Background: Hexaploid oat (Avena sativa L.) is a commercially important cereal crop due to its soluble dietary fiber β-glucan, a hemicellulose known to prevent cardio-vascular diseases. To maximize health benefits associated with the consumption of oat-based food products, breeding efforts have aimed at increasing the β-glucan content in oat groats. However, progress has been limited. To accelerate oat breeding efforts, we leveraged existing breeding datasets (1,230 breeding lines from South Dakota State University oat breeding program grown in multiple environments between 2015 and 2022) to conduct a genome-wide association study (GWAS) to increase our understanding of the genetic control of beta-glucan content in oats and to compare strategies to implement genomic selection (GS) to increase genetic gain for β-glucan content in oat. Results: Large variation for β-glucan content was observed with values ranging between 3.02 and 7.24%. An independent GWAS was performed for each breeding panel in each environment and identified 22 loci distributed over fourteen oat chromosomes significantly associated with β-glucan content. Comparison based on physical position showed that 12 out of 22 loci coincided with previously identified β-glucan QTLs, and three loci are in the vicinity of cellulose synthesis genes, Cellulose synthase-like (Csl). To perform a GWAS analysis across all breeding datasets, the β-glucan content of each breeding line was predicted for each of the 26 environments. The overall GWAS identified 73 loci, of which 15 coincided with loci identified for individual environments and 37 coincided with previously reported β-glucan QTLs not identified when performing the GWAS in single years. In addition, 21 novel loci were identified that were not reported in the previous studies. The proposed approach increased our ability to detect significantly associated markers. The comparison of multiple GS scenarios indicated that using a specific set of markers as a fixed effect in GS models did not increase the prediction accuracy. However, the use of multi-environment data in the training population resulted in an increase in prediction accuracy (0.61–0.72) as compared to single-year (0.28–0.48) data. The use of USDA-SoyWheOatBar-3 K genotyping array data resulted in a similar level of prediction accuracy as did genotyping-by-sequencing data. Conclusion: This study identified and confirmed the location of multiple loci associated with β-glucan content. The proposed genomic strategies significantly increase both our ability to detect significant markers in GWAS and the accuracy of genomic predictions. The β-glucan in oat grain plays an important role in preventing cardiovascular disease. Therefore, it is of great significance to realize the rapid measurement of β-glucan content in oat grain for the rational use of β-glucan. In this study, hyperspectral monitoring models of oat grain β-glucan content were constructed, and the effects of transformation algorithm, sensitive band screening algorithm, and modeling algorithm on the accuracy of the model were studied. The results showed that logarithm(Lg) and standardized normal variables(SNV) transformation can change the value of original spectral reflectance, while first-order differentiation(FD) and second-order differentiation(SD) transformation can also change the changing trend of original spectral reflectance. multivariate scattering correction(MSC), SNV, FD, and SD transformation can improve the correlation between spectral reflectance and oat grain β-glucan content to varying degrees. When using full band spectrum to construct the model, the performance of linear modeling algorithm was better than that of nonlinear modeling algorithm. Compared with uninformative variable elimination(UVE) algorithm, successive projections algorithm(SPA) had better band screening effect. Combining all the modeling results, it was found that MSC and SNV transformation algorithms had good results. Among them, the model based on SNV transformation spectrum and SPA-multiple linear regression(SPA-MLR) reached the highest accuracy, which can realize the hyperspectral monitoring of oat grain β-glucan content. Introduction: It is recognized that the character and the quality of the malt is obtained during kilning. Moreover, the changes occurring during kilning affect mashing and wort quality. Objectives: Till now, in Greece, for the malt production other barley varieties rather than native ones are commonly used. Thus, this research aims to evaluate the effect of kilning temperature in improving malting ability of a Greek barley cultivar. Methods: Barley kernels were malted in the Food Process Engineering Laboratory - Pilot Plant of ATEITh (Thessaloniki). The malted barley was first dried at 40-45°C (dry malt) and then kilned. The kilning of germinated barley was performed at three different temperatures (80, 90 and 100°C for 6h), in order to produce three different malts. Barley, dry malt and malted kernels were analysed for their moisture, ash, protein and β-glucan content whereas their respective worts, for extract content of the malt, colour, specific gravity and specific viscosity. The total fermentable sugars as well as their profile were also determined. Finally, the malts were compared with a commercial malt. Results: Factors such as malt moisture, β-glucan content, malt colour, malt extract and specific viscosity were significantly affected by the kilning process. Contrary, the ash and protein contents in the malt were not significantly affected. The malt produced from the Greek barley cultivar showed β-glucan, specific gravity and specific viscosity values similar to the commercial sample. Conclusions: The present study revealed that the malt and wort quality of the Greek barley cultivar (Seirios) can be significantly improved by optimizing the kilning temperature.

The Cellulose synthase-like F6 (CslF6) gene has been reported as a key factor for β-glucan synthesis in grasses. However, the action of this gene on oat β-glucan content is poorly understood. The objectives of this study were: (i) to quantify the absolute expression of CslF6 transcripts in oats under field conditions; and (ii) to characterize the CslF6 homoeologs in oat genotypes with different levels of β-glucan content. Developing kernels of five oat genotypes were collected under field conditions in two planting dates. The CslF6 absolute quantification was performed using droplet digital PCR considering global and homoeolog-specific expressions. The third exon of the CslF6 gene, which covers approximately 62.2% of the coding region, was sequenced in each homoeolog. The CslF6 expression was highly influenced by planting date, kernel developmental stage, and genotype. High CslF6 transcript number was not associated with high β-glucan content, indicating the action of a complex biosynthesis pathway controlling this trait in oats. Under stressful conditions, the CslF6 expression was more pronounced and associated with β-glucan content. The CslF6 motif has 1767 bases pair and is highly conserved between A and D homoeologs. Single nucleotide polymorphisms were not identified among the assessed genotypes, regardless of the homoeolog. High β-glucan content is one of the main goals of oat breeding programs worldwide. However, the genomic regions and genes controlling β-glucan content in oats are not fully understood. In this sense, the objectives of this study were as follows: (i) to characterize structure and linkage disequilibrium (LD) in a panel of oat germplasm adapted to subtropical environments; and (ii) to identify genomic regions associated with oat β-glucan content. An oat panel with 413 genotypes was evaluated for β-glucan content under subtropical conditions in different years and genotyped using genotyping-by-sequencing. Population structure, LD, and genome-wide association (GWA) analyses were carried out. GWA mapping was performed for each year separately and in a multi-environment model. The UFRGS Oat Panel showed weak population structure and has great potential to elucidate many agronomic traits in subtropical environments. Seven quantitative trait loci (QTL) associated with β-glucan content were identified. These QTL are located on Mrg02, Mrg06, Mrg11, Mrg12, Mrg19, and Mrg20. The QTL located on Mrg02, Mrg06, and Mrg11 seem to be genomic regions syntenic with barley. The use of these QTL may be useful to accelerate the genetic progress of oat β-glucan content in subtropical environments. [ABSTRACT FROM AUTHOR]

The objective of this study was to propose a rapid and nondestructive method for quantitatively detecting the hardness of black highland barley kernels using hyperspectral imaging. Initially, a regression model was established to predict hardness based on β-glucan content. Spectral reflectance within the 400–1000 nm wavelength range was gathered for black highland barley, and six preprocessing techniques were applied. Once preprocessing was completed, three characteristic wavelength screening methods were employed. Finally, three different models were utilized to construct a dependable prediction model for β-glucan content. The results indicated that the one-dimensional convolutional neural network (1D-CNN), in combination with the moving average (MA) preprocessing method, exhibited the best performance. To validate the hardness prediction model, the β-glucan content prediction model was integrated with the hardness regression model. The hardness prediction model attained a coefficient of determination (R2) value of 0.8093 and root mean square error (RMSE) of 0.2643 kg. The visual images exhibit characteristics feature of hardness in different varieties of black highland barley. Recent studies have revealed an interest in the composition of beer biomolecules as a colloidal system and their influence on the formation of beer taste. The purpose of this research was to establish biochemical interactions between the biomolecules of plant-based raw materials of beer in order to understand the overall structure of beer as a complex system of bound biomolecules. Generally accepted methods of analytical research in the field of brewing, biochemistry and proteomics were used to solve the research objectives. The studies allowed us to establish the relationship between the grain and plant-based raw materials used, as well as the processing technologies and biomolecular profiles of beer. The qualitative profile of the distribution of protein compounds as a framework for the formation of a colloidal system and the role of carbohydrate dextrins and phenol compounds are given. This article provides information about the presence of biogenic compounds in the structure of beer that positively affect the functioning of the body. A critical assessment of the influence of some parameters on the completeness of beer taste by biomolecules is given. Conclusion: the conducted analytical studies allowed us to confirm the hypothesis about the nitrogen structure of beer and the relationship of other biomolecules with protein substances, and to identify the main factors affecting the distribution of biomolecules by fractions.

In this study, the β-glucan of milled rice, rice husk, and rice bran fractions, as well as the amylose content of milled rice fraction, from 38 selected rice-paddy grains from six regions of Thailand were quantitatively determined. The milled rice of the Sakon Nakhon (SN) cultivar grown in the northeast contained the highest β-glucan content (0.88 ± 0.03%), followed by the milled rice of the Jow Khao Chiangmai (JKC) cultivar (0.71 ± 0.03%) and rice bran of the Sew Mae Jan (SMJ) cultivar (0.67 ± 0.03%) grown in the north. The results reveal that the rice cultivars from each region showing variation in the β-glucan level in each fraction, which is mainly found in milled rice and rice bran, are similar to those found in other cereal grains, although low amounts are found in the husk. The amylose and β-glucan contents in the milled rice fraction showed a strong negative correlation (r = −0.805; p < 0.0001). This new information about the β-glucan content of Thai rice cultivars could be used for the development of cereal-based functional food.

Grain weight and protein content will be reduced and increased, respectively, when barley is subjected to water stress after anthesis, consequently deteriorating the malt quality. However, such adverse impact of water stress differs greatly among barley genotypes. In this study, two Tibetan wild barley accessions and two cultivated varieties differing in water stress tolerance were used to investigate the genotypic difference in metabolic profiles during grain-filling stage under drought condition. Totally, 71 differently accumulated metabolites were identified, including organic acids, amino acids/amines, and sugars/sugar alcohols. Their relative contents were significantly affected by water stress for all genotypes and differed distinctly between the wild and cultivated barleys. The principal component analysis of metabolites indicated that the Tibetan wild barley XZ147 possessed a unique response to water stress. When subjected to water stress, the wild barley XZ147 showed the most increase of β-amylase activity among the four genotypes, as a result of its higher lysine content, less indole-3-acetic acid (IAA) biosynthesis, more stable H2O2 homeostasis, and more up-regulation of BMY1 gene. On the other hand, XZ147 had the most reduction of β-glucan content under water stress than the other genotypes, which could be explained by the faster grain filling process and the less expression of β-glucan synthase gene GSL7. All these results indicated a great potential for XZ147 in barley breeding for improving water stress tolerance.

Maitake ( Grifola frondosa) was hydrothermally extracted at 4 different temperatures (121, 130, 140, and 150°C) for 30 and 60 min, and the total phenolic content (TPC), antioxidant activity, and β-glucan content of the extracts was evaluated. The highest TPC was detected in the extract at 150°C treated for 60 min with 13.61 mg gallic acid equivalents (GAE)/g. The antioxidant activity of the maitake extracts, evaluated by determining DPPH radical scavenging activity (RSA), ABTS RSA, reducing power, and tyrosinase inhibitory activity increased with increasing treatment temperature and time. The content of β-glucan was the highest (5.13%) in the extract prepared at 140°C for 30min. These results suggest that hydrothermal extraction could be used as a tool to increase the antioxidant activity of maitake extracts. [ABSTRACT FROM AUTHOR]

β–glucan is a non–starchy, water–soluble polysaccharide component of wheat grain that positively affects grain nutrient quality. High β–glucan content in wheat grain also has a cholesterol–lowering effect in humans. The aim of this study was to valuate the effect of nitrogen and irrigation on durum wheat grain β–glucan content. Correlations between durum eat grain β–glucan content, and grain yield and some quality characteristics were examined. Kunduru 1149, K.z.ltan 91, and Çakmak 79 durum wheat cultivars were used as material, and 0–, 75–, and 150–mm irrigation levels with 0, 40, 80, and 120 kg ha-1 nitrogen rates were applied. Higher nitrogen levels generally increased durum wheat grain β–glucan content in both years of the study and the N3 nitrogen level had the greatest effect on the β–glucan content in durum wheat. Higher irrigation levels negatively affected the β–glucan content in the wheat grain. Non–irrigated treatments generally yielded the highest β–glucan content in both years of the study. The response of grain β–glucan content according to cultivar was significant and C1 had the best grain #x03B2;–glucan content of all the cultivars. Correlations between grain β–glucan content, and grain yield, protein content, 1000–grain weight, test weight, seed vitreousness, and the SDS sedimentation were significant. With the exception of protein content in 2005, strong positive correlations were observed between β–glucan content, and grain yield and quality characteristics. These results indicate that higher irrigation levels negatively affected grain &#x03B2 –glucan content in durum wheat, whereas higher nitrogen rates positively affected grain β–glucan content. Two hull-less barley varieties were roller-milled, and breaks (B) and reduction flours (C), shorts, and bran were collected. Shorts, which mainly originate from endosperm cells with a smaller amount of the outer layers, had the largest yield (48.87–51.54%). Ash (0.82–3.10%) and protein (9.95–14.8%) increased from flours toward shorts and bran, while starch decreased (82.31–48.69%). In contrast to clear distribution differences in protein content (bran > shorts > C > B), albumins/globulins content was lowest in bran (0.78–0.90 g/100 gdw), and their distribution between fractions was uneven and genotype dependent. Distribution of hordeins (6.69–10.49 g/100 gdw) was more distinct and generally decreased in order from bran > B > shorts > C. The proportion of nutritionally poor C-hordeins in total hordeins varied from 28.33% to 30.24%, without significant differences between fractions. The β-glucan content varied from 0.80% to 7.49% with decreasing content in the order bran, shorts > C > B. Shorts and bran could be classified as moderate and high β-glucan flour (5.70–7.22%). The total phenolic and antioxidant activities ranged from 0.91 to 2.21 mg GAE/gdw and 28.81–72.06%, respectively. Ferulic and sinapic acids determined by high-performance liquid chromatography (HPLC) were major contributors to the antioxidant activity (45.16–1026.91 ug/gdw and 18.93–206.52 ug/gdw), respectively.
