OBJECTIVE: To probe into the correlation between efficacy, blood concentration and dose of levetiracetam in the treatment of children with epilepsy, so as to provide a certain reference for the medication of levetiracetam in children. METHODS: Levetiracetam blood concentration and epilepsy frequency were analyzed by NONMEN software and oracle crystal ball software. RESULTS: NONMEM software analysis showed that the efficacy tended to become better as the levetiracetam blood concentration increased. When the epilepsy control rate was 50%, the corresponding blood concentration was 24 μg/ mL. When the blood concentration reached 36 μg/ mL, the increase of epilepsy control rate was not significant. Analysis results of oracle crystal ball software showed that the optimal dosage of levetiracetam in the treatment of children with epilepsy was from 20 mg/ (kg·d) to 48 mg/ (kg·d). CONCLUSIONS: The blood concentration of levetiracetam has a certain correlation with the efficacy, but it is not that the higher the drug concentration, the better the efficacy. OBJECTIVE: To explore the optimal drug treatment plan of azithromycin sequential therapy and azithromycin-minocycline therapy in the treatment of Mycoplasma pneumoniae pneumonia by using the pharmacoeconomic analysis methods, and obtain the best effect at the lowest cost. METHODS: Retrospective analysis was performed on 322 children with M. pneumoniae pneumonia over 8 years old admitted into pediatrics of the hospital from Aug. 2016 to Dec. 2018. According to the treatment regimen, all patients were divided into group A (azithromycin sequential therapy) with 160 cases and group B ( azithromycin-minocycline therapy) with 162 cases. Through decision tree and SPSS software, cost-effectiveness analysis of two groups was carried out. RESULTS: The improvement time of cough, fever, pulmonary signs and average length of stay in group A were significantly longer than those in group B, with statistically significant differences (P<0. 05). The total effective rates in group A and group B were respectively 89. 4% (143 / 160) and 92. 6% (150 / 162), with no statistically significant difference (P = 0. 529). The curing costs of one case with M. pneumoniae pneumonia in group A and group B were respectively 3 840 yuan and 3 570 yuan. No obvious adverse drug reactions were found in two groups. CONCLUSIONS: For children with M. pneumoniae pneumonia over 8 years old, there is no significant difference between azithromycin sequential therapy and azithromycin-minocycline therapy, and the cost of azithromycin-minocycline therapy is low. It is very common for abnormal deformation measurements to occur before dam crash. Therefore, in order to ensure the safety operation of the dam, accurate models are needed for data analysis and dam deformation prediction. In this paper, a combination model combining principal component analysis (PCA) and grey wolf optimization (GWO) algorithm with support vector machine (SVM) was established for the characteristics of small samples, short sequences and sequence non-linearity in the dam monitoring data sequence. It used PCA extracted the main features of the factors to simplify the factor relationship and to optimize the input of SVM model. At the same time, GWO algorithm was used to optimize the penalty coefficient and kernel function parameters of SVM model. Combined with the nonlinear fitting ability of support vector machine, the model could better reflect the working mechanism of the dam. Taking a concrete multi-arch dam as an example, this paper established statistical model, standard SVM model, PCA-SVM model and PCA-GWO-SVM model respectively, and analyzed the pre? diction results to verify the feasibility of PCA-GWO-SVM model method.
