目的观察奥沙利铂、氟尿嘧啶筛选前后胃癌SGC7901细胞株中CD44v6/ATP结合转运蛋白G超家族成员2(ABCG2)表达变化。方法取对数生长期的胃癌SGC7901细胞株,分别用0、4、12、20、28、36μg/m L的奥沙利铂及0、30、50、70、90、110μg/m L的氟尿嘧啶进行干预,MTT法得到奥沙利铂、氟尿嘧啶作用于实验细胞株的最适浓度分别为30、28μg/m L。以最适浓度作用于实验细胞株分离出胃癌干细胞,以免疫细胞化学法、半定量RTPCR法、Western blotting法检测以上两种药物筛选前后细胞株中CD44v6/ABCG2的阳性表达率。再以单细胞悬浮成瘤实验、免疫细胞化学法检测次代干细胞级别的实验细胞株中CD44v6/ABCG2的阳性表达率。结果奥沙利铂、氟尿嘧啶筛选的干细胞级别的实验细胞株中CD44v6/ABCG2阳性表达率明显上调,能在低浓度血清培养基中形成悬浮肿瘤球,免疫细胞化学检测其次代细胞中CD44v6/ABCG2的阳性表达率又回到筛选前水平。结论奥沙利铂、氟脲嘧啶筛选后的细胞株中CD44v6/ABCG2阳性表达率明显上调,细胞株具有干细胞的特性。 [ABSTRACT FROM AUTHOR]
