In order to further promote the research and application of BIM technology in caisson gravity wharf project, this paper takes the No. 9 and No. 10 berths project of Dalanping South Operation Area, Dalanping Port Area, Qinzhou Port as an example, applies BIM technology to the construction management of traditional hydraulic wharf, introduces BIM technology in the construction process, and establishes a three-dimensional information model. On this basis, the comprehensive application of BIM technology was carried out for the project, and the effective combination of BIM technology and site construction was realized. With the advantages of visualization and informationization of BIM technology, engineering quantity statistics, construction progress simulation, design optimization, project results display and other work can be realized. BIM technology can empower traditional hydraulic projects, improve management level, enhance performance ability, and obtain good economic and social benefits, which provides a reference for similar projects and has certain promotion and reference significance. Taking the back-silting mud near the Shenzhen-Zhongshan Tunnel immersed tunnel as the research object. Based on the laboratory model test, the rheological characteristics of the back-silting mud were determined, the relationship between shearing force and shearing velocity was determined, and a constitutive model to characterize its rheological characteristics was established. The reliability of the constitutive model was verified by the combination of laboratory small-scale model test and the CEL finite element method. Results show that the back-silting mud in the Shenzhen-Zhongshan Tunnel all satisfy the Herschel-Buckley rheology model; the back-silting mud with gravity density of 12.4 kN/m³ can be sinked to the bottom of the foundation trench smoothly within 1 s under the load of 0.5 kPa; using the Herschel-Buckley rheology model and CEL model can effectively simulate the squeezing and removing characteristics of the back-silting mud under pressure. 14. RNA 干扰沉默高迁移率族蛋白B1 的表达 对结直肠癌 LoVo 细胞增殖和侵袭的抑制 作用

- Abstract
Objective To inquire into the influence of silencing HMGB1 expression by small interfering RNA (siRNA) on cell growth,proliferation, invasion and metastasis of colorectal cancer LoVo cells both in vitro and in vivo. Methods Lentivirus-mediated HMGB1 siRNA was transfected into LoVo cells to silence the HMGB1 expression. The HMGB1 mRNA and protein expression after siRNA transfection was detected by RT-PCR and Western blot. MTT assay was used to observe the cell proliferation and to draw a growth curve. Cell cycle was measured by flow,cytometry. The ability of invasion and speed of cell migration were evaluated by transwell chamber invasion and cell scratch assay. The influence of HMGB1 silencing on the proliferation of LoVo cells in vivo was observed in LoVo tumor-bearing nude mice. Results Lentivirus- mediated siRNA was successfully transfected into colorectal cancer cell line LoVo. The expression of HMGB1 mRNA and protein in the HMGBl-siRNA group wee 0.24±0.04 and 0.21 ±0.03,respectively. Compared with the HMGB1-siRNA-Neg group (0.82±0.13,1.15±0.18) and control group (0.93±0.15, 1.21 ±0.20), the difference was significant ( P<0.05). MTT assay show-ed that the cell proliferation in the HMGBl-siRNA group was significantly inhibited when compared with that in the HMGB1-siRNA-Neg group and control group (P<0.05). Flow,cytometry showed that the proliferation index (PI) of HMGB1-siRNA group w,as 38.27± 1.32,significantly lower than 54.66± 1.74 in the HMGB1-siRNA-Neg group and 57.43± 1.29 in the control group (P <0.05). The transwell assay showed that the number of penetrated cells in the HMGB1-siRNA group w,as 14.0±3.5,significantly low,er than 51.0±6.7 in the HMGB1-siRNA-Neg group and 68.0±5.3 in the control group (P<0.05). Similarly, the scrape wound recovered significantly slower in the HMGB1- siRNA group (83.61±23.21) pm than that in the other tww groups (202.86±46.46) pm and (214.58±57.38) pm (P<0.05). The nude mouse xenograft tumor experiment show,ed that the final tumor volume w,as (521 ±34) mm3 in the HMGB1-siRNA group,significantly smaller than that in the HMGB1-siRNA-Neg group of (763±46) mm3 and control group of ( 802 ± 51) mm3 ( P < 0.05 ). Conclusions Lentivirus-mediated HMGBl- siRNA can effectively inhibit the HMGB1 expression in colorectal cancer LoVo cells both in vitro and in vivo. HMGB1 gene silencing can slow the growth of colorectal cancer cells, extend the cell proliferation cycle, decrease their invasion and migration, and significantly inhibit the growth of xenograft tumor in nude mice. 15. 滚转机构在0.6 m 跨超声速风洞中的应用研究.

- Abstract
A rolling device is developed for the continuous change of model's rolling angle, the improvement of test efficiency and test range in the 0.6 m×0.6 m transonic and supersonic wind tunnel. The designs of rolling device, hollow built-in torque servomotor and the software and hardware of control system are introduced. The key technical problems are analyzed and summarized, and the conclusions are finally validated by a wind tunnel test. The test data shows that the rolling device and the central support of the normal force test, as model's supporting mechanisms, can be used to acquire better repeatable data. Meanwhile, the system has a high level control precision (rolling angle≤±3′), and the engineering reliability of the loads meets the wind tunnel test requirements. [ABSTRACT FROM AUTHOR]
