In soybeans, the integument or seed coat is an important modulator between the external and internal environment. It plays a fundamental role in seed vitality, and its lignin content may influence the seed quality. The objective of this experiment was to evaluate the quality of soybean seeds from a partial diallel and their reciprocals and its relationship with lignin content, seed coat thickness, and deposition location. The seeds were also evaluated for physiological quality through germination and accelerated aging tests. The lignin content was quantified by absorbance, and the integument thickness was analyzed and measured using scanning electron microscopy. The most contrasting cultivars for lignin content were analyzed using fluorescence microscopy and histochemical techniques. Cultivars and their reciprocals differed in seed physiological quality. We found differences in the genotypes for integument thickness. Using histochemical techniques, autofluorescence was observed in the same regions identified as lignified. Positive staining for lignin was observed in the hilum region. Among the genotypes, we found variations in the physiological quality of seeds (germination and accelerated aging test), lignin content, and integument thickness.

Abstract: The selection of soybean genotypes with seeds of high physiological quality is key to increasing the likelihood of establishment and success in the field and thus reaping higher yields. The aim of this study was to evaluate the genetic control of the physiological quality of soybean seeds from a partial diallel cross. Six previously selected soybean cultivars, group 1 (CD 201, CA 115, MS 8400) and group 2 (CD 202, Syn 1263, Syn 1279), were intercrossed by a partial diallel cross, totalizing 24 treatments. Seeds from these cultivars and crosses were evaluated for seed physiological quality based on germination tests, first germination count, accelerated aging, seedlings emergence, and emergence speed index. The lignin content in the soybean seed coat was evaluated. The effects on general and specific combining ability and reciprocal effects were analyzed. There were significant additive and non-additive effects of the genes on the seed quality traits and pronounced effects on the reciprocal traits, which suggest the presence of a maternal effect. Lignin content was not correlated with the physiological test results in the studied genotypes.

Abstract Background The study of cold tolerance in maize seeds and seedlings through physiological quality assessments, as well as the genetic control associated with this trait, allows an early characterization of genotypes. Here we studied the genetic control for cold tolerance during the germination process in maize seeds and genes influenced by this stress. Results Six maize lines were used, three classified as tolerant and three as susceptible to low germination temperature. A field was developed to produce the hybrid seeds, in a partial diallel scheme including the reciprocal crosses. For the expression analysis, seeds from two contrasting lines were used, as well as their hybrid combination and their reciprocal crosses, on dried and moistened seeds at 10 °C for 4 and 7 days. It was evaluated the catalase (CAT) and esterase (EST) enzymes, heat-resistant proteins and the genes Putative stearoyl-ACP desaturase (SAD), Ascorbate Peroxidase (APX), Superoxide Dismutase (SOD) and Mitogen Activated Protein Kinase (ZmMPK5). The estimated values ​​for heterosis, general and specific combining abilities and reciprocal maternal and non-maternal effects were carried out and it showed that there is heterosis for germination at low temperatures, also the non-additive genes were more important and there was a reciprocal effect. Conclusions There is a greater expression of the CAT and EST enzymes in moistened seeds at seven days and there is less expression of heat-resistant proteins and the SAD gene at seven days of moistening. Also, there are variations in the expression of the APX, SOD and ZmMPK5 genes in dried and moistened seeds, as well as among the genotypes studied.

Drought stress is a major limiting factor for the development of maize, and the identification of the expression of genes related to this stress in seeds and seedlings can be an important tool to accelerate the selection process. The expression of genes related to tolerance to water deficit in seeds and in different tissues of maize seedlings were evaluated. Four tolerant genotypes (91-T, 32-T, 91x75-T, 32x75-T) and four non-tolerant genotypes (37-NT, 57-NT, 37x57-NT and 31x37-NT) were seeded in a substrate with 10% (stress) and 70% (control) water retention capacity. The expression of 4 enzymes were evaluated: catalase (CAT), peroxidase (PO), esterase (EST), and heat-resistant protein (HRP), as well as the relative expression of 6 genes: ZmLEA3, ZmPP2C, ZmCPK11, ZmDREB2A/2.1s, ZmDBP3 and ZmAN13 were evaluated in seed, shoots and roots of seedlings submitted or not to stress. There was variation in the expression of CAT, PO, SOD, EST and HRP enzymes among the evaluated genotypes and also in the different tissues evaluated. Higher expression of the CAT and PO was observed in the shoots. There was a greater expression of the EST in the genotypes non-tolerant to water deficit. HRP was expressed only in seeds. In the aerial part of maize seedlings, classified as tolerant, higher expression of genes ZmLEA3 and ZmCPK11 was observed. There was a higher expression of the ZmAN13 and ZmDREB2A/2.1S genes in roots developed under stress conditions and a higher expression of the ZmPP2C gene in seeds of line 91-T, which is classified as tolerant to drought stress.

The objective of this work was to evaluate the effects of different storage conditions on the physiological and sanitary quality and oil content of castor bean (Ricinus communis) seeds. Seeds of castor bean plants of the Guarani, and IAC-80 cultivars were stored in two environments (cold room, and room conditions), using three package types (multifoliate Kraft paper bag, and polyethylene bag, and polyethylene bag with vacuum at 1 atm). In addition, another storage condition was evaluated: cryopreservation (-196 °C) in foil paper bags. Seed quality was evaluated before storage and at 4, 8, and 12 months after storage by testing their 7-day and 14-day germination, emergence, health, water content, and oil content. The experiment was conducted in a completely randomized design, with 7×4 factorial arrangement consisting of seven storage conditions and four evaluation times. Cryopreservation is the ideal condition for maintaining the seed physiological quality of the Ricinus communis cultivars used throughout storage. The oil content of the R. communis seeds decreases, and the incidence of Aspergillus spp. and Fusarium spp. fungi increases throughout storage, regardless of the storage conditions.

Abstract: The aim of this study was to investigate the effect of treatment with insecticides and fungicides on the physiological quality of corn seeds, classified in sizes, in different periods of storage. Seeds of the hybrid BM915 PRO, classified in two sizes (CH20/64 and CH24/64), were treated with a mixture of carbendazim + thiram (Derosal Plus®), pirimiphos methyl (Actellic®), deltamethrin (K-obiol®), and water (standard treatment). In addition to the standard treatment adopted by the company, treatments with clothianidin (Poncho®), thiamethoxam (Cruizer®), and fipronil (Shelter®) were added to the spray mixture. The seeds were stored in a non-climate-controlled environment in multi-ply paper packages. The physiological quality of the seeds was evaluated every ninety days over a period of 270 days by the germination test, cold test, and accelerated aging. Sanitary quality was also evaluated through the Blotter test. The physiological quality of seeds of the hybrid BM915 PRO is maintained up to ninety days of storage, regardless of the chemical treatment used and the size of the seeds. The addition of the insecticide to the standard treatment used in chemical seed treatment does not affect the action of the fungicides on the fungi in the seeds.

ABSTRACT Knowing genes related to water deficit is a valuable tool to assist and accelerate the selection process of maize genotypes more adapted to drought conditions. Therefore, this work aimed to evaluate genes expression related to tolerance to water deficit in maize lines, using proteomic and transcriptomic analyses. Two lines previously classified as tolerant (91-T) and non-tolerant (57-NT) to low water availability were used. Dried seeds, ears tips, and seedlings (collected at three, five, and seven days after sowing) of the two maize lines (91-T and 57-NT) were used in proteomic analyses. Seedlings were subject to two contrasting water-availability conditions (70% and 10% of the water retention capacity). The expression of catalase, peroxidase, and esterase enzymes and heat-resistant proteins were evaluated by electrophoresis. Dried seeds, ears tips, and seedlings (collected at seven days after sowing, subject to two contrasting water-availability conditions) of two maize lines (91-T and 57-NT) were used in the transcriptomic analyses. The expression of genes associated with tolerance to low water availability (ZmPP2C, ZmLEA3, and AOX2) was evaluated by the qRT-PCR technique. Results revealed that catalase, peroxidase, and esterase enzymes had a higher expression on the ears tips of the 91-T line. The expression of catalase and esterase enzymes reduced in seven-day-old seedlings in the two lines analyzed. Heat-resistant proteins had greater activity on the dried seeds of the 91-T line. The AOX2 gene was identified as a potential marker for the selection of maize genotypes tolerant to water deficit.

The objective of this study was to evaluate gene expression related to water deficit tolerance in maize lines. For this, lines previously classified as tolerant (91-T and 32-T) and non-tolerant (24-NT and 57-NT) to water deficit were used. The seeds of the four lines were evaluated for emergence and emergence speed index, and the seedlings were evaluated for root and shoot length under two conditions of water availability (70 and 10% substrate water retention capacity). In transcript analysis, the expression of several genes associated with water deficit tolerance, ZmDBP3, ZmALDH9, ZmAN13, and ZmDREB2A, was evaluated by qRT-PCR for the 91-T and 57-NT lines. It can be concluded that soil water deficiency did not reduce root development. However, the shoot length was significantly lower under dry conditions. Through transcript analysis, the genes ZmDBP3 and ZmAN13 were identified as potential markers for the early selection of maize lines tolerant to water deficit.

Desiccation is a technique used by soybean growers, aiming to anticipate the harvest. However, for seed production this technique requires attention, because some desiccant herbicides can generate residues in the seeds damaging its quality. The objective of this study was to evaluate the desiccant effect of different spraying times on the physiological quality of soybean seeds, stored by six months, from different cultivars and lignin contents in the coat. Soybean plants from BRS Silvânia RR, BRS Valiosa RR, BRS 245 RR and BRS 247 RR cultivars were desiccated with Diquat, Paraquat and Glufosinate ammonium herbicides, and water as a control treatment, applied when the seeds presented 30, 40 and 50% of water content. After harvesting and processing, seeds were stored for six months in conventional warehouse. The physiological quality of soybean seeds was tested via germination, emergence, emergence speed index, electrical conductivity and tetrazolium. The physiological quality of seeds from cultivars BRS Valiosa RR and BRS Silvânia RR were lower than the seeds from cultivars BRS 245 and BRS 247. The desiccation, especially when the Glufosinate ammonium herbicide was applied, showed more damaged to the physiological quality of soybean seeds than the other treatments. = A dessecação é uma técnica utilizada pelos produtores de soja com o objetivo de antecipar a colheita. No entanto, para a produção de sementes essa técnica necessita de cuidados, uma vez que alguns dessecantes podem deixar resíduos na semente prejudicando sua qualidade. Objetivou-se com esse estudo avaliar o efeito da aplicação de dessecantes em diferentes épocas sobre a qualidade fisiológica de sementes, após seis meses de armazenamento, de cultivares de soja com diferentes teores de lignina no tegumento. As plantas das cultivares BRS Silvânia RR, BRS Valiosa RR, BRS 245 RR e BRS 247 RR foram dessecadas com os herbicidas Diquat, Paraquat e Glufosinato de amônio, e água como tratamento controle, aplicados quando as sementes apresentavam 30, 40 e 50% de teor de água. Após a colheita e beneficiamento, as sementes foram armazenadas por seis meses em armazém convencional. Para a avaliação da qualidade fisiológica das sementes foram realizados os testes de germinação, emergência, índice de velocidade de emergência, condutividade elétrica e tetrazólio. As sementes das cultivares BRS Silvânia e BRS Valiosa apresentaram menor qualidade fisiológica das sementes do que as cultivares BRS 245 e BRS 247. A dessecação, especialmente quando Glufosinato de amônio foi aplicado, mostrou-se mais prejudicial à qualidade fisiológica das sementes de soja do que os demais tratamentos.

The seeds of a sesame crop contain approximately 50% oil content, one of the main reasons for their cultivation. However, information regarding methodologies for the assessment of quality seeds of this culture are scarce. Therefore, the objective of this study was to adapt the methods of accelerated ageing tests for sesame seeds and investigate changes in the behaviour of isoenzymes. The physiological characterization of the seeds and electrophoretic analysis of the isoenzymes superoxide dismutase, esterase, catalase, alcohol dehydrogenase, malate dehydrogenase and isocitrate lyase were also performed. For the accelerated ageing test, the seeds were subjected to the traditional method, with a saturated NaCl solution, at ageing times of 0, 24, 48, 72, and 96 hours. It was found that it is possible to evaluate the effect of accelerated ageing tests on sesame seeds by using the traditional method for 72 hours and saturated NaCl solution at 45°C for 48 hours. When associated with the activity of isozymes EST, CAT, ADH, and ICL, it was possible to observe that there was a significant variation in the intensity of expression of the bands as the seed deterioration process progressed.

ABSTRACTArtificial chromosome duplication is one of the most important process in the attainment of doubled haploids in maize. This study aimed to evaluate the induction ability of the inducer line KEMS in a tropical climate and test the efficiency of the R1-Navajo marker by flow cytometry to evaluate two chromosome duplication protocols and analyze the development of the doubled haploids in the field. To accomplish this goal, four genotypes (F1 and F2 generations) were crossed with the haploid inducer line KEMS. The seeds obtained were selected using the R1-Navajo marker and subject to two chromosome duplication protocols. Duplication was confirmed using flow cytometry. The percentages of self-fertilized plants after duplication as well as the quantities of doubled haploid seeds obtained after the self-fertilization processes were analyzed. It was observed that the germplasm influences haploid induction but not the duplication rates of the tested protocols. Protocol 2 was more efficient for the duplication of haploids, in the percentage of self-fertilized plants after duplication, and in the attainment of doubled haploid lines. Moreover, the haploid inducer line KEMS can produce haploids in a tropical climate. Other markers, in addition to the R1-Navajo system, should be used in the selection of haploid seeds.

Handroanthus serratifolius seeds are considered orthodox and have some mechanisms that assist in desiccation tolerance, like heat-resistant proteins and the presence of protective systems against free radical production. In order to investigate the drying effect on the quality of Handroanthus serratifolius seeds collected with 28% water content two methods were used: slow drying (30 °C) and fast drying (45 °C) until water contents equaled 20%, 15%, 10% and 5%. Seed quality was assessed by germination test, first count of germination, speed germination, emergence and speed emergence. The expression of heat-resistant proteins and enzymes catalase, esterase and isocitrate lyase was assessed by electrophoresis. The gene expression of sHSP17.5, CAT3 and ICL6 was quantified by real time PCR. Higher values of germination and vigor were obtained in seeds with 20% water content, subjected to fast drying. There was induction of heat-resistant proteins expression for both drying methods. There was variation in the electrophoretic pattern of catalase, esterase and isocitrate lyase in seeds subjected to different treatments. The gene expression in Handroanthus serratifolius seeds varies depending on methods of drying and water content of seeds. The drying of Handroanthus serratifolius seeds until the water content between 15 and 10% compromises their physiological quality.

This study aimed to investigate the desiccation tolerance of Rhamnidium elaeocarpum Reissek seeds through physiological and biochemical alterations. Fruit were collected from the municipality of Jaupaci, Goias State, Brazil, when the water content of their seeds had a 37% wet basis (w.b.) and the seeds were subsequently kept in a drying oven with air circulation at a temperature of 35ºC until a water content was reached of 20 to 12% (w.b.). The following parameters were evaluated: germination, germination speed index, emergence, emergence speed index, electrical conductivity and respiration rate. Furthermore, the electrophoretic profile of the isoenzymes: catalase, peroxidase and esterase, the enzymatic activities of endo-β-mannanase and α-amylase and the electrophoretic analysis of heat-resistant proteins were performed. A higher germination speed and respiratory rate was obtained for seeds with a water content of 12% w.b., and the activity of catalase isoforms was more pronounced in seeds with a higher degree of dehydration, which was in contrast to peroxidase, showed decreased activity. The seeds contained heat-resistant proteins of low molecular weight that ranged from 48.7 to 13.2 kDa. We found that the acquisition of drying tolerance in R. Elaeocarpum Reissek seeds is associated more with catalase than with esterase and peroxidase.

The radish has been gaining importance in the Brazilian market due to its use as a green manure and cover crop, and also due to its potential for the production of biodiesel, however there are only two registered cultivars, which are morphologically very similar. Determination of genetic purity is a prerequisite in marketing seeds as it ensures uniformity of the crop and is important in breeding programs. The aim of this work therefore was to evaluate the similarity and genotype patterns which permit differentiation of the commercial radish cultivars IPR 116 and CATI AL-1000. In order to do this, isoenzyme electrophoretic patterns were analysed in dry seeds, soaked seeds, seedling leaves and young leaves; the isoenzymes used being: superoxide dismutase, catalase, esterase, glutamate-oxalocetate, malate dehydrogenase and isocitrate lyase. Thirty-seven RAPD primers and 10 ISSR primers in leaves of the CATI AL-1000 and IPR-116 cultivars were analysed. Among the isoenzymes under study, the most polymorphic were glutamate oxalocetate, malate dehydrogenase, esterase and superoxide dismutase, with the superoxide dismutase system giving the best characterisation for all stages of development. The catalase isoenzyme system did not make it possible to differentiate between cultivars at any stage of development, and isocitrate lyase was not revealed by the protocol used. In analysis of the markers, 27 RAPD primers and eight ISSR primers showed polymorphism. The results indicate that it is possible to determine reliable descriptors based on isoenzymes at different stages of development of the radish and with the use of RAPD and ISSR primers.

The technology developed by breeding programs is applied to coffee seeds; however, after processing and drying, they lose viability within a short period of time, thus making storage unsuitable. The objective of this research was to evaluate the quality of coffee seeds submitted to different drying methods and moisture contents during storage. The coffee seeds were submitted to conventional drying (slow shade drying) and fast drying in a static drier until they reached a moisture content of 40, 20, 12 and 5%. After this process, the seeds were stored in a cold chamber for 12 months, and seed quality was evaluated before and during storage by the germination test, electrophoretic patterns of heat resistant proteins, and the activity of isoenzyme systems. Conventional drying (slow shade drying) at 20% of moisture content maintains coffee seed quality until 12 months of storage.

Several factors affect the quality of soybean seeds, including the mineral nutrition of plants. Manganese (Mn) is an important nutrient because it has as one of its functions the enzyme activation. The aim of this study was to evaluate enzyme activity in seeds of soybean cultivars produced with foliar application of different doses of Mn. The experiment was conducted at the Universidade Federal de Lavras (UFLA) (Federal University of Lavras) in randomized blocks with three replications and 4 x 4 x 2 factorial arrangement consisting of four soybean cultivars, two conventional and its genetically modified RR derived (BRS Celeste and BRS Baliza RR; BRSGO Jataí and BRS Silvânia RR), four doses of Mn via foliar application (0; 200; 400 and 600 g Mn ha-1) and two stages of application (R1 or R3). In the seeds, the expressions of the enzymes esterase (EST), malate dehydrogenase (MDH), alcohol dehydrogenase (ADH), superoxide dismutase (SOD), peroxidase (PRX) and isocitratelyase (ICL) were determined. For evaluation of physiological quality, the germination test and emergence speed index (ESI) were performed. The Mn content in the seeds was also determined. The expression of the enzymes EST, SOD, PRX and ICL in soybean seeds are affected by foliar application of Mn, regardless of the stages of application. In the seeds of the cultivars that showed better physiological quality, Celeste and Baliza RR, greater expressions of the enzymes ADH and ICL and lower expressions of MDH and of PRX were observed.

Early characterization of genotypes, through the previous assessment of physiological quality of their seeds and their enzymatic systems, can provide parameters capable of aiding in the selection of new maize cultivars tolerant to water stress. Thus, this study was aimed at assessing seed performance of five different maize lines subjected to four different water stress levels. The physiological quality of seeds germinated under the osmotic potentials of 0.0, -0.3, -0.6 and -0.9 MPa was assessed by germination test, number of strong normal seedlings and T50, as well as by the expression of the isozymes superoxide dismutase (SOD), catalase (CAT), glutamate oxaloacetate transaminase (GOT), esterase (EST), malate dehydrogenase (MDH), alcohol dehydrogenase (ADH), and of heat-resistant proteins. The osmotic potential of -0.9 MPa affected the physiological quality and vigor of seeds of all maize lines assessed. The highest values for the germination percent, in the lowest osmotic potentials assessed, were found in the seeds of line 63. However, seeds of line 91 were superior in regard to vigor. Increased expression of the enzymes SOD and CAT, considered as enzymes of the antioxidant system, was observed in seeds of lines 44 and 91. Thus, line 91 was considered as promising for tolerance to water stress.

With the growth in the transgenic market, fast and economically viable methodologies are necessary for undertaking transgene detection tests, both for identification of contamination in seeds and in grain. Seeds from commercial conventional GNZ 2004, and transgenic VT-Pro (MON89034), Roundup Ready (NK603) and Herculex (TC1507) maize cultivars were used. In order to simulate different levels of contamination, the transgenic seeds were mixed with conventional seeds at levels of 0.2%, 0.4%, 1.0% and 1.6% for VT-Pro, and 0.2%, 0.5%, 0.8% and 1.2% for Roundup Ready and Herculex. The lateral flow membrane strip test was performed in the whole seed, endosperm and embryo. For evaluation of the specificity of the technique in detection of the TC1507 event by means of the conventional PCR technique, seeds of the commercial maize hybrid GNZ 2004 were used as negative control, and the maize hybrid 2B655Hx as positive control. In order to simulate different levels of contamination, transgenic seeds were mixed with conventional seeds at the levels of 10%, 5%, 1%, 0.5% and 0.1%. Seeds from each sample were crushed, and then DNA extraction was performed by the CTAB 2% method. Using the immunochromatographic strip, it was possible to evaluate the expression of proteins related to the VT-Pro, Roundup Ready and Herculex events when whole seeds were used at the 0.2% level of contamination, whereas by the conventional PCR technique, it was possible to detect the TC1507 event in samples with 1% contamination.

The physiological quality of maize seeds is affected by the genotype. Thus, the study of expression of genes associated with this characteristic is important in the genotype selection process in breeding programs. The aim of this research was to study the expression of amylase enzymes associated with physiological quality of maize seeds with different genotypes and seed sizes. We further sought to assess the expression of these enzymes in dry and soaked seeds The experiment was conducted in the experimental area and the Central Seed Laboratory of the Department of Agriculture of the Universidade Federal de Lavras. Seeds of four maize inbred lines were used, classified in two sizes. The physiological quality of the seeds was evaluated by means of germination, seedling emergence, seedling emergence speed index and accelerated aging test. Expression of the alpha amylase enzyme was evaluated by the electrophoresis technique and expression of the alpha amylase B73, alpha amylase (LOC542522) and beta amylase 5 (amyb5) genes was studied by the qRT-PCR technique in dry and soaked seeds of the inbred lines. There is differentiated expression of amylase enzymes in maize seeds of inbred lines with different levels of physiological quality. higher expression of amylase enzymes is observed in soaked maize seeds. The expression of transcripts is higher in smaller as wellas in soaked maize seeds of inbred lines.A qualidade fisiológica de sementes de milho é influenciada pelo genótipo. Assim, o estudo da expressão de genes associados a essa característica é importante no processo de seleção de genótipos em programas de melhoramento. O objetivo neste trabalho foi estudar a expressão das enzimas amilases associadas à qualidade fisiológica de sementes de milho, de diferentes genótipos e tamanhos de sementes. Objetivou-se ainda avaliar a expressão dessas enzimas em sementes secas e embebidas. O experimento foi conduzido em área experimental e no Laboratório Central de Sementes do Departamento de Agricultura da Universidade Federal de Lavras. Foram utilizadas sementes de quatro linhagens de milho, classificadas em dois tamanhos. A qualidade fisiológica das sementes foi avaliada por meio dos testes de germinação, emergência de plântulas, índice de velocidade de emergência de plântulas e envelhecimento artificial. A expressão da enzima -amilase foi avaliada pela técnica de eletroforese e a expressão dos genes alpha amylase B73, alpha amylase (LOC542522) e beta amylase 5 (amyb5), foi estudada pela técnica de qRT-PCR em sementes secas e embebidas das linhagens. Há expressão diferenciada das enzimas amilases em sementes de linhagens com diferentes níveis de qualidade fisiológica. Maior expressão das enzimas amilases é observada em sementes de milho de menor tamanho e embebidas.

Injúrias nas estruturas internas da semente limitam sua viabilidade e podem reduzir seu vigor, produzindo plântulas fracas e susceptíveis às condições adversas. Dessa forma, a determinação da qualidade de sementes de Physalis peruviana L. pelo teste de raios X é válida, visando a redução de tempo na separação das sementes e um bom estabelecimento inicial da cultura. Diante do exposto, objetivou-se com o presente trabalho avaliar a qualidade de sementes de fisális pelo teste de raios X em diferentes fases de desenvolvimento. As sementes foram removidas e separadas em lotes de acordo com a fase de maturação dos frutos, sendo frutos com maturação intermediária (50% do fruto maduro), frutos completamente maduros e frutos completamente maduros submetidos a sete dias de repouso após a colheita. Posteriormente foram submetidas ao teste de raios X para determinação da qualidade. O delineamento experimental constituiu-se de oito repetições de 100 sementes por lote. Sementes de fisális de frutos com 50% de maturação apresentam baixa qualidade morfofisiológica quando comparadas às sementes extraídas de frutos completamente maduros. O vigor das sementes é maior quando os frutos são submetidos ao repouso por sete dias. O teste de raios X é eficiente para seleção de sementes não viáveis de fisális.

A dessecação é uma técnica utilizada pelos produtores de soja com o objetivo de antecipar a colheita. No entanto, para a produção de sementes essa técnica necessita de cuidados, uma vez que alguns dessecantes podem deixar resíduos na semente prejudicando sua qualidade. Objetivou-se com esse estudo avaliar o efeito da aplicação de dessecantes em diferentes épocas sobre a qualidade fisiológica de sementes, após seis meses de armazenamento, de cultivares de soja com diferentes teores de lignina no tegumento. As plantas das cultivares BRS Silvânia RR, BRS Valiosa RR, BRS 245 RR e BRS 247 RR foram dessecadas com os herbicidas Diquat, Paraquat e Glufosinato de amônio, e água como tratamento controle, aplicados quando as sementes apresentavam 30, 40 e 50% de teor de água. Após a colheita e beneficiamento, as sementes foram armazenadas por seis meses em armazém convencional. Para a avaliação da qualidade fisiológica das sementes foram realizados os testes de germinação, emergência, índice de velocidade de emergência, condutividade elétrica e tetrazólio. As sementes das cultivares BRS Silvânia e BRS Valiosa apresentaram menor qualidade fisiológica das sementes do que as cultivares BRS 245 e BRS 247. A dessecação, especialmente quando Glufosinato de amônio foi aplicado, mostrou-se mais prejudicial à qualidade fisiológica das sementes de soja do que os demais tratamentos.

O objetivo deste trabalho foi estimar o fluxo gênico em milho transgênico com resistência a insetos, em lavouras comerciais. Amostras de grãos foram coletadas em lavouras com milho convencional e transgênico, nos municípios de: Itumirim, Uberlândia, Paracatu e Tupaciguara, MG; Itapetininga e Pedrinhas, SP; e Assaí e Ponta Grossa, PR. As amostras foram coletadas em lavouras de milho convencional, a partir de 5 m de distância da fonte com o milho transgênico. Foram coletadas dez espigas de plantas individuais por ponto, em quatro repetições, no total de 40 espigas para cada distância amostrada. As análises de fluxo gênico foram realizadas por meio da técnica de reação em cadeia da polimerase (PCR) em tempo real. Em média, 82% da fecundação cruzada ocorreu nos primeiros 30 m. Em Itapetininga, SP, foram observadas as maiores taxas de fecundação cruzada - acima de 10% até a distância de 50 m -, porém, inferiores a 1% na distância de 100 m. O isolamento de 20 m, com dez linhas de bordadura, não é suficiente para garantir taxas de fecundação cruzada inferiores a 1%.

The main objectives of hybrid development programs include incorporating genetic resistance to diseases and increasing grain yield. Identification of Quantitative Trait Loci (QTL) through the statistical analysis of molecular markers allows efficient selection of resistant and productive hybrids. The objective of this research was to identify QTL associated with resistance to gray leaf spot and for grain yield in the germplasm of tropical corn. We used two strains with different degrees of reaction to the disease; the genotypes are owned by GENESEEDS Ltda, their F1 hybrid and the F2 population. The plants were evaluated for gray leaf spot resistance, for grain yield and were genotyped with 94 microsatellite markers. Association of the markers with the QTL was performed by single marker analysis using linear regression and maximum likelihood analysis. It was observed that the additive effect was predominant for genetic control of resistance to gray leaf spot, and the dominant effect in that of grain yield. The most promising markers to be used in studies of assisted selection are: umc2082 in bins 4.03 and umc1117 in bins 4.04 for resistance to gray leaf spot; for grain yield umc1042 in bins 2.07 and umc1058 in bins 4.11.A incorporação de resistência genética a doenças e o aumento na produtividade de grãos estão entre os principais objetivos dos programas de desenvolvimento de híbridos. A identificação de locos de caracteres quantitativos (QTL) por meio de análises estatísticas associadas a marcadores moleculares possibilita a rápida obtenção de híbridos resistentes e produtivos. Nesta pesquisa, objetivou-se identificar locos de caracteres quantitativos (QTL) associados com resistência à cercosporiose e com produção de grãos em germoplasma de milho tropical. Foram utilizadas duas linhagens contrastantes em níveis de reação à doença (genótipos pertencentes à GENESEEDS - Ltda), seu híbrido F1 e a população segregante F2. Essas plantas foram fenotipadas quanto à resistência a doença e quanto a produção de grãos e genotipadas com 94 marcadores de microssatélites. A associação dos marcadores ao QTL foi realizada por meio de análises de marcas individuais, utilizando as metodologias de regressão linear e análise da máxima verossimilhança. Observou-se que o tipo de efeito predominante no controle genético da resist��ncia a cercosporiose foi o aditivo e para o controle genético da produção de grãos foi de dominância. Os marcadores microssatélites mais promissores para serem utilizados em estudos de seleção assistida para resistência a cercosporiose são umc2082 na posição 4.03 e umc1117 na posição 4.04 e para produção de grãos, umc1042 e umc1058 nas posições 2.07 e 4.11.

A caracterização de cultivares de flores é fundamental nos processos de registro e proteção dessas e também na certificação da pureza genética. Nesse trabalho foram realizadas a caracterização e a certificação da pureza genética de cultivares de Gladiolus sp. por meio de marcadores morfológicos. Utilizaram-se onze cultivares de Gladiolus sp., e para a caracterização das cultivares, as parcelas foram constituídas de 25 plantas avaliando-se características dos cormos, folhas e flores. O delineamento experimental adotado foi de blocos casualizados, com quatro repetições Para a certificação da pureza genética, foram instaladas parcelas constituídas de 24 plantas e cormos contendo contaminação varietal em diferentes proporções. Foram utilizadas três cultivares para compor os tratamentos e as avaliações foram conduzidas por três avaliadores que desconheciam a porcentagem existente de contaminação varietal. Foram calculadas as porcentagens de acerto e de erro com base nos resultados obtidos nas avaliações. Foi calculado o desvio padrão da média dos resultados nas quatro repetições, obtidos pelos três avaliadores. Para verificar a significância dos desvios entre os resultados obtidos pelos avaliadores foi utilizado o teste de qui-quadrado ( χ²). Por meio de características do cormo é possível diferenciar quatro cultivares das onze avaliadas. A característica da cor das flores é o descritor morfológico mais seguro para a diferenciação das cultivares de gladíolo. Dependendo das características das cultivares contaminantes e das cultivares em análises é possível utilizar descritores morfológicos para a certificação de pureza genética em cultivares de gladíolo.The characterization of flower cultivars is fundamental in the registration and protection processes and also in the genetic purity certification. In this work, both the characterization and the genetic purity certification of cultivars of Gladiolus sp by means of morphologic markers were made. Eleven cultivars of Gladiolus sp. were used and for the characterization of the cultivars, the plots were made up of 25 plants, evaluating the characteristics of corms, leaves and flowers. The used experimental design was the one of randomized blocks with four replicates. For the genetic purity certification, plots with 24 plants and corms containing varietal contaminations in different proportions were established. Three cultivars were used to make up the treatments, and the evaluations were conducted by three evaluators who were unaware of the existent percentage of varietal contamination. The percentage of mistakes was calculated based upon the obtained results of the evaluations. The standard deviation of the means of the results in the four replicates obtained by the three evaluators was calculated. In order to verify the significance of the deviations among the obtained results by the evaluators, Chi-square test was used. By means of corm characteristics, it is possible to distinguish four cultivars out of the eleven evaluated. The characteristic of flower color is the safest morphological descriptor for distinguishing gladiolus cultivars. Depending on the characteristics of the contaminant cultivars and on the ones under analyses, it is possible to use morphological descriptors for the genetic purity certification in gladiolus cultivars.

A composição química das sementes pode ser influenciada por fatores genéticos, ambientais e pela disponibilidade de nutrientes no solo durante a produção e, como consequência, terem o seu vigor afetado. O potássio pode influenciar a atividade de várias enzimas presentes nas plantas, necessárias em várias reações envolvidas na utilização de energia, síntese de amido, metabolismo do nitrogênio e respiração. Avaliou-se o efeito da adubação potássica e da calagem na composição química, qualidade fisiológica e na atividade de enzimas de sementes de soja. A pesquisa foi desenvolvida nos laboratórios de análises e biotecnologia de sementes do Departamento de Agricultura e no Laboratório de Produção Vegetal do Departamento de Ciências dos Alimentos da Universidade Federal de Lavras (UFLA). Em sementes de soja do cultivar Msoy 8001, produzidas sob 2 níveis de saturação por base (48% e 85%) e quatro doses de K2O por hectare (0, 50, 100 e 200 kg), foram avaliados teores de óleo e proteína, germinação, envelhecimento acelerado e atividade das enzimas envolvidas nos processos de germinação e deterioração de sementes. A adubação potássica não afeta a germinação e o vigor das sementes, enquanto a elevação da saturação por base, proporciona maior vigor de sementes de soja. Há aumento no teor de óleo e redução no teor de proteína em sementes de soja à medida que a dose de K2O é aumentada. Ocorre aumento de teor de proteínas em sementes de soja, produzidas sob maiores níveis de saturação por base. A atividade das enzimas piruvato quinase, esterase e álcool desidrogenase em sementes de soja é afetada pela concentração de potássio e saturação por base no solo.The chemical composition of seeds can be influenced by genetical and environmental factors and by the availability of nutrients in the soil during production, which consequently affect their vigor. Potassium can influence the activity of several enzymes present in plants. These enzymes are necessary in a number of reactions involved in the utilization of energy, starch synthesis, nitrogen metabolism and respiration. The effects of potassium fertilization and liming on the chemical composition, physiological quality and enzymes activities of soybean seeds were evaluated. The research was developed in the Seed Analysis Laboratory of the Agriculture Department and in the Plant Production Laboratory in the Food Science Department of the Federal University of Lavras, Lavras (UFLA). In soybean seeds of cultivar Msoy 8001 produced under two levels of base saturation (48% and 85%) and four doses. ha-1 of K2O, 0, 50, 100 and 200, oil and protein contents, seed germination, aging test and the activity of some enzymes involved in the seed germination and deterioration processes were evaluated. Seed germination and vigor are not influenced by fertilization with potassium. However, base saturation increases the vigor of soybean seeds. Highert doses of potassium increase oil content and reduce protein in soybean seeds. High base saturation level increases the protein content. The activities of the enzymes piruvate kinase, esterase and alcohol dehydrogenase are altered by the doses of potassium and levels of soil base saturation.

O objetivo deste trabalho foi avaliar os efeitos gênicos aditivos e não aditivos para as características da espessura das camadas da testa e da lignina presente nessas camadas, e verificar se os caracteres utilizados para avaliação da qualidade fisiológica das sementes de soja se correlacionam com o teor de lignina do tegumento. Foram usadas três cultivares (M-Soy 8400, CD201, CD206) de alta e três (CD215, CD202, Savana) de baixa qualidade fisiológica, para se obterem sementes híbridas F1 e F2, inclusive os recíprocos, pelo sistema de cruzamento dialelo parcial e as linhagens parentais. Os parâmetros primeira contagem de germinação, contagem final de germinação e velocidade de germinação, utilizados para avaliação da qualidade fisiológica, podem ser correlacionados com o teor de lignina. Foram observadas correlações positivas entre o teor de lignina e a percentagem de plântulas normais no teste de envelhecimento ao 5º e 11º dias após a semeadura, com valores maiores de correlação no 5º dia. A correlação entre velocidade de germinação e teor de lignina foi negativa. A análise do dialelo apontou significância do efeito recíproco, o que indica que a espessura das camadas da testa e a espessura de lignina nessas camadas podem ser explicadas pelo efeito materno.The objective of this work was to evaluate the additive and non-additive genetic effects for thickness of the seed coat layers and of the lignin present in them, and to verify if the characters used to evaluate the physiological quality of the seeds is correlated with the lignin content. Three cultivars (M-Soy 8400, CD201, CD206) with high and three (CD215, CD202, Savana) with low physiological quality were used to obtain F1 and F2 hybrid seeds, including the reciprocals, using partial diallel crosses and the parental lines. First germination count, final germination count and germination rate, used to evaluate the physiological quality, can be correlated with lignin content. Positive correlations were observed between lignin content and the percentage of normal seedlings in the aging test at the 5th and 11th days after sowing, with the highest correlation values at the 5th day. The correlation between germination rate and lignin content was negative. The diallel analysis showed reciprocal effect, showing that the thickness of the seed coat layers and of the lignin contained in these layers can be explained by maternal effect.

The objective of this work was to compare the health and physiological quality of double crosses-maize hybrid seeds obtained from F1 and F2 generations of single crosses hybrids. Field trials were carried out at the Federal University of Lavras – UFLA in two sowing dates, September and December 2001. Four isolated plots were used in each sowing date to produce seeds of the double hybrids DKB333/AG9012 and DKB901/AG9012. In two plots the female parental consisted of the F1 generations of the single hybrids DKB333 e DKB901 and the male parental was the single cross hybrid AG 9012. On the other two plots, the female parental consisted of F2 generations of the single crosses hybrids DKB333 e DKB901. The following agronomic characteristics were measured: plant height, ear insertion, ear weight and seed production. Seeds, size 22, were used to evaluate: germination, cold test, artificial aging, speed of emergence, emergence in substract of soil and sand mixture, seedling dry weight matter and health analysis by the blotter test. The experimental design was completely randomized using a factorial with two hybrids, two generations (F1 and F2), two sowing dates and four replications. The results showed a 28.7% decrease in seed production when the female parental came from the F2 generation instead of the F1 generation. For both hybrids, DKB901/AG9012 and DKB333/AG9012 sowed in September, the decrease in seed production was around 46% for the plants originated from F2 generation when compared to those from F1 generation. In December the reduction was 10.8%. All physiological assays showed a better performance for seeds obtained from F1 generations of the single hybrids. The same was true for the health tests.

Na aquisição e manutenção da tolerância à dessecação de sementes, há vários mecanismos envolvidos, entre eles a indução das proteínas resistentes ao calor. O objetivo deste trabalho foi avaliar mudanças no padrão eletroforético das proteínas resistentes ao calor de sementes de milho submetidas a alta temperatura de secagem, associando-as à sua tolerância. Foram utilizadas sementes de linhagens, híbridos simples e híbridos recíprocos colhidas com teor de água de aproximadamente 35% e secadas a 45°C. Sementes das linhagens secadas à sombra foram utilizadas como controle e sua qualidade fisiológica foi avaliada por meio do teste de germinação. As proteínas resistentes ao calor foram extraídas de eixos embrionários das sementes em tampão Tris HCl 0,05 M. Não foi possível determinar uma banda específica da fração das proteínas resistentes ao calor que possa servir como marcador da tolerância à alta temperatura de secagem. Houve estabilidade nos padrões de bandas das proteínas provenientes de sementes submetidas à secagem artificial e natural, mesmo quando foram observadas variações nos valores de germinação. Os padrões eletroforéticos das proteínas resistentes ao calor foram semelhantes entre as sementes híbridas e os respectivos recíprocos.Several mechanisms are involved in the acquisition and maintenance of desiccation tolerance by the seeds. One of these mechanisms is related to the induction of heat resitant proteins. The objective of this work was to evaluate changes in the electophorectic patterns of the heat resistant proteins of corn seeds submitted to high drying temperature associating their expression with drying tolerance. Seeds of breeding lines, simple hybrids and respective reciprocals hybrids were utilized. The seeds were harvested with approximately 35% of water content and dried at 45°C. Seeds of lines dried under shadow were used as control and the physiological quality was evaluated by germination test. The heat resistant proteins were extracted from embryonic axis of seeds in Tris-HCl 0.05 M buffer. It was not possible to determine a specific band of heat resistant proteins which can be of use as a molecular marker of tolerance to high drying temperature. There was stability in the patterns of the bands of proteins of seeds submitted to artificial drying compared to the ones after shadow drying even for the lines of high variation in the germination values. The electrophorectic patterns of the heat resistant proteins were similar for both the hybrid seeds and respective reciprocals.

Seeds tolerance to high drying temperature is an important characteristic to be evaluated in the corn breeding programs. In this research the genetic control of this characteristic was studied by using the seed physiological characteristics. Twelve lines, previously selected were used, being six tolerant and six intolerant to high drying temperature, in a 6 x 6 partial diallel. The seeds were harvested by hand on corn ears with water content around 35% and dried at 45oC up to 8% water content. The physiological quality of the seeds was evaluated by germination test, cold test without soil, accelerated aging and electrical conductivity. The effects of the general (GCA) and specific (SCA) combining ability, and the reciprocal effects were significant for the high drying temperature tolerance. Inside the genotypical variability observed, the reciprocal effects occurred for 47% and 44,7% for the accelerated aging and electrical conductivity test, respectively, and for 49% for germination and cold tests. In the genetic control for the high drying temperature tolerance maternal effect will probably occur.

Cultivares tolerantes a altas temperaturas de secagem proporcionam redução no tempo de secagem, uma etapa crítica no sistema de produção de sementes de milho (Zea mays L.). Nesta pesquisa, foi avaliada a tolerância à alta temperatura de secagem de sementes de linhagens de milho, por meio de testes de germinação e vigor. As sementes foram colhidas manualmente em espigas com teor de água em torno de 35% e secas artificialmente à 45 C até atingirem 11% de teor de água. Em seguida, foram submetidas aos testes de primeira contagem e contagem final de germinação, envelhecimento acelerado, teste de frio sem solo e de condutividade elétrica. Houve diferenças significativas nos valores de germinação e vigor de sementes das diferentes linhagens, sendo então classificadas em tolerantes e intolerantes. Pelos resultados, conclui-se que a sensibilidade das sementes à injúria por secagem à alta temperatura é dependente da linhagem.High drying temperature tolerant cultivars provide a reduction in the drying period, a critical phase of the corn seeds (Zea mays L.) production system. In this research the tolerance of corn lines seeds to high drying temperature was evaluated by the germination and vigor tests. Seeds were handpicked in ears with water content around 35% and dried artificially at 45ºC up to 11% water content. Then, the seeds were submitted to the first and final germination counting tests, accelerated aging, cold test without soil and electrical conductivity. There were significant differences in the germination and vigor values of seeds from different lines, being classified into tolerant and intolerant. The results permitted to conclude that sensitivity of seeds to high drying temperature injury depends on the lines.

Aiming at obtaining additional subsides to decision-making concerning harvest time and corn seeds drying on ears, biochemical and physiological aspects of seeds harvested at different milk line stages and submitted to artificial drying were studied. The experiments were accomplished at UFLA Seed Laboratory in association with Monsanto do Brasil S.A. Seeds of the hybrids AG-9090, AG-9010, DKB-350 and DKB-747, produced in the 2000 winter in the towns of Iraí de Minas, MG, Campo Florido, MG, Miguelópolis, SP and Guaíra, SP were utilized, respectively. In each field, four rows were demarcated and 100 ears per milk line stage (ML) were harvested at 25% (ML2), 50% (ML3), 75% (ML4) and 100% (ML5) of hardened endosperm. The ears were sampled to determine the seed moisture content. After straw removal, ears were dried at initial temperature of 35oC till they reached 20% of moisture content and 42oC up to 12% of water content. Germination , electric conductivity and accelerated aging tests and cold test without soil, as well as electrophoresis of a-amylase and heat resistant proteins analyses were carried out. These analysis were done for freshly harvested or dried seeds. We concluded that the germinability of corn seeds increases with water loss in the field. It is fully obtained after a moderate drying of the seeds harvested from milk line stage 3 (ML3). In this phase seeds have already achieved desiccation tolerance and have a high physiological quality.

Stable and polymorphic markers are required for the identification and registration of the cultivars. The purpose of this research was to evaluate the heat resistant proteins polymorphism and stability of seeds from corn lines harvested in different years and dried under natural and artificial conditions, in order to employ such proteins in the identification and registration of the cultivars. The seeds showed different physiological quality when evaluated by germination test and the heat resistant proteins were extracted from embryonic axis from seeds of each line, in Tris-HCl 0,05 M buffer. The eletrophoretic patterns of these proteins in SDS-PAGE showed stable bands for each genotype even for the lines with higher variation in the germination test. The most similar corn inbred lines were distinguished, at least by two bands, showing that these proteins were highly polymorphic. The stability and the high level of polymorphism of the heat resistant proteins, make them potential markers in corn cultivars registration and identification programs.

O desequilíbrio nutricional, principalmente dos micronutrientes, tem sido um dos fatores para perdas na produção de sementes. Esses nutrientes desempenham papel em rotas bioquímicas que garantem a formação de lipídeos, proteínas e ainda contribuem na estruturação das membranas celulares. O objetivo deste trabalho foi avaliar a influência da aplicação de Mn na folha e no solo, sobre o rendimento e a qualidade de sementes de soja, cultivares Conquista e Garimpo. A qualidade das sementes foi avaliada por meio de testes de germinação, envelhecimento artificial, condutividade elétrica, índice de velocidade de emergência e pela determinação dos teores de proteína e óleo. Verificaram-se aumentos na produtividade, na germinação, no vigor e nos teores de proteína e óleo das sementes e no teor de Mn na planta, nas duas formas de aplicação do Mn, com maior grau de eficiência da aplicação na folha.

Aiming at evaluating the physiological quality of hybrid seeds of corn harvestedat different milk line stages after drying and storage, the present work was performed at theUniversidade Federal de Lavras (UFLA) seed laboratory together with Monsanto do Brasil S/A. Seeds of the hybrids Ag-9090, AG-9010, DKB-350 and C-747 produced in the winter of2000 in the towns of Iraí de Minas - MG, Campo Florido - MG, Miguelópolis -SP and Guaíra- SP were utilized. In each field were drawn four rows, where 100 ears were harvested per milk line stage (ML), which are ML2, ML 3, ML 4 and ML 5, displaying, respectively, 25%,50%, 75% and 100% of hardened endosperm were harvested . The ears were transported tothe UFLA seed laboratory where they were sampled for determination of water content ofseeds and next, they had their straws removed and were taken to the experimental dryerswhere they were dried at the initial temperature of 35OC to reach 20% of water content followedby temperature of 42OC to 12% of water content. Tests of germination, electric conductivity,cold test without soil and accelerated aging after drying and after 8 month storage wereaccomplished. After drying, values above 95% in the germination tests and above 85% in thevigor tests were found After 8 months storage lower values of germination and vigor for theseeds harvested at ML-2 for all the hybrids evaluated were observed. The results enabled usto conclude that it is safe to harvest corn seeds from the milk line 3 stage ( ML-3) when theyhave 50% of their endosperm solid and a high physiological quality.

Este trabalho teve como objetivo estudar a interferência dos fungos Aspergillus flavus, Fusarium moniliforme e Penicillium spp. sobre padrões eletroforéticos das sementes de milho. Tais padrões são, normalmente, utilizados na identificação de cultivares e na certificação da pureza genética da espécie em estudo. Sementes da cultivar C-805 foram infectadas artificialmente com os referidos fungos; outra parte delas foi tratada com Benomil e Thiabendazol, e ainda outra parte (controle) não foi tratada. As amostras foram acondicionadas em câmara de crescimento (25°C, 95% de umidade relativa) por um período de 30 dias. Na análise eletroforética foi avaliada também uma amostra de sementes que não permaneceu em câmara de crescimento, visando detectar possíveis interferências das condições do ambiente de crescimento sobre os padrões eletroforéticos. Os resultados obtidos permitiram concluir que a infecção das sementes com os fungos Aspergillus flavus, Fusarium moniliforme e Penicillium spp. promove alterações nos padrões eletroforéticos das isoenzimas malato-desidrogenase, esterase, fosfatase ácida, peroxidase e glutamato-oxalacetato-transaminase. A infecção das sementes com Aspergillus flavus promove alterações tanto na intensidade como no número de bandas dos padrões isoenzimáticos da álcool-desidrogenase e malato-desidrogenase.

Nesse trabalho foram avaliados marcadores moleculares de proteína visando à caracterização de variedades de gladíolos.

As pesquisas relacionadas com a certificação da pureza genética em flores são escassas. Nesse trabalho foram realizadas a caracterização e a certificação da pureza genética de variedades de Gladilous sp. por meio de marcadores morfológicos.

Early characterization of genotypes, through the previous assessment of physiological quality of their seeds and their enzymatic systems, can provide parameters capable of aiding in the selection of new maize cultivars tolerant to water stress. Thus, this study was aimed at assessing seed performance of five different maize lines subjected to four different water stress levels. The physiological quality of seeds germinated under the osmotic potentials of 0.0, -0.3, -0.6 and -0.9 MPa was assessed by germination test, number of strong normal seedlings and T50, as well as by the expression of the isozymes superoxide dismutase (SOD), catalase (CAT), glutamate oxaloacetate transaminase (GOT), esterase (EST), malate dehydrogenase (MDH), alcohol dehydrogenase (ADH), and of heat-resistant proteins. The osmotic potential of -0.9 MPa affected the physiological quality and vigor of seeds of all maize lines assessed. The highest values for the germination percent, in the lowest osmotic potentials assessed, were found in the seeds of line 63. However, seeds of line 91 were superior in regard to vigor. Increased expression of the enzymes SOD and CAT, considered as enzymes of the antioxidant system, was observed in seeds of lines 44 and 91. Thus, line 91 was considered as promising for tolerance to water stress.

The radish has been gaining importance in the Brazilian market due to its use as a green manure and cover crop, and also due to its potential for the production of biodiesel, however there are only two registered cultivars, which are morphologically very similar. Determination of genetic purity is a prerequisite in marketing seeds as it ensures uniformity of the crop and is important in breeding programs. The aim of this work therefore was to evaluate the similarity and genotype patterns which permit differentiation of the commercial radish cultivars IPR 116 and CATI AL-1000. In order to do this, isoenzyme electrophoretic patterns were analysed in dry seeds, soaked seeds, seedling leaves and young leaves; the isoenzymes used being: superoxide dismutase, catalase, esterase, glutamate-oxalocetate, malate dehydrogenase and isocitrate lyase. Thirty-seven RAPD primers and 10 ISSR primers in leaves of the CATI AL-1000 and IPR-116 cultivars were analysed. Among the isoenzymes under study, the most polymorphic were glutamate oxalocetate, malate dehydrogenase, esterase and superoxide dismutase, with the superoxide dismutase system giving the best characterisation for all stages of development. The catalase isoenzyme system did not make it possible to differentiate between cultivars at any stage of development, and isocitrate lyase was not revealed by the protocol used. In analysis of the markers, 27 RAPD primers and eight ISSR primers showed polymorphism. The results indicate that it is possible to determine reliable descriptors based on isoenzymes at different stages of development of the radish and with the use of RAPD and ISSR primers.

The aim of this study was adapting the methodology of the accelerated aging test (AA) and electrical conductivity test (EC) to evaluate seed quality of crambe, as well as verifying enzymatic activity in relation to vigor differences. Crambe seed lots, cv. FMS Brilhante, were evaluated by the AA test, for periods of 0, 24, 48, 72 and 96 h, at 42 °C, with and without NaCl saturated solution; and the EC test, by soaking seeds into deionized water (mL) in the ratios of 25/25, 25/50, 50/50 and 50/75 (seed/water) for 2, 4, 6, 8, 10, 12, 14, 16 and 18 h. The electrophoretic profile for isozymes esterase, superoxide dismutase, catalase, isocitrate lyase, alcohol dehydrogenase and malate dehydrogenase was evaluated on polyacrylamide gel (7.5%). The AA test conducted at 42 °C, for 96 h, was efficient to detect differences on seed quality levels; however, the EC test was not suitable to evaluate vigor of those seeds. The bands stained for isozymes esterase, superoxide dismutase and catalase were evident for the more vigorous seeds; and the bands for isozymes isocitrate lyase and alcohol dehydrogenase were less evident for the less vigorous seeds. For enzyme malate dehydrogenase enzyme intensity of bands was similar.

Abstract Among the plant molecular mechanisms capable of effectively mitigating the effects of adverse weather conditions, the heat shock proteins (HSPs), a group of chaperones with multiple functions, stand out. At a time of full progress on the omic sciences, they look very promising in the genetic engineering field, especially in order to conceive superior genotypes, potentially tolerant to abiotic stresses (AbSts). Recently, some works concerning certain families of maize HSPs (ZmHSPs) were published. However, there was still a lack of a study that, with a high degree of criteria, would fully conglomerate them. Using distinct but complementary strategies, we have prospected as many ZmHSPs candidates as possible, gathering more than a thousand accessions. After detailed data mining, we accounted for 182 validated ones, belonging to seven families, which were subcategorized into classes with potential for functional parity. In them, we identified dozens of motifs with some degree of similarity with proteins from different kingdoms, which may help explain some of their still poorly understood means of action. Through in silico and in vitro approaches, we compared their expression levels after controlled exposure to several AbSts' sources, applied at diverse tissues, on varied phenological stages. Based on gene ontology concepts, we still analyzed them from different perspectives of term enrichment. We have also searched, in model plants and close species, for potentially orthologous genes. With all these new insights, which culminated in a plentiful supplementary material, rich in tables, we aim to constitute a fertile consultation source for those maize researchers attracted by these interesting stress proteins.
