Mutations in KRAS are among the most common aberrations in cancer. However, despite considerable research efforts, KRAS remains a challenging therapeutic target. In recent years, there has been a drive to develop KRAS mutant specific drugs. Among the different known mutations, the KRASG12C (glycine 12 to cysteine) has been considered druggable. Studies have shown that due in part to the close proximity of Cysteine 12 to both the nucleotide pocket and the switch regions, thiol-reactive compounds can bind to the active site covalently and inhibit KRASG12C mutation-driven signaling. The absence of this particular cysteine residue in wild-type KRAS makes such an approach very selective towards cancer cells. We have discovered that derivatives of 6-(naphthale-1-yl)-5,6-dihydro-2H-pyran-2-one (klavuzon) have potent inhibitory effects over KRASG12C due to their thiol-reactive property.

p53 ve MDM2 arasındaki etkileşimi engellemek için farmakofor tasarımı kanser terapisinde yeni bir yaklaşım olmaktadır. Genomum koruyucusu olarak bilinen p53 stres altında hücre siklusunda aresti, apaptozu ve DNA tamirini kontrol eder. Bununla birlikte, MDM2 fazla eksprese edildiğinde hücrenin çabuk çoğalmasına ve neticede tümör oluşumuna neden olur.p53 ve MDM2 arasındaki geribesleme mekanizması p53'ün, MDM2'nin N-ucundaki Phe19, Trp23 ve Leu26 aminoasitlerinden oluşan hidrofobik boşluktan etkileşir. Bu çalışmada asimetrik sentez ile yeni kiral 1,4-oksazepan-5-on türevlerinin sentezlenmesi amaçlanmıştır. (R)-2-amino-2-(4-chlorophenyl)acetik asit başlangıç maddesi olarak kullanıldı. Amino grubu tritil grubu ile korundu ve daha sonra maddenin karboksilik asit kısmı LiAlH4 ile N-korunmalı amino alkol elde edmek üzere indirgendi. Dess Martin periodinane alkolün aldehite oksidasyon için kullanıldı, daha sonra 3-klorofenilmagnezyum bromür Grignard reaksiyonu ile aldehite eklendi. Koruyucu tritil grubu TFA ile uzaklaştırıldı ve oluşan aminoalkolün farklı α,β-doymamış karboksilik asit ile bağlanma reaksiyonları HATU ve DIPEA kullanılarak gerçekleştirildi. Tüm denemelere rağmen yedi üyeli 1,4-oksazepan-5-on halkalaşması hiç bir zaman gerçekleştirilemedi. Nonetheless, when over-expressed, MDM2 causes proliferation in the cell and eventually tumorgenesis.The feedback mechanism between p53 and MDM2 arises from the interaction of p53 through the hydrophobic cleft which consists of Phe19, Trp23 and Leu26 aminoacids in the N-terminal of MDM2.In this study, it was aimed to synthesize a new, chiral 1,4-oxazepan-5-one derivatives by asymmetric synthesis. (R)-2-amino-2-(4-chlorophenyl)acetic acid was used as starting material. Amino group was protected by trityl group then the carboxylic acid part was reduced by LiAlH4 to produce N-trityl-protected amino alcohol. Dess Martin periodinane was used for the oxidation of the alcohol to the aldehyde, then 3-chlorophenylmagnesium bromide was added to the aldehyde by Grignard reaction. Deprotection of N-trityl was performed with TFA then, coupling reactions of produced aminoalcohol with different α,β-unsaturated carboxylic acids were performed by HATU and DIPEA. Despite all of the attempts, cyclization to seven membered 1,4-oxazepan-5-one ring was never achieved. 69

In this study, three novel polyoxotungstate clusters were synthesized using hydrothermal method and catalytic activities of these three novel polyoxometalates and in addition two crystals previously synthesized in our group were studied. The first compound, [(4,4’-bpyH2)3][PCuW11O39] yellow crystals, crystallize in the space group P2(1)/n of the monoclinic system with four formula units in a cell. The crystal is a Keggin polyoxometalate and includes free 4,4’-bipyridine groups between the clusters. The second compound, (4,4’-bpyH2)[H2PW12O40]2.H2O colorless crystals, crystallize in the space group Ia-3 of the cubic system with twelve formula units in a cell. The crystal is a Keggin polyoxometalate and consists of two polyoxotungstate clusters, a free 4,4’-bipyridine group and a water molecule. The third and the last novel compound, (4,4’-bpyH2)4[H2P2W18O62]2 dark yellow crystals, crystallize in the space group P-1 of the triclinic system with four formula units in a cell. The crystal is a Wells-Dawson polyoxometalate and contains two polyoxotungstate clusters and free 4,4’-bipyridine groups between clusters. The catalytic study has been carried out using starch hydrolysis reactions by using hydrothermal synthesis and five different crystals were tried and conversion results were obtained. Starch samples converted to D-glucose with glucose yields above 90 wt.%. Catalyst reusability was performed for each crystal. No appreciable loss was observed in activity after five reaction cycles for each crystal., Son zamanlarda, birçok alanda farklı uygulamalarının olması özellikle kimyasal reaksiyonlarda katalizör olarak kullanılmalarından dolayı birçok bilim adamı polioksometal kümeleri içeren organik-inorganik melez malzemelerin hidrotermal sentezi ve karakterizasyonu ile ilgilenmektedir. Bu çalışmada, üç adet yeni polioksotungstat kümesi hidrotermal sentez ile sentezlenmiş ve bu yeni bileşikler ile birlikte iki adet daha önce grubumuzda sentezlenmiş olan kristallerin katalitik aktiviteleri çalışılmıştır. İlk bileşik, [(4,4’-bpyH2)3][PCuW11O39] sarı kristalleri, monoklinik sistemin P2(1)/n uzay grubunda olup, dört formül birimi içeren birim hücrede kristallenmiştir. Kristal Wells-Dawson polioksometal yapısına sahiptir ve iki adet polioksotungstat kümesi ile kümeler arasında serbest halde bulunan 4,4’-bipiridin gruplarını içermektedir. Katalitik çalışmada, hidrotermal sentez ve nişasta hidroliz reaksiyonları ile beş farklı kristal kullanılmış ve dönüşüm sonuçları elde edilmiştir. Nişasta örnekleri kütlece %90 glikoz verimi ile D-glikoza dönüşmüştür. Her bir kristal için yeniden kullanılabilirlik denemeleri yapılmıştır. Beş reaksiyon döngüsü sonucunda her bir kristalin katalitik aktivitesinde kayda değer bir azalışa rastlanmamıştır.

Biyotransformasyon, enzimler ya da canlı organizmalar kullanılarak meydana getirilen kimyasal modifikasyonlardır. Enzimlerin izolasyonu ve saflaştırılmasının zor ve yüksek maliyetli olması, bütün hücre sistemlerinin biyokatalizör olarak kullanımını daha avantajlı kılmaktadır. Mikrobiyal biyokatalistler; steroitler ve triterpenoitler gibi kompleks moleküllerin modifikasyonunda, kimyasal reaksiyonlarla gerçekleştirilmesi zor ya da mümkün olmayan stereoselektif ve regioselektif reaksiyonlar katalizleyebilmeleriyle oldukça ilgi çekicidirler. Özellikle bitkinin toksik savunma metabolitlerini tolere ederek, yaşadıkları çevreye adaptasyon için spesifik enzimler üretebilme yetenekleri endofitik mikroorganizmaları biyotransformasyon çalışmaları için ilgi çekici kılmaktadır. Telomerler, kromozomlarımızın sonunda bulunan ve hücre bölünmesiyle kısalan nükleotit yapılarıdır. Telomeraz ise, yaşa ve strese bağlı olarak kısalan telomer uçlarını onarmaya yardım eden rivers transkriptaz bir enzimdir. Telomeraz enzimi aktivatörleri (TA) barındırdıkları potansiyel ile sağlıklı yaşlanma için ümit verici ajanlar olarak gösterilmektedir ve ileride büyük bir pazar oluşturacağı öngörülmektedir. Bu çalışmada anti-aging etkilerinden dolayı son dönemde önemli hale gelmiş ve sadece Astragalus türlerinde bulunan sikloastragenol (CA) molekülünün türevlerinden biri olan 20(27)-oktanor sikloastragenol (SCG) üzerinde, Astragalus bitkisinden izole edilen endofitik funguslar kullanılarak biyotransformasyon calışmaları gerçekleştirilmiş ve elde edilen yeni metabolitlerin biyolojik aktiviteleri taranmıştır. Sonuç olarak, kromatografik çalışmalar ile 14 biyotransformasyon ürünü izole edilmiş ve metabolitlerin yapıları spektral yöntemler kullanılarak belirlenmiştir. Literatür araştırması sonucunda, 13 bileşiğin doğa için yeni olduğu anlaşılmıştır. Endofitlerin enzim sistemleri; oksidasyon (çoğunlukla hidroksilasyon), halka açılması (metil göçü ve halka genişlemesi veren) ve dehidrojenasyon reaksiyonlarını katalizlemiştir. Biyotransformasyon ürünlerinin 7'si telomeraz enziminin aktivasyonuna yönelik taranmıştır. Bu taramalarda DMSO ile muamele edilen kontrol hücrelerine kıyasla metabolitler 0.1-30 nM doz aralığında 5.43 ile 12.36 kat arasında değişen telomeraz aktivasyonu göstermişlerdir. Biotransformation is the chemical modifications performed by enzymes or living organisms. The difficulty and high cost of enzyme isolation and purification makes it more advantageous to use whole cell systems as biocatalysts. Microbial biocatalysts are particularly interesting in the modification of complex molecules such as steroids and triterpenoids, which can catalyze stereo- and regio- selective reactions that are difficult or impossible to perform with chemical reactions. Specially, ability of endophytic microorganisms to produce specific enzymes for adaptation to their environment by tolerating toxic defense metabolites, makes them interesting for biotransformation studies.Telomeres are nucleotide structures located at the end of chromatids shortening with each cell division. Telomerase is a reverse transcriptase enzyme, and it helps to replenish telomere ends that are truncated by aging and stress factors. The telomerase activators (TA) with their potentials are suggested encouraging agents for healthy aging, and they are projected to generate a huge market in the future. Cycloastragenol is the only natural product in the sector marketed as a potent telomerase activator.In this study, by using endophytic fungi, the biotransformation studies were performed on 20(27)-octanor cycloastragenol. As a result, 14 biotransformation products, were isolated by chromatographic studies, and the structures of the metabolites were established by spectral methods. Based on the literature survey, 13 compounds turned out to be new for nature. Seven metabolites were screened for telomerase activation. In these screenings, metabolites showed telomerase activation ranging from 5.43 to 12.36 fold at doses ranging from 0.1 to 30 nM compared to the control cells treated with DMSO. 180

Statinler yaygın olarak kullanılmakta olan anti-lipidemik ve anti-kolesterol sınıfında yer alan ilaçlardır. Bu ana fonksiyonlarının yanı sıra anti-kanser etkileri in vitro, hayvan ve klinik çalışmalarda ortaya çıkarılmıştır. Bu çalışmanın amacı yapısal olarak birbirinden farklı olan 6 adet statinin (rosuvastatin, pravastatin, simvastatin, lovastatin, fluvastatin ve atorvastatin) A549 hücreleri üzerindeki yapısal etkilerinin bir spektroskopi metoduyla incelenmesidir. Yarı maksimum inhibitör konsantrasyonunu (IC50) belirlemek amacıyla MTT canlılık testi A549 hücrelerine uygulandı. IC50 değerleri simvastatin için 50 µM, atorvastatin ve pravastatin için 150 µM, fluvastatin için 170 µM rosuvastatin ve lovastatin için 200 µM olarak bulunmuştur. Hücreler statinlerin IC5, IC10 ve IC50 değerlerine tabi tutularak tüm hücre ve lipit ekstraktları, makromoleküler fonksiyonel grup seviyesinde değişimlerinin incelenmesinde çok kullanışlı bir yöntem olan FTIR spektroskopisi ile karşılaştırılmıştır. Sonuçlar farklı statinlerin A549 hücreleri üzerinde farklı önemli etkilere sahip olduğunu göstermiştir. Tüm statinler A549 hücreleri DNA'sında ve proteomunda gözlenebilir konformasyonel değişiklikler meydana getirmiştir. Atorvastatin lipidasyona sebep olurken, lovastatin ve pravastatin önemli derecede lipit düşürücü özellik göstermiştir. Hücre lipit ekstraktı sonuçlarına göre, hidrokarbon zincir uzunluğu, doymamışlık indeksi, fosfolipit içeren lipitler ve karbonil indeksi rosuvastatin muamele edilmiş A549 hücreleri hariç artış göstermiştir. Bu çalışmada statinlerin A549 hücreleri üzerinde önemli yapısal ve kompozisyonel değişikliklere sebep olduğu bir spektroskopik değerlendirme ile tespit edilmiştir. Statins are commonly prescribed anti-lipidemic and anti-cholesterol class of drugs. In addition to their major role, they have been found to have anti-cancer effects on in vitro, animal and clinical studies. The aim of this study was to investigate the structural effects of 6 different statins (rosuvastatin, pravastatin, simvastatin, lovastatin, fluvastatin and atorvastatin) on A549 cells by a spectroscopic method. MTT viability tests were carried out to detect the half maximal inhibitory concentrations (IC50) of each statin on A549 cells. The IC50 values were 50 µM for simvastatin, 150 µM for atorvastatin and pravastatin, and 170 µM for fluvastatin, 200 µM for rosuvastatin and lovastatin on A549 cells. The cells were treated with IC5, IC10 and IC50 values of each statins concentration and their whole cell extracts and lipid extracts were compared using FTIR spectroscopy which is one of the most useful techniques to evaluate the structural changes at the macromolecular functional group level. The results indicated that different statins have different prominent effects on A549 cells. All the statins studied caused observable conformational changes on DNA and proteome of A549 cells. Whereas atorvastatin led to lipidation, lovastatin and pravastatin indicated enormous lipid-lowering properties. Based on the cell lipid extracts it was found that hydrocarbon chain length, unsaturation index, phospholipid containing lipids and carbonyl index showed increasing except for rosuvastatin-treated A549 cells. This study indicated that statins caused significant structural and compositional changes on A549 cells based on a spectroscopic evaluation. 77

Meme kanseri dünyada olduğu gibi dünyada ve Türkiye'de de kadınlarda en sık görülen ve ölüm oranı en yüksek olan kanser çeşitlerindendir. Tümör mikroçevresi, sağlıklı ve kanserli hücreleri, hücredışı matriksi, çözünür halde biyolojik faktörleri ve kimyasal etkenleri içerir. Araştırmalar hücre şekli, yapışması, hareketi, büyüme etkenlerine tepki ve ilaçlara karşı direncin, iki ve üç boyutlu düzeneklerde farklı olduğunu göstermiştir. Günümüzde kansere karşı yapılan 100 klinik deneyden sadece 8'i etkili sonuç vermektedir. 3B hücre kültürü sistemlerinin ise, tüpte, canlıda ve klinikte yapılan çalışmalar arasında çok gerekli bir adım oldukları gösterilmiştir. Bu yüzden, kanserin mikroçevresi ile olan etkileşiminin çok daha iyi anlaşılması, bunun için de yeni hücre kültürü düzenekleri gerekmektedir. Yaygın olarak kullanılan 3B düzeneklerin en belirgin eksik yönü, destek doku hücrelerinin kullanılmamasıdır. Geliştirilecek sistemler hem 3B ortam sağlamalı hem de birden fazla hücre çeşidi içermelidir. Bu çalışmada geliştirilen yonga-üstü-laboratuvar aygıtı ile 3B ve üçlü kültürde ilaç tarama yöntemi bu ihtiyaçlara cevap verebilecektir.Farklı çeşitlerde meme kanserlerini temsil eden hücre hatları tek başlarına ve destek doku hücreleri ile beraber, geliştirilen yonga-üstü-laboratuvar aygıtında 3B kültürlenerek; farklı hedefleri olan ilaçların hücrelerin canlılıklarına ve dağılımlarına etkileri belirlenerek bir ilaç tarama yöntemi geliştirilmiştir. Breast cancer is one of the cancers with the highest incidence and mortality rates in women in Turkey as well as in the world. Tumor micro environment comprises of cancer and normal cells, extracellular matrix, soluble biological and chemical factors. Research has shown that cell shape,adhesion, migration, response to growth factors and drugs are different in 2D and 3D culture. Today, only 8 out of 100 anti-cancer clinical trial gives effective results. 3D cell culture systems have shown to be a necessary step between in vitro, in vivo and clinical studies. Therefore, it is necessary to better understand the interactions of cancer cells with their micro environment, for which new cell culture setups are required. The most apparent disadvantage of widely used 3D cell culture setups is the lack of stromal cells. The systems to be developed should both provide a 3D environment and comprise multiple cell types. The drug screen in 3D tri-culture method with a lab-on-a-chip device, that will be developed in this study will be able to answer these needs. Cell lines that represent different breast cancer types alone or together with stromal cells were cultured in 3D in the to be developed lab-on-a-chip; by determining the effects of drugs with different targets on the viability and distribution of cells, a drug screening method is developed. 92

(R)- goniothalamin, isolated from the Goniothalamus plant, is a styryl lactone and has been found to have a selective antiproliferative property on cancer cells in studies conducted. The Michael acceptor feature in the structure of goniothalamin is thought to be covalently bonded to the nucleophilic side chains of the enzymes and show activity in this way. In previous studies, it has been shown that 1-naphthyl substituted 5,6-dihydro- 2H-pyran-2-one derivatives and 4'-methyl klavuzon derivatives exhibit higher cytotoxic activity on cancer cells than goniothalamin. In this study, antiproliferative properties of newly synthesized 2'-alkoxymethyl substituted klavuzon derivatives have been examined and MIA PaCa-2 pancreatic cancer cell lines and HPDEC pancreatic healthy cell lines were used. MTT cell viability tests were performed at the first step of this study. As a result of this study, it has been observed that the 2'-isobutoxymethylklavuzon derivative has selective cytotoxic activity on the MIA PaCa-2 cell line. It showed activity at lower concentrations than goniothalamin. Cytotoxic activities of the compounds are associated with the size of the R group at position 2’-. Methoxymethyl substituted the worst selective activity among these compounds whereas isobutoxy derivative the best selective one. In the second stage of the study, the inhibition on topoisomerase I enzyme was studied. The 2'-alkoxymethyl klavuzon derivatives were found to have Topo I enzyme inhibition properties depending on concentration and time manner. The study continued with choices methoxy and isobutoxy derivatives and these two compounds caused an arrest at G1 phase and DNA damage. Also, isobutoxy derivative induced apoptosis in the MIA PaCa-2 pancreatic cancer cell lines., Anti kanser ajanları üzerine yapılan çalışmaların ana amaçlarından biri ajanın, kanserli hücreler üzerinde yüksek bir sitotoksik aktivite göstermesi ve sağlıklı hücrelerde tam tersi olarak daha az bir sitotoksik etki göstermesi yada mümkünse hiç göstermemesi beklenilmektedir. Goniothalamus bitkisinden izole edilen (R)- goniothalamin, bir stiril lakton olup, yapılan çalışmalarda kanser hücreleri üzerinde seçici antiproliferatif özelliği olduğu bulunmuştur. Goniothalaminin yapısında bulunan Micheal akseptör özelliği, enzimlerin nücleofilik yapıları ile kovalent bağlar kurduğu ve bu şekilde aktivite gösterdiği düşünülmektedir. Daha once yapılan çalışmalarda 1-naftil subsitüeli 5,6-dihidro-2H-piran-2-on türevi ve 4’-metil klavuzon türevleri, kanser hücreleri üzerinde goniothalaminden daha yüksek bir sitotoksik aktivite gösterdiği görülmüştür. Bu çalışmada yeni sentezlenen 2'-alkoksimetil sübstitueli klavuzon türevlerinin antiproliferatif özelliklerine bakılmıştır ve MIA PaCa-2 pankreas kanserli hücre hattı ile HPDEC pancreas sağlıklı hücre hatları kullanılmıştır. Çalışmanın ilk basamağında MTT hücre canlılık testleri yapılmıştır. Bu çalışmanın sonucunda 2’-isobütoksimetilklavuzon türevinin MIA PaCa-2 hücre hattı üzerinde seçici bir sitotoksik aktiviteye sahip olduğu görülmüştür. Goniothalaminden daha düşük konsantrasyonlarda bu aktiviteyi gösterdiği gözlemlenmiştir. Klavuzon türevlerinin sitotoksik aktiviteleri, 2’-konumundaki R grubunun büyüklüğü ile ilişkilidir. Metoksimetil substitueli klavuzon, bileşikler içinde en kötü seçici aktiviteye sahip olan bileşik iken, isobütoksimetil substitueli klavuzon en iyi seçici aktiviteye sahip olan bileşik olduğu görüldü. Çalışmanın ikinci aşamasında topoisomeraz I enzimi üzerindeki inhibisyonuna bakıldı. Bu çalışmada kullanılan 2’-alkoksimetil klavuzon türevlerinin konsantrasyona ve zamana bağlı olarak enzimi inhibe etme özellikleri olduğu görülmüştür. Metil ve isobütoksi türevlendirilmiş klavuzon türevleri ile çalışmalara devam edildi. Ve bu iki bileşik hücrenin G1 fazını bloke etti ve DNA hasarına neden oldu. Klavuzons are 6-(naphthalen-1-yl) substituted 5,6-dihydro-2H-pyran-2-one derivatives showing promising antiproliferative activities in variety of cancer cell lines. In this work, racemic syntheses of nine novel 4′-alkyl substituted klavuzon derivatives were completed in eight steps and anticancer properties of these compounds were evaluated. It is found that size of the substituent has dramatic effect over the potency and selectivity of the cytotoxic activity in cancerous and healthy pancreatic cell lines. The size of the substituent can also effect the CRM1 inhibitory properties of klavuzon derivatives. Strong cytotoxic activity and CRM1 inhibition can be observed only when a small substituent present at 4′-position of naphthalen-1-yl group. However, these substituents makes the molecule more cytotoxic in healthy pancreatic cells rather than cancerous pancreatic cells. Among the tested compounds 1,2,3,4-tetrahydrophenanthren-9-yl substituted lactone was the most cytotoxic compound and its antiproliferative activity was also tested in 3D spheroids generated from HuH-7 cell lines., Scientific and Technological Research Council of Turkey (TUBITAK 113Z146)

Klavuzon is a naphthalen-1-yl substituted α,β-unsaturated δ-lactone derivative, and is one of the anti-proliferative members of this class of compounds. Asymmetric and racemic syntheses of novel α,β-unsaturated δ-lactone derivatives are important to investigate their potential for the treatment of cancer. In this study, asymmetric and racemic syntheses of heteroatom-substituted klavuzon derivatives are reported. The syntheses were completed by a well-known three-step procedure. α,β-Doymamış laktonlar, halkalı ester yapılarına sahip olan lactonların birüyesidir. Bunlar, yapılarındaki doymamış karbonil fonksiyonel grubunun Michaelakseptör özelliklerinden dolayı oldukça değerli bileşiklerdir. Enzimlerin nükleofilikkısımlarının laktonun β-karbonuyla reaksiyona girdiği ve bir kovalent bağ oluşturduğudüşünülmektedir. Eğer bu bağ enziminin aktif bölgesinde gerçekleşirse, aktivitesi geridönüşsüz olarak engellenir. (R)-goniothalamin bir α,β-doymamış lakton olup kanserhücreleri üzerinde seçici bir sitotoksik aktiviteye sahip olduğu gösterilmiştir. Sağlıklıhücrelerde daha az sitotoksiktir. Daha sonra Kasaplar ve iş arkadaşları,goniothalaminlerin yakın akrabaları sayılabilecek klavuzon türevlerini sentezledi. Buçalışmada basit 2'-alkoksimetil sübstitüe klavuzon türevleri sentezlenmiştir.Bu amaçla, normal şartlar altında 2-metil-1-naftoik asit iyodoetan ile reaksiyonasokarak etil-2-metil-1-naftoata dönüştürüldü. Daha sonra, oluşan ester NaBrO3 veNaHSO3 tarafından üretilen Br2 molekülleri ile reaksiyona sokuldu. Bu tepkimeden eldeedilen etil 2-(bromometil)-1-naftoat bazik koşullar altında çeşitli alkollerle reaksiyonasokularak etil 2-alkoksimetil-l-naftoat oluşturuldu. Bu esterler hedef klavuzonürünlerine LiAlH4 ile indirgeme, PCC ile oksidasyon, allilmagnezyum bromür ilavesi,akrilat ester oluşumu ve halka kapanış metatezis reaksiyonları kullanılarak beşbasamakta dönüştürülmüştür. α,β-Unsaturated δ-lactones are the members of lactones, which are cyclic esters.They are quite valuable compounds because of their Michael acceptor property atunsaturated carbonyl functional group. It is believed that soft nucleophilic parts of theenzymes react with the β-carbon of lactone and form a covalent bond. If that is occurredat the active site of an enzyme, its activity is inhibited irreversibly. (R)-goniothalamin isa α,β-unsaturated lactone and it was shown that it has a selective cytotoxic activity overcancer cells. It is less cytotoxic in healthy cells. Later, Kasaplar and coworkerssynthesized the klavuzon derivatives, which can be considered as the close relatives ofgoniothalamins. In this study novel derivatives of simple 2'-alkoxymethyl substitutedklavuzon derivatives were synthesized.For this purpose, we started with 2-methyl-1-naphtoic acid, which can betransferred to ethyl 2-methyl-1-naphtoate by reacting with iodoethane under basiccondition. Next, formed ester reacted with Br2 molecules generated in situ by NaBrO3and NaHSO3. Then formed ethyl 2-(bromomethyl)-1-naphtoate reacted with variousalcohols under basic conditions to form ethyl-2-alkoxymethyl-1-naphtoate. These estersare coverted to the target klavuzon products in five steps that are reduction with LiAlH4,oxidation with PCC, addition of allylmagnesium bromide, acrylate ester formation andring closing metathesis reactions. 136

Hepatoselüler karsinoma (HSK) beşinci en yayın kanser türü olup kanser nedeniyle ölümlerin üçüncü en yüksek nedenidir. HSK ölümcül bir hastalık olup HSK hastalarının 5 yıllık yaşam süreleri %14'tür. Yeni terapötik ajanların keşfi HSK tedavisi için gerekmektedir. Kanser tedavisinde en önemli hedeflerden birisi histonların asetilasyonudur. Histon deasetilazlar (HDAS) ve sirtuinler asetil gruplarını histon proteinlerinden uzaklaştırarak kromatin sıkılaşmasına ve transkripsiyonel represyona sebep olurlar. Yeni HDAS/sirtuin inhibitörleri ile kromatinin tekrar asetillenerek tümör baskılayıcı genlerin tekrar eksprese edilmesi terapötik hedeflerdendir.Bu çalışmada, (R)-4'-metilklavuzon'un HuH-7 parental hücreleri için 1.25 µM, EpCAM+/CD133+ HuH-7 hücreleri için 2,5 µM; EpCAM-/CD133- HuH-7 hücreleri için 1.25 µM olan IC50 değerleri ile sitotoksik olduğu bulunmuştur. (R)-4'-metilklavuzon'un üç hücre populasyonunda 1,00 µM konsantrasyonda G1 fazında hücre döngüsünde birikmeye sebep olduğu ve 24 saatlik inkübasyonda 10 µM konsantrasyonda apoptozu indüklediği gözlemlenmiştir. (R)-4'-metilklavuzon'un in vitro koşullarda sınıf I/II histon deasetilaz enzimlerini inhibe etmediği ancak MACS ve FACS ile ayrımlanmış hücrelerdeki endojen sınıf I/II histon deasetilazları ve/veya sirtuinleri 0,10 µM konsantrasyonda hücre içinde inhibe ettiği bulunmuştur. (R)-4'-metilklavuzon, HuH-7 hücrelerinde p21 ekspresyonunu 3,3 kata kadar arttırabilmiştir. p53/MDM2 kompleks oluşumunu %45 oranında azalttığı görülmüştür. In vitro koşullarda SIRT1 enziminin 100 μM ilaç adayı ile %46 oranında inhibe olduğu bulunmuştur. HuH-7 hücrelerindeki SIRT1 protein seviyelerinin 3,00 µM ilaç adayı ile 24 saatlik inkübasyonunda %240 oranında artmıştır. (R)-4'-metilklavuzon'un CRM1 proteinini inhibe ederek tümör baskılayıcı proteinleri nukleus içinde tutabileceği anlaşılmıştır. 0,10 μM ve 1,00 μM ilaç adayı ile 6 ve 24 saat muamele edilen hücrelerdeki p53 protein seviyelerinin HepG2 hücrelerinde yüksek miktarda artış gösterdiği ancak HuH-7 hücrelerinde çok az bir artış gösterdiği bulunmuştur. Hepatocellular carcinoma (HCC) is the fifth most seen cancer type and the third leading cause of death from cancers. HCC is a fatal disease and HCC patients have a 5-year survival rate of 14%. Discovery and identification of mechanisms of action for new therapeutic agents are required for a better treatment of HCC. One of the most important target in cancer treatment is the epigenetic acetylation of histones. Histone deacetylases (HDAC) and sirtuins provide chromatin compaction and transcriptional repression by removing acetyl groups from histone proteins and non-histone proteins. Re-acetylation of chromatin and re-expression of tumor suppressor genes with the discovery of novel HDAC and/or sirtuin inhibitors are therapeutic targets in cancer research. In this study, (R)-4'-methylklavuzon was found to be cytotoxic in HuH-7 cells with IC50 values of 1.25 µM for HuH-7 parental cells, 2.5 µM for EpCAM+/CD133+ HuH-7 cells and 1.25 µM for EpCAM-/CD133- HuH-7 cells. It was also observed that (R)-4'-methylklavuzon causes cell cycle arrest at G1 phase at 1.00 µM concentration in three cell populations, it induces apoptosis at 10 µM concentration at the end of 24 hours incubation. Besides (R)-4'-methylklavuzon does not inhibit Class I/II histone deacetylase enzymes in vitro whereas it causes inhibition of endogenous histone deacetylase enzymes and/or sirtuins inside the cells sorted by MACS and FACS even at 0.10 µM concentration. (R)-4'-methylklavuzon upregulates p21 expression significantly in HuH-7 cell populations to cause G1 arrest. It also causes 45% inhibition in p53/MDM2 complex formation when examined with pure p53 and pure MDM2 proteins. Further assays indicated that drug candidate causes 46% SIRT1 inhibition at 100 μM concentration in vitro whereas there was no inhibition of HDAC1 enzyme at the same concentration. SIRT1 protein levels in HuH-7 parental cells were upregulated to 240% at the end of 24 hours incubation with 3.00 μM of drug candidate. It was found that (R)-4'-methylklavuzon can also inhibit CRM1 protein providing increased retention of tumor suppressor proteins in the nucleus. p53 was significantly overexpressed at 0.10 and 1.00 μM concentrations within 6 and 24 hours in HepG2 cells but slightly overexpressed in HuH-7 parental cells. 138

An organometallic dye, europium tetrakis dibenzoylmethide triethylammonium (EuD4TEA) and gold nanoparticles (Au NPs) impregnated paper based sensor platform have been utilized for development of fluorescence turn-on cyanide assay in aqueous media. The ordinary filter paper with 6 μ m pore size were employed as solid support that facilitates impregnation of EuD4TEA and gold nanoparticles and provides durability. Detection mechanism relying on two processes (i) dissolution of gold nanoparticles causing fluorescence recovery and (ii) ligand exchange of triethyl amine with CN group stimulating cyanide specific fluorescence enhancement. The paper platform exhibit naked eye distinguishable color transition upon CN− addition from 10−2 to 10−12 M. To standardize the methodology a homemade image processing algorithm has been developed that enabling calibration of color change and quantify CN− concentration. The described algorithm is applicable to Android smart phones and facilitate transforming these devices into a quantitative cyanide detector. The overall methodology provides instrument free cyanide detection and therefore rapid control of water quality and safety at off-field conditions.

Kansere karşı geliştirilen ilaçların etkilerinin incelenebilmesi için hücresel düzeyde yapılan çalışmalar çoklu kuyucuklar kullanılarak yapılmaktadır. Ancak bu klasik sistemler gerçekçi denemeler için gerekli in-vivo benzeri mikroçevre oluşturmakta başarısız olmaktadırlar. Ayrıca klasik hücre kültürü sistemleri artan maliyetlerle yüksek miktarda malzeme kullanmaktadır. Diğer yandan çip-üzeri-laboratuvar sistemleri minimum reaktif hacmi kullanmakta ve daha önemlisi in-vivo mikroçevreyi boyutsal ve zamansal taklit edebilmektedir. Dahası hücrelerin ilaca karşı karşılıklarının 2B ve 3B hücre kültürü denemelerinde değiştiği bilinmektedir. Doksorubisin yaygın olarak kullanılan bir antikanser ilacıdır. Bu çalışmada, yüksek oranda metastatik insan meme kanseri hücre hattı MDA-MB-231 ve normal meme epitel hücre hattı MCF10A tarafından ilaç alımı 2B ve 3B, klasik ve çip-üzeri-hücre kültüründe araştırılmıştır. Farklı ilaç dozlarının 24, 48 ve 72. saatlerde alımı hücrelerin floresan mikroskobundaki resimlerinden sinyal yoğunluğu ölçülerek hesaplanmıştır.Hücre canlılığı analizi için hücreler dapi ile boyanmış ve iki hücre hattı bu sistemler içerisinde karşılaştırılmıştır. Sonuçlara göre, çip içerisindeki 3B hücre kültürü ortamın daha az reaktif tüketimiyle birlikte daha çok in-vivo benzeri ortamın sağladığı ve hücre canlılığının sadece ilaç alımıyla orantılı olmadığı gözlenmiştir. The studies that aim to assess the effects of drugs developed against cancer at the cellular level use multiwell plates. However, these classical systems fail to reproduce the in-vivo like microenvironment necessary for realistic assessment. In addition, classical cell culture systems use high amount of materials increasing cost. On the other hand, lab-on-a-chip systems use minimal volumes of reagents and more importantly can mimic the in-vivo microenvironment via spatial and temporal control. Furthermore, it is known that cell response to drugs can be very different in 2D and 3D cell culture setups. Doxorubicin is a widely used anticancer drug. Here, doxorubicin uptake by highly metastatic human breast cancer cell line MDA-MB-231 and normal mammary epithelial cell line MCF10A were investigated using 2D and 3D, classical and cell-on-a-chip cultures. Drug uptake at 24, 48 and 72 hours various concentrations of the drug determined by measuring signal intensities from fluorescence microscopy images of cells. For cell viability assay, cells were stained with dapi and two cell lines were compared in systems. According to results, it was observed that 3D cell culture environment in chip provides more in-vivo like environment with less reagent consumption and cell viability is not correlated only with drug uptake. 87

Antikanser ajan geliştirme çalışmalarda en önemli konulardan birisi spesifik olarak kanserli hücreleri hedef alıp sağlıklı hücrelere zarar vermeyen bir ajanın keşfidir. Goniothalamin, Goniothalamus bitki türlerinden izole edilmiş bir stiril lakton olup, kanserli hücrelerin çoğalmalarına karşı seçici aktivitesi bulunan bir anti-kanser ajandır. Klavuzon ve türevleri bir tür goniothalamin türevi olarak düşünülebilirler ve goniothalamine oranla kanser hücrelerine karşı daha sitotoksiklerdir. Daha önceki yapı-aktivite ilişkisi çalışmalarında α,β-doymamış δ-lakton yapısının biyolojik aktivite için gerekli olduğu bilinmektedir. Bu grup Michael akseptörü olarak görev aldığından bu tez kapsamında iki farklı hücre içi mekanizmanın geridönüşümsüz inhibisyonu araştırılmıştır.Çalışmanın ilk bölümünde, olası bir timidilat sentaz inhibisyonuna bağlı olarak gerçekleşebilecek timidinsiz ölümün varlığı araştırılmıştır. Farklı konsantrasyonlarda timidin varlığında veya yokluğunda klavuzonla muamele edilmiş hücrelerin canlılıklarında herhangi bir değişiklik olmamıştır, bu nedenle timinsiz ölüm olayının klavuzon türevleri için önemli bir mekanizma olmadığı sonucuna varılmıştır. İkinci bölümde ise, 4'-metilklavuzon ve türevlerinin nükleositoplazmik transport üzerindeki etkilerini araştırmak için HeLa hücreleri üzerinde test edilmiştir. CRM1 proteini tarafından taşınan Riok2 proteininin nükleositoplazmik yerini göstermek için immünositokimya kullanılmıştır. In anti-cancer agent development studies one of the most significant issue is to get an agent that specifically targets cancer cells without any effects on healthy cells. Goniothalamin, that is a styryl lactone isolated from Goniothalamus plant species, is an anti-cancer agent that has selective anti-proliferative activity on cancer cell lines. Klavuzon and derivatives, which can be thought as analogs of goniothalamin, are more cytotoxic in cancer cells compared to goniothalamin. Previous structure activity relationship studies implies that α,β-unsaturated δ-lactone moiety is the source of the biological activity. Since it behaves as Michael acceptor, in this thesis possible irreversible inhibitions of two separate intracellular targets are investigated. In the first part, thymineless death caused by possible thymidylate synthase inhibition has been studied. Anti-proliferative effect of 4'-methylklavuzon in HuH-7 cancer cell line was tested by using MTT. Viability of klavuzon treated cells did not changed significantly in the absence and presence of varying concentration of additional thymidine supplement, and it is concluded that thymineless death is not a crucial mechanism for klavuzon derivatives. In the second part, 4'-methylklavuzon and its derivatives were tested on HeLa cell line to investigate inhibitory effect on the nucleocytoplasmic transport. Immunocytochemistry was used to demonstrate nucleocytoplasmic localization of Riok2 protein which is transferred from nuclei to cytoplasm by CRM1 nuclear export protein. Successfully, all tested klavuzon derivatives inhibit CRM1 nuclear export protein. Potency of the inhibition depends on the size of the alkyl substituent at 4'- position of klavuzon. 56

(R)-goniothalamin, which is a styryl lactone isolated from plants, is an anti-cancer agent that is cytotoxic to cancer cell lines with no or minimal effect on normal cells. Also klavuzon molecule, the 1-naphthyl substituted 5,6-dihydro-2H-pyran-2-one derivative, was tested on cancer cell lines and showed higher cytotoxicity. In the first part of this study anti-proliferative effect of novel 4’-alkyl substituted klavuzon derivatives were tested on MIA PaCa-2 cancer cell line and normal Human Pancreatic Duct Epithelial Cell (HPDEC) line by using MTT, and it is found that cytotoxic activity of the compounds depends on the size of the substituent at position 4 in 1-naphthyl part. While two of them, methyl and ethyl substituted, showed high cytotoxicity to MIA PaCa-2 that IC50 values in nano-molar levels. Next studies were continued with 4’-methylklavuzon derivative. In the second part, the apoptotic effect and cell cycle analysis of 4’-methylklavuzon was studied. Especially at the 10 μM concentration there was increment in both early and late apoptosis. Cell cycle analysis showed that increasing the concentration of molecule caused cell cycle arrest in S and G2 phases. The next part was testing the inhibitory effect of 4’-methylklavuzon on human topoisomerase I enzyme. This enzyme was chosen as one potential target, because 4’-methylklavuzon caused S and G2 phase arrest. Topo I is an actively working enzyme during S phase and inhibition of its activity causes DNA fragmentation and apoptosis. The results showed that there was time dependent inhibition of Topo I enzyme in vitro. The last part of the study was to show DNA fragmentation at cellular level by COMET assay, also known as Single Cell Gel Electrophoresis. After Topo I assay has showed the Topo I inhibition activity of 4’-methylklavuzon, COMET assay was performed. Daha sonraki çalışmalara 4’-metilklavuzon ile devam etmiştir İkinci kısımda ise 4’-metilklavuzonun apoptotik ve hücre döngüsü üzerindeki etkileri çalışılmış ve 10 μM dozdaki uygulamada erken ve geç faz apoptotik hücre sayılarında artma görülmüştür. Hücre döngüsü analizlerinde ise 4’-metilklavuzonun S ve G2 fazlarında döngüyü durdurduğu görülmüştür. Sonraki çalışmada ise 4’-metilklavuzonun insan topoizomeraz I enzimi üzerindeki inhibisyon etkisi incelendi. Yapılan çalışma 4’-metilklavuzonun ön-inkübasyon süresine paralel olarak topoizomeraz I enzimi üzerinde inhibisyon etkisi olduğunu göstermiştir. Son kısımda ise tek hücre düzeyinde COMET testi ile 4’-metilklavuzonun DNA üzerindeki etkilerine bakılmıştır. This study aimed to identify and determine the carotenoids from green microalga, Scenedesmus protuberans using analytical techniques. Identification of carotenoids was realized by comparing their absorption and mass spectral data with those of reference standards available and reported values. Chromatographic data were then combined with the spectroscopic information. The separation of carotenoids was achieved by C30 column and high-performance liquid chromatography-diode array detection was used for their determination. In the present work, the carotenoid content of S. protuberans was found to be 1.45 mg/g of violaxanthin, 2.47 mg/g of all-trans-lutein, 0.15 mg/g of all-trans-α-carotene, 0.55 mg/g of all-trans-β-carotene, and 0.20 mg/g of 9 or 9′-cis-β-carotene. Due to lack of their standards, the amount of all-trans-loroxanthin and cis-isomers of other carotenoids could not be quantified. In order to validate the method, Certified Reference Material (BCR 485-Mixed vegetables) was used. A solid phase extraction sorbent, aminopropyl triethoxysilane-functionalized silica was prepared and used for vanadium speciation prior to inductively coupled plasma mass spectrometric determination. The necessary sorption parameters such as sorption pH, reaction temperature, sorbent amount and sample volume were optimized. The sorption for V(V) and V(IV) under the optimized conditions was 98% (±1) and was lower than 10%, respectively. The sorption process was investigated both from a kinetic perspective and also in terms of Freundlich, Dubinin-Radushkevich and Langmuir isotherm models to characterize the uptake of vanadate ion by the functionalized silica. Desorption from the sorbent was realized with 0.5 mol L−1 thiourea prepared in 0.2 mol L−1 HCl. Among the possible interfering species tested, Te(IV), Se(IV), Se(VI) and Fe(III) showed interference effects with V(V) signal. The validity of the method was first checked via spike recovery experiments with four different types of water; namely ultra pure, bottled drinking, tap, and sea water for different concentrations of V(V). The percent recovery values changed between 87 and 108 for all water types. The accuracy of the proposed methodology was also checked by analyzing a standard reference material (NIST, SRM 1643e) and a good agreement was found between the determined and the certified value. The proposed methodology has also been shown to be applicable for the separate sorption of V(V) in the absence/presence of V(IV) and for the sorption of both V(IV) and V(V) in waters. The amount of V(IV) can then be determined from the difference.

The mechanism of action for α,β-unsaturated lactones can be explained by their Michael acceptor properties. They have the potential of being covalently binding inhibitors by accepting nucleophiles from target proteins. In this work, Michael addition reactions of ethanethiol with 6-bicycloaryl substituted 5,6-dihydro-2H-pyran-2-ones were studied to explore the existence of such interactions. Three of the Michael addition products were isolated and tested over PC3 (human prostate cancer) and MCF-7 (human breast adenocarcinoma) cancer cell lines and no cytotoxicity was observed. It was revealed that biological activity depends on the existence of a Michael acceptor, but potency probably depends upon the 3D structure of the substituent on lactone ring. We described a one-pot vinylogous aldol addition of methyl 3-butenoate to aromatic aldehydes in the presence of silver salts and the TMSCl-TBAF mixture. Bu yapıya sahip moleküller üzerindeki araştırmalar bunların önemli miktarda biyolojik aktivite göstermelerinden dolayı son on yılda artmıştır.Bu çalışma 4'-alkil sübstitüentli klavuzon ve naftalin-1-il türevlendirilmiş 3,6-dihidro-2H-piran-2-on türevlerinin sentezlerini amaçlamaktadır. Sentezler etil 4-brommetil-1-naftoatın hazırlanmasıyla başlamıştır. Etil 4-bromometil-1-naftoat ve Grignard reaktiflerinin bakır katalizörlüğünde reaksiyonları sonucu alkil sübstitüentli esterler elde edilmiştir. Ardından alkillenmiş esterler LiAlH4 ile indirgenip, PCC ile yükseltgenerek alkillenmis naftaldehit türevleri elde edilmiştir. Alkillenmiş naftaldehitler allilmagnezyum bromür ile reaksiyona sokularak homoallilik alkoller elde edilmiştir. Son olarak homoallilik alkoller akriloyil klorür ile baz varlıgında reaksiyona sokulup akrilat esterler elde edilmiş ve 2. nesil Grubbs' katalizörü varlığında halka kapanması reaksiyonu ile klavuzon türevleri elde edilmiştir.Ek olarak, yeni beş β,γ-doymamış lakton türevleri de sentezlenmiştir. Aldehitlerin vinilmagnezyum bromür ile reaksiyonları sonucu allilik alkoller elde edilmiştir. Alkoller vinil asetik asit ve DCC/DMAP katalizör varlığında estere dönüştürüldükten sonra 2. nesil Grubbs' katalizörü ile halka kapanma reaksiyonları çalışılmıştır. The styryl-lactones are a group of secondary metabolites which have α,β-unsaturated δ-lactone moiety in their structures. One of the most important member of this group of compound is goniothalamin. Goniothalamin and its analogs having a common α,β-unsaturated δ-lactone moiety. Investigations on these species have been increased in the last decade because of their interesting biological activities.Present study aims the synthesis of novel 4'-alkyl substituted klavuzon and naphthalen-1-yl substituted 3,6-dihydro-2H-pyran-2-one derivatives. Syntheses were started with the preparation of ethyl 4-(bromomethyl)-1-naphthoate. Copper-catalyzed cross coupling reactions between ethyl 4-bromoethyl-1-naphthoate and Grignard reagents gave alkyl substituted esters. Next, alkylated ester derivatives were converted to alklylated-naphthaldehyde derivatives via reduction with LiAlH4 and then oxidation with PCC. Allylation reaction of corresponding aldehydes with allylmagnesium bromide gave homoallylic alcohols. Finally, homoallylic alcohols were reacted with acryloyl chloride in the presence of a base to produce acrylate esters, then ring closing metathesis by second generation Grubbs' catalyst yielded the final klavuzon derivatives.Additionally, synthesis of new five β,γ-unsaturated δ-lactone derivatives was also completed. Vinylation reaction of corresponding aldehydes with vinylmagnesium bromide gave the allylic alcohols. Coupling reactions of these alcohols with 3-butenoic acid in the presence of DCC/DMAP yielded the ester. Ring closing metathesis of the related ester was studied by using 2nd generation Grubbs' catalyst. 116

Sitiril laktonlar doğal kaynaklardan izole edilen ve çeşitli canserli hücre hatlarına karşı sahip oldukları sitotoksik etkisi ile bilinen yapılardır. Goniothalamin en bilinen sitiril laktonlardan biri olup yapsında α,β-doymamış-δ-lakton bulundurması sebebiyle değişik kanser hücrelerine karşı yüksek sitotoksiteye sahip olduğu gözlemlenmiştir. Bunun yanında goniothalamin sağlıklı hücreler üzerinde de test edilmiş ve minimal etki gözlemlenmiştir. Son zamanlarda yapılan çalışmalarda 5-aril sübstütientli-α,β-doymamış δ-laktonların (R)-goniothalamin yapısından daha sitotoksik olduğu gözlemlenmiştir. Bunun sonucunda naftalen-1-il sübstitüentli α,β-doymamış-δ-laktonlar Çağır araştırma grubu tarafından klavuzon olarak adlandırılmıştır.Bu çalışmada 4'-alkil substitüeli klavuzon türevlerinin sentezi çalışılmıştır. Sentezler Cu katalizörlüğünde etil 4-(bromometil)-1-naftoata Grignard reaktifi ile benzilik alkilasyon gerçekleştirilerek başlanmıştır. Lakton sentezinde literatürde iyi bilinen ve aldehit ile başlayan üç basamaklı sentez yöntemi kullanılmıştır. Bu sebeple daha önce alkil eklenmiş olan 1-naftoat esterleri LiAlH4 kullanılarak önce alkole indirgenmiş ardından PCC kullanılarak aldehite yükseltgenmiştir. Bu aldehitlere allilmagnezyum bromür ile muamele edilmiş ve elde edilen alkoller akriloil klorür ile akrilat estere dönüştürülmüştür. Son olarak 2. Nasil Grubbs katalizörü yardımıyla sentezlenmesi planlanan α,β-doymamış-δ-laktonlar elde edilmiştir. Styryl lactones, which are compounds isolated from natural sources, are known for their cytotoxic property against variety cancer cell lines. Goniothalamin is a well known member of stryl lactones, include α,β-unsaturated δ-lactone in its structure. Being a α,β-unsaturated δ-lactone subunit is an important structural feature. Since, Goniothalamin has a selective cytotoxic property for cancer cell lines with no significant cytotoxic activity toward non-malignant cells. Recent studies show that naphthalen-1-yl substituted α,β-unsaturated δ-lactone derivative has slightly better cytotoxic activity than (R)-goniothalamin. As a result of that naphthalen-1-yl substituted α,β-unsaturated δ-lactones derivative was named as klavuzon by Çağır Research group.In this study it is aimed to synthesize 4'-alkyl substituted klavuzon derivatives. Syntheses were started with the Cu catalayzed addition of Grignard reagents to ethyl 4-(bromomethyl)-1-naphthoate to achieve alkyl substitution. For lactone synthesis a well-known three steps synthesis was used starting from aldehyde. Thus 1-naphthoate esters first reduced to alcohol then oxidized to aldehyde by using LiAlH4 and PCC. These aldehydes were reacted with allymagnesium bromide then formed alcohols were acrylated with acryloyl chloride to yield the corresponding esters. Finally, target molecules were synthesised by ring closing metathesis by using 2nd generation Grubbs' catalyst. 82

Flavonoid içeren çok sayıda bitki gıda, ilaç ve kozmetik endüstrisinde yaygın olarak kullanılmaktadır. Literatür çalışmaları ilk ilaçların bitkisel kaynaklı olduğunu göstermektedir. Flavonoidlerin farklı alanlardaki uygulamaları yeni ekstraksiyon ve tayin yöntemlerinin geliştirilmesini gerektirmektedir.Bu çalışmada, üç farklı katı faz mikroekstraksiyon kaplamaları üretilmiştir. İlk olarak, sol-gel yöntemi ile sentezlenen baicalin- ve imidazole- grupları ile işlevselleştirilmiş kaplamalar yer almaktadır. Bu fiberler hem manuel kaplama ile hem de sol-jel kaplama cihazı (dip coater) kullanarak üretilmiştir. Fonksiyonelleştirilmiş silika fiberler taramalı elektron mikroskobu ile karakterize edilmiştir ve elde edilen sonuçlara göre sol-jel kaplama cihazı ile yapılan kaplamalarda homojen kaplamalar elde edilmiştir. İkinci kaplamalarda ise polistiren kalıp yöntemi uygulanarak sentezlenen monolitik silikayı içermektedir. Sentezlenen monolitik silika daha sonra baicalin- ve imidazole- grupları ile işlevselleştirilmiştir. Son kaplama yönteminde moleküler baskılama yöntemi ile silika sentezlenmiştir. Bu yöntemde, sol-jel ile sentezlenen moleküler baskılı silikada TEOS çapraz bağlayıcı, APTES fonksiyonel monomer, quersetin kalıp molekül ve AlCl3 ise lewis asit olarak kullanılmıştır. Baikalin ile işlevselleştrilmiş monolitik silika ve moleküler baskılı silikanın karakterizasyonu SEM, FTIR, TGA ve elemental analiz yöntemleri ile yapılmıştır. Flavonoidlerin katı faz mikroekstraksiyonundan sonra, bu bileşikler Zorbax Eclipse XDB-C18 (5µm, 4.6x150mm) kolonunda ile 25ºC'de gradient elüsyon uygulayarak ayrılmasından sonra HPLC-DAD yöntemi ile 335 nm tayin edilmiştir. Hazırlanan kaplamalar apigenin, apigetrin, eriodictyol, isoquercitrin, luteolin, and quersetinin katı faz mikroekstraksiyonunda kullanılmıştır. Baikalin ile işlevselleştirilmiş monolitik silika ve moleküler baskılı silika ile kaplanan fiberler ile yüksek ekstraksiyon performansı elde edilmiştir. Many plant species contain flavonoids which are widely utilized in food, pharmaceutical and cosmetics industries as additives. Literature studies show that most of the earliest drugs were of plant origin. Application of flavonoids in many fields necessitates the development of new extraction and determination methods in various samples.In this study, three different types of solid phase microextrcation coatings were produced. The first one is baicalin- and imidazole-functionalized silica fibers via sol-gel method. Both manual and dip-coating were employed. The fibers were examined by SEM and the results show that dip-coating produced more homogeneous coatings. The second one is the polystyrene templated preparation of monolithic silica. Then, it was functionalized with baicalin- and imidazole-groups. The last solid phase microextraction coating is the molecularly imprinted silica. In this case, a sol-gel process was used to synthesize molecularly imprinted silica with TEOS as the crosslinker reagent, APTES as a functional monomer, quercetin as the template molecule, and AlCl3 as a lewis acid. Characterization of baicalin-functionalized monolithic silica and molecularly imprinted silica was achieved by SEM, FTIR, TGA and elemental analysis. SPME extractions were followed by HPLC-DAD at 335 nm after separation of flavonoids on a Zorbax Eclipse XDB-C18 (5µm, 4.6x150mm) column at 25.0°C with gradient elution using methanol and water (containing 0.10% acetic acid). Functionalized silica fibers were utilized in solid phase microextraction of apigenin, apigetrin, eriodictyol, isoquercitrin, luteolin, and quercetin. Their results demonstrated that higher extraction efficiency can be obtained with baicalin-functionalized monolithic silica and molecularly imprinted silica coated fibers. 134

α,β-doymamış lakton türevleri doğadan izole edilen ve çeşitli biyolojik aktivitelerinin kaynağı olarak gösterilen önemli yapı taşlarıdır. Biyolojikçe aktif stiril δ-laktonlar α,β-doymamış δ-laktonlar içinde en iyi bilinenleridir. Birçok çeşitli doğal ürün yapısında α,β-doymamış δ-lakton yapısını içerir. Günümüze kadar kanser hücreleri üzerinde anti-proliferatif özelliğe sahip ve gelecek vaat eden birçok α,β-doymamış-δ-laktonlar ile analogları doğadan izole edilmiş ve sentezlenmiştir.Bu tez çalışmasında, (R)-4'-metilklavuzon ve (R)-2'-metilklavuzon'un büyük ölçekli asimetrik sentezleri başarıyla tamamlanmıştır. Ayrıca heteroatom fonksiyonuna sahip naftalenil sübsititüyeli yeni α,β-doymamış-δ-lakton (5,6-dihidro-2H-piran-2-on) türevlerinin, α,β-doymamış-γ-lakton (furan-2(5H)-on) türevlerinin ve 7 üyeli α,β-doymamış lakton (6,7-dihidro--5H-oksepin-2-on) türevlerinin sentezleri başarılmıştır.Altı üyeli α,β-doymamış-δ-laktonların tek kap yöntemiyle sentezine yönelik yeni bir metot geliştirme çalışmaları yapılmış ve yeni bir vinilogous eklenme yöntemi geliştirilmiştir. Bu metot ile metil büten-3-oat ile başlanarak yeni α,β-doymamış metil esterleri dienolat oluşumu aracılığıyla hazırlanmıştır. α,β-unsaturated lactone derivatives are very important structural core which are isolated from nature and shown as source for several biological activities. Biologically active styryl δ-lactones are well known α,β-unsaturated-δ-lactones. Up to date, many α,β-unsaturated-δ-lactones and their analogues have been isolated and synthesized, exhibiting promising anti-proliferative properties against different cancer cell lines.In this thesis, large scale asymmetric syntheses of (R)-4'-methylklavuzon and (R)-2'-methylklavuzon were completed successfully. Also, syntheses of heteroatom functionalized naphthyl substituted novel α,β-unsaturated-δ-lactone (5,6-dihydro-2H-pyran-2-one) derivatives, α,β-unsaturated-γ-lactone (furan-2(5H)-one) derivatives and 7-membered α,β-unsaturated lactone (6,7-dihydro-5H-oxepin-2-one) derivatives were accomplished.Development of a method for one-pot synthesis of 6-membered α,β-unsaturated-δ-lactone was also studied, and a new vinylogous aldol addition method was developed. By this method, new α,β-unsaturated methyl esters were prepared via dienolate formation starting with methyl buten-3-oate. 140

Sol-gel based amine-functionalized SPME fibers (PDMS-weak anion exchanger) were prepared and used for direct mode extraction of dimethylarsinic acid (DMA), monomethylarsonic acid (MMA), and arsenate (As(V)) from aqueous solutions followed by HPLC-ICPMS determination. Two different methods of coating were employed: (i) electrospinning and (ii) dip coating. Electrospinning was used for the first time for preparation of sol-gel based SPME fibers and was found to be superior in terms of extracted amount of arsenicals, coating homogeneity, accessibility of amine groups on the surface, and preparation time for a single fiber. Various parameters such as solution pH, extraction time, agitation speed, extraction temperature, and ionic strength were studied. Optimum extraction conditions were determined as pH of 5.0, extraction time of 30 min, agitation speed of 700 rpm, and extraction temperature of 20 degrees C. Extraction ability of the novel coating decreased by the addition of NaCl as a consequence of the competition between anionic arsenic species and chloride ions for active sites of the weak anion exchanger. This novel sol-gel coating prepared by electrospinning was found to be promising for SPME applications. Vibrational spectroscopy revealed the alignment of PDMS chains by elongational force under electrospinning process. The chain alignment accordingly orients the pendant amino functional groups perpendicular to the fiber surface, which may develop the free active functional groups available to the medium and lead to the enhancement of the extraction performance. Moreover, the proposed coating strategy through electrospinning might be able to break new ground for various applications in analytical chemistry as well as other disciplines.

Tek kristal seryum oksit nanoparçacıkları seryum nitratın [Ce(NO3)3.6H2O] NaOH ve üre ile sulu çözeltilerinin hazırlanmasının ardından hidrotermal metot ile sentezlenmiştir. Taramalı ve geçirimli elektron mikroskobu ve X-ray kırınımı yöntemleri kullanılarak parçacıkların morfoloji ve kristal özellikleri saptanmıştır.Çalışmanın ilk kısmı reaksiyon zamanı, reaksiyon sıcaklığı, polimer, baz çeşidi ve konsantrasyonu gibi parametrelerin değişiminin parçacık boyutuna ve morfolojisine olan etkilerini içermektedir. Reaksiyon zamanı ve sıcaklığı ve baz konsantrasyonu denemelerinde sadece üre kullanılmıştır. Baz tipleri karşılaştırıldığında, NaOH ile daha küçük parçacık boyutuna ulaşılırken, üre ile daha küçük kristal boyutuna ulaşılmıştır. Polimer etkisi de incelenmiş olup, parçacığın boyut ve morfolojisini değiştiren net bir etkiye rastlanmamıştır. Baz konsantrasyonundaki değişimde ise 1M?dan küçük konsantrasyonlarda kübik ve üçgen prizma şeklinde, 1M?dan büyük konsantrasyonlarda ise topaklanmış küre biçimdeki parçacıklar elde edilmiştir. Reaksiyon zamanı ve sıcaklığının etkileri de araştırılmış, bütün denemelerde topaklanmış düzgün olmayan küre biçiminde parçacıklar gözlemlenmiştir. Bu örnekler için parçacık boyutu ve kristal boyutunda dikkate değer bir fark görülmemiştir. Üre ile sentezlenen parçacıkların mikron altı boyutlarda olmasından dolayı organik reaksiyonlarda NaOH ile sentezlenen CeO2 nanoparçacıkları katalizör olarak kullanılmıştır.İkinci kısımda ise CeO2 nanoparçacıklarının katalitik özellikleri araştırılmıştır. CeO2 nanoparçacıkları 2?-hidroksiçalkondan flavon dönüşümünde katalizör olarak kullanılmıştır. Reaksiyonların sonucunda, toplam 42.5 % oranında dönüşüm saptanmıştır. Ayrıca değişik reaksiyon parametrelerinin flavon dönüşümü üzere olan etkileri de araştırılmıştır. Sonuçlar TLC, GC-MS ve NMR ile takip edilmiştir. Single crystalline cerium oxide nanoparticles were synthesized with hydrothermal method by mixing cerium nitrate [Ce(NO3)3.6H2O] aqueous solution with NaOH and urea. SEM, TEM and XRD characterization methods were used in order to identify morphology.First part of the study includes the work on effect of hydrothermal parameters, such as base type and concentration, surfactant, reaction temperature, and time on particle size and morphology. The reaction time, temperature and concentration trials were carried out for urea only. When base types were compared, smaller particles were synthesized in the presence of NaOH yet smaller crystalline sized particles were obtained in the presence of urea. Effect of surfactant was investigated however there was no significant effect on size or morphology. Effect of concentration was clear, cubic and triangular prism shaped particles obtained for concentrations lower than 1M, and for concentration higher than 1M, agglomerated spherical particles were obtained. Reaction time and temperature were also investigated and all the reactions resulted in agglomerated non-uniform spherical particles. There was no considerable difference in particle and crystalline size for these samples. Due to the sub-micron size of CeO2 synthesized using urea, CeO2 nanoparticles synthesized using NaOH for catalysis.Second part includes the investigation of catalytic property of CeO2 nanoparticles. CeO2 nanoparticles were used as catalyst in the synthesis of flavone from 2?-hydroxychalcone. Total flavone yield was approximately 42.5 %. Several reaction parameters were studied in order to achieve the flavone synthesis. TLC, GC-MS and NMR were used in order to monitor the results of the reactions. 81

Kanser araştırmalarında doğal yollardan izole edilen 5-sübstitüeli,α,β-doymamış laktonlar sitotoksik ve antitümör etkileri sebebiyle bilim adamlarının ilgisini çekmiştir. Bu grup bileşiklerin en önemli üyelerinden biri de goniothalamindir. Goniothalaminin değişik kanser hücrelerine karşı yüksek sitotoksik aktiviteye sahip olması ve sağlıklı hücreler üzerinde minimal etki yaratması, kanser hücrelerine karşı seçici sitotoksik aktivite gösterdiği düşünülmektedir.Bu çalışmanın ilk kısmında sentezlenen 5,6-dihidro piran türevlerinin MCF7, PC3 HCT116 p53+/+ ve p53-/- hücre hatlarındaki antikanser aktiviteleri MTT testi ile belirlenmiş ve FITC işaretlenmiş Annexin V-PI boyaları yardımıyla Akış Sitometrisinde bu türevlerin doz ve zamana bağlı apoptotik etkilerine bakılmıştır. Son bölümde ise seçilmiş yüksek sitotoksik aktivite gösteren anti-kanser bileşiklerin, hücre siklusu ve apoptoz mekanizması üzerindeki etkileri için akış sitometrisi ve western blot analizi ile test edilmiştir. In cancer research, naturally isolated 5-substituted, α,β-unsaturated lactones have become attracted the attention of scientists increasingly due to its biological activities. One of the most important member of this group of compounds is goniothalamin. Goniothalamin has high cytotoxic activity against various cancer cells and causes a minimal effect on healthy cells so it is believed that it has selective cytotoxic activity against cancer cells.In the first part of this study, anticancer activities of novel 5,6-dihydro pyran derivatives were determined by MTT assay in MCF7, PC3, HCT116 p53+/+ and p53-/- cell lines. Apoptotic effects of the selected compounds were also examined with the help of FITC labeled Annexin V-PI in flow cytometry .In the second part, anti-cancer compounds having high cytotoxic compounds were chosen and tested for their effect on cell cycle and apoptosis mechanism by flow cytometry and western blot analysis. 77

The Baeyer-Villiger oxidation experiments provided an interesting 3,5-seco-4-nor-triterpene skeleton via ring opening followed by an unusual rearrangement. Tek kristal seryum oksit nanoparçacıkları Seryum nitratın [Ce(NO3)3.6H2O] CsOH ve RbOH alkali bazlarının sulu çözeltilerinin hazırlanmasının ardından hidrotermal metot ile sentenzelenmiştir.Taramalı ve geçirimli elektron mikroskopu ve X-ray kırınımı yöntemleri kullanılarak parçacıkların morfoloji ve kristal özellikleri saptanmıştır.Çalışmanın ilk kısmı reaksiyon zamanı, reaksiyon sıcaklığı, alkali baz çeşidi ve konsantrasyonu gibi parametrelerin değişiminin paraçacık boyutuna ve morfolojisine olan etkilerini içermektedir. Sonuçlara bakıldığında açıkça söylenebilir ki reaksiyon süresiyle parçacık boyutu doğru orantılı bir şekilde artım göstermiş olup reaksiyon süresi arttığında topaklanmanın yerini homojen dağılım almıştır. Reaksiyon sıcaklığıyla ilgili yapılan denemelerde 120 oC de sentezlenen nanoparçacıkların çubuk şeklinde olduğu, sıcaklık arttıkça kübik bir yapıya dönüştüğü gözlemlenmiştir. Ayrıca baz konsantrasyonunun artışıda parçacıkların büyümesinde etkili bir rol oynamıştır. Bu çalışmalara ilaveten UV-Vis spektrometri ve Florasans Spektrometri cihazları kullanılarak seryum oksit nanoparçacıklarının optik özellikleri araştırılmış olup UV-Vis spektrum sonuçlarından yola çıkılarak RbOH alkali bazıyla sentezlenen nanoparçacıkların CsOH ile sentezlenenlerden daha büyük olduğu tayin edilmiştir. Floresans sonuçlarında ise reaksiyon zamanı azalımının kristal kusurlarını azaltığı gözükmektedir.Son kısımda ise CeO2 nanoparçacıklarının katalitik özellikleri araştırılmaya çalışılmıştır. Nanoparçacıklar 2'-hidroksiçalkondan flavon maddesinin sentezlenmesinde katalizör olarak kullanılmıştır. Sentezi tamamlamak için çözgen, reaksiyon sıcaklığı, nanoparçacık çeşidi ve kullanılan baz gibi parametreler değiştirilmiş, ürün oluşumu GC, GC-MS ve TLC ile takip edilmiş, son ürün NMR ile karakterize edilmiştir. It was prooved that the size of ceria nanoparticles is directly proportional with the reaction time. The reaction temperature is also an important parameter that effect the morphology of nanoparticles. At 120 oC nanoparticles form rod like structure and as time goes they start to form cubic crystals. When the alkali base and concentration was changed the results showed that higher base concentration favors the particles to form bigger structures than that of lower concentrations. In addition, optical properties of CeO2 nanoparticles were studied by using the UV-Vis and Fluorescence Spectrometry. UV-Vis Spectroscopy results show that particle size of CeO2 nanoparticles synthesized in the presence of 8M RbOH are larger than that of synthesized in presence of 8M CsOH. When the reaction time decreases the Ce3+ defect increases based on the results of Fluorescence Spectrometry results.Second part of the study includes catalytic property of CeO2 nanoparticles. Ceria nanoparticles were used as catalyst in the synthesis of flavone from 2?-hydroxychalcone. Several reaction parameters were studied in order to achieve the flavone synthesis. TLC, GC, GC-MS and NMR were used in order to monitor the results of the reactios. 79
