[Objective] Calcium-dependent protein kinases (CDPK or CPK) are widely involved in plant growth and development and environmental stress response. To explore their roles involved in Haematococcus pluvialis growth, development and astaxanthin accumulation, HpCDPK gene family was identified and analyzed. [Method] Bioinformatics tools were used to identify the HpCDPK gene family members, and to analyze the physical and chemical properties, phylogenetic evolution, conserved motifs and functional domains of the encoded proteins, as well as the gene expression profiles under nitrogen deficiency stress. [Result] The results showed that seven members of HpCDPK gene family were identified, and all HpCDPK proteins contained typical EF-hand motifs and protein kinase domains. Phylogenetic analysis showed that these HpCDPKs clustered with the CrCDPKs from Chlamydomonas reinhartii and separated from the AtCDPKs of Arabidopsis thaliana, suggesting that species-specific gene replication occurred in the process of evolution. Transcriptome data analysis showed that HpCDPK gene expression was induced by a variety of stresses, among which the transcription level of HpCDPK7 gene was most significantly up-regulated under nitrogen deficiency. In addition, the analysis of the correlation between HpCDPK and the expression of genes related to carotenoid synthesis showed that HpCDPK was closely related to the expressions of genes relevant to several carotenoid synthesis in H. pluvialis, especially HpCDPK2 was positively correlated with the expression of key genes of astaxanthin synthesis (BKT and BCH). [Conclusion] Seven HpCDPK genes were identified, the expression of HpCDPK7 gene was most significantly up-regulated under nitrogen deficiency and HpCDPK2 may play an active role in the synthesis and accumulation of carotenoids and astaxanthin. The results provide a reference basis for further analysis of the function and molecular mechanism of stress response and carotenoids synthesis and accumulation mediated by HpCDPK in H. pluvialis. Objective: To screen the interacting protein of ubiquitin-conjugating enzyme E2S (UBE2S) and construct the hepatocellular carcinoma (HCC) based on UBE2S interacting protein prognosis model (UIPM), and to discuss the value of UIPM in assessing the prognosis of the HCC patients. Methods: Coimmunoprecipitation (Co-IP) was used to screen the protein complexes binding to Flag-UBE2S. After validation by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blotting methods；liquid chromatography-mass spectrometer (LC-MS) was used to identify the UBE2S interacting proteins； Gene Ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) signaling pathway enrichment analysis were conducted on these proteins； the prognosis-related proteins from The Cancer Genome Atlas (TCGA) were cross-referenced with UBE2S interacting proteins by survival package of R software；the key proteins were extracted through LASSO regression analysis to build the UIPM； the prognostic model risk scoring formula was established. The HCC patients in TCGA were divided into high risk group and low risk group based on median value of the risk scores. The predictive accuracy of UIPM was evaluated by receiver operating characteristic curve (ROC), and the predictive accuracy was further validated by International Cancer Genome Consortium (ICGC) Database； univariate regression analysis and multivariate Cox regression analysis were used to detect whether the UIPM risk score was an independent prognostic factor for HCC. Furthermore, the nomogram model was built. Results: A total of 97 UBE2S interacting proteins were identified through Co-IP combined with LC-MS analysis. The GO functional enrichment analysis and KEGG signaling pathway enrichment analysis results showed that the interacting proteins were closely associated with cysteine-type endopeptidase activity, oxidative stress, and cell death. The TCGA revealed 5 163 HCC prognosis-related proteins；after intersecting with UBE2S interacting proteins, 40 prognosis-related interacting proteins were found. Seven key proteins were determined through LASSO regression analysis, including UBE2S, heat shock protein family A member 8 (HSPA8), heterogeneous nuclear ribonucleoprotein H1 (HNRNPH1), chaperonin containing TCP1 subunit 3 (CCT3), eukaryotic translation initiation factor 2 subunit 1 (EIF2S1), receptor for activated C kinase 1 (RACK1), and actin related protein 2/3 complex subunit 4 (ARPC4), and the UIPM was constructed. There was significant difference in survival rate of the patients between high risk group and low risk group (P<0. 05) . The ROC curve analysis results showed the area under ROC curve (AUC) values of UIPM for predicting 1-year, 2-year, and 3-year survival risk scores of the HCC patients were all greater than 0. 7, indicating the model had high predictive accuracy. This was also confirmed by ICGC Database data. The univariate and multivariate Cox regression analysis results showed that the UIPM risk score was an independent prognostic risk factor for the HCC patients (P<0. 05) . The nomogram results showed good consistency between predicted survival rate and actual survival rate of the patient. Conclusion : A total of 97 interacting proteins that interact with UBE2S may promote the occurence and development of HCC through oxidative stress and dysregulation of ferroptosis pathways. The UIPM risk score is an independent risk factor for the prognosis of HCC and can be used to predict the outcomes of the patients. Current studies have not clarified the relationship between sedentary behavior(SB) and muscle strength(MS), especially the lack of relevant evidence for preschool children in China. A cross-sectional study research method was used to explore the role of SB on MS in preschool children aged 3 ~ 6 years old. Taking children aged 3 ~ 6 years old in Beijing (n = 275) as the study subjects, the muscle strength of upper limbs and lower limbs were evaluated by handgrip strength and a standing broad jump. According to the evaluation results T-score of MS were calculated. Total T score of MS were calculated according to the evaluation results. SB were measured by the ActiGraph GT9X Link. Participants need to wear ActiGraph GT9X Link for continuous 7 days to assess the total, sustained SB time of preschool children􀆳s SB. Results show total time of SB and SB Bouts time negatively correlated with MS after adjusting for confounding factors such as sex, age, household income, father􀆳s and mother􀆳s education. For each 10 min / d increase in the total time of SB, the total T-score of MS decreased by 0. 738(95% CI: - 1. 118, - 0. 357) . For each 10 min / d increase in sustained SB time, the total T-score of MS decreased by 0. 822(95% CI: - 1. 632, - 0. 012). Furthermore, both upper limb MS and lower limb MS were significantly and negatively correlated with the total time of SB. A 10 min / d increase in the total time of SB was associated with a 0. 427(95% CI: - 0. 664, - 0. 189) decrease in the T-score of upper limb MS and a 0. 312(95% CI: - 0. 533, - 0. 091) decrease in the T-score of lower limb MS. The result supports that total time of SB and SB Bouts time negatively correlated with MS. SB is a possible risk factor for MS. Objective To investigate the expression of cytoskeleton⁃associated protein 4 (CKAP4) in clear cell renal cell carcinoma (ccRCC) and its clinical significance. Methods A total of 237 tumor tissues and paracancerous tissues were collected from ccRCC patients. The expression level of CKAP4 protein was detected by immunohistochemistry. The clinicopathological characteristics of ccRCC patients with different CKAP4 protein expression levels were compared by χ2 test. The Kaplan⁃Meier method and Log⁃rank test were used to analyze the differences in the overall survival (OS) and progression⁃free survival (PFS) of patients with different CKAP4 protein expression levels. Cox proportional hazard regression was performed to analyze the influencing factors of OS and PFS in the patients with ccRCC. Results The high expression rate of CKAP4 protein in ccRCC tissues was significantly higher than that in paracancerous tissues (54.0% vs 23.6%, χ² =46.050, P<0.001). CKAP4 protein was highly expressed in the cancer tissues of ccRCC patients with higher Fuhrman grade, deeper tumor invasion, lymph node metastasis, distant metastasis and higher TNM stage (all P<0.05). The ccRCC patients with high expression of CKAP4 protein had significantly lower OS rate (χ² =6.394, P=0.011) and PFS rate (χ² =10.065, P=0.002) than those with low CKAP4 protein expression. High expression of CKAP4 protein was an independent risk factor for postoperative OS (HR= 2.884, 95%CI:1.425-4.765, P<0.001) and PFS (HR=3.512, 95%CI: 1.987-5.326, P<0.001) in the patients with ccRCC. Conclusions CKAP4 protein is highly expressed in ccRCC tissues, and is an independent risk factor for poor prognosis in the patients with ccRCC. [ABSTRACT FROM AUTHOR]

This paper presents a set of test methods for evaluating the performance of active knee-assisted exoskeleton. A motor-driven assist model is established to calculate the theoretical assist efficiency of exoskeleton knee joints. Six healthy examinees are selected to measure comprehensive metabolic indexes of the human body and lower limb muscle activity under the condition of wearing and not wearing exoskeleton. The results of the comprehensive metabolic cost test of the human body showthat, compared with the experiment without wearing the exoskeleton, examinees wearing the exoskeleton see an increase of oxygen consumption of the human body by 3.21% on average, an increase of exhalation of carbon dioxide by 1.27% on average, and an increase of net metabolic power per unit mass by 2.80% on average. The results of the lower limb muscle activity test show that, compared with the experiment without exoskeleton, wearing exoskeleton can effectively reduce the muscle activity of rectus femoris, vastus lateralis and vastus medialis. A controlled weight-bearing test is designed, and the results prove that the weight of the exoskeleton itself cannot be ignored, which increases the comprehensive metabolic cost of the examinees. The exoskeleton can effectively reduce the activity of the wearer’s lower limb muscles, which is basically in line with the theoretical assisting efficiency of exoskeleton knee joints, and the overall power-assisting efficiency is not obvious due to the weight of the wearer. The phenotypic differences of Chlamydomonas reinhardtii wild strain (CC5325) and CRY mutant strain (crcry) under normal CK (Control) and Blue BL (Blue light) cultures were investigated. The PCR verification showed that the crcry mutant was inserted with paromomycin resistance gene AphVIII expression box in the coding region. AphVIII resistance gene was inserted into crcry mutant and successfully expressed in plate and liquid culture system with paromomycin as screening condition. Phenotypic identification showed that there were no significant differences in growth, pigment, photosynthesis and lipid synthesis between wild strain CC5325 and mutant strain crcry under CK culture condition. However, under BL condition, the growth of mutant crcry was significantly inhibited, and the color of algae liquid turned yellow. Chlorophyll a, chlorophyll b and total pigment contents of unit cells decreased significantly, while total carotenoid contents increased significantly. The photosynthetic system was severely suppressed and the total lipid content significantly reduced. In conclusion, plant cry is involved in the blue light response of C. reinhardtii.
