Abstract

IntroductionIt is proved that there are differences between gait performed by females and males, which appear in movements of selected body parts. Despite numerous state-of-the-art studies related to the discriminative analysis of motion capture data, the question of whether measures of signal complexity and uncertainty can extract valuable features for the problem of sex distinction still remains open. It is the subject of the paper.MethodsCorrelation dimension, as well as approximate and sample entropies, are selected to describe motion data. In the numerical experiments, the collected dataset with 884 samples of 25 females and 30 males was used. The measurements took place in the Human Motion Laboratory (HML), equipped with a highly precise motion capture system. Two variants of data representation were investigated-time series that contain joint rotations of taken skeleton model as well as positions of the markers attached to the human body. Finally, a comparative analysis between the populations of females and males using descriptive statistics, non-parametric estimation, and statistical hypotheses verification was carried out.ResultsThere are statistically significant sex differences extracted by the taken measures. In general, the movements of lower limbs result in greater values of correlation dimension and entropies for females, while selected upper body parts play a similar role for males. The dissimilarities are mainly observed in hip, ankle, shoulder, and head movements.DiscussionCorrelation dimension and entropy measures provide robust and explainable features of motion capture data with a valuable description of the human locomotion system. Thus, beyond the importance of discovered differences between females and males, their interpretation and understanding are also known.

Abstract

Abstract The ability to uncover characteristics based on empirical measurement is an important step in understanding the underlying system that gives rise to an observed time series. This is especially important for biological signals whose characteristic contributes to the underlying dynamics of the physiological processes. Therefore, by studying such signals, the physiological systems that generate them can be better understood. The datasets presented consist of 33,000 time series of 15 dynamical systems (five chaotic and ten non-chaotic) of the first, second, or third order. Here, the order of a dynamical system means its dimension. The non-chaotic systems were divided into the following classes: periodic, quasi-periodic, and non-periodic. The aim is to propose datasets for machine learning methods, in particular deep learning techniques, to analyze unknown dynamical system characteristics based on obtained time series. In technical validation, three classifications experiments were conducted using two types of neural networks with long short-term memory modules and convolutional layers.

Abstract

Abstract Background The majority of genes in the human genome is present in two copies but the expression levels of both alleles is not equal. Allelic imbalance is an aspect of gene expression relevant not only in the context of genetic variation, but also to understand the pathophysiology of genes implicated in genetic disorders, in particular, dominant genetic diseases where patients possess one normal and one mutant allele. Polyglutamine (polyQ) diseases are caused by the expansion of CAG trinucleotide tracts within specific genes. Spinocerebellar ataxia type 3 (SCA3) and Huntington’s disease (HD) patients harbor one normal and one mutant allele that differ in the length of CAG tracts. However, assessing the expression level of individual alleles is challenging due to the presence of abundant CAG repeats in the human transcriptome, which make difficult the design of allele-specific methods, as well as of therapeutic strategies to selectively engage CAG sequences in mutant transcripts. Results To precisely quantify expression in an allele-specific manner, we used SNP variants that are linked to either normal or CAG expanded alleles of the ataxin-3 (ATXN3) and huntingtin (HTT) genes in selected patient-derived cell lines. We applied a SNP-based quantitative droplet digital PCR (ddPCR) protocol for precise determination of the levels of transcripts in cellular and mouse models. For HD, we showed that the process of cell differentiation can affect the ratio between endogenous alleles of HTT mRNA. Additionally, we reported changes in the absolute number of the ATXN3 and HTT transcripts per cell during neuronal differentiation. We also implemented our assay to reliably monitor, in an allele-specific manner, the silencing efficiency of mRNA-targeting therapeutic approaches for HD. Finally, using the humanized Hu128/21 HD mouse model, we showed that the ratio of normal and mutant HTT transgene expression in brain slightly changes with the age of mice. Conclusions Using allele-specific ddPCR assays, we observed differences in allele expression levels in the context of SCA3 and HD. Our allele-selective approach is a reliable and quantitative method to analyze low abundant transcripts and is performed with high accuracy and reproducibility. Therefore, the use of this approach can significantly improve understanding of allele-related mechanisms, e.g., related with mRNA processing that may be affected in polyQ diseases.

Abstract

This work is focused on the preliminary stage of the 3D drone tracking challenge, namely the precise detection of drones on images obtained from a synchronized multi-camera system. The YOLOv5 deep network with different input resolutions is trained and tested on the basis of real, multimodal data containing synchronized video sequences and precise motion capture data as a ground truth reference. The bounding boxes are determined based on the 3D position and orientation of an asymmetric cross attached to the top of the tracked object with known translation to the object’s center. The arms of the cross are identified by the markers registered by motion capture acquisition. Besides the classical mean average precision (mAP), a measure more adequate in the evaluation of detection performance in 3D tracking is proposed, namely the average distance between the centroids of matched references and detected drones, including false positive and false negative ratios. Moreover, the videos generated in the AirSim simulation platform were taken into account in both the training and testing stages.

Abstract

During the ovarian cycle in domestic dogs, females do not accept males during the first days of estrus but become attractive to males from the beginning of proestrus, with this attractiveness persisting until the end of the estrus phase. It is believed that increased estradiol is responsible for the female attractiveness to the males. In this paper we describe the case of strong, but atypical attractiveness of a castrated male to various, adult, intact males, influenced by the emitted semiochemical signals. Any significant changes in the level of hormones typically involved in the process connected with estrus and responsible for sexual arousal in the males were assessed. The case animal was a 4 year old castrated male Border Collie that was extremely attractive to various males, which presented high levels of sexual arousal, with intensive sniffing and licking of the preputial area, specific vocalization, increased salivation and, finally, mating attempts. Clinical examination of the castrated male revealed a lack of testes in the scrotum and abdominal cavity confirmed by USG.
