[Objective]DNA barcoding technology was one of the latest advances in molecular identification. In order to establish the DNA barcoding technology of Chinese orchid, the rapid identification of the DNA barcoding of Chinese orchid was realized. [Method]The feasibility of PCR amplification methods for several common DNA barcoding standard sequences such as ITS, ITS2, psbA-trnH, rbcL and matK in Chinese orchid was verified by electrophoresis imaging and BLASTN nucleotide similarity sequence analysis. [Result]The method could be effectively used to amplify the barcode sequences such as ITS2, psbA-trnH, rbcL and matK in Chinese orchid and obtained the sequence information through bidirectional sequencing. The ITS standard sequence required the redesign of suitable primers due to the non-specific amplification of the primers. [Conclusion]Using this method, common DNA barcoding standard sequences such as ITS2, psbA-tmH, rbcL, and matK can be obtained and simplify the PCR reaction procedure, which provides a reference for further study of Chinese orchid DNA barcoding technology.
