness focus of South Cameroon. Methods: The study site was divided into two areas: (i) the south-west experimental area, which included vector control, and (ii) the eastern part as the non-intervention area. After compiling the baseline entomological data (tsetse densities and trypanosome infection rates), around 2000 Tiny Targets were deployed in the South-West area and replaced every six months for two years. Post-intervention surveys were conducted every six months to determine tsetse densities and levels of trypanosome infections with PCR-based methods. Results: Following the intervention, tsetse mean catches decreased by 61% after six months, and up to 73% after twelve months (pre-intervention: 2.48 flies/trap/day, 95%CI [1.92–3.14]; 12-months post-intervention: 0.66 tsetse/trap/day, 95%CI [0.42–0.94]). This decrease was not sustained after 18 months, and the mean catch doubled compared to that after 12 months. After 24 months, the mean catches still increased by 17% (18 months: 1.45 tsetse/trap/day, 95%CI [1.07–1.90] and 24 months: 1.71 tsetse/trap/day, 95%CI [1.27–2.24]). In the non-intervention area, a variation in tsetse catches was observed during the two years, with a general increase from 2.43 [0.73–5.77] to 3.64 [1.47–7.70] tsetse/trap/day. In addition, trypanosome infection rates dropped by 75% in both areas (P-value < 0.001) from 21.20% to 5.06% and from 13.14% to 3.45% in intervention and control areas respectively. Conclusion: Tiny targets have proven useful in reducing tsetse population densities and trypanosome infection rates, providing evidence for the integration of this tool in current strategies towards trypanosomiasis elimination in Campo. The non-sustained decrease of tsetse densities after one year may indicate reinvasions from neighbouring breeding sites or that the intervention area was not large enough. Our results show the need to scale up by accessing difficult breeding sites and extend the tiny targets to the whole transborder focus. During routine activities of farming, fishing, and hunting in the forest, villagers are in close contact with tsetse flies which bite them and transmit the disease. The tsetse fly display high densities in Campo, increasing the risk of disease transmission to the population. Recently, deltamethrin-treated blue-black fabric screens called Tiny Targets were developed to reduce tsetse densities when installed in the environment. These Tiny Targets significantly contributed to decrease sleeping sickness cases in many countries. In this study, baseline data were collected on tsetse densities in Campo and after population sensitization, around 2000 Tiny Targets were installed and maintained for two years with help of 10 villagers who received prior training. The Tiny Targets were replaced every six months and monitoring activities showed that tsetse density decreased by 73% after one year with a slight recovery in the second year. Trypanosome infections in tsetse flies also decreased by 75%. This study showed the positive impact of Tiny Targets in Campo in limiting trypanosome circulation and suggested to scale up the area covered by the tsetse control, to improve its efficiency. Background: African trypanosomiasis is a tsetse-borne parasitic infection that affects humans, wildlife, and domesticated animals. Tsetse flies are endemic to much of Sub-Saharan Africa and a spatial and temporal understanding of tsetse habitat can aid surveillance and support disease risk management. Problematically, current fine spatial resolution remote sensing data are delivered with a temporal lag and are relatively coarse temporal resolution (e.g., 16 days), which results in disease control models often targeting incorrect places. The goal of this study was to devise a heuristic for identifying tsetse habitat (at a fine spatial resolution) into the future and in the temporal gaps where remote sensing and proximal data fail to supply information. Methods: This paper introduces a generalizable and scalable open-access version of the tsetse ecological distribution (TED) model used to predict tsetse distributions across space and time, and contributes a geospatial Bayesian Maximum Entropy (BME) prediction model trained by TED output data to forecast where, herein the Morsitans group of tsetse, persist in Kenya, a method that mitigates the temporal lag problem. This model facilitates identification of tsetse habitat and provides critical information to control tsetse, mitigate the impact of trypanosomiasis on vulnerable human and animal populations, and guide disease minimization in places with ephemeral tsetse. Moreover, this BME analysis is one of the first to utilize cluster and parallel computing along with a Monte Carlo analysis to optimize BME computations. This allows for the analysis of an exceptionally large dataset (over 2 billion data points) at a finer resolution and larger spatiotemporal scale than what had previously been possible. Results: Under the most conservative assessment for Kenya, the BME kriging analysis showed an overall prediction accuracy of 74.8% (limited to the maximum suitability extent). In predicting tsetse distribution outcomes for the entire country the BME kriging analysis was 97% accurate in its forecasts. Conclusions: This work offers a solution to the persistent temporal data gap in accurate and spatially precise rainfall predictions and the delayed processing of remotely sensed data collectively in the − 45 days past to + 180 days future temporal window. As is shown here, the BME model is a reliable alternative for forecasting future tsetse distributions to allow preplanning for tsetse control. Furthermore, this model provides guidance on disease control that would otherwise not be available. These 'big data' BME methods are particularly useful for large domain studies. Considering that past BME studies required reduction of the spatiotemporal grid to facilitate analysis. Both the GEE-TED and the BME libraries have been made open source to enable reproducibility and offer continual updates into the future as new remotely sensed data become available. Simple Summary: Tsetse flies are large biting insects that inhabit much of tropical Africa and have a significant economic impact as the biological vectors of trypanosomes, which cause serious diseases to humans and livestock. A large array of bacteria, termed collectively symbionts, inhabit the internal organs of the flies' body. These bacterial symbionts are involved in important aspects of the flies' biology, including nutrition and reproduction. For instance, the main bacterial symbiont Wigglesworthia provides nutritional supplements necessary for host fertility and development, while Wolbachia is known to affect the reproduction of flies by causing a series of abnormalities. Therefore, the symbionts of tsetse flies show promising signs for exploitation and can be used for the development of innovative tools for the control of the flies and the diseases they carry. In this work, we used next-generation sequencing to characterize in detail the bacterial communities of four tsetse fly species. Their bacterial communities differed significantly, depending on the origin and the developmental stage of the flies. Certain important bacteria, such as Wigglesworthia and Sodalis, were present in all species and exhibited a high number of interactions with the other members of the bacterial community. Finally, Wolbachia was mostly present in G. morsitans samples. Tsetse flies (Glossina spp.; Diptera: Glossinidae) are viviparous flies that feed on blood and are found exclusively in sub-Saharan Africa. They are the only cyclic vectors of African trypanosomes, responsible for human African trypanosomiasis (HAT) and animal African trypanosomiasis (AAT). In this study, we employed high throughput sequencing of the 16S rRNA gene to unravel the diversity of symbiotic bacteria in five wild and three laboratory populations of tsetse species (Glossina pallidipes, G. morsitans, G. swynnertoni, and G. austeni). The aim was to assess the dynamics of bacterial diversity both within each laboratory and wild population in relation to the developmental stage, insect age, gender, and location. Our results indicated that the bacterial communities associated with the four studied Glossina species were significantly influenced by their region of origin, with wild samples being more diverse compared to the laboratory samples. We also observed that the larval microbiota was significantly different than the adults. Furthermore, the sex and the species did not significantly influence the formation of the bacterial profile of the laboratory colonies once these populations were kept under the same rearing conditions. In addition, Wigglesworthia, Acinetobacter, and Sodalis were the most abundant bacterial genera in all the samples, while Wolbachia was significantly abundant in G. morsitans compared to the other studied species. The operational taxonomic unit (OTU) co-occurrence network for each location (VVBD insectary, Doma, Makao, and Msubugwe) indicated a high variability between G. pallidipes and the other species in terms of the number of mutual exclusion and copresence interactions. The successful implementation of area-wide integrated tsetse control using the Sterile Insect Technique (SIT) relies on various factors. These include adapting and mass-producing the wild strain in the laboratory, suppressing the wild population in the target area, and releasing competent sterile male flies into the targeted area. Two important factors that can influence the effectiveness of this strategy are the presence of tsetse endosymbionts, specifically Sodalis glossinidius, and the infection of salivary gland hypertrophy virus (SGHV). SGHV infection directly hampers the expansion of Glossina pallidipes colonies in mass-rearing facilities by negatively impacting reproduction (fertility and fecundity), and overall growth. The viral infections in other tsetse species indirectly affect G. pallidipes by serving as a source of infection, as these species are less susceptible to clinical manifestations of the virus. The role of S. glossinidius in this context remains inconclusive despite previous laboratory and field studies, highlighting the need for further research. In addition to knowledge generation, it is crucial to introduce healthy fly populations from the wild into the insectary for sustainable mass-rearing production. In this study, our objective was to determine the prevalence of Sodalis, SGHV, and trypanosome infections in wild populations of tsetse fly G. pallidipes and investigate potential interactions among them. We analyzed 146 dissected midgut and mouthparts samples from non-teneral flies collected from the Makao Wildlife Management Area (WMA) interface. Our results revealed a prevalence of trypanosome infection at 12.3%, SGHV infection at 7.5%, and Sodalis at 51.4%. Co- existence of SGHV infection and Sodalis was observed in 9.6% of the flies. We did not find a statistically significant association of trypanosome infections occurrence with either SGHV infection or Sodalis. However, a significant association with trypanosome infection was observed when the both Sodalis and SGHV infection co-existed in the tsetse flies. Furthermore, we identified a negative correlation between Sodalis and SGHV infections. In conclusion, our study highlights the association between the co- existence of Sodalis and SGHV and the prevalence of trypanosome infections in wild populations of G. pallidipes. These findings contribute to our understanding of the interactions among these microbes, which is crucial for the development and implementation of effective tsetse control strategies using the Sterile Insect Technique (SIT). Thermal stress during development can prime animals to cope better with similar conditions in later life. Alternatively, negative effects of thermal stress can persist across life stages and result in poorer quality adults (negative carryover effects). As mean temperatures increase due to climate change, evidence for such effects across diverse taxa is required. Using Glossina morsitans morsitans, a species of tsetse fly and vector of trypanosomiasis, we asked whether (i) adaptive developmental plasticity allows flies to survive for longer under food deprivation when pupal and adult temperatures are matched; or (ii) temperature stress during development persists into adulthood, resulting in a greater risk of death. We did not find any advantage of matched pupal and adult temperature in terms of improved starvation tolerance, and no direct negative carryover effects were observed. There was some evidence for indirect carryover effects—high pupal temperature produced flies of lower body mass, which, in turn, resulted in greater starvation risk. However, adult temperature had the largest impact on starvation tolerance by far: flies died 60% faster at 31°C than those experiencing 25°C, consequently reducing survival time from a median of 8 (interquartile range (IQR) 7–9) to 5 (IQR 5–5.25) days. This highlights differences in temperature sensitivity between life stages, as there was no direct effect of pupal temperature on starvation tolerance. Therefore, for some regions of sub‐Saharan Africa, climate change may result in a higher mortality rate in emerging tsetse while they search for their first blood meal. This study reinforces existing evidence that responses to temperature are life stage specific and that plasticity may have limited capacity to buffer the effects of climate change. Background: Tsetse flies are cyclical vectors of African trypanosomiasis (AT). The flies have established symbiotic associations with different bacteria that influence certain aspects of their physiology. Vector competence of tsetse flies for different trypanosome species is highly variable and is suggested to be affected by bacterial endosymbionts amongst other factors. Symbiotic interactions may provide an avenue for AT control. The current study provided prevalence of three tsetse symbionts in Glossina species from Cameroon, Chad and Nigeria. Results: Tsetse flies were collected and dissected from five different locations. DNA was extracted and polymerase chain reaction used to detect presence of Sodalis glossinidius, Spiroplasma species and Wolbachia endosymbionts, using species specific primers. A total of 848 tsetse samples were analysed: Glossina morsitans submorsitans (47.52%), Glossina palpalis palpalis (37.26%), Glossina fuscipes fuscipes (9.08%) and Glossina tachinoides (6.13%). Only 95 (11.20%) were infected with at least one of the three symbionts. Among infected flies, six (6.31%) had Wolbachia and Spiroplasma mixed infection. The overall symbiont prevalence was 0.88, 3.66 and 11.00% respectively, for Sodalis glossinidius, Spiroplasma species and Wolbachia endosymbionts. Prevalence varied between countries and tsetse fly species. Neither Spiroplasma species nor S. glossinidius were detected in samples from Cameroon and Nigeria respectively. Conclusion: The present study revealed, for the first time, presence of Spiroplasma species infections in tsetse fly populations in Chad and Nigeria. These findings provide useful information on repertoire of bacterial flora of tsetse flies and incite more investigations to understand their implication in the vector competence of tsetse flies. Human African Trypanosomiasis (HAT) is still endemic in the Republic of Congo. Although the incidence of cases has significantly decreased over years, the disease still persists in some active foci. Factors contributing to the maintenance of the disease such as the existence of an animal reservoir or population knowledge are still not well known. It is in this context that a study focusing on the knowledge and practices of the population with regard to HAT as well as on the prevalence of trypanosomes infecting animals was undertaken in three active HAT foci in the Republic of Congo. The study was performed using field surveys conducted from November 2019 to June 2021. Domestic animal blood was examined by microscopy and PCR to detect the presence of trypanosomes. A structured questionnaire was administered to the population to assess their knowledge and practices concerning HAT in these endemic foci. More than half of the animals examined were found to be infected with trypanosomes (51.22%). The main trypanosome species infecting animals were Trypanosoma congolense savannah (67.2%) and Trypanosoma brucei (s.l.) (32.8%). No trypanosomes infecting humans were detected. Concerning household surveys, more than half of the respondents (52.9%) were fully aware of the mode of transmission and symptoms of the disease. The majority of people preferred to wear clothes covering the whole body and to use locally made soap as repellents to protect themselves from tsetse fly bites. This study suggests frequent circulation of animal trypanosomes in domestic animals and the use of personal measures to protect against tsetse fly bites. Updating information on the HAT animal reservoir and population knowledge alongside regular monitoring of the tsetse fly populations and the use of traps to control tsetse flies are crucial to drive efforts towards the elimination of gHAT in the Republic of Congo. This study aims to analyze, map, and identify the prevalence of, service provision for, and risk distribution and control for Human African Trypanosomosis (HAT), or sleeping sickness, in the endemic areas of Greater Equatoria Region (GER), including Eastern, Central, and Western Equatoria States of South Sudan. Passive and active screening data, detection data, and existing facilities and centers for sleeping sickness were used to assess the prevalence, screening coverage, and overall risk in the region for the 2016–2018 period. In addition, historical literature and surveillance information were used. The results show that 0.43% (N = 14,552) of the total at-risk population (N = 3,399,400) of GER were subjected to passive or active screening for Gambian HAT (gHAT), which showed an infection rate of 0.30%. Out of the total area of 196,211 km2, 58.77% of the region (115,311 km2) was found to be endemic to HAT. The population remains at high or very high risk for the disease in Western Equatoria State due to a number of active historic gHAT foci. With relative peace currently prevailing in the region, there is need to reinforce the leadership of South Sudan's health ministry with sufficient internal and external resources to support its activities. Abortion rates were assessed among 170, 846 tsetse (154,228 Glossina pallidipes and 19,618 Glossina morsitans morsitans) sampled in Zimbabwe in 1988–1999. The study produced improved estimates of abortion rates and how these varied with fly age and size and temperatures experienced during pregnancy. An abortion was diagnosed if the uterus was empty and the largest oocyte <0.82 of the expected mature length. Abortion rates for G. pallidipes and G. m. morsitans were 0.64% (95% ci: 0.59–0.69) and 0.83% (0.62–1.10) for trapped flies and 2.03% (1.77–2.31) and 1.55% (1.20–1.98) for flies from artificial refuges. Abortion rates increased with increasing temperature and decreased with increasing wing length and wing fray. Contrary to laboratory findings, abortion rates did not increase in the oldest flies. Percentages of tsetse with empty uteri, regardless of abortion status, were significantly higher than estimated abortion percentages. For tsetse from traps, 4.01% (95% ci: 3.90–4.13) of G. pallidipes and 2.52% (2.14–2.95) of G. m. morsitans had empty uteri; for flies from artificial refuges, the percentages were 12.69% (12.07–13.34) and 14.90% (13.82–16.02), respectively. Between 1990 and 1999, at Rekomitjie Research Station, Zambezi Valley, Zimbabwe, 29,360 female G. pallidipes were dissected to determine their ovarian category and trypanosome infection status. Overall prevalences were 3.45 and 2.66% for T. vivax and T. congolense , respectively, declining during each year as temperatures increased from July – December. Fits to age-prevalence data using Susceptible-Exposed-Infective (SEI) and SI compartmental models were statistically better than those obtained using a published catalytic model, which made the unrealistic assumption that no female tsetse survived more than seven ovulations. The improved models require knowledge of fly mortality, estimated separately from ovarian category distributions. Infection rates were not significantly higher for T. vivax than for T. congolense. For T. congolense in field-sampled female G. pallidipes , we found no statistical support for a model where the force of infection was higher at the first feed than subsequently. The long survival of adult female tsetse, combined with feeding at intervals ≤3 days, ensures that post-teneral feeds, rather than the first feed, play the dominant role in the epidemiology of T. congolense infections in G. pallidipes. This is supported by estimates that only about 3% of wild hosts at Rekomitjie were harbouring sufficient T. congolense to ensure that tsetse feeding off them take an infected meal, so that the probability of ingesting an infected meal is low at every meal. Swifts (Apodidae) are an unusual group of birds that spend most of their lives in flight, landing only when breeding. Although this aerial lifestyle greatly reduces their likelihood of being bitten by vectors and infected by vector-born parasites, swifts can still be heavily infested during breeding by nest-based vectors such as louse flies (Hippoboscidae). Here, we investigated host, vector, and vector-borne parasite relationships in the three most widespread swift species in the Western Palearctic (WP): common swifts (Apus apus), pallid swifts (A. pallidus), and alpine swifts (Tachymarptis melba), their nest-based louse flies (Crataerina pallida and C. melbae) and avian haemosporidians (genera Haemoproteus, Plasmodium, and Leucocytozoon). Studies of haemosporidian infections in Apodidae remain limited, with clear evidence of infection found to date in just four Neotropical and one Australasian species. The possible role of louse flies in transmitting haemosporidian infections has never been tested in swifts. We assessed the occurrence of haemosporidian infection by PCR screenings of DNA from blood samples from 34 common swifts and 44 pallid swifts from Italy, and 45 alpine swifts from Switzerland. We also screened 20 ectoparasitic louse flies present on 20 birds and identified them by both morphological features and cytochrome oxidase subunit 1 (COI) barcodes. Our results provide no evidence of haemosporidian infection in the 123 swifts tested or in the two louse fly species we identified. Our findings are consistent with available knowledge showing no haemosporidian occurrence in WP swift species and that the most likely infection route for these highly aerial species (via louse fly ectoparasites during nesting) is unlikely. Background: Tsetse-transmitted African trypanosomiasis is a debilitating and fatal disease of humans and livestock if left untreated. While knowledge of the spatial distribution patterns of tsetse is essential for the development of risk-based vector control strategies, existing distribution maps in Zambia are more than 40 years old and were based on coarse spatial resolution data. The recently developed vehicle-mounted sticky trap (VST) provides an alternative sampling device to aid in updating existing distribution maps but has not been applied outside an experimental setting and is limited to motorable tracks. Therefore, the objective of the present study was to demonstrate the effectiveness of utilizing the VST for area-wide surveys of Glossina morsitans and to use the occurrence records to predict its spatial distribution in Zambia under current environmental conditions using Maxent. Methodology/Principal findings: Two-sided all-blue VST baited with butanone and 1-octen-3-ol was used to survey 692 and 1020 km of transect routes in G. m. centralis Machado and G. m. morsitans Westwood previously published distribution in Zambia. Maxent species distribution technique was used to predict the potential distribution of the two subspecies using current climatic and environmental data which was then compared to the historical distribution. A total of 15,602 tsetse were captured with G. m. morsitans (58%) being the most abundant. G. m. centralis and G. pallidipes Austin represented 39 and 2% of the catch respectively, and G. brevipalpis Newstead was also detected. The predicted potential distribution for G. m. centralis was 80,863 km2 while that of G. m. morsitans was 70,490 km2 representing a 47 and 29% reduction compared to their historical distributions, respectively. Conclusion/Significance: The VST is effective for sampling G. morsitans outside experimental settings and is recommended for use as an additional tsetse survey tool. The spatial distribution of G. morsitans in Zambia has reduced by 101,051 km2 due to temperature and land cover changes. Camel trypanosomosis is a life-threatening disease with adverse effects on camel health, production, and working efficiency. Despite this, camel trypanosomosis has received much less attention in Ethiopia compared with the disease in cattle and other animals. This cross-sectional study was conducted to evaluate the prevalence of camel trypanosomosis, identify the potential risk factors, and determine the importance of trypanosomosis in causing anemia in camels in the Gorodola and Liben districts in the Guji Zone of Oromia Regional State in Ethiopia. To this end, blood samples were collected from randomly selected 450 camels in heparinized capillary tubes and analyzed for the presence of Trypanosoma evansi using the buffy coat technique and Giemsa-stained thin smears. T. evansi infection was detected in 24 (5.3%) of the 450 camels examined. Out of the four variables analyzed in this study, two factors, such as body condition (BC) score and age, were found to be significantly (P < 0.05) associated with trypanosomosis in camels. A higher prevalence of trypanosomosis was observed in camels in poor BC (13.22%) than in camels in good (4.62%) or moderate (1.01%) BC. Likewise, adult camels (8.09%) were infected more frequently than young camels (1.12%), whereas no trypanosomes were detected in camel calves under 2 years of age. No significant statistical difference was found between the two districts, and male and female camels (P > 0.05). Statistically, the mean packed cell volume was significantly lower (P < 0.05) in parasitemic camels (26.5 % ± 7) as compared with aparasitemic camels (33.9 % ± 9.1). In conclusion, the current study conducted during a dry season showed a moderate prevalence of trypanosomosis in camels. Further studies using more sensitive and specific diagnostic tests, such as miniature anion-exchange centrifugation technique, serology, or molecular tests, are needed to establish a true epidemiological dataset on the prevalence and seasonality of the disease and its vectors in the study area to recommend viable control measures. Pathogen transfers between wild and domestic animals and between animals and humans are increasing. Their dramatic consequences for public and veterinary health as well as for conservation call for innovative and user-friendly methods for pathogen surveillance in wildlife. Xenosurveillance, a method based on the use of invertebrates (e.g., mosquitoes, hematophagous flies, leeches, cadaveric arthropods) to sample animal tissues (e.g., blood) and the associated pathogens, is one of these tools. Previously, we demonstrated that hematophagous flies, such as tsetse flies, could be useful to detect and identify the etiological agents of malaria in a diverse range of mammals in Gabon. However, we did not assess whether this method can be also used to detect viruses. In the present study, we experimentally fed tsetse flies (Glossina fuscipes fuscipes) rabbit blood containing different viruses of medical or veterinary importance (Zika, Dengue, Chikungunya, African swine fever, Bluetongue, and peste des petits ruminants viruses). Then, we used quantitative PCR (i) to determine for how long viral nucleic acid fragments remained detectable in the tsetse midgut during blood digestion and (ii) to compare two blood meal preservation methods (i.e., FTA cards and RNAlater solution) tested using tsetse flies engorged with blood and dengue-2 virus. All viruses remained detectable for 6 days after feeding, although the detection probability significantly decreased over time. FTA cards and RNAlater solution gave similar results in terms of virus detection. Background: Tsetse-transmitted African animal trypanosomiasis is recognised as an important disease of ruminant livestock in sub-Saharan Africa, but also affects domestic pigs, with Trypanosoma simiae notable as a virulent suid pathogen that can rapidly cause death. Trypanosoma simiae is widespread in tsetse-infested regions, but its biology has been little studied compared to T. brucei and T. congolense. Methods: Trypanosoma simiae procyclics were cultured in vitro and transfected using protocols developed for T. brucei. Genetically modified lines, as well as wild-type trypanosomes, were transmitted through tsetse flies, Glossina pallidipes, to study T. simiae development in the tsetse midgut, proventriculus and proboscis. The development of proventricular trypanosomes was also studied in vitro. Image and mensural data were collected and analysed. Results: A PFR1::YFP line successfully completed development in tsetse, but a YFP::HOP1 line failed to progress beyond midgut infection. Analysis of image and mensural data confirmed that the vector developmental cycles of T. simiae and T. congolense are closely similar, but we also found putative sexual stages in T. simiae, as judged by morphological similarity to these stages in T. brucei. Putative meiotic dividers were abundant among T. simiae trypanosomes in the proboscis, characterised by a large posterior nucleus and two anterior kinetoplasts. Putative gametes and other meiotic intermediates were also identified by characteristic morphology. In vitro development of proventricular forms of T. simiae followed the pattern previously observed for T. congolense: long proventricular trypanosomes rapidly attached to the substrate and shortened markedly before commencing cell division. Conclusions: To date, T. brucei is the only tsetse-transmitted trypanosome with experimentally proven capability to undergo sexual reproduction, which occurs in the fly salivary glands. By analogy, sexual stages of T. simiae or T. congolense are predicted to occur in the proboscis, where the corresponding portion of the developmental cycle takes place. While no such stages have been observed in T. congolense, for T. simiae putative sexual stages were abundant in the tsetse proboscis. Although our initial attempt to demonstrate expression of a YFP-tagged, meiosis-specific protein was unsuccessful, the future application of transgenic approaches will facilitate the identification of meiotic stages and hybrids in T. simiae. Morphometric analysis of wings has been suggested for identifying and controlling isolated populations of tsetse (Glossina spp), vectors of human and animal trypanosomiasis in Africa. Single-wing images were captured from an extensive data set of field-collected tsetse wings of species Glossina pallidipes and G. m. morsitans. Morphometric analysis required locating 11 anatomical landmarks on each wing. The manual location of landmarks is time-consuming, prone to error, and infeasible for large data sets. We developed a two-tier method using deep learning architectures to classify images and make accurate landmark predictions. The first tier used a classification convolutional neural network to remove most wings that were missing landmarks. The second tier provided landmark coordinates for the remaining wings. We compared direct coordinate regression using a convolutional neural network and segmentation using a fully convolutional network for the second tier. For the resulting landmark predictions, we evaluate shape bias using Procrustes analysis. We pay particular attention to consistent labelling to improve model performance. For an image size of 1024 × 1280, data augmentation reduced the mean pixel distance error from 8.3 (95% confidence interval [4.4,10.3]) to 5.34 (95% confidence interval [3.0,7.0]) for the regression model. For the segmentation model, data augmentation did not alter the mean pixel distance error of 3.43 (95% confidence interval [1.9,4.4]). Segmentation had a higher computational complexity and some large outliers. Both models showed minimal shape bias. We deployed the regression model on the complete unannotated data consisting of 14,354 pairs of wing images since this model had a lower computational cost and more stable predictions than the segmentation model. The resulting landmark data set was provided for future morphometric analysis. The methods we have developed could provide a starting point to studying the wings of other insect species. We use a two-step approach, first removing wings that are missing landmarks due to damage. We thereby provide accurate landmarks, unbiased for wing shape. We compared two modern deep-learning models to locate landmarks and showed how data manipulation techniques improve performance. We deployed the final system to generate the landmark data for the tsetse wing images and provide the trained models for transfer learning on similar tasks with smaller data sets. Our methods will allow researchers to study tsetse wing shape in relation to other biological data and to gauge how wing shape characterises tsetse populations. Controlling tsetse flies is critical for effective management of African trypanosomiasis in Sub-Saharan Africa. To enhance timely and targeted deployment of tsetse control strategies a better understanding of their temporal dynamics is paramount. A few empirical studies have explained and predicted tsetse numbers across space and time, but the resulting models may not easily scale to other areas. We used tsetse catches from 160 traps monitored between 2017 and 2019 around Shimba Hills National Reserve in Kenya, a known tsetse and trypanosomiasis hotspot. Traps were divided into two groups: proximal (<1.0 km)) to and distant (> 1.0 km) from the outer edge of the reserve boundary. We fitted zero-inflated Poisson and generalized linear regression models for each group using as temporal predictors rainfall, NDVI (Normalized Difference Vegetation Index), and LST (land surface temperature). For each predictor, we assessed their relationship with tsetse abundance using time lags from 10 days up to 60 days before the last tsetse collection date of each trap. Tsetse numbers decreased as distance from the outside of reserve increased. Proximity to croplands, grasslands, woodlands, and the reserve boundary were the key predictors for proximal traps. Tsetse numbers rose after a month of increased rainfall and the following increase in NDVI values but started to decline if the rains persisted beyond a month for distant traps. Specifically, tsetse flies were more abundant in areas with NDVI values greater than 0.7 for the distant group. The study suggests that tsetse control efforts beyond 1.0 km of the reserve boundary should be implemented after a month of increased rains in areas having NDVI values greater than 0.7. To manage tsetse flies effectively within a 1.0 km radius of the reserve boundary, continuous measures such as establishing an insecticide-treated trap or target barrier around the reserve boundary are needed. Limited reliable information on tsetse temporal dynamics hinders the implementation of effective control strategies. We used two empirical modelling strategies (zero-inflated Poisson and generalized linear Poisson) to link tsetse abundance with environmental and weather data, to understand and explain tsetse temporal dynamics in a heterogeneous environment. Tsetse numbers were consistently high within 1.0 km of the reserve outer boundary due to favorable breeding sites, resting sites, and host availability. Beyond 1.0 km of the reserve boundary, tsetse numbers increased after a month of increased rainfall, but started to decline if rainfall persisted beyond a month. For these localities, higher tsetse numbers were also found in areas with NDVI values above 0.7. African trypanosomes proliferate as bloodstream forms (BSFs) and procyclic forms in the mammal and tsetse fly midgut, respectively. This allows them to colonise the host environment upon infection and ensure life cycle progression. Yet, understanding of the mechanisms that regulate and drive the cell replication cycle of these forms is limited. Using single-cell transcriptomics on unsynchronised cell populations, we have obtained high resolution cell cycle regulated (CCR) transcriptomes of both procyclic and slender BSF Trypanosoma brucei without prior cell sorting or synchronisation. Additionally, we describe an efficient freeze-thawing protocol that allows single-cell transcriptomic analysis of cryopreserved T. brucei. Computational reconstruction of the cell cycle using periodic pseudotime inference allowed the dynamic expression patterns of cycling genes to be profiled for both life cycle forms. Comparative analyses identify a core cycling transcriptome highly conserved between forms, as well as several genes where transcript levels dynamics are form specific. Comparing transcript expression patterns with protein abundance revealed that the majority of genes with periodic cycling transcript and protein levels exhibit a relative delay between peak transcript and protein expression. Camels are considered an important food source in North Africa. Trypanosomiasis in camels is a life-threatening disease that causes severe economic losses in milk and meat production. Therefore, the objective of this study was to determine the trypanosome genotypes in the North African region. Trypanosome infection rates were determined by microscopic examination of blood smears and polymerase chain reaction (PCR). In addition, total antioxidant capacity (TAC), lipid peroxides (MDA), reduced glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) were determined in erythrocyte lysate. Furthermore, 18S amplicon sequencing was used to barcode and characterizes the genetic diversity of trypanosome genotypes in camel blood. In addition to Trypanosoma, Babesia and Thelieria were also detected in the blood samples. PCR showed that the trypanosome infection rate was higher in Algerian samples (25.7%) than in Egyptian samples (7.2%). Parameters such as MDA, GSH, SOD and CAT had significantly increased in camels infected with trypanosomes compared to uninfected control animals, while TAC level was not significantly changed. The results of relative amplicon abundance showed that the range of trypanosome infection was higher in Egypt than in Algeria. Moreover, phylogenetic analysis showed that the Trypanosoma sequences of Egyptian and Algerian camels are related to Trypanosoma evansi. Unexpectedly, diversity within T. evansi was higher in Egyptian camels than in Algerian camels. African sleeping sickness, Chagas disease, and leishmaniasis are life-threatening diseases that together affect millions of people around the world and are caused by different members of the protozoan family Trypanosomatidae. The most studied member of the family is Trypanosoma brucei , which is spread by tsetse flies and causes African sleeping sickness. Nucleotide metabolism in T. brucei and other trypanosomatids is significantly different from that of mammals and was recognized as a target for chemotherapy already in the 1970–1980s. A more thorough investigation of the nucleotide metabolism in recent years has paved the way for identifying nucleoside analogues that can cure T. brucei brain infections in animal models. Specific features of T. brucei nucleotide metabolism include the lack of de novo purine biosynthesis, the presence of very efficient purine transporters, the lack of salvage pathways for CTP synthesis, unique enzyme localizations, and a recently discovered novel pathway for dTTP synthesis. This review describes the nucleotide metabolism of T. brucei , highlights differences and similarities to other trypanosomatids, and discusses how to exploit the parasite-specific features for drug development. Tsetse flies significantly impact public health and economic development in sub-Saharan African countries by transmitting the fatal disease African trypanosomiasis. Unusually, instead of laying eggs, tsetse birth a single larva that immediately burrows into the soil to pupate. Where the female chooses to larviposit is, therefore, crucial for offspring survival. Previous laboratory studies suggested that a putative larval pheromone, n-pentadecane, attracts gravid female Glossina morsitans morsitans to appropriate larviposition sites. However, this attraction could not be reproduced in field experiments. Here, we resolve this disparity by designing naturalistic laboratory experiments that closely mimic the physical characteristics found in the wild. We show that gravid G. m. morsitans were neither attracted to the putative pheromone nor, interestingly, to pupae placed in the soil. By contrast, females appear to choose larviposition sites based on environmental substrate cues. We conclude that, among the many cues that likely contribute to larviposition choice in nature, substrate features are a main determinant, while we failed to find evidence for a role of pheromones. Simple Summary: Tsetse flies transmit Trypanosoma parasites, which cause severe diseases in humans and animals. To reduce the number of tsetse flies, male flies are sterilized through irradiation and released into the field to compete with wild male flies for mating with wild females. Recently, two insect-specific viruses were discovered in mass-reared Glossina morsitans morsitans tsetse flies. The aim of this study was to evaluate whether these viruses are affected by the irradiation treatment. The results showed that irradiation did not change the number of viral genome copies in the flies. Tsetse flies are cyclic vectors of Trypanosoma parasites, which cause debilitating diseases in humans and animals. To decrease the disease burden, the number of flies is reduced using the sterile insect technique (SIT), where male flies are sterilized through irradiation and released into the field. This procedure requires the mass rearing of high-quality male flies able to compete with wild male flies for mating with wild females. Recently, two RNA viruses, an iflavirus and a negevirus, were discovered in mass-reared Glossina morsitans morsitans and named GmmIV and GmmNegeV, respectively. The aim of this study was to evaluate whether the densities of these viruses in tsetse flies are affected by the irradiation treatment. Therefore, we exposed tsetse pupae to various doses (0–150 Gy) of ionizing radiation, either in air (normoxia) or without air (hypoxia), for which oxygen was displaced by nitrogen. Pupae and/or emerging flies were collected immediately afterwards, and at three days post irradiation, virus densities were quantified through RT-qPCR. Generally, the results show that irradiation exposure had no significant impact on the densities of GmmIV and GmmNegeV, suggesting that the viruses are relatively radiation-resistant, even at higher doses. Social motility (SoMo) is the ability of Trypanosoma brucei procyclic forms to generate and respond to pH gradients. The differential responses of early and late procyclic forms to pH results in the self-organisation of communities on semi-solid surfaces. pH gradients occur in the tsetse fly host and might be used by the parasite to guide migration. pH sensing requires specific adenylate cyclases and a cAMP response protein, CARP3. T. brucei encodes a large number of adenylate cyclases that are likely to sense other environmental signals, enabling parasites to home into specific tissues in both hosts. Recent studies showed that the formation of elegant geometric patterns by communities of Trypanosoma brucei on semi-solid surfaces, dubbed social motility (SoMo) by its discoverers, is a manifestation of pH taxis. This is caused by procyclic forms generating and responding to pH gradients through glucose metabolism and cAMP signalling. These findings established that trypanosomes can sense and manipulate gradients, potentially helping them to navigate through host tissues. At the same time, the host itself and bystanders such as endosymbionts have the potential to shape the environment and influence the chances of successful transmission. Background: Human African trypanosomiasis (HAT) is a neglected tropical disease caused by Trypanosoma brucei gambiense transmitted by tsetse flies in sub-Saharan West Africa. In southern Chad the most active and persistent focus is the Mandoul focus, with 98% of the reported human cases, and where African animal trypanosomosis (AAT) is also present. Recently, a control project to eliminate tsetse flies (Glossina fuscipes fuscipes) in this focus using the sterile insect technique (SIT) was initiated. However, the release of large numbers of sterile males of G. f. fuscipes might result in a potential temporary increase in transmission of trypanosomes since male tsetse flies are also able to transmit the parasite. The objective of this work was therefore to experimentally assess the vector competence of sterile males treated with isometamidium for Trypanosoma brucei brucei. Methods: An experimental infection was set up in the laboratory, mimicking field conditions: the same tsetse species that is present in Mandoul was used. A T. b. brucei strain close to T. b. gambiense was used, and the ability of the sterile male tsetse flies fed on blood with and without a trypanocide to acquire and transmit trypanosomes was measured. Results: Only 2% of the experimentally infected flies developed an immature infection (midgut) while none of the flies developed a metacyclic infection of T. b. brucei in the salivary glands. We did not observe any effect of the trypanocide used (isometamidium chloride at 100 mg/l) on the development of infection in the flies. Conclusions: Our results indicate that sterile males of the tested strain of G. f. fuscipes were unable to cyclically transmit T. b. brucei and might even be refractory to the infection. The data of the research indicate that the risk of cyclical transmission of T. brucei by sterile male G. f. fuscipes of the strain colonized at IAEA for almost 40 years appears to be small. African trypanosomosis remains one of the major diseases affecting food security and human healthy living in sub-Saharan Africa. This study screened domesticated animals for the presence of pathogenic trypanosomes in 1,050 slaughtered animals comprising 858 and 100 cattle respectively from Zango and Kawo abattoirs and 92 camels from Zango as Kawo abattoir neither slaughters nor sell camel meat. Both abattoirs are government-owned and located in Kaduna metropolis, Kaduna State, Nigeria. The animals examined were sourced from Bauchi, Yobe, Katsina, Kaduna, Kano, and Chad Republic. Blood samples were collected from each animal at the point of slaughter and examined using the buffy coat technique and Leishman-stained thick film for trypanosome identification. Packed cell volume (PCV) determination was conducted to assess the level of anaemia. Results indicated that 25 cattle and 2 camels (27) had an overall trypanosome infection rate of 2.57%. The trypanosomes observed were mainly Trypanosoma brucei, Trypanosoma congolense, and Trypanosoma vivax. This study showed that sources of livestock slaughtered at various abattoirs can be a useful indicator in quantifying trypanosomosis risk. Continuous sensitization and awareness creation among the herders at the grass-root level is recommended for effective disease management towards increased livestock production and productivity required to sustain the rural economy and improve the livelihoods of the local populace. During infection of mammalian hosts, African trypanosomes thwart immunity using antigenic variation of the dense Variant Surface Glycoprotein (VSG) coat, accessing a large repertoire of several thousand genes and pseudogenes, and switching to antigenically distinct copies. The parasite is transferred to mammalian hosts by the tsetse fly. In the salivary glands of the fly, the pathogen adopts the metacyclic form and expresses a limited repertoire of VSG genes specific to that developmental stage. It has remained unknown whether the metacyclic VSGs possess distinct properties associated with this particular and discrete phase of the parasite life cycle. We present here three novel metacyclic form VSG N-terminal domain crystal structures (mVSG397, mVSG531, and mVSG1954) and show that they mirror closely in architecture, oligomerization, and surface diversity the known classes of bloodstream form VSGs. These data suggest that the mVSGs are unlikely to be a specialized subclass of VSG proteins, and thus could be poor candidates as the major components of prophylactic vaccines against trypanosomiasis. Author summary: The African trypanosome is a single-celled parasite causing African Sleeping Sickness in humans and related diseases in animals. It has a unique protein coat made up mostly of a single protein, called the Variant Surface Glycoprotein, or VSG. The African trypanosome possesses several thousand different VSG genes. As the immune system of an infected host learns to recognize one coat protein and kill the trypanosomes, the parasite switches to a different VSG protein that is not recognized by the immune system. This allows the parasite to keep thriving in the host as this process repeats. This paper presents molecular images of a subset of VSG proteins that are on the surface of the organism when it is transferred to hosts by the bite of the tsetse fly—the first step in infection. We show that these so-called metacyclic VSGs are neither special nor unique compared to those VSGs that take over in the bloodstream during the course of infection. Background: Trypanosomes are protozoan parasites of vertebrates that are of medical and veterinary concern. A variety of blood-feeding invertebrates have been identified as vectors, but the role of ticks in trypanosome transmission remains unclear. Methods: In this study, we undertook extensive molecular screening for the presence and genetic diversity of trypanosomes in field ticks. Results: Examination of 1089 specimens belonging to 28 tick species from Europe and South America led to the identification of two new trypanosome strains. The prevalence may be as high as 4% in tick species such as the castor bean tick Ixodes ricinus, but we found no evidence of transovarial transmission. Further phylogenetic analyses based on 18S rRNA, EF1-α, hsp60 and hsp85 gene sequences revealed that different tick species, originating from different continents, often harbour phylogenetically related trypanosome strains and species. Most tick-associated trypanosomes cluster in a monophyletic clade, the Trypanosoma pestanai clade, distinct from clades of trypanosomes associated with transmission by other blood-feeding invertebrates. Conclusions: These observations suggest that ticks may be specific arthropod hosts for trypanosomes of the T. pestanai clade. Phylogenetic analyses provide further evidence that ticks may transmit these trypanosomes to a diversity of mammal species (including placental and marsupial species) on most continents. South Africa's research incentive system rewards faculty members and affiliates for publishing in outlets that are "accredited" by the Department of Higher Education and Training. This arguably perverse incentive makes academic research a potentially aggressive numbers game. It is compounded by factors such as undue delay in peer review, difficulty in securing expert evaluators, and poor understanding of who constitutes a "peer". Despite the "publish or perish" pressure on researchers and the prohibition of the parallel submission of manuscripts by many journals, there is negligible research on publishing problems in South African journals. Informed by a literature survey, editorial experience and conversations with colleagues, this article seeks a dialogue about these problems, which it characterises as a tsetse fly perched on the scrotum. On the one hand, the current incentive system commodifies outputs and diverts attention from building knowledge with socially responsive research to building the financial coffers of universities and authors. On the other hand, inappropriate editorial interpretations of a "peer" and the gratis nature of peer evaluation encumber the publishing process. We argue that this situation erodes academic excellence, encourages the growth of predatory journals, and potentially harms national development. [ABSTRACT FROM AUTHOR]

Simple Summary: The sterile insect technique (SIT) is an environment-friendly, species-specific pest control method by which target insects are mass-produced in a factory and are made infertile by irradiation—usually with gamma rays. However, gamma sources are becoming more difficult and expensive to purchase, and the regulations surrounding these types of irradiators are becoming stricter. Therefore, there is now increasing interest in alternatives, such as X-ray irradiators. Following a recent technical evaluation of a blood X-ray unit, the aim of this research was to assess the biological responses of a selection of major SIT target insect species to irradiation in the X-ray unit as compared to gamma ray irradiation. It was found that all the insects responded similarly to X-rays as to gamma rays and that the X-ray unit is suitable for small- to medium-sized SIT programs. The sterile insect technique (SIT) is based on the inundatory field release of a target pest following their reproductive sterilization via exposure to radiation. Until recently, gamma irradiation from isotopic sources has been the most widely used in SIT programs. As isotopic sources are becoming increasingly expensive, especially for small programs, and regulations surrounding their procurement and shipment increasingly strict, irradiation capacity is one of the limiting factors in smaller or newly developing SIT projects. For this reason, the possibility of using X-ray irradiators has been evaluated in the recent decade. The availability of "off-the-shelf" blood X-ray irradiators that meet the technical requirements for insect irradiation can provide irradiation capacity for those SIT projects in which the acquisition of gamma ray irradiators is not feasible. Following the recent technical characterization of a Raycell MK2 X-ray blood irradiator, it was found in this study, that MK2 instruments were suitable for the sterilization of fruit flies, tsetse flies and mosquitoes, inducing comparable, even slightly higher, sterility levels compared to those achieved by gamma ray irradiation. Background: With the largest cattle population in Africa and vast swathes of fertile lands infested by tsetse flies, trypanosomosis is a major challenge for Ethiopian farmers. Managing the problem strategically and rationally requires comprehensive and detailed information on disease and vector distribution at the national level. To this end, the National Institute for Control and Eradication of Tsetse and Trypanosomosis (NICETT) developed a national atlas of tsetse and African animal trypanosomosis (AAT) for Ethiopia. Methods: This first edition of the atlas focused on the tsetse-infested areas in western Ethiopia. Data were collected between 2010 and 2019 in the framework of national surveillance and control activities. Over 88,000 animals, mostly cattle, were tested with the buffy-coat technique (BCT). Odour-enhanced traps were deployed in approximately 14,500 locations for the entomological surveys. Animal- and trap-level data were geo-referenced, harmonized and centralized in a single database. Results: AAT occurrence was confirmed in 86% of the districts surveyed (107/124). An overall prevalence of 4.8% was detected by BCT in cattle. The mean packed cell volume (PCV) of positive animals was 22.4, compared to 26.1 of the negative. Trypanosoma congolense was responsible for 61.9% of infections, T. vivax for 35.9% and T. brucei for 1.7%. Four tsetse species were found to have a wide geographic distribution. The highest apparent density (AD) was reported for Glossina pallidipes in the Southern Nations, Nationalities and People's Region (SNNPR) (3.57 flies/trap/day). Glossina tachinoides was the most abundant in Amhara (AD 2.39), Benishangul-Gumuz (2.38), Gambela (1.16) and Oromia (0.94) regions. Glossina fuscipes fuscipes and G. morsitans submorsitans were detected at lower densities (0.19 and 0.42 respectively). Only one specimen of G. longipennis was captured. Conclusions: The atlas establishes a reference for the distribution of tsetse and AAT in Ethiopia. It also provides crucial evidence to plan surveillance and monitor control activities at the national level. Future work on the atlas will focus on the inclusion of data collected by other stakeholders, the broadening of the coverage to tsetse-free areas and continuous updates.
