BACKGROUND: Construction of tissue engineering vascular structures is a key to the regeneration of complex tissues and organs. The use of three-dimensional printing technology to construct vascular structures has become a hotspot. OBJECTIVE: A highly connected and perfusable vascular network was rapidly deposited by a three-dimensional bioprinter with a coaxial nozzle using alginate/silk fibroin bioink. METHODS: A mixed solution containing 5% sodium alginate and 5% silk fibroin as the bio-ink, and a mixed solution containing 5% calcium chloride and 13% F127 as the crosslinker, alginate/silk fibroin gel was printed using a bio-printer and subjected to optical coherence tomography and scanning electron microscopy. The solution containing 5% sodium alginate and 5% silk fibroin mixed with the suspension of human liver cancer cells (C3A) as the bio-ink, and a mixed solution containing 5% calcium chloride and 13% F127 as the crosslinker, the cell-containing sodium alginate/silk fibroin gel was printed on a bio-printer. Then, the printed scaffold was placed in the medium for 24 hours, and then stained by Calcein-AM and observed under a fluorescence microscope. RESULTS AND CONCLUSION: (1) Optical coherence tomography: a multi-layer composite hollow tube network formed the structure of the scaffold. The gel filament had a complete hollow structure, and the channels were interconnected. The structure was similar to the hollow passage of the blood vessel, which was beneficial to the transportation of nutrients and metabolic waste. (2) Scanning electron microscope: the hollow pipe structure with the diameter of about 400 μm arranged in parallel, and the pipe boundaries were clear. The microporous structure caused by the removal of F127 was formed between the boundaries. (3) Fluorescence microscope: the cells were evenly dispersed in the material on both sides of the channel, and the cells grew well in the scaffold with the survival rate of above 95%. (4) These results suggest that three-dimensional bioprinting technology based on coaxial nozzles and sodium alginate/silk fibroin bio-ink can be used to construct vascularized functional tissues in the future. ObjectiveTo establish a microRNA-mRNA differential expression network for alcoholic hepatitis (AH), and to investigate new targets for the diagnosis and treatment of AH. MethodsDifferentially expressed microRNAs and mRNAs between AH patients and normal controls were screened out. Related software including TargetScan, DIANA, MIRDB, PICTAR, and miRWalk 2.0 was used to search for the target genes of differentially expressed microRNA, and a key microRNA-mRNA network was established using the differentially expressed mRNAs that changed in an opposite way to microRNA. The Database for Annotation, Visualization and Integrated Discovery was used for the gene ontology (GO) and Kyoto Encyclopedia of Genes and Genome (KEGG) analyses of target genes. The GCBI online software (www.gcbi.com.cn) was used for enrichment analysis of target genes and core network establishment. The GeneMANIA database in Cytoscape software (genemania.org) was used to perform a protein-protein interaction analysis of key target genes. The above three methods were compared in terms of the search for key pathways involved in the development of AH. ResultsA key microRNA-mRNA network was established with 5 differentially expressed microRNAs including hsa-mir-21-5p, hsa-mir-148a-3p, and hsa-mir-30e-5p and 51 target genes including collagen type IV alpha 1 chain (COL4A1), thrombospondin-2 (THBS2), and integrin alpha 6 (IGTA6). A protein-protein interaction network of key target genes was established. The GO analysis and various pathway analyses showed that the PI3K-Akt pathway and local adhesion were closely associated with AH. ConclusionDuring the development of AH, there are complex interactions between the related proteins of key target genes. COL4A1 and THBS2 may promote the development of AH by activating ITGA6 to regulate the PI3K-Akt pathway and the process of local adhesion. The establishment of the microRNA-mRNA network reveals the key links in the development of AH and highlights the focus of research. The discovery of the genes associated with the PI3K-Akt pathway in AH is expected to provide new targets for the diagnosis and treatment of AH. [ABSTRACT FROM AUTHOR]

Objective To investigate the effects of mutated isocitrate dehydrogenase 1 (IDHl) on TAZ (transcriptional coactivator with PDZ-binding motif protein) expression in human astrocytoma and to explore the relevant mechanisms. Methods Glioblastoma (GBM) U87MG cells transfected with mutated IDH1-132H and wild type IDH1-132R (WT) were used to detected the TAZ protein expression by Western blot; The expression of TAZ protein and its cytoplasmic binding protein 14-3-3e were investigated in a cohort of GBM cases, which including 14 IDH1 wild type cases and 7 IDH1 mutated cases. In vitro, qRT-PCR was used to detect TAZ mRNA expression; Western blot was used to detect the levels of LATS1, phosphorylated TAZ and 14-3-3e protein. Results We found that the expression of TAZ protein was decreased in IDH1 mutated cells compared with those of IDH1 wild type cells. This result was further verified in vivo by using immunohistochemistry to detect the expression of TAZ in human GBM samples with or without IDH1 mutation. Immunohistochemistry also showed that TAZ cytoplasmic binding protein 14-3-3e was increased; qRT-PCR showed that TAZ mRNA level was decreased in IDH1 mutated cells; and subsequent Western blot showed the levels of LATS1, TAZ phosphorylation and the 14-3-3e were increased in IDH1 mutant cells. Conclusions IDH1 mutation in gliomacould reduce TAZ mRNA and its protein expression, increase the phosphorylation level of TAZ protein by activating Hippo signaling pathway, thereby furtherinhibit TAZ expression.
