Objective: To investigate the changes of interleukin-8 (IL-8)and tumor necrosis factor-a (TNF-a)levels in gingival sulcus fluid of patients with chronic periodontitis and theirs clinical significance, to provide reference for clinical diagnosis and treatment. Methods: Selected 60 cases of patients with chronic periodontitis who were collected and treated in our hospital from December 2013 to December 2015 as experiment group, and 60 cases of patients with periodontal health as control group at the same time. Collected the gingival sulcus fluid and recorded the gingival index(GI), gingival sulcus bleeding index(SBI), plaque index(PLI), probing depth(PD) and clinical attachment loss (CAL) at the times of physical examination of the control group, before and after the routine periodontal therapy of patients in the experimental group. Measured and compared the IL-8 and TNF-a levels between the patients of two groups. Results: The GI, SBI, PLI, PD and CAL periodontal clinical indexes of patients in experimental group before treatment were significantly higher than the control group, the difference was statistically significant (P<0.05); After treatment, the periodontal clinical indexes of patients in experimental group were significantly lower than before treatment, the difference was statistically significant (P<0.05), but there was no significant difference compared with the control group (P>0.05). The IL-8 and TNF-α levels in gingival sulcus fluid of patients in experimental group before treatment were significantly higher than the control group, the differences were statistically significant(P<0. 05); After treatment, the IL-8 and TNF-a levels in gingival sulcus fluid of patients in experimental group were significantly lower than before treatment, the differences were statistically significant(P<0.05). The IL-8 levels in gingival sulcus fluid of patients in experimental group before treatment have positively correlation with PD of periodontal clinical indicators (r=0.495, P=0.027), the TNF-a levels have positively correlation with SBI and PD of periodontal clinical indicators (r=0.512, 0.673; P=0.019, 0.012). Conclusion: The IL-8 and TNF-a in gingival sulcus fluid of patients with chronic periodontitis have high levels, which may be associated with the development of chronic periodontitis, and it has positive clinical significance for clinical diagnosis of chronic periodontitis.
