Objective: To study the effect of resveratrol (RES) on the apoptosis, invasion and migration of human osteosarcoma cell line MG-63 through JAK2/STAT3 signaling pathway. Methods: MG-63 cells were cultured in vitro, and MG-63 cells were treated with different concentrations of RES. Annexin V-FITC/PI double staining was used to detect the effects of RES at different times and different concentrations on the apoptosis of MG-63 cells. Scratch test and Transwell invasion test were used to detect the effects of different time and different concentration of RES on the invasion and migration ability of MG-63 cells. Western blotting was used to detect the expression of phosphorylated protein tyrosine kinase 2 (p-JAK2), phosphorylated protein tyrosine kinase 3 (p-STAT3), apoptosis-associated protein B lymphocytoma-2(Bcl-2), Bcl-2 family of pro-apoptotic proteins(Bax), matrix metalloproteinase(MMP)-2 and MMP-9 in MG-63 cells after treated with RES at different times points and concentrations. Results: The higher the RES concentration, and the longer the time, the higher the apoptosis rate of MG-63 cells (P<0.05). The higher the RES concentration, the weaker the migration and invasion ability of MG-63 cells (P<0.05). After RES treatment of MG-63 cells, the expression of p-JAK2, p-STAT3, Bcl-2, MMP-2 and MMP-9 were significantly decreased, while the expression of Bax was significantly increased, and the expression levels of pp-JAK2, p-STAT3, Bax, Bcl-2, MMP-2 and MMP-9 were RES concentration-dependent (P<0.05). Conclusions: RES may promote the apoptosis of human osteosarcoma MG-63 cells by regulating the JAK2/STAT3 signaling pathway, and inhibit the invasion and migration ability of osteosarcoma cells. To investigate the mechanism of MiRNA-125a-5p on inflammatory response, apoptosis and nerve regeneration after spinal cord injury (SCI). Methods: The SCI model of 6-week-old rats was established. 60 rats were randomly divided into control group (n=20), SCI group (n=20) and MiRNA-125a-5p group (n=20). Only laminectomy was performed in the control group, rats in SCI group were injected with the same amount of normal saline, and rats in MiRNA-125a-5p group were injected with MiRNA-125a-5pagomir. The relative RNA expression levels of MiRNA-125a-5p and nuclear factor kB (NF-kB) were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The expressions of inflammatory factors interleukin-1 (IL-1), chemokine receptor-4 (CXCR-4)and monocyte chemokine-1 (MCP-1) were detected by enzyme-linked immunosorbent assay (ELISA). The expressions of apoptosis protein and neuroregulatory factor protein were detected by Western blot. Basso Beattie Bresnahan (BBB) motor ability rating scale was used to evaluate the neuromotor function of rats. Results: Compared with the control group, the expression of MiRNA-125a-5p decreased and the expression of NF-kB increased in SCI group (P<0.05); Compared with SCI group, the expression of MiRNA-125a-5p increased and the expression of NF-kB decreased in MiRNA-125a-5p group (P<0.05). Compared with the control group, the levels of IL-1, CXCR-4and MCP-1 in SCI group were significantly higher (P<0.05). Compared with SCI group, the levels of IL-1, CXCR-4 and MCP-1 in MiR-NA-125a-5p group were significantly lower (P<0.05). Compared with the control group, the expressions of caspase-3, cleaved PARP andbcl-2 protein in SCI group were increased, and the expressions of NCAM1, NL1 and NRG1 protein were decreased (P<0.05); Compared with SCI group, the expression of caspase-3 and cleaved PARP protein in MiRNA-125a-5p group decreased, and the expression of bcl-2,NCAM1, NL1 and NRG1 protein increased (P<0.05). Compared with the control group, the neuromotor function score of SCI group decreased significantly after the 7th day (P<0.05); Compared with SCI group, the neuromotor function scores of rats in MiRNA-125a-5pgroup increased after the 7th day, and the difference was statistically significant (P<0.05). Conclusion:MiRNA-125a-5p inhibits NF-kB signaling pathway, reduces inflammatory response and apoptosis, promotes nerve regeneration and improves the recovery of neuromotor function in rats.
