2012年6月～2013年4月,对上海动物园的9只圈养马来熊进行日粮表观消化率的研究,分春、夏、冬3个季节,每季采样10 d,5 d预实验,5 d正式实验,分析不同季节成年马来熊对日粮营养成分的表观消化率及夏季不同年龄段之间马来熊对日粮营养成分表观消化率的差异。结果表明,成年马来熊对日粮干物质、脂肪和蛋白质的表观消化率分别为71.41%、70.95%和 66.18%,且夏、冬两季及夏、春两季存在极显著差异(PP>0.05)。夏季不同年龄组之间,幼年组马来熊对干物质、脂肪、蛋白质的表观消化率最高,分别为(82±6)%、(85±6)%和(80±7)%,老年组最低,分别为(71±3)%、(78±4)%和(70±5)%; 对干物质的消化率,幼年组、成年组均与老年组之间存在显著差异(PP

2011年4～5月,对徐州金桥禽业发展有限公司养殖的赤麻鸭所产的44枚卵进行了人工孵化研究。其中43枚卵受精,出雏35枚,4枚在孵化中期死胚,5枚为27 d落盘后啄壳困难而死亡,受精卵的孵化率为81.40%。孵化温度前期为38.5℃; 中期为38.2℃; 后期为37.8℃,落盘后温度降为37.5℃。赤麻鸭卵鲜重为(79.80±6.72)g,长径为(66.10±2.53)mm,短径为(46.40±1.45)mm,卵形指数为1.43±0.06,出雏后的卵壳等剩余物质重(12.60±1.07)g,新生雏鸟重占鲜卵重的(67.20±3.71)%。卵总失重率为17%。孵化期间的卵重(y)与孵化天数(x)之间的线性方程为y=-0.9071x+81.019(R2=0.9997,P﹤0.01),根据卵失重方程可以比较准确地推算出赤麻鸭卵的孵化天数。

2011年46月,在徐州金桥珍禽有限公司,采用全日制瞬时扫描法对繁殖良好的一对赤麻鸭在繁殖期的行为日节律及时间分配进行了研究,并用独立样本t检验进行了不同性别在繁殖各期行为的差异性分析。结果表明:繁殖的不同时期赤麻鸭行为时间分配存在明显的性别差异。繁殖前期雌雄赤麻鸭之间的筑巢行为差异极显著（P0.05）,2只赤麻鸭几乎形影不离。赤麻鸭繁殖期行为呈现一定的日活动节律,采食行为集中在上午和下午的喂食时间7:008:00及16:0017:00这2个时间段内,孵化行为所占比例最低,此时雌性赤麻鸭出巢取食;一天中,休息行为在中午呈现一个高峰;警戒行为在清晨达到最高峰。

Objective To observe the expression changes of Notch1 signaling pathway-related proteins (transcription factor notch1, NICD, and its downstream protein HES1), and the apoptosis of neurons after inhibition of Notch1 signaling pathway in brain tissues of immature SD rats with acute hypoxic-ischemic brain injury, so as to investigate the role and mechanism of Notch1 signaling pathway in neonatal brain injury. Methods Eighteen three-day-old Sprague-Dawley rats were randomly divided into the Notch1 inhibition group, model group, and control group (6 rats), respectively. Rats in the Notch1 inhibition group were intraperitoneally injected with DAPT (100 mg/kg) 30 min before modeling, and then were treated with ischemia and hypoxia. Rats in the model group were intraperitoneally injected with 1% DMSO before hypoxia and ischemia treatment. Rats in the control group received sham surgery. Notch1 signaling pathway proteins (notch1, NICD, and HES1) and apoptotic proteins (Caspas-3, Bax, and Bcl-2) were detected by Western blotting. The changes of brain structure and apoptosis were observed by Niss staining and TUNEL staining. Results Compared with the control group, the expression levels of transcription factor NICD of Notch1 signaling pathway and its downstream regulatory protein HES1 increased (both P<0. 05), the expression level of pro-apoptotic protein Caspas-3 increased (P<0. 05), and the expression level of anti-apoptotic protein Bcl-2 decreased in the model group (P<0. 05) . Compared with the model group, the expression levels of NICD and HES1 protein decreased (both P<0. 05), the expression level of Caspas-3 protein decreased (P<0. 05), and the expression level of Bcl-2 increased in the Notch1 inhibition group (P<0. 05). Compared with the model group, the brain tissue structure of the Notch1 inhibition group was significantly improved and the number of apoptotic neurons decreased (both P<0. 05) . Conclusions Notch1 signaling pathway-related proteins NICD and HES1 increased, the expression of pro-apoptotic protein Caspase-3 increased, and the expression of anti-apoptotic protein Bcl-2 decreased in SD rats with acute hypoxic-ischemic brain injury. After the inhibition of Notch1 signaling pathway, the apoptosis of nerve cells decreased in SD rats with acute hypoxic-ischemic brain injury. The study was to select suitable silage maize varieties for planting in the western region of Heilongjiang. The agronomic traits, biomass and nutritional quality of 28 varieties were comprehensively evaluated in this experiment by membership function method. The results showed that the highest plant height was Yu Silage 23, which was 340.10 cm. The highest ear position was Silage 5102, which was 161.60 cm. The highest hay yield was M115S with 25 724.23 kg/hm2. The crude protein, starch, acid detergent fiber and neutral detergent fiber contents of the 28 participating varieties ranged from 6.75% to 9.21%, 17.90% to 27.99%, 20.81% to 28.38% and 42.86% to 56.94%. Comprehensive analysis of 28 major agronomic traits, biological yield and nutritional quality of silage maize by the subordinate function method showed that Meng Silage 266, M114D, Bei Silage 1, Yu Silage 23, and Zhongdan Silage 29 were outstanding in the western Heilongjiang region, and were suitable for popularization of planting in this region and areas with similar climatic environments. Extracellular vesicles（EVs） are vesicles with phospholipid bilayer structure actively secreted by cells, which naturally carry various contents derived from mother cells. EVs retain the biological characteristics of mother cells and reflect the status of mother cells during their secretion. In order to understand the biogenesis of EVs and their biological functions in the body, scientists have made many attempts. However, direct detection of EVs has some difficulties and current detection techniques usually use EVs contents as detection objects to achieve the purpose of indirect characterization. The purpose of disease diagnosis can be achieved by analyzing EVs in body fluids, and detailed characterization of EVs can promote the development of precision medicine. EVs belong to endogenous substances and can be used as carriers to deliver drugs, with the advantages of increasing efficacy, reducing drug toxicological effects and passing through biological barriers. When EVs applied as a drug or a carrier for delivering drug, challenges to be addressed include（i） clear characterization of the contents of EVs,（ii） development of suitable assays to monitor EVs in vivo to determine and optimize the dose, route of administration, and potential toxicity of EVs, and（iii） determination of the kind and amount of membrane composition of EVs. The diameter of EVs is nanoscale and the content is small, but it is of great value in clinical application. The selection of appropriate analytical techniques in the application will enable a comprehensive characterization of EVs and further advance our understanding of EVs biology to develop novel diagnostic and therapeutic approaches. In this review, the principles, applications, advantages and disadvantages of analytical techniques in the study of EVs are introduced, with emphasis on the characterization techniques of EVs. Objective To investigate the influence of poor lifestyle on developmental coordination disorder (DCD) in preschool children in a Chinese context and whether there is a dose-response relationship. Methods A total of 3 605 children from 15 kindergartens were randomly selected from 90 kindergartens in urban areas of Yangzhou city by random cluster sampling method. The general characteristics, birth situation and lifestyle of preschool children were collected by electronic questionnaire. The motor coordination ability of the research subjects was assessed by the little developmental coordination disorder questionnaire (little DCDQ). Multivariate logistic regression analysis was carried out with DCD as the dependent variable and lifestyle variable as the independent variable to infer the relationship between poor lifestyle and DCD of preschool children. Results In this study, the detection rate of abnormal DCD was 8.9%, and the detection rate of suspicious DCD was 9.7%. Preschoolers who slept with night lights on (OR=1.38，95%CI: 1.01-1.89) and had high frequency of fast food (OR=3.44, 95%CI: 1.56-7.60) had a higher risk of suspicious DCD. Preschoolers who did not sleep enough (OR=1.68, 95%CI: 1.30-2.18), spent too much time watching video (OR=1.83, 95%CI: 1.16-2.89), did not share meals with their families (OR=2.18，95%CI: 1.31-3.64), and did not exercise enough outdoors (OR=1.90, 95%CI: 1.15-3.14) had a higher risk of abnormal DCD. The increasing adverse lifestyle was associated with higher risk of suspicious and abnormal DCD in preschoolers. Conclusion Adverse lifestyle is associated with suspicious and abnormal DCD in preschool children, which might be dose-responsive, providing theoretical support for the prevention of DCD in children through lifestyle intervention. To study the influence of long-term mine earthquakes on the stress-strain state of slope rock mass and the possibility of influence accumulation, a mine earthquake scene was set to simulate the change of rock mass state in the process of mine earthquakes. The scene is set as 45 years, divided into 9 simulations, and the period is 5 years. In each period, there are 3 times mine earthquakes with magnitude 3 randomly. The representative rock mass state characterization quantities, including plastic zone, displacement and the maximum principal stress, are selected to analyze the law. This paper studies the action forms of mine earthquakes on open pit mine slope, analyzes the reasons for the change of rock mass state caused by these actions, and establishes the relationship between the plastic zone, displacement, the maximum principal stress and mine earthquake vibration. The results show that the displacement of rock mass is mainly affected by mine earthquakes, followed by the plastic zone, and the influence on the principal stress is very small. It has a great influence on the plastic zone and displacement of rock mass with large elevation difference, and the cumulative effect of displacement is obvious, while the cumulative effect of plastic zone is small. It has little effect on the principal stress and does not accumulate. In the long run, the weakening of slope rock mass caused by mine earthquakes is a long-term cumulative process. Objective: To observe the efficacy of agomelatine combined with sertraline in the treatment of depression with insomnia and its effects on sleep quality score, polysomnography (PSG) monitoring parameters and serum neurotransmitters. 80 patients with depression with insomnia who came to our hospital from April 2020 to December 2021 were selected as the observation objects. They were randomly divided into experimental group and control group with 40 patients in each group. The patients in the control group were treated with sertraline, and the patients in the experimental group were treated with agomelatine combined with sertraline. The efficacy, Pittsburgh sleep quality index (PQSI), PSG related index parameters, Hamilton Depression Scale (HAMD) score, serum neurotransmitter levels changes of the two groups were compared. Adverse reactions of the two groups during treatment were recorded. The total clinical effective rate of the experimental group was 90.00% (36/40), which was higher than 67.50% (27/40) of the control group, and the difference was statistically significant (P<0.05). PSQI and HAMD scores of the two groups decreased at 8 weeks after treatment, and the change degree of the experimental group was greater than that of the control group (P<0.05). 8 weeks after treatment, the total sleep time (TST), sleep efficiency (SE), the percentage of 3+4 in non REM sleep phase (SWS), and the sleep time in REM sleep phase (RT) increased, while the percentage of non REM sleep phase 1 (S1) and the percentage of non rem sleep phase 2 (S2) decreased in the two groups, and the degree of change in the experimental group was greater than that in the control group (P<0.05). 8 weeks after treatment, the norepinephrine (NE) and 5-hydroxytryptamine (5-HT) levels in two groups increased, and the degree of increase in the experimental group was greater than that in the control group (P<0.05). There was no difference in the incidence of adverse reactions between the two groups (P>0.05). Agomelatine combined with sertraline in the treatment of depression with insomnia can effectively improve the symptoms of depression and insomnia, and regulate the levels of serum neurotransmitters. Objective To observe the effect of miR-145-5p on renal injury in rats with renal ischemia/reperfusion (I/R）injury，and to explore whether its renal protective effect is related to the regulation of autophagy through protein ki‐ nase B/target of rapamycin (AKT/mTOR）signaling pathway. Methods Forty SD rats were randomly divided into the Sham group，I/R group，I/R+agomir group，and I/R+agomir+LY group，with 10 rats in each group. Renal I/R models were established in all groups except the Sham group. The miR-145-5p agomir (10 nmol/L）was injected via tail vein 3 days before I/R operation in the I/R+agomir and I/R+agomir+LY groups. LY294002 (50 mg/kg）was simultaneously in‐ traperitoneally injected in the I/R+agomir+LY group，while the same volume of PBS was injected through tail vein in the Sham and I/R groups. The levels of serum urea nitrogen (BUN）and creatinine (Scr)，renal tissue injury score，miR-145- 5p expression，apoptosis rate of renal tissue，and the expression levels of pro-apoptotic protein Bax and anti-apoptotic pro‐ tein Bcl-2 were compared. The fluorescence intensity of autophagy-related protein LC3B，the expression levels of LC3Ⅱ/ LC3Ⅰ，Beclin1 and Atg5，and the values of p-AKT/AKT and p-mTOR/mTOR were compared. Results Compared with the Sham group，the expression of miR-145-5p in the I/R group decreased，and the expression levels of miR-145-5p in the I/R+agomir and I/R+agomir+LY groups increased (all P<0. 01). Compared with the Sham group，apoptosis rate and renal tissue injury score in the other three groups increased，and the increase was more obvious in the I/R+agomir and I/R+agomir+LY groups (all P<0. 05). Compared with the Sham and I/R+agomir groups，serum BUN and Scr levels，Bax expression，LC3B fluorescence intensity，LC3Ⅱ/LC3Ⅰ and Beclin1 expression in the I/R and I/R+agomir+LY groups significantly increased，while the expression levels of Bcl-2 and Atg5，p-AKT/AKT and p-mTOR/mTOR significantly de‐ creased (all P<0. 05). There were no significant differences in the above indexes between Sham group and I/R+agomir group，and between I/R group and I/R+agomir+LY group (all P>0. 05). Methods The colorectal carcinoma (CRC) cell HCT116 was divided into blank control, negative control (NC), si-GAS5 and si-GAS5 +miR-21 inhibitor groups. The quantitative real-time polymerase chain reaction (qRT-PCR) was used to test the expressions of miR-21 and GAS5 at 48 h after transfection. The binding site of GAS5 and miR-21 was determined by luciferase reporter array. Cell proliferation ability was detected by CCK-8 assay. Cell colony ability was detected by colony formation assay. Cell invasion and migration abilities were detected by Transwell assay. Cell cycle and apoptosis were examined by flow cytometer (FCM). The protein levels of EMT associated factors including Snail, N-cadherin, vimentin, E-cadherin, stem cell related factors including CD44, SOX2, Oct2, and PTEN/ Akt signal pathway associated factors were examined by western blotting. Results The expression levels of miR-21 in blank, NC, si-GAS5 group were 1.00±0.10, 1.00±0.10, 1.80±0.20, the absorbance values were 0.51±0.02, 0.50±0.01 and 0.65±0.01, the cell clones were 90±4, 91±5, 200±8, the invaded cells were 118±3, 119±3, 150±4, the migrated cells were 110±2, 108±2, 127±2, the cell ratios in G1 phase were (49.3±2.1)%, (50.1± 2.0)% and ( 42.2± 1.1)%, the cell ratios in S phase were ( 19.2± 1.2)%, (20.2±1.1)% and (28.3±2.2)%, the cell apoptotic ratios were (14.4±2.2)%, (14.5±2.1)% and (7.2±1.3)%. These results indicated that inhibition of GAS5 up regulated the expression level of miR- 21, promoted cell proliferation, invasion and migration, decreased G1 -phase cells and increased S-phase cells, and suppressed cell apoptosis (P<0.05). Moreover, inhibition of GAS5 up regulated the expressions of Snail, N-cadherin, vimentin, Sox2, CD44, Oct2 and p-Akt in HCT-116 cells (P<0.05), while down regulated the expressions of E-cadherin and PTEN (P<0.05). Inhibition of miR-21 reversed the impact of GAS5 knockdown on PTEN/ Akt signaling pathway (P<0.05). Conclusion GAS5 can act as a competing endogenous RNA for miR-21, and down regulation of GAS5 can promote the development of CRC by activating the miR-21/ PTEN/ Akt signaling pathway and promoting the acquisition of EMT and tumor cell stemness.
