Objective To investigate the potential effects of polyphyllin Ⅱ （PP Ⅱ） on A431 human squamous-cell carcinoma cells. Methods Human squamous cell carcinoma cell line A431 were cultured with or without PP Ⅱ. The cell proliferation was measured by MTT assay. The change of cell morphology was observed by transmission electron microscopy. The change of cell cycles was measured by flow cytometry. Results The proliferation of A431 human squamous-cell carcinoma cells was significantly inhibited by PP Ⅱ with dose- and time-dependent manners. The ultrastructure changes of A431 cells were observed. The cell cycles of A431 human squamous-cell carcinoma cells were arrested in S phase. Conclusion Polyphyllin Ⅱ have potentially anticancer effect on A431 human melanoma cells in vitro. Inducing the cell cycle arresting and apoptosis of cells may be the potential mechanisms.
